Abstract. Life cycles, ploidy levels, reproductive modes, and regeneration capacities of laboratory populations of the southern Brazil freshwater planarians Girardia tigrina and Girardia schubarti were studied. The mating behavior and life cycle of different populations from both species were analyzed regarding their karyotype, body size, modes of reproduction, and regeneration potential. Reproduction was evaluated according to fecundity, fertility, and fissiparity indices. In both species we observed that diploid planarians are ∼25% larger than triploid or mixoploid ones and that sexually reproducing populations have more offspring than asexual ones. Cocoon incubation time was shorter for G. tigrina , and G. schubarti showed a higher frequency of spontaneous malformations. G. tigrina was both more fecund (produced more cocoons) and more fertile (produced more hatchlings) than G. schubarti . The effects of alternative food sources (liver or egg yolk), demographic density, and water–salt concentration were evaluated in different populations of both species. All evaluated environmental factors had effects on fecundity and fertility indices and were more prominent for G. schubarti than for G. tigrina . Our data suggest that the populations of G. schubarti were more sensitive to environmental factors, and thus may be useful as bioindicators.  相似文献

The photodocumentation technique was used weekly to observe until the end of the regenerative process of eyes in three species of Dugesia. Morphometric data based on the measurement of 96 samples of the species: Dugesia tigrina, D. schubarti, and D. anderlani were analyzed. A comparative analysis was made through graphic representation of statistical parameters of the following morphometric characters: the length of the transversal and longitudinal axes of the macula alba, pigment spot, and ocular opening. An interesting feature appeared in D. schubarti: the average of ocular opening during the regenerative process is bigger than in the original samples.  相似文献

The karyotypes of Biomphalaria tenagophila collected from Rio de Janeiro, Brazil were studied using the air-drying method. Somatic cells of this species had 2n=36. The 18 chromosome pairs were identified and classified into 3 groups. The diploid cell has 7 pairs of metacentric, 8 pairs of submetacentric, and 3 pairs of subtelocentric chromosomes. Observed chromosomes ranged from 2.4 to 6.4 µm, and the total length was 122.3 µm. This is the first report on the chromosome of B. tenagophila.  相似文献

Cytogenetic studies were performed in two syntopic species of Characidium, C. lauroi and Characidium sp. cf. C. alipioi, from Ribeir?o Grande, Paraíba do Sul river basin. Both species have diploid number 2n=50 chromosomes, but differ in chromosome shape, C-banding pattern and location of nucleolar organizing regions. In Characidium sp. cf. C. alipioi a new type of ZW sex chromosome system composed of equal sized metacentric chromosomes is reported for the first time in the genus Characidium. Species of Characidium with a sex chromosome system form a monophyletic group. Variations in this system are interpreted as resulting from geographic isolation among allopatric species.  相似文献

This is the first record of Culex (Culex) brethesi (Dyar) in Rio Grande do Sul state, Brazil. The species was identified from specimens collected in a sand bar vegetation with the aid of a Nasci's trap, during an expedition for surveillance of the West Nile Virus in July of 2006, in the city of Mostardas, Rio Grande do Sul, Brazil.  相似文献

Six of the seven possible primary trisomics in Agropyron cristatum were produced. Based on morphology, arm length ratios, and C-banding patterns, they were identified as primary trisomics for chromosomes A, B, C, D, E, and G. Agropyron cristatum is one of several species constituting the crested wheatgrass complex. All species in this complex contain one basic genome (P). A study was conducted to produce and identify a primary trisomic series that will be used to map genes to individual chromosomes. A population of 157 plants were generated by crossing autotriploids (PPP) with diploid (PP) A. cristatum: 58 were diploid (2n = 14), 76 were primary trisomies (2n = 15), 17 were double trisomic (2n = 16), 4 were triple trisomics (2n = 14 + 3), 1 was telocentric trisomic (2n = 14 + 1 telo), and 1 was tetratrisomic (2n = 14 + 4). Karyotype analysis of acetoorcein-stained chromosomes was carried out using the CHROMPAC III computer program; for analysis of C-banded karyotypes, the computer imaging analysis program PCAS (Plant Chromosome Analysis System) was used to identify the primary trisomics. Of the 47 primary trisomics analyzed, 21 plants had one extra satellited chromosome E, 18 with the satellited D chromosome, 3 each for chromosomes B and G, and 1 each for chromosomes C and A. Chromosome pairing was studied in trisomies B, D, E, and G. Trisomics for chromosomes B and G were similar in their mieotic behavior. Each had a trivalent frequency of about 60% and pollen stainability of less than 40%. Trisomics for chromosomes D and E had a trivalent frequency of about 30% and pollen stainability of over 70%.  相似文献

The hermaphroditic flatworm, Polycelis nigra, is characterized by two reproductive biotypes which differ with respect to ploidy; sexual individuals are diploid (n = 8, 2× = 16) and pseudogamous parthenogenetic individuals are polyploid (typically 3×). We have collected and karyotyped individuals from 15 sampling sites (13 in mid to northern Italy, one in Great Britain and one in The Netherlands). We found that biotypes can exist alone or in sympatry, and identified purely diploid, mixed diploid-polyploid, and purely polyploid populations. Karyotype data show that in addition to the normal autosome complement, B chromosomes of differing morphology as well as stable aneuploid chromosomes (extra-A) were found almost exclusively in polyploids (11 of 12 sites). We extensively sampled Lago di Toblino (northern Italy), a pure polyploid population characterized by a submetacentric to metacentric, mitotically stable B chromosome, as well as a stable extra-A chromosome. Here, individuals having 1–3 B chromosomes were more abundant (61%) than those having no B's, implying that B chromosome infection has little detrimental effect when occurring in low numbers. Furthermore, 66% of individuals from this population possessed extra-A chromosomes, although it is unclear whether these elements are aneuploid autosomes or B chromosomes of different morphology. The ubiquity of these chromosomes, within asexuals in particular, is suggestive of a correlation between the origination of the elements and the evolution of polyploidy, or may reflect increased tolerance of parthenogenetic genomes to aneuploidy. Five specimens of Rhamdia quelen collected from the Lindóia Stream, PR, Brazil, were cytogenetically analyzed. The diploid chromosome number found was 58, including 30 metacentric, 16 submetacentric, 10 subtelocentric, and 2 acrocentric chromosomes. Supernumerary or B chromosomes, frequently observed in this fish group, were not detected. One of the individuals was triploid, with 3n = 87. A silver-stained nucleolar organizer region was found on a pair of submetacentric chromosomes of the diploid specimens, and on three chromosomes of the triploid individual, confirming triploidy. Treatment with fluorochrome chromomycin A(3) revealed fluorescent bands coincident with those of the silver-stained nucleolar organizer region, in both diploid and triploid individuals, showing that this is a GC-rich region. Heterochromatin distribution was visualized by the C-banding technique, mainly in the terminal chromosome regions of the individuals and was also observed in the pericentromeric regions of some chromosomes and at both telomeres.  相似文献

B chromosomes are additional dispensable chromosomes that may be present in some individuals, populations, or species, which have probably arisen from the A chromosomes but follow their own evolutionary pathway. Supposedly, B chromosomes do not contain major genes except for ribosomal DNA (rDNA) sequences that have been mapped on the supernumerary chromosomes of many plants and animals. This paper is a new report of B chromosome occurrence in plants. B chromosomes with nucleolar organizing regions (NORs) were found in a diploid sample of Nierembergiaaristata D. Don (sub nom. N. stricta Miers) (2n = 2x = 16). This is an extreme case in which B chromosomes possess not only strong nucleolar activity, as revealed by conventional staining methods, AgNOR and fluorescence banding, and fluorescent in situ hybridization (FISH), but also show nucleolar competition with the A chromosomes. The observed phenomenon could be analogous to the nucleolar dominance or 'differential amphiplasty' phenomenon that occurs in interspecific hybrids.  相似文献

A large number of genera in the tropical fish family Characidae are incertae sedis. Cytogenetic analysis was made of four of these species: Astyanax eigenmanniorum, Deuterodon stigmaturus, Hyphessobrycon luetkenii, and H. anisitsi, collected from various hydrographic basins: hydrographic system from Laguna dos Patos/RS, Tramandaí basin/RS and Tibagi River basin/PR. The first two species were collected in their type locality in the State of Rio Grande do Sul. The 2n = 48 karyotype was observed only in A. eigenmanniorum, while the other species had 2n = 50 chromosomes, with different karyotypic formulas. There was weak heterochromatin staining in the pericentromeric region of A. eigenmanniorum, D. stigmaturus and H. luetkenni chromosomes. In H. anisitsi, heterochromatin appeared to be more abundant and distributed in the pericentromeric and terminal regions of the chromosomes; three pairs showed more evident heterochromatic blocks. There were multiple Ag-NORs in all populations, visualized by FISH with an 18S rDNA probe. While D. stigmaturus and H. luetkenii had conserved AgNOR, CMA3 and 18S rDNA sites, the other two species showed intra- and interindividual variation at these sites. The karyotype variability was high, as is common in this group of fish. Different species arising from isolated hydrographic basins maintain an elevated level of karyotype differentiation, mainly with respect to chromosome structure, heterochromatin distribution and rDNA localization. This is the first report with cytogenetic data for D. stigmaturus and H. luetkenii.  相似文献

A comparison of R-banding patterns obtained by 5-bromodeoxyuridine incorporation was made between the chromosomes of two fish species of the genus Astyanax (Characiformes: Tetragonopterinae), A. altiparanae with 2n = 50 chromosomes, and A. schubarti with 2n = 36 chromosomes. The two species present the highest and the lowest chromosome numbers found in this fish genus, respectively, for which the modal chromosome number is 50. R-band homeology was detected, involving eleven chromosomes of A. schubarti and seventeen chromosomes of A. altiparanae, indicating a close chromosomal relationship between the two species, in spite of their great difference in chromosome number. A chromosome fusion in the past history of the group was hypothesized as a possible cause of the discrepant chromosome numbers of the two species.  相似文献

The longnose dace, Rhinichthys cataractae, is a primary freshwater fish inhabiting riffle habitats in small headwater rivers and streams across the North American continent, including drainages east and west of the Continental Divide. The mitochondrially encoded cytochrome b gene (1140 bp) and 2298–2346 bp of the nuclear‐encoded genes S7 and RAG1 were obtained from 87 individuals of R. cataractae (collected from 17 sites throughout its range) and from several close relatives. Phylogenetic analyses recovered a monophyletic R. cataractae species‐group that contained Rhinichthys evermanni, Rhinichthys sp. ‘Millicoma dace’, and a non‐exclusive R. cataractae. Within the R. cataractae species‐group, two well‐supported lineages were identified, including a western lineage (containing R. evermanni, R. sp. ‘Millicoma dace’ and individuals of R. cataractae from Pacific slope drainages) and an eastern lineage (containing individuals of R. cataractae from Arctic, Atlantic, and Gulf slope drainages). Within the eastern lineage of R. cataractae, two well‐supported groups were recovered: a south‐eastern group, containing individuals from the Atlantic slope, southern tributaries to the Mississippi River, and the Rio Grande drainage; and a north‐eastern group, containing individuals from the Arctic slope and northern tributaries to the Mississippi River. Estimates of the timing of divergence within the R. cataractae species‐group, combined with ancestral area‐reconstruction methods, indicate a separation between the eastern and western lineages during the Pliocene to early‐Pleistocene, with a direction of colonization from the west of the Continental Divide eastward. Within the southern portion of its range, R. cataractae likely entered the Rio Grande drainage during the Pleistocene via stream capture events between the Arkansas River (Mississippi River drainage) and headwaters of the Rio Grande. A close relationship between populations of R. cataractae in the Rio Grande drainage and the adjacent Canadian River (Mississippi River drainage) is consistent with hypothesized stream capture events between the Pecos (Rio Grande drainage) and Canadian rivers during the late‐Pleistocene. The population of R. cataractae in the lower Rio Grande may have become separated from other populations in the Rio Grande drainage (upper Rio Grande and Pecos River) and Canadian River during the late‐Pleistocene, well before initiation of recent and significant anthropogenic disturbance within the Rio Grande drainage. © 2014 The Linnean Society of London, Biological Journal of the Linnean Society, 2014, 111 , 317–333.  相似文献

Specimens of Astyanax scabripinnisfrom three different altitudes (1920, 1800 and 700?m) along the Ribeirão Grande stream in the Campos do Jordão region (São Paulo State, Brazil) were investigated. The same diploid number, 2n?=?50, was detected in the three populations, with the following karyotypic constitution: 6M, 22SM, 10ST and 12A. The populations located at 1920 and 1800?m altitude presented a high incidence of B chromosomes varying in number (0–2), shape (meta- and submetacentrics), size (large and small) and sex-related frequency (they were more frequent among females). The two morphologically variant B chromosomes probably evolved from a metacentric macrochromosome, which is the most commonly observed B chromosome in several A. scabripinnispopulations.  相似文献

The leafcutter ants, which consist of Acromyrmex and Atta genera, are restricted to the New World and they are considered the main herbivores in the neotropics. Cytogenetic studies of leafcutter ants are available for five species of Atta and 14 species of Acromyrmex, both including subspecies. These two ant genera have a constant karyotype with a diploid number of 22 and 38 chromosomes, respectively. The most distinct Acromyrmex species from Brazil is A. striatus, which is restricted to the southern states of Santa Catarina and Rio Grande do Sul. Several cytogenetic and phylogenetic studies have been conducted with ants, but the karyotypic characterization and phylogenetic position of this species relative to leafcutter ants remains unknown. In this study, we report a diploid number of 22 chromosomes for A. striatus. The phylogenetic relationship between A. striatus and other leafcutter ants was estimated based on the four nuclear genes. A. striatus shared the same chromosome number as Atta species and the majority of metacentric chromosomes. Nuclear data generated a phylogenetic tree with a well-supported cluster, where A. striatus formed a different clade from other Acromyrmex spp. This combination of cytogenetic and molecular approaches provided interesting insights into the phylogenetic position of A. striatus among the leafcutter ants and the tribe Attini.  相似文献

A unique derived character distinguishes this species from its congeners and all other planarians known to date, namely separate openings of the vasa deferentia at the tip of the penial papilla. Chromosome number, meiotic behavior, and pollen viability were analyzed in 15 species of two genera, Vriesea and Aechmea, native to Rio Grande do Sul, Brazil. This study is the first cytogenetic analysis of these taxa. The chromosome numbers are all n = 25, consistent with the proposed base number of x = 25 for Bromeliaceae. All examined taxa displayed regular bivalent pairing and chromosome segregation at meiosis. Observed meiotic abnormalities include univalents in metaphase I; missing or extra chromosomes and precocious division of centromeres in metaphase II; laggards in telophase I and anaphase II/telophase II. The high pollen viability (>88%) reflects a regular meiosis.  相似文献

Here, we use vertebral microchemistry to investigate the habitat use patterns of the longnose stingray Hypanus guttatus in the northern and northeastern regions of Brazil, and to evaluate the existence of potential nurseries of the species. Samples were collected in Maranhão, Rio Grande do Norte and Pernambuco states between 2008 and 2019. Trace element concentrations of Ba:Ca, Mg:Ca, Mn:Ca and Sr:Ca in vertebrae were determined through laser ablation inductively coupled plasma mass spectrometry. We used one-step permutational ANOVAs to evaluate multi and single-element differences in element:Ca ratios among life stages and sexes across and within sites. Element signatures differed among all sites for both single and multi-element settings. However, there were few differences among life stages within sites, except for Mn:Ca between life stages for Maranhão state, and between sexes for Sr:Ca for both Pernambuco and Rio Grande do Norte, and Ba:Ca for Rio Grande do Norte. Furthermore, all multi and single-element analyses across locations were significant and highlight the strong differences by sites. We also performed a nonmetric multidimensional scaling analysis, which demonstrated the strong differences for samples from Rio Grande do Norte and the other two sites. By analysing the lifetime transects of each sample, we observed that there was little variation in all element:Ca ratios within sites. Therefore, we argue that the longnose stingray likely does not display extensive habitat use shifts and fulfils much of its life cycle within each area. Finally, we obtained no evidence of nurseries for any site and suggest this is likely a pattern across the species distribution. Future studies investigating habitat use with samples from larger specimens (disc width > 90 cm, >15 years) are needed to better understand the species habitat use patterns across all size ranges.  相似文献

Karyotypes and Giemsa N banding patterns of 13 species of different ploidy wheats have been studied by means of a new method of preparing mitotic chromosome sample in plant. Chromosomes of all diploids and Emmer are metacentric chromosomes, while submetacentric chromosomes occur only in T. timopheevii and T. aestivum. The number of satellite chromosome in the polyploid wheats is not the case of the amount of diploid. Diploid species do not show any N-bands except for T. speltoides. The banding pattern of T. timopheevii is different from that of Emmer. According to the results, authors consider: 1. In the evolution of polyploid wheats, certain variances arise frmn morphological and structural variation of chromosomes; 2. T. speltoides is of importance to the evolution of polyploid wheats and may not only involve the origin of the B genome but also the G genome; 3. The origin of the B genome perhaps is polyphyletic, i.e. 2 or more amphidiploids included T. speltoides crossing each other result in the present of B genome by recombination or chromosome substitution, and 4. T. dicoccoide and T. timopheevii may have evolved from a common ancestor.  相似文献

The centromere is a repeat-rich structure essential for chromosome segregation; with the long-term aim of understanding centromere structure and function, we set out to identify cotton centromere sequences. To isolate centromere-associated sequences from cotton, (Gossypium hirsutum) we surveyed tandem and dispersed repetitive DNA in the genus. Centromere-associated elements in other plants include tandem repeats and, in some cases, centromere-specific retroelements. Examination of cotton genomic survey sequences for tandem repeats yielded sequences that did not localize to the centromere. However, among the repetitive sequences we also identified a gypsy-like LTR retrotransposon (Centromere Retroelement Gossypium, CRG) that localizes to the centromere region of all chromosomes in domestic upland cotton, Gossypium hirsutum, the major commercially grown cotton. The location of the functional centromere was confirmed by immunostaining with antiserum to the centromere-specific histone CENH3, which co-localizes with CRG hybridization on metaphase mitotic chromosomes. G. hirsutum is an allotetraploid composed of A and D genomes and CRG is also present in the centromere regions of other AD cotton species. Furthermore, FISH and genomic dot blot hybridization revealed that CRG is found in D-genome diploid cotton species, but not in A-genome diploid species, indicating that this retroelement may have invaded the A-genome centromeres during allopolyploid formation and amplified during evolutionary history. CRG is also found in other diploid Gossypium species, including B and E2 genome species, but not in the C, E1, F, and G genome species tested. Isolation of this centromere-specific retrotransposon from Gossypium provides a probe for further understanding of centromere structure, and a tool for future engineering of centromere mini-chromosomes in this important crop species.  相似文献

Three new species of Habenaria (Orchidaceae) section Nudae from Rio Grande do Sul, southern Brazil, are described and illustrated: Habenaria australis, H. kleinii and H. sobraliana. These are the first records of H. sect. Nudae for the state. Based on the examination of living and dried specimens, a total of 33 species and 400 collections of Habenaria were recorded for Rio Grande do Sul. Based on this survey, an updated checklist of the genus Habenaria for Rio Grande do Sul has been compiled. Four species are known only from this state, while seven other species are restricted to southern Brazil, Argentina and Uruguay. Habenaria hieronymi, previously known only from Argentina, is recorded for the first time from Brazil, and H. brachyphyton, H. ekmaniana and H. melanopoda are new records for Rio Grande do Sul.  相似文献
