T-DNA插入产生的水稻迟抽穗突变体的遗传分析   总被引:9,自引:0,他引:9  
在筛选和鉴定水稻T-DNA插入纯合体的过程中,观察到一个迟抽穗的突变体.对该突变体进行遗传分析的结果表明,分离群体后代中出现正常抽穗、迟抽穗和特迟抽穗3种类型,分离比率为1:2:1,符合一对基因的不完全显性遗传;对突变体及其后代分离群体做Basta抗性检测及PCR分子检测,证实该突变体是由T-DNA插入所引起的,突变性状与T-DNA共分离.该材料可用于插入座位的基因克隆.  相似文献

NaCl胁迫初期 ,Na 主要在根和叶鞘中积累。相应地 ,根和叶鞘液泡膜ATP酶和焦磷酸酶水解活性、依赖ATP和PPi的质子泵活性及Na /H 逆向转运活性均明显增加 ,根和叶鞘的生长没有受到抑制。NaCl胁迫后期 ,Na 开始向地上部分运输并在叶片中积累。此时 ,叶片液泡膜质子泵和Na /H 逆向转运活性开始增加 ,根和叶鞘的Na/K比增加 ,其液泡膜ATP酶和焦磷酸酶水解活性、质子泵活性和Na /H 逆向转运活性下降。相应地 ,根和叶鞘的生长也下降。当保温介质中Na/K比超过 1时 ,液泡膜微囊ATP酶和焦磷酸酶活性均随Na/K比的增加而下降。表明非盐生植物液泡膜质子泵在盐胁迫的初期对Na 在液泡内的积累及其耐盐性起重要作用  相似文献

Basal exocrine pancreatic secretion was studied in anaesthetized and conscious chickens and spontaneous secretion was studied in anaesthetized chickens. Results are compared with those of other vertebrates to estimate the specific pattern of this secretion in birds. The flow of pancreatic juice was greater in conscious chickens than when anaesthetized (Table 1). Amylase activity was greater than that of the other species and was of the same order in anaesthetized and conscious birds (Table 1). A spontaneous exocrine pancreatic secretion was seen to remain after eliminating a large part of the tonic influences (Table 2).  相似文献

对番茄(Lycopersicoin esculentum Mil1.)突变体Epinastics(Epi)及其野生亲本VFN幼苗、成长植株和果实生长发育与成熟特性进行了系统研究和比较分析.Epi突变体从幼苗到果实都有乙烯过量合成,在黄化幼苗部分三重反应、成长植株叶柄严重上位生长、器官脱落和果实加速成熟等多方面表现出明显增强的乙烯反应,与已知的乙烯反应模式相符.强烈的顶端优势、紧凑的植株形态提示乙烯在植物的顶端优势调节中可能起着重要的作用.Epi突变体黄化幼苗项勾缩小、叶片和花瓣衰老延迟,与传统的乙烯反应不符,提示植物不同的生长发育特性由不同的乙烯信号转导途径调控.  相似文献

Naturally occurring new triterpenoid saponins reported from mid-1996 to March, 2007 are reviewed including their physical constants and plant sources, and are compiled in Table 1. New saponins are arranged in Table 1 on the basis of the skeletal structures of their aglycones, e.g., oleanane type, ursane type, lupane type, hopane type, taraxastane type, cycloartane type, lanostane type, tirucallane type, dammarane type, cucurbitane type, and holostane type. The known triterpenoid saponins and prosapogenins of the new saponins, the biological and pharmacological activities of which were published during 1996-2007, are also reviewed together with their plant sources listed in Table 2 according to the skeletal structures of their aglycones in the same fashion as in Table 1. The plant and animal sources of both new and known bioactive triterpenoid saponins are collected in Table 3 in alphabetical order. The biological and pharmacological activities such as antiallergic, antiatherosclerosis and antiplatelet, antibacterial, anticomplementary, antidiabetic, contraceptive, antifungal, anti-inflammatory, antileishmanial, antimalarial/antiplasmodial, anti-obesity, anti-proliferative, antipsoriatic, antispasmodic, antisweet, antiviral, cytotoxic/antitumor, detoxication, gastroprotective, haemolytic, hepatoprotective, immunomodulatory, anti-enzyme, anti-osteoporotic, insecticidal, insulin-like, membrane-porosity, molluscicidal, neuropharmacological, anti-endothelial dysfunction, snake venom antidote, and sweet activities of these saponins or derived prosapogenins are discussed briefly after Table 3.  相似文献

荞麦与大豆叶片中草酸含量差异及其可能的原因   总被引:11,自引:0,他引:11  
用1/5浓度Hoagland营养液培养荞麦和大豆幼苗l0 d后,荞麦叶片、根及其分泌物中的草酸含量均明显高于大豆,说明荞麦叶片的草酸形成能力强.荞麦叶片中存在少量的草酸氧化酶活性,而大豆中未检测到该酶活性,表明荞麦具有一定的降解草酸的能力.乙醇酸氧化酶(GO)催化乙醇酸氧化的活性两种植物之间虽差异不明显,但该酶催化乙醛酸氧化的活性荞麦显著高于大豆.荞麦GO对乙醛酸的Km值明显低于大豆GO,同时其乙醛酸含量也较高,因此其叶片中由乙醛酸形成草酸的速率应高于大豆.由此认为,由乙醛酸氧化生成草酸可能是植物草酸合成限速步骤之一,其反应速率高低可能导致不同种类植物叶片中草酸含量的差异.  相似文献

本实验利用Solt-Farber顺序诱发大鼠肝癌,观察肝组织中GST活性及GST-P含量在化学诱癌中的变化,并观察性激素对大鼠化学诱发肝癌的早期病变中GST-P表达的作用。结果显示无论是GST活性或GST-P的含量,在诱癌至第三周开始升高,第五周升至最高。利用此模式,选择诱癌至第五周,免疫组化法检测各种处理后肝组织中GST-P的表达。发现睾丸假切除的雄性大鼠经化学诱癌后,肝中有高的GST-P表达,睾丸假切除的雄性大鼠诱癌合并雌二醇处理,明显降低肝组织GST-P阳性灶的面积和数量;合并睾丸酮处理,虽减少GST-P阳性灶的面积,但其数量略有升高。与睾丸假切除后诱癌的雄性大鼠相比,切除睾丸的大鼠经诱癌,有更低的GST-P阳性灶的面积;睾丸切除合并雌二醇处理,GST-P阳性灶的面积进一步降低。与仅化学诱癌的卵巢假切除雌性大鼠比,卵巢切除鼠诱癌后,GST-P阳性灶的面积稍有增加;对卵巢切除合用睾丸酮的大鼠诱癌,阳性灶的面积进一部增加。无论性腺切除与否,雄性大鼠比雌性大鼠有更高的GST-P表达。这些结果提示雌激素可抑制而雄激素则可促进化学诱癌大鼠肝中GST-P的表达。这一结果可能与临床上男性较女性易患肝癌有关。  相似文献

利用携带pC3301质粒(含bar和gus基因)的超毒根癌农杆菌菌株EHA105对普通小麦(Triticum aestivum L.)扬麦158进行了遗传转化,对筛选中的抗生素浓度、菌液浓度、共培养温度和时间、幼胚预培养时间、乙酰丁香酮(AS)浓度、洗涤用液及筛选方式等影响转化的几个重要因素进行了讨论.对从294个小麦幼胚外植体中转化得到的5株成活植株进行了PCR和Southern blot分析,结果表明其中2株小麦基因组中整合了外源DNA,转化频率为0.68%.  相似文献

美味猕猴桃和中华猕猴眺子叶愈伤组织系A_(16)N_1,A_(11)B_2和A_(14)N_7,A_(14)B_2的生理分析表明:愈伤组织系A_(16)N_1和A_(14)N_7的原生质体再生细胞能持续分裂。而愈伤组织系A_(11)B_2和A_(14)B_2的原生质体再生细胞不能持续分裂。前两个愈伤组织系的多胺含量高,酚酸含量低,超氧物歧化酶活性高。过氧化物酶活性低,可溶性蛋白质和氨基酸含量高,说明用于取得并分离原生质体的材料的生理状态对原生质体生长、分化有影响。  相似文献

野生动物资源本底是自然保护区开展生物多样性有效管理的基础。为了掌握脊椎动物资源现状和评估保护成效,在2017和2018年,利用样线法和红外相机陷阱法等方法,对陕西青木川国家级自然保护区脊椎动物资源进行了实地调查,并采用专家打分法对该保护区保护成效进行赋值评估。结果表明:(1)青木川自然保护区现已记录野生脊椎动物342种,隶属5纲32目91科233属,其中,国家I级、II级重点保护野生动物分别为10种和41种;与2002年保护区建立初期相比,增加了4目9科46属83种,其中,鸟类物种数增加最多,达55种,两栖类仅增加1种。(2)青木川镇游客总量从2009年的5万余人次/年,发展到2017年的162万人次/年,保护区外围的旅游开发对该保护区脊椎动物及栖息地均有一定的潜在影响。(3)过去30年内土地利用模式变化明显,2001年未建立保护区时,该区域农业用地面积约占总面积的33%,建立保护区后至2018年,农业用地仅占保护区总面积的1%,使得区域内脊椎动物的栖息地面积显著增加。(4)保护区的保护成效显著,主要干扰因素发生变化,表现为保护区脊椎动物物种总数明显增加,栖息地适宜性和区域完整性显著增强,主要干扰因素类型已由早期的农林牧渔活动转变为现在的外围旅游开发。  相似文献

Summary Of two strains ofParamecium aurelia Syngen4, one differs from the normal form by a gene-induced cytpolasmic mutation (Mo-strain), the other by a simple gene mutation (snaky). Both strains have a very similar character: the cells fail to separate with a variable frequency and monster formation results (Table 1).The phenotype of the two strains can be completely normalised by low oxygen tension (Table 2) or by the presence of carbon monoxide. The CO effect is reversible by light (Table 3, 4). 2,4-Dinitrophenol reduces the frequency of monster formation, sometimes to zero (Table 5). Iodoacetic acid, on the other hand, causes a tenfold increase in monster formation over the controls under certain conditions (Table 6).The facts are discussed in relation to the Pasteur phenomenon.  相似文献

Ca植物春小麦幼苗叶片累积脯氨酸的数量随水分胁迫时间和强度递增,累积的数量与品种的抗旱性没有相关性。C_4植物玉米和高粱累积脯氨酸的数量低于小麦,但高粱多于玉米。CAM植物落地生根脯氨酸含量略有变化。旱生植物细枝岩黄芪的同化枝累积脯氨酸的数量高于中生植物钻天杨和槐的叶片,而梭梭的同化校则低于槐而高于钻天杨。表明这些植物累积脯氨酸的数量与它们的抗旱性无夫。因此似不宜用脯氨酸数量的多少作为植物抗旱性的生理指标。  相似文献

利用根癌土壤农杆菌EHA105/pCAMBIA2301对来自大豆成熟种子的胚尖外植体进行遗传转化,并对农杆菌侵染时间长短以及乙酰丁香酮(AS)浓度等影响转化频率的条件进行了探讨.发现浸染时间以20 h为佳,乙酰丁香酮最佳浓度为200 umo1/L,并探讨了恢复培养的重要性.分别从3个大豆品种合丰35、合丰39、东农42得到了转基因植株,GUS染色及Southern杂交结果证明外源基因整合到大豆基因组中,获得转基因大豆的频率达6.4%~12.1%.  相似文献

Using Solt-Farber method for the induction of rat hepatocarcinoma, the changes of the activity of glutathione S-transferase (GST) and the content of placenta form GST (GST-P) were studied during hepatocarcinogenesis, then the effects of sex hormones on the hepatic expression of GST-P were observed using immunohistochemical method. The results showed that both GST activity and GST-P content began to increase at the 3rd week, and reached the highest level at the 5th week (Table 1). Therefore, the 5th week was selected for the study of GST-P expression in the livers of rats treated with different protocol (Fig. 1). It was found that GST-P was highly expressed in the livers of sham-castrated male rats after chemically induced hepatocarcinoma (PLATE I, Fig. 1A, Table 2). When estradiol was administrated to these rats, both the number and area of GST-P positive(+) foci decreased significantly (PLATE I, Fig. 1B, Table 2). While testosterone was administrated instead of estradiol, the decrease of the area but slight increase of the number of GST-P positive foci were found (PLATE I, Fig. 1C, Table 2). After orchiectomy, the areas of GST-P (+) foci in carcinogen treated liver of male rats were smaller than those in rats with sham-orchiectomy and same carcinogen treatment (PLATE I, Fig. 2A, B, Table 3). When the orchiectomized male rats were administrated with estradiol, the areas of GST-P (+) foci decreased further (PLATE I, Fig. 2C, Table 3). In contrast, after ovariectomy of the female rats, the areas of GST-P (+) foci in carcinogen treated livers were slightly increased as compared with those in the rats with sham-ovariectomy and same carcinogen treatment (PLATE I, Fig. 2D, E, Table 3). While the ovariectomized female rats were administrated with testosterone, the areas of GST-P (+) foci increased further (PLATE I, Fig. 2F, Table 3). Regardless of whether castrations were done or not, GST-P expression in livers of male rats induced hepatocarcinoma was higher than in livers of female rats (PLATE I, Fig. 2A, B, D, E, Table 3). These results indicated that estrogen may inhibit but androgen may promote the GST-P expression in the rat liver during hepatocarcinogenesis. This may be related to the higher incidence of liver carcinoma in male than in female.  相似文献

PEG/Hoagland(-0.1 MPa)溶液根际胁迫处理5叶期玉米幼苗24h,明显刺激第5幼叶生长区细胞H?的分泌,比对照高约1.5倍,钒酸钠对此过程有强烈抑制作用。用水溶性多聚物(PEG4000/Dextran T 500)两相分配法分离玉米第5幼叶生长区的质膜,胁迫处理增加了质膜H~+—ATPase对底物的亲和力,K_m降低到对照的?;活力增加约1.5倍。亚胺环己酮对胁迫处理引起的质膜H~+—ATP_(ase)活性增加没有抑制作用,不同程度的胁迫处理会导致膜制剂磷脂/蛋白比率的变化,其比率在0.83~1.05之间时.随比率的增加,质膜 H~+—ATP_(ase)活性迅速上升;比率在1.05~1.37之间时,随比率值增加,活性迅速下降。暗示质膜的磷脂含量变化可能是胁迫导致质膜 H~+—ATP_(ase)活性增加的主要原因。  相似文献

Summary The coat protein of the RNA containing bacteriophage fr has been hydrolyzed and its amino acid composition determined (Table 1). Furthermore, the protein was split with trypsin and the tryptic peptides were separated by column chromatography on Dowex 1 (Figure 1) and purified by paper chromatography and electrophoresis.The amino acid composition of all but one tryptic peptide are given in Table 2. The large peptide T13 which is much more difficult to purify than all other peptides, was isolated by several methods. Its amino acid composition is shown in Table 3. All tryptic peptides are compiled in Table 4.Amino acid sequences have been fully or partially determined for 9 tryptic peptides (Table 5) and the others are presently being investigated.These findings are compared with the results from other RNA phages, especially f2. It is concluded from the available data that the relationship between the coat proteins of the RNA phages is similar to that between the various naturally occurring strains of tobacco mosaic virus whose amino acid sequences are known.

怒江上游三种裂腹鱼类摄食及消化器官比较研究   总被引:1,自引:0,他引:1  
于2017年5和6月以及9和10月在怒江西藏段收集了怒江裂腹鱼(Schizothorax nukiangensis)194尾、裸腹叶须鱼(Ptychobarbus kaznakovi)152尾,热裸裂尻鱼(Schizopygopsis thermalis)117尾。综合运用3 种多元分析方法分析3 种鱼类摄食及消化器官形态的种间差异,结果显示,主要种间差异部位为头部及肠道;种间形态指标均为显著性差异(P < 0.05);3种裂腹鱼的头部形态已有了一定程度的分化。鱼类食物组成及食物竞争情况研究表明,怒江裂腹鱼和裸腹叶须鱼属杂食性偏动物食性鱼类,两者食物重叠指数较高(0.91);热裸裂尻鱼属杂食性偏植物食性鱼类。食物多样性指数的种间差异明显。3种裂腹鱼营养及空间生态位均有分化,摄食与水温、流速和海拔等环境因素密切相关。  相似文献
