We examined diet and diel energy intake of rainbow trout, Oncorhynchus mykiss, of different lengths captured by electrofishing between 1991 and 1997 in the Lee's Ferry tailwater, Colorado River, below Glen Canyon Dam, Arizona. Trout diets reflected a depauperate food base and indicated limited potential of different fish size-groups to partition food resources. As evidenced by relative stomach volumes of ingested matter, mid-sized and large trout tended to consume more algae than did small fish, suggesting that they consumed diets of lower nutritional quality. An energy intake model indicated that median consumers among mid-sized and large fish generally failed annually to surpass estimated maintenance energy requirements and that median consumers among mid-sized trout failed to meet or exceed maintenance requirements during all seasons. In contrast, median consumers among small trout met or surpassed maintenance energy requirements during most years and in summer. Results support a hypothesis that larger rainbow trout in lotic systems are food-limited more often than smaller fish.  相似文献

Real-time polymerase chain reaction was used to monitor the expression of specific genes and confirm that an alteration in level of expression correlates with changes in diet, metabolism, or immune response. To test these probe and primer sets for their ability to monitor changes in gene expression, groups of fish were experimentally challenged either via daylength, vitamin availability, or with pathogenic microorganisms. The probes and primers sets were then used to analyze expression levels of specific gene products from isolated RNA. The expression of certain genes such as myosin in muscle correlated significantly with protein intake and seasonal daylength. Other genes involved with growth and metabolism such as insulin-like growth factor and pyruvate kinase showed lower levels of significance in correlating with planes of nutrition and varied with diet and treatment. However differences for pyruvate kinase were found in early tests done with animals receiving a vitamin-reduced diet. For factors relating immunological status after infection with microbiological pathogens, several factors such as MX-1 and CD-8 correlated with dose of the viral pathogen infectious hematopoeitic necrosis virus. Association of pathogen dose with immunological expression level was much less pronounced when fish were tested with bacterial microorganisms. This work will aid in evaluating the effects of diet on fish health and nutrient utilization and for evaluating the effects of selection of trout strains.  相似文献

黑素细胞皮质激素受体(MC4R)是跨膜G蛋白偶联受体。MC4R在人和鼠的体重、能量稳态和采食量的调控中具有重要作用,是第一个发现的与人类显性遗传疾病性肥胖相关的靶位点。虹鳟(Oncorhynchus mykiss)属于冷水性鱼类,具有很好的药用和食用价值,但生长缓慢。本研究根据斑马鱼的MC4R基因保守区的核苷酸序列设计引物,通过PCR扩增出虹鳟的MC4R基因,纯化后测序。本实验测出虹鳟MC4R基因968bp,并发现其与其它鱼类的MC4R进行了同源性分析,构建基因进化树。  相似文献

Terrestrial plant resources are increasingly used as substitutes for fish meal and fish oil in fish feed in order to reduce the reliance of aquaculture on marine fishery resources. Although many studies have been conducted to assess the effects of such nutritional transition, no whole breeding cycles of fish fed diets free from marine resources has been reported to date. We therefore studied the reproductive performance of trout after a complete cycle of breeding while consuming a diet totally devoid of marine ingredients and thus of n-3 long chain polyunsaturated fatty acids (n-3 LC-PUFAs) that play a major role in the formation of ova. Two groups of female rainbow trout were fed from first feeding either a commercial diet (C, marine and plant ingredients), or a 100% plant-based diet (V, blend of plant proteins and vegetable oils). Livers, viscera, carcasses and ova were sampled at spawning and analyzed for lipids and fatty acids. Although the V-diet was devoid of n-3 LC-PUFAs, significant amounts of EPA and DHA were found in livers and ova, demonstrating efficient bioconversion of linolenic acid and selective orientation towards the ova. Some ova were fertilized to assess the reproductive performance and offspring survival. We observed for the first time that trout fed a 100% plant-based diet over a 3-year breeding cycle were able to produce ova and viable alevins, although the ova were smaller. The survival of offspring from V-fed females was lower (-22%) at first spawning, but not at the second. Our study showed that, in addition to being able to grow on a plant-based diet, rainbow trout reared entirely on such a diet can successfully produce ova in which neo-synthesized n-3 LC-PUFAs are accumulated, leading to viable offspring. However, further adjustment of the feed formula is still needed to optimize reproductive performance.  相似文献

Freshwater salmonids exposed to low environmental pH typically suffer a net loss of ions, primarily Na+ and Cl, across the gills, resulting in reduced plasma and tissue ion concentrations. However, in recent experiments in our laboratory, juvenile rainbow trout, Oncorhynchus mykiss, fed a ration of 1% body weight d–1 or greater showed no ionoregulatory disturbance during chronic, sublethal acidification. This raised the possibility that these fish had acclimated to low pH in that they would be better able to withstand further, more severe acidification than fish that had no prior experience of acid conditions: previous studies had concluded that such acclimation does not occur. This hypothesis was tested by measuring unidirectional ion fluxes during a 24h acute acid challenge (pH 4.2) in juvenile rainbow trout that had previously been exposed to either ambient pH 6.2 (naive fish) or sublethal low pH 5.2 (acid pre-exposed fish) for 90 days, and fed a ration of either 1.0 or 0.25% d–1 (wet basis). No mortalities were observed during the acute acid challenge in the fish fed the higher ration and no differences between the two groups in the response of Na+ fluxes were observed. Sodium influx in both groups was significantly inhibited throughout the challenge and Na+ net flux was significantly stimulated over the first 6h. Prior to the acute acid challenge, the fish fed the lower ration that had previously been exposed to pH 5.2 had significantly lower plasma ion concentrations than those fish previously exposed to pH 6.2. Both groups suffered mortalities; those of the naive fish (22% by 24h) being markedly lower than those of the acid pre-exposed fish (68% by 24h). However, there were no significant differences in either Na+ or Cl fluxes between the two groups of fish during the acid challenge: both showed significant inhibition of ion influxes and significantly greater net ion losses, resulting in reduced plasma ion concentrations. These results indicate that rainbow trout are unable to acclimate to environmental acidification irrespective of the availability of dietary salts.  相似文献

The use of microsatellites for studies of population structure, as markers in genome mapping, and for parentage control has become increasingly popular in recent years. However, the presence of null alleles can lead to confounding results when using microsatellites. In the Omy3DIAS microsatellite, the presence of a null allele was discovered by analysis of family material. The null allele was sequenced after amplification with new primers located farther away from the repeat sequence. The null allele was shown to be caused by a deletion of a 4-bp sequence, which was part of a repetitive sequence within one of the primer recognition sites. As this phenomenon has been seen in other cases of null alleles, this observation leads to the recommendation to avoid repetitive sequences of any kind within primer sequences. Nine percent of the rainbow trout (Salmo gairdneri) from a hatchery source have a greater than 100-fold increase in expression of a phosphoglucomutase (PGM) locus, Pgm1, in the liver but have normal expression of this locus in other tissues. The results of genetic crosses are consistent with a single regulatory gene with additive inheritance being responsible for the differences in the amount of PGM activity in the liver.—The allele responsible for the expression of Pgm1 in the liver is apparently a recent mutation. This is supported by its restricted distribution in rainbow trout and the absence of liver Pgm1 expression in closely related species. This genetic system is valuable for future analysis of the control of gene expression and in determining the relative evolutionary importance of genetic variation at structural and regulatory genes.  相似文献

Smears were taken from the eyes, spleen and blood from rainbow trout collected from a fish farm and classified as clinically sick, under treatment, recovered and apparently healthy. The smears were examined microscopically and the bacteria present identified as Aeromonads or Streptococci. The results indicated a high incidence of Aeromonads in the eye and the spleen but not the blood in all groups including those apparently healthy while streptococci were present in the blood as well as the eye and the spleen.  相似文献

Effects of melanocyte-stimulating hormone (MSH) and beta-endorphin on lipid mobilization were examined in the rainbow trout (Oncorhynchus mykiss). Plasma levels of fatty acid (FA) were measured after intra-arterial administration of alpha-MSH, desacetyl-alpha-MSH, beta-MSH, or beta-endorphin through a cannula in the dorsal aorta. Desacetyl-alpha-MSH at 1 ng/g body weight resulted in an increase in plasma FA levels 1-3 hr after the injection, whereas the other three peptides showed no significant effect at the same dose. There was no significant change in plasma levels of cortisol after administration of any of the peptides. Lipolytic enzyme activity in the liver was significantly increased in a dose-related manner 1 hr after single intra-peritoneal injection of desacetyl-alpha-MSH. The direct effect of desacetyl-alpha-MSH on lipolysis was examined in liver slices incubated in vitro. Lipase activity in the liver slice was stimulated in the medium containing desacetyl-alpha-MSH in a dose-related manner. The results indicate that desacetyl-alpha-MSH is a potent stimulator of lipid mobilization in the rainbow trout.  相似文献

As global aquaculture fish production continues to expand, an improved understanding of how environmental factors interact in fish health and production is needed. Significant advances have been made toward economical alternatives to costly fishmeal-based diets, such as grain-based formulations, and toward defining the effect of rearing density on fish health and production. Little research, however, has examined the effects of fishmeal- and grain-based diets in combination with alterations in rearing density. Moreover, it is unknown whether interactions between rearing density and diet impact the composition of the fish intestinal microbiota, which might in turn impact fish health and production. We fed aquacultured adult rainbow trout (Oncorhynchus mykiss) fishmeal- or grain-based diets, reared them under high- or low-density conditions for 10 months in a single aquaculture facility, and evaluated individual fish growth, production, fin indices, and intestinal microbiota composition using 16S rRNA gene sequencing. We found that the intestinal microbiotas were dominated by a shared core microbiota consisting of 52 bacterial lineages observed across all individuals, diets, and rearing densities. Variations in diet and rearing density resulted in only minor changes in intestinal microbiota composition despite significant effects of these variables on fish growth, performance, fillet quality, and welfare. Significant interactions between diet and rearing density were observed only in evaluations of fin indices and the relative abundance of the bacterial genus Staphylococcus. These results demonstrate that aquacultured rainbow trout can achieve remarkable consistency in intestinal microbiota composition and suggest the possibility of developing novel aquaculture strategies without overtly altering intestinal microbiota composition.  相似文献

Feeding behavior of adult rainbow trout (Oncorhynchus mykiss)is released by visual and/or chemical stimuli. Detection ofeither a conditioned visual or a conditioned chemical stimuluscreates an excitatory feeding state within the central nervoussystem which turns on feeding behavior composed of swimming,turning and biting/snapping actions. Particular amino acidsthat are highly effective physiological taste stimuli that arealso detected through olfaction (e.g. L-proline, L-alanine,L-leucine) release the initial sequence of food searching andbiting/snapping behaviors; however, an effective olfactory,but poor gustatory, stimulus (e.g. L-arginine) is rarely effectivebehaviorally. After bilateral removal of the paired olfactoryorgans, visual stimuli alone release the entire set of feedingbehavior patterns. Since amino acids that are highly potentphysiological taste stimuli do not release either feeding behavioror reflex biting/snapping actions in adult anosmic rainbow trout,it is postulated that the olfactory system detects potent tastestimuli and provides the afferent input for arousal and therelease of all feeding activity patterns. Cytochalasin B action has been investigated in polyploidisation and gynogenesis in Atlantic salmon and rainbow trout. Solutions of 10, 20, 30 and 50 mg/1 were used to treat eggs from 35 h° to the 2-, 4- and 8-cell stages for rainbow trout, from 35 h° to the 2- and 4-cell stages, and 50 h° to the 8-cell stage for salmon. Survival rate was low in all treated groups. Low frequencies of polyploid and gynogenetic individuals were observed in embryos and alevins by means of two methods: DNA analysis by flow cytometry and chromosome counting. All polyploids observed were triploids without evidence of mosaicism. A few spontaneous diploid gynogenetic rainbow trout were registrated, as well as spontaneous triploid salmon, in controls.  相似文献

The older literature suggests that the development of the blood vascular system in teleosts differs from that of other vertebrates. The evidence, however, came mostly from studies of salmonid embryos beyond the stages when blood cells had begun to circulate, which overlooked earlier developmental stages. The development of the blood vessels of the rainbow trout are illustrated from the time of the first heartbeat to the stage of eye pigment formation. Unless they can be injected with a dye solution, the earliest vessels to develop remain invisible until blood flow makes them visible. By the time of the first heartbeat stage, the embryo has a dorsal aorta, caudal artery and vein, a few transverse vessels, and even the beginning of a vitelline network. One feature peculiar to teleosts is the development of the intermediate cell mass, from which the erythrocytes and a temporary capillary network are formed rather than from the yolk sac. Development of the early posterior cardinal and the subintestinal vein occurs much as in other vertebrates. Previous investigators missed these earliest phases of development because of the difficulty of making them visible. Early formation and transformation of the vascular system of the rainbow trout generally conforms to that seen in vertebrates, except as modified by the temporary presence of the intermediate cell mass and the specialized teleostean yolk mass. With the reduction of the intermediate cell mass, the primary circulatory system for yolk utilization is transformed into a secondary one for respiratory and metabolic functions, as happens usually among vertebrates. The distribution of 35S-labelled sulfadiazine and 14G-labelled trimethoprim was studied in rainbow trout by use of whole body autoradiography and liquid scintillation. As compared to mammals, gastrointestinal absorption and elimination were slow. Accumulation in the skin and the uveal tract of the eye was observed for both drugs tested. The results also indicated that the bile was an important route of excretion. Considerable radioactivity was still present in the skin at 144 hr. survival time.  相似文献

采用显微记数法和使用日立7170A全自动生化分析仪及贝克曼CX-3电解质分析仪,检测水温14℃、19℃和23℃时虹鳟血液红细胞数量和血清有机成分与无机微量元素的含量或活性。结果表明:水温14—23℃范围内,虹鳟红细胞数量随水温上升急剧减少;血清中谷丙转氨酶活性随水温上升而升高,谷草转氨酶活性随水温上升先升高后降低;血清中总蛋白、球蛋白含量随水温上升而增加,白蛋白、血糖和总胆固醇含量随水温上升先增加后减少,甘油三酯含量随水温上升而减少;血清中铜元素含量变化比率大,钾、磷、钙、镁、钠、氯、铁元素含量变化不显著。分析了虹鳟在温水环境中生化因子变化规律,为冷水性鱼类虹鳟温水驯化和基因工程技术提供可借鉴的技术资料。  相似文献
