Abstract Radioisotope equilibration techniques have been used to determine the intracellular concentration of K+, Na+ and Cl?, together with the unidirectional ion fluxes across the plasmalemma of Porphyra purpurea. Influx and efflux of 42K+, 24Na+ and 36C1? are biphasic, the rapid, initial uptake and loss of tracer from individual thalli being attributable to desorption from extracellular regions. Cellular fluxes are slower and monophasic, cells discriminating in favour of K+ and Cl? and against Na+. A comparison between the equilibrium potential of individual ion species and the measured membrane potential demonstrates that there is an active component of K+ and Cl? influx and Na+ efflux. ‘Active’ uptake and ‘passive’ loss of K+ and Cl? are reduced when plants are kept in darkness, suggesting that a fraction of the transport of K+ and Cl? may be due to ‘exchange diffusion’ (K+/K+ and Cl?/Cl?antiport).  相似文献

Static and dynamic light-scattering measurements are reported on zinc-insulin at room temperature (21 ± l°C) and pH = 6.88 in 0.1M NaCl aqueous solution. The experiments were performed at very low concentration, in the range 0.12 × 10?4 to 0.90 × 10?4 g cm?3. Within experimental error, we find no evidence for a critical micellar concentration in this system. The aggregation phenomenon starts immediately after preparation of the solutions, and takes several days to come to stable equilibrium. The concentration dependence of the diffusion coefficients, D z, = Do (1 — kDC), is negative, and kD was observed to decrease as a function of time, while the aggregate size was found to increase. The equivalent concentration coefficient, ?2BM W, obtained from static light scattering, showed a similar behavior, and, within experimental error, was found to be numerically equal to kD. From the relation found between the diffusion coefficient at infinite dilution and the molecular weight of the aggregates, log D0 = ?0.240 log M w ? 5.077, we deduce that the insulin aggregates are compact structures with a characteristic radius of 0.71 Å/(dalton)1/3, surrounded by a hydration layer of a thickness of 8.0 Å. The Na+ efflux from Na+-rich yeast cells into a cation-free medium is largely balanced by the excretion of organic anions. In the presence of Rb+, K+, or high levels of H+ (pH 3–4), the Na+ efflux is increased and the organic anion excretion is suppressed so that stoichiometric cation exchanges occur. H+ participates in the exchanges, moving into or out of the cells depending on the external pH and on the concentration of external Rb+(K+). The total cation efflux is dependent on the external Rb+ concentration in a "saturation" relationship, but the individual cations in the efflux stream are not. The discrimination factor in the efflux pathway between H+ and Na+ is very large (of the order of 10,000), and between Na+ and K+ considerable (of the order of 50). For the latter pair, the recycling of K+ from the cell wall space is an important factor in the discrimination. In addition, the Na+ efflux as a function of Na+ content follows a sigmoidal curve so that the discrimination factor is increased at high levels of cellular Na+. Although the influx and efflux pathways behave as a tightly coupled system, the mechanism of coupling is not entirely clear. A single system with different cation specificities and kinetic behaviors on the inside and outside faces of the membrane could account for the data.  相似文献

Abstract Effects of temperature on the ionic relations and energy metabolism of Chara corallina were investigated. Measurements were made of the ionic content, tracer ion fluxes, and photosynthetic and dark CO2 fixation in isolated cells, and of O2 exchange in photosynthesis and respiration in isolated shoot apices. The total intracellular concentration of K+, Na+ and Cl? was the same in cells held for 5 days in non-growing medium at 15°C (the growth temperature) as in those held at 25°C or 5°C. The tracer influx in the light of all ions tested (Rb+, Na+, CH3NH3+, Cl? and H2PO4?) was lower at 5°C than at 15°C in experiments in which cells were subjected to 5°C for less than 24 h in toto. The influx at 25°C was greater than that at 15°C for H2PO?4, there was no difference between the two temperatures for Na+, while the influx at 25°C was less than that at 15°C for Cl?, Rb+ and CH3NH3+ For Cl? and H2PO?4 similar results were found in later experiments with cells grown at 20—23°C. Photosynthetic CO2 fixation and O2 evolution, and respiratory O2 uptake, are greater at 25°C, and lower at 5°C, than they are at the growth temperature of 15°C. In longer-term pretreatments at the different temperatures, tracer Cl? influx at 15°C and particularly at 25°C were lower than in short-term experiments, while the influx at 5°C was higher. It was concluded from these experiments, and from previous data on H+ free energy differences across the plasmalemma, that (1) the maintenance of internal ion concentrations involves a close balancing of influx and efflux of K+, Na+ and Cl? at all experimental temperatures; (2) the regulation of the tracer fluxes of the ions is kinetic rather than thermodynamic and (3) that the tracer fluxes at low temperatures are not restricted by the rate at which respiration or photosynthesis can supply energy to them.  相似文献

Agropyron elongatum [Host. (Beauv.)] [cv. Arizona Glendale, was grown in liquid medium salinized with either NaCl, KCI, or a 50:50 mixture of these two salts at osmotic potentials ranging from 0 to –1.6 MPa. The amount of growth in 21 days was measured, and extracts were made of the shoots at this time. The extracts were assayed for low-molecular-weight organic compounds (glucose, fructose, sucrose, be-taine, proline) and inorganic solutes (Na+, K+, Cl?, P.). The purpose was to determine if there was any correlation between the harmful effect of salinity on growth and the concentrations of solutes in tissues. Growth inhibition of A. elongatum was roughly proportional to the osmotic potential of the growth medium and was independent of the ionic composition of the salinizing salts. Total monovalent cation (the sum of Na+ and K+) concentrations and the ratio of these two cations in leaves were mainly a function of the ionic compostion of the salt in growth media, and, to a lesser degree, of osmotic potentials. F At an osmotic potential of –0.2 MPa, total monovalent cation in leaves was the same as in non-stressed plants. However, if the salinizing salt contained NaCl, there was an increase in foliar Na+ with a balancing decrease in K+. At stress levels between –0.4 and –1,6 MPa, and, if the media were salinized with either 100% NaCl or a 50:50 mixture of NaCl and KCI, total monovalent cation concentrations remained constant at a value that was twice that in non-stressed plants. Although total monovalent cation concentrations were equal in plants grown under these two salinity conditions, the K+/Na+ ratios shifted from a value of 1:2 in plants grown in 100% NaCl to 3:1 in plants subjected to the 50:50 mixture. If 100% KCI was used to salinize media, total monovalent cation was 80% of its concentration in NaCl-treated plants in the range of –0.4 to -1.2 MPa. At –1.6 MPa due to 100% KCI, total monovalent cation was double that in plants subjected to -0.4 MPa. In the range of osmotic potentials from–0.2 to –1.2 MPa, the chloride:cation ratio was 1:2. At –1.6 MPa the ratio changed to 3:4. Proline started accumulating in leaves of A. elongatum when the tissue concentration of total monovalent cation exceeded 200 μ (g fresh weight)?1. Above this threshold value of total monovalent cation, the proline concentration of leaves was 6% of the amount of total monovalent cation that exceeded 200 umol (g fresh weight)1.  相似文献

Summary When tracer Na+ is added to the solution bathing the apical side of isolated epithelia the observed transepithelial tracer influx increases with time until a steady state is reached. The build-up of the tracer flux follows a single exponential course. The halftime for this build-up under control conditions was 0.92 ±0.06 min, and in the presence of ouabain 4.51±0.7 min. It is shown that the calculated Na+-transport pool is located in the cells. The Na+-transport pool under control conditions was 35.6 ±3.4 nmol/cm2, which corresponds to an intracellular Na+ concentration of 7.9mm. Activation of the active Na+ transport by addition of antidiuretic hormone resulted in a highly significant increase in the Na+ transport pool, and inhibition of the transcellular Na+ transport with amiloride resulted in a decrease in the Na+-transport pool.Furthermore, the active Na+ transport increased along anS-shaped curve with increasing intracellular Na+ concentration (Na+-transport pool). The Na+ pump was found to be half saturated at an intracellular Na+ concentration of 12.5mm.  相似文献

Right-side-out plasma membrane vesicles were isolated from wheat roots using an aqueous polymer two-phase system. The purity and orientation of the vesicles were confirmed by marker enzyme analysis. Membrane potential (Ψ)-dependent 22Na+ influx and sodium/proton (Na+/ H+) antiport-mediated efflux across the plasma membrane were studied using these vesicles. Membrane potentials were imposed on the vesicles using either K+ gradients in the presence of valinomycin or H+ gradients. The ΔΨ was quantified by the uptake of the lipophilic cation tetraphenylphosphonium. Uptake of Na+ into the vesicles was stimulated by a negative ΔΨ and had a Km for extrav-esicular Na+ of 34.8 ± 5.9 mol m3. The ΔΨ-dependent uptake of Na+ was similar in vesicles from roots of hexaploid (cv. Troy) and tetraploid (cv. Langdon) wheat differing in a K+/Na+ discrimination trait, and was also unaffected by growth in 50 mol m?3 NaCl. Inhibition of ΔΨ-dependent Na+ uptake by Ca2+ was greater in the hexaploid than in the tetraploid. Sodium/proton antiport was measured as Na+-dependent, amiloride-inhibited pH gradient formation in the vesicles. Acidification of the vesicle interior was measured by the uptake of 14C-methylamine. The Na+/H+ antiport had a Km, for intravesicular Na+ of between 13 and 19 mol m?3. In the hexaploid, Na+/H+ antiport activity was greater when roots were grown in the presence of 50 mol m?3NaCl, and was also greater than the activity in salt-grown tetraploid wheat roots. Antiport activity was not increased in a Langdon 4D chromosome substitution line which carries a trait for K+/Na+ discrimination. It is concluded that neither of the transport processes measured is responsible for the Na+/K+ discrimination trait located on the 4D chromosome of wheat.  相似文献

At 0°C, when Na+ was the only cation present in the incubation medium, increasing the Na+ concentration from 3 to 10 mM enhanced the affinity of [3H]l-[2-(di-phenylmethoxy)ethyl]-4-(3-phenyl-2-propenyl)piperazine ([3H]GBR 12783) for the specific binding site present in rat striatal membranes without affecting the 5max. For higher Na+ concentrations, specific binding values plateaued and then slightly decreased at 130 mM Na+. In a 10 mM Na+ medium, the KD and the Bmax were, respectively, 0.23 nM and 12.9 pmol/mg of protein. In the presence of 0.4 nM [3H]GBR 12783, the half-maximal specific binding occurred at 5 mM Na+. A similar Na+ dependence was observed at 20°C. Scatchard plots indicated that K+, Ca2+, Mg2+, and Tris+ acted like competitive inhibitors of the specific binding of [3H]GBR 12783. The inhibitory potency of various cations (K+, Ca2+, Mg2+, Tris+, Li+ and choline) was enhanced when the Na+ concentration was decreased from 130 to 10 mM. In a 10 mM Na+ medium, the rank order of inhibitory potency was Ca2+ (0.13 mM) > Mg2+ > Tris+ > K+ (15 mM). The requirement for Na+ was rather specific, because none of the other cations acted as a substitute for Na+. No anionic requirement was found: Cl-, Br-, and F- were equipotent. These results suggest that low Na+ concentrations are required for maximal binding; higher Na+ concentrations protect the specific binding site against the inhibitory effect of other cations.  相似文献

Sorghum bicolor L. Moench, RS 610, was grown in liquid media salinized with NaCl, KCl, Na2SO4, K2SO4 or with variable mixtures of either NaCl/KCl or Na2SO4/K2SO4 at osmotic potentials ranging from 0 to -0.8 MPa. The purpose was to study the effects of different types and degrees of salinity in growth media on growth and solute accumulation. In 14-day-old plants the severity of leaf growth inhibition at any one level of osmotic potential in the medium increased according to the following order: NaCl < Na2SO4 < KCl = K2SO4. Inhibition of growth by mixtures of Na+ and K+ salts was the same as by K+ salts alone. Roots responded differently. Root growth was not affected by Na+ salts in the range of 0 to -0.2 MPa while it was stimulated by K+ salts. The major cation of leaves was K+ because S. bicolor is a Na+-excluder, while Na+ was the major cation in roots except at low Na+/K+ ratios in media. Anions increased in tissues linearly in relation to total monovalent cation, but not with a constant anion/cation ratio. This ratio increased as the cation concentrations in tissues increased. Sucrose in leaf tissue increased 75 fold in Chloride-plants (plants growing in media in which the only anion of the salinizing salts was Cl?) and 50 fold in Sulphate-plants (the only anion of the salinizing salts was SO42-). Proline increased 60 and 18 fold in Chloride- and Sulphate-plants, respectively, as growth media potentials decreased from 0 to -0.8 MPa. The concentrations of both sucrose and proline were directly proportional to the amount of total monovalent cation in the tissue. Sucrose concentrations began increasing when total monovalent cations exceeded 100 μmol (g fresh weight)?1 (the monovalent cation level in non-stressed plants), but proline did not start accumulating until monovalent cation concentrations exceeded 200 μmol (g fresh weight)?1. Therefore, sucrose seemed to be the solute used for osmotic adjustment under mild conditions of saline stress while proline was involved in osmotic adjustment under more severe conditions of stress. Concentrations of inorganic phosphate, glucose, fructose, total amino acids and malic acid fluctuated in both roots and leaves in patterns that could be somewhat correlated with saline stress and, sometimes, with particular salts in growth media. However, the changes measured were too small (at most a 2–3 fold increase) to be of importance in osmotic adjustment.  相似文献

Swelling of nonenergized heart mitochondria suspended in acetate salts appears to depend on the activity of an endogenous cation/H+ exchanger. Passive swelling in acetate shows a characteristic cation selectivity sequence of Na+ >Li+ >K+, Rb+, Cs+, or tetramethylammonium, a sharp optimum at pH 7.2–7.3, activation by Ca2+, and loss of activity on aging which can be related to loss of endogenous K+. The reaction is nearly insensitive to either addition of exogenous Mg2+ or removal of membrane Mg2+ with EDTA. Each of these characteristics of passive swelling in acetate salts is duplicated in chloride media when tripropyltin is added to induce Cl?/OH? exchange. In contrast to nonenergized mitochondria, swelling of respiring mitochondria has been postulated to depend on electrophoretic uptake of cations in response to an interior negative membrane potential. Respiration-dependent swelling in acetate shows an indistinct cation selectivity sequence with Li+ and Na+ supporting higher rates of swelling at higher efficiency than K+, Rb+, and Cs+. The high rates of respiration-dependent swelling in Li+ and Na+ are inhibited by low levels of exogenous Mg2+ (Ki of 5–10 μm), but a significant swelling with almost no cation selectivity persists in the presences of 2 mm Mg2+. Removal of membrane Mg2+ by addition of EDTA strongly activates the rate of respiration-dependent swelling and converts a sigmoid dependency of swelling rate on Li+ concentration to a hyperbolic one with a Km of about 14 mm Li+. The cation selectivity and Mg2+ dependence of the reaction induced in chloride salts by tripropyltin are identical to these properties in acetate. Energy-dependent swelling in acetate shows optimum activity at pH 6.5 which appears related to the availability of free acetic acid, since the corresponding reaction induced in chloride shows a broad optimum at about pH 7.5. These studies support the concept that monovalent cations enter nonenergized mitochondria by electroneutral exchange with protons but penetrate respiring mitochondria by electrophoretic movement through one or more uniport pathways. They further suggest that both a Mg2+-sensitive uniport with high activity for Na+ and Li+ and a Mg2+-insensitive pathway with little cation discrimination are available in the membrane.  相似文献

Abstract The effect of salinity on the growth and ion concentrations in a number of tobacco cultivars is described. Sodium chloride, at a concentration of 200 mol m?3, hardly affected the fresh weight, but significantly reduced the dry weight. The difference in the response of fresh and dry weights to salt was due to a change in succulence (water per unit leaf area); the latter increased with increasing leaf Na+ and Cl? concentration. Under saline conditions, increasing the external Na+: Ca? ratio by decreasing the Ca2+ concentration increased the accumulation of Na+ and Cl? into the leaf tissue.  相似文献

Abstract: A previous study of the metabolism of 6-[18F]-fluoro-l -3,4-dihydroxyphenylalanine (FDOPA) in rats pretreated with carbidopa contained information amenable to kinetic analysis. Using these data, tracer transfer coefficients and metabolic rate constants were estimated. After intravenous injection, FDOPA in circulation was O-methylated (kD0 = 0.055 min?1), and the metabolite (O-methyl-FDOPA) escaped from plasma with a rate constant (kM?1) of 0.01 min?1. The initial clearance of FDOPA to striatum (KD1) was 0.07 ml g?1 min?1, and the equilibrium distribution volume (VDe) was 0.67 ml g?1. The initial clearance of O-methyl-FDOPA to striatum (KM1) was 0.08 ml g?1 min?1, and the equilibrium distribution volume (VMe) was 0.75 ml g?1. The rate constant of FDOPA decarboxylation (kD3) was 0.17 min?1 in striatum. The elimination of 6-[18F]fluorodopamine (FDA) from striatum suggested an apparent rate constant for monoamine oxidase activity (k7) of 0.055 min?1. 6-[18F]Fluorohomovanillic acid (FHVA) was formed from 6-[18F]fluoro-l -3,4-dihydroxyphenylacetic acid with a rate constant (k11) of 0.083 min?1, and FHVA was eliminated from striatum (k9) with a rate constant of 0.12 min?1. The steady-state concentration ratios of FDA and its metabolites were shown to be functions of these rate constants.  相似文献

Summary The transepithelial fluxes, conductances and permeabilities of Li+, Na+, K+, Cs+, NH 4 + and H3CNH 3 + were studied under ionic concentrations ranging from 12 to 250mm inBufo arenarum gallbladders. When these measurements are carefully corrected in order to get only the component due to the paracellular cation channels, the following results are obtained: (1) The permeability ratios (cationic/anionic) are a decreasing function of salt concentration. (2) The partial conductances through paracellular cationic channels show nonlinear saturable concentration kinetics. (3) Moreover, partial conductance kinetics of K+, Cs+ and NH 4 + present a maximum followed, at higher concentratons, by a negative-slope region. (4) The selectivity sequences obtained from biionic potentials do not agree with those obtained from partial conductance measurements. (5) The unidirectional22Na tracer flux (serosal to mucosal) is inhibited by 63% when the K+ symmetrical concentration in the bathing solutions is raised from 25 to 200mm. (6) When the unidirectional42K fluxes (serosal to mucosal) at 200mm KCl Na-free solutions are compared with K+ partial conductance by means of the Hodgkin and Keynes (Hodgkin, A.L., Keynes, R.D. 1955.J. Physiol London 128:61–88) expression, then factor is 2.0. These results indicate that cations do not follow the independence principle and behave as in single-file diffusion multi-ion pores when crossing the paracellular cation channels ofBufo arenarum gallbladder epithelium.  相似文献

The effects of diet and different constant temperatures on hemolymph cation concentrations (Na+, K+, Mg2+, Ca2+) have been studied in Morimus funereus larvae collected from natural habitat, fed natural (oak or beech bark) or artificial diet, as well as in larvae reared from hatching on an artificial diet. In the hemolymph of larvae maintained under natural conditions Mg2+ was dominant, whereas Na+ concentration was very low. In their natural diets concentrations of Na+ and K+ were very low, while those of Ca2+ and Mg2+ were high. In larvae continuously reared on an artificial diet, hemolymph Mg2+ concentration was significantly decreased and Na+ concentration increased more than fourfold compared to the results obtained in oak-fed larvae. Na+ and K+ are the dominant cations in the artificial diet. The concentrations of K+ and Ca2+ in the hemolymph of larvae fed natural or artificial diet are nearly identical, suggesting the existence of an internal regulatory mechanism in this insect for these cations. The hemolymph cation concentrations of M. funereus larvae are predominantly dependent upon the diet consumed, much less upon the environmental temperatures. The most stable concentrations of cations were observed in larvae continuously fed an artificial diet and exposed to different constant temperatures. There was much less stability in the hemolymph cation concentration in oak larvae fed either natural or artificial food after their transfer to constant temperatures. With respect to the response to the external factors studied, the most sensitive are the Na+ concentrations, the most stable seems to be K+. © 1992 Wiley-Liss, Inc.  相似文献

Summary A transport system for branched-chain amino acids (designated as LIV-II system) inPseudomonas aeruginosa requires Na+ for its operation. Coupling cation for this system was identified by measuring cation movement during substrate entry using cation-selective electrodes. Uptakes of Na+ and Li were induced by the imposition of an inwardly-directed concentration gradient of leucine, isoleucine, or valine. No uptake of H was found, however, under the same conditions. In addition, effects of Na+ and Li+ on the kinetic property of the system were examined. At chloride salt concentration of 2.5mm, values of apparentK m andV max for leucine uptake were larger in the presence of Na+ than Li+. These results indicate that the LIV-II transport system is a Na+(Li+)/substrate cotransport system, although effects of Na+ and Li+ on kinetics of the system are different.  相似文献

In solutions containing DNA and cations of more than one type, the competitive interactions of these cations with DNA can be modeled as an ion exchange process that can be described quantitatively by means of the theoretical approach reported in this paper. Under conditions of experimental interest the radial distribution function of each type of counterion is calculated from the results of canonical Monte Carlo (MC) simulations using the primitive model for DNA (having a helical charge distribution) and for the electrolyte ions. These ions consist of monovalent coions, monovalent counterions intended to represent Na+, and counterions of a second type designated Mz+, having variable size and charge (z ≥ 1). The competitive association of these counterions with DNA is described in terms of D, a parameter analogous to an ion exchange equilibrium quotient. Values of D are calculated from the results of our MC simulations and compared with corresponding predictions of the Poisson–Boltzmann (PB) cell model and with results inferred from analyses of previously published nmr measurements. Over typical experimental concentration ranges (0.02M < [Na+] < 0.20M, 0.001 < [Mz+] < 0.160M), DMC and DPB both are predicted to be relatively independent of the bulk ion concentrations. For various specifications of the size and charge of the competing cation (Mz+), DMC and DPB exhibit similar trends, although the MC simulations consistently predict that the cations bearing a higher charge density than that of Na+ are somewhat stronger competitors than indicated by the PB calculations. For monovalent and divalent competitors of varying radii, theoretical predictions of D are compared with values obtained by fitting nmr measurements. If the hard-sphere radii specified in the simulations are the (hydrated) ionic radii determined from conductance measurements, then the MC predictions and the corresponding nmr results are in reasonable agreement for various monovalent competitors and for a divalent polyamine, but not for Ca2+ and Mg2+.  相似文献

Abstract: The effect of hypoxia on Na+,K+-ATPase and Na+-K+-Cl? cotransport activity in cultured rat brain capillary endothelial cells (RBECs) was investigated by measuring 86Rb+ uptake as a tracer for K+. RBECs expressed both Na+,K+-ATPase and Na+-K+-Cl? cotransport activity (4.6 and 5.5 nmol/mg of protein/min, respectively). Hypoxia (24 h) decreased cellular ATP content by 43.5% and reduced Na+,K+-ATPase activity by 38.9%, whereas it significantly increased Na+-K+-Cl? cotransport activity by 49.1% in RBECs. To clarify further the mechanism responsible for these observations, the effect of oligomycin-induced ATP depletion on these ion transport systems was examined. Exposure of RBECs to oligomycin led to a time-dependent decrease of cellular ATP content (by ~65%) along with a complete inhibition of Na+,K+-ATPase and a coordinated increase of Na+-K+-Cl? cotransport activity (up to 100% above control values). Oligomycin augmentation of Na+-K+-Cl? cotransport activity was not observed in the presence of 2-deoxy-d -glucose (a competitive inhibitor of glucose transport and glycolysis) or in the absence of glucose. These results strongly suggest that under hypoxic conditions when Na+,K+-ATPase activity is reduced, RBECs have the ability to increase K+ uptake through Na+-K+-Cl? cotransport.  相似文献

In order to identify physiological components that contribute to salinity tolerance, we compared the effects of Na+, Mg2+ and K+ salts (NaCl, Na2SO4, MgCl2, MgSO4, KCl and K2SO4), Ca2+ (CaSO4), mannitol and melibiose on the wild type and the single-gene NaCl-tolerant mutants stl1 and stl2 of Ceratopteris richardii. Compared with gametophytic growth of the wild type, stl2 showed a low level of tolerance that was restricted to Na+ salts and osmotic stress. stl2 exhibited high tolerance to both Na+ and Mg2+ salts, as well as to osmotic stress. In response to short-term exposure (3 d) to NaCl, accumulation of K+ and Na+ was similar in the wild type and stl1. In contrast, stl2 accumulated higher levels of K+ and lower levels of Na+. Ca2+ supplementation (1.0 mol m?3) ameliorated growth inhibition by Na+ and Mg2+ stress in wild type and stll, but not in stl2. In addition, under Na+ stress (175 mol m?3) wild-type, stll and stl2 gametopbytes maintained higher tissue levels of K+ and lower levels of Na+ when supplemented with Ca2+ (1.0 mol m?3). stl2 gametophytes were extremely sensitive to K+ supplementation. Growth of stl2 was greater than or equal to that of the wild type at trace concentrations of K+ but decreased substantially with increasing K+ concentration. Supplementation with K+ from 0 to 1.85 mol m?3 alleviated some of the inhibition by 75 mol m?3 NaCl in the wild type and in stl1. In stl2, growth at 75 mol m?3 NaCl was similar at 0 and 1.85 mol m?3 K+ supplementation. Although K+ supplementation above 1.85 mol m?3 did not alleviate inhibition of growth by Na+ in any genotype, stl2 maintained greater relative tolerance to NaCl at all K+ concentrations tested.  相似文献

Abstract Atriplex amnicola was grown at 25, 200 or 400 mol m3 NaCl. Root tissues at different stages of development were investigated for concentrations of K+, Na+ and Mg2+, and in some cases for Cl?. Sugar and starch concentrations were measured for plants grown at 25 or 400 mol m3 NaCl. In the ‘slightly vaeuolated’ root tips, Na+ was only 40 mol m?3 at an external concentration of 400 mol m?3 NaCl. The concentrations of K+ were not affected substantially by external NaCl between 25 mol m?3 and 400 mol m?3. The ‘highly vacuolated’ root tissues had substantially higher concentrations of K+, Na+ and Cl? in plants grown at 200 and 400 mol m 3 NaCl than in plants grown at 25 mol m?3 NaCl. Concentrations of Cr and of the sum of the cations in recently expanded tissue were similar to those in the bulk of the roots, consisting mainly of old cells. However, the K+: Na+ decreased with age; at 400 mol m?3 external NaCl with a K+: Na+ of 0.012, the K+: Na+ in recently expanded 12 mm root tips was as high as 1.6, compared with 0.7 for the bulk of the roots. These ion data were used to estimate cytoplasmic and vacuolar concentrations of K+ and Na +. Such calculations indicated that between 25 mol m3 and 400 mol m?3 external NaCl the concentration of the sum of (Na++K+) in the cytoplasm was maintained at about 180–200 mol m?3 (cell water basis). In contrast, the (Na++ K+) concentration in the vacuole was 170 mol m?3 for plants grown at 25 mol m?3 NaCl and 420 mol 400 mol m?3 NaCl. The expanding root (issues exhibited greatly decreased soluble sugars and starch between dusk and dawn. Ai both times, sugar and starch concentrations in these tissues were 2.5–4.0 times greater in plants grown at 400 mol m?3 NaCl compared with plants grown at 25 mol m?3 NaCl. In contrast, carbohydrate concentrations in expanded root tissues were very similar at 25 and 400 mol m?3 and showed little diurnal fluctuation. This paper considers the causes for the slower growth of A. amnicola at 400 than at 25 mol m”3 NaCl, using the data for the roots described here, and those for the shoots presented in the preceding paper (Aslam et al., 1986). There is no support for possible adverse effects by high internal ion concentrations. Instead, there may be deficiencies in supply of organic solutes for osmotic regulation; during part of the night a limited supply of such solutes may well restrict the rate of expansion of cells in plants growing at 400 mol m?3 NaCl. There is insufficient evidence to decide whether this limitation in the expanding tissues is particularly prominent for the roots or for the shoots.  相似文献

The content of the heterosides floridoside and digeneaside and of the main ions Na+, K+, and Cl? was estimated in 20 species of the Rhodophyceae. Methods for quantitative determination of the heterosides are described. The floridoside content is in the range of 1.5–8% on a dry weight basis (Catenella: up to 22%); the content of digeneaside, exclusively found in species of the Ceramiales, is lower, in the range of 1–2.2% on a dry weight basis. All species investigated have Cl? as main anion, while there is a remarkable diversity in cation composition. Na+ was the major cation in 12 of the species investigated, the others having K+ as main cation.  相似文献
