The accumulation of the molecular forms of acetylcholinesterase (AChE) has been studied in leg muscles during embryonic chick development and in cell cultures initiated with myoblasts obtained from embryos at different stages of development. The collagen-tailed, A12 form appears in leg muscles as soon as day 5 in ovo. An early excision of the lumbar zone of the neural tube at day 2 1/2 in ovo severely delayed the morphological development. In leg muscles dissected at day 12 in ovo from operated embryos, we found that the total amount of AChE activity and particularly the proportion of A12 form were dramatically reduced.

Muscle cells were grown in vitro in a medium supplemented with fetal calf serum. In these conditions, chick muscle cells unequivocally synthesize the A12 form when they originated from muscles which accumulated this form in vivo. In contrast, myoblasts obtained from 5-day old embryo leg muscles did not produce the A12 form either in aneural cultures or in the presence of nerve cells. In relation with previous observations concerning chick myogenesis, we discuss the possibility that this difference reflects the existence of two types of myoblasts. This hypothesis would also explain the results of cocultures performed with nerve cells and normal or demedullated leg muscle myoblasts.  相似文献

The appearance of ESR signals from Photosystem I (PS I) electron acceptors A1 and A0 in water or deuterium oxide suspension was followed using a low-temperature photoaccumulation technique. In deuterated samples the A1 signal was narrowed by a factor of 0.66 compared with the control. This effect was fully reversible upon resuspension of treated samples in H2O. The narrow ESR signal from deuterated A1 had similar power saturation characteristics to the normal signal; however, a signal from a second component resolved by deuteration was saturated at higher microwave powers than the control. The power saturation behaviour of A1 in un-modified reaction centres indicated that it is an anionic semiquinone in a ‘protic’ environment. Deuteration reversibly modified the relative extents of reduction of iron sulphur electron acceptors A and B such that centre B became the more stable electron acceptor. The g-value and line-width of iron sulphur centre X was not modified by deuteration although it appeared to become more efficiently reduced. These results are discussed in the light of current evidence from optical, electron spin polarisation and extraction experiments that suggest that A1 is a quinone, probably vitamin K-1.  相似文献

The effects of brain ischemia on the maximum binding capacity (Bmax) and affinity (Kd) of A1 receptors were studied in the rat cerebral cortex, with an in vitro approach. The results were correlated with changes in 3H-adenosine release, studied under identical experimental conditions. Fifteen minutes of in vitro ‘ischemia’ (hypoxic, glucose-free medium) induced a significant increase in both Bmax (2398±132 fmol/mg protein, 151% of the control, P<0.05) and in Kd (2.43±0.12 nM, 161% of the control, P<0.01). At the same time, an increase in tritium efflux from [3H]-adenosine labeled cerebral cortex slices to 324% of the control was observed. A trend toward normalization was evident 5–15 min after ‘reoxygenation’ (restoring normal medium), but the binding parameters were still altered after 60 min (Bmax 2110±82 fmol/mg protein, Kd 2.26±0.14 nM, P<0.01 vs the corresponding control) as was adenosine release (196% of the control). These findings suggest that the increased availability of adenosine and its receptors may be a defense mechanism against ischemic injury, while the reduced affinity of A1 receptors, possibly due to desensitization, may be a sign of ischemia-induced cellular damage.  相似文献

Phospholipase A2 selectively hydrolyses the ester linkage at the sn-2 position of phospholipids forming lysocompounds. This bioconversion has importance in biotechnology since lysophospholipids are strong bioemulsifiers. The aim of the present work was to study the kinetic behaviour and properties of immobilized phospholipase A2 from bee venom adsorbed into an ion exchange support. The enzyme had high affinity for CM-Sephadex® support and the non-covalent interaction was optimum at pH 8. The activity of immobilized phospholipase A2 was comparatively evaluated with the soluble enzyme using a phospholipid/Triton X-100 mixed micelle as assay system. The immobilized enzyme showed high retention activity and excellent stability under storage. The activity of the immobilized system remained almost constant after several cycles of hydrolysis. Immobilized phospholipase A2 was less sensitive to pH changes compared to soluble form. The kinetic parameters obtained (Vmax 883.4 μmol mg−1 min−1 and a Km 12.9 mM for soluble form and Vmax = 306 μmol mg−1 min−1 and a Km = 3.9 for immobilized phospholipase A2) were in agreement with the immobilization effect. The results obtained with CM-Sephadex®-phospholipase A2 system give a good framework for the development of a continuous phospholipid bioconversion process.  相似文献

Effects of cyclopentenone prostaglandins, Δ12-prostaglandin (PG) J2 and PGA2 on the expression of N-myc in relation to the effects on cell cycle progression were investigated using human neuroblastoma cell line GOTO. Both PGs suppressed M-myc expression within several hours prior to inducing G1 arrest. The N-myc suppression with Δ12-PGJ2 was continued but with PGA2 it was gradually released, followed by the release of G1 arrest. These results suggest that Δ12-PGJ2 and PGA2 inhibit cell cycle progression in strong association with N-myc suppression and Δ12-PGJ2 is more potent and has a longer effect than PGA2.  相似文献

1. The ‘oxygenated’ compound of cytochrome c oxidase used in our experiments is more stable than the compound of previous reports. It is quantitatively reversible to ferrous oxidase.

2. It is best formed with an excess of O2 after reduction with a minimum amount of dithionite. It can also be formed at low O2 tension, but then contains some ferric oxidase.

3. Its formation from ferrocyanide-reduced oxidase remains incomplete and subsequent reduction by dithionite is also incomplete.

4. Cyanide does not inhibit its formation from ferrous oxidase. If only ferricytochrome a but no ferricytochrome a3 is reduced in the presence of cyanide by dithionite, there is no reaction with O2.

5. The anaerobic reduction of ‘oxygenated’ oxidase by dithionite is monophasic and fast. In contrast, that of ferric oxidase is biphasic, with an initial fast reduction of ferricytochrome a followed by a much slower reduction of ferricytochrome a3. The rate of cytochrome a, but not that of cytochrome a3 reduction depends on dithionite concentration.

6. In the presence of dissolved O2, the ferric oxidase reduction comes to a temporary standstill when one-third of the absorbance increase at 444 mμ has been reached.

7. Ethyl hydrogen peroxide reacting with ferrous oxidase forms a compound similar to the ‘oxygenated’ compound.

8. Hydrogen donors known to react with peroxidase-H2O2 complexes, particularly pyrogallol, accelerate the transformation of ‘oxygenated’ to ferric oxidase, though not at a rate comparable to that of cytochrome c.

We describe herein the discovery and development of a series of 4-arylthieno[3,2-d]pyrimidines which are potent adenosine A2A receptor antagonists. These novel compounds show high degrees of selectivity against the human A1, A2B and A3 receptor sub-types. Moreover, a number of these compounds show promising activity in vivo, suggesting potential utility in the treatment of Parkinson’s disease.  相似文献

Pseudomonas fluorescens strain GRS1, PRS9 and their cold tolerant mutants were examined for their tricalcium phosphate (TCP) solubilizing activity in NBRIP (broth) media at 10°C and 25°C. Invariably, all the cold tolerant mutants of GRS1 and PRS9 were found more efficient than their respective wild type counterparts for ‘P’ solubilization activity at 10°C as compared to 25°C. ‘P’ solubilization potential of CRM was found maximum among all the strains followed by CRPF6 and CRPF4. To the best of out knowledge, this is the first report regarding low temperature ‘P’ solubilization activity.  相似文献

The (−)-(11R,2′S)-enantiomer of the antimalarial drug mefloquine has been found to be a reasonably potent and moderately selective adenosine A2A receptor antagonist. Further investigation of this compound has led to the discovery of a series of keto-aryl thieno[3,2-d]pyrimidine derivatives, which are potent and selective antagonists of the adenosine A2A receptor. These derivatives show selectivity against the A1 receptor. Furthermore, some of these compounds have been shown to have in vivo activity in a commonly used model, suggesting the potential for the treatment of Parkinson’s disease.  相似文献

Earlier work of several laboratories established that the yields of radiation-induced ring and dicentric chromosomes are greater when human peripheral blood lymphocytes are irradiated in GH1 some hours after phytohemagglutinin stimulation than if they are irradiated in G0 before stimulation. Post-treatment of lymphocytes irradiated in G0 with the DNA polymerase inhibitor aphidicolin, which is effective against both pol and pol δ, produces a similar increase in ring and dicentric yield. We found that aphidicolin post-treatment was much less effective in increasing ring and dicentric yield increases in cells irradiated in G1 four to five hours after stimulation. Because we had earlier found specific inhibitors of DNA pol ineffective in producing increased yields in either G0 or G1 lymphocytes, we conclude that much of the G0 to G1 increase in yields is mediated by pol δ.  相似文献

The cytotoxic and mutagenic effect of X-irradiation was determined with Chinese hamster ovary cells arrested in the G0/G1 phase of the cell cycle through 9 days incubation in serum-free medium. In comparison with exponential phase cultures, the arrested cells showed increased cytotoxicity and mutation induction over the dose range of 50–800 rad. Exponential cultures showed a linear mutant frequency-survival relationship while the arrested cells showed a biphasic linear relationship. A post irradiation holding period of 24 h does not result in any change in the mutant frequency. The increased sensitivity of the arrested cells to the mutagenic effects of X-rays appears to be a cell-cycle phase phenomenon. Upon readdition of serum, the arrested cells re-enter the cell cycle in a synchronous manner, reaching S phase at 10–12 h. Cells irradiated at 5 h after serum addition, i.e. in G1, show a similar does response for mutant frequency, while those irradiated at 10 h or later, i.e. in late G1, S or G2, show lower mutation induction. These observations are consistent with a chromosome interchange mechanism of mutation induction by X-rays, possibly through interactions between repairing regions of the DNA. Irradiation of cells in the G0/G1 phase allow more time for such interactions in the absence of semiconservative DNA replication.  相似文献

Ultrastructural changes in the mitochondria of intact Ehrlich ascites tumor cells were observed after stimulation by valinomycin of the energy-dependent transport of K+ into mitochondria. The mitochondria in cells taken directly from the animal displayed an orthodox configuration. After repeated washings of the cells, the mitochondria were converted to the ‘condensed’ or ‘aggregated’ state. The addition of valinomycin resulted in a transformation of mitochondria from the condensed to orthodox and markedly swollen forms. Alterations in cell size, O2 uptake, and K+ content accompanied the changes in mitochondrial morphology.  相似文献

We have carried out a comparative study of the developmental profiles of the enzyme acetylcholinesterase, and of its collagen-tailed and globular structural forms, solubilized in the presence of 1 M NaCl, 1% (w/v) sodium cholate and 2 mM EDTA, in the chick retina and optic lobes. The overall acetylcholinesterase activities, both per mg protein and per embryo or chick, are substantially higher in tectum than in retina, from embryonic day 16. The A12 collagen-tailed form of the enzyme is present in similar amounts in the embryonic retina and optic tectum; however, while the A12 activity increases significantly in retina after birth, both by percentage and in absolute terms, the tectal tailed enzyme follows a declining developmental profile, reaching a minimum after 6 months of life. On the other hand, the globular G4 species shows developmental profiles, both in retina and tectum, rather similar to those obtained for the overall enzyme activity, while the G2 and G1 forms are present in comparable concentrations in both tissues. Besides, G4 is the predominant globular form in the chick optic lobe after hatching, G2 and G1 being enriched in the embryonic tectum. In the case of retina, however, all the globular forms contribute more evenly to the total acetylcholinesterase activity, along the developmental period considered.The potential significance of some of the postnatal developmental profiles is discussed in terms of the progressive adjustment of retina and tectum to the requirements of visual function.  相似文献

The importance of the 2′-hydroxyl and 2-amino groups of guanosine residues for the catalytic efficiency of a hammerhead ribozyme has been investigated. The three guanosines in the central core of a hammerhead ribozyme were replaced by deoxyinosine, inosine, and deoxyguanosine, and ribozymes containing these analogues were chemically synthesized. Most of the modified ribozymes are drastically descreased in their cleavage efficiency. However. deletion of the 2-amino group at G8 (replacement with inosine, deoxyguanosine, deoxyinosine) caused little alteration in the catalytic activity relative to that obtained with the unmodified ribozyme. Whereas, deletion of the 2′-amino group at G12 and G5 (replacement with inosine, deoxyinosine, and deoxyguanosine) resulted in ribozymes with drastic decrease in the catalytic activity relative to that obtained with the unmodified ribozyme. In contrast, two uridine residues, U7 and U4, in the ribozyne sequence were replaced by deoxyuridine (dU). The dU4 complex resulted in a decrease in the catalytic rate, with relative cleavage activity that ws about half that observed for the native complex. By comparison, the dU7 complex exhibited a relative cleavage activity within 3.3-fold of that observed with native ribozyme/substrate complex. This result suggests that the 2′-hydroxyl group at U 7 is not essential for activity.

However, deletion of the 2-amino group at G8 or deletion of the 2′-hydroxyl group at G12 caused little alteration in the catalytic activity relative to that obtained with the unmodified ribozyme. In contrast, two uridine residues, U7 and U4, in the ribozyme sequence were replaced by deoxyuridine (dU). The local structures of ‘host’ and ‘guest’ layers of MoS2 intercalated with M(OH)2 (M=Mn, Co and Ni) prepared via interaction of single-layer MoS2 dispersions and solutions of M2+ salts were studied by X-ray absorption spectroscopy. According to M K-edge extended X-ray absorption fine structure (EXAFS) and X-ray absorption near-edge structure (XANES) results, the electronic structure and atomic environment of the M atoms in the intercalates are similar to that of the crystalline hydroxides M(OH)2. In the Ni intercalate, Mo K-edge EXAFS revealed a structural change of the ‘host’ MoS2 layers similar to that reported for water dispersions of MoS2 single layers. S K-edge XANES data indicate that the change is associated with increased electron density on the S atoms in the matrix. SO42− and Mo″ (4 < n < 6) were detected in the intercalated materials exposed to air, suggesting that transition metal intercalation may increase the susceptibility of the MoS2 layers to oxidation.  相似文献

A study of the Fe(II) complexes of P-3A (1) and (−)-desacetamido P-3A (2) abilities to cleave duplex DNA was conducted through examination of single-strand and double-strand cleavage of supercoiled φX174 RFI DNA (Form I) in the presence of O2 to produce relaxed (Form II) and linear (Form III) DNA, respectively. Like Fe(II)-bleomycin A2 and deglycobleomycin A2, Fe(II)-1 and 2 effectively produced both single- and double-strand cleavage of supercoiled φX174 DNA. Unlike Fe(II)-bleomycin A2 or deglycobleomycin A2, Fe(II)-1 and 2 were found to cleave duplex w794 DNA with no discemible sequence selectively suggesting that the polynucleotide recognition of the C-terminus tetrapeptide S subunit of the bleomycins including the bithiazole may dominate the bleomycin A2 DNA cleavage selectivity.  相似文献

The tissue distribution of pertussis toxin-sensitive GTP-binding proteins was examined using specific antibodies raised against the purified -subunit of G0 from bovine brain or against synthetic peptides predicted from cDNAs for distinct Gi subtypes. GTP-binding proteins were partially purified from membrane fractions prepared from rabbit tissues including brain, heart, liver, lung, erythrocytes and neutrophilis. Brain contained both G0 and G1. Gil was also found to be abundant in heart. All peripheral tissues contained tissues contained readily detectable amounts of G12, whereas only barely detectable amounts of Gi2 were found in brain. Gi3 was found to be prominent in erythorocytes and exists as a minor component of G proteins in neutrophils and liver. Thus, Gi2 appears to be widely disseminated in peripheral rabbit tissues, while other pertussis toxin substrates are more limited in their distribution.  相似文献

It is shown by X-ray studies that the compound Ni(HPOB)(NO3)2(MeOH)9 [where HPOB=hexaxis(N-pyridin-4-one)benzene] contains [Ni(MeOH)6]2+ cations hydrogen-bonded to the oxygen atoms of the pyridone units in HPOB, with the resulting six-connectivity at both metal and HPOB producing a three-dimensional network array essentially topologically equivalent to the -Po structure. The pyridone rings in the HPOB molecules are arranged orthogonally to the central C6 ring and the nitrate anions form an unusual (NO3 −)(HPOB)(NO3 −) ‘sandwich’ by a combination of π-stacking and C---HO hydrogen bonds.  相似文献

Estrogenic potency of six triterpene glycosides, Holothurin A, Holotoxin A1, Frondoside A, Cucumarioside A2-2 and Cauloside C, that are natural products and semi-synthesized Ginsenoside-Rh2, were examined with yeast two-hybrid system, including expressed genes of human estrogen receptor, hER, the co-activator TIF2 and lacZ as a reporter gene. Only Ginsenoside-Rh2 exhibited significant moderate estrogenic activity in the concentration range of 10−7 to 10−6 M. Its effect was approximately 30% of the activity of 17β-estradiol applied at half-effective concentration. This indicates Ginsenosides-Rh2 is a weak phytoestrogen. The sea cucumber triterpene glycosides, Holothurin A, Holotoxin A1, Cucumarioside A2-2 and Frondoside A, and plant glycoside Cauloside C had no appreciable estrogenic activity. Data obtained by yeast two-hybrid assay reflect structure–activity relationship between tested compounds and 17β-estradiol. Only Ginsenoside-Rh2 has some similarity in chemical structure with 17β-estradiol that might explain affinity of this glycoside to the hER receptor.  相似文献

Straws of five wheat cultivars, including soft and hard and dwarf and semi-dwarf varieties, were treated with sulphur dioxide (SO2), and the effect of treatment on monosaccharide composition and cell wall (CW) degradation by rumen microorganisms was studied. Despite agrobotanical differences, the monosaccharide profiles of the straw hydrolysates were similar. Glucose, xylose, uronic acids and arabinose comprised about 98% of the total monosaccharides and the ratio of glucose to xylose was generally 2:1. The major and general effect of SO2 treatment was expressed in solubilization of the matrix polysaccharides, cellulose being hardly affected. More than 20% of the permanganate lignin was solubilized by SO2 in four of the cultivars. The ‘Lakhish’ cultivar had the highest lignin content (13.3%), but its lignin proved resistent to SO2 treatment. Organic matter digestibility in vitro (IVD) was maximally increased in four varieties (excluding the ‘Lakhish’ straw) to 77–84%, and the IVD of monosaccharides to 88–93%. The treatment effect on the ‘Lakhish’ variety was weaker, the IVD of organic matter and monosaccharides was only increased to 67 and 77%, respectively. Other straw batches of the ‘Lakhish’ variety have shown a strong response to SO2 treatment similar to that found in the straw batches of the other four cultivars. The present study provides support to previous conclusions about the major biodegradation obstacles in wheat straw. The results of this study imply that mild SO2 treatment is generally effective in increasing the degradability of wheat straw by rumen microorganisms.  相似文献
