为研究不同浓度磺胺嘧啶胁迫下鱼肝脏、肾脏、鳃、肠道、肌肉等组织的病理变化, 以异育银鲫为研究对象, 采用室内模拟半静态法分别将其置于低(1 μg/L)、中(100 μg/L)、高(10 mg/L)三个不同浓度的磺胺嘧啶水环境中养殖28d。结果显示, 不同磺胺嘧啶浓度胁迫下, 异育银鲫的肝脏、肾脏、鳃、肠道、肌肉组织均产生了不同程度的病理变化, 主要表现为肝细胞淤血, 空泡化, 核固缩, 细胞溶解坏死; 肾小管上皮细胞空泡化, 肾小球细胞坏死, 肾间质淤血; 鳃小片上皮细胞肿大、增生, 鳃丝成板状, 鳃小片结构消失; 肠道微绒毛脱落, 黏膜下层增厚, 黏膜皱襞破损; 肌纤维排列疏松、紊乱、断裂。各组织中肝脏、鳃、肾脏损伤尤为严重, 且随着磺胺嘧啶处理时间的延长, 剂量的增加均会导致组织损伤加重。试验可为水产养殖过程中合理使用磺胺类药物提供一定的理论指导。  相似文献

Our study was designed to evaluate effects of an herbicide, pendimethalin on biochemical biomarkers and histopathological indices of the freshwater fish Channa punctata Bloch. Fish were acutely exposed (96 h) to sub-lethal concentrations (0.5 and 0.8 ppb of pendimethalin). Various oxidative stress indicators such as thiobarbituric acid reactive substances levels and protein carbonyl content, as well as antioxidant defenses parameters, such as glutathione-S-transferase (GST), catalase (CAT), reduced glutathione (GSH) and non-protein thiols (NP-SH) levels were studied, using the liver, kidney and gill tissues. Pendimethalin exposure increased lipid peroxidation and protein oxidation processes. There was significant inhibition in levels of GSH and NP-SH. The activity of antioxidant enzymes GST and CAT depleted in all the tissues in a dose dependent manner. The histopathological change in the gill showed necrosis and atrophy of primary and secondary gill lamellae. The tissue damages like degeneration of cytoplasm in hepatocytes, atrophy, formation of vacuoles, are some histopathological changes observed in the liver. The changes in histoarchitechture observed in the kidney included necrosis, cellular hypertrophy and granular cytoplasm. The present study demonstrates the disturbances in antioxidant armamentarium and importance of study in the potential risk assessment of herbicides on fish species.  相似文献

Pathological alterations in various organs of rohu (L. rohita) fingerlings following acute (0, 7.50, 11.25 and 13.75 mg/kg body weight) and subchronic (0, 1.25 and 2.50 mg/kg body weight) single i.p. aflatoxin B1 exposure for 10 and 90 days, respectively, were investigated. Mortality (dose-dependent) was marked only during acute toxicosis. The changes observed in various organs were dose and time dependent. The acute dose groups revealed toxic changes viz., necrotic and vascular changes in liver and gill lamellae; meningitis, congestion in brain, degeneration and inflammatory reaction in heart along with degenerative to necrotic changes in kidney tubules and sloughing of the intestinal mucosa. During subchronic exposure to this toxin, preneoplastic lesions in liver along with changes in spleen, intestine, gill and pancreas were recorded. With low doses of aflatoxin, the fish did not reveal any mortality or external signs other than catchexia and increased pigmentation on scales. In composite culture practice of Indian major carps, this could be of economic significance.  相似文献

Changes in the levels of nitrogen metabolic profiles in gill, brain, liver and muscle tissues of fish, Labeo rohita exposed to sublethal concentration (10 micrograms/L) of fenvalerate for 6, 12, 24 and 48 hours were studied. Ammonia content was decreased, whereas urea and glutamine levels were increased in the tissues of fenvalerate exposed fish. Changes were more pronounced with an increase in the period of exposure. Within the tissues, the alterations in excretory products were in the following order: liver greater than muscle greater than brain greater than gill. The survivability of fish in the polluted habitats might be due to the operation of compensatory mechanism in the metabolic profiles such as detoxification and transamination of more toxic ammonia by conversion to less toxic compounds like urea and glutamine.  相似文献

Freshwater zebrafish and brackish water rainbow trout were exposed to different concentrations of cadmium for up to 6 weeks. The gill morphology was examined by light and electron microscopy, and a morphometric analysis was performed. The morphometric study of the secondary lamellae revealed an increase in the portion exterior to the basal lamina, resulting in an increased diffusion distance, after exposure to cadmium concentrations of 10μg l−1 and above. In both species this was due to an increase in volume of the non-tissue spaces of the secondary lamellar epithelium. Furthermore, the water space between neighbouring secondary lamellae was clearly reduced. Morphological examination revealed some gross alterations compared with control fish. These initially consisted in curling of the secondary lamellae and finally resulted in local teleangiectasia. Partial lifting of the secondary lamellar epithelium from the pillar cells resulted in large non-tissue spaces which were invaded by leucocytes. The first sign of degeneration was observed in the chloride cells, which were characterized by a dispersed cytoplasm and a smooth apical plasma membrane.  相似文献

Histological and protein pattern changes in the gill of the common carp, Cyprinus carpio were investigated following exposure to sublethal concentrations of the organochlorine insecticide, endosulfan. Hyperplasia, lamellar fusion, curling of the tips of primary lamellae, exudation of erythrocytes and bulging of the tips of primary lamellae were the histological lesions observed. Protein pattern of the gill varied at different sampling periods characterised by disappearance of certain fractions and occurrence of additional fractions. It was concluded that the severity of histological changes and protein pattern variations seen in the gill was dependent on both the duration of exposure and the concentrations of endosulfan used (over the range 5—25 % of the 96 hr LC50 value, 5·199ppb).  相似文献

Active compound oleandrin extracted from Nerium indicum (Lal Kaner) leaf has potent piscicidal activity. The piscicidal activity of oleandrin on freshwater fish C. punctatus was both time and dose dependent. Exposure to sub-lethal doses of oleandrin for 24hr and 96hr to fish caused significant alteration in the level of total protein, total free amino acid, nucleic acid, glycogen, pyruvate, lactate and enzyme protease, phosphatases, alanine aminotransferase, aspartate aminotransferase and acetylcholinesterase activity in liver and muscle tissues. The alterations in all the above biochemical parameters were also significantly time and dose dependent. The results show a significant recovery in all the above biochemical parameters, in both liver and muscle tissues of fish after the 7th day of the withdrawal of treatment. Toxicity persistence test of oleandrin on juvenile Labeo rohita shows that fish seed of common culturing carp can be released into rearing ponds after three days of oleandrin treatment. It supports the view that the oleandrin is safer and may be useful substitute of other piscicides for removing the unwanted freshwater fishes from aquaculture ponds.  相似文献

Experiments on hybrid red tilapia Oreochromis sp. were conducted to assess histopathological effects induced in gill tissues of 96 h exposure to waterborne lead (5.5 mg/L). These tissues were investigated by light and scanning electron microscopy. Results showed that structural design of gill tissues was noticeably disrupted. Major symptoms were changes of epithelial cells, fusion in adjacent secondary lamellae, hypertrophy and hyperplasia of chloride cells and coagulate necrosis in pavement cells with disappearance of its microridges. Electron microscopic X-ray microanalysis of fish gills exposed to sublethal lead revealed that lead accumulated on the surface of the gill lamella. This study confirmed that lead exposure incited a difference of histological impairment in fish, supporting environmental watch over aquatic systems when polluted by lead.  相似文献

The evaluation of cytotoxic and apoptotic activities of silver nanoparticles (Ag-NPs) synthesized by aqueous extract of Prosopis farcta was investigated against lung (A549) and colon (HT-29) cell lines. The cytotoxic activity of nanoparticles was performed using MTT assay, while their apoptotic activity was tested using TUNEL method. The obtained results of MTT showed that the cell viability of A549 was dependent on the nanoparticles concentration and incubation time. Therefore, although the cytotoxic effect increased as the Ag-NPs concentration and incubation time heightened, yet the viability of HT-29 cells seems to be dependent only on the incubation time. The apoptotic results of the nanoparticles showed more than 50% of apoptosis on A549 and HT-29 cell lines, which in this case, HT-29 demonstrated 100% apoptosis at concentrations of more than 400 µg/ml. It seems that Ag-NPs synthesized using P. farcta extract can serve as anti-cancer agent in the treatment many cancers through creating or discovering new drug forms.  相似文献

In order to test whether histopathological changes of gills demonstrated a good dose–response relationship with water copper levels, juvenile Nile tilapia Oreochromis niloticus, of both sexes and similar mass (36·3 + 7·7 g), were kept in dechlorinated tap water (temperature 25° C, range ±1° C; pH 6·5–7·5; hardness 74·5 mg l?1 CaCO3) and exposed to 40 and 400 μg l?1 of copper. Gill samples were collected after 3, 7, 14 and 21 days. Six major histopathological changes (oedema, lifting, changes in filament epithelium thickness, lamellar fusion, vasodilatation and aneurisms) and three minor ones (proliferation of the lamellar epithelium, necrosis and adjacent lamellar fusion) were found and their prevalence estimated. The extent and severity of each histopathological change were used to develop a severity gradation scale (SGS). Semi‐quantitative analysis of the histopathological changes and measurements of gill copper deposition levels revealed a good dose– and time–response relationship. Oedemas and aneurisms were significantly correlated with acute exposure periods and lamellar fusion with chronic exposure. Epithelial lifting and changes in filament epithelial thickness were seen at lower and higher metal concentrations, respectively. The data also revealed that the SGS profile of each lesion was dependent of gill copper burden.  相似文献

Compartmentation of liver, kidney muscle and gill tissues in relation to glucose-6-phosphatase and fructose 1,6-diphosphatase was examined in the fishes Labeo rohita, Clarias batrachus and Channa punctatus. The anterior region of the right and left lobes of the liver contained the maximum of fructose 1,6-diphosphatase and glucose-6-phosphatase, while the minimum was in the right and left lobes of gill tissue. Herbivore fish had the highest gluconeogenic enzyme content followed by carnivore and piscivore species. The observed enzymatic variations in the three fish species were discussed.  相似文献

The tissue damage induced by various organic pollutants in aquatic animals is well documented, but there is a dearth of information relating to the histological alterations induced by copper in the spiny lobster. In the present study, intermoult juveniles of the spiny lobster Panulirus homarus (average weight 150–200 g) were exposed to two sublethal concentrations of the copper (9.55 and 19.1 μg/l) for a period of 28 days. The muscle, hepatopancreas, midgut, gills, thoracic ganglion and heart of the lobsters were then dissected out and processed for light microscopic studies. Exposure to copper was found to result in several alterations in the histoarchitecture of the muscle, hepatopancreas, midgut, gills, thoracic ganglion and heart of P. homarus. The alterations included disruption and congestion of muscle bundle in muscle tissue; blackened haemocytes; distended lumen and F cell; necrosis of the tubules of the hepatopancreas; disarrangement of circular muscle of the midgut; accumulation of haemocytes in the haemocoelic space; swelling and fusion of lamellae; abnormal gill tips; hyperplastic, necrotic, and blackened secondary gill lamellae of the gills; damaged neurosecretory cell and sensory and motor fibre; necrotic of the thoracic ganglion; dispersedly arranged muscle bands; clumped satellite cells and nucleus of the heart. The results obtained suggest that the muscle, hepatopancreas, midgut, gills, thoracic ganglion and heart of lobsters exposed to copper were structurally altered. Such alterations could affect vital physiological functions, such as absorption, storage and secretion of the hepatopancreas, digestion of gut and respiration, osmotic and ionic regulations of the gills, which in turn could ultimately affect the survival and growth of P. homarus. Thus, all possible remedial measures should be adopted to prevent the occurrence of copper contamination in the aquatic environment.  相似文献

The aim of the present study was to investigate the effects on gill morphology of the explosive 2,4,6-trinitrotoluene (TNT) in a model fish, the European eel, Anguilla anguilla (Linnaeus, 1758) to assess potential detrimental effects in marine fish due to its presence in dumping areas. Juvenile specimens of A. anguilla were exposed in vivo for 6 and 24 h to 0.5, 1 and 2.5 mg/l nominal concentrations of TNT using dimethyl sulfoxide (0.1‰) as solvent carrier. Histological analysis of gills indicated that TNT induced several structural lesions. After 6h of exposure at 0.5 and 1 mg/l TNT, oedema of some secondary lamellae was evident: this change at the highest dose of 2.5 mg/l led to epithelial lifting and detachment from the endothelium and rupture of the branchial membrane and vascular congestion. After 24 h at 0.5 mg/l, increased oedema in secondary lamellae, extensive epithelial detachment and vascular congestion and dilation of lamellar capillaries and pooling of blood at 1 mg/l were observed. At 2.5 mg/l, epithelial hyperplasia, fusion of adjacent lamellae, obliteration of interlamellar spaces by means of tight junctions were also observed. Moreover, chloride cells proliferated along secondary lamellae, and mucus hypersecretion was evident. The overall results clearly indicate that gills are sensitive targets of TNT.  相似文献

In this study we investigated the acute toxicity of Zinc Nano Particles (ZnO NPs) and bulk ZnO on tilapia fish (Oreochromis mossambicus). Oreochromis mossambicus was exposed to the different concentration of ZnO NPs, ZnO and mixed solution of both ZnO NPs and ZnO (20 ppb, 20 ppb, 20 ppb) respectively for 96 h. A very high impact was recorded in hematological parameters which shows significant increased (p < 0.05) in count of white WBCs and platelets in all the experimental groups compared tocontrol group. The count of RBCs, Hb, hematocrit and MCHC were significantly decreased. The remarkable changes which were recorded during this study were histopathological lesions in the gills of exposed fish including, disorganization of gill lamella, cartilaginous core disruption, lifting of epithelium, loss of secondary gill lamellae, blood congestion, fusion of secondary gills lamellae, shortening of secondary gills lamellae, atrophy and curling. Disassembly were seen in plasma membrane of liver along with blood congestion, pyknosis, necrosis, hyperplasia and formation of vacuoles. Intestinal alterations which were observed include shortening of villi, necrosis, detachment and fusion of villi and extreme goblet cells formation. It is concluded from the present study that high level of ZnO NPs, ZnO and mixed solution has a strong tendency to alter hematological parameters, histological architecture, therefore, the indiscriminate use of ZnO NPs and ZnO can subsidize in reducing the population of Oreochromis mossambicus in natural water bodies.  相似文献

Some fish species, such as killifish, that normally inhabit temperate water environments are also found in extreme thermal environments such as thermal springs. The extent of the adaptations involved is not known. In the present laboratory study, we exposed killifish (Aphanius dispar) acclimated to a normal thermal environment to elevated temperatures (37–40 °C) in which related killifish species live permanently. Our objective was to determine whether there is evidence that killifish have heat-shock characteristics that make permanent adaptation likely. The fish was exposed to this temperature for a period of 44 days and then compared with control fish kept at their normal temperature (24 °C) with respect to growth, survival and histopathology of gill and heart tissues. At the end of the experimental time, the percentage of body weight gain and specific growth rate were significantly lower in fish kept in thermal stress compared with the control group. Feed conversion ratio (FCR) was also significantly affected by water temperature, so that during thermal stress the values of FCR were negative. Fish condition (Condition factor: CF) did not differ significantly between both groups at the end of the experiment. On days 11 and 33, however, CF was significantly lower in the thermal stress group. The gill showed blood congestion in primary lamellae and shortened secondary lamellae in fish kept at 37–40 °C. No specific alterations were found in the cardiac tissue of fish kept in thermal stress conditions. Under thermal stress, 40% of fish survived until the end of the experiment. A preliminary conclusion drawn from this work is that A. dispar, which lives at normal temperatures, shows evidence of adaptability to elevated temperatures that could be a factor in the ability of killifish to adapt permanently over time to thermal environment.  相似文献

Silver nanoparticles possess antibacterial effect for various bacteria; however mechanisms of the interaction between Ag-NPs and bacterial cells remain unclear. The aim of our study was to obtain direct evidence of Ag-NPs penetration into cells of Gram-negative bacterium S. typhimurium and Gram-positive bacterium S. aureus, and to study cell responses to Ag-NPs. The Ag-NPs (most 8–10 nm) were obtained by gas-jet method. S. typhimurium (7.81 × 107 CFU), or S. aureus (8.96 × 107 CFU) were treated by Ag-NPs (0.05 mg/l of silver) in orbital shaker at 190 rpm, 37 °C. Bacteria were sampled at 0.5, 1, 1.5, 2, 5 and 23 h of the incubation for transmission electron microscopy of ultrathin sections. The Ag-NPs adsorbed on outer membrane of S. typhimurium and cell wall of S. auereus; penetrated and accumulated in cells without aggregation and damaging of neighboring cytoplasm. In cells of S. aureus Ag-NPs bound with DNA fibers. Cell responses to Ag-NPs differed morphologically in S. typhimurium and S. aureus, and mainly were presented by damage of cell structures. The cytoplasm of S. aureus became amorphous, while S. typhimurium showed lumping and lysis of cytoplasm which led to formation of “empty” cells. Other difference was fast change of cell shape in S. typhimurium, and late deformation of S. aureus cells. The obtained results showed how different could be responses induced by the same NPs in relatively simple prokaryotic cells. Evidently, Ag-NPs directly interact with macromolecular structures of living cells and are exert an active influence on their metabolism.  相似文献

Linear alkyl benzene sulphonate (LAS), one of the main ingredients used in synthetic detergents to enhance their cleansing properties. Indiscriminate and untreated discharge of detergents and their residues in both lantic and lotic habitats pose a variety of ecological threats and also adversely affect aquatic fauna. In vivo, LAS metabolism and biotransformation occurs via monooxygenases in liver, leading to Reactive Oxygen Species, ROS, production and consequently oxidative stress by disturbing cellular antioxidant enzymatic equilibrium. Present study aims to evaluate the activities of two widely distributed antioxidant enzymes viz., catalase (CAT) and superoxide dismutase (SOD) and ROS induced histological impairments in liver of freshwater fish, Channa punctatus. For the estimation of oxidative stress and hepatic impairments, well acclimatized fishes were divided in three groups. Fish of group G1 serves as control whereas fish of the other two groups, G2 and G3 were exposed to two fractions, 1/20th and 1/10th of 96 h LC50 of LAS for 24, 48, 72 and 96 h of exposure periods. Our results showed a significant induction in CAT and SOD activities in liver tissue of C. punctatus in a dose and time dependent manner. ROS induced histopathological impairments in hepatic tissues are characterized by loosely arranged, irregularly distributed and degenerated hepatocytes with increased vacuolization and pyknotic nuclei. The results are quite suggestive that LAS intoxication generates oxidative stress by ROS production which brings about histopathological impairments in exposed fish.  相似文献

This study was carried out to determine the protective effects of lithium borate (LTB) on blood parameters and histopathological findings in experimentally induced acute cadmium (Cd) toxicity in rats. Twenty-eight male Wistar albino rats were used, weighing 200–220 g, and they were randomly divided into four groups, including one control and the following three experimental groups: a Cd group (0.025 mmol/kg), a LTB group (15 mg/kg/day orally for 5 days), and a LTB + Cd group (15 mg/kg/day orally for 5 days and Cd 0.025 mmol/kg by intraperitoneal injection on the fifth day). All the rats in the study were anesthetized with ketamine at the end of the sixth day, blood was taken from their hearts, and then the rats were decapitated. The values in the control and LTB group were usually close to each other. White blood cell (WBC), neutrophil %, and C-reactive protein (CRP) levels increased in the Cd and LTB + Cd groups while lymphocyte and monocyte levels decreased in a statistically significant manner, in comparison to the other groups. It was determined that the levels of red blood cells (RBCs), hematocrit (Htc), and hemoglobin (Hb) did not change in the groups. The levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in the Cd and LTB + Cd groups significantly increased, in comparison to the other groups, while the glucose, alkaline phosphatase (ALP), albumin (ALB), and total protein (TP) levels decreased. According to histopathological findings in the control and LTB groups, the liver and kidney tissues were found to have normal histological structures. In the Cd group, severe necrotic hemorrhagic hepatitis, mild steatosis, and mononuclear cell infiltration were detected in the liver. In the LTB + Cd group, degeneration and mild mononuclear cell infiltration were found in the liver. Regarding the kidney tissue in the Cd group, severe intertubular hyperemia in both kidney cortex and medulla, as well as degeneration and necrosis in the tubulus epithelium, was observed. In the LTB + Cd group, mild interstitial hyperemia and mononuclear cell infiltration was detected. Resultantly, it can be said that LTB at this dose has non-toxic effects and some beneficial effects for liver and kidney damage caused by acute Cd toxicity.  相似文献

The effects of 96-h sublethal exposure of nitrite (1, 2, 4, 8 and 10.4 mg l(-1)) on selected enzymatic activities in serum and tissues of fingerlings of catla (Catla catla), rohu (Labeo rohita) and mrigal (Cirrhinus mrigala) were studied for the first time in these species. All three species responded almost identically to nitrite exposure. With increasing nitrite concentration, reduction in activities was observed in acetylcholinesterase (AChE) in brain and liver; alkaline phosphatase (ALP) in serum, brain and gill; and acid phosphatase (ACP) in gill, while progressive increase in alanine aminotransferase (ALAT) and aspartate aminotransferase (ASAT) activities in brain, gill and serum, and ACP activity in serum and brain was observed. Lactate dehydrogenase (LDH) activity increased in gill, liver, kidney, brain and serum of all three species with increasing nitrite concentration up to 8 mg l(-1) followed by reduction at 10.4 mg l(-1). The study revealed nitrite stress causing alteration in activities of all measured tissue and serum enzymes in the fingerlings, and so stresses the need for proper management of this particular nutrient in water during carp culture.  相似文献

研究采用组织学方法观察和比较了达里诺尔湖瓦氏雅罗鱼(Leuciscus waleckii, 碱水种)和松花江瓦氏雅罗鱼(淡水种)在相同碳酸盐碱度胁迫下(CA0、CA30和CA50)鳃组织结构的差异, 探究瓦氏雅罗鱼碱水种耐高碱特性与鳃组织结构微观调整的适应性关系。结果显示, 随着碱度增加, 碱水种鳃丝变宽、鳃小片变长、鳃小片间距变大(P<0.05); 淡水种鳃丝变宽、鳃小片间距变大(P<0.05), 鳃小片长度在CA30时显著变长(P<0.05), 而在CA50时与对照组无明显差异(P>0.05)。碱水种在CA30和CA50的氯细胞数量与对照组相比明显增加, CA50的氯细胞排列更加紧密并且有叠加现象, 扁平上皮细胞变大, 细胞表面增厚; 淡水种在CA30时的氯细胞数量明显多于CA50, 但在碱度胁迫下, 鳃小片出现破损, 扁平上皮细胞、柱细胞和血细胞融合、脱落现象严重。另外, 在碱水种和淡水种鳃耙上皮细胞中发现了大量黏液细胞分泌, 随着碱度增加, 黏液细胞由大而稀疏变为小而密集, 其中碱水种的黏液细胞数量较淡水种多, 而且排列更为整齐、密集。鳃组织学研究结果表明, 瓦氏雅罗鱼碱水种通过保持鳃组织结构和生理功能的完整性达到对高碱环境的长期适应, 而淡水种则因鳃细胞融合、脱落造成生理功能丧失, 不能长期适应高碱环境。研究结果可为淡水鱼类在盐碱水的移植驯化提供依据和指导。  相似文献
