The study of infectious disease has been aided by model organisms, which have helped to elucidate molecular mechanisms and contributed to the development of new treatments; however, the lack of a conceptual framework for unifying findings across models, combined with host variability, has impeded progress and translation. Here, we fill this gap with a simple graphical and mathematical framework to study disease tolerance, the dose response curve relating health to microbe load; this approach helped uncover parameters that were previously overlooked. Using a model experimental system in which we challenged Drosophila melanogaster with the pathogen Listeria monocytogenes, we tested this framework, finding that microbe growth, the immune response, and disease tolerance were all well represented by sigmoid models. As we altered the system by varying host or pathogen genetics, disease tolerance varied, as we would expect if it was indeed governed by parameters controlling the sensitivity of the system (the number of bacteria required to trigger a response) and maximal effect size according to a logistic equation. Though either the pathogen or host immune response or both together could theoretically be the proximal cause of pathology that killed the flies, we found that the pathogen, but not the immune response, drove damage in this model. With this new understanding of the circuitry controlling disease tolerance, we can now propose better ways of choosing, combining, and developing treatments.  相似文献

Although behavioral and endocrine consequences of acute exposure to stressors have been extensively studied, little is known about how simultaneous exposure to two different stressors interacts to induce short- and long-term effects. In the present experiment we studied this interaction in adult male rats exposed to cat fur odor (impregnated cloth) or immobilization on boards either separately or simultaneously. We reasoned that exposure to the odor of a potential predator while immobilized, may potentiate its negative consequences as compared to exposure to only one of the stressors. Exposure to cat odor elicited the expected reduction of activity and avoidance of the area where the impregnated cloth was located. The endocrine response (plasma levels of ACTH and corticosterone, as a measure of the hypothalamic-pituitary-adrenal axis, HPA) was markedly greater after immobilization than after cat fur odor and no additive effects were found by simultaneous exposure to both stressors. Cat odor, but not immobilization, increased anxiety-like behavior as evaluated in the elevated plus-maze 7 days after the stressors, with no evidence of enhanced HPA activation. In addition, cat odor exposure resulted in long-lasting (8 days later) fear conditioning to the box containing a clean cloth, which was reflected by hypoactivity, avoidance of the cloth area and enhanced HPA activation. All these effects were similarly observed in rats exposed simultaneously to cat odor and immobilization. In rats only exposed to immobilization, only some weak behavioral signs of fear conditioning were found, but HPA activation in response to the context paired to immobilization was enhanced to the same extent as in cat odor-exposed animals, supporting a certain degree of endocrine conditioning. The present results did not reveal important behavioral interactions between the two stressors when animals experienced both simultaneously, whereas some interactions were found regarding HPA activation. The widespread use of brominated flame retardants (BFRs) has led to increasing concentrations in environmental samples. Previous reports suggested endocrine activity of these compounds. Because thyroid hormones and sex steroids are known to regulate the development of the nervous system, we examined neurobehavioral and endocrine effects of gestational exposure to 2,2′4,4′,5-pentabrominated diphenyl ether (PBDE99) and lifetime exposure to tetrabromobisphenol A (TBBPA) or hexabromocyclododecane (HBCD) in rats. Treatment with PBDE99 (0, 1, or 10 mg/kg bw) reduced serum testosterone and estradiol in male offspring and affected sexual development in both sexes. Exposed males exhibited increased sweet preference, indicating feminization of this sexually dimorphic behavior. In TBBPA and HBCD experiments, a benchmark design was used with dose ranges of 0-3000 mg/kg bw/day and 0–100 mg/kg bw/day, respectively. Because thyroid hormones are critical for auditory development, brainstem auditory evoked potentials (BAEPs) were recorded in young adult offspring. TBBPA exposure elevated BAEP thresholds in females and prolonged latencies in both sexes. Benchmark doses were comparable for auditory effects and decreased plasma T4. HBCD increased BAEP thresholds and delayed BAEP waves only in males, but the relation to thyroid hormones remains to be determined. HBCD also reduced latencies in haloperidol-induced catalepsy, suggesting effects on the dopaminergic system.  相似文献

In this paper we address the interrelated questions of why and how certain features of an organism’s environment become meaningful to it. We make the case that knowing the biology is essential to understanding the foundation of meaning-making in organisms. We employ Miguel Nicolelis et al’s seminal research on the mammalian somatosensory system to enrich our own concept of brain-objects as the neurobiological intermediary between the environment and the consequent organismic behavior. In the final section, we explain how brain-objects advance the ongoing discussion of what constitutes a biosemiotic system. In general, this paper acknowledges Marcello Barbieri’s call for biology to make room for meaning, and makes a contribution to that end.  相似文献

Current methods for estimating past climatic patterns from pollen data require that the vegetation be in dynamic equilibrium with the climate. Because climate varies continuously on all time scales, judgement about equilibrium conditions must be made separately for each frequency band (i.e. time scale) of climatic change. For equilibrium conditions to exist between vegetation and climatic changes at a particular time scale, the climatic response time of the vegetation must be small compared to the time scale of climatic variation to which it is responding. The time required for vegetation to respond completely to climatic forcing at a time scale of 104 yr is still unknown, but records of the vegetational response to climatic events of 500-to 1000-yr duration provide evidence for relatively short response times. Independent estimates for the possible patterns and timing of late-Quaternary climate changes suggest that much of the vegetational evidence previously interpreted as resulting from disequilibrium conditions can instead be interpreted as resulting from the individualistic response of plant taxa to the different regional patterns of temperature and precipitation change. The differences among taxa in their response to climate can lead a) to rates and direction of plant-population movements that differ among taxa and b) to fossil assemblages that differ from any modern assemblage. An example of late-Holocene vegetational change in southern Quebec illustrates how separate changes in summer and winter climates may explain the simultaneous expansion of spruce (Picea) populations southward and beech (Fagus) populations northward.  相似文献

Evolutionary biologists often argue that menopause evolved in the human female as the result of selection for a postreproductive phase of life, during which increased maternal investment in existing progeny could lead to enhanced survivorship of descendents. Adaptive theories relating menopause to enhanced maternal investment are known as the mother (first-generation) and grandmother (second-generation-offspring) hypotheses. Although menopause—universal midlife termination of reproduction—has not been documented in primates other than humans, some researchers have argued that postreproductive alloprimates also have a positive impact on the survivorship of first and second generation progeny. We tested the maternal investment hypotheses in Japanese macaques by comparing the survivorship of offspring, final infants, and great-offspring of females that terminated reproduction before death with females that continued to reproduce until death. SURVIVAL analyses revealed no significant difference in the survivorship of descendents of postreproductive and reproductive females, though final infants of postreproductive females were 13% more likely to survive than final infants of females that reproduced until death were. We also explored possible differences between these two groups of females, other than survivorship of progeny. We found no difference in dominance rank, matrilineal affiliation, body weight, infant sex ratio, age at first birth, fecundity rate or lifetime reproductive success. However, postreproductive females are significantly longer-lived than reproductive females and as a result experienced more years of reproduction and produced more infants in total. Apart from final infants, offspring survival is marginally lower in postreproductive females. Since offspring survival is not significantly enhanced in postreproductive females, the greater number of infants produced did not translate into greater lifetime reproductive success. Our findings fail to support the maternal investment hypotheses and instead suggest that reproductive termination in this population of Japanese macaques is most closely associated with enhanced longevity and its repercussions.  相似文献

Biodiversity is globally recognised as a cornerstone of healthy ecosystems, and biodiversity conservation is increasingly becoming one of the important aims of environmental management. Evaluating the trade-offs of alternative management strategies requires quantitative estimates of the costs and benefits of their outcomes, including the value of biodiversity lost or preserved. This paper takes a decision-analytic standpoint, and reviews and discusses the alternative aspects of biodiversity valuation by dividing them into three categories: socio-cultural, economic, and ecological indicator approaches. We discuss the interplay between these three perspectives and suggest integrating them into an ecosystem-based management (EBM) framework, which permits us to acknowledge ecological systems as a rich mixture of interactive elements along with their social and economic aspects. In this holistic framework, socio-cultural preferences can serve as a tool to identify the ecosystem services most relevant to society, whereas monetary valuation offers more globally comparative and understandable values. Biodiversity indicators provide clear quantitative measures and information about the role of biodiversity in the functioning and health of ecosystems. In the multi-objective EBM approach proposed in the paper, biodiversity indicators serve to define threshold values (i.e., the minimum level required to maintain a healthy environment). An appropriate set of decision-making criteria and the best method for conducting the decision analysis depend on the context and the management problem in question. Therefore, we propose a sequence of steps to follow when quantitatively evaluating environmental management against biodiversity.  相似文献

Few studies have compared the sensitivity of trauma questionnaires to disaster inventories for assessing the prevalence of exposure to natural disaster or associated risk for post-disaster psychopathology. The objective of this analysis was to compare reporting of disaster exposure on a trauma questionnaire (Brief Trauma Questionnaire [BTQ]) to an inventory of disaster experience. Between 2011 and 2014, a sample of 841 reproductive-aged southern Louisiana women were interviewed using the BTQ and completed a detailed inventory about exposure to hurricanes and flooding. Post-traumatic stress disorder (PTSD) symptomology was measured with the Post-Traumatic Stress Checklist, and depression with the Edinburgh Depression Scale. The single question addressing disaster exposure on the BTQ had a sensitivity of between 65% and 70% relative to the more detailed questions. Reporting disaster exposure on the BTQ was more likely for those who reported illness/injury due to a hurricane or flood (74%-77%) or danger (77-79%), compared to those who reported damage (69-71%) or evacuation (64-68%). Reporting disaster exposure on the BTQ was associated with depression (odds ratio [OR] 2.29, 95% confidence interval [CI] 1.43-3.68). A single question is unlikely to be useful for assessing the degree of exposure to disaster across a broad population, and varies in utility depending on the mental health outcome of interest: the single trauma question is useful for assessing depression risk.  相似文献

Our understanding of plant meiosis is rapidly increasing thanks to the model Arabidopsis thaliana. Here we present the results of a screening for meiotic mutants carried out with a library containing 30,719 T-DNA insertion lines. An average of one mutant per 1000 lines was recovered. Several phenotypic classes could be distinguished and are presented. In parallel, 39 proteins known to be involved in meiosis in non-plant organisms were chosen and a search was performed for homologue sequences in the completed Arabidopsis thaliana genome. Approximately 30% of the meiotic related sequences showed similarities with one or several Arabidopsis putative genes. The relevance of forward versus reverse genetics in order to characterize meiotic functions is discussed.  相似文献

This paper undertakes a direct, comprehensive assessment of the long-term sustainability of desertification rehabilitation in China under a plausible but worst case scenario where governmental interventions, in the form of payments for environmental services (PES), will cease. The analysis is based on household behavior as well as experimental data. Our econometric results highlight the main obstacles to the sustainability of rehabilitation programs subsequent to cessation of government intervention, including specific shortfalls in households’ preference for a free ride, budget constraints, attitudes, tolerance of and responsibility for desertification, and dissatisfaction with governmental actions. We conclude that desertification rehabilitation is not sustainable in China without continued governmental intervention. The results of this study are intended to support policy makers as they consider future directions for rehabilitation sustainability.  相似文献

The aim of this work was to study the exposure to magnetic fields of children living at different distances from a power line and to evaluate how well theoretical calculations compared with actual exposure. Personal exposure instruments were carried for 24 h by 65 schoolchildren living 28–325 m from a 300 kV transmission line; the current load was 200–700 A. About half of the children attended a school far from the power line, whereas the other half attended a school located about 25 m from the line. Exposure to magnetic fields was analyzed for three categories of location: at home, at school, and at all other places. Time spent in bed was analyzed separately. The results indicated that children who lived close to a power line had a higher magnetic field exposure than other children. The power line was the most important source of exposure when the magnetic field due to the line was greater than about 0.2 μT. Exposure at school influenced the 24 h time-weighted average results considerably in those cases where the distance between home and power line was very different from the distance between school and power line. The calculated magnetic field, based on line configuration, current load, and distance between home and power line, corresponded reasonably well with the measured field. However, the correlation depends on whether home only or 24 h exposure is used in the analysis and on which school the children attended. The calculated magnetic field seems to be a reasonably good predictor of actual exposure and could be used in epidemiological studies, at least in Norway, where the electrical system normally results in less ground current than in most other countries. Although chronic exposure to high doses of trichloroethene causes tumors of the lung, liver, and kidney in experimental animals, the epidemiology data in humans exposed to trichloroethene as a whole fail to support a causal association between trichloroethene exposure and cancers of the lungs, liver, or kidneys in humans at environmentally relevant concentrations. Environmentally relevant concentrations of trichloroethene are defined as 50 ppb (50 µg/L) in water or 5 ppb (27 µg/m3) in air. Tumor induction by trichloroethene in rodents exposed to very high doses over their whole lifespan has been observed in the kidney of rats and in the lung and liver of mice. Mechanistic data demonstrate that species-specific processes are involved in the carcinogenicity associated with chronic trichloroethene exposure in rodents. Based on these data and the results of recent well-conducted epidemiology studies, it can be concluded that humans exposed to trichloroethene at environmentally relevant concentrations are not at an increased risk for developing cancer.  相似文献

Lithium is widely used in medicine, but its administration can cause numerous side effects. The present study aimed at the evaluation of the possible application of selenium, an essential and antioxidant element, as a protective agent against lithium toxicity. The experiment was performed on four groups of Wistar rats: I (control)—treated with saline, II (Li)—treated with lithium (Li2CO3), III (Se)—treated with selenium (Na2SeO3) and IV (Li?+?Se)—treated with lithium and selenium (Li2CO3 and Na2SeO3) in the form of water solutions by stomach tube for 6 weeks. The following biochemical parameters were measured: concentrations of sodium, potassium, calcium, magnesium, phosphorus, iron, urea, creatinine, cholesterol, glucose, total protein and albumin and activities of alkaline phosphatase, aspartate aminotransferase and alanine aminotransferase in serum as well as whole blood superoxide dismutase and glutathione peroxidase. Morphological parameters such as red blood cells, haemoglobin, haematocrit, mean corpuscular volume, mean corpuscular haemoglobin, mean corpuscular haemoglobin concentration, platelets, white blood cells, neutrophils as well as lymphocytes were determined. Lithium significantly increased serum calcium and glucose (2.65?±?0.17 vs. 2.43?±?0.11; 162?±?31 vs. 121?±?14, respectively), whereas magnesium and albumin were decreased (1.05?±?0.08 vs. 1.21?±?0.15; 3.85.?±?0.12 vs. 4.02?±?0.08, respectively). Selenium given with lithium restored these parameters to values similar to those observed in the control (Ca—2.49?±?0.08, glucose—113?±?26, Mg—1.28?±?0.09, albumin—4.07?±?0.11). Se alone or co-administered with Li significantly increased aspartate aminotransferase and glutathione peroxidase. The obtained outcomes let us suggest that the continuation of research on the application of selenium as an adjuvant in lithium therapy seems warranted.  相似文献

Might the gravity levels found on other planets and on the moon be sufficient to provide an adequate perception of upright for astronauts? Can the amount of gravity required be predicted from the physiological threshold for linear acceleration? The perception of upright is determined not only by gravity but also visual information when available and assumptions about the orientation of the body. Here, we used a human centrifuge to simulate gravity levels from zero to earth gravity along the long-axis of the body and measured observers'' perception of upright using the Oriented Character Recognition Test (OCHART) with and without visual cues arranged to indicate a direction of gravity that differed from the body''s long axis. This procedure allowed us to assess the relative contribution of the added gravity in determining the perceptual upright. Control experiments off the centrifuge allowed us to measure the relative contributions of normal gravity, vision, and body orientation for each participant. We found that the influence of 1 g in determining the perceptual upright did not depend on whether the acceleration was created by lying on the centrifuge or by normal gravity. The 50% threshold for centrifuge-simulated gravity''s ability to influence the perceptual upright was at around 0.15 g, close to the level of moon gravity but much higher than the threshold for detecting linear acceleration along the long axis of the body. This observation may partially explain the instability of moonwalkers but is good news for future missions to Mars.  相似文献

The distribution, density and species composition of juvenile shrimps were studied in three mangrove zones of Pichavaram, situated on the southeast coast of India. Nine species of shrimps dominated with Metapenaeus monoceros, M. brevicornis and Penaeus indicus were recorded. This shrimp resource was higher in mangrove-lined waters than that in neritic waters. This field observation was experimentally proved using litter bags. The average catch around the litter bags was 0.80 individuals day–1 for Rhizophora leaves, 1.6 ind. day–1 for Avicennia leaves and 0.16 ind. day–1 in control. In general, abundance of juvenile shrimps was the highest around the leaves that were decomposed for 40 days and colonized with high counts of nitrogen-fixing azotobacters. Finally, a vegetation trap technique using mangrove leaves has been proposed for efficient schooling of juvenile shrimps. There was about 3-fold increase in number of shrimp juveniles around the trap than at control waters.  相似文献
