Soil contamination with radiocaesium is a significant problem at any countries when a nuclear accident occurred. Recently, phytoextraction technique is developed to remediate the contaminated environment. However, the application is limited by the availability of the contaminant for root uptake. Therefore, a green house trial experiment of soil amendment with ethylene diamine tetraacetic acid (EDTA) has been conducted to examine 134Cs availability for root uptake. Two groups of Indian mustard (Brassica juncea) were cultivated in 134Cs contaminated soil. The soil in the first group was treated with EDTA amendment, while the other was not. Plant growth was observed gravimetrically and the 134Cs concentration in soil as well as plants were determined using gamma spectrometry. The plant uptake capacity was determined as transfer factor (Fv), and the Fv values of 0.22 ± 0.0786 and 0.12 ± 0.039 were obtained for the soil treated with and without EDTA amendment, respectively. The phytoextraction efficiency of the plant cultivated in 134Cs contaminated soil both with and without EDTA amendment was low. The EDTA amendment to the soil seems to enhance the 134Cs availability for root uptake of Indian mustard and can still be considered to assist the field phytoremediation of contaminated soil.  相似文献

Spring wheat (Triticum aestivum L. cv. Tonic) was grown for 16 days in a sandy loam soil which was contaminated with 137Cs. The soil was fertilised with K at three rates (0,1 and 2 mmol K per 950 g dry soil) and with NO3 --N at two rates (0 and 2 mmol per 950 g dry soil) in a factorial design. The 137Cs Activity Concentration (AC) in the shoot tissue significantly reduced 8.2-fold (nil N treatment, p<0.001) and 9.3-fold (highest N dose, p<0.001) with increasing K supply. In contrast, the K application increased the 137Cs AC in soil solution 1.7 fold (nil N treatment) or had no significant effect (highest N dose). At similar K application, the application of N increased the 137Cs AC in the shoot compared to the control. This effect is most probably due to the increased NH4 + concentration in soil solution which increased the 137Cs AC in soil solution. The soil solution composition (137Cs and K concentration) in the rhizosphere was estimated from the average soil solution composition at day 16 and solute transport calculations. The 137Cs AC in the shoot tissue was predicted from the estimated soil solution composition in the rhizosphere and the relationship between K concentration and 137Cs uptake derived from a nutrient solution experiment. The predictions of 137Cs AC's in the shoot are qualitatively correct for the fertiliser effects but underestimate the observations between 1.4 and 9.9 fold.  相似文献

The uptake of radionuclides 60Co, 32P and 134Cs by some aquatic plants (Lemna gibba, Ceratophyllum demersum and Potamogeton crispus) present in the Ismailia Canal at different pH values and for different contamination periods was studied. The experimental results showed that the uptake generally decreases as the water pH increased. The tested macrophytes proved to be reliable biological indicators.  相似文献

Field-grown maize hybrids were assessed for variability in 137Cs accumulation in vegetative parts of young and mature maize shoots and grains during 2 years with contrasting climatic conditions. Trials were carried out at different sites in the Tula region of Russia, which is characterized by a highly homogenous soil classified as Luvic Chernozem according to FAO/UNESCO, and average contamination levels of about 509–564 Bq 137Cs kg−1 soil. In the first year, 19 hybrids were tested. The two hybrids with the highest and the two with the lowest 137Cs concentration ratios (C r) were also tested in the second year, together with another 11 hybrids. All samples were additionally assessed for their potassium content. In both investigation periods 137Cs accumulation in vegetative shoots and grains was found to vary up to more than twofold between hybrids. However, C r values of those hybrids that showed a relatively low 137Cs accumulation in the first year were not necessarily low in the second year, and the ratio between the 137Cs C r of low- and high-accumulating hybrids was much smaller than in the year before. In both vegetative shoots and grains the variance caused by the different years was larger than the genotypic variance, thus indicating the limits of genotype selection for this trait. Significant correlations were determined between the 40K and 137Cs C r values in the same tissue, but for one hybrid indications for uncoupling of the two traits were found. Average Cs/K ratios in young shoots, mature shoots and grains were 0.06, 0.05 and 0.02, respectively, indicating tissue- and stage-specific regulation of accumulation within each plant. The influence of Bifezh (a hexacyanoferrate compound as veterinary feed additive) on the transfer of 137Cs to meat was investigated in broiler chickens after application of an artificially contaminated feed mixture. The reduction factors reached 2.23 and 8.46 for single and multiple 137Cs administration, respectively. The elimination rate of 137Cs in the decontamination period was only slightly increased by Bifezh. The reduction effect of Bifezh was considerably and positively limited by various amounts of the additive during both radiocaesium accumulation and decontamination. No significant differences in the reduction effect between breast and thigh meat were observed. In conclusion, the hexacyanoferrate-cellulose binder Bifezh can be recommended as an alternative chemical measure for reducing 137Cs contamination of broiler chicken meat, although administration rates may need to be enhanced over those currently recommended.  相似文献

D1- and D2-types of dopamine receptors are located separately in direct and indirect pathway striatal projection neurons (dSPNs and iSPNs). In comparison, adenosine A1-type receptors are located in both neuron classes, and adenosine A2A-type receptors show a preferential expression in iSPNs. Due to their importance for neuronal excitability, Ca2+-currents have been used as final effectors to see the function of signaling cascades associated with different G protein-coupled receptors. For example, among many other actions, D1-type receptors increase, while D2-type receptors decrease neuronal excitability by either enhancing or reducing, respectively, CaV1 Ca2+-currents. These actions occur separately in dSPNs and iSPNs. In the case of purinergic signaling, the actions of A1- and A2A-receptors have not been compared observing their actions on Ca2+-channels of SPNs as final effectors. Our hypotheses are that modulation of Ca2+-currents by A1-receptors occurs in both dSPNs and iSPNs. In contrast, iSPNs would exhibit modulation by both A1- and A2A-receptors. We demonstrate that A1-type receptors reduced Ca2+-currents in all SPNs tested. However, A2A-type receptors enhanced Ca2+-currents only in half tested neurons. Intriguingly, to observe the actions of A2A-type receptors, occupation of A1-type receptors had to occur first. However, A1-receptors decreased CaV2 Ca2+-currents, while A2A-type receptors enhanced current through CaV1 channels. Because these channels have opposing actions on cell discharge, these differences explain in part why iSPNs may be more excitable than dSPNs. It is demonstrated that intrinsic voltage-gated currents expressed in SPNs are effectors of purinergic signaling that therefore play a role in excitability.  相似文献

Spring wheat plants were grown in a 137Cs labelled nutrient solution, either in the presence or absence of NH4 as a secondary N source. Between 11 and 64 days after sowing (DAS), plants were harvested on nine occasions. The plants supplied with NH4 and NO3 had lower root 137Cs Activity Concentrations (AC) than those supplied with NO3 only. Shoot AC were equal in both nutrition treatments. Shoot and root 137Cs AC (dry weight basis) showed the same trends with plant age in both nutrition treatments. Shoot AC almost doubled between 11 and 28 DAS after which they gradually decreased concomitant with a similar decrease in K concentrations. Root AC were always higher than shoot AC and increased to a maximum at 35 DAS after which they fluctuated. Expressed on a tissue water basis, the 137Cs AC varied less during plant age than did dry weight based AC. Furthermore, root and shoot AC expressed on a tissue water basis were almost equal. It is shown that the initial increase in 137Cs AC in both root and shoot can largely be explained by the initial dilution of absorbed 137Cs in the unlabelled seedling tissues. No correlation was found between K and 137Cs distribution among ears, leaves, stems and roots in 64 old wheat plants. Mycotoxins are secondary metabolites which can form on various foodstuffs through the growth of certain fungi. Ochratoxin A (OTA) and the aflatoxins B1 B2, G1 and G2 have been detected in low concentrations in cocoa and cocoa products. As regards the question of in what stages of the cocoa production process a contamination with the mycotoxin-producing moulds and the formation of mycotoxins takes place, it is assumed that in the case of cocoa the contamination is not concerning the individual beans but the fermentation units. A model test was carried out to provide information on the process by which a possible carryover of the above-mentioned mycotoxins to cocoa beans occurs during the fermentation process. For this purpose fresh cocoa beans were left to soak in an artificial mycotoxin-containing fermentation solution. The mycotoxin levels in the cocoa beans were regularly determined over a period of 12 days. New findings were made as regards the migration of mycotoxins during the fermentation process. We interpret the divergent uptake behaviour of the mycotoxins to indicate that the transport of OTA and that of aflatoxins does not take place in the same manner. The comet assay is one of the most versatile and popular tools for evaluating DNA damage. Its sensitivity to low dose radiation has been tested in vitro, but there are limited data showing its application and sensitivity in chronic exposure situations. The influence of the internal contamination caused by the Chernobyl accident on the level of DNA damage was evaluated by the comet assay on lymphocytes of 56 Ukrainian children. The study was performed during 2003 on children with demonstrable 137Cs internal contamination caused by food consumption. The children were selected for the study immediately after a 137Cs whole body counter measurement of internal contamination. The minimal detectable amount of 137Cs was 75 Bq. The control group included 29 children without detectable internal contamination, while in the exposed group 27 children with measured activity between 80 and 4037 Bq and committed effective dose between 54 and 3155 μSv were included. Blood samples were taken by a finger prick. The alkaline version of the comet assay was used, in combination with silver stained comets and arbitrary units (AU), for comet measurement. Factors such as disease, medical treatment, surface contamination of children's living location, etc., were considered in the study. Non-significant differences (p > 0.05) in DNA damage in control (9.0 ± 5.7 AU) versus exposed (8.5 ± 4.8 AU) groups were found. These results suggest that low doses of 137Cs internal contamination are not able to produce detectable DNA damage under the conditions used for the comet assay in this study. Further studies considering effects of high exposure should be performed on chronically exposed people using this assay.  相似文献

Four forms of bovine adrenodoxin with modified amino-termini obtained by direct expression of cDNAs in Escherichia coli are Ad(Met1), Ad(Met−1), Ad(Met−12), and Ad(Met6). The shoulder numbers represent this site of translation initiator Met at the amino-termini. The adrenodoxins, except for Ad(Met−1), were purified from the cell lysate and the ratios of A414-to-A276 of the purified proteins were over 0.92. NADPH-cytochrome c reductase activities of the three forms of adrenodoxin in the presence of adrenodoxin reductase were the same as that of purified bovine adrenocortical adrenodoxin. However, as cytochrome P-450SCC reduction catalyzed by Ad(Met0) was about 60% or that by Ad(Met1), the contribution of the amino-terminal region for the electron transfer or binding to cytochrome P-450SCC would need to be considered.  相似文献

In many Swedish lakes, the fallout of 137Cs from the Chernobyl accident in April 1986 has largely accumulated in the sediments. The availability and transfer of deposited 137Cs to biota is influenced by factors such as resuspension. The frequency of resuspension and the 137Cs-content of different fish species was studied in three shallow lakes in Uppland, central Sweden, and in one deeper lake in northern Sweden. Resuspension was measured by the use of sediment traps. Sedimentation rates measured from the traps in the shallower lakes were 5–10 times higher than normal for this type of lake, indicating that resuspension was an important factor. The decrease of the 137Cs-content in muscle tissue of pike, perch and roach was slow in each of the shallow lakes. 137Cs decreased by about 30% over a period of 2 years in the shallowest lake (maximum depth 4 m), whereas 137Cs decreased by 50% in the deeper lakes (maximum depth 10 m). The slower rates of decline of 137Cs in biota from the shallow lakes, are probably a function of sediment dynamics (mainly influenced by lake morphometry, wind direction and strength). They may be influenced, also, by bioavailability of resuspended sediment material. In the deepest northern lake, much of the 137Cs-containing material collected in the sediment traps originated from the catchment area. Resuspension was minimal, and the high activity of 137Cs in the sediment had no effect on content or decline of 137Cs in lake fish.  相似文献

Extracellular adenosine activates P1 receptors (A1, A2A, A2B, A3) on cellular membranes. Here, we investigated the involvement of P1 receptor-mediated signaling in differentiation to regulatory T cells (Treg). Treg were induced in vitro by incubating isolated CD4+CD62L+ naïve murine T cells under Treg-skewing conditions. Antagonists of A1 and A2B receptors suppressed the expression of Foxp3, a specific marker of Treg, and the production of IL-10, suggesting the involvement of A1 and A2B receptors in differentiation to Treg. We also investigated the effect of these antagonists on T cell activation, which is essential for differentiation to Treg, and found that A1 antagonist, but not A2B antagonist, suppressed T cell activation. We conclude that A1 and A2B receptors are both involved in differentiation to Treg, but through different mechanisms. Since A2B antagonist blocked differentiation to Treg without suppressing T cell activation, it is possible that blockade of A2B receptor would facilitate tumor immunity.  相似文献

European beavers (Castor fiber) from two regions were examined to identify exposure to persistent environmental contaminants. A reference group was comprised of six animals from the Forest Division of Srokowo, and an exposed group was comprised of five animals from the vicinity of a former military airport operated in 1918–1986—both from Warmia land in Poland. 137Cs in beavers' muscles was considered negligible for the overall condition and reproduction of the animals. Elevated levels of Cd and Pb occurred in the kidneys and liver of beavers from a former military airport area. dl-PCBs were found in the liver of all specimens, and pCB no. 118 was a dominant compound. Beavers as animals living both in water and land habitats can be good bioindicators of heavy metal environmental pollution caused by human activity. Despite the significant contamination of former military sites in northeastern Poland, the beaver population shows an upward trend, reflecting the excellent adaptation of this species to a changing environment.  相似文献

The synthesis, crystal structure and magnetic properties of the charge transfer salt [Fe(Cp)2][Ni(dsit)2], dsit = (2-thioxo-1,3-dithiole-4,5-diselenolato) are described. The crystal structure consists of layers composed by parallel chains, where side-by-side pairs of cations, alternate with face-to-face pairs of anions, ?D+D+AAD+D+AA?. The acceptors in the AA pairs are strongly dimerized through a Ni-Se bond. Short interdimer contacts (Se-Se and S-S) give rise to a porous 3D network, and the [Fe(Cp)2]+pairs are located in the cavities of the acceptors network. The [Fe(Cp)2]+ donors are relatively isolated leading to weak DA and DD interactions and the magnetic behavior of this compound can be basically described as the sum of two contributions: one from nearly independent donors, and the second from the acceptors network, with strong AA AFM coupling, both intradimer and between dimers in adjacent chains.  相似文献

Anatomical evidence indicates that γ-aminobutyric acid (GABA)-ergic and opioidergic systems are closely linked and act on the same neurons. However, the regulatory mechanisms between GABAergic and opioidergic system have not been well characterized. In the present study, we investigated whether there are changes in GABAA receptors in mice lacking μ-opioid receptor gene. The GABAA receptor binding was carried out by autoradiography using [3H]-muscimol (GABAA), [3H]-flunitrazepam (FNZ, native type 1 benzodiazepine) and [35S]-t-butylbicyclophosphorothionate (TBPS, binding to GABAA-gated chloride channels) in brain slices of wild type and μ-opioid receptor knockout mice. The binding of [3H]-FNZ in μ-opioid receptor knockout mice was significantly higher than that of the wild type controls in most of the cortex and hippocampal CA1 and CA2 formations. μ-Opioid receptor knockout mice show significantly lower binding of [35S]-TBPS than that of the wild type mice in few of the cortical areas including ectorhinal cortex layers I, III, and V, but not in the hippocampus. There was no significant difference in binding of [3H]-muscimol between μ-opioid receptor knockout and wild type mice in the cortex and hippocampus. These data indicate that there are specific regional changes in GABAA receptor binding sites in μ-opioid receptor knockout mice. These data also suggest that there are compensatory up-regulation of benzodiazepine binding site of GABAA receptors in the cortex and hippocampus and down-regulation of GABA-gated chloride channel binding site of GABAA receptors in the cortex of the μ-opioid receptor knockout mice.  相似文献

A recently identified novel Gαolf variant, XLGαolf, is shown to functionally couple to the human adenosine A2A receptor (A2AR). In Sf9 cells expressing A2AR, β1, and γ2, co-expression of XLGαolf increased NECA-induced [35S]GTPγS binding from approximately 130% to 300% of basal levels. Pharmacological characteristics of A2AR ligands on these cells were evaluated by using [3H]ZM241385- and [35S]GTPγS- binding assays. The rank order of the equilibrium binding constants (Kd or Ki) of adenosine receptor ligands were [3H]ZM241385 ≈ CGS15943 < MRS1220 < < CV1808 ≈ NECA < CGS21680 ≈ adenosine < IBMECA < HEMADO ≈ CPA ≈ CCPA. The rank order of EC50 values for agonists were CV1808 ≈ NECA < adenosine ≈ CGS26180 < IBMECA < HEMADO ≈ CPA ≈ CCPA. This pharmacology is consistent with the literature for A2AR and suggests that Sf9 cells co-expressing A2AR, β1, γ2, and XLGαolf could serve as a heterologous expression system for A2AR drug screening.  相似文献

The pH dependence of emission peak temperature and decay time of thermoluminescence arising from S2QB and S2QA recombinations demonstrates that a stabilization of S2QB occurs at low pH whereas stabilization of S2QA occurs at high pH. Based on comparative analysis of thermoluminescence parameters of the two types of recombination, we suggest that in the pH range between 5.3 and 7.5, Em(S2/S1) and Em(QA/QA ) are constant, but Em(QB/QB ) gradually increases with decreasing pH, while in the pH range between 7.5 and 8.5, an unusual change occurs on S2QA charge pair, which is interpreted as either a decrease in Em(S2/S1) or an increase in Em(QA/QA ).  相似文献

The ultrafast (< 100 fs) conversion of delocalized exciton into charge-separated state between the primary donor P700 (bleaching at 705 nm) and the primary acceptor A0 (bleaching at 690 nm) in photosystem I (PS I) complexes from Synechocystis sp. PCC 6803 was observed. The data were obtained by application of pump-probe technique with 20-fs low-energy pump pulses centered at 720 nm. The earliest absorbance changes (close to zero delay) with a bleaching at 690 nm are similar to the product of the absorption spectrum of PS I complex and the laser pulse spectrum, which represents the efficiency spectrum of the light absorption by PS I upon femtosecond excitation centered at 720 nm. During the first ∼ 60 fs the energy transfer from the chlorophyll (Chl) species bleaching at 690 nm to the Chl bleaching at 705 nm occurs, resulting in almost equal bleaching of the two forms with the formation of delocalized exciton between 690-nm and 705-nm Chls. Within the next ∼ 40 fs the formation of a new broad band centered at ∼ 660 nm (attributed to the appearance of Chl anion radical) is observed. This band decays with time constant simultaneously with an electron transfer to A1 (phylloquinone). The subtraction of kinetic difference absorption spectra of the closed (state P700+A0A1) PS I reaction center (RC) from that of the open (state P700A0A1) RC reveals the pure spectrum of the P700+A0 ion-radical pair. The experimental data were analyzed using a simple kinetic scheme: An* [(PA0)*A1 P+A0A1] P+A0A1, and a global fitting procedure based on the singular value decomposition analysis. The calculated kinetics of transitions between intermediate states and their spectra were similar to the kinetics recorded at 694 and 705 nm and the experimental spectra obtained by subtraction of the spectra of closed RCs from the spectra of open RCs. As a result, we found that the main events in RCs of PS I under our experimental conditions include very fast (< 100 fs) charge separation with the formation of the P700+A0A1 state in approximately one half of the RCs, the ∼ 5-ps energy transfer from antenna Chl* to P700A0A1 in the remaining RCs, and ∼ 25-ps formation of the secondary radical pair P700+A0A1.  相似文献

从陕北地区石油污染土壤中分离鉴定得到两株不动杆菌属(Acinetobacter sp.)的高效石油降解菌A.sp 1和A.sp 2,分别从盐浓度、pH值、氮源、磷源和接种量等因素进行研究以确定其最佳石油降解条件,并进一步通过GC-MS(Gas ChromatographyMass Spectrometer)方法分析其在最佳条件下对原油组分的不同降解性能。结果显示:A.sp 1在盐浓度为1%、pH值为6—7、磷源为KH2PO4和K2HPO4、氮源为尿素和接种量为4%的条件下,最高降解率可达到60%。A.sp 2在盐浓度为1%、pH值为7—9、磷源为KH2PO4和K2HPO4、氮源为硝酸铵和接种量为8%的条件下,最高降解率可达到67%。GC-MS分析结果表明,菌株A.sp 1对石油烃类C21—C25有明显的降解效果,菌株A.sp 2对石油烃类C20—C30的降解效果较好。  相似文献

A series of Ca6AlP5O20 doped with rare earths (Eu and Ce) and co‐doped (Eu, Ce and Eu,Mn) were prepared by combustion synthesis. Under Hg‐free excitation, Ca6AlP5O20:Eu exhibited Eu2+ (486 nm) emission in the blue region of the spectrum and under near Hg excitation (245 nm), Ca6AlP5O20:Ce phosphor exhibited Ce3+ emission (357 nm) in the UV range. Photoluminescence (PL) peak intensity increased in Ca6AlP5O20:Eu,Ce and Ca6AlP5O20:Eu, Mn phosphors due to co‐activators of Ce3+ and Mn2+ ions. As a result, these ions played an important role in PL emission in the present matrix. Ca6AlP5O20:Eu, Ce and Ca6AlP5O20:Eu, Mn phosphors provided energy transfer mechanisms via Ce3+ → Eu2+ and Eu2+ → Mn2+, respectively. Eu ions acted as activators and Ce ions acted as sensitizers. Ce emission energy was well matched with Eu excitation energy in the case of Ca6AlP5O20:Eu, Ce and Eu ions acted as activators and Mn ions acted as sensitizers in Ca6AlP5O20:Eu, Mn. This study included synthesis of new and efficient phosphate phosphors. The impact of doping and co‐doping on photoluminescence properties and energy transfer mechanisms were investigated and we propose a feasible interpretation.
