Summary The orchid Prasophyllum fimbria is pollinated by nectar-feeding native bees and wasps. The pollinia are patially separated from the viscidium by a stipe so that pollinia can be labelled with coloured histochemical stains without interfering with pollinarium removal. Pollen flow was monitored by following the movement of the coloured pollen in several populations of P. fimbria in Western Australia. Statistical analysis confirmed that pollen labelling did not interfere with pollinarium removal or subsequent pollination of the labelled flower. Fifty eight labelled pollinaria were removed by vectors from 16 test spikes, with a total of 125 flowers on 47 spikes receiving labelled pollen. An average of 2 flowers received pollen for every pollinium removed but up to 6 flowers received pollen from a single collinium. No significant differences between mean vector flights and pollen flow distances were detected. On average, geitonogamous transfers only accounted for 22% of all pollinations. This is a simple and inexpensive technique for the direct labelling of pollen with minimal disruption to the pollination system and may have applications in other plant families.  相似文献

A simple technique for the isolation of deletion mutants of phage lambda.   总被引:5,自引:0,他引:5  
We describe a simple technique for isolating deletion mutants of phage lambda and use it to dissect a cloned fragment of foreign DNA. The technique is based on our previous finding that the normally essential product of lambda head gene D is dispensible for phage growth if the DNA content of the phage is less than 82% that of lambda wild-type (Sternberg and Weisberg, 1977). A significant fraction of the few phage that form plaques when a D amber mutant is plated on a nonsuppressing host contains deletions that reduce the phage chromosome size to less than 82% that of wild-type. It is possible to isolate deletions ranging in size from less than 1.5 kb to 14 kb (3 to 27% of wild-type lambda), and the size range can be restricted by an appropriate choice of the DNA content of the starting phage. This method, unlike the older EDTA or heat resistance methods, permits the scoring of deletions because of the absence of phenotypic variants. We investigated the effect of several host and phage mutations on deletion frequency and type and have determined that a host polA mutation increases the frequency of deletions about 30-50-fold without changing the type of deletions. A host mutD mutation or thymine deprivation increases deletion frequency about 10-fold. In contrast, a host ligts mutation has no effect on the frequency of deletions. We have also determined that the size of the smallest lambda chromosome packageable in a plaque-forming phage particle is 72-73% that of lambda wild-type.  相似文献

Patterns of reproductive isolation in Mediterranean deceptive orchids   总被引:2,自引:0,他引:2  
The evolution of reproductive isolation is of central interest in evolutionary biology. In plants, this is typically achieved by a combination of pre- and postpollination mechanisms that prevent, or limit, the amount of interspecific gene flow. Here, we investigated and compared two ecologically defined groups of Mediterranean orchids that differ in pollination biology and pollinator specificity: sexually deceptive orchids versus food-deceptive orchids. We used experimental crosses to assess the strength of postmating prezygotic, and postzygotic reproductive isolation, and a phylogenetic framework to determine their relative rates of evolution. We found quantitative and qualitative differences between the two groups. Food-deceptive orchids have weak premating isolation but strong postmating isolation, whereas the converse situation characterizes sexually deceptive orchids. Only postzygotic reproductive isolation among food-deceptive orchids was found to evolve in a clock-like manner. Comparison of evolutionary rates, within a common interval of genetic distance, showed that the contribution of postmating barriers was more relevant in the food-deceptive species than in the sexually deceptive species. Asymmetry in prezygotic isolation was found among food-deceptive species. Our results indicate that postmating barriers are most important for reproductive isolation in food-deceptive orchids, whereas premating barriers are most important in sexually deceptive orchids. The different rate of evolution of reproductive isolation and the relative strength of pre- and postmating barriers may have implication for speciation processes in the two orchid groups.  相似文献

Genomic DNA was isolated from as little as 2 mg dry biomass of Magnaporthe grisea by microwave treatment within 30 s. The quantity of DNA was good enough for PCR analysis and Dot blot hybridization. This technique can be used for various studies, such as DNA fingerprinting to study the population structure of the phytopathogen in different regions, and for a quick screening of M. grisea transformants.  相似文献

Abstract Light and fluorescence microscopy studies indicated that chlorophyll was absent from the guard cells of the lady slipper orchids, Paphiopedilum insigne (Wall.) Pfitz, P. insigne (hybrid), P. venustum (Wall.) Pfitz and P. harrisseanum Hort. In the guard cells of P. aureum hyeanum Hort., however, very slight red fluorescence suggested that chlorophyll and hence chloroplasts were present. Ultrastructural studies of the lower epidermis of P. insigne (hybrid) confirmed the absence of chloroplasts in guard and epidermal cells although plastids of an unusual structure were found in these cells. In fully developed epidermal cells the plastids contained large amounts of a fibrous, possibly proteinaceous substance, spherical, lightly staining vesicles and an electron-dense material located in reticulate and non-reticulate regions. Additionally, latticed crystalline inclusions and plasto-globuli were occasionally observed in the epidermal cell plastids. In plastids of fully developed guard cells the fibrous material, starch and plastoglobuli were present. From the earliest stages of development of the epidermal tissue starch was present in both epidermal cell and guard cell plastids. At maturity, however, starch had accumulated to greater levels in the guard cell plastids and had entirely disappeared in the epidermal cell plastids. In differentiating epidermal tissue, plasmodesmata were found between neighbouring epidermal cells and between guard and epidermal cells. At maturity, plasmodesmata between guard and epidermal cells were not observed. Mitochondria were particularly abundant in guard cells. Large oil drops developed in guard and epidermal cells, being especially abundant in the former at maturity. Our results confirm the observations of Nelson & Mayo (1975) that certain lady slipper orchids possess functional stomata the guard cells of which do not contain chloroplasts.  相似文献

A simple method for the isolation of plant protoplasts   总被引:1,自引:0,他引:1  
A simple protoplast isolation protocol that was designed to recover totipotent plant protoplasts with relative ease has been described. The key elements of the protocol are, tissue digestion at slightly elevated temperatures and use of protoplast-releasing enzymes that are stable and efficient at higher temperatures. Besides enzymes, the protoplast isolation cocktail consisted of an osmoticum (mannitol or MgSO4), and a protectant (CaCl2 2H2O), all dissolved in distilled water. The protocol has ensured reproducibility, higher yields and is gentle on protoplasts as the protoplasts obtained were amenable to cell wall regeneration and cell division. Plant regeneration was demonstrated forNicotiana tabacum cv. Thompson from protoplasts isolated by this method. Wall regeneration and cell division were obtained in other species. The merits of the protocol are, simple and easy-to-handle procedure, non-requirement of preconditioning of donor plant and explants, incubation without agitation, satisfactory yields, culturability of the protoplasts isolated and applicability of the protocol to a large number of species including mucilage-containing plants.  相似文献

Summary Activated phytochrome lowers the free amino acid pools of all plants investigated by about 15%. The action of red light is preferentially directed to Asp, Glu and Phe. Exogenously supplied Leu-U-14C is incorporated more quickly into protein of red-light-treated samples compared with dark controls. In contrast, red light decreases the amount of Asp-U-14C incorporated into protein, but increases the amount of 14CO2 respired after feeding with Asp-U-14C. Red light has no effect on the amount of 14CO2 respired after feeding with Leu-U-14C. A technique for endotracheal intubation in rabbits, which eliminates the need for tracheostomies, special devices, laryngoscopes and guide wires, is described. Arabidopsis thaliana has been widely used as a model plant in gene function analysis. However, its tiny flower and curved embryo sac make it difficult to study gene expression during megagametogenesis, fertilization, and early embryogenesis, especially in the screening of mutants from those developmental processes. The techniques currently available are sectioning and whole-mount clearing of ovules; however, sectioning is time consuming and laborious for quantitative analysis, and whole-mount clearing, makes clear cytological observation impossible. Reported here is a simple and efficient method based on enzymatic isolation of embryo sacs that enables both quantitative analysis and elaborate cytological observation for gene expression investigation and mutant screening.  相似文献

Summary It was shown that Chaetomium was isolated from four types of soil; Cinebar, Ephrata, Milville, and quartz sand, more frequently when using alcohol agar as the growth medium than it (Chaetomium) was isolated from these soils when using either Czepak's-Dox agar or peptone-dextrose agar plus rose bengal and streptomycin.
