The association pattern of marked tetrasomes of Secale chromosome 1R at meiotic first metaphase was analyzed. Two of the four chromosomes were identical with terminal C-bands at both arms; the other two were also identical but lacked C-bands and were homologous or homeologous with the first two. Four different types of heterozygotes for 1R were studied: (i). autotetraploid hybrids between genetic variants within Secale cereale subsp. cereale, (ii). tetraploid hybrids between subspecies of Secale cereale, (iii). tetraploid hybrids between species of Secale, and (iv). autotetrasomes of S. cereale in a wheat background. Earlier observations that heterozygous associations (banded with unbanded) had consistently higher chiasma frequencies than homozygous associations were extended and confirmed. To analyze this phenomenon more closely, the possible relations between this correlation and several other meiotic phenomena were studied. For this analysis, three genetically different autotetraploid hybrids within S. cereale were selected that differed with respect to the relation between pairing type and chiasma frequency. Special attention was given to different patterns of interference and other meiotic phenomena in the two chromosome arms of chromosome 1R. No relations between such phenomena and the relation between pairing type and chiasma frequency could be established. A hypothesis is formulated assuming that long-distance homologue attraction is concentrated in a limited number of sites and that in different genotypes, different patterns of active sites are present. Moderately weak attraction sites can pair with strong homologous sites under favorable genetic conditions, but two weak sites cannot. Then, heterozygotes have more effective pairing initiation and consequently chiasma formation than homozygotes. Under less favorable conditions, only strong sites are effective, and then, homozygotes pair better, but the chiasma frequency is lower. A model of the forces involved in homologue attraction is presented.  相似文献

研究甘蔗属与滇蔗茅属间远缘杂交F1染色体遗传行为具有重要科学意义,可为发掘利用滇蔗茅野生优异基因资源提供细胞学依据。本研究采用常规压片技术对可育父本及不育杂交F1花粉母细胞减数分裂过程进行比较观察,结果显示可育父本云南95-19减数分裂正常,而不育杂交F1分裂异常;进一步对F1花粉母细胞减数分裂过程进行基因组荧光原位杂交(GISH,genome in situ hybridization)分析,结果表明:滇蔗茅与甘蔗属热带种的亲缘关系较远,双亲染色体在F1细胞中不能进行同源配对,终变期,15条滇蔗茅染色体以单价体形式存在,花粉母细胞减数分裂细胞中存在滞后染色体、染色体丢失和不均衡分离现象。甘蔗-滇蔗茅属间远缘杂交F1花粉母细胞减数分裂异常因而杂交F1花粉完全败育。  相似文献

The meiotic pairing behaviour of four B isochromosomes of Crepis capillaris was studied by synaptonemal complex (SC) surface spreading of pollen mother cells. The four B chromosomes form a tightly associated group, separate from the standard chromosomes, throughout zygotene and pachytene. All four B chromosomes are also folded around their axis of symmetry, the centromere, and the eight homologous arms are closely aligned from the earliest prophase I stages. A high frequency of multivalent pairing of the four B chromosomes is observed at pachytene, in excess of 90%, mirroring the situation observed at metaphase I but exceeding the frequency expected (76.2%) on the assumption of random pairing among the eight B isochromosome arms with a single distal pairing initiation site per arm. The higher than expected frequency of multivalents is due to the occurrence of multiple pairing initiations along the B isochromosome arms, resulting in high frequencies of pairing partner switches. Pairing of the standard chromosome set is frequently incomplete in the presence of four B chromosomes, and abnormalities of SC structure such as thickening and splitting of axes and lateral elements are also frequently seen. Similarly, B chromosomes show partial pairing failure, the extent of which is correlated with pairing failure in the standard chromosome set. The B chromosomes themselves also show abnormalities of SC structure. Both standard and B chromosomes show non-homologous foldback pairing of regions that have failed to pair homologously.by D. Mechanisms of two F1 hybrids (D46A × DTP-4 and D46A × Dminghui63) of autotetraploid rice (2n = 4x = 48) showing extremely high pollen fertility 87.40% and 85.97%, respectively, seed set 82.00% and 79.00%, respectively and extremely high level of heterosis were analyzed cytologically. The chromosome pairing of D46A × DTP-4 and D46A × Dminghui63 was normal at metaphase I(MI), and had almost no I or III, with an average of 0.020I +14.36 II 6.44rod+7.91ring) +0.01III + 4.80 IV + 0.01VIII and 0.06 I + 17.67 II (11.01rod + 6.67ring)] + 0.06 III +3.10IV+0.01VI, respectively. The most frequent chromosome configurations were 10II+7IV and 12II+bIV. The bivalent frequency was less frequent in hybrids than that in restoring parents, and the same results were gained from univalents, trivalent and multivalents. However, the quadrivalent frequency was significantly higher in hybrids than that in restoring parents at MI. The other meiotic phases progressed normally, except for low percentages of PMCs with lagging chromosomes at AI and low percentages of PMCs with micronuclei at telophaseI (TI) and telophaseII (TII). PMCs with lagging chromosomes at AI and PMCs with micronuclei at TI and TII showed negative correlation between pollen fertility and seed set. Above 90% of the PMCs could form normal microspores, which resulted in the production of viable pollen grains, abnormal microspores were observed including penta-fission and hexa-fission. Based on these results we suggest that the two F1 hybrids had better behaviors of chromosome pairing and genetic stability than autotetraploid rice and other autotetraploid plants ever studied.  相似文献

Amphiploids (2n = 6x = 42) of Ae. kotschyi and Ae. biuncialis with self-compatible S. cereale were produced from F1 sterile hybrids (2n = 3x = 21) through colchicine treatment and callus tissue regeneration. The amphiploids resembled the F1 plants in overall morphology, but were larger in all respects and self-fertile. The spikelets consisted mostly of 3 well-developed florets. Selfed seeds were obtained from some colchicine-doubled sectors and callus regenerates. Most of the produced seeds were well developed. Backcrosses between amphiploids and rye (2x and 4x) resulted in obtaining (Ae. biuncialis x S. cereale amphiploid) x S. cereale hybrids via embryo culture. The BC1 plants (2n = 4x = 28 and 2n = 5x = 35, respectively) were phenotypically intermediate between the parents and vigorous in vegetative growth. Some seeds were obtained only from the 35-chromosome BC1 hybrids.  相似文献

An attempt to transfer genes from droughttolerant Diplotaxis harra, a wild relative of Brassica species, to an elite oil-yielding cultivar, B-85, of mustard (Brassica juncea) was made through protoplast fusion, as the two plant systems are sexually incompatible. By following the standard protocol for PEG-mediated protoplast fusion followed by high pH, high Ca++, DMSO treatment and appropriate cell-culture technique, 16 presumptive somatic hybrid plants could be regenerated. Chromosomal analysis of four such somatic hybrids revealed that three of them were asymmetric. Analysis of morphological characters, meiotic chromosomes, and esterase isoenzyme pattern revealed that all the somatic hybrids were different from each other. Furthermore four chromosomes of each genome could undergo homoeologous pairing at meiosis indicating the possibilities for genetic recombination and chromosomal rearrangements. Irregular distribution of chromosomes at anaphase-II at meiosis has been a consistent feature of these plants. Eventually, pollen of all the somatic hybrids showed complete infertility preventing the recovery of any selfed seed. Nevertheless, ovule fertility of one somatic hybrid was not totally impaired as it had set some seeds upon backcrossing with the B. juncea parent. The esterase isoenzyme banding pattern of 24 individual progeny plants of this backcross provided evidence for their recombinant nature. It was thus confirmed that a transfer of genetic traits from Diplotaxis harra to B. juncea had indeed taken place. Furthermore, it was conceptualised that a transfer of alien genes through the protoplast-fusion technique is primarily possible in situations where meiotic pairing of the chromosomes of the two participating genomes generates recombinant gametocytes which can pass through subsequent filial generations.  相似文献

Diploid populations of Aegilops mutica and Aegilops speltoides containing B chromosomes have been used as male parents in crosses with aneuploid genotypes of Triticum aestivum to investigate the effect of B chromosomes on meiotic homologous and homoeologous chromosome pairing. F1 hybrids of T. aestivum/Ae. mutica and T. aestivum/Ae. speltoides segregated into four classes with regard to the degree of meiotic chromosome pairing, irrespective of the presence of B chromosomes. The B chromosomes do not introduce factors altering the level of pairing other than that due to the natural allelic and gene variation occurring in the diploids. Similarly no reduction in pairing of homologous chromosomes was observed in genotypes in which pairs of homologues co-existed with B chromosomes. However, a significant drop in chiasma frequency was observed in F1 hybrids of T. aestivum × Ae. mutica with B chromosomes and T. aestivum × Ae. mutica nullisomic for wheat chromosome 5D with B chromosomes, in temperature regimes of 12° C. No asynapsis occurred in similar hybrids in the absence of Mutica B chromosomes at low temperatures. The low-temperature sensitive phase lies early in the pre-meiotic interphase. In this instance the Mutica B chromosomes are interacting with specific gene loci of the A chromosomes. Synaptic pairing has been observed between A and B chromosomes in Ae. mutica. A high frequency of pollen mother cells with twice the number of chromosomes was observed in hybrids in the presence of Mutica B chromosomes due to failure of spindle formation at the last pre-meiotic mitosis. Meiotic spindle irregularities occurred in hybrids containing Speltoides B chromosomes. Hybrids of Ae. speltoides + B's X Ae. mutica + B's displayed the mitotic and meiotic spindle abnormalities introduced by the presence of the B chromosomes of each parent.  相似文献

Electrically induced protoplast fusion was used to produce somatic hybrids between Brassica napus L. and Sinapis alba L. Seven hybrids were obtained and verified by the simple sequence repeat and cleaved amplified polymorphic sequence analysis of the genefael, indicating that the characteristic bands from S. alba were present in the hybrids. The hybridity was also confirmed by chromosome number counting because the hybrids possessed 62 chromosomes, corresponding to the sum of fusion-parent chromosomes. Chromosome pairing at meiosis was predominantly normal, which led to high pollen fertility,ranging from 66% to 77%. All hybrids were grown to full maturity and could be fertilized and set seed after self-pollination or back-crosses with B. napus. The morphology of the hybrids resembled characteristics from both parental species. An analysis of the fatty acid composition in the seeds of F1 plants was conducted and the seeds were found to contain different amounts of erucic acid, ranging from 11.0% to 52.1%.  相似文献

Interspecific crossing between L. temulentum L. and L. persicum Boiss. & Hohen. ex Boiss. was performed to clarify their interfertility based on the results of chromosome pairing, pollen fertility and seed set. Both parents were normal with a high percentage of chromosome association of ring bivalents in contrast to rod bivalents at metaphase I, pollen fertility and seed set, but F1 hybrids showed different proportions of them for each crossing combination. Chromosome affinity expressed by pairing was certainly a factor affecting the pollen fertility or seed set in F1 hybrids, but it was not the most important. The positive correlation was generally found between pollen fertility and seed set of F1 hybrids. The L. persicum accession with relatively high interfertility with L. temulentum was supposed to be derived from natural hybridization between L. temulentum and L. persicum. The degree of cytogenetic differentiation between L. temulentum and L. persicum existed because of lower chromosomal pairing, pollen fertility and seed set, but their F1 hybrids were partially fertile.  相似文献

The present study was conducted to investigate the effectiveness of the PhI gene transferred from Aegilops speltoides into bread wheat cultivar Chinese Spring (CS) in inducing homoeologous chromosome pairing in interspecific crosses using the Chinese Spring line, CS(PhI), carrying the gene. Chinese Spring, as well as CS(PhI), were crossed as female parents with three accessions of Ae. kotschyi (UUSS), one accession of Secale cereale (RR), two amphiploids of Triticum durum-Ae. caudata (AABBCC), and one amphiploid of Triticum durum-Ae. umbellulata (AABBUU). Meiotic metaphase I chromosome pairing was studied in all the interspecific crosses with CS as well as CS(PhI). There was significant increase in chiasma frequency in all the crosses with CS(PhI) over those with CS. The extent of induced homoeologous chromosome pairing by PhI in crosses of CS(PhI) with S. cereale was higher than with those of Ae. kotschyi, as indicated by higher chiasma frequency per pollen mother cell. Significant reduction in frequency of univalents and increase in bivalents (>14), multivalents, and chiasma frequency in crosses of amphiploids with CS(PhI) as compared to those of CS indicated induced homoeologous pairing between C and D, D and U, and C, D, and U genomes with AB genomes in the presence of PhI. The results of the present study unequivocally demonstrate the effectiveness of PhI gene transferred from Ae. speltoides in hexaploid wheat in inducing homoeologous chromosome pairing and suggest that the line CS(PhI) can be effectively used for precise transfer of useful alien genetic variations with least linkage drag.  相似文献

以不同类型的大白菜与紫甘蓝为材料,运用蕾期授粉结合胚挽救技术获得大白菜与紫甘蓝的种间杂种,并对其进行细胞学鉴定.结果表明,大白菜与紫甘蓝杂交,获得了57株F1代幼苗;对根尖染色体数量进行观察和杂种花粉特性调查发现,其中47株具有预期的染色体数目,2n =19,鉴定为真杂种,其花粉败育;另有6株具有38条染色体,鉴定为种间异源双二倍体,应该是发生了染色体自然加倍,其花粉可育.可育的杂种F1代与大白菜回交,获得了大白菜-紫甘蓝BC1代材料.田间观测结果显示,杂种F1代植株综合性状均介于双亲之间,BC1代植株包球明显,综合性状偏向母本大白菜.  相似文献

34个小麦品种(系)与黑麦进行了杂交,结果表明:亲和性在供试材料间存在明显差异,其中12个小麦品种表现了与中国春相似的亲和性;对34个小麦/黑麦属间杂种(F_1)的回交结实率和开放授粉条件下的自交结实率的统计表明,杂种育性普遍较差,但不同组合间存在明显不同,其回交与自交结实率的变异幅度分别为0—4.50%和0—3.087粒/穗;小麦与黑麦属间杂种(F_1)育性与杂交亲和性间存在明显相关,回交和自交结实率以及能够回交和自交结实组合的出现频率均随亲和性的增加而表现增加的趋势;在小麦品种演化过程中,亲和性与杂种育性均表现降低的趋势;小麦/黑麦属间杂种(F_1)PMC MI染色体配对水平普遍较低,平均交叉结仅为0.359,但组合间存在一定差异,变幅为0—1.3;平均99.936%的杂种(F_1)花粉表现败育,仅有0.064%的杂种(F_1)花粉能被KI—I_2正常染色,杂种(F_1)染色体配对水平与正常染色花粉频率、正常染色花粉频率与自交结实率间存在一定程度的相关性,相关系数分别为0.209和0.205。  相似文献

Twenty-two intergeneric hybrids from a cross between Brassica napus (AACC, 2n = 38) cultivar Oro and the ornamental crucifer Orychophragmus violaceus (OO, 2n = 24) were produced without embryo rescue. The plants were classified into three groups based on morphological and cytological observations and RAPD banding patterns. Plants of Group I had morphological traits of both parents and 2n = 29 chromosomes. In these plants, 62.1% of the pollen mother cells (PMCs) had the pairing configuration 1 III + 9 II + 8 I; the remaining PMCs had 10 II + 9 I. The plants possessed 97.6-98.8% B. napus specific and 9.2-11.7% O. violaceus specific RAPD fragments. Plants of Group II exhibited novel morphological traits and possessed 2n = 35, 36, or 37 chromosomes. Plants of Group III were morphologically similar to B. napus and possessed 2n = 19, 37, 38, or 39 chromosomes. Plants of Group II and Group III had 94.1-99.4% B. napus specific RAPD fragments and no O. violaceus specific RAPD fragments. Chromosome fragments were observed in PMCs of most of the F1 plants in all groups. Based on the cytological results and RAPD analysis, it is suggested that genome doubling and chromosome elimination occurred in the intergeneric hybrids of B. napus x O. violaceus.  相似文献

Seven enzymatic systems in F1 Aegilops kotschyi and Ae. biuncialis x Secale cereale hybrids, Aegilops kotschyi x S. cereale amphiploids and their parental species (Ae. kotschyi, Ae. biuncialis and S. cereale) were analysed by starch and polyacrylamide gel electrophoresis. Five of them (phosphoglucose isomerase, glutamic oxalacetic transaminase, esterase, acid phosphatase, and diaphorase) were polymorphic and two (malic dehydrogenase and superoxide dismutase) were monomorphic. Several isophorms of phosphoclucose isomerase, esterase, acid phosphatase, and diaphorase were detected in some hybrids and amphiploids, but absent in the parents. The role of regulators, translocations and recombination is discussed in relation to the origin of these new isophorms. Some parental isozymes were absent both in hybrids and amphiploids, probably as a result of the suppression of structural genes in new combinations of the three genomes.  相似文献

Hybrids between Aegilops kotschyi and Ae. biuncialis with Secale cereale were synthesized. Five Ae. kotschyi and four Ae. biuncialis accessions, as well as one inbred and four self-compatible forms of Secale cereale were used for crossing. The hybrids were produced directly from cultured embryos or through embryo callus culture. Sixty hybrids, 11 involving Ae. kotschyi and 49 Ae. biuncialis, had a stable somatic chromosome number 2n = 3x = 21. The plants showed good vegetative vigour and tillering capacity. Morphologically the hybrids were intermediate between their parents and completely sterile. In vitro propagation of Ae. kotschyi and Ae. biuncialis x S. cereale hybrids revealed that their capacity for callus production and plantlet regeneration - varies.  相似文献

Common wheat cv. Meri was crossed to a set of 21 Chinese Spring monosomic lines to characterize resistance to powdery mildew and to determine the chromosomal location of the gene(s). Monosomic F1 plants were allowed to self-pollinate and to produce F2 seeds. Seedlings of F2 and F3 plants and their parents were inoculated with isolates Ns 2 and 9 of Erysiphe graminis f. sp. tritici. Analysis of obtained data revealed that one major dominant gene conferring resistance is located on chromosome 1B of cv. Meri. The new gene is designated by symbol Pm28. On the basis of the trivalent configuration frequency (without univalent) at the 1st metaphase of meiosis it was found that two reciprocal translocations involving chromosomes 2A/5A and 5B/5D differentiate cv. Meri from cv. Chinese Spring. In the F1 monosomic hybrids, genes causing a decrease in pairing are found on chromosomes 4D and 6D, and genes enhancing pairing--on chromosomes 3A and 7B.  相似文献

Summary The meiotic behaviour of hybrids between Secale cereale carrying B chromosomes and S. vavilovii has been studied in order to estimate the effects of B chromosomes on hybrid meiotic pairing. The possible effect of Bs on the meiotic pairing of the offspring from backcrosses with S. vavilovii has been studied also. The results obtained clearly indicate that no detectable differences existed in chromosome pairing of hybrids with or without B chromosomes. The hypothetical existence of epistatic genes on cereale genome masking the effect of Bs has been rejected after the results obtained in backcrosses. Therefore, lack of qualitative genes controlling interspecific pairing on rye B chromosomes has been concluded. A quantitative effect of B chromosomes was detected only when they were in alien cytoplasm.  相似文献

The cytogenetic characteristics of Pseudoplatystoma corruscans and Pseudoplatystoma reticulatum and their F1, F2 and backcross hybrids were assessed by using chromosome banding techniques. The diploid number of 56 chromosomes was constant in all species and lineages, with a karyotypic formula containing 20 metacentric, 12 submetacentric, 12 subtelocentric and 12 acrocentric chromosomes. Nucleolar organizer regions (NORs) were identified in two subtelocentric chromosomes in the parents and hybrids, with partial nucleolar dominance in F1 and F2 specimens. Heterochromatic blocks were detected in the terminal and centromeric regions of some chromosomes in all individuals. For parental and hybrid lineages, 18S ribosomal clusters corresponding to NORs and 5S ribosomal genes were identified in distinct pairs of chromosomes. The striking conservation in the chromosomal macrostructure of the parental species may account for the fertility of their F1 hybrids. Similarly, the lack of marked alterations in the chromosomal structure of the F1 hybrids could account for the maintenance of these features in post-F1 lineages.  相似文献

Natural and synthetic hybrids are reported betweenPanicum, scribnerianum Nash and two other species,P. pacificum Hitchc. & Chase andP. occidentale Scribn. & Merr. The hybrids are morphologically intermediate between the parents, and those in whichP. pacificum is one parent strongly resemble the rareP. shastense Scribn. & Merr. All the parent plants studied cytologically had regular meiosis and a gametic number ofn = 9 chromosomes. The hybrids usually showed good pairing of chromosomes in meiosis, but irregularities such as univalents and lagging chromosomes were common. In the hybrids generally less than 7% of the pollen grains stained with cotton blue in lactophenol, and the plants set no seed, whereas the parent plants generally had over 90% pollen stainability and a high percentage of seed set. Pollen fromP. shastense, like that of the hybrids, was 3–4% stainable.Panicum shastense is believed to be based on a first generation sterile hybrid and the nameP. xshastense is proposed.  相似文献
