胚胎干细胞是从动物胚胎内细胞团分离的具有全能性的细胞.研究证明分离的小鼠胚胎干细胞在体外可以分化成心肌细胞,这一发现为小鼠胚胎干细胞向各种特化心脏细胞(动脉样细胞、血管样细胞、窦样细胞、心室样细胞、蒲肯野氏样细胞)分化提供了依据,使基因功能的研究在体外成为可能.1998年末人类ES细胞的成功培养奠定了心脏细胞的再生性治疗的战略基础.主要综述了目前ES细胞体外分化心肌细胞的进程,讨论了此进程对心脏基因研究的促进作用.  相似文献

目前胚胎干细胞(ESCs)建系的取材来源包括桑椹胚的卵裂球、囊胚的内细胞团(ICM)、上胚层细胞和原始生殖细胞(PGCs),甚至从新生鼠睾丸细胞也分离得到类ES样细胞系。这就提出了一个问题,什么是ESCs最接近的体内细胞来源。传统观念常常把ESCs等同于ICM细胞,也有学者认为ESCs更象上胚层细胞,而在已知的分子标记基因方面,ESCs所具有的特征更接近体内早期生殖细胞。不清楚ESCs最接近的体内细胞来源,可能是制约许多品系小鼠和大多哺乳类动物建系成功率提高的原因之一。ESCs系与EG细胞系的分离条件不同表明,加强对ESCs多能性维持基因调控研究具有重要意义。本文从ESCs的经典概念及其发展,早期胚胎细胞和生殖细胞发育规律,早期胚胎细胞、早期生殖细胞和ESCs的关系等方面进行综合分析,认为ESCs可能有多种接近的体内细胞来源。进一步应通过对ESCs建系不同的取材细胞和不同品系的ESCs间进行比较研究,以便弄清ESCs的来源和转化机制,为提高不同物种ESCs建系效率提供理论支持。  相似文献

目的:为探索鸡胚胎干细胞培养的优化条件,比较不同饲养层对鸡胚胎干细胞离体培养的效果。方法:用传至第2代的鸡胚成纤维细胞与鸭胚成纤维细胞,经丝裂霉素处理后制作饲养层,比较这2种饲养层以及不用饲养层对鸡胚胎干细胞离体培养效果的影响。结果:在以鸡胚成纤维细胞和鸭胚成纤维细胞作为饲养层的培养体系中,鸡胚胎干细胞均可保持良好的生长状态,而且2种饲养层对鸡胚胎干细胞克隆形成的影响差异不显著(P0.05)。结论:鸡胚成纤维细胞和鸭胚成纤维细胞均可作为较好的饲养层细胞用于鸡胚胎干细胞的离体培养。  相似文献

Unequal segregation of parental chromosomes in embryonic stem cell hybrids   总被引:4,自引:0,他引:4  
Chromosome segregation was studied in 14 intra- and 20 inter-specific hybrid clones generated by fusion of Mus musculus embryonic stem (ES) cells with fibroblasts or splenocytes of DD/c mice or Mus caroli. As a control for in vitro evolution of tetraploid karyotype we used a set of hybrid clones obtained by fusion of ES cells (D3) with ES cells (TgTP6.3). Identification of the parental chromosomes in the clones was performed by microsatellite analysis and in situ hybridization with labeled species-specific probes. Both analyses have revealed three types of clones: (i) stable tetraploid, observed only for ES x ES cell hybrids; (ii) bilateral loss of chromosomes of both ES and somatic partners; (iii) unilateral segregation of chromosomes of the somatic partner. Observed unilateral segregation was extensive in ES-splenocyte cell hybrids, but lower in ES-fibroblast hybrid clones. Developmental state of the somatic partner is presumably responsible for directional chromosome loss. Nonrandom segregation implies that initial differences in the parental homologous chromosomes were not immediately equalized implying at least transient persistence of the differentiated epigenotype.  相似文献

Long embryonic periods are assumed to reflect slower intrinsic development that are thought to trade off to allow enhanced physiological systems, such as immune function. Yet, the relatively rare studies of this trade-off in avian offspring have not found the expected trade-off. Theory and tests have not taken into account the strong extrinsic effects of temperature on embryonic periods of birds. Here, we show that length of the embryonic period did not explain variation in two measures of immune function when temperature was ignored, based on studies of 34 Passerine species in tropical Venezuela (23 species) and north temperate Arizona (11 species). Variation in immune function was explained when embryonic periods were corrected for average embryonic temperature, in order to better estimate intrinsic rates of development. Immune function of offspring trades off with intrinsic rates of embryonic development once the extrinsic effects of embryonic temperatures are taken into account.  相似文献

目的开发一种新的培养人胚胎干细胞(hESCs)的包被基质,使hESCs的培养更加简便。方法用甲醇固定的小鼠胚胎成纤维细胞(MEF)作为包被基质,人胚胎干细胞系X-01在该基质上生长,每隔5~6 d传代一次,培养10代后,对人胚胎干细胞特性进行检测,包括细胞形态、碱性磷酸酶染色、相关多能性基因的表达和分化能力。结果 hESCs在新的基质上生长良好,经10次传代后仍能保持典型的hESCs克隆形态。碱性磷酸酶染色阳性,免疫荧光染色Oct4、SSEA4、Tra-1-60为阳性,体外分化可形成拟胚体。结论此种固定的基质可以大量制备,长期保存,并可以长期维持hESCs的未分化状态,为人胚胎干细胞的体外扩增探索出了一个新的途径。  相似文献

In hybrid cells, not only are the nuclear genomes of parent cells fused, but their cytoplasm is as well. Mitochondrial DNA (mtDNA) is a convenient marker of cytoplasm that allows us to gain insight into the organization of hybrid-cell cytoplasm. We analyzed the parental mtDNA in hybrid cells resulting from the fusion of Mus musculus embryonic stem (ES) cells with splenocytes and fetal fibroblasts of DD/c mice or with splenocytes of M. caroli. Identification of parental mtDNA in hybrid cells was based on polymorphism among parental mtDNA for certain restriction endonucleases. We found that intra- and interspecific ES cell-splenocyte hybrid cells either entirely or partially lost mtDNA derived from a somatic partner, whereas ES cell-fibroblast hybrids retained mtDNA from both parents in similar ratios with a slight bias. The loss of somatic mitochondria by ES-splenocyte hybrids implies a nonrandom segregation of parental mitochondria, which was supported by a computer simulation of genetic drift. In contrast, ES cell-fibroblast hybrids show bilateral random segregation of the parental mitochondria judging from the analysis of mtDNA in single cells. Preferential segregation of somatic mitochondria does not depend on the differences in sequences of the parental mtDNA, but rather on the replicative state of parental cells.  相似文献

鸡胚胎生殖细胞在鼠胚成纤维细胞饲养层上的生长   总被引:1,自引:0,他引:1  
目的:探讨以鼠胚成纤维细胞为饲养层分离、培养鸡胚胎生殖细胞的方法和条件。方法:分离、培养12.5~13.5d鼠胚成纤维细胞。分离孵化5.5d鸡胚原始生殖细胞,原代培养时不使用饲养层,与性腺基质细胞共培养;继代培养时将其置于鼠胚成纤维细胞饲养层上,在含生长因子、分化抑制因子的培养体系中培养胚胎生殖细胞。结果:鼠胚成纤维细胞可连续传代18代以上(4个月),3~15代细胞可以用作饲养层细胞。分离的鸡胚胎生殖细胞在饲养层上可增殖形成典型胚胎生殖细胞集落,并能连续在体外培养超过9代。集落未分化标志高碘酸希夫反应(PAS)呈强阳性,体外分化实验表明胚胎生殖细胞具有多能性。结论:用鼠胚成纤维细胞作为饲养层能获得可连续增殖的胚胎生殖细胞。  相似文献

Summary A mammalian embryonic cell surface glycoprotein (ESGp), whose expression and biochemical structure seem to be developmentally regulated, has been isolated and characterized. The molecule expressed in two cell through morula stage mouse embryos has a molecular weight, by electrophoretic analyses, of 90 kDa. At the blastocyst stage, however, the molecule migrates as a broad, heterogeneous band ranging from 90 to 110 kDa. Evidence obtained from studies of embryonal carcinoma (EC) cells indicates that this band is actually a composite of three distinct molecules (molecular weight 90, 95, and 105 to 110 kDa), each of which is synthesized uniquely by one of the different cell types of the blastocyst: the embryonic ectoderm and visceral and parietal endoderms, respectively. A survey of various mouse tissues and cell lines has revealed that undifferentiated cells express the low molecular weight form (90 kDa) characteristic of embryonic ectoderm, whereas differentiated cells and adult tissues express the high molecular weight form (110 kDa) characteristic of parietal endoderm. Only the EC visceral endoderm cell analogues have been shown to express the intermediate molecule (95 kDa). In embryos, the antigen is uniformly distributed over the cell surface during early cleavage stages (two to eight cell); just before compaction, however, it seems to redistribute and becomes polarized at the outside exposed edges of blastomeres. In cultured EC cells, ESGp is found only in areas of cell-to-cell contact; free-standing surfaces of cells are negative for expression. It is possible, therefore, that ESGp may be involved in the intercellular adhesion of both EC cells and compacting embryos. Three and four independent cell lines with limited pluripotency were obtained from the inner cell mass cells of blastocysts and primordial germ cells of common voles, respectively. The results of cytogenetic analysis suggest that all these lines originated from the embryos of F1 Microtus rossiaemeridionalis × M. arvalis males and had a great number of near-triploid cells already during the early passages. The cells of these lines, like those of the inner cell mass, were characterized by the alkaline phosphatase activity. Nine independent cell lines were obtained as a result of hybridization of the mouse embryonic stem cells and vole splenocytes: eight lines and one line from hybridization with the M. kirgisorum and M. rossiaemeridionalis splenocytes, respectively. The cells of these lines expressed some properties of embryonic stem lines had a chromosome complement similar to the sum of two initial diploid sets of the mouse and vole.  相似文献

The embryonic development of two ctenopods Sida crystallina and Diaphanosoma brachyurum has been investigated by observing living embryos removed from the female brood pouches. The sequence of morphological changes was analysed, as was the time at which the activity of certain organs began. The timing of these events at 21–22 °C is documented for both species. During embryogenesis four membranes are cast off. The growth in length of embryos began only after the shedding of the external egg envelope. Growth rates throughout embryogenesis are documented. A new outline of ctenopod embryogenesis is proposed. It includes four stages (= instars) demarcated by the shedding of membranes (= moults), as is commonly accepted for juvenile and mature animals in ctenopods and other crustacean groups. These instars are well distinguished morphologically. Their characteristics are presented. Differences in the duration of embryogenesis in Sida and Diaphanosoma are explained by greater extension of the first, and especially the fourth, instars of Sida, while the duration of the second and third instars are approximately the same in both. A list of instar features is given which may be used to determine the instar (and approximate age) of embryos in the broud pouch of ctenopod females from natural populations. We studied the embryonic development of the phoronid Phoronis ijimai Oka, 1897. The egg cleavage is radial. The fourth and fifth cleavage furrows extend along the meridian of the egg. The blastula is flattened. Gastrulation occurs by a combination of epiboly, bending, and invagination. The mesoderm originates from two sources. The anterior mesoderm arises through immigration and gives rise to the first and second coeloms. The third coelomic mesoderm originates enterocoelically from the hindgut. The newly hatched larva has preoral and postoral ciliary bands, which can be compared with the corresponding ciliary bands of dipleurula and with the prototroch and metatroch of trochophore larvae.  相似文献

Developmental potential was assessed in 8 intra-specific and 20 inter-specific hybrid clones obtained by fusion of embryonic stem (ES) cells with either splenocytes or fetal fibroblasts. Number of chromosomes derived from ES cells in these hybrid clones was stable while contribution of somatic partner varied from single chromosomes to complete complement. This allowed us to compare pluripotency of the hybrid cells with various numbers of somatic chromosomes. Three criteria were used for the assessment: (i) expression of Oct-4 and Nanog genes; (ii) analyses of teratomas generated by subcutaneous injections of the tested cells into immunodeficient mice; (iii) contribution of the hybrid cells in chimeras generated by injection of the tested cells into C57BL blastocysts. All tested hybrid clones showed expression of Oct-4 and Nanog at level comparable to ES cells. Histological and immunofluorescent analyses demonstrated that most teratomas formed from the hybrid cells with different number of somatic chromosomes contained derivatives of three embryonic layers. Tested hybrid clones make similar contribution in various tissues of chimeras in spite of significant differences in the number of somatic chromosomes they contained. The data indicate that pluripotency is manifested as a dominant trait in the ES hybrid cells and does not depend substantially on the number of somatic chromosomes. The latter suggests that the developmental potential derived from ES cells is maintained in ES-somatic cell hybrids by cis-manner and is rather resistant to trans-acting factors emitted from the somatic one.  相似文献

为建立一种能够一次性分离任意两个植入前胚胎之间全部的差别表达的基因的方法 ,在已有的单胚胎操作技术的基础之上 ,对单个植入前胚胎抑制性消减杂交 (singlepreimplantationembryosuppressionsubtractivehybridization ,SPE SSH)方法进行了初步的探索 ,分离到OM2和MⅡ d 2的基因片段 ,经GenBank和文献检索发现 ,这两个基因具有在MⅡ期和 2细胞期特异性表达的特点 .利用cDNA阵列所进行的鉴定也获得了同样的结果 ,而且所使用的材料极少 ,说明SPE SSH是一种强有力的分离和识别早期发育相关基因片段的实验技术 .若与单个卵裂球分离技术相结合 ,还可用于分离和识别人类早期分子诊断的标记性基因  相似文献

目的 体外建立人胚胎干细胞传代培养方法,研究人胚胎干细胞细胞化学染色特性.方法 以小鼠胚胎成纤维细胞作为饲养层传代培养人胚胎干细胞,检测人胚胎干细胞、自发分化克隆及拟胚体的细胞化学染色特性.结果 人胚胎干细胞在小鼠胚胎成纤维细胞饲养层上传30代以上其形态保持不变;人胚胎十细胞碱性磷酸酶、过碘酸-雪夫反应、α-醋酸萘酚酯酶染色阳性,自发分化克隆细胞阳性程度明显减弱;人胚胎干细胞形成的拟胚体碱性磷酸酶染色弱阳性,过碘酸-雪夫反应、α-醋酸萘酚酯酶染色阳性.结论 小鼠胚胎成纤维细胞能支持人胚胎干细胞传代培养,细胞化学染色结果能初步鉴别人胚胎干细胞未分化特性.  相似文献

管氏肿腿蜂Scleroderma guani Xiao et Wu的卵在24℃和RH60%~70%条件下发育约140h孵化。根据胚胎外形的发育特点,可将整个发育过程可分5个阶段:早期发育阶段、胚胎伸长期、原躯原头分化阶段、器官形成阶段和胚胎成熟期。胚层形成趋于简单化,未形成解剖学上比较完整的呼吸、排泄及循环等系统。口道形成后,胚胎体积逐渐增加,这表明胚胎在发育过程中吸收了寄主的营养物质。  相似文献
