The aim of this study was to investigate the effects of aluminum (Al) exposure on the reproductive function in female rats. Forty female Wistar (5 weeks old) rats, weighing 110–120 g, were divided randomly into four groups. They were orally administrated with 0, 64.18, 128.36, and 256.72 mg aluminum chloride (AlCl3) per kilogram body weight in drinking water for 120 days. Levels of Al, estrogen (E2), progestogen (P), testosterone (T), follicle-stimulating hormone (FSH), and luteinizing hormone (LH) in serum were measured at the end of experiment. The results showed that levels of E2, P, FSH, and LH were significantly lower and Al concentration was significantly higher in all three Al-treated groups than those in the control group (GC). The level of T was significantly higher in the low- and medium-dose groups (GL and GM) (P < 0.05) but not in high-dose group (GH) compared with GC. The results suggest that the reproductive function of female rats is inhibited under long-term Al exposure in an Al dose-dependent manner.  相似文献

The influence of zinc (Zn) on the serum levels of triiodothyronine (T3), thyroxine (T4), thyroid-stimulating hormone (TSH) and sodium (Na), potassium (K), and calcium (Ca) was evaluated following ethanol toxicity to the rats. To achieve this, male Wistar rats (150–195 g) were given 3 ml of 30% ethanol orally, and zinc was given in the form of zinc sulfate (227 mg/l) in their drinking water daily for 8 weeks. Ethanol feeding resulted in a slight decrease in T3 and T4 levels and a significant increase in thyroid-stimulating hormone concentration, which may be due to the direct stimulatory effect of ethanol on thyroid. Interestingly, when zinc was given to these rats, all the above levels were brought quite close to their normal levels, thus indicating the positive role of zinc in thyroid hormone metabolism. Serum Zn and Ca levels were found to be reduced, but Na levels were raised upon ethanol feeding. Restoration of normal levels of these metals upon zinc supplementation to ethanol fed rats confirms that zinc has potential in alleviating some of the altered thyroid functions following ethanol administration.  相似文献

In most communities, there is a constant exposure to environmental pollutants with probable negative impact on the development of the nervous system. Among these pollutants are the sodium fluoride (NaF) and aluminum chloride (AlCl3) which may represent a real threat to the proper functioning of the brain. This study comprises two fundamentally different strategies; in the first one, pregnant rats were administered a daily dose of NaF (0.15 g /L) or AlCl3 (500 mg/L) in the drinking water either separately or in combination with each other from day 6 of gestation until just after weaning. In the second approach, the male rats born to mothers exposed to the pollutants were divided into two groups. In the first, rats were continued to be treated with the same pollutants administered to them in the drinking water at the same dose level until the age of 70 days. The rats of the second group were supplied with drinking water without either one of the pollutants for a similar period of time. The rats exposed to NaF separately or in combination with AlCl3 during the prenatal life and subsequently through the postnatal stages exhibited disturbance in the locomotor activities. This was concomitant with alterations in plasma, PTH, ACTH, and estradiol levels. Additionally, the serum levels of LH and testosterone were altered in the two groups treated with sodium fluoride during the prenatal and up to the weaning periods or in the group which continued to have the NaF until day 70 after birth.  相似文献

The effects of the acute immobilization stress on follicle-stimulating hormone (FSH), luteinizing hormone (LH), estradiol, and testosterone levels in rats were evaluated. The male and female rats were grouped in control and stress groups (7 animals in each group). Vaginal smears were taken daily from female rats and the rats in the proestrus were involved in the experiments. After a 1-h immobilization stress, blood samples from the heart were taken and the serum FSH, LH, estradiol and testosterone levels were measured by radioimmunoassay (RIA). The results were statistically analyzed using the Mann–Whitney U-test. A statistically significant increase of the levels of FSH and estradiol, but not of LH, was found in the female rats after the stress. A statistically significant increase of the FSH concentration was also revealed in the male rats, but there were no significant changes of the LH and testosterone levels. The scientific and applied (medical) aspects of the data obtained are discussed.  相似文献

Since melatonin injections administered near the end of the daily photoperiod influence both gonadal and thyroid hormones in the female hamster, the present study was designed to compare the effects of melatonin and hypothyroidism on the reproductive system and to determine whether thyroid status influenced the action of melatonin on the regulation of the hormones of reproduction. The effects of daily melatonin injections were determined in control hamsters, in hamsters rendered hypothyroid with thiourea, and in hypothyroid hamsters receiving thyroxin (T4) hormone replacement. As previously reported, melatonin injections disrupted estrous cyclicity, disrupted the normal pattern of gonadotropin secretion, and resulted in atrophy of the uterus and vagina. These changes coincided with depressed serum and pituitary prolactin (PRL), and depressed levels of estradiol. The effects of melatonin on uterus, vagina, ovary, and on gonadotropin levels were not prevented by T4 replacement, with the exception of a melatonin-induced increase in serum follicle-stimulating hormone (FSH). This suggested that the cessation of estrous cyclicity was not primarily a result of thyroid deficiency. Hypothyroidism, however, like melatonin, resulted in a reduced number of developing and mature follicles and corpora lutea in the ovaries, and in reduced uterine weight. It also produced follicular atresia, reduced the circulating levels of estradiol, and resulted in reduced incidence of estrus smears. T4 replacement, for 2 weeks, prevented the decline in mature follicles and corpora lutea, reduced the extent of follicular atresia, increased circulating levels of estradiol, and increased uterine weight. PRL and luteinizing hormone (LH) data also provided evidence for antagonistic effects of melatonin and T4 in female hamsters. These data raise the question whether melatonin-induced changes in circulating levels of T4 play a role in the seasonal cycles of reproductive competence in the female hamster.  相似文献

Numerous studies have suggested that estradiol (E) improves spatial memory as female rats with E perform better than those without E. However there is an inverse relationship between E and luteinizing hormone (LH) levels and LH could play a role. We examined whether treatment with the LH homologue human chorionic gonadotropin (hCG), would impair spatial memory of adult E-treated female rats. In the object location memory task, ovariectomized (ovxed) rats treated with E and either a single high dose (400 IU/kg) or a lower repeated dose of hCG (75 IU/kg hourly for 8 h) showed spatial memory disruption compared to ovxed rats treated with estradiol alone. Impairment was attributed to memory disruption as performance improved with shortened delay between task exposure and testing. Tests on another spatial memory task, the Barnes maze, confirmed that hCG (400 IU/kg) can impair memory: although E + veh treated animals made significantly fewer hole errors across time, E + hCG-treated did not. In humans, high LH levels have been correlated with Alzheimer's disease (AD). Because brain amyloid-beta (Aβ) species have been implicated as a toxic factor thought to cause memory loss in AD, we analyzed whether hCG-treated animals had increased Aβ levels. Levels of Aβ from whole brains or hippocampi were assessed by Western blot. hCG treatment to E-implanted females significantly increased soluble Aβ40 and Aβ42 levels. These results indicate that high levels of LH/hCG can impair spatial memory, and an increase in brain Aβ species may account for the memory impairment in hCG-treated rats.  相似文献

Plasma ANP (atrial natriuretic peptide), thyroid hormones, and calcium and magnesium levels as well as heart tissue calcium and magnesium concentrations were determined in male Wistar rats after exposure of 114 rats at low temperature (4°C) and 95 rats at high temperature (35–36°C) for 28 d. Plasma ANP, triiodothyronine (T3), thyroxine (T4), thyroid-stimulating hormone (TSH), free T3, and free T4 were estimated by radioimmunoassay, and plasma and heart tissue levels of Ca and Mg by flame atomic absorption spectrophotometry. Results were compared to a control group exposed at 20–22°C (76 rats). All the above parameters in control rats did not show statistically significant variations during the study. A significant increase of plasma ANP, T3, T4, Ca, and Mg concentrations developed during cold exposure, whereas a gradual decrease of plasma ANP, T3, T4, and Mg concentrations was revealed during hot exposure. A significant increase of heart tissue Mg concentrations developed during hot exposure. Results also indicate that plasma ANP and T3 levels are proportionally related, whereas an inverse relationship exists between plasma ANP and T3 levels and heart Mg concentrations, in both cold and hot exposed rats. In conclusion, ANP and thyroid hormones in relation to Ca and Mg play an important role in temperature adaptation.  相似文献

Rats, when vitamin A deficient, had increased plasma T3, T4 and free thyroxine indexes. Pituitary TSH and hypothalamic TRH content were increased in vitamin A deficient animals compared to pair-fed controls. The plasma TSH response to TRH was normal in the vitamin A deficient rats. Basal prolactin, LH and FSH levels did not differ significantly in the two groups. Both groups had significant increases in LH and FSH after LRH. Vitamin A deficiency produces biochemical hyperthyroidism. Our data are consistent with an abnormality in thyroid hormone feedback on the hypothalamic pituitary axis.  相似文献

Growth and the endocrine status of the pituitary and thyroid glands were studied in rats born and raised in a hypobaric chamber at a simulated high altitude of 3800 m (SHA); comparisons were drawn with similar rats at sea level. From birth until 40 days of age, SHA rats weighed significantly less than controls with the most striking growth impairment found in female SHA rats. Relative organ weights of anterior pituitary glands, ovaries and uteri from 40-day-old female SHA rats were significantly less than controls. Pituitary content of growth hormone (GH) was reduced in 40-day-old female SHA rats while the content of follicle-stimulating hormone (FSH) and luteinizing hormone (LH) were significantly increased over sea level controls. Plasma levels of GH, LH, FSH and thyrotropin (TSH) and pituitary TSH levels did not differ from control values. However, thyroidal uptake of 131I and plasma protein-bound 131I were significantly reduced in SHA rats as compared with controls. It is suggested that (1) the continuous exposure of developing female rats to hypoxia significantly impairs pituitary function and reproductive maturation, and (2) that despite other environmental factors acting on the developing organism at high altitude, growth retardation in rats born and raised at high altitudes is primarily a consequence of hypoxia.  相似文献

Light regulates numerous physiological functions and synchronizes them with the environment, in part by adjusting secretion of different hormones. We hypothesized that constant light (CL) would disturb pituitary‐thyroid axis. Our aim was to determine morphological and functional changes in this endocrine system in such extreme conditions and, based on the obtained results, to propose the underlying mechanism(s). Starting from the thirtieth postnatal day, female Wistar rats were exposed to CL (600 lx) for the following 95 days. The controls were maintained under the regular laboratory lighting conditions. After decapitation, pituitaries and thyroids were prepared for further histomorphometric, immunohistochemical, and immunofluorescence examinations. Concentration of thyroid stimulating hormone (TSH), total T4 and T3 (TH) were determined. Thyroid tissue of light‐treated rats was characterized by microfollicular structure. We detected no change in total thyroid volume, localization and accumulation of thyroglobulin, thyroid peroxidase, and sodium‐iodide symporter in the follicular epithelium of CL rats. The volume of follicular epithelium and activation index were increased, while volume of the colloid and serum levels of TH decreased. In the pituitary, the relative intensity of TSH β‐immunofluorescence signal within the cytoplasm of thyrotrophs increased, but their average cell volume and the relative volume density decreased. Serum TSH was unaltered. We conclude that exposure of female rats to CL induced alterations in pituitary‐thyroid axis. Thyroid tissue was characterized by microfollicular structure. Serum TH levels were reduced without accompanying increase in serum TSH. We hypothesize that increased secretion and clearance of TH together with unchanged or even decreased hormonal synthesis, resulted in decreased serum TH levels in CL group. We assume this decrease consequently led to increased synthesis and/or accumulation of pituitary TSH. However, decreased average TSH cell volume and relative volume density, together with unchanged serum TSH, point to additional, negative regulation of thyrotrophs. J. To investigate the influence of chronic ethanol consumption on circulating thyroid hormone levels, male and female rats were given 20% ethanol as the only drinking solution daily for 8 weeks. Blood ethanol levels ranged 30–45 mg/L. In male rats serum T4 decreased from the initial mean ± SD value of 5.2±1.4 to3.0 ±0.7 μg/dl; T3 decreased from initial value of 97±14 to 66±11 ng/dl and rT3 decreased from initial value of 19±9 to 10±1 ng/dl after 8 weeks of ethanol ingestion. Under similar experimental conditions, female rats showed a significant decrease in serum T4 and rT3 levels; however, T3 levels decreased slightly but not significantly as compared to initial values. The results indicate adverse effect of chronic ethanol intake on serum thyroid hormone levels in rats.  相似文献

Free radical production and lipid peroxidation are potentially important mediators in testicular physiology and toxicology. Polychlorinated biphenyls (PCBs) are global environmental contaminants that cause disruption of the endocrine system in human and animals. The present study was conducted to elucidate the protective role of vitamin C and E against Aroclor 1254-induced changes in Leydig cell steroidogenesis and antioxidant system. Adult male rats were dosed for 30 days with daily intraperitoneal (ip) injection of 2 mg/kg Aroclor or vehicle (corn oil). One group of rats was treated with vitamin C (100 mg/kg bw/day) while the other group was treated with vitamin E (50 mg/kg bw/day) orally, simultaneously with Aroclor 1254 for 30 days. One day after the last treatment, animals were euthanized and blood was collected for the assay of serum hormones such as luteinizing hormone (LH), thyroid stimulating hormone (TSH), prolactin (PRL), triiodothyronine (T3), thyroxine (T4), testosterone and estradiol. Testes were quickly removed and Leydig cells were isolated in aseptic condition. Purity of Leydig cells was determined by 3β-hydroxysteroid dehydrogenase (3β-HSD) staining method. Purified Leydig cells were used for quantification of cell surface LH receptors and steroidogenic enzymes such as cytochrome P450 side chain cleavage enzyme (P450scc), 3β-hydroxysteroid dehydrogenase (3β-HSD) and 17β-hydroxysteroid dehydrogenase (17β- HSD). Leydig cellular enzymatic antioxidants superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR), γ-glutamyl transpeptidase (γ-GT), glutathione-S-transferase (GST) and non-enzymatic antioxidants such as vitamin C and E were assayed. Lipid peroxidation (LPO) and reactive oxygen species (ROS) were also estimated in Leydig cells. Aroclor 1254 treatment significantly reduced the serum LH, TSH, PRL, T3, T4, testosterone and estradiol. In addition to this, Leydig cell surface LH receptors, activities of the steroidogenic enzymes such as cytochrome P450scc, 3β-HSD, 17β-HSD, antioxidant enzymes SOD, CAT, GPX, GR, γ-GT, GST and non-enzymatic antioxidants such as vitamin C and E were significantly diminished whereas, LPO and ROS were markedly elevated. However, the simultaneous administration of vitamin C and E in Aroclor 1254 exposed rats resulted a significant restoration of all the above-mentioned parameters to the control level. These observations suggest that vitamin C and E have ameliorative role against adverse effects of PCB on Leydig cell steroidogenesis.  相似文献

These studies attempted to elucidate the relationship between estradiol and luteinizing hormone (LH) secretion in chronically underfed (R) adult female rats. Examination of the response to ovariectomy revealed a significant delay in the onset of the postcastration increase in LH secretion in R females compared to control (C) animals. Chronic estrogen treatment in the form of Silastic capsules containing varying doses of E2. The response of C females was dose-dependent, ranging from complete suppression at 10 micrograms E2/animal to an absence of inhibition at 2.4 micrograms E2/animal. The acute response of LH secretion to E2 administration in the ovariectomized female indicated an increased suppression of plasma LH at 6 and 24 h after a single s.c. injection of estradiol benzoate (EB) in R compared to C animals. There was no difference between R and C rats in the ratio of free to protein-bound estradiol in the serum. The results of these studies suggest that the negative feedback efficacy of estrogen on LH secretion is significantly enhanced by reduced food intake in adult female rats and may be responsible for the loss of reproductive cyclicity in these animals.  相似文献

Intravenous injection of 600 μg PGE2 or PGI2 significantly increased serum LH and prolactin levels in estradiol treated ovariectomized rats. There was no effect on serum FSH concentration. PGE2 and PGI2 stimulated LH release in a non-dose dependent manner, while prolactin levels were positively correlated with the dose administered following PGI2 treatment. 6-keto-PGF at a comparable dose had no effect on pituitary hormone levels. Subcutaneous administration of 1 mg/kg or 60 mg/kg PGI2 for seven days significantly depressed serum LH level both in male and female rats. These doses had no effect on serum FSH or prolactin levels.  相似文献

The hypothalamic pituitary ovarian axis in adult female rats with 131-I induced hypothyroidism was studied before and after triiodothyronine (T3) replacement. Forty days after 131-I, hypothyroid (H) rats showed irregular cycles with predominantly diestrous vaginal smears, atrophied and underweight ovaries, and decreased serum T3, T4, LH and estradiol (E2). T3 replacement restored normal cycles and ovary weight and increased serum E2 levels above control values, while LH levels remained below the limit of detection of the RIA. The GnRH stimulation test performed on the day that the rats exhibited diestrous vaginal smears elicited a greater increase in FSH than in LH in H rats and a greater increase in LH than in FSH in both H-T3 treated and control rats. The data suggest that the lack of thyroid hormones in adult female rats seems to produce a reversion of sexual hormones to a prepubertal pattern, while T3 treatment restored normal estrous cycles and ovarian function.  相似文献

Studies using both Fisher 344 and Sprague-Dawley (SD) rat lines have shown that gestational and/or lactational maternal lead (Pb) exposure causes delayed reproductive maturation in their respective female offspring. Because these studies utilized different experimental regimens for dosing and for monitoring Pb levels, it has not been possible to determine which rat line provides the best model for low level Pb toxicity studies. This study was designed to address this issue. Adult Fisher and SD female rats were dosed with either a solution of PbAc containing 12 mg of Pb/ml or sodium acetate (NaAc) for controls. Dosing began 30 days prior to breeding and continued until their pups were weaned at 21 days of age. At the time of breeding and through weaning the blood lead (BPb) levels in the Fisher dams averaged 37.3 microg/dl and the SD dams averaged 29.9 microg/dl. Pb delayed the timing of puberty (p < 0.01) in Fisher offspring, and suppressed serum levels of luteinizing hormone (LH, p < 0.001) and estradiol (E2, p < 0.01). These effects did not occur in the SD offspring. Doubling the dose given to the SD rats increased their BPb levels to 62.6 microg/dl, yet there were still no effects noted. These results indicate that Fisher offspring are more sensitive to maternal Pb exposure with regard to puberty related insults than are SD rats, suggesting that the Fisher line may be a more reliable rodent model to study the effects of low level Pb toxicity.  相似文献

The effects of extremely low-frequency (ELF) magnetic fields on sex hormones of adult female Spague-Dawley rats were investigated. Adult female rats were exposed to a 50 Hz sinusoidal magnetic field at approximately 25 microT (rms) for 18 weeks before they returned to their normal life with unexposed counterparts. Serum level of Luteinizing Hormone (LH), Follicle Stimulating Hormone (FSH), progesterone, and estrogen were measured before, after, and during the exposure. Body and uterine weights were not affected by the field. A significant reduction in absolute and relative ovarian weights in exposed rats was observed when compared with sham-exposed controls (P < 0.05). The reduction in the levels of gonadotropins (FSH and LH) was significant after six weeks of exposure (P < 0.005). FSH levels were affected only on week 6 of exposure while LH remained affected during at 12 and 18 weeks (P < 0.05). Interestingly, no significant effects were found at 6 and 12 weeks after removing the field. The level of progesterone and estrogen was significantly decreased after 12 weeks of exposure (P < 0.05), while no other effects on progesterone level was observed during exposure or after removing the exposure. The level of estrogen was also significantly reduced at 12 weeks after removing the field (P < 0.05). These results suggest possible adverse effect on mammalian fertility and reproduction. The effects of ELF-MF on sex hormones were shown to be partly reversible.  相似文献

Immune system disorders are often accompanied by alterations in the reproductive axis. Several reports have shown that administration of bacterial lipopolysaccharide (LPS) has central inflammatory effects and activates cytokine release in the hypothalamus where the luteinizing hormone releasing hormone (Gn-RH) neurons are located. The present study was designed to investigate the effect of repeated LPS administration on the neuroendocrine mechanisms of control of the reproductive axis in peripubertal female rats (30-day-old rats). With this aim, LPS (50 μg/kg weight) was administered to the animals during 25, 27 and 29 days of age and sacrificed on 30 day of life. Gn-RH, γ?amino butyric acid (GABA) and glutamic acid (GLU), two amino acids involved in the regulation of Gn-RH secretion, hypothalamic content were measured. LH and estradiol serum levels were also determined and the day of vaginal opening examined. The results showed a significant increase in Gn-RH and GLU content (p?<?0.0001), shared by a reduction of GABA one (p?<?0.0001). LH and estradiol serum levels were decreased (p?<?0.01, p?<?0.001) and delay in the day of vaginal opening was also observed in treated animals. Present results show that repeated LPS administration impaired reproductive function, modifying the neuroendocrine mechanisms of control of the axis in peripubertal female rats.  相似文献

We examined the effect of acclimation to moderate hyperthermic environment on the ACTH, TSH, T3, T4 and corticosterone level, as well as the relative weight of hypophysis, thyroid and adrenal glands in streptozotocin-diabetic rats. Increased activity of the hypothalamo-pituitary-adrenocortical (HPA) axis has been demonstrated in diabetic animals, whereas insulin treatment restores the changes. Heat acclimation reduces the level of ACTH and corticosterone in control animals and moderates the hormonal disturbances caused by diabetes. Simultaneously, our study revealed impairment in the activity of the hypothalamo-pituitary-thyroid (HPT) axis. Acclimation to 35±1 °C resulted in significantly lower T3 and T4 levels in control, diabetic and insulin-treated animals. Relative weight of the hypophysis, thyroid and adrenal glands is decreased in heat-acclimated rats. Our assumption is that there might be a cross tolerance between diabetes and heat acclimation on a hormonal level.  相似文献

The objective of this study was to evaluate the transdermal efficiency of iodide microemulsion in treating iodine deficiency using rats as an animal model. Animals were fed either iodine-deficient diet (20 μg/kg iodide) or control diet (200 μg/kg iodide) over a 17-month period. At month 14, iodide microemulsion was applied topically in iodine-deficient group and physiological evaluations of thyroid gland functions were characterized by monitoring the thyroid hormones (T3, T4), thyroid-stimulating hormone (TSH), iodide ion excretion in urine, and the overall rat body weights in both groups. Moreover, morphological evaluations of thyroid gland before and after treatment were performed by ultrasound imaging and through histological assessment. Prior to microemulsion treatment, the levels of T3, T4, and TSH in iodine-deficient group were statistically significant as compared to that in the control group. The levels of T3 and T4 increased while TSH level decreased significantly in iodine-deficient group within the first 4 weeks of treatment. After treatment, iodide concentration in urine increased significantly. There was no statistical difference in weight between the two groups. Ultrasound imaging and histological evaluations showed evidence of hyperplasia in iodine-deficient group. Topical iodide microemulsion has shown a promising potential as a novel delivery system to treat iodine deficiency.  相似文献
