Action of some phytohormones on the respiration and on the absorption of phosphate by aging potato tuber discs. Discs of potato tuber incubated in aerated medium show an increase of the rates of respiration and phosphate absorption with aging time; the rates increase by two and nine respectively during the time period between 5 and 24 h of aging. Adenine or some N-6 substituted adenines [benzylaminopurine (BAP), furfurylaminopurine (FAP), methylaminopurine (MAP)], which present variable degrees of cytokinin activity, partially inhibit the increase of the rate of phosphate absorption and, to a lesser extent, the increase of the rate of respiration. Also abscisic acid (ABA), indole 3-acetic acid (IAA), and gibberellic acid (GA3) produce inhibition of the increase of the rate of phosphate absorption with varied effects on the respiration. With regard to phosphate uptake, the effects of ABA, 1AA and GA3 were additive to those of BAP. The effects on respiration were different from the effects on phosphate uptake, so that there is no direct relationship between inhibition of respiration and inhibition of phosphate uptake.  相似文献

Although fruit set and development are induced by applications of gibberellins, final fruit weight of gibberellin-induced parthenocarpic fruit is often less than that of pollinated fruit. We examined changes in the activities of sucrose-metabolizing enzymes and sugar accumulation in developing fruits of cultivated blueberry (Vaccinium ashei Reade) and their correlation with fruit growth upon pollination or exogenous applications of gibberellic acid (GA3). The objective was to determine if differences in fruit growth could be attributed to differences in enzyme activities and subsequent sugar accumulation in fruits. The fruit development period of GA3-treated fruits was 15 days longer than that of pollinated fruits. At maturity, GA3-treated fruit accumulated an average of 180 mg dry weight while pollinated fruit accumulated 390 mg dry weight. Dry weight accumulation in nonpollinated fruits was negligible and these fruits abscised by 45 days after bloom (DAB). The total carbon (C) cost (dry weight C + respiratory C) for fruit development was 109 and 244 mg C fruit-1 for GA3-treated and pollinated fruits, respectively. Hexose concentration increased to 100 mg (g fresh weight)-1 at ripening in both GA3-treated and pollinated fruits. Nonpollinated fruits reached a maximum hexose concentration at 45 DAB. Sucrose phosphate synthase (EC 2.4.1.14) and sucrose synthase (EC 2.4.1.13) activities reached a maximum of ≤5.0 μmol (g fresh weight)-1 h-1 in both GA3-treated and pollinated fruits. Soluble acid invertase (EC 3.2.1.26) activity increased to about 60 μmol (g fresh weight)-1 h-1 in both GA3-treated and pollinated fruits at ripening, while in nonpollinated fruits, a maximum soluble acid invertase activity of 0.12 μmol (g fresh weight)-1 h-1 was measured at 24 DAB. Insoluble acid invertase activity declined during the early stages of fruit growth and remained relatively low throughout fruit development. Neutral invertase activity was low throughout development, increasing to 5 μmol (g fresh weight)-1 h-1 at ripening in GA3-treated and pollinated fruits. Our studies demonstrate that blueberry fruit development does not appear to be limited by sucrose metabolizing enzyme activity and/or the ability to accumulate sugars in either GA3-treated or pollinated fruits.  相似文献

Gibberellic acid (GA3) inhibition of anthocyanin accumulation by carrot cell-suspension cultures was reversed by supplying dihydroquercitin or naringenin to the culture and not by supplying 4-coumaric acid or malonic acid. This suggested that gibberellic acid was inhibiting chalcone synthase, chalcone isomerase, or acetyl CoA carboxylase. Acetyl-CoA-carboxylase specific activity was the same in GA3-treated and untreated cultures and was not detected in cultures treated with uniconazole, an inhibitor of gibberellic acid biosynthesis. Chalcone-isomerase specific activity was lower in GA3-treated cultures than in untreated cultures and was lower in uniconazole-treated cultures than in the GA3-treated cultures. The total chalcone synthase activity in extracts from GA3- and from uniconazole-treated cells was not significantly different from that in extracts of untreated tissue. When these extracts were chromatographed on a Mono Q column, three peaks of chalcone synthase activity were found in extracts of nontreated cells, whereas only two of these peaks were detected in extracts of GA3-treated cells. The extracts from GA3-treated cells did not contain the peak of chalcone synthase activity that, in untreated cells, preceded the main peak. The correlation between the absence of this peak and the inhibition of anthocyanin accumulation suggests that this form of chalcone synthase is responsible for anthocyanin synthesis and that GA3 prevents this form from appearing in the cells.  相似文献

Growth of carrot and radish seedlings in nutrient culture was inhibited by pretreatment with three calmodulin inhibitors. There was little selective effect on specific organs, shoots, tap root and fibrous roots over a range of concentrations. Although pretreatment with CaCl2 (0.5 mM) did not affect growth of untreated seedlings, it partially reduced the inhibitory effects of trifluoperazine over the concentration range 0.01–0.05 mM. Trifluoperazine reduced the growth of GA3-treated seedlings but did not overcome the modifying effect of GA3 in favouring shoot/root ratio; ABA exacerbated its inhibitory effect on overall seedling growth and particularly on tap root development.Abbreviations GA3 gibberellic acid - ABA abscisic acid - CaCl2 calcium chloride - GAs gibberellins - Tfp trifluoperazine  相似文献

Capillary electrophoresis revealed that the endogenous level of ACC (1-aminocyclopropane-1-carboxylic acid) in the gametophytes of Anemia phyllitidis was elevated during GA3-induced male determination, whereas AOA (aminooxyacetic acid, specific inhibitor of ACC synthase) in untreated as well as in the GA3-treated gametophytes decreased concentration of ACC. The mechanism of ethylene involvement in controlling antheridiogenesis reflected at the level of ACC, which is supposed to mediate interactions between ethylene and gibberellins, is proposed.  相似文献

Summary Embryoless wheat (Triticum aestivum L.) half-seeds on incubation with gibberellic acid (GA3) showed a 2- to 2.5fold stimulation of monophenolase activity. The enzyme activity was not released into the incubation medium in GA3-treated half-seeds. The effect of GA3 was counteracted by the addition of abscisic acid (ABA) to the half-seeds. Adenosine-3,5-cyclic monophosphate and its structural analogues were ineffective in increasing the monophenolase activity. Actinomycin D and cycloheximide showed no inhibitory effecton the monophenolase activity in controls as well as in GA3-treated half-seeds, but on the contrary caused a 2- to 3fold stimulation of enzyme activity similar to that observed in endosperm treated with GA3 alone. However, there was no additive or synergistic enhancement of monophenolase activity when GA3 was tested in combination with cycloheximide or actinomycin D. GA3- or cyclic AMP-treated half-seeds showed no stimulation of o-diphenolase activity.  相似文献

The influence of after-ripening, sodium nitroprusside, potassium ferricyanide, cyanide, paclobutrazol and nitrite on germination of seeds of Nicotiana benthamiana was investigated as well as the influence of plant hormones such as gibberellins and abscisic acid. Dormancy of N. benthamiana seeds was broken by all treatments except treatments with abscisic acid, paclobutrazol and gibberellic acid (GA3). Gibberellins had an interesting effect on dormancy breakage of studied seeds which was dependent on use of particular gibberellin: GA3 or GA4+7. Unlike GA3, GA4+7 had broken seed dormancy.  相似文献

Respiration rate of foliarPelargonium discs was insensitive to ageing. The addition of BAP or GA3 to the ageing medium did not produce any effect. The presence of GA3 or BAP in the ageing medium induced an increase (27 %) or a decrease (45 %) of the phosphate uptake. The analysis of phosphorylated compound labelling showed that these two hormones decreased32P incorporation in the non-acid soluble fraction and increased32P incorporation in the acid soluble organic fraction. GA3 and BAP had little effect on the distribution of radioactivity between the different acid soluble compounds, but they increased the ATP level. These results suggest that both GA3 and BAP increase the basal metabolism, but they seem to act differently on the development of the uptake mechanism during ageing.  相似文献

The N-substituted phthalimide AC 94377 (1-(3-chlorophthalimido)-cyclohexanecarboxamide) was equally effective as a mixture of the gibberellins A4 and A7 (GA4/7) in breaking dormancy and stimulating germination of celery seeds when either was used in combination with ethephon or daminozide as a seed soak. Whereas seedlings emerging from GA4/7-treated seeds became etiolated in comparison with those from untreated seeds, those from AC 94377-treated seeds showed normal development. Preharvest sprays of gibberellic acid (GA3) increased the height of mature plants in comparison with untreated controls by about 16 per cent whereas AC 94377 was ineffective. The yield from GA3-treated plots was about 10 per cent greater than that from AC 94377-treated plots.  相似文献

Tomato (Lycopersicon esculentum Mill.) plants homozygous for the mutant pro gene, exhibiting the distinctive procera phenotype, appeared virtually identical to gibberellic acid (GA3)-treated isogenic normal plants. The pro gene and GA3 caused analogous increases in internode length, and in the length and number of cells in the outer cell layers of each internode. Internode number was also increased by pro and GA3 over the period of the experiment. Despite their greater length, the internodes of GA3-treated and pro plants reached their final size within a time period similar to that of internodes of untreated normal plants. The aleurone of RB-3 shrunken-2 (sh2) maize kernels is deficient in α-amylase activity during germination, but exogenous applications of gibberellic acid (GA3) (0.001–10 μm ) induced low levels of activity. The highest activity was measured in the aleurone of kernels treated with 10 μm GA3 (14,600 ± 945 units), but was lower than untreated Starchy (Su) aleurone tissues (35,280 ± 5,010 units). On isoelectric focusing gels, no α-amylase isozymes were detected in the untreated sh2 aleurone using starch zymograms or immunoblots, but the 1.0 and 10 μm mm GA3 treatments induced nearly all the isozymes (eight to ten) present in the Su aleurone. There was a very low level of α-amylase mRNA in the untreated sh2 aleurone, an intermediate level in the 1.0 μm GA3-treated sh2 aleurone, and the highest level in the untreated Su aleurone. On the confocal microscope, the 1.0 μm GA3-treated aleurone cells had enhanced levels of cytoplasmic membranes and RNA compared to untreated sh2 aleurone cells. The 1.0 μm GA3 treatment also induced shoot elongation in the sh2 seedlings. The data demonstrate that the sh2 aleurone is deficient in its function to produce α-amylases, and exogenous GA3 can partially restore cell function in the sh2 kernels.  相似文献

Xanthium plants were grown vegetatively and their developmental stages were designated by a previously described plastochron index (PI). Internodes of plants, both treated with gibberellic acid (GA3) and untreated, were marked with India ink and photographed during 3 successive days. The relative elemental rates of elongation d(dX/dt)/dX were estimated between 15.7 and 19.0 plastochrons. The rate of growth of the GA3-treated internodes was at least twice that of the control. The emerging pattern of acropetal internode elongation was similar in both GA3-treated and control plants. Only rates of growth were significantly higher in the GA3-treated plants. The acropetal pattern of internode elongation was the opposite of the basipetal pattern observed in Xanthium leaves but followed the acropetal pattern observed in Helianthus and Phaseolus internode growth.  相似文献

Tissue-culture-propagated own-rooted cv. Spartan apple trees (Malus domestica Borkh.) planted in 1979 were treated in 1983 and 1985 via a soil-line trunk drench with the plant growth retardant paclobutrazol [(2RS, 3RS)-1-(4-chlorophenyl)-4.4-dimethyl-2-(1,2, 4-triazol-1-yl) pentan-3-ol]. Seeds of immature fruits from untreated and treated trees were sampled in 1989 ca 75 days after full bloom. After seeds were freeze-dried, gibberellins (GAs) were extracted, purified and fractionated via C18 reversed-phase high-performance liquid chromatography (HPLC). Gibberellins A1, A3, A4, A7, A8, A9, A15, A17, A19, A20, A24, A34, A35, A44, A51, A53, A54, A61, A62, A63 and A68 were identified by using C18 HPLC, gas chromatography-selected ion monitoring and Kovats retention indices. Eight of the GAs identified were also quantified by using deuterated internal standards. The paclobutrazol applications caused a 55% reduction of vegetative shoot elongation in 1989, but both treated and untreated trees had developed a biennial bearing pattern by that time (heavy bloom or “on year’in 1989). Levels of early 13-hydroxylation pathway GAs, viz. GA53, GA19, GA20, GA1 and also GA3, were not altered by treatment. However, GA4, GA7 and GA9 were increased 13.4, 6.5 and 3.8 times, respectively, in seeds of fruit from treated compared to untreated trees.  相似文献

The patterns of protein synthesis in barley aleurone layers treated with gibberellic acid (GA3) and abscisic acid (ABA) are compared with the patterns observed in wheat germ in vitro translation assays directed by RNA isolated from similarly treated layers. When used alone, GA3 and ABA both induce the formation of new translatable mRNAs and cause new proteins to be synthesized. The effects of GA3 are more dramatic than those of ABA. In GA3-treated tissues, overall protein synthesis is redirected to produce large quantities of α-amylase and a few other GA3-induced proteins, while other protein synthesis is reduced or stopped. Large amounts of new translatable mRNA for α-amylase are also induced such that the dominant in vitro translation product is α-amylase. These changes are blocked by the simultaneous addition of ABA to the tissue. In GA3 plus ABA-treated layers, few changes in protein synthesis in vivo are observed when compared to protein synthesis in untreated tissue, although the induction of mRNA for α-amylase and the other GA3-induced mRNAs does occur. This indicates that ABA does not interfere with GA3 induction of translatable mRNAs but prevents the translation of these mRNAs in vivo. Thus ABA and potentially GA3 regulate the translation of proteins in vivo in barley aleurone layers.  相似文献

Seeds from heavily fruiting (on-year), mature untreated, and paclobutrazol-treated apple trees (Malus domestica Borkh. cv. Spartan) were sampled in mid-June 1987, mid-July 1987, and mid-July 1990. After seeds were freeze-dried, gibberellins (GAs) were extracted, purified, and fractionated via C18 reversed-phase high-performance liquid chromatography (HPLC). Nine GAs (GA1, GA3, GA4, GA7, GA8, GA9, GA19, GA20, and GA53) were quantified by the use of deuterated GA internal standards. Paclobutrazol trunk drench treatments reduced vegetative shoot elongation in the seasons that seeds were sampled by 55% or more. Between June 17, 1987 and July 15, 1987, the dry weight of seeds from both untreated and treated trees increased about 2.5 times and there were reductions, on a per seed basis, of GA4 in seeds from both untreated and treated trees, of GA7 in seeds from treated trees, and of GA9 in seeds from untreated trees. However, GA9 increased in seeds from treated trees. Changes in levels of some of the early-13-hydroxylation pathway GAs (GA15 GA3, GA8, GA19, GA20, and GA53) also occurred during the month. For mid-July harvested seeds, the pattern, with some exceptions, was that 2 years after paclobutrazol treatment (1987), levels of early-13-hydroxylation pathway GAs in seeds from treated trees were lower compared to controls but after 5 years (1990) their levels tended to increase. For the non-13-hydroxylated GAs (GA4, GA7, and GA9), 2 years after paclobutrazol treatment, GA4 levels were equal in seeds from untreated and treated trees, GA7 decreased in seeds from treated trees compared with controls, but GA9 levels increased. Levels of these three GAs were higher in seeds from treated trees 5 years after treatment (1990) but levels of GA4, GA7, and GA9 dramatically increased in seeds from treated trees 4 years after treatment (1989), as we previously reported.  相似文献

Satsuma [Citrus unshiu (Mak) Marc.] and Clementine [Citrus reticulata (Hort.) Ex. Tanaka, cv. Oroval] are two related species of seedless mandarins which differ in their tendency to set parthenocarpic fruits. Satsuma fruits naturally set parthenocarpically whereas Clementine mandarins show very low ability to set fruit in the absence of cross-pollination. The endogenous levels of gibberellins (GAs) and free and conjugated indole-acetic acid (IAA) and abscisic acid (ABA) throughout early stages of fruit development were investigated in seedless cultivars of both species. Analyses performed by full-scan combined gas chromatography-mass spectrometry (GC-MS) of extracts from ovaries at anthesis demonstrated the presence of GA19, GA20, GA29, GA1, GA8, GA3 and iso-GA3 in Satsuma mandarin, whereas only GA29, GA3 and trace levels of GA8 were detected in Clementine. At this developmental stage GA-like substances, as estimated by bioassay, reached their highest levels in Satsuma, while Clementine mandarins contained relatively lower levels. In both species the highest levels of free IAA were found at petal-fall stage at which time free ABA levels also peaked. Developing fruits of Clementine had higher amounts of both free IAA and ABA. In Satsuma, levels of conjugated IAA remained low throughout reproductive development whereas in Clementine they increased as the free form declined. In contrast, conjugated ABA was at low levels in Clementine but reached higher concentrations in Satsuma. These results suggest that in these mandarins the potential for setting parthenocarpic fruits is mainly influenced by the hormonal status of the fruit during the later stages of cell division and early stages of cell enlargement. Thus, the condition of low ability to set parthenocarpic fruits appears to be associated with lower levels of active GAs, lower capability to catabolize ABA to conjugated ABA and higher ability to conjugate IAA during this period.  相似文献

Early foliar spray treatments containing gibberellic acid(GA3) significantly lower the concentration ofthe bitter flavonoid naringin in fruit tissues. Sprayscontaining a surfactant and different levels ofGA3 (5, 50, 100, and 500 ppm) or abscisic acid(ABA) (5, 25, and 50 ppm) were applied to young,developing fruit on mature grapefruit (Citrusparadisi) trees during the period from April to June,beginning just after fruit set. The fruit were allowedto mature and were harvested early the following year.Harvested fruit were evaluated for weight, juicecharacteristics, and flavonoid concentrations.GA3 application resulted in larger mature fruit,which yielded juice with the same soluble solids valueas juice from control fruit, but with slightly loweracid percentages and lower concentrations of naringin.ABA treatment had little effect on juice solublesolids, acid content and naringin content except atthe highest concentration of 50 ppm, which lowerednaringin levels slightly in juice.  相似文献

Ethylene, indol-3-acetic acid (IAA), gibberellin-like substances (GAs) and abscisic acid (ABA) were analysed in extracts from normal, seed-containing and parthenocarpic tomato fruits throughout fruit development. Parthenocarpic fruit growth was induced with an auxin (4-CPA), morphactin (CME) or gibberellic acid (GA3) and compared with that of pollinated control fruits. Fruit growth was only affected by the treatment with GA3, decreasing size and fresh weight by 60%. The peak sequence of hormones during fruit development was ethylene-GAs-IAA-ABA. Seeded fruits contained the highest levels of IAA and ABA but the lowest levels of GAs. Also, in seeded fruits, a high proportion of IAA and ABA was found in the seeds whereas this was not the case for GAs.Hormone levels of tomato fruits may be successfully, easily and reproducibly altered by inducing parthenocarpic fruit growth and thus eliminating development of seeds which are a major source of hormone synthesis. In spite of markedly changed hormone levels, there was no obvious relationship between fruit growth and extractable hormones per se. However, the results indicate that a high ratio of GAs: auxins is unfavourable for growth of tomato fruits.  相似文献

GA17, GA19, GA20, GA29, GA44 and 13-hydroxy-GA12, now named GA53, were identified by GC-MS in immature seeds of Vicia faba (broad bean). Also identified were a GA catabolite, two polyhydroxykauranoic acids, and abscisic, phaseic and dihydrophaseic acids. The GAs of Vicia are hydroxylated at C-13, in common with those of other legumes. However the GAs of Vicia are not hydroxylated at C-3, nor do they appear to be readily conjugated. In these respects Vicia resembles Pisum, another member of the tribe Viciae. Gibberellic-acid (GA3) treatment of azukibean epicotyls resulted in alterations of the direction of newly deposited microfibrils, on the cell walls. Cells having transverse microfibrils on the inner surface of the wall were observed more frequently in GA3-treated epicotyls than in untreated or water-treated ones. This effect of GA3 was negated by simultaneously supplied colchicine. A crossed polylamellate structure was observed in the inner portion of the walls of GA3-treated cells, but not in the inner portion of the walls of colchicine-treated cells.
