Net photosynthesis, dark respiration, and growth of Rumex patientia L. exposed to a ultraviolet irradiance (288-315 nanometers) simulating a 0.18 atm·cm stratospheric ozone column were determined. The ultraviolet irradiance corresponding to this 38% ozone decrease from normal was shown to be an effective inhibitor of photosynthesis and leaf growth. The repressive action on photosynthesis accumulated through time whereas leaf growth was retarded only during the initial few days of exposure. Small increases in dark respiration rates occurred but did not continue to increase with longer exposure periods. A reduction in total plant dry weight and leaf area of approximately 50% occurred after 22 days of treatment, whereas chlorophyll concentrations remained unaltered.  相似文献

Pisum sativum L. was exposed to ultraviolet-B (UV-B) radiation (280-315 nm) in greenhouse and controlled environment chambers to examine the effect of this radiation on photosynthetic processes. Net photosynthetic rates of intact leaves were reduced by UV-B irradiation. Stable leaf diffusion resistances indicated that the impairment of photosynthesis did not involve the simple limitation of CO2 diffusion into the leaf. Dark respiration rates were increased by previous exposure to this radiation. Electron transport capacity as indicated by methylviologen reduction was also sensitive to UV-B irradiation. The ability of ascorbate-reduced 2,6-dichlorophenolindophenol to restore much of the electron transport capacity of the UV-B-irradiated plant material suggested that inhibition by this radiation was more closely associated with photosystem II than with photosystem I. Electron micrographs indicated structural damage to chloroplasts as well as other organelles. Plant tissue irradiated for only 15 minutes exhibited dilation of thylakoid membranes of the chloroplast in some cells. Some reduction in Hill reaction activity was also evidenced in these plant materials which had been irradiated for periods as short as 15 minutes.  相似文献

Summer squash plants {Cucurbita pepo L., cv. ‘Early ProlificStraightneck’) hydroponically cultured under optimal boronnutrition (+ B)3 until age 5 d were transferred to hydroponicsolutions supplemented with excess boron (+ + B)3. As boronaccumulated in the leaves with time, leaf conductance to watervapour, 14CO2 fixation, and chlorophyll content of the oldestleaf became significantly less than in the + B-control plantsof the same age; shoot growth essentially ceased. Boron alsoaccumulated in the roots; concomitant inhibition of root elongationand lateral root development resulted. These metabolic changesall occurred prior to the accumulation of sufficient boron tocause the appearance of any symptoms characteristic of borontoxicity. The effects of enhanced UV-B radiation on growth and photosynthetic activities were investigated in fronds of the aquatic fern Azolla microphylla Kaulf. The fronds were exposed to UV-B radiation intermittently once in 3 d during 12 d. Biomass and relative growth rate of UV-B treated Azolla plants and the heterocyst frequency of the UV-B treated symbiont decreased resulting in an increase in doubling time over the control. The doubling time was 3.08 d for control and 3.35 d for UV-B irradiated plants. Chl and carotenoid contents per unit fresh mass and photosystem 2 (PS2) activity also decreased under UV-B treatment. Measurements of photosynthetic activity in terms of fluorescence kinetics and PS2 mediated O2 evolution showed that the aquatic fern Azolla is sensitive to UV-B damage.  相似文献

The effect of ultraviolet-B (280 to 320 nm) irradiation on physiological activities of Anabaena flos-aquae and the water fern Azolla caroliniana has been studied under conditions where lethal effects of irradiation are absent. Nitrogenase activity, measured as acetylene reduction, specifically declined during irradiation with low levels of ultraviolet-B, whereas other physiological activities were unaffected. These findings indicate that measurement of acetylene reduction by cyanobacteria may serve as a specific, sensitive biochemical assay to assess environmental ultraviolet-B effects due to depletion of stratospheric ozone.  相似文献

In comparison with primary leaves of French bean plants grown under a photon flux density of 100 μeinstein m-2 s-1 (LP), leaves grown under 400 μeinstein m-2 s-1 (HP) were thicker (contained 82 to 104% more dry matter per blade area), had 44 to 48% higher stomatal frequency, 18 to 26% more chlorophyll (a + b) per leaf area unit and 31 to 42% less chlorophyll (a + b) per dry matter unit, 41% higher photosynthetic and 38% higher transpiration rates at light saturation, 33% higher stomatal conductance and 40% higher Photosystem 2 (H2O → K3[Fe(CN)6]) activity of isolated chloroplasts. There were no significant differences in the Photosystem 1 (TMPD/Ascorbate → MV) activity per unit amount of chlorophyll. Higher growth irradiance increased the ratio of frequencies of stomata in the upper/lower epidermes.  相似文献

Laboratory experiments were conducted to study the effects of soil moisture content, planting density, plant age, and the growth of multiple generations on the bioconcentration of weathered p,p'-DDE by the plant Cucurbita pepo. As soil moisture content increased from 7.4% to 29.9% (by weight), rates of contaminant accumulation by plant roots were increased by more than a factor of 2. Higher planting density also led to higher uptake, as the root bioconcentration factor (BCF, dry-weight ratio of contaminant concentration in the tissue to that in the soil) increased by 15-fold as the number of plants per pot was raised from 1 to 3. Concentrations of the compound in plant roots were inversely related to plant age, with root BCF declining by approximately a factor of 3 as plants aged from 14 to 28 d. Finally, no change in the bioavailability of the compound was observed in successive generations of plants grown in the same contaminated soil. The results suggest that phytoremediation is influenced by a number of factors and that the cleanup of contaminated soil can be enhanced by an understanding of environmental and other conditions affecting plant growth and bioconcentration.  相似文献

Two cultivars of rice (Oryza sativa L.) IR-36 and Fujiyama-5 were grown at ambient (360 microbars) and elevated CO2 (660 microbars) from germination through reproduction in unshaded greenhouses at the Duke University Phytotron. Growth at elevated CO2 resulted in significant decreases in nighttime respiration and increases in photosynthesis, total biomass, and yield for both cultivars. However, in plants exposed to simultaneous increases in CO2 and ultraviolet-B (UV-B) radiation, CO2 enhancement effects on respiration, photosynthesis, and biomass were eliminated in IR-36 and significantly reduced in Fujiyama-5. UV-B radiation simulated a 25% depletion in stratospheric ozone at Durham, North Carolina. Analysis of the response of CO2 uptake to internal CO2 concentration at light saturation suggested that, for IR-36, the predominant limitation to photosynthesis with increased UV-B radiation was the capacity for regeneration of ribulose bisphosphate (RuBP), whereas for Fujiyama-5 the primary photosynthetic decrease appeared to be related to a decline in apparent carboxylation efficiency. Changes in the RuBP regeneration limitation in IR-36 were consistent with damage to the photochemical efficiency of photosystem II as estimated from the ratio of variable to maximum chlorophyll fluorescence. Little change in RuBP regeneration and photochemistry was evident in cultivar Fujiyama-5, however. The degree of sensitivity of photochemical reactions with increased UV-B radiation appeared to be related to leaf production of UV-B-absorbing compounds. Fujiyama-5 had a higher concentration of these compounds than IR-36 in all environments, and the production of these compounds in Fujiyama-5 was stimulated by UV-B fluence. Results from this study suggest that in rice alterations in growth or photosynthesis as a result of enhanced CO2 may be eliminated or reduced if UV-B radiation continues to increase.  相似文献

The amino acid sequence of plastocyanin from marrow was determined. It consists of a single polypeptide chain of mol.wt. 10284 containing 99 amino acid residues. The sequence was determined by using a Beckman 890C automatic sequencer and by dansyl–phenyl isothiocyanate analysis of peptides obtained by the enzymic digestion of purified CNBr fragments. The sequence is in good agreement with the amino acid composition, except that fewer residues of glutamic acid were found in the sequence than were suggested by the composition. Evidence for histidine-37 was weaker than for the rest of the sequence. A `tree' of phylogenetic affinities was constructed by using several higher-plant plastocyanin sequences.  相似文献

Grape (Vitis vinifera cv Silvaner) vine plants were cultivated under shaded conditions in the absence of ultraviolet (UV) radiation in a greenhouse, and subsequently placed outdoors under three different light regimes for 7 d. Different light regimes were produced by filters transmitting natural radiation, or screening out the UV-B (280-315 nm), or screening out the UV-A (315-400 nm) and the UV-B spectral range. During exposure, synthesis of UV-screening phenolics in leaves was quantified using HPLC: All treatments increased concentrations of hydroxycinnamic acids but the rise was highest, reaching 230% of the initial value, when UV radiation was absent. In contrast, UV-B radiation specifically increased flavonoid concentrations resulting in more than a 10-fold increase. Transmittance in the UV of all extracted phenolics was lower than epidermal UV transmittance determined fluorimetrically, and the two parameters were curvilinearly related. It is suggested that curvilinearity results from different absorption properties of the homogeneously dissolved phenolics in extracts and of the non-homogeneous distribution of phenolics in the epidermis. UV-B-dependent inhibition of maximum photochemical yield of photosystem II (PSII), measured as variable fluorescence of dark-adapted leaves, recovered in parallel to the buildup of epidermal screening for UV-B radiation, suggesting that PSII is protected against UV-B damage by epidermal screening. However, UV-B inhibition of CO(2) assimilation rates was not diminished by efficient UV-B screening. We propose that protection of UV-B inactivation of PSII is observed because preceding damage is efficiently repaired while those factors determining UV-B inhibition of CO(2) assimilation recover more slowly.  相似文献

Attached leaves of pumpkin (Cucurbita pepo L.) were treated in high or moderate light at room temperature or a 1°C. The symptoms of photoinhibition appearing during light treatments at room temperature could be attributed to a decrease in the primary activity of PSII. However, when the light treatment was given at 1°C, the quantum yield of photosynthetic oxygen evolution decreased much more than would be expected from the decrease in the ratio of variable to maximum fluorescence at 77°K. Also, light treatment at 1°C lowered the chloroplast wholechain electron transfer capacity much more than it affected PSII electron transport (H2O to paraphenylbenzoquinone). Light treatments at both room temperature and 1°C led to an increase in Bmax, which indicates an increase in the proportion of PSIIβ centers. PSI was not affected by the light treatments, and the treatments in the dark at 1°C caused only minor changes in the measured properties of the leaves. We conclude that high light always inhibits the primary activity of PSII, but at low temperature there is greater inhibition of electron transfer from primary electron accepting plastoquinone of PSII to the plastoquinone pool, which leads to a drastic decrease in the quantum yield of oxygen evolution in the chilling-sensitive pumpkin.  相似文献

Entomopathogenic nematodes (EPNs) can be highly effective biocontrol agents, but their efficacy can be reduced due to exposure to environmental stress such as from ultraviolet (UV) radiation. Our objectives were to 1) compare UV tolerance among a broad array of EPN species, and 2) investigate the relationship between reduced nematode viability (after exposure to UV) and virulence. Nematodes exposed to a UV radiation (254 nm) for 10 or 20 min were assessed separately for viability (survival) and virulence to Galleria mellonella. We compared 9 different EPN species and 15 strains: Heterorhabditis bacteriophora (Baine, fl11, Oswego, and Vs strains), H. floridensis (332), H. georgiana (Kesha), H. indica (HOM1), H. megidis (UK211), Steinernema carpocapsae (All, Cxrd, DD136, and Sal strains), S. feltiae (SN), S. rarum (17C&E), and S. riobrave (355). In viability assessments, steinernematids, particularly strains of S. carpocapsae, generally exhibited superior UV tolerance compared with the heterorhabditids. However, some heterorhabditids tended to be more tolerant than others, e.g., H. megidis and H. bacteriophora (Baine) were most susceptible and H. bacteriophora (Vs) was the only heterorhabditid that did not exhibit a significant effect after 10 min of exposure. All heterorhabditids experienced reduced viability after 20 min exposure though several S. carpocapsae strains did not. In total, after 10 or 20 min exposure, the viability of seven nematode strains did not differ from their non-UV exposed controls. In virulence assays, steinernematids (particularly S. carpocapsae strains) also tended to exhibit higher UV tolerance. However, in contrast to the viability measurements, all nematodes experienced a reduction in virulence relative to their controls. Correlation analysis revealed that viability among nematode strains is not necessarily related to virulence. In conclusion, our results indicate that the impact of UV varies substantially among EPNs, and viability alone is not a sufficient measure for potential impact on biocontrol efficacy as other characters such as virulence may be severely affected even when viability remains high.  相似文献

UV‐B dose responses of two lines of pea were quantified at 2.3, 4.6, 6.9 and 9.2 kJ m−2 day−1 UV‐B (weighted according to Caldwell's generalised plant action spectrum) in controlled environments providing near‐field doses of photosynthetic radiation. Increasing UV‐B significantly increased UV‐B absorbing compounds in both lines. In the UV‐B sensitive line, JI1389, increasing UV‐B significantly inhibited most aspects of plant morphology and biomass. In the more UV‐B‐tolerant line, Scout, increasing UV‐B significantly reduced foliage area but had no effect on above‐ground biomass, although root biomass was significantly increased. Reduced plant height in JI1389 was caused by shorter internodes, in turn due to reduced cell number but not cell length. UV‐B had no significant effects on photosynthesis in either line. Significant dose responses were linear for the growth of the main stem in JI1389 but remaining significant dose responses were better fitted by quadratics with maximum UV‐B effects occurring in the range 5–7 kJ m−2 day−1 PAS300, due to stimulation of branch growth at the highest dose. However, growth stimulation by UV‐B was confined to PAS300 doses which at temperate latitudes would result only from rather extreme ozone depletions. We conclude that investigations using relatively low UV‐B doses, rather than those well above the current maximum, may be the best approach to both understanding of the fundamental mechanisms of plant responses to UV‐B and quantifying the magnitude of responses to stratospheric ozone depletion.  相似文献

The shape of the fitness function relating the decline in fitness with coefficient of inbreeding (f) can provide evidence concerning the genetic basis of inbreeding depression, but few studies have examined inbreeding depression across a range of f using noncultivated species. Futhermore, studies have rarely examined the effects of inbreeding depression in the maternal parent on offspring fitness. To estimate the shape of the fitness function, we examined the relationship between f and fitness across a range off from 0.000 to 0.875 for components of both male and female fitness in Cucurbita pepo ssp. texana. Each measure of female fitness declined with f, including pistillate flower number, fruit number, seed number per fruit, seed mass per fruit, and percentage seed germination. Several aspects of male fitness also declined with f, including staminate flower number, pollen number per flower, and the number of days of flowering, although cumulative inbreeding depression was less severe for male (0.34) than for female function (0.39). Fitness tended to decline linearly with f between f = 0.00 and f = 0.75 for most traits and across cumulative lifetime fitness (mean = 0.66), suggesting that individual genes causing inbreeding depression are additive and the result of many alleles of small effect. However, most traits also showed a small reduction in inbreeding depression between f = 0.75 and f = 0.875, and evidence of purging or diminishing epistasis was found for in vitro pollen-tube growth rate. To examine inbreeding depression as a maternal effect, we performed outcross pollinations on f = 0.0 and f = 0.5 mothers and found that depression due to maternal inbreeding was 0.07, compared to 0.10 for offspring produced through one generation of selfing. In at least some families, maternal inbreeding reduced fruit number, seed number and mass, staminate flower number, pollen diameter, and pollen-tube growth rate. Collectively these results suggest that, while the fitness function appears to be largely linear for most traits, maternal effects may compound the effects of inbreeding depression in multigenerational studies, though this may be partially offset by purging or diminishing epistasis.  相似文献

Nectar resorption and sugar translocation were studied in Cucurbita pepo (Cucurbitaceae) and Platanthera chlorantha (Orchidaceae) by micro-autoradiography. In both species, nectar was resorbed in pollinated and unpollinated flowers and ovules developing into seeds were found to be the main sugar sink. In C. pepo, the mobility of resorbed sugars in pollinated female flowers was higher than in unpollinated ones; male flowers showed lower mobility of resorbed sugar. In P. chlorantha, radioactivity was detected in pollinated flowers below and above labelled unpollinated ones: the nearer the flower, the stronger the accumulation of label in developing fruits.  相似文献

To test the hypothesis that leaf surface wax influences plant responses to UV-B, 6 lines of cultivated pea (Pisum sativum L.), selected as having more or less wax, were grown at 0 or 6.5 kJ m-2 day-1 plant-weighted UV-B against a background of 850–950 μmol m-2 s-1 photosynthetically active radiation. In the 4 lines with least leaf surface wax the amount of wax on adaxial and abaxial leaf surfaces was increased following exposure to 6.5 kJ m-2 day-1 UV-B, but UV-B decreased surface wax in Scout, which had the greatest wax deposits. On the adaxial leaf surface, UV-B radiation caused a shift in wax composition from alcohols to esters and hydrocarbons and the ratio of short to long chain length alkyl ester homologues was increased. There was no evidence of a shortening in carbon chain length of hydrocarbons, primary alcohols or fatty acids due to UV-B and no significant correlation between wax amount and UV reflectance from leaves. UV-B induced significant increases in UV-absorbing compounds in the expanded leaves and buds of most lines. UV-B reduced the growth of all lines. Foliage area (leaves plus stipules) declined by 5–30%, plant dry weight by 12–30%, and plant height by 24–38%. Reductions in growth occurred in the absence of any changes in chlorophyll fluorescence or photosynthetic rate. UV-B also had no major effect on carbon allocation patterns. The effects of UV-B on growth appeared to be due to changes in tissue extension and expansion. Indeed, many of the responses to UV-B observed in this study of pea appear more consistent with indirect effects being expressed in developing tissues rather than through the direct action of UV-B on mature tissues. There was no evidence that wax amount or biochemistry was associated with the sensitivity of the lines to UV-B radiation. Furthermore, induction of pigments was not correlated with changes in growth. However, lines with the greatest constitutive amounts of pigments in unexpanded bud tissues were most tolerant of elevated UV-B.  相似文献

Numerous branched plasmodesmata (pd) are present between bundle-sheath cells (BSCs) and specialized companion cells known as intermediary cells (ICs) in the minor-vein phloem of melon (Cucumis melo L.) and squash (Cucurbita pepo L.). These pd were found to be secondary, i.e., they form across existing walls. Sink, sink-source transition, and source tissues were sampled from developing and mature leaves. In sink tissue, IC precursors divide to produce the two to four ICs and associated sieve elements which are present by the time of the sink-source transition. Plasmodesmata along the interface between the IC precursor and adjacent BSCs in sink tissue are unbranched and few in number. Before the leaf tissue undergoes the sink-source transition, the number of pd channels (individual branches of pd) becomes more numerous. This increase in number of pd channels occurs at least in part and perhaps entirely by branching, resulting in more channels on the IC-side than on the BSC-side. In melon there is a 12-fold increase in the number of pd channels within the IC-side of the interface and a corresponding 9-fold increase in pd channels within the BSC-side. Thus, secondary pd form by the time of the sink-source transition and may be involved in phloem loading and photoassimilate export. Cyamopsis tetragonoloba L. seedlings were subjected to continuous ultraviolet (UV)-B radiation for 18 h and post-irradiated with "white light" (WL) and UV-A enhanced fluorescent radiations. UV-B treatment alone reduced plant growth, pigment content, and photosynthetic activities. Supplementation of UV-A promoted the overall seedling growth and enhanced the synthesis of chlorophyll and carotenoids with a relatively high photosystem 1 activity. Post UV-B irradiation under WL failed to photoreactive the UV-B damage whereas a positive photoregulatory effect of UV-A was noticed in electron transport rates and low temperature fluorescence emission spectra.  相似文献
