By means of microanatomical methods the inferior tracheobronchial lymph nodes have been investigated in 48 Wistar rats in 2 weeks and 3 months after discontinuance of inhalation of dimethylsulfate (DMS) vapours for 2 and 14 days by the animals in concentration 2.0 mg/m3, that is to say during rehabilitation period. Comparison of relative parameters of the structural components areas and cell composition of the lymph nodes has been carried out. During rehabilitation period after DMS inhalation for 2 days the cortical and medullary areas in histological preparations do not essentially differ from corresponding parameters of an acute experiment (2 days, 2.0 mg/m3, without rehabilitation). Amount (%) of cells with mitotic figures in the lymphoid nodules++ increases in 2 weeks and in 3 months. Contents of poorly differentiated cells during rehabilitation periods increase in the cortical plateau, but keeps nearly at the same low level as during the acute experiment in the lymphoid nodules++. In 2 weeks after DMS influence for 14 days, the cortical and medullary area in the histological preparations reach the control levels. In the lymphoid nodules++ a relative amount of reticular, poorly differentiated, mitotically dividing cells increases, and in the medullary cords contents of middle and small lymphocytes become greater in comparison with the acute experiment (14 days, 2.0 mg/m3, without rehabilitation).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

By means of histological methods inferior tracheobronchial lymph nodes have been investigated in 48 Wistar rats in 2 weeks and 3 months after cessation of inhalation of DNSV at maximum possible dose (0.1 mg/m3) for 2 and 14 days. Cell composition of the lymph nodes and cross sections of structural components have been compared. The cross section of the cortex decreases, while that of the medulla increases in comparison with corresponding parameters of the acute experiment. In 2 weeks and 3 months after DMSV effect for 2 days the part (%) of poorly differentiated cells and middle lymphocytes increases, while content of plasma cells in the medullary cords decreases. During rehabilitation after DMSV effect for 14 days a low level plateau is noted (as in the acute experiment). However, in lymphoid nodules++ within content of the poorly differentiated cells and middle lymphocytes in the cortical germinative centers in 2 weeks and 3 months the number of the poorly differentiated and reticular cells increases essentially, as well as mitotically dividing cells (in comparison to the acute experiment). In the medullary cords of the lymph nodes after 3 months of rehabilitation content of immature plasma cells is essentially higher than in 2 weeks of rehabilitation and than in the acute experiment. During rehabilitation the level of cells destruction in the lymph nodes noticeably decreases in comparison to the acute experiment.  相似文献

Cytoarchitectonics and rearrangement of cellular composition in functionally different zones of the rat tracheobronchial lymph nodes have been studied at a short effect of the industrial carbon disulfide vapour at a maximally admissible concentration (MAC) (1 mg/m3 and 10 mg/m3), as well as after the effect of MAC of carbon disulfide in the inhaled air for 4, 7, 14 days. The inhalation of the carbon disulfide vapours results in certain changes of the cytoarchitectonics of the lymph nodes studied in dependence on concentration and duration of the substance effect in the air inhaled. The effect of the MAC carbon disulfide, when the experiment lasts for 2 days, is accompanied with a decreased lymphocytopoiesis in all the structural components of the node and with a sharp intensification in the number of plasma cells of the medullary cords, that demonstrates local immunocytopoiesis. An opposite picture is noted, when carbon disulfide is applied in an elevated concentration (10 mg/m3)--plasmic reaction is sharply inhibited in the medullary cords, lymphocytopoiesis is noticeably increased, certain signs of allergic reaction are also seen. A prolonged effect of carbon disulfide vapours (up to 14 days) in MAC results in lymphocytopenia of the nodes, in increasing destructive processes and in decreasing cell proliferation. Differences in reaction of the structural components are revealed in the nodes studied at the toxic effect: in the cortical plateau and in the medullary cords suppression of the lymphocytopoietic activity is noted, and in the medullary cords--inhibition of immunocytopoiesis.  相似文献

The construction and cell composition of the cortex and medulla have been morphometrically studied after 1-, 3-, 5-, 7- and 14-aliquot injection of medical doses of hydrocortisone (0.25 mg per 100 g of body mass). The body mass of the animals decreases, as does the size of the mesenteric lymph nodes. The lymphocyto- and immunocytopoietic functions intensify in the medullary cords with a simultaneous inhibition of these processes in the cortical plateau and in the lymphoid noduli. On the 5th and 7th days there is some increase in the section area of the cortex, while that of the medulla decreases. The part of small lymphocytes in these structures grows large. Therefore, it is possible to suppose a definite tendency towards restoration of the structural components within two weeks.  相似文献

In the experiment performed on 127 dogs by means of cytospectrofluorometric analysis, using fluorochrome acridine orange in dynamics up to 1 year, changes in the level of chromatin activation and RNA content have been studied in lymphocytes of the germinative centers and the crown of lymphoid nodules, in the paracortical zone and medullary cords of the regional and contralateral popliteal lymph nodes, after subcutaneous injection of antigen (BCG vaccine, 0.2 mg/kg) into the lateral area of the foot of the left pelvic extremity. The immune response is accompanied with a periodical increase in the level of chromatin activation and RNA content in populations of lymphocytes in the regional and contralateral popliteal lymph nodes with maximum in 6 h, 3-7 days, 1-3 months after the antigen injection. The intensity of these processes has an unequal level in lymphoid cells of various structural components; it is higher in lymphocytes of the contralateral lymph node.  相似文献

Caprine hemal nodes were studied by light microscopy after glutaraldehyde fixation and epoxy resin embedding. A node consisted of a capsule, subcapsular and other sinuses, cortex, medulla and hilus. Elements of circulating blood filled the interstices of the reticular meshwork and associated macrophages which traversed the lumina of subcapsular and medullary sinuses. The latter were rare in 1-month-old goats, progressively increased in number and size in 2- to 4-month-old goats and coalesced with each other and the subcapsular sinus in adult animals. The cortical tissue appeared as lymphoid nodules. Circumferential lymphatic vessels abutted on outer margins of the nodules and gave origin to several radial lymphatics which branched and anastomosed between the medullary blood sinuses. Medullary cords were organized around the radial lymphatics. A single efferent lymphatic was formed at the hilum by confluence of the radial lymphatics. Our study, in contrast to earlier reports, shows that caprine hemal nodes possess efferent lymphatics. The present data suggest that the hemal nodes are involved, in addition to classical functions, in blood storage by hemoconcentration.  相似文献

Blood vessels, that bring blood to various areas of the human superficial inguinal lymph nodes are predominantly arterioles and precapillaries. They are often arranged radially from the hilus to the capsule and from the capsule towards the portal thickening. The arteries and arterioles of the portal and capsular trabeculae reach the paracortical zone, occupying an intermediate position between the medullary cords and the cortex of the lymph node. The arterioles of the paracortical zone, passing between the cortex and the medullary cords, acquire an arcuate appearance. In both directions from them (into central and peripheral areas of the node) precapillaries branch off at a right angle. The cortex is supplied with blood by the arteriolar branches of the paracortical zone and the capsule of the node. The cortical precapillaries branch into capillaries either within the lymphoid nodules, or along their periphery. In the medullary cords those arterioles branch, that get from the portal thickening, portal trabeculae and paracortical zone.  相似文献

In the lymphoid tissue of the iliac lymph node at an aseptic inflammation in the uterus the greatest changes have been revealed in 48 h from the moment, when the foreign body has been introduced. They are manifested at the tissue level (increasing content of macrophages, lymphoblasts, large lymphocytes, plasmoblasts, immature plasmocytes, mast cells, neutrophils), as well as at the organic level (increasing amount of secondary lymph noduli, increasing area of the paracortical zone, decreasing area of the cortical plateau, of the medullary intermediate sinus, medullary cords). Use of carbomineral sorbents ensures correction of the inflammation; this is demonstrated as reduction of reactive changes in the regional lymph node.  相似文献

Inherited deficiency of the enzyme adenosine deaminase (ADA) has been found in a significant proportion of patients with severe combined immunodeficiency disease and inherited defect generally characterized by a deficiency of both B and T cells. Two questions are central to understanding the pathophysiology of this disease: (1) at what stage or stages in lymphocyte development are the effects of the enzyme deficiency manifested; (2) what are the biochemical mechanisms responsible for the selective pathogenicity of the lymphoid system. We have examined the stage or stages of rat T-cell development in vivo which are affected by an induced adenosine deaminase deficiency using the ADA inhibitors, erythro-9-(2-hydroxy-3-nonyl)adenine (EHNA) and 2'-deoxycoformycin (DCF). In normal rats given daily administration of an ADA inhibitor, cortical thymocytes were markedly depleted; peripheral lymphocytes and pluripotent hemopoietic stem cells (CFU-S) all were relatively unaffected. Since a deficiency of ADA affects lymphocyte development, the regeneration of cortical and medullary thymocytes and their precursors after sublethal irradiation was used as a model of lymphoid development. By Day 5 after irradiation the thymus was reduced to 0.10-0.5% of its normal size; whereas at Days 9 and 14 the thymus was 20-40% and 60-80% regenerated, respectively. When irradiated rats were given daily parenteral injections of the ADA inhibitor plus adenosine or deoxyadenosine, thymus regeneration at Days 9 and 14 was markedly inhibited, whereas the regeneration of thymocyte precursors was essentially unaffected. Thymus regeneration was at least 40-fold lower than in rats given adenosine or deoxyadenosine alone. Virtually identical results were obtained with both ADA inhibitors, EHNA and DCF. The majority of thymocytes present at Day 9 and at Day 14 in inhibitor-treated rats had the characteristics of subcapsular cortical thymocytes which are probably the most ancestral of the thymocytes. Thus, an induced ADA deficiency blocked the proliferation and differentiation of subcapsular cortical thymocytes which are the precursors of cortical and medullary thymocytes.  相似文献

Longitudinal histological sections of lymphoid patches (LP) in the small intestine, stained after van Gieson and with hematoxylin--eosin, obtained from 105 corpses of persons at the age of 0 up to 90 years have been studied. The LP parenchyma is presented as accumulation of lymphoid nodules++ with a germinative center, or without it and diffuse lymphoid tissue. The lymphoid nodules++ situate both in the mucous membrane and in the submucous tela of the small intestine. The area of the LP components in the newborns is the greatest--84% from the whole area of the LP section. During the human life it decreases and by the old age it makes 47%. The lymphoid nodules++ area with the germinative center in the newborns makes 21% from the area of the LP section, in children from 1 to 3 years of age it increases up to 50%, then it gradually decreases and in the elderly age it makes only 3.8%. The diffuse lymphoid tissue in the LP composition is revealed in all age periods. Size of the lymphoid nodules++ patches, situating in the jejunum and ileum walls in the newborns is the same. In children at the age of 10 days--12 years the LP nodules++ in the ileum are larger than those in the sejunum. The size of the LP noduli increase up to the period of the second childhood then decreases. The height of the lymphoid noduli in persons of mature, elderly and old ages decreases twice in comparison with the children of 3-12 years of age.  相似文献

Interrelations of the blood and lymphatic systems of the uterus have been examined in rats at pregnancy complicated with the ++phlebo-occlusive syndrome. Blood stream impediment in the caudal vena cava results in increasing diameter of the arterioles, capillaries and venules of the endo- and myometrium. Certain disturbances of blood circulation in the uterus at the ++phlebo-cclusive syndrome in the pregnancy animals, as a rule, reflect in the uterine lymph outflow; this is demonstrated as dilatation and deformity of the lymphatic vessels and capillaries, appearance of protrusions of the lymphatic vessels wall. There is a definite co-ordination in the reaction to the caudal vena cava occlusion in the intra-, ++extra-organic uterine lymphatic bed and in its regional lymph nodes, manifested as the following morphological signs: decreasing relative volume, that the lymphoid tissue occupies and increasing relative volume of the cortical and medullary intermediate sinuses.  相似文献

Using 68 3-month-old male albino rats, it was established that the pattern of the changing interrelationships of B- and T-cell areas in the spleen and in the popliteal, inguinal and medial iliac lymph nodes regional for the experimental limb during the initial stage after hind limb autotransplantation, with or without sciatic nerve alloplasty, represents a universal type of initial response of the peripheral immune organs to external challenge which takes place in three steps: 1) an increase in the number and size of the lymph nodules, 2) enlargement of T-cell areas, 3) an increase in the number of structures containing antibody-forming cells (the medullary cords in the lymph nodes and the splenic cords). Sciatic nerve alloplasty gives rise to expansion of the medullary cords in the lymph nodes and the marginal zone and cords in the spleen, with parallel significant enlargement of T-cell areas.  相似文献

By means of mathematical methods, quantitative and qualitative changes were studied in different structural components of the mesenteric (ileocecal) lymph nodes in normal monkeys (Macaca rhesus) and under per os administration of Salmonella typhi murium, streptomycin-dependent. Cellular composition was calculated in the cortical plateau, cortical (lymphoid) cords and in follicules. Average percent of every cell type was determined. Vaccine administration, was stated to inhibit cytopoiesis in the cortical plateau and in the follicules with light centers. An inverse correlation was noted between the content of small and medium size lymphocytes. Different reactivity of certain structural components in the lymph nodes was demonstrated. As a response to the vaccine administration, plasmocellular acidophilic and macrophagal reactions were most pronounced in the cortical (lymphoid) cords.  相似文献

The investigation has been performed on 44 Wistar rats, some of them at the age of 2 months have been subjected to thymectomy. In 3, 6 and 12 months in the spleen, axillary and popliteal lymph nodes density of cell populations, position of lymphatic nodules have been determined, and in the liver--activity of key enzymes of gluconeogenesis. In the animals with the deficiency of the thymic hormones the most manifested changes are seen in 3 and 12 months: decrease in cell population density in the lymph nodes and in their T-dependent zones, decrease in amount of small lymphocytes, in density of the lymphoid nodules arrangement. The dynamics of processes is also followed at analysis of the key enzymes activity of gluconeogenesis in the liver: glucose-6-phosphatase activity is decreased in 3 months after thymectomy, phosphoenolpyruvate carboxycinase activity--in 12 months. Fructose-1,6-diphosphotase activity is not changed. The thymic hormones deficiency is supposed to disturb the ratio of cyclic nucleotides and in this way participate both in inhibition of the immune function and in decrease of metabolic activity mediated by cyclic adenosine monophosphate.  相似文献

With the aim to study effectiveness+ of endolymphatic (EL) administration of ampicillin (AC), using the model of an acute diffuse septic peritonitis in dogs, the morphological and morphometrical investigation has been performed concerning the state of the lymph nodes (LN), which are regional as regards the pathological focus (pelvic) and remote (tracheobronchial, mesenteric) and hemomicrocirculatory bed (HMCB) of the small intestine mesentery. All LN groups studied are involved in the pathological process, that produces certain increasing disturbances in the structure and cell composition in LN. In 6 h the changes are especially manifested in the pelvic LN, and in 18 h--in the animals without application of AC, or at its intramuscular injection LN lose their typical structure. Their dimensions and number of lymphoid nodules++ and medullary cords decrease, a sharp impoverishment of lymphocytes in LN is observed. By this time critical disturbances in the HMCB structure develop; they are characterized as presence of great amount of avascular areas in the mesentery, extended capillary loops, plasmatic saturation of interstitium. When AC is injected endolymphatically, simultaneously with peritonitis modelling T- and B-dependent zones in LN are preserved, a high volumetric part of lymphocytes is kept in all groups of LN, structure and function of HMCB are normalized. The pronounced delay in development and decreasing manifestation of infective-toxic disorder in LN and HMCB depend on effective concentrations of the antibiotic, produces in the lymphatic system.  相似文献

Summary Ontogenetic differentiation of the human thymus was investigated in 50 embryos by means of light and electron microscopic methods in an attempt to clarify the morphogenesis of the complicated microecology of thymic tissue. At the 8th gestational week (g.w.), the primordium of the thymus contains almost exclusively undifferentiated epithelial cells. At the 10th g.w., the epithelial cells in the central part are spindle-shaped. During the subsequent weeks the cortical region of the thymus becomes separated into lobes by mesenchymal septa containing hemopoietic precursor cells and large electronlucent cells with irregularly shaped nuclei. The latter cells are also found in the deeper presumptive medullary regions of the thymus; they differentiate into interdigitating reticulum cells (IDC). The permeation of the medulla of the thymus by non-epithelial IDC occurs concurrently with the formation of cortical and medullary epithelial cells. Between the 12th and 14th g.w. the cortical and medullary differentiation is completed. At this time-stage cortical small lymphocytes differ in morphological shape from medullary lymphocytes, the latter acquiring the appearance of immunocompetent T cells and establishing intimate contact with the IDC.These findings indicate that the thymic cortex and medulla contain different epithelial cells. In addition, the thymic medulla displays cells characterized by the morphology of typical interdigitating reticulum cells of peripheral lymphoid tissue. The morphological development and plasticity of embryonic and postnatal rat adrenal medullary cells were studied in homologous adrenal grafts to the anterior chamber of the eye. The eyes of recipient rats were adrenergically denervated 10 days prior to grafting by extirpation of the superior cervical ganglion in order to increase levels of NGF and NGF-like activities in the iris. Grafts taken at the 15th day of embryonic development (E15), i.e., at the beginning of immigration of medullary progenitor cells into the adrenal cortical anlagen, contained no cortical or mature medullary cells after 2 weeks in oculo. Numerous sympathoblastic cells, however, were located at the anterior surface of the iris. E 16 and E 17 transplants showed abundant mature cortical tissue after 2 weeks. Small groups of medullary cells with the ultrastructural characteristics of mature pheochromoblasts or young chromaffin cells were interspersed among cortical cells without forming a discrete medulla. Neuronal cells were exclusively found outside the cortical cell mass. Sympathoblasts grew at the surface of the iris, while young sympathetic nerve cells, which were invested by Schwann cells and received synaptic axon terminals, were embedded into the stroma of the iris. Grafting of E 21 adrenals yielded very similar results except that, in a few instances, young chromaffin cells were located outside the cortex and sympathetic nerve cells were seen to be in close contact with cortical cells. In transplants of adult medullary cells typical mature adrenaline and noradrenaline cells were clearly distinguishable after 8 weeks even in the absence of cortical cells. The only indication of phenotypical changes in these cells was the formation by some of them, of neuritic processes which could be visualized in glyoxylic acid-treated whole mounts of irises. These results are compatible with the idea that embryonic adrenal medullary cells have the environmentally controlled potential to develop along the neuronal or endocrine line, but could also be interpreted in terms of a selection of a specific subpopulation with predetermined potentialities by a specific microenvironment. Moreover, these results suggest that increasing differentiation of medullary cells is accompanied by progressive restrictions in their genetic program, which eventually prevent full transdifferentiation of mature chromaffin into neuronal cells.  相似文献

A field experiment has been performed in white noninbred rats in order to study morphometrically structural reconstructions and cellular composition characteristics in the submandibular lymph nodes on the 1st, 3d, 7th, 14th, 21st, 30th and 50th days after lifting them to the altitude of 3,375 m above sea-level. The data obtained demonstrate a stable increase in small lymphocytes contents during the days of the observations; they are accompanied with an increasing specific area of the folliculi and a decreasing area of the cortical plateau. An essential decrease in the plasmic cells and dividing cells contents noted during the whole experiment is regarded to demonstrate an inhibition of the organism's immune reactivity at hypoxic hypoxia. Certain time periods are revealed which are accompanied, during the adaptation period to the high-mountain conditions, with general reactions of definite cell types and the lymph node structures: the 1st-7th days; the 14th-30th days; the 45th day and more. According to the character of the cell reactions in the experiment performed, it is possible to arrange the main structures of the submandibular lymph nodes in the following order: folliculi, cortical plateau, sinuses, medullary cords.  相似文献

Influence of strong sulfide baths of the sanatorium "Talgi" to the popliteal and superficial inguinal lymph nodes (LN) has been studied in 35 rats (140-150 g of body mass) and 15 animals have been used as the control. The slices are stained with hematoxyli-eosin, after van Gieson, Romanovsky-Giemsa, Kurnik and silver nitrate impregnation after Foot. The section area of the LN decreases, but amount of lymphoid nodules increases (especially in deep layers of the cortex). In the germinative centers amount of middle lymphocytes and mitoses becomes larger. In the internodular zones of the popliteal nodes amount of macrophages and eosinophiles enlarges significantly, and in the inguinal-amount of mast cells and eosinophiles. In the medullary sinuses amount of macrophages declines. In the medullary cords amount of immature plasma cells and blasts increases, while amount of mature cells decreases.  相似文献

American alligator (Alligator mississippiensis) ovary development is incomplete at hatching. During the months following hatching, the cortical processes of oogenesis started in ovo continues and folliculogenesis is initiated. Additionally, the medullary region of the gonad undergoes dramatic restructuring. We describe alligator ovarian histology at hatching, 1 week, 1 month, and 3 months of age in order to characterize the timing of morphological development and compare these findings to chicken ovary development. At hatching, the ovarian cortex presents a germinal epithelium containing oogonia and a few primary oocytes irregularly scattered between somatic epithelial cells. The hatchling medulla shows fragmentation indicative of the formation of lacunae. By 1 week of age, oocytes form growing nests and show increased interactions with somatic cells, indicative of the initiation of folliculogenesis. Medullary lacunae increase in diameter and contain secretory material in their lumen. At 1 month, nest sizes and lacunar diameters continue to enlarge. Pachytene oocytes surrounded by somatic cells are more frequent. Trabeculae composed of dense irregular connective tissue divide cortical nests. Three months after hatching oocytes in meiotic stages of prophase I up to diplotene are present. The ovary displays many enlarged follicles with oocytes in diplotene arrest, thecal layers, lampbrush chromosomes, and complete layers of follicular cells. The medulla is an elaborated complex of vascularized lacunae underlying the cortex and often containing discrete lymphoid aggregates. While the general morphology of the alligator ovary is similar to that of the chicken ovary, the progression of oogenesis and folliculogenesis around hatching is notably slower in alligators. Diplotene oocytes are observed at hatching in chickens, but not until 3 months in alligators. Folliculogenesis is completed at 3 weeks in chickens whereas it is still progressing at 3 months in alligators.  相似文献
