This paper describes the association of two bioreactors: one photoautotrophic and the other heterotrophic, connected by the gas phase and allowing an exchange of O2 and CO2 gases between them, benefiting from a symbiotic effect. The association of two bioreactors was proposed with the aim of improving the microalgae oil productivity for biodiesel production. The outlet gas flow from the autotrophic (O2 enriched) bioreactor was used as the inlet gas flow for the heterotrophic bioreactor. In parallel, the outlet gas flow from another heterotrophic (CO2 enriched) bioreactor was used as the inlet gas flow for the autotrophic bioreactor. Aside from using the air supplied from the auto- and hetero-trophic bioreactors as controls, one mixotrophic bioreactor was also studied and used as a model, for its claimed advantage of CO2 and organic carbon being simultaneously assimilated. The microalga Chlorella protothecoides was chosen as a model due to its ability to grow under different nutritional modes (auto, hetero, and mixotrophic), and its ability to attain a high biomass productivity and lipid content, suitable for biodiesel production. The comparison between heterotrophic, autotrophic, and mixotrophic Chlorella protothecoides growth for lipid production revealed that heterotrophic growth achieved the highest biomass productivity and lipid content (>22%), and furthermore showed that these lipids had the most suitable fatty acid profile in order to produce high quality biodiesel. Both associations showed a higher biomass productivity (10–20%), when comparing the two separately operated bioreactors (controls) which occurred on the fourth day. A more remarkable result would have been seen if in actuality the two bioreactors had been inter-connected in a closed loop. The biomass productivity gain would have been 30% and the lipid productivity gain would have been 100%, as seen by comparing the productivities of the symbiotic assemblage with the sum of the two bioreactors operating separately (controls). These results show an advantage of the symbiotic bioreactors association towards a cost-effective microalgal biodiesel production.  相似文献

Auxin autotrophic and heterotrophic lines of tobacco calli may differ not only in their indoleacetic acid (IAA) synthetizing abilities and sensitivities to exogenous auxins, but also in their gene expression patterns. Auxin autotrophic callus tissues from the leaf protoplasts of transgenic Nicotiana tabacum SR1 plants involving mas1′::GUS gene fusion were generated and the growth of cultures was compared with that of the heterotrophic lines of the same transgenic tissues on MS medium containing different concentrations of IAA or 2,4‐d . The mas1′::GUS gene fusion expression was investigated, together with the glutathione S‐transferase activities (GST, EC 2.5.1.18) in auxin autotrophic and heterotrophic tobacco calli. Both the mas1′ promoter and GST gene promoters contain ocs or ocs‐like elements, responsible for both auxin and ethylene/wound inducibility. The mas1′ promoter exhibited a much higher expression activity in the heterotrophic cultures growing on IAA than in the autotrophic ones, but in contrast with the natural auxin, the mas1′::GUS activity decreased at elevated 2,4‐d concentrations in the heterotrophic tissues and increased with increasing 2,4‐d concentrations in the autotrophic lines. The induction of GST activity by different exogenous auxin concentrations was much higher in the autotrophic lines, especially in the case of 2,4‐d . Higher concentrations of external 2,4‐d resulted in increased ethylene production, which displayed different kinetics in the two types of calli. The ethylene‐inducing 2,4‐d concentrations increased the growth of the heterotrophic, but decreased that of the autotrophic lines. Blocking the ethylene receptors and hence the signal perception by 2,5‐norbornadiene (NBD) in the heterotrophic tissues increased the 2,4‐d ‐induced mas1′ promoter and GST activities, suggesting that the gaseous hormone counteracted the auxin response pathway. This was not found in the autotrophic tissues, where NBD decreased the mas1′‐driven GUS activity. The GST activities were slightly decreased, or almost independent of the action of ethylene. It is suggested that the cross‐talk between the auxin‐ and ethylene‐induced signal transduction pathways may differ in the auxin autotrophic and heterotrophic lines.  相似文献

Nitrifying bacteria, cyanobacteria, and algae are important microorganisms in open pond wastewater treatment systems. Nitrification involving the sequential oxidation of ammonia to nitrite and nitrate, mainly due to autotrophic nitrifying bacteria, is essential to biological nitrogen removal in wastewater and global nitrogen cycling. A continuous flow autotrophic bioreactor was initially designed for nitrifying bacterial growth only. In the presence of cyanobacteria and algae, we monitored both the microbial activity by measuring specific oxygen production rate (SOPR) for microalgae and cyanobacteria and specific oxygen uptake rate (SOUR) for nitrifying bacteria. The growth of cyanobacteria and algae inhibited the maximum nitrification rate by a factor of 4 although the ammonium nitrogen fed to the reactor was almost completely removed. Terminal restriction fragment length polymorphism (T‐RFLP) analysis indicated that the community structures of nitrifying bacteria remained unchanged, containing the dominant Nitrosospira, Nitrospira, and Nitrobacter species. PCR amplification coupled with cloning and sequencing analysis resulted in identifying Chlorella emersonii and an uncultured cyanobacterium as the dominant species in the autotrophic bioreactor. Notwithstanding their fast growth rate and their toxicity to nitrifiers, microalgae and cyanobacteria were more easily lost in effluent than nitrifying bacteria because of their poor settling characteristics. The microorganisms were able to grow together in the bioreactor with constant individual biomass fractions because of the uncoupled solids retention times for algae/cyanobacteria and nitrifiers. The results indicate that compared to conventional wastewater treatment systems, longer solids retention times (e.g., by a factor of 4) should be considered in phototrophic bioreactors for complete nitrification and nitrogen removal. Partial nitrification (PN) has been considered as one of the promising processes for pretreatment of ammonium-rich wastewater. In this study, a kind of novel carriers with enhanced hydrophilicity and electrophilicity was implemented in a moving bed biofilm reactor (MBBR) to start up PN process. Results indicated that biofilm formation rate was higher on modified carriers. In comparison with the reactor filled with traditional carriers (start-up period of 21 days), it took only 14 days to start up PN successfully with ammonia removal efficiency and nitrite accumulation rate of 90 and 91%, respectively, in the reactor filled with modified carriers. Evident changes of spatial distributions and community structures had been detected during the start-up. Free-floating cells existed in planktonic sludge, while these microorganisms trended to form flocs in the biofilm. High-throughput pyrosequencing results indicated that Nitrosomonas was the predominant ammonia-oxidizing bacterium (AOB) in the PN system, while Comamonas might also play a vital role for nitrogen oxidation. Additionally, some other bacteria such as Ferruginibacter, Ottowia, Saprospiraceae, and Rhizobacter were selected to establish stable footholds. This study would be potentially significant for better understanding the microbial features and developing efficient strategies accordingly for MBBR-based PN operation.

Heterotrophic growth of microalgae presents significant economic advantages over the more common autotrophic cultivation. The efficiency of growth and nitrogen, phosphorus, and glucose uptake from synthetic wastewater was compared under heterotrophic, autotrophic, and mixotrophic regimes of Chlorella vulgaris Beij. immobilized in alginate beads, either alone or with the bacterium Azospirillum brasilense. Heterotrophic cultivation of C. vulgaris growing alone was superior to autotrophic cultivation. The added bacteria enhanced growth only under autotrophic and mixotrophic cultivations. Uptake of ammonium by the culture, yield of cells per ammonium unit, and total volumetric productivity of the culture were the highest under heterotrophic conditions when the microalga grew without the bacterium. Uptake of phosphate was higher under autotrophic conditions and similar under the other two regimes. Positive influence of the addition of A. brasilense was found only when light was supplied (autotrophic and mixotrophic), where affinity to phosphate and yield per phosphate unit were the highest under heterotrophic conditions. The pH of the culture was significantly reduced in all regimes where glucose was consumed, similarly in heterotrophic and mixotrophic cultures. It was concluded that the heterotrophic regime, using glucose, is superior to autotrophic and mixotrophic regimes for the uptake of ammonium and phosphate. Addition of A. brasilense positively affects the nutrient uptake only in the two regimes supplied with light.  相似文献

The addition of commercial nitrifying bacterial products has resulted in significant improvement of nitrification efficiency in recirculating aquaculture systems (RAS). We developed two nitrifying bacterial consortia (NBC) from marine and brackish water as start up cultures for immobilizing commercialized nitrifying bioreactors for RAS. In the present study, the community compositions of the NBC were analyzed by universal 16S rRNA gene and bacterial amoA gene sequencing and fluorescence in situ hybridization (FISH). This study demonstrated that both the consortia involved autotrophic nitrifiers, denitrifiers as well as heterotrophs. Abundant taxa of the brackish water heterotrophic bacterial isolates were Paenibacillus and Beijerinckia spp. whereas in the marine consortia they were Flavobacterium, Cytophaga and Gramella species. The bacterial amoA clones were clustered together with high similarity to Nitrosomonas sp. and uncultured beta Proteobacteria. FISH analysis detected ammonia oxidizers belonging to β subclass of proteobacteria and Nitrosospira sp. in both the consortia, and Nitrosococcus mobilis lineage only in the brackish water consortium and the halophilic Nitrosomonas sp. only in the marine consortium. However, nitrite oxidizers, Nitrobacter sp. and phylum Nitrospira were detected in both the consortia. The metabolites from nitrifiers might have been used by heterotrophs as carbon and energy sources making the consortia a stable biofilm.  相似文献

Limitations in available techniques to separate autotrophic (root) and soil heterotrophic respiration have hampered the understanding of forest C cycling. The former is here defined as respiration by roots, their associated mycorrhizal fungi and other micro‐organisms in the rhizosphere directly dependent on labile C compounds leaked from roots. In order to separate the autotrophic and heterotrophic components of soil respiration, all Scots pine trees in 900 m2 plots were girdled to instantaneously terminate the supply of current photosynthates from the tree canopy to roots. Högberg et al. (Nature 411, 789–792, 2001) reported that autotrophic activity contributed up to 56% of total soil respiration during the first summer of this experiment. They also found that mobilization of stored starch (and likely also sugars) in roots after girdling caused an increased apparent heterotrophic respiration on girdled plots. Herein a transient increase in the δ13C of soil CO2 efflux after girdling, thought to be due to decomposition of 13C‐enriched ectomycorrhizal mycelium and root starch and sugar reserves, is reported. In the second year after girdling, when starch reserves of girdled tree roots were exhausted, calculated root respiration increased up to 65% of total soil CO2 efflux. It is suggested that this estimate of its contribution to soil respiration is more precise than the previous based on one year of observation. Heterotrophic respiration declined in response to a 20‐day‐long 6 °C decline in soil temperature during the second summer, whereas root respiration did not decline. This did not support the idea that root respiration should be more sensitive to variations in soil temperature. It is suggested that above‐ground photosynthetic activity and allocation patterns of recent photosynthates to roots should be considered in models of responses of forest C balances to global climate change.  相似文献

Membrane‐aerated biofilm reactors performing autotrophic nitrogen removal can be successfully applied to treat concentrated nitrogen streams. However, their process performance is seriously hampered by the growth of nitrite oxidizing bacteria (NOB). In this work we document how sequential aeration can bring the rapid and long‐term suppression of NOB and the onset of the activity of anaerobic ammonium oxidizing bacteria (AnAOB). Real‐time quantitative polymerase chain reaction analyses confirmed that such shift in performance was mirrored by a change in population densities, with a very drastic reduction of the NOB Nitrospira and Nitrobacter and a 10‐fold increase in AnAOB numbers. The study of biofilm sections with relevant 16S rRNA fluorescent probes revealed strongly stratified biofilm structures fostering aerobic ammonium oxidizing bacteria (AOB) in biofilm areas close to the membrane surface (rich in oxygen) and AnAOB in regions neighbouring the liquid phase. Both communities were separated by a transition region potentially populated by denitrifying heterotrophic bacteria. AOB and AnAOB bacterial groups were more abundant and diverse than NOB, and dominated by the r‐strategists Nitrosomonas europaea and Ca. Brocadia anammoxidans, respectively. Taken together, the present work presents tools to better engineer, monitor and control the microbial communities that support robust, sustainable and efficient nitrogen removal.  相似文献

The marine heterotrophic roseobacter Phaeobacter gallaeciensis DSM 17395 was grown with glucose in defined mineral medium. Relative abundance changes of global protein (2‐D DIGE) and metabolite (GC‐MS) profiles were determined across five different time points of growth. In total, 215 proteins were identified and 147 metabolites detected (101 structurally identified), among which 60 proteins and 87 metabolites displayed changed abundances upon entry into stationary growth phase. Glucose breakdown to pyruvate apparently proceeds via the Entner–Doudoroff (ED) pathway, since phosphofructokinase of the Embden–Meyerhof–Parnas pathway is missing and the key metabolite of the ED‐pathway, 2‐keto‐3‐desoxygluconate, was detected. The absence of pfk in other genome‐sequenced roseobacters suggests that the use of the ED pathway is an important physiological property among these heterotrophic marine bacteria. Upon entry into stationary growth phase (due to glucose starvation), sulfur assimilation (including cysteine biosynthesis) and parts of cell envelope synthesis (e.g. the lipid precursor 1‐monooleoylglycerol) were down‐regulated and cadaverine formation up‐regulated. In contrast, central carbon catabolism remained essentially unchanged, pointing to a metabolic “stand‐by” modus as an ecophysiological adaptation strategy. Stationary phase response of P. gallaeciensis differs markedly from that of standard organisms such as Escherichia coli, as evident e.g. by the absence of an rpoS gene.  相似文献

Despite their role in soil functioning, the ecology of nitrite‐oxidizing bacteria, NOB, and their response to disturbances such as those generated by agricultural practices are scarcely known. Over the course of 17 months, we surveyed the potential nitrite oxidation, PNO, the abundance of the Nitrobacter‐ and Nitrospira‐like NOB (by quantitative PCR) and the community structure of the Nitrobacter‐like NOB (by PCR‐DGGE and cloning‐sequencing targeting the nxrA gene) in soils for four treatments: after establishment of tillage on a previously no‐tillage system, after cessation of tillage on a previously tillage system, and on control tillage and no‐tillage systems. Key soil variables (moisture, organic carbon content and gross mineralization – i.e. ammonification – measured by the 15N dilution technique) were also surveyed. PNO was always higher for the no‐tillage than tillage treatments. Establishment of tillage led to a strong and rapid decrease in PNO whereas cessation of tillage did not change PNO even after 17 months. PNO was strongly and positively correlated to the abundance of Nitrobacter‐like NOB and was also strongly related to gross mineralization, a proxy of N‐availability; in contrast, PNO was weakly and negatively correlated to the abundance of Nitrospira‐like NOB. Selection of a dominant population was observed under no‐tillage, and PNO was loosely correlated to the community structure of Nitrobacter‐like NOB. Our results demonstrate that Nitrobacter‐like NOB are the key functional players within the NOB community in soils with high N availability and high activity level, and that changes in PNO are due to shifts between Nitrospira‐like and Nitrobacter‐like NOB and to a weaker extent by shifts of populations within Nitrobacter‐like NOB.  相似文献

  • 2 The picoplankton was differentiated into autotrophic algae (<1–3μm) and heterotrophic bacteria (<0.2–1 μm) using differential filtration through a 1 μm pore size Nuclepore filter.
  • 3 Efficient separation of these distinct metabolic constituents of picoplankton was obtained. A good correlation (r= 0.81, P < 0.001) was found between physical separation of bacterial and picoalgal cells from fluorescence microscopy and the distribution of heterotrophic metabolic activity between different cell size fractions measured by uptake of 14C-glucose.
  • 4 Picoplankton community production was differentiated into the ‘absolute’ autotrophic production by picoalgae, corrected for overestimation due to retention of bacteria with the picoalgae, and the heterotrophic component, bacterial uptake of ‘extracellular organic carbon’ (EOC), derived from the entire phytoplankton community.
  • 5 The heterotrophic contribution to picoplankton community production ranged from 88 to 1%, mean value 55% of total. Autotrophic picoplankton production was dominant in June and July, but in August and September heterotrophic uptake of EOC was the major input to picoplankton community production.

Summary The kinetics of H2S oxidation in bioreactors with separately packed autotrophic Thiobacillus thioparus CH11 and heterotrophic Pseudomonas putida CH11 were evaluated. The reaction rates were determined to be first-order below 20 ppm, zero-order above 60 ppm, and fractional-order in the intermediate concentration ranges for the Thiobacillus thioparus CH11 bioreactor, and first-order below 35 ppm, zero-order above 80 ppm, and fractional-order in the intermediate concentration ranges for the Pseudomonas putida CH11 bioreactor. The saturation constants for H2S by Thiobacillus thioparus CH11 and Pseudomonas putida CH11 were calculated to be 30.3 ppm and 44.2 ppm, respectively.  相似文献

Relative growth rate, isocitrate lyase activity, chlorophyll, protein, lipid, and soluble carbohydrate contents were investigated in Chlamydomonas humicola Lucksch during auto-, mixo-, and heterotrnphic growth. Mixotrophic cells have a relative growth rate of 1.66 d –1as compared to 0.78 d –1 and 0.21 d –1 for hetero- and autotrophic cells, respectively. Addition of acetate to autotrophic cells resulted in an increase in cell dry weight during the first day, followed by a rapid decrease and stabilization at 40 pg·cell –1. Cellular yield of mixotrophu cells, on a dry weight basis, was 6.6 times that of heterotrophic cells and 21.9 limes that of autotrophic ones. After 4 d, mixotrophic cells were characterized by higher chlorophyll (3.6% dry weight [d.w.]) and protein (58.6% d.w.) contents and lower lipid (4.8% d.w.) and soluble carbohydrate (1.3% d.w.) contents than those of autotrophic (2.6% d.w. chlorophyll, 31.0% d.w. protein, 10.2% d.w. lipid, and 6.5% d.w. soluble carbohydrate) and heterotrophic (1.5% d.w. chlorophyll, 36.9% d.w. protein, 5.6% d.w. lipid, and 6.0% d.w. soluble carbohydrate) cells. The ratio of chlorophyll a/b was highest in heterotrophic cells due to lower chlorophyll b content. Isocitrate lyase activity, a key enzyme in ecetate assimitation, could not be detected in autotrophic cells. Addition of 10 mM acetate to the culture medium of hetero- and mixotrophic cells resulted in increased isocitrate lyase activity with a maximum after 24 h, followed by a decline in activity over a 7-d period. After 7 d of growth, only 0.01 mM acetate was found in the culture medium of mixotrophic cells as compared to 3.2 mM in the medium of heterotrophic ones, from an initial concentration of 10 mM.  相似文献

Nitrous oxide (N2O) was previously deemed as a potent greenhouse gas but is actually an untapped energy source, which can accumulate during the microbial denitrification of nitric oxide (NO). Compared with the organic electron donor required in heterotrophic denitrification, elemental sulfur (S0) is a promising electron donor alternative due to its cheap cost and low biomass yield in sulfur-driven autotrophic denitrification. However, no effort has been made to test N2O recovery from sulfur-driven denitrification of NO so far. Therefore, in this study, batch and continuous experiments were carried out to investigate the NO removal performance and N2O recovery potential via sulfur-driven NO-based denitrification under various Fe(II)EDTA-NO concentrations. Efficient energy recovery was achieved, as up to 35.5%–40.9% of NO was converted to N2O under various NO concentrations. N2O recovery from Fe(II)EDTA-NO could be enhanced by the low bioavailability of sulfur and the acid environment caused by sulfur oxidation. The NO reductase (NOR) and N2O reductase (N2OR) were inhibited distinctively at relatively low NO levels, leading to efficient N2O accumulation, but were suppressed irreversibly at NO level beyond 15 mM in continuous experiments. Such results indicated that the regulation of NO at a relatively low level would benefit the system stability and NO removal capacity during long-term system operation. The continuous operation of the sulfur-driven Fe(II)EDTA-NO-based denitrification reduced the overall microbial diversity but enriched several key microbial community. Thauera, Thermomonas, and Arenimonas that are able to carry out sulfur-driven autotrophic denitrification became the dominant organisms with their relative abundance increased from 25.8% to 68.3%, collectively.  相似文献

Ecosystem respiration (Reco) is one of the largest terrestrial carbon (C) fluxes. The effect of climate change on Reco depends on the responses of its autotrophic and heterotrophic components. How autotrophic and heterotrophic respiration sources respond to climate change is especially important in ecosystems underlain by permafrost. Permafrost ecosystems contain vast stores of soil C (1672 Pg) and are located in northern latitudes where climate change is accelerated. Warming will cause a positive feedback to climate change if heterotrophic respiration increases without corresponding increases in primary production. We quantified the response of autotrophic and heterotrophic respiration to permafrost thaw across the 2008 and 2009 growing seasons. We partitioned Reco using Δ14C and δ13C into four sources–two autotrophic (above – and belowground plant structures) and two heterotrophic (young and old soil). We sampled the Δ14C and δ13C of sources using incubations and the Δ14C and δ13C of Reco using field measurements. We then used a Bayesian mixing model to solve for the most likely contributions of each source to Reco. Autotrophic respiration ranged from 40 to 70% of Reco and was greatest at the height of the growing season. Old soil heterotrophic respiration ranged from 6 to 18% of Reco and was greatest where permafrost thaw was deepest. Overall, growing season fluxes of autotrophic and old soil heterotrophic respiration increased as permafrost thaw deepened. Areas with greater thaw also had the greatest primary production. Warming in permafrost ecosystems therefore leads to increased plant and old soil respiration that is initially compensated by increased net primary productivity. However, barring large shifts in plant community composition, future increases in old soil respiration will likely outpace productivity, resulting in a positive feedback to climate change.  相似文献

冬水田-水稻是川中丘陵区传统的稻田种植模式,冬水田种植模式转变是实现多熟种植及机械化的重要途径。为探究冬水田-水稻种植模式转旱作过程中作物季及休闲期土壤呼吸速率及其组分构成,试验设置冬水田-水稻转旱作(FTD)、冬水田-水稻(FR)和冬闲田-玉米(FM)3种不同种植模式,采用根排除法和静态明箱-气相色谱法原位取样测定作物季及季后休闲期土壤呼吸及其组分,并通过测算净生态系统生产力(NEP)进而判断冬水田-水稻转旱作过程的农田系统碳汇强度。结果表明:(1)FTD显著提高了土壤总呼吸速率及其自养和异养呼吸速率,从而提高了其累积排放量(P<0.05)。与FR相比,FTD的土壤总呼吸及其自养和异养呼吸的累积排放量分别提高了13.14倍、11.32倍和15.56倍(P<0.05);与FM相比,FTD的土壤总呼吸及其自养和异养呼吸的累积排放量分别提高了70.56%、40.83%和115.47%(P<0.05)。(2)与FR和FM相比,FTD均降低了土壤呼吸及其组分的温度敏感性(Q10),且土壤总呼吸的温度敏感性介于异养呼吸和自养呼吸之间。(3)FR,FM和FTD的净生态系统生产力(NEP)均为正值,其数值分别为7911.66 kg/hm2,5667.89 kg/hm2和1583.46 kg/hm2,均表现为大气CO2的碳汇,但与FR与FM相比,FTD显著降低了其净生态系统生产力,呈现出较弱的碳汇。  相似文献

1. Nutrient spiralling provides a conceptual framework and a whole‐system approach to investigate ecosystem responses to environmental changes. We use spiralling metrics to examine how the coupling of nitrogen and phosphorus uptake varies between streams dominated by either heterotrophic (i.e. bacteria‐dominated) or autotrophic (algal‐dominated) microbial communities. 2. Algae generally exhibit greater capacity to store nutrients than bacteria because of differences in cellular structures. These differences led us to hypothesise that the uptake of N and P in heterotrophic ecosystems should have reduced stoichiometric variation in response to changes in supply N : P compared to autotrophic ecosystems when assimilation dominates nutrient uptake. 3. To test this hypothesis, we used an array of serial nutrient additions in several streams in the South Fork Eel River watershed in Northern California. In one set of experiments, N and P were added alone and simultaneously in separate experiments to two small, heterotrophic streams to assess uptake rates and interactions between nutrient cycles. In a second set of experiments, N and P were added simultaneously at a range of N : P in one heterotrophic and one autotrophic stream to assess differences in uptake responses to changes in supply N : P. 4. Results of these experiments suggest two important conclusions. First, increased N supply significantly shortened P uptake lengths, while P addition had little impact on N uptake in both streams, indicating that uptake of non‐limiting nutrients is tightly coupled to the availability of the limiting element. Second, changes in P uptake and uptake ratios (UN : UP) with increased supply N : P supported our hypothesis that heterotrophic streams are more homeostatic in their responses to changes in nutrient supply than autotrophic streams, suggesting that physiological controls on nutrient use scale up to influence ecosystem‐scale patterns in nutrient cycling.  相似文献

Microbial electrosynthesis is a relatively new research field where microbial carbon dioxide fixation based on the energy supplied by a cathode is investigated. Reaction media used in such bioelectrochemical systems have to fulfill requirements of classical biotechnology as well as electrochemistry. The design and characterization of a medium that enables fast electroautotrophic growth of Cupriavidus necator in microbial electrosynthesis was investigated in detail. The identified chloride‐free medium mainly consists of low buffer concentration and is supplied with trace elements. Biotechnologically relevant parameters, such as high‐specific growth rates and short lag phases, were determined for growth characterization. Fast growth under all conditions tested, i.e. heterotrophic, autotrophic and electroautotrophic was achieved. The lag phase was shortened by increasing the FeSO? concentration. Additionally, electrochemical robustness of the reaction media was proven. Under reductive conditions, no deposits on electrodes or precipitations in the media were observed and no detectable hydrogen peroxide evolved. In the bioelectrochemical system, no lag phase occurred and specific growth rate of C. necator was 0.09 h?¹. Using this medium shortens seed train drastically and enables fast electrobiotechnological production processes based on C. necator .  相似文献

1. Five oligotrophic clear‐water lakes on the Faroe Islands were studied during August 2000. Algal and bacterial production rates, community respiration, and CO2 saturation were determined. In addition, we examined the plankton community composition (phytoplankton and heterotrophic nanoflagellates) and measured the grazing pressure exerted by common mixotrophic species on bacteria. 2. High respiration to primary production (6.6–33.2) and supersaturation of CO2 (830–2140 μatm) implied that the lakes were net heterotrophic and that the pelagic heterotrophic plankton were subsidised by allochthonous organic carbon. However, in spite of the apparent high level of net heterotrophy, primary production exceeded bacterial production and the food base for higher trophic levels appeared to be mainly autotrophic. 3. We suggest that the observed net heterotrophy in these lakes was a result of the oligotrophic conditions and hence low primary production in combination with an input of allochthonous C with a relatively high availability. 4. Mixotrophic phytoplankton (Cryptomonas spp., Dinobryon spp. and flagellates cf. Ochromonas spp.) constituted a large percentage of the plankton community (17–83%), possibly as a result of their capacity to exploit bacteria as a means of acquiring nutrients in these nutrient poor systems.  相似文献
