利用SSR分子标记技术分析了佛坪国家级自然保护区秦岭箭竹(Fargesia qinlingensis)的克隆多样性和克隆结构,以探讨小尺度范围内秦岭箭竹自然居群遗传变异的分布特征,对该种开花特性、高山地区生态环境维护和大熊猫的保护提供重要依据。结果表明7对SSR引物共扩增出79个位点,其中多态性位点77个,多态位点百分率(PPB)为97.47%。秦岭箭竹的142个分株共形成107个克隆,最大克隆可达5 m。克隆多样性略高于其他克隆植物的平均值(D=0.62,G/N=0.17,E=0.68),基因型比率(G/N)、Simpson指数(D)、平均克隆大小(N/G)和Fager均匀性指数(E)分别为0.7535、0.9680、1.3271和0.5109。克隆空间结构分析表明秦岭箭竹的克隆构型为密集型,各克隆呈镶嵌性分布,同一克隆的分株排列紧密。克隆聚类分析表明各克隆之间聚类不明显,总体上来自同一样地的克隆被聚为一类。空间自相关分析显示在空间距离为36 m范围内,分株比基株有更显著的空间遗传结构,空间自相关系数r的取值范围分别为0.084—0.626和0.024—0.288,说明克隆繁殖在一定程度上限制了空间遗传结构的范围。样地内秦岭箭竹个体在空间距离小于44 m时存在显著的正相关空间结构,特别是在4 m处表现出最大的空间自相关系数(r=0.626),表明空间距离相距4 m内的个体最有可能属于同一克隆,4 m比5 m更能表现出清晰的克隆结构,X-轴截距为52.280,代表了秦岭箭竹不规则克隆的平均最小长度。秦岭箭竹的克隆多样性和克隆结构与初始苗补充、花粉散播方式和微环境差异有关。  相似文献

很多竹类植物是典型的克隆植物,也是大熊猫的食物。研究典型竹子种群克隆结构的形成和发展对竹林的生产和抚育具有理论和实践意义,可为预测该竹林群落的演替趋势和大熊猫保护提供科学依据。利用SSR标记研究不同年龄A(7龄)、B(30龄)和C(60龄)巴山木竹种群的克隆结构和多样性,探讨小尺度范围内不同年龄巴山木竹种群的克隆结构及斑块的建立和发展。8对SSR引物共扩增出了118个位点,3个种群样地的256个样本共检测到了49个克隆(基因型),A、B和C种群分别检测出31、10个和8个克隆。随着种群年龄的增长,巴山木竹克隆面积增加,克隆数量减少;A和B样地各克隆分布格局为团块状,而C样地克隆既有团块状又有离散状。这一结果显示出在幼苗定居的初期,基株可能以短距离的克隆延伸为主从而呈现出团块状;而随着年龄的增长,克隆面积不断扩大,当复轴混生型的巴山木竹克隆受到强大的压迫时,基株可能会进行较多的单轴和长距离克隆延伸,呈现出离散状。Mantel检测和空间自相关分析都支持3个样地在小尺度范围内存在明显的克隆空间遗传结构。3个样地在10 m等级下显著的正相关空间遗传结构距离为3.1、28、48 m,X-轴截距为9.051、30.698和50.536,空间自相关系数的范围分别为0.1—0.167、0.008—0.703和0.006—0.735。由此可推断,随着年龄的增长,巴山木竹克隆斑块的规模在不断地扩大,同一克隆的分株数量增加,在均匀取样情况下,正相关空间遗传结构距离范围内取到具有相同基因型的可能性越大。A、B和C 3样地的基因型比率(G/N)为1、0.14和0.055,Simpson多样性指数(D)分别为1、0.876和0.744。这说明巴山木竹幼苗期基因型比例远远高于成年的竹林,随着年龄的增长巴山木竹克隆多样性虽有所降低,但由于有性繁殖的作用仍然保持了较高的多样性。聚类和主坐标分析均表明总体上各样地的克隆被聚为一类,但不同样地少数克隆的基因型有重叠和聚集,可推断出不同巴山木竹种群之间可能存在着基因流动和近似的克隆起源。  相似文献

Three different approaches were used to assess the kinship structure of two epiphytic bryophytes, Orthotrichum speciosum and O. obtusifolium, that have different dispersal strategies. The two species were sampled in a 200 ha landscape where species occurrence and host trees had been mapped previously. Local environmental conditions at sampled trees were recorded and kinship between individuals was calculated based on amplified fragment length polymorphism (AFLP)-marker data. We did not detect any association between AFLP-markers and investigated environmental conditions. In both species, significant kinship coefficients were found between individuals up to 300-350 m apart which shows that both species have a restricted dispersal range. The spatial kinship structure was detected with both autocorrelation analysis and generalized additive models (GAMs), but linear regression failed to detect any structure in O. speciosum. Although the dioecious O. obtusifolium is currently the more common species it may, none the less, due to its restricted dispersal range and reproduction mode, become threatened in the future by current silvicultural practices which enhance the distance between host trees and decrease their life span. Finally, GAMs seem most appropriate for analysing spatial genetic structure because the effects of local environmental conditions and spatial structure can be analysed simultaneously, no assumption of a parametric form between kinship coefficient and distance is required, and spatial data resolution is not lost in the arbitrary choice of distance classes characterizing autocorrelation analysis.  相似文献

BACKGROUND AND AIMS: In plant populations the magnitude of spatial genetic structure of apparent individuals (including clonal ramets) can be different from that of sexual individuals (genets). Thus, distinguishing the effects of clonal versus sexual individuals in population genetic analyses could provide important insights for evolutionary biology and conservation. To investigate the effects of clonal spread on the fine-scale spatial genetic structure within plant populations, Hosta jonesii (Liliaceae), an endemic species to Korea, was chosen as a study species. METHODS: Using allozymes as genetic markers, spatial autocorrelation analysis of ramets and of genets was conducted to quantify the spatial scale of clonal spread and genotype distribution in two populations of H. jonesii. KEY RESULTS: Join-count statistics revealed that most clones are significantly aggregated at < 3-m interplant distance. Spatial autocorrelation analysis of all individuals resulted in significantly higher Moran's I values at 0-3-m interplant distance than analyses of population samples in which clones were excluded. However, significant fine-scale genetic structure was still observed when clones were excluded. CONCLUSIONS: These results suggest that clones enhance the magnitude of spatial autocorrelation due to localized clonal spread. The significant fine-scale genetic structure detected in samples excluding clones is consistent with the biological and ecological traits exhibited by H. jonesii including bee pollination and limited seed dispersal. For conservation purposes, genetic diversity would be maximized in local populations of H. jonesii by collecting or preserving individuals that are spaced at least 5 m apart.  相似文献

Clarification of clonal growth pattern is critical for understanding the population dynamics and reproductive system evolution of clonal plant species. The contribution of clonality to the spatial genetic structure (SGS) within populations is also an important issue. I examined the spatial distribution of genetic variability within two populations of the coastal plant Carex kobomugi using seven microsatellite loci. Genotyping of 226 and 140 ramets within 14 × 40 m and 14 × 34 m plots on two populations revealed 36 and 33 multilocus genotypes, respectively. To quantify the extent of intermingling among clones, for each genet, I calculated the dominance of ramets belonging to a particular genet within a spatial range of the genet. Furthermore, I analyzed spatial distribution of genotypes within 2 × 2 m and 1 × 2 m quadrats using second-order spatial statistics. These analyses indicated that clones are highly intermingled, suggesting a low level of spatial interaction among clones. Spatial autocorrelation analysis of kinship coefficient including all pairs of ramets showed significantly stronger SGS than analysis considering only pairs between different genets. I conclude that clonal propagation largely contributes to SGS at a fine scale.  相似文献

Abstract The extent of clonality within populations strongly influences their spatial genetic structure (SGS), yet this is hardly ever thoroughly analysed. We employed spatial autocorrelation analysis to study effects of sexual and clonal reproduction on dispersal of the dioecious seagrass Cymodocea nodosa. Analyses were performed both at genet level (i.e. excluding clonal repeats) and at ramet level. Clonal structure was characterized by the clonal subrange, a spatial measure of the linear limits where clonality still affects SGS. We show that the clonal subrange is equivalent to the distance where the probability of clonal identity approaches zero. This combined approach was applied to two meadows with different levels of disturbance, Cadiz (stable) and Alfacs (disturbed). Genotypic richness, the proportion of the sample representing distinct genotypes, was moderate (0.38 Cadiz, 0.46 Alfacs) mostly due to dominance of a few clones. Expected heterozygosities were comparable to those found in other clonal plants. SGS analyses at the genet level revealed extremely restricted gene dispersal in Cadiz (Sp = 0.052, a statistic reflecting the decrease of pairwise kinship with distance), the strongest SGS found for seagrass species, comparable only to values for selfing herbaceous land plants. At Cadiz the clonal subrange extended across shorter distances (20-25 m) than in Alfacs (30-35 m). Comparisons of sexual and vegetative components of gene dispersal suggest that, as a dispersal vector within meadows, clonal spread is at least as important as sexual reproduction. The restricted dispersal and SGS pattern in both meadows indicates that the species follows a repeated seedling recruitment strategy.  相似文献

The current approaches to the study of clonal plants are reviewed. Most studies concentrate at the level of the ramet and clonal fragment exploring the “microscopic” view of clonal plants, dealing with the translocation of resources, clonal integration, plasticity of growth etc. The information gained, by this approach can be used in the understanding of higher levels of organization within the clonal system either with the help of spatially explicit modelling techniques, or by using means and distributions of size within a population instead of studying individual ramets separately. Plant scientists use the term clone with two meanings, viz. (a) a set of physiologically connected, but potentially independent ramets, and (b) a set of genetically identical, but potentially physically separated individuals. The overlap of these terms differs between individual plant species, depending on the extent of physical separation of the ramets and the degree of physiological integration between the ramets; the lower the frequency of ramet separation, the closer are the physiological and genetic concepts of the clone. Three critical areas seem to be neglected in clonal plant research: (a) the interrelationship between hierarchical levels in clonal plants, (b) the particular spatial structure of their environment, and (c) the importance of clonal plants in different ecological communities.  相似文献

Sex-biased dispersal is expected to generate differences in the fine-scale genetic structure of males and females. Therefore, spatial analyses of multilocus genotypes may offer a powerful approach for detecting sex-biased dispersal in natural populations. However, the effects of sex-biased dispersal on fine-scale genetic structure have not been explored. We used simulations and multilocus spatial autocorrelation analysis to investigate how sex-biased dispersal influences fine-scale genetic structure. We evaluated three statistical tests for detecting sex-biased dispersal: bootstrap confidence intervals about autocorrelation r values and recently developed heterogeneity tests at the distance class and whole correlogram levels. Even modest sex bias in dispersal resulted in significantly different fine-scale spatial autocorrelation patterns between the sexes. This was particularly evident when dispersal was strongly restricted in the less-dispersing sex (mean distance <200 m), when differences between the sexes were readily detected over short distances. All tests had high power to detect sex-biased dispersal with large sample sizes (n ≥ 250). However, there was variation in type I error rates among the tests, for which we offer specific recommendations. We found congruence between simulation predictions and empirical data from the agile antechinus, a species that exhibits male-biased dispersal, confirming the power of individual-based genetic analysis to provide insights into asymmetries in male and female dispersal. Our key recommendations for using multilocus spatial autocorrelation analyses to test for sex-biased dispersal are: (i) maximize sample size, not locus number; (ii) concentrate sampling within the scale of positive structure; (iii) evaluate several distance class sizes; (iv) use appropriate methods when combining data from multiple populations; (v) compare the appropriate groups of individuals.  相似文献

Genetic substructuring in plant populations may evolve as a consequence of sampling events that occur when the population is founded or regenerated, or if gene dispersal by pollen and seeds is restricted within a population. Silene tatarica is an endangered, perennial plant species growing along periodically disturbed riverbanks in northern Finland. We investigated the mechanism behind the microspatial genetic structure of S. tatarica in four subpopulations using amplified fragment length polymorphism markers. Spatial autocorrelation revealed clear spatial genetic structure in each subpopulation, even though the pattern diminished in older subpopulations. Parentage analysis in an isolated island subpopulation indicated a very low level of selfing and avoidance of breeding between close relatives. The mean estimated pollen dispersal distance (24.10 m; SD = 10.5) was significantly longer and the mean seed dispersal distance (9.07 m; SD = 9.23) was considerably shorter than the mean distance between the individuals (19.20 m; SD = 13.80). The estimated indirect and direct estimates of neighbourhood sizes in this subpopulation were very similar, 32.1 and 37.6, respectively. Our results suggested that the local spatial genetic structure in S. tatarica was attributed merely to the isolation-by-distance process rather than founder effect, and despite free pollen movement across population, restricted seed dispersal maintains local genetic structure in this species.  相似文献

BACKGROUND AND AIMS: Many alpine plant species combine clonal and sexual reproduction to minimize the risks of flowering and seed production in high mountain regions. The spatial genetic structure and diversity of these alpine species is strongly affected by different clonal strategies (phalanx or guerrilla) and the proportion of generative and vegetative reproduction. METHODS: The clonal structure of the alpine plant species Salix herbacea was investigated in a 3 x 3 m plot of an alpine meadow using microsatellite (simple sequence repeat; SSR) analysis. The data obtained were compared with the results of a random amplified polymorphic DNA (RAPD) analysis. KEY RESULTS: SSR analysis, based on three loci and 16 alleles, revealed 24 different genotypes and a proportion of distinguishable genotypes of 0.18. Six SSR clones were found consisting of at least five samples, 17 clones consisting of more than two samples and seven single genotypes. Mean clone size comprising at least five samples was 0.96 m(2), and spatial autocorrelation analysis showed strong similarity of samples up to 130 cm. RAPD analysis revealed a higher level of clonal diversity but a comparable number of larger clones and a similar spatial structure. CONCLUSIONS: The spatial genetic structure as well as the occurrence of single genotypes revealed in this study suggests both clonal and sexual propagation and repeated seedling recruitment in established populations of S. herbacea and is thus suggestive of a relaxed phalanx strategy.  相似文献

Spatial structure and fine-scale genetic structure were analyzed for the medicinal plant American ginseng (Panax quinquefolius L.) to more fully understand biological processes within wild populations. P. quinquefolius has been harvested for more than 250 years and is now considered threatened or rare throughout its range. Plants within four protected and four unprotected populations were significantly clumped based on Ripley's univariate analysis. Analysis with Ripley's bivariate test determined that juvenile plants were significantly clumped with adult plants at the shortest distance classes in all populations. Although plants were highly clumped, we found that significant fine-scale genetic structure was restricted to the shortest distance classes based on estimates of coancestry (f(ij)). In most cases, estimates of f(ij) were more significant among juveniles than among adults, especially at the shortest distance classes. The spatial structure of ginseng seems to result from the establishment and persistence of plants in favorable microhabitats coupled with limited seed dispersal around maternal individuals. There were no differences in patterns of fine-scale genetic structure between protected and unprotected populations.  相似文献

采用空间自相关分析方法对浙江天台山亚热带常绿阔叶林优势种甜槠种群全部个体及不同年龄级个体的小尺度空间遗传结构进行了分析,以探讨甜槠种群内遗传变异的分布特征及其形成机制。根据11个ISSR引物所提供的多态位点,经GenAlEx 6软件计算地理坐标和遗传距离矩阵在10个距离等级下的空间自相关系数。在样地内,甜槠种群内个体在空间距离小于10 m时存在显著的正空间遗传结构,其X-轴截矩为9.945。甜槠种群的空间遗传结构与其种子短距离传播和广泛的花粉传播有关。Ⅰ年龄级、Ⅱ年龄级和III年龄级个体在空间距离小于10 m时存在显著的正空间遗传结构,其X-轴截矩分别为11.820、9.746和9.792。当距离等级为5 m时,其空间自相关系数r分别为0.068、0.054和0.070。Ⅳ年龄级个体在所有空间距离等级中均不存在显著的空间遗传结构。甜槠是多年生、长寿命植物,自疏作用是导致IV年龄级个体空间遗传结构消失的主要原因。  相似文献

Higher levels of genetic diversity of river macrophytes are expected in downstream parts because of potential accumulation of various genotypes from upstream sites. We assessed the clonal diversity and spatial genetic structure of fennel pondweed (Potamogeton pectinatus or Stuckenia pectinata) populations with emphasis on the estimation of dispersal via clonal propagules along a river in connection to upstream ponds. We analysed genetic diversity of 354 plant shoots sampled in 2005 and 2006 at three pond and five river sites in the Woluwe river catchment (Belgium). Nine microsatellite DNA markers revealed 88 genets of which 89% occurred in only one site. Clonal propagule dispersal was detected up to 10 km along the river. Few multilocus genotypes were repeatedly present along a major part of the river indicating vegetative spread. Populations of ponds contained a higher amount of clonal diversity, indicating the importance of local seed recruitment. A fine-scaled spatial genetic structure indicated that most seedling recruitment occurred at a distance <5 m in pond populations whereas clones in river sites were unrelated and showed no spatial autocorrelation. The clonal diversity decreased along the river from upstream to downstream due to establishment of few large clones.  相似文献

In previous investigations, natural layering of Japanese stone pine (Pinus pumila) was suggested by the occurrence of adventitious roots. However, there is no genetic evidence so far that this species actually produces offspring by natural layering. We, therefore, investigated clonal structure and spatial genetic structure within a 38×18 m plot on Mt. Aino-dake, using allozyme, random amplified polymorphic DNA (RAPD), and inter-simple sequence repeat (ISSR) analyses. We found 24 genets, with stems found to be genetically identical in multiple tests, which extended later-ally against the direction of the slope, indicating that there were clonal structures originating from elongation of ramified stems and subsequent natural layering. The results suggest, however, that less than one third of the 200 stems analyzed from this site were clonaly propagated. We also analyzed spatial genetic structure by spatial autocorrelation. Many of the spatial autocorrelation coefficients were significantly positive in short distance classes. We concluded that the species has genetic structures which largely originate from clonal propagation and avian seed dispersal.  相似文献

Restricted seed dispersal frequently leads to fine‐scale spatial genetic structure (i.e., FSGS) within plant populations. Depending on its spatial extent and the mobility of pollinators, this inflated kinship at the immediate neighbourhood can critically impoverish pollen quality. Despite the common occurrence of positive FSGS within plant populations, our knowledge regarding the role of long‐distance pollination preventing reproductive failure is still limited. Using microsatellite markers, we examined the existence of positive FSGS in two low‐density populations of the tree Pyrus bourgaeana. We also designed controlled crosses among trees differing in their kinship to investigate the effects of increased local kinship on plant reproduction. We used six pollination treatments and fully monitored fruit production, fruit and seed weight, proportion of mature seeds per fruit, and seed germination. Our results revealed positive FSGS in both study populations and lower fruit initiation in flowers pollinated with pollen from highly‐genetically related individuals within the neighbourhood, with this trend intensifying as the fruit development progressed. Besides, open‐pollinated flowers exhibited lower performance compared to those pollinated by distant pollen donors, suggesting intense qualitative pollen limitation in natural populations. We found positive fine‐scale spatial genetic structure is translated into impoverished pollen quality from nearby pollen donors which negatively impacts the reproductive success of trees in low‐density populations. Under this scenario of intrapopulation genetic rescue by distant pollen donors, the relevance of highly‐mobile pollinators for connecting spatially and genetically distant patches of trees may be crucial to safeguarding population recruitment.  相似文献

Question: Are flower production and associated phenological variables (onset, end, duration, and three measures of flowering synchrony) randomly distributed in space or, alternatively, is there a neighbourhood structure (spatial autocorrelation) in the values of these variables? To which extent does spatial autocorrelation affect the correlation tests between phenological traits? Location: A tree savanna reserve in Southeastern Brazil (22°15′S,47°08′W). Methods: The flowering season of Chromolaena odorata was followed for all (96) individuals in a completely mapped area of 3000 m2. The phenological traits were estimated by counting flower heads in anthesis on individual plants every seven days for 14 weeks. Results: Flowering time was unimodally distributed, but with different peak dates depending on whether individual flower heads or plants were counted. Three phenological traits and canopy closure above the plants showed some degree of spatial autocorrelation, which caused loss of up to 35% of degrees of freedom in nine of 36 correlation tests. Such a decrease in the degrees of freedom resulted in loss of significance for correlations in three pairs of variables. Conclusions: We hypothesize that the spatial autocorrelation in phenological traits between C. odorata neighbours may be driven by genetic similarity among neighbouring plants and/or spatial structuring of environmental factors. Because location and distance between samples may affect their statistical independence, we suggest that spatial autocorrelation should be taken into account in future studies of plant phenology, e.g. by using effective sample size in statistical tests.  相似文献

Conservation of forest genetic resources requires intensive knowledge of the spatial arrangement of genetic diversity. In this study, we used four natural Prunus avium stands in Germany with contrasting for densities to understand patterns of spatial genetic structure. To this end, we genotyped adults and saplings at eight microsatellite markers, 54 AFLP loci and at the gametophytic incompatibility locus. We estimated levels of clonal propagation, spatial genetic structure and gene dispersal. High mortality occurred among young clonal individuals, as depicted by the lower clonal diversity in saplings. Contrasting levels of spatial genetic structure were observed among markers, ontogenic stages and populations. AFLP were more efficient for detecting spatial autocorrelation but did not allow us to differentiate low and high density populations, while high density populations showed substantially stronger spatial genetic structure at microsatellite loci. Furthermore, kinship decreased with tree age only in low density stands. We discuss the present results in terms of population history, pollen and seed dispersal and population density. Although conspecific density seems to be an interesting indicator of genetic diversity for conservation programmes, we still need to disentangle the relative influence of clonal propagation and density on the strength of spatial genetic structure. Simulation studies are needed to further address this question.  相似文献

The information on diversity and spatial distribution of clones of an invasive clonal plant is crucial for the understanding of its clonal structure and invasive history. In this paper, random amplified polymorphic DNA (RAPD) markers were used to explore the clonal diversity and clonal structure of Eichhornia crassipes (Mart.) Solms in natural populations, and their possible effects on the plant success as an invader are also discussed. Five populations covering the entire distribution area in China were studied, sampling 43 individuals per population at an interval of 1 m in a sampling plot. Twelve RAPD primers produced 69 reproducible bands, with 22 being polymorphic. Only five RAPD phenotypes (clones) were detected in these five populations, but each population consisted of at least three clones, contrary to the traditional expectations that E. crassipes populations should be monoclonal. The diversity of clones within populations is thought to be mainly resulted from multiple introductions by humans. The evenness of distribution of clones varied slightly and most clones were widespread, suggesting clonal growth is the predominant mode of regeneration in all the populations. A single clone dominated each population and this clone might be the first one introduced into China or the genotype with a higher phenotypic plasticity, which could survive and reproduce via clonal growth in various habitats. The clones in each population were highly intermixed, especially in river populations, suggesting this species has a guerilla clonal structure which can be facilitated by water current.  相似文献
