棉铃虫Helicoverpa armigera主要借助于性信息素通讯完成雌雄识别,实现交配和种群繁衍。关于棉铃虫感受性信息素机制的研究一直是我国化学生态学领域的热点和重心,研究结果有助于开发和改进棉铃虫防治的性引诱剂。本文将对棉铃虫雄虫感受雌虫释放的性信息素的机制进行综述,以期为深入研究棉铃虫及其他相关昆虫的性信息素感受的分子和神经机理提供参考。棉铃虫雌虫性信息素腺体合成和释放多种长链、饱和或非饱和的脂肪醛和醇等化合物,其中Z11-16:Ald为主要性信息素成分,Z9-16∶Ald和Z9-14∶Ald为次要性信息素成分,不同组分按一定比例混合可明显增强对雄性棉铃虫的引诱效果,而化合物Z11-16∶OH和高剂量的Z9-14∶Ald对性信息素引诱活性具有明显的抑制效果。相应地,雄性棉铃虫触角上A,B和C 3种类型的毛形感器能够感受这些信息化合物。A类型毛形感器内表达受体OR13感受Z11-16∶Ald,B类型毛形感器内表达OR14b感受Z9-14∶Ald,C类型毛形感器内表达OR6和OR16感受Z9-16∶Ald,Z9-14∶Ald,Z11-16:Ac和Z11-16∶OH。受体的表达位置和...  相似文献

本文用单细胞电反应的记录方法,测定了棉铃虫Heliothis armigera Hubner雄蛾触角的毛形感器对其性信息素组分及雌蛾腹部提取物的反应,发现顺-11-十六碳烯醛和顺-9-十六碳烯醛能引起反应。对前者发放大脉冲,对后者发放小脉冲,对雌蛾腹部提取物发放大、小两种脉冲,但以大脉冲为主。  相似文献

【目的】探索斜纹夜蛾Spodoptera litura触角叶结构及其神经元对植物气味和性信息素的神经识别机制。【方法】利用共聚焦激光技术扫描斜纹夜蛾成虫触角叶结构,同时采用多通道电生理技术(multi-unit recording,MR)记录斜纹夜蛾触角叶对6种寄主植物气味化合物(苯甲醛、苯甲醇、苯乙醛、水杨醛、乙酸叶醇酯和己烯醛)及性信息素顺9反11十四碳二烯乙酸酯和顺9反12十四碳二烯乙酸酯的反应;并在风洞中测定分析斜纹夜蛾对上述化合物的定向行为反应。【结果】共聚焦激光扫描结果显示,雄性和雌性斜纹夜蛾触角叶内分别密集地分布有67和66个神经纤维球;而雌性斜纹夜蛾触角叶内的纤维球总体积和平均体积都高于雄性。负责识别和追踪性信息素的扩大型纤维球复合体(macroglomerular complex,MGC)只在雄性斜纹夜蛾触角叶内发现。MR试验结果显示斜纹夜蛾触角叶内神经元具有3种自发放电模式:稀疏放电(不规则的放电频率)、温和放电(宽而慢的放电频率)和密集放电(暴发性的放电频率)。同时,斜纹夜蛾触角叶神经元对所有刺激的气味表现出3种反应类型:兴奋性、抑制性和无反应。神经元对气味的兴奋性和抑制性反应以及无反应取决于刺激化合物的结构和浓度。雌虫的触角叶神经元对性信息素和单一的植物气味表现出很小的反应,而雄虫对两种性信息素以及苯甲醛、苯甲醇、苯乙醛和水杨醛具有很强的兴奋性反应。斜纹夜蛾风洞试验也显示绝大部分的斜纹夜蛾雄虫都选择停留在性信息素和芳香族化合物上,这与MR的结果一致。【结论】神经元的反应强度和刺激化合物浓度之间的关系根据不同的神经元和刺激化合物而有所不同。在测试的浓度范围内,雄虫触角叶神经元对性信息素的反应强度随着浓度的增加而加强,但是除乙酸叶醇酯外,对其他植物气味的反应强度在测试的浓度范围内没有显著的变化。  相似文献

应用压力注射,在Agrotis segetum雄蛾触角叶(AL)中33个对性信息素有反应的MGC神经元上探计了对性信息素反应模式的形成机制,压力注射100mmol/L GABA进入AL神经网引起神经元一个慢的超极化电位,并有一个长时程的放电抑制相,与用性信息素刺激诱导的神经元分应很相似,但GABA并不影响神经元对性信息素刺激的去极化反应,低Cl^-溶液可减弱AL神经元对性信息素刺激的超极化反应,甚至使超极化相逆转为兴奋反应,抑制相消失。压力注射Bicuculline使神经元放电频率增加。压力注射Bicuculline的同时给予性信息素刺激,可使性信息素刺激所致的神经元放电增加进一步加强;Bicuculline可使性信息素刺激引起的神经元超极化幅度变小,放电抑制时间变短,甚至其抑制相完全被逆转为正常放电,无超极化反应和抑制相存在,结果表明,AL神经元对性信息系反应的超极化相与GABA受体有关。  相似文献

In moths, sex pheromone components are detected by pheromone-specific olfactory receptor neurons (ph-ORNs) housed in sensilla trichodea in the male antennae. In Grapholita molesta, ph-ORNs are highly sensitive and specific to the individual sex pheromone components, and thus help in the detection and discrimination of the unique conspecific pheromone blend. Plant odors interspersed with a sub-optimal pheromone dose are reported to increase male moth attraction. To determine if the behavioral synergism of pheromone and plant odors starts at the ph-ORN level, single sensillum recordings were performed on Z8-12:Ac and E8-12:Ac ph-ORNs (Z-ORNs and E-ORNs, respectively) stimulated with pheromone–plant volatile mixtures. First, biologically meaningful plant-volatile doses were determined by recording the response of plant-specific ORNs housed in sensilla auricillica and trichodea to several plant odorants. This exploration provided a first glance at plant ORNs in this species. Then, using these plant volatile doses, we found that the spontaneous activity of ph-ORNs was not affected by the stimulation with plant volatiles, but that a binary mixture of sex pheromone and plant odorants resulted in a small (about 15%), dose-independent, but statistically significant, reduction in the spike frequency of Z-ORNs with respect to stimulation with Z8-12:Ac alone. The response of E-ORNs to a combination of E8-12:Ac and plant volatiles was not different from E8-12:Ac alone. We argue that the small inhibition of Z-ORNs caused by physiologically realistic plant volatile doses is probably not fully responsible for the observed behavioral synergism of pheromone and plant odors.  相似文献

We used single-sensillum recordings to characterize male Heliothis subflexa antennal olfactory receptor neuron physiology in response to compounds related to their sex pheromone. The recordings were then followed by cobalt staining in order to trace the neurons' axons to their glomerular destinations in the antennal lobe. Receptor neurons responding to the major pheromone component, (Z)-11-hexadecenal, in the first type of sensillum, type-A, projected axons to the cumulus of the macroglomerular complex (MGC). In approximately 40% of the type-A sensilla, a colocalized receptor neuron was stained that projected consistently to the posterior complex 1 (PCx1), a specific glomerulus in an 8-glomerulus complex that we call the Posterior Complex (PCx). We found that receptor neurons residing in type-B sensilla and responding to a secondary pheromone component, (Z)-9-hexadecenal, send their axons to the dorsal medial glomerulus of the MGC. As in the type-A sensilla, we found a cocompartmentalized neuron within type-B sensilla that sends its axon to a different glomerulus of the PCx4. One neuron in type-C sensilla tuned to a third pheromone component, (Z)-11-hexadecenol, and a colocalized neuron responding to (Z)-11-hexadecenyl acetate projected their axons to the anteromedial and ventromedial glomeruli of the MGC, respectively.  相似文献

【目的】围食膜(peritrophic membrane, PM)是昆虫抵御随食物摄入的病原微生物入侵的第一道天然屏障。本研究旨在鉴定出农业重大害虫棉铃虫Helicoverpa armigera围食膜的总蛋白成分,为进一步揭示昆虫围食膜的形成机制及研发新颖的害虫控制策略奠定基础。【方法】剥离棉铃虫5龄幼虫PM,用三氟甲磺酸(trifluoromethane sulfonic acid, TFMS)处理,采用液质联用技术(LC-MS/MS)鉴定围食膜蛋白质组,然后对鉴定结果进行生物信息学分析。【结果】本研究共鉴定出棉铃虫幼虫围食膜蛋白质169个,是目前鉴定最多的棉铃虫围食膜蛋白。通过GO分析,可以将这些鉴定的蛋白分为细胞组分、分子功能和生物学过程三大类;KEGG富集结果显示,鉴定蛋白可以富集在12条代谢通路中;蛋白互作分析(protein protein interaction, PPI)结果表明,以ACC和CG3011等蛋白为核心可以形成蛋白互作网络。【结论】本研究鉴定了169个棉铃虫幼虫围食膜蛋白质,并对其进行了GO, KEGG和PPI分析,结果有助于人们全面理解昆虫围食膜的分子结构和功能。  相似文献

【目的】本研究旨在探究拟除虫菊酯类杀虫剂对棉铃虫Helicoverpa armigera幼虫肠道菌群结构及代谢的影响,丰富对杀虫剂作用机理的认识。【方法】分别对棉铃虫2和3龄幼虫饲喂普通人工饲料(对照组, SS)、含2%七氟菊酯(Ⅰ型拟除虫菊酯)粉剂饲料(七氟菊酯处理组,Te)和含2.5%溴氰菊酯(Ⅱ型拟除虫菊酯)乳油饲料(溴氰菊酯处理组, DM),然后提取3龄幼虫肠道菌群基因组DNA;利用Illumina MiSeq二代高通量测序技术对肠道细菌的16S rDNA的V3-V4变异区进行测序,分析其肠道细菌的多样性和丰富度;利用qPCR验证16S rDNA测序分析结果。取2和3龄幼虫肠道,匀浆后进行Biolog-Eco实验,分析肠道细菌对Eco板上31种碳源的代谢情况。【结果】16S rDNA测序结果表明,棉铃虫3龄幼虫肠道细菌主要是厚壁菌门(Firmicutes)、变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)和蓝藻菌门(Cyanobacteria)。与对照组相比,溴氰菊酯处理组和七氟菊酯处理组的棉铃虫幼虫肠道细菌的α多样性指数没有显著性改变,但是菌群结构发生了变化:在门水平,拟杆菌门的相对丰度减少,厚壁菌门和蓝藻菌门的相对丰度增加,qPCR验证结果亦支持16S rDNA测序分析的这个结果;在属水平,拟杆菌属Bacteroides、普氏菌属Prevotella和假单胞菌属Pseudomonas等的相对丰度降低,狭义梭菌属Clostridium sensu stricto 1、埃希菌属-志贺氏菌属Escherichia-Shigella和盐单胞菌属Halomonas等的相对丰度增加,其中盐单胞菌属Halomonas的相对丰度显著增加。Biolog-Eco结果表明,与对照组相比,溴氰菊酯处理组中2龄幼虫对羧酸类碳源的代谢能力下降;溴氰菊酯处理组和七氟菊酯处理组中3龄幼虫对DL-α-磷酸甘油、肝糖和L-苯丙氨酸等碳源的利用能力下降。【结论】结果显示,拟除虫菊酯类杀虫剂对棉铃虫肠道菌群的结构和代谢能力有明显影响,拟除虫菊酯类杀虫剂使棉铃虫肠道有益菌的相对丰度下降,而使致病菌的相对丰度增加。短时间拟除虫菊酯处理未造成抗药性菌群的丰度增加。qPCR检测结果与16S rDNA测序分析结果相似。Ⅰ型和Ⅱ型拟除虫菊酯类杀虫剂对棉铃虫肠道菌群结构和代谢功能的影响不同。  相似文献

棉铃虫酪氨酸羟化酶基因的分子特性及功能分析   总被引:1,自引:0,他引:1  
【目的】酪氨酸羟化酶(tyrosine hydroxylase, TH)是黑色素形成的关键酶,在昆虫表皮骨化过程中扮演重要角色。本研究旨在获得棉铃虫Helicoverpa armigera TH基因序列,并研究其分子特性、表达模式和功能,为更深入探析该基因作用机理奠定基础。【方法】通过生物信息学和分子生物学技术获得了棉铃虫TH基因序列,利用qRT-PCR分析该基因在棉铃虫不同生长发育阶段的表达模式;利用qRT-PCR技术,分别测定了蜕皮激素20E(400 ng/头)处理不同时间和RNAi成功干扰蜕皮激素受体基因(EcR)前提下再用20E(400 ng/头)处理后,棉铃虫5龄幼虫TH表达量变化;采用生物化学方法检测鞣化激素(30 μg/mg组织)和环腺苷酸(cAMP, 200 ng/mg 组织)处理后棉铃虫幼虫脂肪体中TH活性。【结果】获得了棉铃虫酪氨酸羟化酶基因TH (GenBank登录号: MF440319) cDNA片段,长2 270 bp,开放阅读框1 686 bp,编码561个氨基酸残基。该基因在棉铃虫整个发育期均表达,其中在卵期第3天、2龄幼虫第1天、3-5龄蜕皮期、预蛹期和成虫羽化第1天表达量相对较高。研究还发现,400 ng/头 20E注射能够促进TH的转录;在成功干扰并调低幼虫EcR转录水平的前提条件下(对照仅注射dsGFP)再注射20E,对TH表达量无明显影响;而鞣化激素(30 μg/mg组织)和cAMP(200 ng/mg组织)均显著提高了TH的酶活性。【结论】20E在转录水平参与了TH的表达;鞣化激素和cAMP均能够提高TH活性,在蛋白水平上对TH进行调控。  相似文献

Phytophagous insects detect volatile compounds produced by host and non-host plants, using species-specific sets of olfactory receptor neurons (ORNs). To investigate the relationship between the range of host plants and the profile of ORNs, single sensillum recordings were carried out to identify ORNs and corresponding active compounds in female Uraba lugens (Lepidoptera: Nolidae), an oligophagous eucalypt feeder. Based on the response profiles to 39 plant volatile compounds, 13 classes of sensilla containing 40 classes of ORNs were identified in female U. lugens. More than 95% (163 out of 171) of these sensilla contained 16 classes of ORNs with narrow response spectra, and 62.6% (107 out of 171) 18 classes of ORNs with broad response spectra. Among the specialized ORNs, seven classes of ORNs exhibited high specificity to 1,8-cineole, (±)-citronellal, myrcene, (±)-linalool and (E)-β-caryophyllene, major volatiles produced by eucalypts, while nine other classes of ORNs showed highly specialized responses to green leaf volatiles, germacrene D, (E)-β-farnesene and geranyl acetate that are not produced by most eucalypts. We hypothesize that female U. lugens can recognize their host plants by detecting key host volatile compounds, using a set of ORNs tuned to host volatiles, and discriminate them from non-host plants using another set of ORNs specialized for non-host volatiles. The ORNs with broad response spectra may enhance the discrimination between host and non-host plants by adding moderately selective sensitivity. Based on our finding, it is suggested that phytophagous insects use the combinational input from both host-specific and non-host specific ORNs for locating their host plants, and the electrophysiological characterization of ORN profiles would be useful in predicting the range of host plants in phytophagous insects.  相似文献

棉铃虫Helicoverpa armigera和烟青虫H. assulta属于可同域发生的近缘种昆虫,通过产生比例相反的两种性信息素化合物——顺9-十六碳烯醛和顺11-十六碳烯醛维持种间生殖隔离。本研究应用外源不饱和脂肪醇及乙酸酯在棉铃虫和烟青虫性信息素腺体进行在体转化,利用气相色谱法分析转化产物,从酶学角度探讨了上述两近缘种昆虫性信息素腺体组分差异的形成原因。实验结果表明,两种昆虫信息素腺体表皮伯醇氧化酶对外源顺9-十六碳烯醇、顺11-十六碳烯醇和反10-十六碳烯醇无催化专一性,说明末端氧化过程对于醛类性信息素组分特定比例的形成不起作用。棉铃虫性信息素腺体组织具有较高的乙酸酯酶活性,可水解外源乙酸酯,但烟青虫性信息素腺体乙酸酯酶活性很低。这些发现对于进一步了解两种昆虫的生殖隔离机制有重要参考价值。  相似文献

The neurophysiology and antennal lobe projections of olfactory receptor neurons housed within short trichoid sensilla of female Heliothis virescens F. (Noctuidae: Lepidoptera) were investigated using a combination of cut-sensillum recording and cobalt-lysine staining techniques. Behaviorally relevant odorants, including intra- and inter-sexual pheromonal compounds, plant and floral volatiles were selected for testing sensillar responses. A total of 184 sensilla were categorized into 25 possible sensillar types based on odor responses and sensitivity. Sensilla exhibited both narrow (responding to few odors) and broad (responding to many odors) response spectra. Sixty-six percent of the sensilla identified were stimulated by conspecific odors; in particular, major components of the male H. virescens hairpencil pheromone (hexadecanyl acetate and octadecanyl acetate) and a minor component of the female sex pheromone, (Z)-9-tetradecenal. Following characterization of the responses, olfactory receptor neurons within individual sensilla were stained with cobalt lysine (N=39) and traced to individual glomeruli in the antennal lobe. Olfactory receptor neurons with specific responses to (Z)-9-tetradecenal, a female H. virescens sex pheromone component, projected to the female-specific central large female glomerulus (cLFG) and other glomeruli. Terminal arborizations from sensillar types containing olfactory receptor neurons sensitive to male hairpencil components and plant volatiles were also localized to distinct glomerular locations. This information provides insight into the representation of behaviorally relevant odorants in the female moth olfactory system. 害虫发生高峰期、 发生量的准确预测和田间防治适期的确定与种群雌虫卵巢结构及卵子发生过程密切相关。为了明确棉铃虫Helicoverpa armigera卵巢结构及卵子发生过程, 本研究利用光学体视显微镜和透射电子显微镜, 对棉铃虫成虫卵巢管和卵子的超微结构进行了研究, 并确定了发育级别划分标准。结果表明: 根据卵巢的形状、 卵的产生过程、 卵黄沉积情况等将棉铃虫卵巢发育程度分为6个级别, 即发育初期(0级)、 卵黄沉积前期(Ⅰ级)、 卵黄沉积期(Ⅱ级)、 成熟待产期(Ⅲ级)、 产卵盛期(Ⅳ级)和产卵末期(Ⅴ级)。根据卵子发生过程中滋养细胞、 卵母细胞的变化, 将卵子发生期分为3个阶段: 卵黄发生前期、 卵黄发生期和卵黄成熟期。本研究首次对棉铃虫的卵子发生进行电子显微观察, 并完善了棉铃虫卵巢发育的分级标准, 为进一步研究棉铃虫的生殖发育机理提供了理论参考, 对田间棉铃虫种群发生期和发生量的预测预报也有重要的实践参考价值。  相似文献
