Studies on the Active Transport of Calcium in Human Red Cells   总被引:7,自引:3,他引:4  
The Ca++ transport mechanism in the red cell membrane was studied in resealed ghost cells. It was found that the red cell membrane can transport Ca++ from inside the cell into the medium against great concentration gradient ratios. Tracing the movement of 45Ca infused inside red cells indicated that over 95% of all Ca++ in the cells was transported into media in 20 min incubation under the optimum experimental conditions. The influence of temperature on the rate constant of transport indicated an activation energy of 13,500 cal per mole. The optimum pH range of media for the transport was between 7.5 and 8.5. As energy sources, ATP1, CTP, and UTP were about equally effective, GTP somewhat less effective, and ITP least effective among the nucleotides tested. The Ca++ transport does not appear to involve exchange of Ca++ with any monovalent or divalent cations. Also, it is not influenced by oligomycin, sodium azide, or ouabain in high concentrations, which inhibit the Ca++ transport in mitochondria or in sarcoplasmic reticulum. In these respects, the Ca++ transport mechanism in the red cell membrane is different from those of mitochondria and the sarcoplasmic reticulum.  相似文献

Sarcoplasmic reticulum fragments (S.R.F.) were isolated from skeletal and heart muscles. These fragments were found to take up Ca++ very actively from media. When monophasic square waves were passed through the S.R.F. suspension, the Ca++ uptake by S.R.F. was decreased. When the suspension was stimulated electrically after the Ca++ was taken up by S.R.F., the initiation and the cessation of the stimulation were followed by the release and re-uptake of Ca++ by S.R.F., respectively. The degree of inhibition of the Ca++ uptake as well as of the Ca++ release by electrical stimulation was dependent on the voltage and the frequency of stimulation. The presence of inorganic phosphate or oxalate modified the influence of electrical stimulation on the release and the uptake of Ca++ by S.R.F. Attempts were made to observe the release of Ca++ by electrical stimulation from unfractionated sarcoplasmic reticulum remaining in myofibers, and the interaction of the released Ca++ with myofibrils in vitro. For this purpose, the glycerol-extracted fiber was selected as a muscle model, since it contains both sarcoplasmic reticulum and myofibrils. It was found that electrical stimulation of skeletal and heart glycerol-extracted fibers resulted in the contraction of fibers. It appeared that the contraction of glycerol fibers by electrical stimulation was caused by the Ca++ release from sarcoplasmic reticulum by stimulation.  相似文献

The effects of epinephrine and caffeine on isometric rabbit papillary muscle preparations were observed after maximum peak tension was produced by adjustment of initial length and Ca++ concentration. Without change in peak tension, epinephrine caused increased positive dP/dt (p < .005), increased negative dP/dt (p < .001) and decreased time to peak tension (TP) (p < .001), while caffeine resulted in decreased positive dP/dt (p < .02), decreased negative dP/dt (p < .005) and prolonged TP (p < .001). Caffeine added to muscles being perfused with epinephrine reversed the epinephrine effects. The data suggest that epinephrine increases the influx of Ca++ into the cell and the uptake of Ca++ by the sarcoplasmic reticulum, and that caffeine exerts opposite effects on Ca++ exchange. However, an additional mechanism not dependent on changes in Ca++ flux is suggested. Epinephrine and caffeine may directly effect the rates of actin-myosin interaction, the former agent increasing, the latter decreasing the rates of attachment and detachment of actin-myosin bridges.  相似文献

A study of the intracellular transport of calcium in rat heart   总被引:4,自引:0,他引:4  
The distribution of in vivo injected 45Ca++ in the subcellular fractions of rat heart has been studied. Most of the radioactivity of the cell was found to be associated with the subcellular organelles; only a small fraction was recovered in the soluble phase. Mitochondria contained the greatest part of the total radioactivity associated with the subcellular organelles. After injection of 45Ca++ the specific activity of the mitochondrial calcium pool was several times higher than that of the calcium of the sarcoplasmic reticulum. Pentachlorophenol has been administered to rats to uncouple oxidative phosphorylation in heart mitochondria in vivo and its effect on the distribution of 45Ca++ in the heart studied. Under these conditions, it has been found that mitochondria contained much less 45Ca++ than the controls; this decrease was paralleled by an increase of the radioactivity associated with the microsomes and with the final supernatant. Experiments in which 45Ca++ was added to heart homogenates at 0° indicated that 45Ca++ also became bound to mitochondria and the other subcellular structures at 0°. However, PCP had no effect on the distribution of radioactivity among the subcellular fractions under these conditions. The results suggest that (1) energy-linked movements of Ca++ take place in mitochondria of the intact rat heart, (2) a part of the uptake of 45Ca++ by mitochondria does not depend on metabolism, and, (3) the movements of Ca++ in heart mitochondria in vivo are probably more active than those in the sarcoplasmic reticulum.  相似文献

The capacities and affinities of intracellular calcium-binding sites have been studied in frog ventricles, in which the concentration of Ca++ in the sarcoplasm can be controlled as a result of treatment with EDTA. The total calcium content of calcium-depleted and nondepleted muscles at rest and muscles generating considerable tension was 0.8, 1.4, and 5.4 µmol/g of muscle, respectively. Net movement of calcium into or out of the cells occurred without change in tension when the sarcoplasmic concentration of Ca++ was either of two values, less than 10-7 M or approximately 5 x 10-7 M. These data can be explained by the presence of two groups of intracellular calcium sinks which compete with the contractile proteins, one with a capacity of about 0.6 µmol/g and an affinity constant greater than 107 M-1 and a second with a capacity of 4.0 µmol/g and an affinity constant of about 2 x 106 M-1. The higher affinity calcium is released by anoxia, oligomycin, or abrupt changes in sarcoplasmic Ca++. Muscles soaked in Sr-Ringer's contain electron densities in the sarcoplasmic reticulum and to a lesser extent in the mitochondria.  相似文献

Ruthenium red, a powerful inhibitor of Ca2+ transport by mitochondria, does not inhibit the active Ca2+ uptake by sarcoplasmic reticulum isolated from rabbit skeletal muscle promoted by 5 mM ATP-Mg in the presence or absence of potassium oxalate. Although concentrations of ruthenium red up to 100 μM do not affect the active uptake of Ca2+, 25 μM of the inorganic dye inhibit the passive binding of Ca2+ by about 50%. This inhibitory effect is observed in sarcoplasmic reticulum even after its lipid fraction is extracted with acetone.Although active Ca2+ uptake by sarcoplasmic reticulum is not inhibited by ruthenium red, in the absence of oxalate it inhibits significantly the Ca2+-dependent ATPase activity but not the Mg2+-ATPase. However, if potassium oxalate is present, the Ca2+-stimulated ATPase is not sensitive to the dye. It is not clear how oxalate functions to protect the Ca2+-ATPase against the inhibitor effect of ruthenium red.The high sensitivity to ruthenium red of the Ca2+ transport mechanism in mitochondria as compared to the Ca2+ transport in sarcoplasmic reticulum may be useful in determining the extent to which each organelle functions in the cell to regulate intracellular free Ca2+.  相似文献

Initial velocities of energy-dependent Ca++ uptake were measured by stopped-flow and dual-wavelength techniques in mitochondria isolated from hearts of rats, guinea pigs, squirrels, pigeons, and frogs. The rate of Ca++ uptake by rat heart mitochondria was 0.05 nmol/mg/s at 5 µM Ca++ and increased sigmoidally to 8 nmol/mg/s at 200 µM Ca++. A Hill plot of the data yields a straight line with slope n of 2, indicating a cooperativity for Ca++ transport in cardiac mitochondria. Comparable rates of Ca++ uptake and sigmoidal plots were obtained with mitochondria from other mammalian hearts. On the other hand, the rates of Ca++ uptake by frog heart mitochondria were higher at any Ca++ concentrations. The half-maximal rate of Ca++ transport was observed at 30, 60, 72, 87, 92 µM Ca++ for cardiac mitochondria from frog, squirrel, pigeon, guinea pig, and rat, respectively. The sigmoidicity and the high apparent Km render mitochondrial Ca++ uptake slow below 10 µM. At these concentrations the rate of Ca++ uptake by cardiac mitochondria in vitro and the amount of mitochondria present in the heart are not consistent with the amount of Ca++ to be sequestered in vivo during heart relaxation. Therefore, it appears that, at least in mammalian hearts, the energy-linked transport of Ca++ by mitochondria is inadequate for regulating the beat-to-beat Ca++ cycle. The results obtained and the proposed cooperativity for mitochondrial Ca++ uptake are discussed in terms of physiological regulation of intracellular Ca++ homeostasis in cardiac cells.  相似文献

Comparative studies were carried out on the Ca2+-transport systems of mitochondria and sarcoplasmic reticulum from longissimus dorsi muscle of genetically selected malignant hyperthermia-prone and normal pigs in order to identify the biochemical lesion responsible for the enhanced release of Ca2+ in the sarcoplasm occurring in porcine malignant hyperthermia. Mitochondria isolated from longissimus dorsi muscle of malignant hyperthermia-prone pigs contained a significantly (P < 0.001) higher amount of endogenous long-chain fatty acids. Similar amounts of endogenous mitochondrial phospholipase A2 were observed in both types of pigs, but the total activity in malignant hyperthermia-prone pigs was at least twice that of normal. Spermine, a phospholipase A2 inhibitor, lowered the activity in both types of mitochondria to a similar final level. Mitochondria of malignant hyperthermia-prone pigs showed a significantly (P < 0.001) higher oligomycin-insensitive (Ca2+ + Mg2+)-ATPase activity, but the Mg2+-ATPase and the (Ca2+ + Mg2+)-ATPase activities were similar in both types of pigs. Sarcoplasmic reticulum isolated from longissimus dorsi muscle of malignant hyperthermia-prone pigs showed a significantly higher (Ca2+ + Mg2+)-ATPase activity and a lower rate of Ca2+ uptake; the maximal amount and the rate of Ca2+ uptake by sarcoplasmic reticulum of malignant hyperthermia-prone pigs were half that of normal. Mitochondria from longissimus dorsi muscle of malignant hyperthermia-prone pigs inhibited the Ca2+-transport system of the sarcoplasmic reticulum of longissimus dorsi from both normal and malignant hyperthermia-prone pigs, but mitochondria from normal pigs had no influence on the sarcoplasmic reticulum from either type. Experimental evidence favours the concept that long-chain fatty acids released from skeletal muscle mitochondria by endogenous mitochondrial phospholipase A2 are responsible for the enhanced release of Ca2+ from mitochondria (Cheah, K.S. and Cheah, A.M. (1981) Biochim. Biophys. Acta 634, 70–84), and also additional release of Ca2+ from sarcoplasmic reticulum into the sarcoplasm during porcine malignant hyperthermia syndrome.  相似文献

Quinidine potentiates twitch tension and (at higher concentrations) causes contracture of skeletal muscle whereas the same drug reduces tension development of cardiac muscle. To gain insight into the possible differences in the excitation-contraction coupling mechanism of the two types of muscle the effect of quinidine on calcium accumulation by isolated sarcoplasmic reticulum from skeletal and cardiac muscle was investigated. In a medium containing ATP, Mg++, oxalate, and 45Ca, pharmacologically active concentrations of the drug inhibited calcium accumulation by both skeletal and cardiac sarcoplasmic reticulum. The inhibition of the rates of calcium, uptake by the skeletal muscle preparation ranged from 11% with 10-4 M quinidine to 90% with 10-3 M quinidine. With the cardiac muscle preparation the inhibition ranged from 16% with 3 x 10-6 M quinidine to 100% with 10-3 M quinidine. With both preparations the inhibition of calcium transport was accompanied by an inhibition of the Ca++-activated ATPase activity of the sarcoplasmic reticulum. The effect of quinidine on the skeletal sarcoplasmic reticulum supports the hypothesis that this compound produces twitch potentiation and contracture by interfering with intracellular calcium, sequestration. Its effect on cardiac sarcoplasmic reticulum. has been interpreted in terms of the hypothesis that cardiac contractility is a function of the amount of calcium released from the sarcoplasmic reticulum which is in turn dependent upon the absolute calcium content of the reticulum. Hence, following inhibition of calcium transport there would be less calcium available for coupling.  相似文献

Summary Secretagogues of pancreatic enzyme secretion, the hormones pancreozymin, carbamylcholine, gastrin I, the octapeptide of pancreozymin, and caerulein as well as the Ca++-ionophore A 23187 stimulate45Ca efflux from isolated pancreatic cells. The nonsecretagogic hormones adrenaline, isoproterenol, secretin, as well as dibutyryl cyclic adenosine 3,5-monophosphate and dibutyryl cyclic guanosine 3,5-monophosphate have no effect on45Ca efflux. Atropine blocks the stimulatory effect of carbamylcholine on45Ca efflux completely, but not that of pancreozymin. A graphical analysis of the Ca++ efflux curves reveals at least three phases: a first phase, probably derived from Ca++ bound to the plasma membrane; a second phase, possibly representing Ca++ efflux from cytosol of the cells; and a third phase, probably from mitochondria or other cellular particles. The Ca++ efflux of all phases is stimulated by pancreozymin and carbamylcholine. Ca++ efflux is not significantly effected by the presence or absence of Ca++ in the incubation medium. Metabolic inhibitors of ATP production, Antimycin A and dinitrophenol, which inhibit Ca++ uptake into mitochondria, stimulate Ca++ efflux from the isolated cells remarkably, but inhibit the slow phase of Ca++ influx, indicating the role of mitochondria as an intracellular Ca++ compartment. Measurements of the45Ca++ influx at different Ca++ concentrations in the medium reveal saturation type kinetics, which are compatible with a carrier or channel model. The hormones mentioned above stimulate the rate of Ca++ translocation.The data suggest that secretagogues of pancreatic enzyme secretion act by increasing the rate of Ca++ transport most likely at the level of the cell membrane and that Ca++ exchange diffusion does not contribute to the45Ca++ fluxes.With the technical assistance of C. A fluorescent chelate probe and a Millipore filtration technique have been used to study the effects of β-bungarotoxin (β-toxin) on passive and active Ca++ uptake and ATPase in fragmented sarcoplasmic reticulum (SR) of rabbit skeletal muscle. β-Toxin at 3 × 10?6 M did not affect ATPase activity. In the absence of ATP, β-Toxin increased the passive uptake of Ca++; in the presence of ATP, active Ca++ uptake was inhibited. The effect of β-toxin in SR can be detected at concentrations as low as 10?9 M. The results suggest that β-toxin induces Ca++ leakage in SR membranes.  相似文献

The relative importance of heart mitochondria in regulating intracellular [Ca2+] in cardiac muscle is controversial. In a new approach to the question, we have measured the energy-linked 45Ca uptake of an unusual myocardial tissue preparation in which the cells appear to be intact yet the sarcolemmae are highly permeable to exogenous solutes. Inhibitors of mitochondrial energy metabolism were used to estimate the mitochondrial contribution to rate and extent of total cell uptake. At 6.6μM Ca, which is close to the probable intracellular [Ca] range, inhibitors of mitochondrial energy metabolism did not diminish initial rates of 45Ca uptake by myocardial fragments, if ATP was present to drive Ca2+ sequestration by the sarcoplasmic reticulum. The ultimate extent of uptake was reduced somewhat, however. Similar uptake profiles were obtained in the presence of carbonyl cyanide m-chlorophenyl-hydrazone, CN?, and atractyloside, each of which acts at a different locus to inhibit mitochondrial Ca2+ transport. These data suggest that the mitochondria cannot control beat-to-beat [Ca2+] oscillations, because at μM Ca concentrations, the Ca2+ uptake rate of mitochondria insitu is slow in comparison to the extra-mitochondrial (sarcoplasmic reticulum) uptake rate.  相似文献

The fluorescent chelate probe technique is employed to observe the accumulation and binding of Ca++ to isolated sarcoplasmic reticulum from skeletal and cardiac muscle. Chlorotetracycline serves as a fluorescent chelate probe which chelates to membrane bound Ca++ giving rise to an intensely fluorescence adduct. An increase in fluorescence of chlorotetracycline is caused by ATP induced Ca++ transport in both skeletal and cardiac muscle microsomes. The fluorescence spectra indicate that Ca++ lies on the membrane surface in a relatively polar environment.  相似文献

Ca2+ transport by sarcoplasmic reticulum vesicles was examined by incubating sarcoplasmic reticulum vesicles (0.15 mg/ml) at 37°C in, either normal medium that contained 0.15 M sucrose, 0.1 M KCl, 60 μM CaCl2, 2.5 mM ATP and 30 mM Tes at pH 6.8, or a modified medium for elimination of ADP formed from ATP hydrolysis by including, in addition, 3.6 mM phosphocreatine and 33 U/ml of creatine phosphokinase. In normal medium, Ca2+ uptake of sarcoplasmic reticulum vesicles reached a plateau of about 100 nmol/mg. In modified medium, after this phase of Ca2+ uptake, a second phase of Ca2+ accumulation was initiated and reached a plateau of about 300 nmol/mg. The second phase of Ca2+ accumulation was accompanied by phosphate uptake and could be inhibited by ADP. Since, under these experimental conditions, there was no significant difference of the rates of ATP hydrolysis in normal medium and modified medium, extra Ca2+ uptake in modified medium but not in normal medium could not be explained by different phosphate accumulation in the two media. Unidirectional Ca2+ influx of sarcoplasmic reticulum near steady state of Ca2+ uptake was measured by pulse labeling with 45Ca2+. The Ca2+ efflux rate was then determined by subtracting the net uptake from the influx rate. At the first plateau of Ca2+ uptake in normal medium, Ca2+ influx was balanced by Ca2+ efflux with an exchange rate of 240 nmol/mg per min. This exchange rate was maintained relatively constant at the plateau phase. In modified medium, the Ca2+ exchange rate at the first plateau of Ca2+ uptake was about half of that in normal medium. When the second phase of Ca2+ uptake was initiated, both the influx and efflux rates started to increase and reached a similar exchange rate as observed in normal medium. Also, during the second phase of Ca2+ uptake, the difference between the influx and efflux rates continued to increase until the second plateau phase was approached. In conditions where the formation of ADP and inorganic phosphate was minimized by using a low concentration of sarcoplasmic (7.5 μg/ml) and/or using acetyl phosphate instead of ATP, the second phase of Ca2+ uptake was also observed. These data suggest that the Ca2+ load attained by sarcoplasmic reticulum vesicles during active transport is modulated by ADP accumulated from ATP hydrolysis. ADP probably exerts its effect by facilitating Ca2+ efflux, which subsequently stimulates Ca2+ exchange.  相似文献

Sheep or guinea pig antisera against the purified Ca++ transport ATPase of sarcoplasmic reticulum inhibit Ca++ transport due to a complement-dependent damage of the membrane, which causes massive leakage of Ca++. The Ca++-activated ATPase activity is only slightly affected even at ten times higher antibody concentration than that required for inhibition of Ca++ transport. Antibodies prepared against the Ca++ binding protein (C1 protein) have no influence upon either ATPase activity or Ca++ transport and ferritin-labeled anti-C1 antibodies do not bind to microsomes.  相似文献

Endoplasmic reticulum membranes were isolated from roots of garden cress (Lepidium sativum L. cv Krause) using differential and discontinuous sucrose gradient centrifugation. The endoplasmic reticulum fraction was 80% rough endoplasmic reticulum oriented with the cytoplasmic surface directed outward and contaminated with 12% unidentified smooth membranes and 8% mitochondria. Marker enzyme analysis showed that the activity for endoplasmic reticulum was enriched 2.4-fold over total membrane activity while no other organelle activity showed an enrichment. All evidence indicated that the fraction was composed of highly enriched endoplasmic reticulum membranes. Ca2+ uptake activity was measured using the filter technique described by Gross and Marmé (1978). The results of these experiments showed an ATP-dependent, oxalate-stimulated Ca2+ uptake into vesicles of the endoplasmic reticulum fraction. The majority of the transport activity was microsomal since specific inhibitors of mitochondrial Ca2+ transport (ruthenium red, LaCl3 and oligomycin) inhibited the activity by only 25%. Sodium azide showed no inhibition. The transport was likely directly coupled to ATP hydrolysis since there was no inhibition with carbonylcyanidem-chlorophenylhydrazone. The transport activity was specific for ATP showing only 36% and 29% of the activity with inosine diphosphate and guanosine 5′-triphosphate, respectively. The results indicate a Ca2+ transport function located on the endoplasmic reciculum of garden cress roots.  相似文献

The Ca++ ionophore A23187 had no effect on the release of amylase by mouse pancreas fragments in the absence of Ca++ but when Ca++ was re-added to the medium amylase release was observed in a pattern which mimicked that produced by normal stimulants. Uptake of 45Ca++ by pancreatic fragments was increased by A23187. Tetracaine and dinitrophenol at concentrations which block cholinergic stimulated enzyme release blocked ionophore induced release whereas atropine did not. None of the inhibitors studied affected the ionophore induced Ca++ uptake.  相似文献

The effects of spegazzinine, a dihydroindole alkaloid, on mitochondrial oxidative phosphorylation were studied.Spegazzinine inhibited coupled respiration and phosphorylation in rat liver mitochondria. The I50 was 120 μM. Uncouplers released the inhibition of coupled respiration. Arsenate-stimulated mitochondrial respiration was partially inhibited by spegazzinine. The stimulation of mitochondrial respiration by Ca2+ and the proton ejection associated with the ATP-dependent Ca2+ uptake were not affected by the alkaloid.Oxidative phosphorylation and the Pi-ATP exchange reaction of phosphorylating beef heart submitochondrial particles were strongly inhibited by spegazzinine (I50, 50 μM) while the ATP-dependent reactions, reduction of NAD+ by succinate and the pyridine nucleotides transhydrogenase were less sensitive (I50, 125 μM). Oxygen uptake by submitochondrial particles was not affected.The 2,4-dinitrophenol-stimulated ATPase activity of rat liver mitochondria was not affected by 300 μM spegazzinine, a concentration of alkaloid that completely inhibited phosphorylation. However, higher concentrations of spegazzinine did partially inhibit it. The ATPase activities of submitochondrial particles, insoluble and soluble ATPases were also partially inhibited by high concentrations of spegazzinine.The inhibitory properties of spegazzinine on energy transfer reactions are compared with those of oligomycin, aurovertin and dicyclohexylcarbodiimide. It is concluded that spegazzinine effects are very similar to the effects of aurovertin and that its site of action may be the same or near the site of aurovertin.  相似文献
