Quantitative data on modes of transmission are a crucial element in understanding the ecology of microorganisms associated with animals. We investigated the transmission patterns of a gamma-proteobacterium informally known as pea aphid Bemisia-like symbiont (PABS), also known as T-type, which is widely but not universally distributed in natural populations of the pea aphid, Acyrthosiphon pisum. The vertical transmission of PABS to asexual and sexual morphs and sexually produced eggs was demonstrated by a diagnostic PCR-based assay, and the maximum estimated failure rate was 2%. Aphids naturally lacking PABS acquired PABS bacteria administered via the diet, and the infection persisted by vertical transmission for at least three aphid generations. PABS was also detected in two of five aphid honeydew samples tested and in all five siphuncular fluid samples tested but in none of 15 samples of salivary secretions from PABS-positive aphids. However, PABS-negative aphids did not acquire PABS when they were cocultured with PABS-positive aphids; the maximal estimated level of horizontal transmission was 18%. A deterministic model indicated that the force of infection by a horizontal transmission rate of 3% is sufficient to maintain a previously described estimate of the prevalence of PABS-positive aphids (37%), if the vertical transmission rate is 98%. We concluded that PABS infections in A. pisum can be maintained by high vertical transmission rates and occasional horizontal transmission, possibly via the oral route, in the absence of selection either for or against aphids bearing this bacterium.  相似文献

Abstract.  1. Pea aphids, Acyrthosiphon pisum , harbour a range of facultative accessory bacteria (secondary symbionts), including those informally known as PASS (R-type), PAR, PABS (T-type), and PAUS (U-type).
2. To explore the relationship between possession of these bacteria and ecologically important traits of A. pisum , correlations between the accessory bacteria found in 47 parthenogenetic clones of A. pisum and the host plant on which each clone was collected and its susceptibility to natural enemies were surveyed.
3. The bacterial complement varied with plant of collection. PAUS (U) was present in all of 12 clones affiliated to Trifolium but was otherwise rare, while PABS (T) and PASS (R) occurred at significantly higher frequency in clones from Lotus and Vicia , respectively, than clones from other plants.
4. Possession of PABS (T) was associated strongly with resistance to the parasitoid Aphidius eadyi and weakly with resistance to Aphidius ervi . Aphids carrying PAUS (U) were more resistant to the fungal pathogen Pandora ( Erynia ) neoaphidis , although this correlation was complicated by a strong association with host-plant use.  相似文献

Abstract:  In order to establish the host range of the pea aphid subspecies, Acyrthosiphon pisum ssp. destructor , and hence from which plant species pea crops are likely to become infested, the performance of this aphid on different leguminous plants was assessed. The plant species used were: Lotus uliginosus , Medicago sativa , Melilotus officinalis , Ononis repens , Sarothamnus scoparius , Trifolium hybridum , Trifolium pratense , Trifolium repens , Vicia cracca and Vicia faba . Vicia faba and Trifolium hybridum were the plants on which aphids reached the greatest size, took the least time to reach maturity, and experienced the lowest mortality. The time taken for the aphids to develop to maturity was negatively correlated with adult size, whereas survival to maturity was positively correlated with adult size. The host preference of the aphids was also assessed. The plant species selected as hosts by alatae were those on which their offspring performed best.  相似文献

Abstract.  1. The parasitoid Aphidius ervi and the entomopathogenic fungus Pandora neoaphidis both require successful invasion of an aphid host to complete their life cycle. A shorter developmental period allows P. neoaphidis to out-compete A. ervi. Aphidius ervi may reduce this fitness cost by avoiding aphid colonies containing P. neoaphidis . Here the response of A. ervi towards P. neoaphidis was assessed using sequential experiments designed to replicate different stages of parasitoid foraging behaviour.
2. Entry rate experiments showed that A. ervi entered aphid colonies containing P. neoaphidis -sporulating cadavers and that there was no significant difference in the attraction of A. ervi to aphid-damaged Vicia faba plants containing either healthy Acyrthosiphon pisum or P. neoaphidis -sporulating cadavers.
3. Observational behavioural experiments indicated that the presence of P. neoaphidis did not affect the search time or total foraging time of A. ervi on V. faba plants infested with either healthy A. pisum or P. neoaphidis -sporulating cadavers.
4. In Petri dish bioassays using aphids infected with P. neoaphidis over a period of 120 h, A. ervi showed no difference in attack rate against uninfected aphids or living aphids infected with P. neoaphidis for 1, 24, 48, 72, or 96 h. However, sporulating cadavers (120 h infection) were not attacked.
5.  Aphidius ervi appears only able to detect the presence of P. neoaphidis once the host is dead and sporulation has started. The fitness of A. ervi may therefore be severely reduced when foraging in P. neoaphidis -infected aphid colonies.  相似文献

Bacterial 16S rDNA amplified by PCR from the pea aphid Acyrthosiphon pisum included a sequence with >98% similarity to secondary symbionts in the whitefly Bemisia tabaci. The 'pea aphid Bemisia-like bacterium' (PABS) and B. tabaci secondary symbionts are estimated to have diverged 17-34 million years ago, a time considerably more recent than the common ancestor of aphids and whitefly and suggestive of horizontal transmission of this bacterial lineage. PABS was scored in both the gut and ovaries of aphids by PCR and identified as a small rod by in situ hybridisation. PABS was not universal in pea aphids: 2/3 laboratory strains and 13/35 of field aphids were PABS-positive. It is suggested that the incidence of PABS in pea aphids is determined by the balance between loss (processes may include occasional failure of vertical transmission and selection against PABS-positive aphids) and horizontal transfer between insects.  相似文献

Plant viruses modify the development of their aphid vectors by inducing physiological changes in the shared host plant. The performance of hymenopterous parasitoids exploiting these aphids can also be modified by the presence of the plant pathogen. We used laboratory and glasshouse microcosms containing beans (Vicia faba) as the host plant to examine the interactions between a plant virus (pea enation mosaic virus; PEMV) and a hymenopterous parasitoid (Aphidius ervi) that share the aphid vector/host Acyrthosiphon pisum. Neither PEMV-infection of V. faba, nor the carriage of PEMV virions by A. pisum, affected the growth or morphology of the aphid, or the oviposition behaviour and development of A. ervi. The presence of developing Aphidius ervi larvae within Acyrthosiphon pisum did not affect the ability of the aphids to transmit PEMV. However, by reducing their longevity, parasitism ultimately decreased the time viruliferous aphids were able to inoculate plants. In terms of virus dispersal, parasitized aphids exhibited more movement around experimental arenas than unparasitized controls, causing a slight increase in the proportion of beans infected with PEMV. Exposure to adult Aphidius ervi caused Acyrthosiphon pisum to rapidly drop off bean plants and disperse to new hosts, resulting in considerably higher plant infection rates (70%) than that seen in control arenas (25%). The results of this investigation demonstrate that when parasitoids are added to a plant-pathogen-vector system, benefits to the host plant due to reduced herbivore infestation must be balanced against the consequences of parasitoid-induced aphid dispersal and a subsequent increase in the level of plant infection.  相似文献

The pea aphid, Acyrthosiphon pisum, shows significant reproductive isolation and host plant specialization between populations on alfalfa and clover in New York. We examine whether specialization is seen in pea aphids in California, and whether fitness on alternative host plants is associated with the presence of bacterial symbionts. We measured the fitness of alfalfa- and clover-derived aphids on both types of plants and found no evidence for specialization when all aphid lineages were considered simultaneously. We then screened all aphids for the presence of four facultative bacterial symbionts: PAR, PASS, PABS and PAUS. Aphids with PAUS were host-plant specialized, having twice as many offspring as other aphids on clover, and dying on alfalfa. Other aphids showed no evidence of specialization. Additionally, aphids with PABS had 50% more offspring than aphids with PASS when on alfalfa. Thus, specialist and generalist aphid lineages coexist, and specialization is symbiont associated. Further work will resolve whether PAUS is directly responsible for this variation in fitness or whether PAUS is incidentally associated with host-plant specialized aphid lineages.  相似文献

The bacterial communities of aphids were investigated by terminal restriction fragment length polymorphism and denaturing gradient gel electrophoresis analysis of 16S rRNA gene fragments generated by PCR with general eubacterial primers. By both methods, the gamma-proteobacterium Buchnera was detected in laboratory cultures of six parthenogenetic lines of the pea aphid Acyrthosiphon pisum and one line of the black bean aphid Aphis fabae, and one or more of four previously described bacterial taxa were also detected in all aphid lines except one of A. pisum. These latter bacteria, collectively known as secondary symbionts or accessory bacteria, comprised three taxa of gamma-proteobacteria (R-type [PASS], T-type [PABS], and U-type [PAUS]) and a rickettsia (S-type [PAR]). Complementary analysis of aphids from natural populations of four aphid species (A. pisum [n = 74], Amphorophora rubi [n = 109], Aphis sarothamni [n = 42], and Microlophium carnosum [n = 101]) from a single geographical location revealed Buchnera and up to three taxa of accessory bacteria, but no other bacterial taxa, in each aphid. The prevalence of accessory bacterial taxa varied significantly among aphid species but not with the sampling month (between June and August 2000). These results indicate that the accessory bacterial taxa are distributed across multiple aphid species, although with variable prevalence, and that laboratory culture does not generally result in a shift in the bacterial community in aphids. Both the transmission patterns of the accessory bacteria between individual aphids and their impact on aphid fitness are suggested to influence the prevalence of accessory bacterial taxa in natural aphid populations.  相似文献

Habitat choice plays a critical role in the processes of host range evolution, specialization, and ecological speciation. Pea aphid, Acyrthosiphon pisum, populations from alfalfa and red clover in eastern North America are known to be genetically differentiated and show genetic preferences for the appropriate host plant. This species feeds on many more hosts, and here we report a study of the genetic variation in host plant preference within and between pea aphid populations collected from eight genera of host plants in southeastern England. Most host-associated populations show a strong, genetically based preference for the host plant from which they were collected. Only in one case (populations from Vicia and Trifolium) was there little difference in the plant preference spectrum between populations. All populations showed a significant secondary preference for the plant on which all the aphid lines were reared: broad bean, Vicia faba, previously suggested to be a "universal host" for pea aphids. Of the total genetic variance in host preference within our sample, 61% could be attributed to preference for the collection host plant and a further 9% to systematic differences in secondary preferences with the residual representing within-population genetic variation between clones. We discuss how a combination of host plant preference and mating on the host plant may promote local adaptation and possibly ecological speciation, and whether a widely accepted host could oppose speciation by mediating gene flow between different populations.  相似文献

We investigated the compatibility of host plant resistance to the black bean aphid in the faba bean crop with the use of the predatory ladybird beetle for biological control under laboratory and greenhouse conditions. Greenhouse experiments indicated that apteriform Aphis fabae reproduced on Vicia faba major (susceptible) and on 79S4 (partially resistant) cultivars at different rates. During the entire experimental period, aphids built up higher populations on V. faba major than on 79S4 cultivar. Aphid numbers on 79S4 were about 37% of those on V. faba major after 14 days. Release of a newly hatched Coccinella septempunctata larva onto each plant significantly reduced aphid density to 32.8% and 57.2% on V. faba major and 79S4 on day 14, respectively. Partial resistance combined with predation was more effective in lessening aphid numbers on faba bean than either the predator or the plant resistance alone. Laboratory tests showed that prey, A.   fabae , raised on susceptible cultivar was more suitable for the predator as food source, enhancing the development rate and fecundity than aphids fed on the partially resistant cultivar. Consumption of aphids reared on susceptible cultivar significantly increased the female fecundity and fertility of C. septempunctata by 37.7% and 33.2%, respectively, more than those fed with aphids from partially resistant cultivar. Pre-oviposition time was shortened by 4.5 days, and oviposition period was extended by 11.4 days. Feeding the predator on aphids from the partially resistant cultivar prolonged the embryonic larval developmental time and the time required from egg laying to adult emergence by 19.8, 10.1 and 32.5 h, respectively. Adult longevity was not influenced by the aphid source. The results are discussed in relation to the compatible utilisation of host plant resistance and biological control in the integrated management of aphids.  相似文献

Phytophagous insects frequently use multiple host-plant species leading to the evolution of specialized host-adapted populations and sometimes eventually to speciation. Some insects are confronted with a large number of host-plant species, which may provide complex routes of gene flow between host-adapted populations. The pea aphid (Acyrthosiphon pisum) attacks a broad range of plants in the Fabaceae and it is known that populations on Trifolium pratense and Medicago sativa can be highly specialized at exploiting these species. To find out whether adaptation to a broad range of co-occurring hosts has occurred, we tested the performance of pea aphid clones collected from eight host-plant genera on all of these plants in a reciprocal transfer experiment. We provide evidence for pervasive host-plant specialization. The high performance of all aphid clones on Vicia faba suggests that this host plant could be a site of gene flow between different populations that could limit further host-associated divergence. The genetic variance in host-plant usage was partitioned into within- and among-population components, which represent different levels of host adaptation. Little evidence of within-population trade-offs in performance on different plant species was found.  相似文献

Plant penetration by the stylets of six clones of the pea aphid, Acyrthosiphon pisum, on Vicia faba (acceptable to all clones) and Pisum sativum (acceptable to 3/6 clones) was investigated by the DC electrical penetration graph technique. In a 10 h recording period, 93% of 144 aphids exhibited sustained feeding on phloem sap. Significant interclonal differences were observed for the incidence of potential drops (indicative of brief punctures of plant cells) and the duration of waveform E1 (insect salivation into a sieve element). In addition, the total duration of the sieve element phase and the duration of completed bouts of sustained feeding differed between the two test plants, in a fashion varying between clones. However, these differences could not be related to the acceptability of plants to the different aphid clones. The duration of the stylet pathway phase preceding the first sustained feeding on phloem sap did not vary significantly with either aphid clone or plant. It is concluded that the resistance of P. sativum to certain A. pisum clones does not arise from factors impeding either stylet penetration through the plant tissues or the maintenance of feeding from the sieve elements. It is proposed that host plant affiliation of A. pisum may be mediated primarily by specific olfactory or gustatory cues, before the aphid initiates stylet penetration of the plant.  相似文献

Abstract.  1. To clarify the use of honeydew in foraging for aphids by larvae of the ladybird beetle, Coccinella septempunctata L., searching behaviour of ladybird larvae for Aphis craccivora Koch and Acyrthosiphon pisum Harris and the abundance of honeydew under aphid colonies were examined in laboratory experiments.
2. More larvae responded by climbing the plants with aphids than responded to plants without aphids. When the plants were replaced with sticks, in order to exclude visual and olfactory cues from plants and aphids, more larvae of C. septempunctata climbed sticks above the area that contained honeydew than climbed sticks above the area that did not contain honeydew. Then, ladybird larvae use honeydew as a contact kairomone when foraging for aphids.
3.  Aphis craccivora deposited a larger number of honeydew droplets beneath the plants than did similar numbers of A. pisum. Thus, C. septempunctata larvae licked more frequently the honeydew of A. craccivora than that of A. pisum and spent longer searching on the area containing honeydew of A. craccivora than that of A. pisum . Consequently, a larger number of larvae climbed a stick above honeydew of A. craccivora than that of A. pisum.
4. It may be also considered that C. septempunctata larvae can distinguish honeydew of the two aphid species and respond more strongly to A. craccivora than A. pisum.  相似文献

Many theoretical and empirical studies have shown that species diversity in a trophic level can impact the capture of limited resources in ways that cascade up or down a food web. Only recently, however, have ecologists begun to consider how diversity at multiple trophic levels might act in concert to have opposing or reinforcing effects on resource use. Here, we report the results of an empirical study of a model, tritrophic food web in which we manipulated the diversity of host plant species ( Medicago sativa , Trifolium pratense and Vicia faba ) and natural enemy species ( Harmonia axyridis , Coleomegilla maculata and Nabis sp.) of a widespread herbivorous pest (the pea aphid, Acyrthosiphon pisum ) in laboratory microcosms. We found that increasing natural enemy richness from one to three species increased the proportion of aphids consumed by 0.14. This effect of enemy diversity was due to facilitative interactions and/or a reduction in intraspecific competition in the more diverse assemblages. We also found an independent and additive main effect of host plant richness, with the proportion of aphids consumed by natural enemies decreasing by 0.14 in plant polycultures. A reduction in predator efficiency on a single host plant, Vicia faba , appeared to be responsible for this plant diversity effect. Aphid population sizes were, therefore, simultaneously determined by a top-down effect of natural enemy diversity, and an opposing bottom-up effect of host plant diversity that modified enemy–prey interactions. These results suggest that population sizes in nature, and biotic controls over insect pests, are influenced by species diversity at multiple trophic levels.  相似文献

Abstract.  1. In ecological speciation , adaptation to variation in the external environment provides the crucial push that starts the process of genetic divergence and eventually leads to speciation. This emphasis on the role of ecological specialisation in speciation events has brought with it a renewed interest in its proximate mechanisms in recently diverged groups such as host races. Here, the proximate mechanisms of feeding specialisation are investigated in two host races of the pea aphid Acyrthosiphon pisum .
2. Using alfalfa and clover extracts, enclosed in diet chambers or applied on whole plants, it is shown that feeding specialisation depends on recognition of stimulants specific to the host plant, not on deterrents or toxins specific to the non-host plants.
3. Because pea aphids mate on their host plant, feeding specialisation leads to de facto assortative mating. This study suggests that behavioural recognition of host-specific chemicals, rather than avoidance of deterrents or/and plant toxins, contributes to gene flow restriction between the alfalfa and clover host races.  相似文献

Abstract 1. Motivated by a community study on aphids and their fungal pathogens, three hypotheses were tested experimentally to investigate the influence of the fungal pathogen, Erynia neoaphidis Remaudière and Hennebert, on aphid population and community ecology.
2. Field experiments were performed in 2 years to test whether two susceptible aphid species on different host plants might interact through the shared fungal pathogen. No strong pathogen-mediated indirect interactions (apparent competition) between populations of pea aphid Acyrthosiphon pisum Harris and nettle aphid Microlophium carnosum Buckton were detected.
3. In the first of the field experiments, pea aphids exposed to the fungus showed a weak tendency to produce more winged dispersal morphs than control populations not exposed to the fungus. In a laboratory test, however, no support was found for the hypothesis that the presence of volatiles from fungus-infected cadavers promotes production of winged offspring.
4. The response of the pea aphid parasitoid Aphidius ervi Halliday to colonies containing hosts infected 1 and 3 days previously was assessed. Wasps initiated fewer attacks on 1-day-old infected colonies than on healthy colonies, with the numbers on 3-day-old fungus-infected colonies intermediate.  相似文献

Sympatric populations of insects adapted to different host plants are good model systems not only to study how they adapt to the chemistry of their food plant, but also to investigate whether morphological modifications evolved enabling them to live successfully on a certain plant species. 2. The pea aphid, Acyrthosiphon pisum (Harris) encompasses at least 11 genetically distinct sympatric host races, each showing a preference for a certain legume species. The leaflet surfaces of these legumes differ considerably in their wax coverage. 3. It was investigated whether the attachment structures of three pea aphid genotypes from different host races are adapted to the different surface properties of their host plants and whether they show differences in their attachment ability on the respective host and non‐host plants. 4. The surface morphology of plants and aphid tarsi was examined using SEM (scanning electron microscopy). The ability of the aphids to walk on specific surfaces was tested using traction force measurements. 5. The presence of wax blooms on the leaflets lowers the aphids' attachment ability considerably and diminishes their subsequent attachment on ‘neutral’ surfaces like glass. The pea aphid host races differ in their ability to walk on certain surfaces. However, the genotype from the adapted aphid host race was not necessarily the one with the best walking performance on their host plant. All aphids, regardless of the original host plant, were most efficient on the neutral control surface glass. The general host plant Vicia faba was the plant with the most favourable surface for all aphid host races.  相似文献

Aphidius ervi and Aphidius eadyi, two parasitoids of the pea aphid Acyrthosiphon pisum, were attracted to components of the aphid sex pheromone in laboratory bioassays. Pre-test experience with host aphids in the presence of aphid sex pheromone did not affect the response of A. ervi to pheromone in a 4-way olfactometer, compared with that of naive parasitoids. Aphidius ervi females exposed only to the pheromone prior to testing did not respond in the olfactometer, suggesting habituation to the foraging cue by the parasitoid. In a wind tunnel, aphid sex pheromone increased the attraction of A. ervi to the plant-host complex (Vicia faba/A. pisum), suggesting an additive effect when two different foraging cues are present simultaneously.  相似文献

We used a model plant-aphid system to investigate whether the aphid-specific entomopathogenic fungus Pandora neoaphidis responds to aphid-induced defence by the broad-bean plant, Vicia faba. Laboratory experiments indicated that neither in vivo sporulation, conidia size nor the in vitro growth of P. neoaphidis was affected by Acyrthosiphon pisum-induced V. faba volatiles. The proportion of conidia germinating on A. pisum feeding on previously damaged plants was significantly greater than on aphids feeding on undamaged plants, suggesting a direct functional effect of the plant volatiles on the fungus. However, there were no significant differences in the infectivity of P. neoaphidis towards A. pisum feeding on either undamaged V. faba plants or plants previously infested with A. pisum. Therefore, these results provide no evidence to suggest that P. neoaphidis contributes to plant indirect defence strategies.  相似文献
