具抗菌活性放线菌菌株JSM 20.1的分离和鉴定   总被引:1,自引:1,他引:0  
从药用植物银杏(Ginkgo biloba)根际土壤中分离到1株放线菌菌株JSM20.1,其发酵产物具有广谱抗菌活性。该菌株在多数培养基上生长良好,具有链霉菌属的典型形态特征。菌丝多分枝,不断裂;基内菌丝黄白色至棕褐色,气生菌丝黄白色至棕红色;孢子链直且长,偶有波曲,孢子柱状、光滑、不运动。在察氏、马铃薯浸汁、葡萄糖-天门冬酰胺、酵母膏-麦芽膏(ISP2)、燕麦(ISP3)、无机盐淀粉(ISP4)和甘油-天门冬酰胺(ISP5)琼脂上均产生可溶性色素,而在营养琼脂上不产生可溶性色素。生长温度范围为7~40℃,最适生长温度28℃;生长pH范围为5.0~10.0,最适pH7.0;能在含0~3%(质量与体积比)NaCl的培养基上生长,而在无NaCl的ISP2培养基生长最佳。根据其形态学特征、生理生化特征、细胞化学分类特征和基于16SrRNA基因序列的系统发育分析结果,菌株JSM20.1被鉴定为链霉菌属的有效发表种Streptomyces cyaneofuscatus的1个菌株。  相似文献

2株具抗菌活性的稀有放线菌的筛选和鉴定   总被引:1,自引:0,他引:1  
从药用植物根际土壤样品中分离得到了一批放线菌菌株,通过对其进行了抗茵活性筛选,发现菌株45725具有较好的抗耻垢分枝杆菌活性,菌株06-2230具有较强的抗绿脓杆菌活性。菌株45725和06-2230在酵母-麦芽膏琼脂(ISP2)、燕麦琼脂(ISP3)、无机盐淀粉琼脂培养基(ISP4)、葡萄糖天冬素琼脂培养基(ISP5)和马铃薯培养基(PDA)上生长良好,基生菌丝丰富,无气生菌丝。2株菌的最适生长温度为28℃,最适生长pH值7.0~7.5。综合2株菌的形态学、生理生化、细胞化学分类特征和基于16SrRNA基因序列的系统发育分析和DNA—DNA杂交结果,菌株45725和06—2230分别是多形态放线菌属的2个不同种。  相似文献

海洋放线菌ACMA006的发酵产物具有很强的抗肿瘤活性和较好的抑菌效果, 但对人正常细胞毒性较弱。该菌株在大多数培养基上生长良好, 产生可溶性色素;基质菌丝在培养早期出现横隔, 随后发生断裂;最适生长温度为28°C。基于16S rDNA序列的系统发育分析表明, 菌株ACMA006属于链霉菌属(Streptomyces)的成员, 与该属的合格发表种卡伍尔链霉菌华盛顿亚种(S.cavourensis subsp. washingtonensis)的16S rDNA序列相似性最高, 同源性达100%, 但在部分形态特征和理化特性上存在较大差异。初步确定ACMA006属于卡伍尔链霉菌华盛顿亚种的一个海洋变种。  相似文献

从福建东海海滩土样品中分离到一株海洋放线菌FIM02-523, 该菌株的发酵产物具有抗肿瘤活性。菌株FIM02-523在多数培养基上生长良好, 橙色-暗棕色, 无气生菌丝, 不产生可溶性色素。系统发育、化学分类特征、形态特征、生理生化特性等分析表明菌株FIM02-523是小单孢菌属(Micromonospora), 可能是模式菌种青铜小单孢菌(Micromonospora chalcea)的一个菌株。  相似文献

具抗肿瘤活性放线菌菌株YIM 90022的分离和系统发育分析   总被引:10,自引:3,他引:7  
从青海盐碱土壤样品中分离到一株兼性嗜碱放线菌YIM 90022,该菌株的发酵产物具有很强的体外抗胃癌、肺癌、乳腺癌、皮肤癌、肾癌和子宫癌肿瘤细胞株活性。基于16S rRNA基因序列的系统发育分析表明,菌株YIM90022属于拟诺卡氏属(Nocardiopsis)的成员,与该属的4个有效发表种N.exhalansDSM 44407T,N.prasinaDSM43845T,N.metallicusDSM 44598T和N.listeriDSM 40297T系统发育关系最密切,与其分别以98.8%,98.5%,98.4%和97.8%的16S rRNA基因核苷酸序列相似性聚为一簇。但菌株YIM 90022不与这4个有效种中任何一个单独相聚,形成了一个独立亚分枝。结合形态特征、生理生化特性、细胞化学分类特征,以及rep-PCR基因指纹分析等方面的研究结果,菌株YIM 90022可能为拟诺卡氏菌属的一个潜在新种。菌株YIM 90022在大多数培养基上生长良好,气生菌丝和基内菌丝丰富,在酵母膏麦芽膏琼脂、燕麦片琼脂等培养基中产生可溶性色素。生长pH范围6.0~12.0,最适pH 8.5;能在含0~15%NaCl(W/V)的培养基上生长。  相似文献

菌株SCY311是从河南省凤凰山土壤样品中分离到的对多种植物病原真菌具有拮抗活性的一株放线菌。为了明确其分类地位, 在形态特征、培养特征、生理生化特征、细胞壁组分测定等传统分类学方法的基础上, 测定和分析了菌株的16S rRNA基因序列。结果表明, 菌株SCY311在高氏一号培养基上生长良好, 基内菌丝呈褐色; 气生菌丝灰色至鼠灰色, 不产生可溶性色素, 无吸水现象; 孢子链卷曲, 末端形成闭合或开放螺旋; 孢子椭圆或圆柱状, 表面形成结节状突起; 生理生化特征和在国际链霉菌计划(ISP)培养基上的培养特  相似文献

为了从土壤环境中筛选产生抗菌活性物质的放线菌,采用琼脂扩散法对分离自湖南省高望界自然保护区森林土壤的169株放线菌进行抗菌活性检测,采用16S RNA基因序列分析法进行系统发育分析,采用常规方法进行生物学特性研究。结果表明,有47株受试菌株的发酵产物显示抗菌活性阳性(27.8%),其中6株具有较强抗菌活性。形态观察结果表明,这6株菌株均为典型的产丝产孢放线菌,在多数培养基上生长良好,气生菌丝和孢子丝丰富。系统发育分析表明,6株菌株均属于链霉菌属(Streptomyces),归为5个物种。菌株JSM 147612、JSM 147786、JSM 147831和JSM 147846分别与链霉菌属的已知物种S.violascens(序列相似性为99.93%)、S.malachitospinus(99.65%)、S.anulatus(99.72%)和S.aureus(99.38%)系统发育关系最为密切;菌株JSM 147823和JSM 147842之间的序列相似性为99.86%,应该属于同一物种,与它们系统发育关系最为密切的是S.albidoflavus(相似性分别为99.72%和99.58%)。  相似文献

一株氨氧化链霉菌的分类鉴定及其氨氧化特性的研究   总被引:2,自引:0,他引:2  
从硝化反应器中分离获得一株链霉菌。根据其形态特征、培养特征、生理生化特性,(G+C)mol%含量以及16S rDNA序列和DNA杂交结果,将其归入链霉菌属中的比基尼链霉菌(Streptomycesbikiniensis)。该菌株既能在YD培养基上异养生长,也能在无机培养基上自养生长,异养生长速率(Vmax为0.39mg/L\5d)明显高于自养生长速率(Vmax为0.22mg/L.d)。异养生长时,氨氮主要用于合成细胞物质;自养生长时,部分氨氮用于合成细胞物质,部分氨氮转化成亚硝酸盐。在无机培养基上自养生长时,最适氨浓度为118mgN/L。最适生长pH值为9.36,最适氨氧化pH值为9.29。最适生长温度为31℃,最适氨氧化温度为40.6℃。提高溶解氧浓度有利于该菌株生长和氨氧化,菌体生长对溶解氧浓度的敏感性高于氨氧化。  相似文献

研究培养条件对来自海洋的12株链霉菌抑菌活性的影响。结果表明,用海水(含3.5%NaCl)制备的高氏1号培养基(pH72)、22℃培养时,能表现抑菌活性的菌株数最多;但减少培养基的营养成分、改变氮源、NaCl浓度、pH和培养温度,部分菌株可表现新的抑菌活性,或使原有的抑菌活性有较大提高;新表现或提高的抑菌活性绝大部分都是针对革兰氏阳性菌和白色假丝酵母。本文结果提示在海洋放线菌抗生素产生菌的筛选中,应注意培养条件的选择。  相似文献

为选育链霉菌11371的高产Tetramycin菌株,摸索该菌株的原生质体的制备与再生。结果表明,链霉菌11371原生质体制备和再生的最佳条件为菌丝生长培养液中甘氨酸浓度为0.7%,培养温度为28℃,培养时间为42h,溶菌酶浓度为3mg/mL,酶解温度为37℃,酶解时间为90min。最佳再生培养基为R2YE培养基。  相似文献

采用响应面法对产生抑菌活性物质的波赛链霉菌(Streptomyces peucetius)菌株JMC 06001的发酵培养基进行优化。首先采用Minnimum Run Equireplicated Res IV设计对初始发酵培养基的8个营养因素进行筛选,获得影响产生抑菌活性物质的3个主要影响因素:葡萄糖、大豆粉和NaCI;然后用最陡爬坡实验快速逼近最大响应区域;最后,结合Box-Behnken设计及响应面分析,确定主要影响因素的最佳浓度,得出该菌株产抑菌活性物质的最优发酵培养基配方为:葡萄糖1.2%,麦芽糖0.7%,蛋白胨0.9%,大豆粉1.4%,NaCl3.7%,CaCO3 0.1%,复合盐A液2.0%,复合盐B液0.1%,起始pH值7.0。用优化后的培养基发酵所得发酵液对敏感指示菌藤黄八叠球菌的抑菌圈直径达31.5mm,与预测值的相对偏差仅为1.59%,与用初始发酵培养基发酵所得发酵液的抑菌效果(抑菌圈直径26.5mm)相比提高了18.9%。  相似文献

使用藤黄八叠球菌(Sarcina lutea)、枯草芽胞杆菌(Bacillus subtilis)、大肠埃希菌(Escherichia coli)、黑曲霉(Asperillus niger)、产气肠杆菌(Enterobacter aerogenes)、变形杆菌(Proteus vulgaris)、白色念珠菌(Candidaalbicans)、青霉(Penicilliumsp.)、铜绿假单胞菌(P.aeruginosa)9种指示菌,采用杯碟法检测抑菌活性。研究了波赛链霉菌JMC 06001菌株(Streptomyces peucetiusJMC 06001)发酵液和菌丝体中活性物质的抑菌谱,对由该菌株产生的抑菌物质的温度、pH值、抗紫外线方面的稳定性进行了考察。利用硅胶柱层析和薄层层析对抑菌物质的分离也做了初步的研究。结果表明,该菌株的发酵产物对部分革兰阳性菌、革兰阴性菌和部分真菌有较为明显的抑制作用。稳定性试验结果显示,该菌株产生抑菌活性物质的最适温度为40℃左右,最佳pH值在7.0左右,具有较好的热稳定性和较宽的pH作用范围,抗紫外线辐射能力较强。通过硅胶柱层析分离和薄层色谱检测,显示该菌次生代谢产物中的抑菌活性物质包含了多种成分。  相似文献

将绿色荧光蛋白基因转入饭豆根瘤茵JMCl402后,与AMF(Arbuscular Mycordaizal Fungi)共同接种饭豆,分别用砂培和土培检测饭豆根瘤茵的侵染途径和定殖动态,并与单接饭豆根瘤茵比较。结果表明,饭豆根瘤茵先吸附在根表,然后从卷曲的根毛侵入。AMF能促进饭豆根瘤茵在根内定殖,抑制饭豆根瘤茵在根表定殖。  相似文献

The journal Primates was founded by Kinji Imanishi (1902–1992) in 1957: It is the oldest and longest-running international primatology journal in the world. In this series of dialogues between Tetsuro Matsuzawa, Editor-in-Chief of Primates and the General Director of the Japan Monkey Centre (JMC) and Juichi Yamagiwa, former Editor-in-Chief of Primates and the Museum Director of the JMC, we look back at the achievements of our spiritual ancestors in primate research and talk about the back story of Imanishi and his fellow primatologists: founding the JMC as a research institute focused on primates and launching this journal. Advances in magnetic resonance imaging (MRI) and computational analysis technology have enabled comparisons among various primate brains in a three-dimensional electronic format. Results from comparative studies provide information about common features across primates and species-specific features of neuroanatomy. Investigation of various species of non-human primates is important for understanding such features, but the majority of comparative MRI studies have been based on experimental primates, such as common marmoset, macaques, and chimpanzee. A major obstacle has been the lack of a database that includes non-experimental primates’ brain MRIs. To facilitate scientific discoveries in the field of comparative neuroanatomy and brain evolution, we launched a collaborative project to develop an open-resource repository of non-human primate brain images obtained using ex vivo MRI. As an initial open resource, here we release a collection of structural MRI and diffusion tensor images obtained from 12 species: pygmy marmoset, owl monkey, white-fronted capuchin, crab-eating macaque, Japanese macaque, bonnet macaque, toque macaque, Sykes’ monkey, red-tailed monkey, Schmidt’s guenon, de Brazza’s guenon, and lar gibbon. Sixteen postmortem brain samples from the 12 species, stored in the Japan Monkey Centre (JMC), were scanned using a 9.4-T MRI scanner and made available through the JMC collaborative research program (http://www.j-monkey.jp/BIR/index_e.html). The expected significant contributions of the JMC Primates Brain Imaging Repository include (1) resources for comparative neuroscience research, (2) preservation of various primate brains, including those of endangered species, in a permanent digital form, (3) resources with higher resolution for identifying neuroanatomical features, compared to previous MRI atlases, (4) resources for optimizing methods of scanning large fixed brains, and (5) references for veterinary neuroradiology. User-initiated research projects beyond these contributions are also anticipated.  相似文献

Junctophilins (JPs) play an important role in the formation of junctional membrane complexes (JMC) in striated muscle by physically linking the transverse-tubule and sarcoplasmic reticulum (SR) membranes. Researchers have found five JP2 mutants in humans with hypertrophic cardiomyopathy. Among these, Y141H and S165F are associated with severely altered Ca(2+) signaling in cardiomyocytes. We previously reported that S165F also induced both hypertrophy and altered intracellular Ca(2+) signaling in mouse skeletal myotubes. In the present study, we attempted to identify the dominant-negative role(s) of Y141H in primary mouse skeletal myotubes. Consistent with S165F, Y141H led to hypertrophy and altered Ca(2+) signaling (a decrease in the gain of excitation-contraction coupling and an increase in the resting level of myoplasmic Ca(2+)). However, unlike S165F, neither ryanodine receptor 1-mediated Ca(2+) release from the SR nor the phosphorylation of the mutated JP2 by protein kinase C was related to the altered Ca(2+) signaling by Y141H. Instead, abnormal JMC and increased SOCE via Orai1 were found, suggesting that the hypertrophy caused by Y141H progressed differently from S165F. Therefore JP2 can be linked to skeletal muscle hypertrophy via various Ca(2+) signaling pathways, and SOCE could be one of the causes of altered Ca(2+) signaling observed in muscle hypertrophy.  相似文献

To determine the tubular sites and mechanisms involved in enhanced renal phosphate (P(i)) reabsorption seen in the juvenile animal, renal micropuncture experiments were performed in acutely thyroparathyroidectomized adult (>14 wk old) and juvenile (4 wk old) male Wistar rats fed either a normal P(i) diet (NPD, 0.6% P(i)) or low P(i) diet (0.07% P(i)) for 2 days, in the presence and absence of parathyroid hormone (PTH). P(i) reabsorption was greater in proximal convoluted (PCT) and straight tubules (PST) of the juvenile compared with adult rats fed NPD, whether or not PTH was present. These findings were consistent with a greater P(i) uptake in brush-border membrane (BBM) vesicles from both superficial (SC) and outer juxtamedullary (JMC) cortices of juvenile animals. Western blot analysis revealed a 2- and 1.8-fold higher amount of NaPi-2 protein in the SC and JMC, respectively, in juvenile rats. Immunofluorescence microscopy also indicated that NaPi-2 protein expression was present in the proximal tubule (PT) BBM to a greater extent in juvenile rats. Dietary P(i) restriction in juvenile rats resulted in a significant increase in P(i) reabsorption in the PCT and PST segments. NaPi-2 expression in the PT BBM was also increased, as was the expression of intracellular NaPi-2 protein. These studies indicate that P(i) reabsorption in both the PCT and PST segments of the renal tubule contributes to the attenuated response to PTH in the normal juvenile animal. In addition, dietary P(i) restriction in the juvenile rat upregulates BBM NaPi-2 expression, which is associated with a further increase in proximal tubular P(i) reabsorption.  相似文献

将从饭且根瘤中分离的饭豆根瘤菌(Rhizobium sp.CYY3302,Rhizobium sp.HCY9101,Rhizobium SP.JMC1402)与ANF(Arbuscular Mycorrltizal Fungi)共同接种于饭豆,进行饭豆、玉米田间小区各种试验。结果表明,接种饭豆根瘤菌和AMF的处理与未接种的处理相比,饭豆的结瘸率比对照提高52%-134%;饭豆及玉米的菌根感染率比对照分别增加43.1%-80%和46.8%-97.6%;饭豆的产量提高了54%-67%,而玉米的产量提高了2.4%-19.5%。研究结果还表明:豆科作物接种根瘸菌,即使在种过豆科作物的老区,也是有效的。  相似文献

The complete genomic sequence of a bluetongue virus serotype 4 (BTV-4) strain (strain YTS-4), isolated from sentinel cattle in Yunnan Province, China, is reported here. This work is the first to document the complete genomic sequence of a BTV-4 strain from China. The sequence information will help determine the geographic origin of Chinese BTV-4 and provide data to facilitate future analyses of the genetic diversity and phylogenetic relationships of BTV strains.  相似文献
