The 5-flanking regions of the Ca-sensitive casein-encoding gene family were analysed for DNA variants by automated DNA sequencing of 13 cows belonging to seven breeds. About 1 kbp of each 5-flanking region, including non-coding exon I, was amplified by PCR and sequenced bidirectionally. A total number of 34 variable sites (17 for the s1, 10 for the s2, and 7 for the casein encoding gene) was identified. Variants were computer-analysed for location in putative regulatory sites in order to predict potential influences on gene expression.  相似文献

Summary Mesembryanthemum crystallinum L., an inducible crassulacean acid metabolism (CAM) plant, was grown for approximately 5 weeks following germination in well-watered, non-saline soil in a controlled-environment chamber. During this time, plants were characterized by C3 photosynthetic carbon metabolism. After the initial 5 weeks, CAM was induced by a combination of high soil salinity and reduced soil water content. One group of plants was allowed to engage in CAM by being continuously exposed to normal CO2-containing air (about 350–400 ppm). A second group of plants was deprived of ambient CO2 each night (12 h dark period) until completion of their life cycle, thereby minimizing potential carbon gain via dark CO2 fixation. The capacity to express CAM under conditions of drought and salinity stress markedly improved reproductive success: plants kept in normal CO2-containing air produced about 10 times more seeds than plants kept in CO2-free air during dark periods. Seeds from plants deprived of ambient CO2 overnight had more negative 13C values than seeds from plants kept in normal air.  相似文献

Temperature-induced lipocalins (TIL) have been invoked in the defense from heat, cold and oxidative stress. Here we document a function of TIL for basal protection from salinity stress. Heterologous expression of TIL from the salt resistant poplar Populus euphratica did not rescue growth but prevented chlorophyll b destruction in salt-exposed Arabidopsis thaliana. The protein was localized to the plasma membrane but was re-translocated to the symplast under salt stress. The A. thaliana knock out and knock down lines Attil1-1 and Attil1-2 showed stronger stress symptoms and stronger chlorophyll b degradation than the wildtype (WT) under excess salinity. They accumulated more chloride and sodium in chloroplasts than the WT. Chloroplast chloride accumulation was found even in the absence of salt stress. Since lipocalins are known to bind regulatory fatty acids of channel proteins as well as iron, we suggest that the salt-induced trafficking of TIL may be required for protection of chloroplasts by affecting ion homeostasis.  相似文献

In order to identify DNA variants within the 5-flanking region of the bovine -casein (Cn)-encoding gene, this area of the gene from 13 cows belonging to seven breeds (Holstein Friesian, Brown Swiss, German Simmental, Jersey, Galloway, Scottish Highland and Ceylon Dwarf Zebu) was analysed. For each individual, about 1 kb of the 5-flanking region including exon I was amplified by polymerase chain reaction (PCR). The biotinylated PCR product was immobilized on magnetic beads followed by direct bidirectional sequencing using an automated DNA sequencer. 该文首次在针叶树种马尾松(Pinus massoniana)中采用cDNA末端快速克隆(RACE)技术克隆,并鉴定出1个CYP735A基因。结果表明:马尾松CYP735A基因(PmCYP735A) cDNA全长为1 744 bp,包括1 647 bp的开放阅读框,44 bp的5'端非翻译区和53 bp的3'端非翻译区。该基因编码蛋白由548个氨基酸残基组成,其二级结构含有丰富的α-螺旋和无规则卷曲。该基因编码蛋白不含跨膜区域,且无信号肽酶切位点,在399~406个和475~484个氨基酸残基存在P450超家族保守特征序列ETLRLYP(ExxRxxP)和血红素结合区域(Heme-binding region) FSFGPRKCVG (FxxGxRxCxG)。系统进化树分析表明,马尾松PmCYP735A与水稻、玉米、拟南芥CYP735A蛋白归属于同一小的进化枝,可可、毛果杨、麻风树和橡胶树等的CYP735A同源蛋白相对集中的定位于另一进化分支。实时定量PCR检测发现,PmCYP735A基因在马尾松根和茎中的表达量显著高于叶,该基因响应外源生长素NAA诱导表达,随着诱导时间呈现先上升再下降的表达趋势。以上结果有助于深入探究CYP735A基因家族的生物学功能及其在物种间的表达调控异同,为进一步挖掘马尾松优异基因资源奠定基础。  相似文献

For the detection of polymorphisms within the 5-flanking region of the -lactoglobulin (-LG) -encoding gene a nucleotide sequence containing 795 bp of the promoter and 59 bp of exon I was cloned and sequenced. After comparing the sequence from the DNA of 11 diverse cows (different breeds and milk-protein yields), 14 singlebp substitutions were identified within the 5-flanking region and two in the 5-untranslated region (5-UTR) of exon I. Some of the variants are located in potential binding sites for trans-acting factors or in the 5-UTR. A PCR-based RFLP analysis was performed, and the genotypes of an additional 60 cows were identified at five variable 5-flanking sites. The results reveal three frequent combinations between the A and B alleles of the protein-coding region and the novel 5-flanking DNA variants. This finding may explain the differences of the protein-variant-dependent -LG synthesis (A>B) observed in vivo. A sequence comparison of the bovine and ovine promoters reveals an homology of 92.8% and shows a higher degree of conservation between positions -600 and -300.  相似文献
