Alpha-amylase inhibitor (alpha AI) protects seeds of the common bean (Phaseolus vulgaris) against predation by certain species of bruchids such as the cowpea weevil (Callosobruchus maculatus) and the azuki bean weevil (Callosobruchus chinensis), but not against predation by the bean weevil (Acanthoscelides obtectus) or the Mexican bean weevil (Zabrotes subfasciatus), insects that are common in the Americas. We characterized the interaction of alpha AI-1 present in seeds of the common bean, of a different isoform, alpha AI-2, present in seeds of wild common bean accessions, and of two homologs, alpha AI-Pa present in seeds of the tepary bean (Phaseolus acutifolius) and alpha AI-Pc in seeds of the scarlet runner bean (Phaseolus coccineus), with the midgut extracts of several bruchids. The extract of the Z. subfasciatus larvae rapidly digests and inactivates alpha AI-1 and alpha AI-Pc, but not alpha AI-2 or alpha AI-Pa. The digestion is caused by a serine protease. A single proteolytic cleavage in the beta subunit of alpha AI-1 occurs at the active site of the protein. When degradation is prevented, alpha AI-1 and alpha AI-Pc do not inhibit the alpha-amylase of Z. subfasciatus, although they are effective against the alpha-amylase of C. chinensis. Alpha AI-2 and alpha AI-Pa, on the other hand, do inhibit the alpha-amylase of Z. subfasciatus, suggesting that they are good candidates for genetic engineering to achieve resistance to Z. subfasciatus.  相似文献

Bruchid larvae cause major losses in grain legume crops throughout the world. Some bruchid species, such as the cowpea weevil and the Mexican bean weevil, are pests that damage stored seeds. Plant lectins have been implicated as antibiosis factors against insects, particularly the cowpea weevil, Callosobruchus maculatus. Talisia esculenta lectin (TEL) was tested for anti-insect activity against C. maculatus and Zabrotes subfasciatus larvae. TEL produced ca. 90% mortality to these bruchids when incorporated in an artificial diet at a level of 2% (w/w). The LD(50) and ED(50) for TEL was ca. 1% (w/w) for both insects. TEL was not digested by midgut preparations of C. maculatus and Z. subfasciatus. The transformation of the genes coding for this lectin could be useful in the development of insect resistance in important agricultural crops.  相似文献

Bruchid beetle larvae cause major losses in grain legume crops throughout the world. Some bruchid species, such as the cowpea weevil (Callosobruchus maculatus) and the Mexican bean weevil (Zabrotes subfasciatus), are pests that damage stored seeds. The Mediterranean flour moth (Anagasta kuehniella) is of major economic importance as a flour and grain feeder; it is often a severe pest in flour mills. Plant lectins have been implicated as antibiosis factors against insects. Bauhinia monandra leaf lectin (BmoLL) was tested for anti-insect activity against C. maculatus, Z. subfasciatus and A. kuehniella larvae. BmoLL produced ca. 50% mortality to Z. subfaciatus and C. maculatus when incorporated into an artificial diet at a level of 0.5% and 0.3% (w/w), respectively. BmoLL up to 1% did not significantly decrease the survival of A. kuehniella larvae, but produced a decrease of 40% in weight. Affinity chromatography showed that BmoLL bound to midgut proteins of the insect C. maculatus. 33 kDa subunit BmoLL was not digested by midgut preparations of these bruchids. BmoLL-fed C. maculatus larvae increased the digestion of potato starch by 25% compared with the control. The transformation of the genes coding for this lectin could be useful in the development of insect resistance in important agricultural crops.  相似文献

Resistance of wild and cultivated rice bean (Vigna umbellata [Thunberg] Ohwi and Ohashi) to three bruchid species, Callosobruchus chinensis L., Callosobruchus maculatus F., and Callosobruchus analis F., was evaluated. All but three accessions of cultivated, and all wild rice bean accessions tested, exhibited complete resistance to all three bruchid species. Rice bean seeds with seed coat removed also showed complete resistance to the three bruchid species. Results indicate that physical attributes and/or chemical(s) present in the seed coat of rice bean are not the main factors responsible for resistance. Feeding tests were performed by using artificial beans prepared with varying proportions of rice bean (resistant) and azuki bean (susceptible) flour. Number of bruchid adults that emerged decreased, and larval developmental period (days) was extended, when artificial beans with an increasing proportion of rice bean flour were used. These tests revealed that a chemical compound(s) contained in the cotyledon of rice bean has an inhibitory effect on the growth of these bruchid species. The results also indicate that the chemical(s) in rice bean cotyledon is most effective against C. maculatus.  相似文献

Abstract.  1. Previous studies have produced conflicting results with respect to the genetic lability of host preference in the seed beetle Callosobruchus maculatus .
2. In this study, replicate lines of an Asian population were kept on an ancestral host (mung bean) or switched to a novel host (cowpea). After 40+ generations, lines were assayed for host preference (in choice tests) and host acceptance (under no-choice conditions), and were compared to African lines chronically associated with cowpea.
3. Host preference diverged in the expected direction. When presented a mixture of cowpeas and mung beans, females from the cowpea lines laid a greater fraction of their eggs on cowpea than did females from the mung bean lines. Preference for cowpea was nearly as strong in the cowpea lines as it was in the cowpea-adapted African lines.
4. In contrast, the experimental host shift did not affect long-term host acceptance. African females laid more eggs if given cowpeas than if given mung beans, but realised fecundities in the cowpea and mung bean lines were similar on the two hosts. Females from all lines laid more eggs if they were reared on cowpea than on mung bean, but rearing host had no effect on either relative host acceptance or host preference.
5. Comparisons with earlier studies suggest that the lability of host preference varies among beetle populations, which precludes generalisation at the species level. Because lines were maintained under no-choice conditions, modification of host preference probably occurred via a lower acceptance threshold for the novel host, without a concomitant change in the long-term acceptance of the ancestral host.  相似文献

The influences of both predator density and elapsed time between initial infestation and introduction of predators were determined for suppression of bruchids infesting stored grain legumes by Xylocoris flavipes (Reuter) (Hemiptera: Anthocoridae). Predator density treatments consisted of zero, one, two, three, or five male:female pairs of adult X. flavipes added to experimental arenas 0, 24, or 120 h after bruchid introduction. Suppression of Acanthoscelides obtectus approached eradication with all predator treatments. For all other bruchid species evaluated (Callosobruchus analis, C. chinensis, C. maculatus, and Zabrotes subfasciatus), the most effective predator density and addition time (five predator pairs at 0 h) resulted in an approximately 50% reduction of emerging F1 bruchids compared with progeny produced in arenas without predators. The predator addition time of 0 h, when predators were added to experimental arenas simultaneously with the pest species, was determined to be the universally most efficacious treatment time. Predator density above one pair but lower than five pairs was less influential overall when X. flavipes was added 24 or 120 h after initial bruchid infestation; however, maximum suppression was achieved at approximately two predator pairs and not significantly improved on with increased predator density. Our results indicate that the most effective biological control of pest bruchids would occur when X. flavipes is added as soon as possible after legumes are stored.  相似文献

Development and reproductive success of the solitary egg parasitoid Uscana lariophaga Steffan were examined after development in eggs of the bruchid storage pest Callosobruchus maculatus Fabricius reared at either low or high densities on cowpea seeds and laid at day 1 and 4 of maternal life. Both bruchid larval competition and maternal age negatively affected egg size, but the latter more than the former. Uscana lariophaga reared in small hosts developed slower, were smaller and produced fewer eggs compared to parasitoids reared in large hosts. Fecundity of the parasitoid was heavily influenced by host egg size. This was reflected in the values for the intrinsic rate of increase of U. lariophaga, which differed for wasps that developed in host eggs laid by bruchid females of different age. A range of accessions (182) of wild, semi-cultivated and cultivatedPhaseolus were subjected to attack by two Old World bruchids,Callosobruchus maculatus and Caliosobruchus chinensis and twoNew World bruchids Zabrotes subfasciatus and Acanthoscelidesobtectus. Proteinase activities in the larval midguts of the bruchids Callosobruchus maculatus and Zabrotes subfasciatus were investigated. Both midgut homogenates showed a slightly acidic to neutral pH optima for the hydrolysis of fluorogenic substrates. Proteolysis of epsilon-aminocaproil-Leu-Cys(SBzl)-MCA was totally inhibited by the cysteine proteinase inhibitors E-64 and leupeptin, and was activated by 1.5 mM DTT in both insects, while hydrolysis of the substrate Z-ArgArg-MCA was inhibited by aprotinin and E-64, which suggests that it is being hydrolysed by serine and cysteine proteinases. Gel assays showed that the proteolytic activity in larval midgut of C. maculatus was due to five major cysteine proteinases. However, based on the pattern of E-64 and aprotinin inhibition, proteolytic activity in larval midgut of Z. subfasciatus was not due only to cysteine proteinases. Fractionation of the larval midgut homogenates of both bruchids through ion-exchange chromatography (DEAE-Sepharose) revealed two peaks of activity against Z-ArgArg-MCA for both bruchid species. The fractions from C. maculatus have characteristics of cysteine proteinases, while Z. subfasciatus has one non-retained peak of activity containing cysteine proteinases and another eluted in a gradient of 250-350 mM NaCl. The proteolytic activity of the retained peak is higher at pH 8.8 than at pH 6.0 and corresponds with a single peak that is active against N-p-tosyl-GlyGlyArg-MCA, and sensitive to 250 microM aprotinin (90% inhibition). The peak contains a serine proteinase which hydrolyzes alpha-amylase inhibitor 1 from the common bean (Phaseolus vulgaris). Arch.  相似文献

It is largely known that the range of an insect diet is mostly determined by oviposition behavior, mainly in species with endophytic larvae such as Zabrotes subfasciatus. However, the proximate factors determining host choice and the subsequent steps leading to the expansion or reduction of the host number and occasional host shifts are largely unknown. We analyzed various factors determining host preference of Z. subfasciatus through the evaluation of: (i) oviposition preference of a wild population of Z. subfasciatus on the usual host (bean) and unusual hosts (lentil, chickpea and soy), and the performance of the offspring; (ii) artificial selection for increasing preference for hosts initially less frequently chosen; (iii) comparison of oviposition behavior between two different populations (reared for ∼30 generations in beans or chickpeas, respectively); (iv) oviposition timing on usual and unusual hosts; and (v) identification of preference hierarchies. We found that when using unusual hosts, there is no correlation between performance and preference and that the preference hierarchy changes only slightly when the population passes through several generations on the less frequently accepted host. We also found a positive response to artificial selection for increasing oviposition on the less preferred host; however, when the host-choice experiment involved two varieties of the usual host, the response was faster than when the choice involved usual and unusual hosts. Finally, beetles reared on an unusual host (chickpea) for 26 generations showed similar good fitness on both usual and unusual hosts, indicating that the use of a new host does not necessarily result in the loss of performance on the original host. Nevertheless, this population showed lower fitness on the usual host than that of the original population, suggesting an underlying partial trade-off phenomenon which may contribute to a broadening of diet of this insect species.  相似文献

本研究采用聚丙烯酰胺凝胶垂直板型电泳法 ,分析了 3种瘤背豆象属豆象 :绿豆象 Callosobruchuschinensis L.、灰豆象 C.phaseoli ( Gyllenhal)、鹰嘴豆象 C.analis ( Fabricius)幼虫的酯酶同工酶。从酶谱的酶带数、迁移率、酶活性强弱 (酶带染色深浅 )、酶含量 (酶带宽窄 )等方面的比较发现 ,3种瘤背豆象属豆象幼虫的酯酶同工酶酶谱具有明显的差异。由此探讨利用酯酶同工酶电泳法鉴别豆象的可能性  相似文献

Wolbachia endosymbionts are widespread among insects and other arthropods, often causing cytoplasmic incompatibility and other reproductive phenotypes in their hosts. Recently, possibilities of Wolbachia-mediated pest control and management have been proposed, and the bean beetles of the subfamily Bruchinae are known as serious pests of harvested and stored beans worldwide. Here we investigated Wolbachia infections in bean beetles from the world, representing seven genera, 20 species and 87 populations. Of 20 species examined, Wolbachia infections were detected in four species, Megabruchidius sophorae, Callosobruchus analis, C. latealbus and C. chinensis. Infection frequencies were partial in M. sophorae but perfect in the other species. In addition to C. chinensis described in the previous studies, C. latealbus was infected with two distinct Wolbachia strains. These Wolbachia strains from the bean beetles were phylogenetically not closely related to each other. Among world populations of C. chinensis, some Taiwanese populations on a wild leguminous plant, Rhynchosia minima, exhibited a peculiar Wolbachia infection pattern, suggesting the possibility that these populations comprise a distinct host race or a cryptic species.  相似文献

Alpha-amylase Inhibitors were isolated from Ficus sp. (Gameleira) seeds by acetone fractionation and Sephadex G-50. Two inhibitors (alpha-PPAI and alpha-ZSAI) were tested against alpha-amylases from coleopteran larvae. alpha-PPAI was active to alpha-amylases of Callosobruchus maculatus (52%) and Zabrotes subfasciatus (53%). alpha-ZSAI was strongly active to Z. subfasciatus (100%) of and Mimosestes mimosae (98%). The alpha-ZSAI is a glycoprotein of approximately 50 kDa with an IC50 value of 0.074 microg microl(-1).  相似文献

通过建立网室近自然实验种群,结合野生个体观察记述了云南玉溪九香虫Coridius chinensis (Dallas)种群的各虫期形态特征、在云南玉溪红塔区的年生活史、寄主范围、取食偏好、成虫及若虫的行为习性,为该虫的保育和规模化养殖提供依据。结果显示,九香虫卵有绿色和粉色两种生态型,若虫有5个龄期;在云南玉溪红塔区1年仅发生1代,仅以成虫态越冬;红塔区九香虫野生个体主要以佛手瓜和南瓜等葫芦科植物为食;取食偏好选择试验表明该虫最喜取食葫芦,冬瓜和南瓜次之;雌雄成虫可多次交配多次产卵,单雌单次平均产卵量为16.3±6.0粒。研究表明,云南玉溪红塔区的九香虫种群为典型的一化性昆虫,休眠期可达8个月;该虫食性较广,可取食多科植物,但以葫芦科瓜类为主;羽化成虫每年9月中旬至10月上旬陆续从佛手瓜、南瓜等寄主植物上迁至枯树皮下、柴堆等隐蔽场所越冬,翌年4月下旬陆续解除休眠,迁至佛手瓜、南瓜等作物上取食繁殖。  相似文献

Eupelmus vuilleti (Hymenoptera; Eupelmidae) is a host feeding ectoparasitoid of fourth-instar larvae or pupae of Callosobruchus maculatus (Coleoptera; Bruchidae) infecting Vigna unguiculata seed and pods (Fabacae). Parasitoid females are synovigenic, i.e. they are born with immature eggs and need to feed from the host in order to sustain egg production. In this study, the role of sterols obtained through host feeding in parasitoid oogenesis are examined. Quantitative and qualitative analyses of the sterol contents in each partner of the tritrophic interaction show that a parasitoid female's larval sterol contents is sufficient to produce only 30% of the total number of eggs laid throughout a female's life cycle. In a second step, by manipulating the composition of the sterols hemolymph in the host, it is shown that cholesterol obtained through adult nutrition plays a crucial role in the eggs viability but does not affect the egg production quantitatively. This result has important implications for understanding both the nutrient allocation strategy in this species and the impact of cholesterol in parasitoid reproduction.  相似文献

Although variation in oviposition preference and specificity for host plants has been demonstrated within populations of a variety of oligophagous insect species, it is unknown whether genetic variation in host choice is lost within populations of monophagous species. Analysis of a locally monophagous butterfly species, Papilio oregonius, and a locally oligophagous species, P. zelicaon, showed significant variation in oviposition preference within populations of both species. Females of both species chose primarily their native hosts. Nonetheless, the percentages of eggs laid by individual females among the plant species and the number of plant species on which individual females laid eggs differed significantly among isofemale strains within populations. Moreover, some females within all isofemale strains of both species laid a few eggs on Foeniculum vulgare, an umbelliferous species that does not occur in the native habitats of these populations but is a host for Papilio species in other geographic areas. The results suggest that local monophagy and oligophagy in these species reflect the relative ranking among potential plant species. Both populations harbor variation in oviposition choice that could allow for host shifts if these populations invaded new habitats.  相似文献

This study examined the effect of single, double, and triple matings on female lifetime fecundity in two closely related species of beetle, Callosobruchus analis and C. maculatus. Multiple mating resulted in elevated lifetime fecundities in both species, although the pattern of elevation differed between the two species. When oviposition resource was plentiful the elevation was apparent after a second but not a third copulation for C. maculatus and after a second and after a third for C. analis. By altering the availability of oviposition sites to C. maculatus females, we were able to alter the pattern of fecundity elevation between matings. When oviposition sites were limited, fecundity increased after a third mating but not a second mating. We suggest that the pattern of fecundity elevation associated with multiple mating in these species is primarily the result of oviposition stimulants delivered by males, rather than a nutritional donation brought about by the metabolism of ejaculates. We also suggest that the intra- and interspecific differences in the relationship between mating frequency and fecundity are the result of differences in egg maturation rates.  相似文献

Abstract. This paper concerns the effects of mate and seed availability on the rate of egg maturation in the bean weevil Callosobruchus maculatus (F.). Egg maturation starts before emergence from the seed and, provided that both oviposition sites and mates are available, eggs are laid at a rate determined by the number of oviposition sites, and mature at a similar rate. If seeds or mates are absent then a small number of eggs are laid, but oocytes continue to mature until the oviduct-capacity is approached. The number of eggs that a female can store is dependent on her body weight and does not correlate with the number of ovarioles. If, after a period in which oocyte development has been halted, conditions for egg-laying become suitable, then egg maturation can be re-started, but only after the oviducts have been emptied of eggs. The rate of egg maturation is then similar to that for females of the same age which have been maturing eggs since emergence.  相似文献

In West Africa, Uscana lariophaga (Hymenoptera: Trichogrammatidae) parasitizes the eggs of Callosobruchus maculatus (Coleoptera: Bruchidae), an important pest of stored cowpea. The impact of the egg parasitoid on pest populations was evaluated in clay pots used in traditional storage in Niger. At the beginning of the storage period cowpeas were infested with different densities of larval instars and adults of C. maculatus and inoculated with one density of U. lariophaga. The higher the initial densities of C. maculatus, the better the egg parasitoid was able to establish itself and to control the population of C. maculatus, limiting the damage to cowpea. After three months of storage, the egg parasitoid significantly reduced the number of C. maculatus adults by 68 at the lowest and 86 percent at the highest initial density of the beetle; the percentage of damaged beans was reduced by 13 and 19% respectively.  相似文献

Females of Callosobruchus spp. are known to produce sex pheromones that attract males. These sex pheromones cannot be adopted for use in pest management without first investigating the responses of the males in the windless conditions of storage environments. Consequently, behavioural bioassays of Callosobruchus subinnotatus Pic males were conducted in an olfactometer in the absence of air-flow. Under these conditions males were found to be able to follow odour trails to the source. However, the latency period was longer in diffusional bioassays than for insects in a Y-tube olfactometer that provided directional wind cues. The highest percentage of males reached the pheromone source when components of the pheromones, (E)-3-methyl-2-heptenoic acid (E32A) and (Z)-3-methyl-2-heptenoic acid (Z32A), were formulated in a 50:50 or 25:75 ratio. Males of C. maculatus (Fabricius) responded to sex pheromone of C. subinnotatus, but males of C. subinnotatus did not respond to that of C. maculatus. The two sex pheromone components of C. subinnotatus are also constituents of C. maculatus sex pheromone. These two components may be potentially useful in monitoring the populations of both species in stored beans. It is postulated that (Z)-3-methyl-3-heptenoic acid (Z33A), the major component of the sex pheromone of C. maculatus, must have acted as an antagonist inhibiting response of C. subinnotatus to the sex pheromone of C. maculatus.  相似文献
