The feeding motivation of the common European common toad (Bufo bufo) can be quantified by the feeding sequence of arousal-orientation-approach-fixate-snap. Previous work has found that the optic tectum is an important structure responsible for the mediation of feeding behaviors, and combined electrical and visual stimulation of the optic tectum was found to increase the animals feeding behaviors. However, the pretectal thalamus has an inhibitory influence upon the optic tectum and its lesion results in disinhibited feeding behaviors. This suggests that feeding behavior of anurans is also subject to influence from the pretectal thalamus. Previous studies involving the application of DC stimulation to brain tissue has generated slow potential shifts and these shifts have been implicated in the modulation of the neural mechanisms associated with behavior. The current study investigated the application of DC stimulation to the diencephalon surface dorsal to the lateral posterodorsal pretectal thalamic nucleus in Bufo bufo, in order to assess effects on feeding motivation. The application of DC stimulation increased the incidence of avoidance behaviors to a visual prey stimulus while reducing the prey catching behavior component of approach, suggesting that the DC current applied to the pretectum increased the inhibition upon the feeding elements of the optic tectum. This can be explained by the generation of slow potential shifts.  相似文献

The intermediate and deep layers of the superior colliculus (SC) are known for their role in initiating orienting behaviors. To direct these orienting functions, the SC of some animals (e.g., primates, carnivores) is dominated by inputs from the distance senses (vision, audition). In contrast, the rodent SC relies more heavily on non-visual inputs, such as touch and nociception, possibly as an adaptive response to the proximity of dangers encountered during their somatosensory-dominant search behaviors. The ferret (a carnivore) seems to employ strategies of both groups: above ground they use visual/auditory cues, but during subterranean hunting ferrets must rely on non-visual signals to direct orienting. Therefore, the present experiments sought to determine whether the sensory inputs to the ferret SC reveal adaptations common to functioning in both environments. The results showed that the ferret SC is dominated (63%; 181/286) by visual/auditory inputs (like the cat), rather than by somatosensory inputs (as found in rodents). Furthermore, tactile responses were driven primarily from hair-receptors (like cats), not from the vibrissae (as in rodents). Additionally, while a majority of collicular neurons in rodents respond to brief noxious stimulation, no such neurons were encountered in the ferret SC. A small proportion (4%; 13/286) of the ferret SC neurons were responsive to long-duration (>5 s) noxious stimulation, but further tests could not establish these responses as nociceptive. Collectively, these data indicate that the ferret SC is best adapted for the animal's visuallacoustically guided activities and most closely resembles the SC of its phylogenetic relative, the cat.  相似文献

The origin of the mammalian neocortex in usually considered as an improvement in the structure of the brain. Alternatively, I suggest that the mammalian neocortex arose as a consequence of contingent adaptations in which there was no specific selection for more elaborate cognitive abilities. In primitive mammals, the adaptation to nocturnal life produced a reduction of the optic tectum (superior colliculus). In addition, the development of the olfactory system triggered the development of the cerebral cortex. It is proposed that, since both the optic tectum and the cerebral cortex are laminar structures, the growing cortex replaced the tectum in many integratory functions. When mammals reinvaded diurnal niches, the optic tectum did not redevelop, and the cerebral cortex remained the main integratory and perceptual system. This is a case of irreversible reduction of an organ. In reptiles and especially in birds, although there was also an increase in brain size (associated with higher cognitive capacities), the optic tectum grew in size and complexity and the forebrain grew largely as a nonlaminar structure (except the Wulst in birds). Therefore, the origin of the cerebral cortex resulted from the combination of adaptations to nocturnality and the development of olfactory-driven behavior, and its origin is not directly related to higher cognitive capacities.  相似文献

The model described is an extension of a previous model of the optic tectum (Arbib & Lara, 1982; Lara, Arbib & Cromarty, 1982; Lara & Arbib, 1982) and takes into consideration anatomical, physiological and behavioral studies in anurans, as well as earlier modelling efforts (Ewert & Von Seelen, 1974; Didday, 1976). Computer simulations were conducted to analyze how interactions among retina, optic tectum and pretectum may give frogs and toads the ability to discriminate between prey and predator stimuli. Results from simulations have allowed us to reproduce empirical observations, to suggest new experiments, and to postulate what neural mechanisms might be involved in some phenomena related to prey-catching orienting behavior, with direction invariance of prey-predator recognition being a consequence of tectal architecture, and size preference and response latency depending on the motivational state of the animal.  相似文献

The developing chick optic tectum is a widely used model of corticogenesis and angiogenesis. Cell behaviors involved in corticogenesis and angiogenesis share several regulatory mechanisms. In this way the 3D organizations of both systems adapt to each other. The consensus about the temporally and spatially organized progression of the optic tectum corticogenesis contrasts with the discrepancies about the spatial organization of its vascular bed as a function of the time. In order to find out spatial and temporal correlations between corticogenesis and angiogenesis, several methodological approaches were applied to analyze the dynamic of angiogenesis in the developing chick optic tectum. The present paper shows that a typical sequence of developmental events characterizes the optic tectum angiogenesis. The first phase, formation of the primitive vascular bed, takes place during the early stages of the tectal corticogenesis along which the large efferent neurons appear and begin their early differentiation. The second phase, remodeling and elaboration of the definitive vascular bed, occurs during the increase in complexity associated to the elaboration of the local circuit networks. The present results show that, apart from the well-known influence of the dorsal-ventral and radial axes as reference systems for the spatial organization of optic tectum angiogenesis, the cephalic-caudal axis also exerts a significant asymmetric influence. The term cortico-angiogenesis to describe the entire process is justified by the fact that tight correlations are found between specific corticogenic and angiogenic events and they take place simultaneously at the same position along the cephalic-caudal and radial axes.  相似文献

Previous studies have shown that intracranial administration of the amphibian neuropeptide arginine vasotocin (AVT) to male rough-skinned newts (Taricha granulosa) can stimulate masculine sexual behaviors. The present study was conducted to determine whether concentrations of endogenous AVT in specific brain areas are correlated with seasonal changes in sexual behavior. Immunoreactive (ir) AVT was measured in the dorsal preoptic area and in the optic tectum throughout the course of the reproductive cycle. In the optic tectum, irAVT levels were low throughout the summer, fall, and winter, but rose over fivefold in the spring when sexual behaviors are most prevalent in male newts. In contrast, irAVT levels in the dorsal preoptic area were high in the summer, but declined in the fall and fluctuated widely during the winter and spring. Thus, there are site-specific changes in irAVT concentrations in the brain of male newts during the seasonal reproductive cycle. The finding that irAVT concentrations in the optic tectum are seasonally correlated with newt sexual behaviors provides further evidence that AVT in this brain area may be important in the expression of sexual behavior.  相似文献

Calretinin is a calcium-binding protein which participates in a variety of functions including calcium buffering and neuronal protection. It also serves as a developmental marker of retinal ganglion cells (RGCs). In order to study the role of calretinin in the development and regeneration of RGCs, we have studied its pattern of expression in the retina at different developmental stages, as well as during optic nerve regeneration by means of immunohistochemistry. During development, calretinin is found for the first time in RGCs when they connect with the optic tectum. Optic nerves from adult zebrafish were crushed and after different survival times, calretinin expression in the retina, optic nerve tract and optic tectum was studied. From the day of crushing to 10 days later, calretinin expression was found to be downregulated within RGCs and their axons, as was also observed during the early developmental stages of RGCs, when they are not committed to a definite cell phenotype. Moreover, 13 days after lesion, when the regenerating axons arrived at the optic tectum, a recovery of calretinin immunoreactivity within the RGCs was observed. These results indicate that calretinin may play an important role during optic nerve regeneration, Thus, the down-regulation of Calretinin during the growth of the RGC axons towards the target during development as well as during their regeneration after injury, indicates that an increase the availability of cytosolic calcium is integral to axon outgrowth thus recapitulating the pattern observed during development.  相似文献

The laminated structure of the optic tectum is formed by radial and tangential cell migration during development. Studies of developing chick optic tectum have revealed two streams of tangential cell migration in the middle and superficial layers, which have distinctive origins, migratory paths, modes of migration, and destinations. We will review the process of the two types of tangential migrations, in order to elucidate their roles in the formation of the optic tectum layers.  相似文献

The developmental profiles of acetylcholinesterase and choline acetyltransferase in chick optic tectum and retina cell aggregates, over a 30-day period, have been determined and compared with the corresponding developmental curves obtained in vivo. Both acetylcholinesterase and choline acetyltransferase activities in retina cell aggregates and the acetylcholinesterase activity in optic tectum cell aggregates usually lie between 40 and 90% of the values measured in vivo for the same cell (tissue) type and developmental age. However, the choline acetyltransferase activity in tectum aggregates increases only during the first 7 days of culture, and then decreases to reach a low value of 8% of that measured in vivo, by day 24. This fact, which is associated with widespread degeneration and cell death, could be attributed to the condition of natural deafferentiation occurring in a tectum cell aggregate. A parallel has been drawn between this behavior of a tectum cell aggregate and the effect of early embryonic eye removal on the development of the contralateral optic tectum in vivo. The alar plate of the prosencephalon of the quail embryo was heterotopically transplanted into the alar plate of the mesencephalon of the chick embryo at the 7–10 somite stage. Chick and quail cells in chimeric brains were distinguished after Feulgen-Rossenbeck staining and/or immunohistochemical staining with a species specific monoclonal antibody MAb-37F5 which recognized cytoplasmic components of chick brain cells. Neural connections between the transplant and the host were studied by monoclonal antibodies, MAb39-B11, which recognizes a species specific antigen on chick nerve fibers, and MAb-29B8, which reacts to 160 kD neurofilaments of both chick and quail.
When the transplant was completely integrated into the host mesencephalon, the transplant developed a laminar morphology closely resembling that of the optic tectum. Immunohistochemical staining with MAb-39B11 showed that the host optic nerve fibers innervated both the host tectum and the tectum-like transplant. However, optic nerve fibers did not invade transplants that failed to develope a laminar structure characteristic of the tectum. These findings suggest that the prosencephalon has a capacity to differentiate into the optic tectum at the 7–10 somite stage.  相似文献

Abstract: The effect of unilateral eye extirpation on the development of the chick optic tectum has been studied in both the embryo and the newly hatched chick. Although the prevention of normal afferentation of the embryonic tectum retarded its growth, there appeared to be a significant increase of muscarinic acetylcholine binding site in the noninnervated tectum. This phenomenon was repeated also in the posthatch denervated system wherein the functioning optic nerve is severed. A significant increase in the number of binding sites as well as reduced dissociation constant of the interactions of this receptor with [3H]quinuclindinyl benzilate was found in the deafferented optic tectum. This may suggest the presence of a denervation-supersensitivity-like modulation. Similar increases were not detected with other binding sites studied in either the noninnervated embryonic or deafferented posthatch optic lobes. The possibility that acetylcholine is a primary neurotransmitter of the optic system is discussed.  相似文献

The localization and proliferative response of optic tectum matrix cells has been studied in adult newt following an experimental lesion on an optic lobe. The results show that 15 days after the lesion the cells in division, autoradiographically labelled, are located in the periventricular layer. Thirty days after the lesion the labelled cells are also found in the innermost grey layers; at 90 days the injured optic tectum regains the cytoarchitecture characteristic of this centre, with labelled cells, whether in the external or in the internal pyriform layers. In all the stages the labelled cells are also found in the periventricular layers of the controlateral optic tectum, in the dorsal pallium and in the striatum. The quantitative data exhibit the existence of a direct relationship between the number of proliferating cells in the injured optic lobe and the extent of the lesion. These data show the possibility of active cellular proliferation for the reconstruction of the lesioned nervous area and for restoration of the characteristic histological structure.  相似文献

Summary. Goldfish retinal explant outgrowth in the presence of fetal calf serum is stimulated by taurine. In the absence of it, but with glucose in the medium, length of neurites is still elevated by the amino acid. Using the medium in the presence of glucose, but in the absence of fetal calf serum, we explored the effect of optic tectum medium from cultures of them coming from goldfish without crush of the optic nerve or 3, 5, 10, 14 and 20 days after crush. Retinal explants, intact or from goldfish with crush of the optic nerve 10 days prior to starting the culture, were employed in order to measure the possible effect of optic tectum media and the inter action with taurine. In other type of experiments the optic nerve was crushed 1, 2, 4, 7 and 10 days before dissection of the optic tectum, and then co-cultured with intact or 10 days post-crush retinal explants. Optic tectum media produced a time-dependent effect on outgrowth in lesioned retinas with a maximum effect around 5 days after the lesion for the corresponding optic tectum. Taurine, 4 mM, did not further affect the outgrowth in the presence of optic tectum media, but did significantly increase length of neurites either in intact or in post-lesion retinas. Co-culture of optic tectum at different days post-lesion and retinas at 10 days post-lesion increased the outgrowth around 4 days post-lesion, in a preparation resulting in mutual effects of both types of tissues. The addition of taurine in these conditions did not further increase outgrowth, rather inhibited it according to the time after lesion of optic nerve corresponding to the co-cultured optic tectum. The effect of taurine was concentration-dependent, since 0.2 mM was more effective than 2 or 4 mM in the presence of optic tectum with lesion of 2 days. These results demonstrate the time-course of the regeneration processes in the visual system of goldfish, indicating the crucial periods after crush in which the tectum could produce stimulation and later decrease or no effect on outgrowth from the retina. In addition, they are evidences of the interaction between taurine and optic tectum production of time-produced specific agents. The mechanisms underlying these effects are closely related to calcium, as it was demonstrated by the addition of extracellular or intracellular chelators to the medium, which inhibited the effects of the optic tectum and the trophic properties of taurine in this system. The inhibitor of taurine transport, guanidoethylsulfonate, also decreased the stimulatory effects of the optic tectum and of taurine, indicating an interaction of substances produced by the tectum with taurine, and an effect of taurine mediated through its entrance to the cells. Overall, retinal explants outgrowth in the absence of fetal calf serum, the interaction of agents of the optic tectum and taurine modulates outgrowth from the retina, and these effects are mediated by calcium levels and by the levels of intracellular taurine.  相似文献

Morphological and histological abnormalities were observed in the regenerating optic tecta of Bufo regularis larvae after partial excision of the left tectum and total excision of the right tectum. They were found in both the left and the right tectum. Invagination of the tectal tissue into the optic ventricle, masses of blood capillaries and gaps or cavities in the tectal tissue were observed. The size of the optic tecta was reduced and the shape and structure of the dorsal aspect of the midbrain were highly anomalous.  相似文献

The right optic tectum of four larval stages of Bufo regularis was subjected to partial and total excision to determine the regenerative capacity of the optic tectum. Intense regenerative capacity can be observed at stage 50; the regenerated part grows to a considerable size and its structure is comparable to that of the intact part. This regenerative capacity gradually diminishes and from stage 55 onwards the decrease is pronounced. At stage 57 regenerative capacity is severly reduced and the regenerated part, if present, always has an anomalous structure.  相似文献

Optic nerve activity helps determine the placement of retinal ganglion cell terminals in the optic tectum of the frog. We investigated whether the presence of this nerve might also influence a characteristic of its target structure, neurotransmitter biosynthesis. We performed unilateral optic nerve transections on adult animals and assayed the percent and intensity of substance P- and serotoninlike immunoreactive (SP-ir and 5-HT-ir, respectively) cells in the deafferented and afferented tectal lobes. Regeneration of the optic nerve was prevented. The percent of SP-ir cells in the afferented tectal lobes was significantly less than that in the deafferented ones either 6 weeks or 5 months following optic nerve lesion. Comparison to normal animals indicated that the change in SP-ir expression was due to a decrease in the percent of immunoreactive cells in the afferented tecta ipsilateral to the optic nerve lesion. The serotoninlike immunoreactivity of tectal cells was also significantly different in the two lobes following optic nerve lesions. This difference resulted from an increase in the percent of 5-HT-ir cells in the deafferented tectum. In addition, the intensity of 5-HT-ir cells in the deafferented lobe was significantly greater than in the afferented one. The staining intensity of SP-ir cells underwent only a transient, relative decrease in the deafferented tectum. We conclude that the optic nerve does regulate substance P and serotonin expression in the tectum, but that this regulation likely occurs through different pathways. © 1996 John Wiley & Sons, Inc.  相似文献

To study the adaptative capabilities of the retinotectal system in birds, the primordium of one optic tectum from 12-somite embryos of Japanese quail was transplanted either homotopically, to replace the ablated same primordium, or heterotopically, to replace the ablated dorsal diencephalon in White Leghorn chick embryos of the same stage. The quail nucleolar marker was used to recognize the transplants. The cytoarchitecture of the tecta and the retinal projections from the eye contralateral to the graft were studied on the 17th or 18th day of incubation in the chimeric embryos by autoradiographic or horseradish peroxidase tracing methods. Morphometric analysis was applied to evaluate the percentage of the tectal surface receiving optic projections. It was observed that: (i) quail mesencephalic alar plate can develop a fully laminated optic tectum even when transplanted heterotopically; (ii) retinal ganglion cells from the chick not only recognize the tectal neurons of the quail as their specific targets in homotopic grafts, but the optic fibers deviate to innervate the heterotopically grafted tectum; (iii) in the presence of a graft, the chick retina is unable to innervate a tectal surface of similar or larger size than that of the control tectum; (iv) tectal regions devoid of optic projections, whether formed by donor or by host cells, always present an atrophic lamination; (v) the diencephalic supernumerary optic tectum competes with and prevails over the host tectum as a target for optic fiber terminals.  相似文献

Adult neurogenesis attracts broad attention as a possible cure for neurological disorders. However, its regulatory mechanism is still unclear. Therefore, they have been studying the cell proliferation mechanisms of neural stem cells (NSCs) using zebrafish, which have high regenerative potential in the adult brain. The presence of neuroepithelial‐type NSCs in the optic tectum of adult zebrafish has been previously reported. In the present study, it was first confirmed that NSCs in the optic tectum decrease or increase in proportion to projection of the optic nerves from the retina. At 4 days after optic nerve crush (ONC), BrdU‐positive cells decreased in the optic tectum's operation side. In contrast, at 3 weeks after ONC, BrdU‐positive cells increased in the optic tectum's operation side. To study the regulatory mechanisms, they focused on the BDNF/TrkB system as a regulatory factor in the ONC model. It was found that bdnf was mainly expressed in the periventricular gray zone (PGZ) of the optic tectum by using in situ hybridization. Interestingly, expression level of bdnf significantly decreased in the optic tectum at 4 days after ONC, and its expression level tended to increase at 3 weeks after ONC. They conducted rescue experiments using a TrkB agonist and confirmed that decrease of NSC proliferation in the optic tectum by ONC was rescued by TrkB signal activation, suggesting stimuli‐dependent regulation of NSC proliferation in the optic tectum of adult zebrafish. © 2016 Wiley Periodicals, Inc. The superior colliculus in mammals or the optic tectum in amphibians is a major visual information processing center responsible for generation of orientating responses such as saccades in monkeys or prey catching avoidance behavior in frogs. The conserved structure function of the superior colliculus the optic tectum across distant species such as frogs, birds monkeys permits to draw rather general conclusions after studying a single species. We chose the frog optic tectum because we are able to perform whole-cell voltage-clamp recordings fluorescence imaging of tectal neurons while they respond to a visual stimulus. In the optic tectum of amphibians most visual information is processed by pear-shaped neurons possessing long dendritic branches, which receive the majority of synapses originating from the retinal ganglion cells. Since the first step of the retinal input integration is performed on these dendrites, it is important to know whether this integration is enhanced by active dendritic properties. We demonstrate that rapid calcium transients coinciding with the visual stimulus evoked action potentials in the somatic recordings can be readily detected up to the fine branches of these dendrites. These transients were blocked by calcium channel blockers nifedipine CdCl2 indicating that calcium entered dendrites via voltage-activated L-type calcium channels. The high speed of calcium transient propagation, >300 μm in <10 ms, is consistent with the notion that action potentials, actively propagating along dendrites, open voltage-gated L-type calcium channels causing rapid calcium concentration transients in the dendrites. We conclude that such activation by somatic action potentials of the dendritic voltage gated calcium channels in the close vicinity to the synapses formed by axons of the retinal ganglion cells may facilitate visual information processing in the principal neurons of the frog optic tectum.  相似文献
