The evolution of sophisticated differentiations of the gastro-intestinal tract enabled herbivorous mammals to digest dietary cellulose and hemicellulose with the aid of a complex anaerobic microbiota. Distinctive symbiotic ciliates, which are unique to this habitat, are the largest representatives of this microbial community. Analyses of a total of 484 different 18S rRNA genes show that extremely complex, but related ciliate communities can occur in the rumen of cattle, sheep, goats and red deer (301 sequences). The communities in the hindgut of equids (Equus caballus, Equus quagga), and elephants (Elephas maximus, Loxodonta africanus; 162 sequences), which are clearly distinct from the ruminant ciliate biota, exhibit a much higher diversity than anticipated on the basis of their morphology. All these ciliates from the gastro-intestinal tract constitute a monophyletic group, which consists of two major taxa, i.e. Vestibuliferida and Entodiniomorphida. The ciliates from the evolutionarily older hindgut fermenters exhibit a clustering that is specific for higher taxa of their hosts, as extant species of horse and zebra on the one hand, and Africa and Indian elephant on the other hand, share related ciliates. The evolutionary younger ruminants altogether share the various entodiniomorphs and the vestibuliferids from ruminants.  相似文献

The genus Equus is richly represented in the fossil record, yet our understanding of taxonomic relationships within this genus remains limited. To estimate the phylogenetic relationships among modern horses, zebras, asses and donkeys, we generated the first data set including complete mitochondrial sequences from all seven extant lineages within the genus Equus. Bayesian and Maximum Likelihood phylogenetic inference confirms that zebras are monophyletic within the genus, and the Plains and Grevy’s zebras form a well-supported monophyletic group. Using ancient DNA techniques, we further characterize the complete mitochondrial genomes of three extinct equid lineages (the New World stilt-legged horses, NWSLH; the subgenus Sussemionus; and the Quagga, Equus quagga quagga). Comparisons with extant taxa confirm the NWSLH as being part of the caballines, and the Quagga and Plains zebras as being conspecific. However, the evolutionary relationships among the non-caballine lineages, including the now-extinct subgenus Sussemionus, remain unresolved, most likely due to extremely rapid radiation within this group. The closest living outgroups (rhinos and tapirs) were found to be too phylogenetically distant to calibrate reliable molecular clocks. Additional mitochondrial genome sequence data, including radiocarbon dated ancient equids, will be required before revisiting the exact timing of the lineage radiation leading up to modern equids, which for now were found to have possibly shared a common ancestor as far as up to 4 Million years ago (Mya).  相似文献

Predators influence prey populations not only through predation itself, but also indirectly through prompting changes in prey behaviour. The behavioural adjustments of prey to predation risk may carry nutritional costs, but this has seldom been studied in the wild in large mammals. Here, we studied the effects of an ambush predator, the African lion (Panthera leo), on the diet quality of plains zebras (Equus quagga) in Hwange National Park, Zimbabwe. We combined information on movements of both prey and predators, using GPS data, and measurements of faecal crude protein, an index of diet quality in the prey. Zebras which had been in close proximity to lions had a lower quality diet, showing that adjustments in behaviour when lions are within short distance carry nutritional costs. The ultimate fitness cost will depend on the frequency of predator–prey encounters and on whether bottom-up or top-down forces are more important in the prey population. Our finding is the first attempt to our knowledge to assess nutritionally mediated risk effects in a large mammalian prey species under the threat of an ambush predator, and brings support to the hypothesis that the behavioural effects of predation induce important risk effects on prey populations.  相似文献

The presence of constitutive alkali-labile sites (ALS) has been investigated using a protocol of DNA breakage detection-fluorescence in situ hybridization and comet assay in spermatozoa of donkey (Equus asinus) and stallion (Equus caballus). These results were compared with those obtained using a similar experimental approach using somatic cells. The relative abundance of ALS was of the order of four times more in spermatozoa than in somatic cells. Alkali-labile sites showed a tendency to cluster localized at the equatorial-distal regions of the sperm. The amount of hybridized signal in the ALS in the sperm of donkey (Equus asinus) was 1.3 times greater than in stallion (Equus caballus), and the length of the comet tail obtained in donkey sperm was 1.6 times longer than that observed in stallion (P < 0.05); however, these differences were not appreciated in somatic cells. In conclusion, ALS localization in sperm is not a randomized event and a different pattern of ALS distribution occurs for each species. These results suggest that ALS represents a species-specific issue related to chromatin organization in sperm and somatic cells in mammalian species, and they might diverge even with very short phylogenetic distances.  相似文献

The fauna yielded by the Nihowan lower pleistocene lacustrine deposits includes two species of Equus beside one Hipparion. Equus sanmeniensis was described in 1930 by Teilhard de Chardin and Piveteau; this species is characterized by its large size and the presence of cups on all incisors. Equus teilhardi nov. sp. is smaller and its lower incisors are devoid of cups. It is the first time that such an absence is noted among eurasiatic Equus; till now only some californian, south-american and african species were known to have lowers incisors without cups. The finding of several Equids in the same deposits and the significance of the lack of cups are briefly discussed.  相似文献

Vibrio fischeri proliferates in a sessile, stable community known as a biofilm, which is one alternative survival strategy of its life cycle. Although this survival strategy provides adequate protection from abiotic factors, marine biofilms are still susceptible to grazing by bacteria-consuming protozoa. Subsequently, grazing pressure can be controlled by certain defense mechanisms that confer higher biofilm antipredator fitness. In the present work, we hypothesized that V. fischeri exhibits an antipredator fitness behavior while forming biofilms. Different predators representing commonly found species in aquatic populations were examined, including the flagellates Rhynchomonas nasuta and Neobodo designis (early biofilm feeders) and the ciliate Tetrahymena pyriformis (late biofilm grazer). V. fischeri biofilms included isolates from both seawater and squid hosts (Euprymna and Sepiola species). Our results demonstrate inhibition of predation by biofilms, specifically, isolates from seawater. Additionally, antiprotozoan behavior was observed to be higher in late biofilms, particularly toward the ciliate T. pyriformis; however, inhibitory effects were found to be widespread among all isolates tested. These results provide an alternative explanation for the adaptive advantage and persistence of V. fischeri biofilms and provide an important contribution to the understanding of defensive mechanisms that exist in the out-of-host environment.  相似文献

Species composition and distribution of large intestinal ciliates was investigated in the feces from 13 Cypriot wild donkeys, free-living in the Karpaz national park, Northern Cyprus. We identified 16 ciliate genera and 22 species. This is the first report on intestinal ciliates in Cypriot wild donkeys, and no endemic species were observed. The genus Cycloposthium occurred in all animals. The mean number of ciliates was 3.0±2.5×104 cells ml?1 of feces and the mean number of ciliate species per host was 6.5±4.8. Characteristics of the wild donkey ciliates was almost identical to those reported in other equids from various regions around the world. We thus conclude that there is no pronounced geographic variation in the intestinal ciliate fauna of equids.  相似文献

Once a diverse family, the Equidae family is now reduced to a single genus, Equus. From the seven extant species of the genus, the African wild ass (Equus africanus) is the most threatened with extinction (last survey indicated 600 individuals). In this work we tested 25 published microsatellite primer pairs isolated from the horse genome on 22 African wild ass (E. africanus) individuals from wildlife reserves and zoos. From the 25 loci tested, 15 amplified well and showed moderate allelic richness (5.06, mean number of alleles) and moderately high expected heterozigosity (0.59). Although all possible loci pairs showed no significant gametic disequilibrium (P?>?0.007), deviations from Hardy–Weinberg proportions were found in 2 out of the 15 analysed microsatellite loci (AHT5 and VHL20). Here, we propose these polymorphic markers to be used as a standard set in future studies on population and conservation genetics of the African wild ass.  相似文献

The Villafranchian fossil record of Perissodactyla is of a great interest because the appearance of the genus Equus and disappearance of the genus Tapirus mark important faunal turnovers. Here, we provide new data on Italian Villafranchian Tapiridae, Rhinocerotidae, and Equidae with updates from the last comprehensive review; most relevant are those of the Rhinocerotidae and Equidae. At present only two Villafranchian rhinoceros’ species are documented in Italy: Stephanorhinus jeanvireti and Stephanorhinus etruscus. The two species can be distinguished by several morphological features and by the dimensions of their bones. Stephanorhinus etruscus occurred throughout the Villafranchian; despite the numerous records, its remains are not enough to investigate morphometric trends. The Equidae include seven species including new occurrences of Hipparionine horses and of the genus Equus. The statistical analyses performed on the Equus species show two different evolutionary trends, namely the stenonine lineage “Equus livenzovensis - Equus stenonis” and the small-to-medium-sized lineage “Equus senenzensis - Equus stehlini”, suggesting a different evolution of these species. Moreover, the analyses show close relationships between the Italian Equus stenonis samples and three European Equus stenonis subspecies (E. stenonis vireti, E. stenonis guthi, and E. stenonis pueblensis). We begin to reconsider the validity of E. stenonis subspecies identification in order to provide new perspectives on the taxonomy of this species.  相似文献

Crop damage caused by herbivorous wildlife species on farms located within conservation landscapes, is a driver of human-wildlife conflict (HWC). Guarding of farms, whereby farmers spend the night out in the fields, in areas adjacent to protected areas is, therefore, very common in many African and Asian countries. Furthermore, guarding is often combined with other crop protection measures, but little is known about the efficacy of these measures.We examined the effect that different traditional and advanced crop protection measures (active and passive guarding strategies, barriers and combinations of measures) had on the magnitude of damaged crops. For this, we examined the cost of crop damage caused by a total of 20 wildlife species in two African and two Asian study areas, where different protection types were applied. Data was compared with the cost of crop damage on unprotected fields. We continuously used a standardised HWC assessment scheme over six years (2009–2014), based on site observations and measurements in addition to interviews with victims.The analysis of crop damage costs revealed substantial losses, especially from that caused by elephants (Loxodonta africana and Elephas maximus) and other large herbivores, such as zebra (Equus quagga) and common eland (Taurotragus oryx). Once wildlife had entered the farms, it was found that crop protection measures by farmers were only able to reduce damage costs when applied as a communal, strategic guarding system. Surprisingly, all other traditional crop protection strategies have proven ineffective in reducing crop damage costs. Electrical fences actually increased the risk of crop damage when combined with guarding and the chasing of wildlife strategies. Therefore, we recommend reviewing the practice of traditional guarding strategies and the effectiveness of fences. Furthermore, we emphasise the need for objective evaluation of HWC mitigation strategies in the long-term.  相似文献

The present paper is based on original and literature data. In Croatia the first studies on the occurrence of ixodid species were made about 80 years ago. The number of tick species recorded in Croatia considerably increased during the 1950s, 60s, 70s and 80s of the past century. A total of 21 species of hard tick belonging to 5 genera have been recorded in Croatia. Ixodes is the best represented genus, with seven species recorded. Haemaphysalis is represented by six species, followed by Rhipicephalus with four species. Dermacentor and Hyalomma are represented by two species each. The ticks were collected on 47 different host species. Eleven tick species were collected on Bos taurus and Ovis aries, followed by Capra hircus and Equus caballus with 8 species and Canis lupus familiaris with 6 species. On the remaining 42 host species one, two or three tick species were collected. The most widespread tick is Ixodes ricinus which was found on 25 different host species.  相似文献

The nature of Chlorella symbioses in invertebrates and protists has attracted much interest, but the uncertain taxonomy of the algal partner has constrained a deeper ecological understanding of this symbiosis. We sequenced parts of the nuclear 18S rDNA, the internal transcribed spacer (ITS)‐1 region, and the chloroplast 16S rDNA of several Chlorella isolated from pelagic ciliate species of different lakes, Paramecium bursaria symbionts, and free‐living Chlorella to elucidate phylogenetic relationships of Chlorella‐like algae and to assess their host specificity. Sequence analyses resulted in well‐resolved phylogenetic trees providing strong statistical support for a homogenous ‘zoochlorellae’ group of different ciliate species from one lake, but clearly different Chlorella in one of those ciliate species occurring in another lake. The two Chlorella strains isolated from the same ciliate species, but from lakes having a 10‐fold difference in underwater UV transparency, also presented a distinct physiological trait, such as the ability to synthesize UV‐absorbing substances known as mycosporine‐like amino acids (MAAs). Algal symbionts of all P. bursaria strains of different origin resolved in one clade apart from the other ciliate symbionts but split into two distinct lineages, suggesting the existence of a biogeographic pattern. Overall, our results suggest a high degree of species specificity but also hint at the importance of physiological adaptation in symbiotic Chlorella.  相似文献

Two endemic subterranean freshwater amphipod species have been discovered in groundwater of the volcanic active zone in Iceland, Crangonyx islandicus and Crymostygius thingvallensis. At least five different ciliate sequences were isolated from C. islandicus and the analysis of variation at the 18S ribosomal DNA gene suggests that they present previously unsequenced species, from the orders Apostomatida and Philasterida. Apostome ciliates are well known to be exuviotrophic epibionts on crustaceans. Analyses of ciliate epibionts from different groundwater amphipod species from North America and mainland Europe revealed distinct groups of ciliates and higher diversity on the epigean amphipod Crangonyx pseudogracilis. Analysis of geographic patterns of the most common ciliate within Iceland revealed population differentiation supporting limited current connectivity between the different groundwater systems. Our study reports an occurrence of previously undescribed ciliate species in a groundwater ecosystem characterized by low species diversity.  相似文献

The population of the Asiatic wild dog or dhole (Cuon alpinus) in the wild has been estimated to be less than 2500 mature individuals. This has led to the IUCN assessing the Asiatic wild dog as Endangered. The killing of Asiatic wild dogs in retaliation for livestock depredation is one of the major threat for the species across its geographic range. Understanding the prey preference of endangered large carnivores is imperative for making informed conservation decisions and devising management strategies. We studied the prey preference of the Asiatic wild dog by reviewing studies that have been conducted on the diet and prey abundance of the species across its distributional range. Nine available studies conducted in eight protected areas of three countries, in which 5265 scats were studied, were reviewed. Across studies, we tested whether prey consumption was based on prey availability or prey body mass. The value of Jacobs’ index was determined for each prey species and tested against the proportion of available prey and body mass. Sambar (Rusa unicolor) and chital (Axis axis) were found to be significantly preferred by the Asiatic wild dog, and the preferred prey body mass range was between 40 and 60 kg. The prey preference of the Asiatic wild dog was mainly influenced by the availability of prey.  相似文献

To genetically assess the Australian distribution and frequency of Eimeria species in wild rabbits, with a primary focus on Eimeria intestinalis and Eimeria flavescens as possible additional agents of rabbit biocontrol, the distal colon and faecal samples from wild rabbits sourced from 26 Australian locations with mean annual rainfalls of between 252 mm and 925 mm were analysed using amplicon sequencing of the ITS1 region. Contrary to previous microscopy studies which had only detected E. flavescens on mainland Australia at Wellstead in south-west Western Australia, we detected this species at all 23 effectively sampled sites. The more pathogenic E. intestinalis was only found at 52.2% of sites. Three unique Eimeria genotypes were detected that did not align to the 11 published sequences using a pairwise-match threshold of 90%, and may represent unsequenced known species or novel species. One genotype we termed E. Au19SH and was detected at 20 sites, E. Au19CO was detected at eight sites, and E. Au19CN was detected in one rabbit at Crows Nest (Qld). Site diversity ranged from only five Eimeria species at Boboyan (ACT) to 13 unique sequences at Cargo (NSW). Eimeria diversity in individual rabbits ranged from 11 unique sequences in a rabbit at Wellstead (WA) and a rabbit at Cargo (NSW), to one in 17 rabbits and zero in six rabbits. The three rabbit age classes averaged 4.3 Eimeria species per rabbit. No relationship was found between the number of Eimeria species detected and mean annual rainfall. As Eimeria species were found to be fairly ubiquitous at most sites they appear to be an unlikely additional candidate to assist the control of pest rabbits in Australia.  相似文献

Vertebrate pigmentation is a fundamentally important, multifaceted phenotype. Zebrafish, Danio rerio, has been a valuable model for understanding genetics and development of pigment pattern formation due to its genetic and experimental tractability, advantages that are shared across several Danio species having a striking array of pigment patterns. Here, we use the sister species D. quagga and D. kyathit, with stripes and spots, respectively, to understand how natural genetic variation impacts phenotypes at cellular and organismal levels. We first show that D. quagga and D. kyathit phenotypes resemble those of wild-type D. rerio and several single locus mutants of D. rerio, respectively, in a morphospace defined by pattern variation along dorsoventral and anteroposterior axes. We then identify differences in patterning at the cellular level between D. quagga and D. kyathit by repeated daily imaging during pattern development and quantitative comparisons of adult phenotypes, revealing that patterns are similar initially but diverge ontogenetically. To assess the genetic architecture of these differences, we employ reduced-representation sequencing of second-generation hybrids. Despite the similarity of D. quagga to D. rerio, and D. kyathit to some D. rerio mutants, our analyses reveal a complex genetic basis for differences between D. quagga and D. kyathit, with several quantitative trait loci contributing to variation in overall pattern and cellular phenotypes, epistatic interactions between loci, and abundant segregating variation within species. Our findings provide a window into the evolutionary genetics of pattern-forming mechanisms in Danio and highlight the complexity of differences that can arise even between sister species. Further studies of natural genetic diversity underlying pattern variation in D. quagga and D. kyathit should provide insights complementary to those from zebrafish mutant phenotypes and more distant species comparisons.  相似文献

Parasites can shape the foraging behaviour of their hosts through cues indicating risk of infection. When cues for risk co-occur with desired traits such as forage quality, individuals face a trade-off between nutrient acquisition and parasite exposure. We evaluated how this trade-off may influence disease transmission in a 3-year experimental study of anthrax in a guild of mammalian herbivores in Etosha National Park, Namibia. At plains zebra (Equus quagga) carcass sites we assessed (i) carcass nutrient effects on soils and grasses, (ii) concentrations of Bacillus anthracis (BA) on grasses and in soils, and (iii) herbivore grazing behaviour, compared with control sites, using motion-sensing camera traps. We found that carcass-mediated nutrient pulses improved soil and vegetation, and that BA is found on grasses up to 2 years after death. Host foraging responses to carcass sites shifted from avoidance to attraction, and ultimately to no preference, with the strength and duration of these behavioural responses varying among herbivore species. Our results demonstrate that animal carcasses alter the environment and attract grazing hosts to parasite aggregations. This attraction may enhance transmission rates, suggesting that hosts are limited in their ability to trade off nutrient intake with parasite avoidance when relying on indirect cues.  相似文献

Calmodulin (CaM) phosphorylated at different serine/threonine and tyrosine residues is known to exert differential regulatory effects on a variety of CaM-binding enzymes as compared to non-phosphorylated CaM. In this report we describe the preparation and characterization of a series of phospho-(Y)-mimetic CaM mutants in which either one or the two tyrosine residues present in CaM (Y99 and Y138) were substituted to aspartic acid or glutamic acid. It was expected that the negative charge of the respective carboxyl group of these amino acids mimics the negative charge of phosphate and reproduce the effects that distinct phospho-(Y)-CaM species may have on target proteins. We describe some physicochemical properties of these CaM mutants as compared to wild type CaM, after their expression in Escherichia coli and purification to homogeneity, including: i) changes in their electrophoretic mobility in the absence and presence of Ca2+; ii) ultraviolet (UV) light absorption spectra, far- and near-UV circular dichroism data; iii) thermal stability in the absence and presence of Ca2+; and iv) Tb3+-emitted fluorescence upon tyrosine excitation. We also describe some biochemical properties of these CaM mutants, such as their differential phosphorylation by the tyrosine kinase c-Src, and their action as compared to wild type CaM, on the activity of two CaM-dependent enzymes: cyclic nucleotide phosphodiesterase 1 (PDE1) and endothelial nitric oxide synthase (eNOS) assayed in vitro.  相似文献

In contrast to the domestic horse, whose digestive physiology has been thoroughly investigated, knowledge on the digestive physiology of wild equids is scarce. Comparisons between the domestic horse and the domestic donkey suggest that wild asses might achieve higher digestibilities. This could derive from longer retention times or a greater difference in the mean retention time (MRT) of particles vs. fluid (the selectivity factor (SF)). Here, we measured MRT of a solute (fluid; MRTsolute) and a particle (<2 mm; MRTparticle) marker in five captive male Somali wild asses (Equus africanus somaliensis) fed a diet of 95% grass hay. At a mean dry matter intake of 94 ± 3 g kg?0.75 day?1, MRTsolute was 33.3 ± 5.4 h and MRTparticle 39.6 ± 3.9 h, resulting in a SF of 1.21 ± 0.14. For their food intake, Somali wild asses appeared to have slightly higher MRTparticle than expected based on domestic equid data, in contrast to Grevy zebras (Equus grevyi), potentially indicating higher capacities of the digestive tract. However, considering data on domestic horses, donkeys, and zebra, there was no evident difference in the SF of wild equids compared to domestic ones. Together with an absence of reported anatomical differences in the digestive tract of wild and domestic equids, the data suggest a general similarity in the digestive physiology of equid species that contrasts with the diversity in the digestive physiology of ruminants, and that might be one contributing factor to a lack of sympatric, niche-differentiated equid species.  相似文献

Adaptive evolution requires both raw genetic material and an accessible path of high fitness from one fitness peak to another. In this study, we used an introgression line (IL) population to map quantitative trait loci (QTL) for leaf traits thought to be associated with adaptation to precipitation in wild tomatoes (Solanum sect. Lycopersicon; Solanaceae). A QTL sign test showed that several traits likely evolved under directional natural selection. Leaf traits correlated across species do not share a common genetic basis, consistent with a scenario in which selection maintains trait covariation unconstrained by pleiotropy or linkage disequilibrium. Two large effect QTL for stomatal distribution colocalized with key genes in the stomatal development pathway, suggesting promising candidates for the molecular bases of adaptation in these species. Furthermore, macroevolutionary transitions between vastly different stomatal distributions may not be constrained when such large-effect mutations are available. Finally, genetic correlations between stomatal traits measured in this study and data on carbon isotope discrimination from the same ILs support a functional hypothesis that the distribution of stomata affects the resistance to CO2 diffusion inside the leaf, a trait implicated in climatic adaptation in wild tomatoes. Along with evidence from previous comparative and experimental studies, this analysis indicates that leaf traits are an important component of climatic niche adaptation in wild tomatoes and demonstrates that some trait transitions between species could have involved few, large-effect genetic changes, allowing rapid responses to new environmental conditions.  相似文献
