The existence of antigenic differences between cell populations in the local growth of the 3LL tumor (L-3LL) and its lung metastases (M-3LL) was studied. Normal C57BL/6 spleen cells sensitized in vitro for 5 days against L-3LL monolayers lysed preferentially L-3LL targets but not M-3LL tumor cell targets. Conversely, anti-M-3LL-sensitized lymphocytes killed M-3LL targets more efficiently than they killed L-3LL targets. Furthermore, spleen cells from mice bearing subcutaneous L-3LL tumors were significantly more cytotoxic to L-3LL targets than to M-3LL targets and vice versa. M-3LL cells were found also to be more resistant in vitro and in vivo to natural killer cells than were L-3LL tumor cells. M-3LL cells were more resistant than L-3LL cells to hybrid resistant mechanisms when they were inoculated into F1 (C3Heb X C57BL/6) or F1 (BALB/c X C57BL/6) mice. Anti-M-3LL lymphocytes generated both in vitro and in vivo, but not anti-L-3LL lymphocytes, admixed with L-3LL or M-3LL tumor cells and inoculated into footpads of syngeneic recipients suppressed the development of lung metastases. These results suggest that metastatic cells are indeed phenotypic variants of the local growing tumor cell populations. Presumably, these variants are selected for their capacity to home to and grow in the lungs, and for their resistance to specific immune effects initially evoked against the local tumor and to nonspecific natural killer cells. These data may prove to be of importance with respect to any rational approach to the problem of immunotherapy.  相似文献

Analogous to protans, the two types of deutan color-defectives—the dichromats (deuteranopes) and the anomalous trichromats (deuteranomalous)—do not differ in spectral sensitivity in the red-green range at threshold (either in the dark or against bright colored backgrounds). However, luminosity curves obtained by heterochromatic brightness matching show the latter to be slightly more sensitive in the blue-green, and slightly less so in the red, than the former. Experiment proves that these differences are due (at least in part) to contributions of cones containing the deuteranomalous anomalous pigment which are missing from the deuteranope's eye. The absorption spectrum of the anomalous pigment can be inferred with assumptions (analogous to those already made with protanomalous trichromats) about how the different cone mechanisms pool their responses to yield luminosity. Two alternatives thus revealed are (a) the normal red pigment in dilute solution or (b) a spectrum very similar to that of the normal red pigment but shifted slightly toward the short wave end of the spectrum. Since the spectrum inferred by (a) has the same λmax as the normal red pigment, (a) predicts that deuteranomalous observers will require a negative red primary when matching monochromatic lights of wavelengths near the λmax. Previous studies have provided evidence that color measurements enable on site quantification of superficial biofilms, thereby avoiding the need for sampling. In the present study, the efficiency of color measurements to evaluate to what extent pigment production is affected by environmental parameters such as light intensity, combined nitrogen and nutrient availability, was tested with two cyanobacteria, Nostoc sp. strains PCC 9104 and PCC 9025, which form biofilms on stone. Both strains were acclimated, in aerated batch cultures for 2 weeks, to three different culture media: BG-11, BG-110, and BG-110/10 at either high or low light intensity. The content of chlorophyll a, carotenoids, and phycocyanins was measured throughout the experiment, together with variations in the color of the cyanobacteria, which were represented in the CIELAB color space. The results confirmed that the CIELAB color parameters are correlated with pigment content in such a way that variations in the latter are reflected as variations in color.  相似文献

The squirrel monkey (Saimiri sciureus) exhibits a polymorphism of colour vision: some animals are dichromatic, some trichromatic, and within each of these classes there are subtypes that resemble the protan and deutan variants of human colour vision. For each of ten individual monkeys we have obtained (i) behavioural measurements of colour vision and (ii) microspectrophotometric measurements of retinal photopigments. The behavioural tests, carried out in Santa Barbara, included wavelength discrimination, Rayleigh matches, and increment sensitivity at 540 and 640 nm. The microspectrophotometric measurements were made in London, using samples of fresh retinal tissue and a modified Liebman microspectrophotometer: the absorbance spectra for single retinal cells were obtained by passing a monochromatic measuring beam through the outer segments of individual rods and cones. The two types of data, behavioural and microspectrophotometric, were obtained independently and were handed to a third party before being interchanged between experimenters. From all ten animals, a rod pigment was recorded with lambda max (wavelength of peak absorbance) close to 500 nm. In several animals, receptors were found that contained a short-wave pigment (mean lambda max = 433.5 nm): these violet-sensitive receptors were rare, as in man and other primate species. In the middle- to long-wave part of the spectrum, there appear to be at least three possible Saimiri photopigments (with lambda max values at about 537,550 and 565 nm) and individual animals draw either one or two pigments from this set, giving dichromatic or trichromatic colour vision. Thus, those animals that behaviourally resembled human protanopes exhibited only one pigment in the red-green range, with lambda max = 537 nm; other behaviourally dichromatic animals had single pigments lying at longer wavelengths and these were the animals that behaviourally had higher sensitivity to long wavelengths. Four of the monkeys were behaviourally judged to be trichromatic. In a haplontic green alga, mating group A of the Closterium ehrenbergii Meneghini ex Ralfs species complex, viability of meiotic progeny was studied by isolating two gone cells from a single germinating zygospore. In F1 progeny of a cross between mating-type plus M-16-4a and mating-type minus M-16-4b, studied in six independent experiments, percentage survivals varied little from 86 to 96 with a mean of 93 ± 1.4 SE. In F2 progenies of crosses among eight mating-type plus and eight mating-type minus F1 clones of the M-16-4a · M-16-4b cross, percentage survival varied considerably from 24 to 100, with a mean of 70.8 ± 2.2. In B1 progenies of the above eight mating-type plus F1 clones, survival values were significantly different between backcrosses to the recurrent M-16-4b (range = 32–83, mean ± SE = 58.3 ± 6.8) and backcrosses to a genetically unrelated mating-type minus, R-13-20, (85–97, 92 ± 1.6). Also in B1 progenies of the above mating-type minus F1 clones, survival values were significantly different between backcrosses to the recurrent M-16-4a (56–90, 68.3 ± 4,4) and backcrosses to a genetically unrelated mating-type plus, R-13-131 (78–93, 86.1 ± 1.6). Clearly, viabilities of meiotic progeny differed considerably between outbreedings (M-16-4a × M-16-4b, and F, clones × R-13-20 or R-13-131) and inbreedings (F2 and F1 clones × M-16-4a, or M-16-4b). These data suggest the presence of hidden deleterious genetic factors that may reduce viability of zygospore progeny if inbred between a pair of wild-type strains from the normally outbreeding mating group A of the C. ehrenbergii species complex.  相似文献

Light-dark cycles are the most important time cue for the circadian system to entrain the endogenous circadian clock to the environmental 24 h cycle. Although photic entrainment of circadian rhythms is mediated by the eye in mammals, photoreceptors implicated in circadian photoreception remain unknown. In our previous study, retinally degenerate CBA/J (rd/rd) mice were found to have lower circadian photo-sensitivity for phase-shifting the locomotor activity rhythms than normal CBA/N(+/+) mice. In the present study, the spectral sensitivity for phase-shifting the rhythms was examined in order to characterize the photopigments involved in circadian photoreception of these mice. The spectral sensitivity of CBA/J-rd/rd mice clearly fitted to the Dartnall nomogram for a retinal1-based pigment with a maximum at 480 nm, while the best fitted nomogram had a maximum at 500 nm in CBA/N- +/+ mice. These results suggest that circadian photopigments involved in CBA/J-rd/rd and CBA/N- +/+ mice may be different.  相似文献

1. Protanopes and deuteranopes show one maximum of wavelength discrimination which occurs near their neutral point in the region of 500 mµ (blue-green for color-normal). The value of the just discriminable wavelength interval Δλ is about 1 mµ at this point and is much like the normal. To either side of this, Δλ rises. It increases rapidly on the short-wave side, and slowly on the long-wave side, rising to about 50 mµ at the two ends of the spectrum. 2. The brightness distribution in the spectrum for dichromats falls only partly outside the range established for color-normals. The protanope curve is narrower than normal, and its maximum lies nearly 15 mµ to the left of it. The deuteranope curves are about the same width as the normal, and their maxima lie slightly but definitely to the right of it. The main difference between protanope and deuteranope spectrum sensitivity lies on the red side of brightness curves, where the deuteranope is strikingly higher. This difference furnishes the only reliable diagnostic sign which may be applied to an individual dichromat for separating the two types. 3. The average position for the neutral point of twenty-one protanopes is 496.5 mµ; of twenty-five deuteranopes 504.3 mµ. The range of variation in the position of neutral point is twice as great for the deuteranope as for the protanope. 4. Dichromatic gauging of the spectrum cannot yield unique mixture values for any wavelength because of the large stretches of poor wavelength discrimination. Data have therefore been secured which locate the spectral ranges that can match specific mixtures of two primaries when brightness differences are eliminated. The form of the data is much the same for a protanope and for a deuteranope; the only difference is in the relative brightness of the primaries. 5. Previously accepted anomalies in the spectral matching of dichromats which have led to the rejection of the third law of color mixture for them, have been eliminated. They are shown to have been due to the non-uniqueness of color matches and the usually disparate brightnesses of the primaries. Color mixture matches for dichromats are valid at all brightnesses.  相似文献

Threshold spectral sensitivities (in the dark, or against bright colored backgrounds) are identical in the red-green range for both protanopes (dichromats) and protanomalous trichromatic color defectives. The latter, however, must have an additional photolabile cone pigment in the red-green range, and its presence is revealed by heterochromatic brightness matching through the spectrum (i.e. luminosity curves). The absorption spectrum of the anomalous cone pigment can be inferred from the protanomalous and protanopic luminosity curve, given reasonable assumptions as to how the different cone mechanisms pool their responses. Depending upon these assumptions, the pigment inferred is either (a) dilute solution of the normal red pigment (assumed density 1.0 for the deuteranope) or (b) similar in its absorption spectrum to the normal green pigment but shifted slightly toward the long wave end of the spectrum. Experimental attempts to choose between these alternatives have so far proved equivocal though (b) seems more likely on the basis of indirect evidence.  相似文献

The alkali metal (M=Li, Na, and K) para-substituted (M-1), meta-substituted (M-2) or ortho-substituted (M-3) derivatives of phenylpentazole (PhN5) were studied using density functional theory. The substituted metals improve the energy barrier for decomposition of the N5 ring of PhN5 by 19.3?~?65.0 kJ/mol. M-3 has the ionic N-M bond, which is not found for M-1 and M-2. M-1 and M-2 have similar electrostatic potentials and dispersion interactions between metal and N5 ring. The comparable intramolecular interactions of M-1 and M-2 lead to similar N5 ring stability. Compared to M-1 and M-2, M-3 has a more negative charge on N5 ring and stronger dispersion interaction. The stronger intramolecular interactions of M-3 result in the higher N5 ring stability. For M-1 and M-2, different metals have slight affects on N5 ring stability. For M-3, N5 ring stability decreases in the order of Li > Na > K. The substituted metal lowers E g of PhN5.  相似文献

Retinal extracts of Gekko gekko were found to contain two retinene1 photopigments, one with maximum absorption at about 521 mµ, the second with a maximum in the region of 478 mµ. These pigments were assayed by the method of partial bleaching and their spectral characteristics studied by examining their difference spectra. The 478 mµ pigment was present in the extracts as 8 per cent of the total photopigment concentration. The two pigment systems were shown to be biochemically independent and to have different properties. Unlike the 521 mµ pigment, for example, the 478 mµ pigment was found to resist the action of NH2OH and, within the cells, to be unaffected by sucrose solutions. These solutions destroyed or altered the 521 system so that extracts of sucrose-treated retinae were found to contain significantly less 521 photopigment. In digitonin solution the 521 pigment was unaffected by sucrose treatment. Both pigments were extracted from separated, washed outer segments and so are considered to be visual pigments. This dual system accounts for the spectral sensitivity of this gecko as determined by Denton. A search was made, but no evidence was secured for the presence of a photopigment absorbing at longer wavelengths. Electoretinographic data suggest, however, that an elevated sensitivity at longer wavelengths occurs in some geckos so that a continued search is justified for a third photopigment.  相似文献

The photochemical behavior of intact stream periphyton communities in France was evaluated in response to the time course of natural light. Intact biofilms grown on glass substrata were collected at three development stages in July and November, and structural parameters of the biofilms were investigated (diatom density and taxonomy). At each season, physiological parameters based on pigment analysis (HPLC) and pulse‐amplitude‐modulated (PAM) chl fluorescence technique were estimated periodically during a day from dawn to zenith. Regardless of the community studied, the optimal quantum yield of PSII (Fv/Fm), the effective PSII efficiency (ΦPSII), the nonphotochemical quenching (NPQ), and the relative electron transport rate (rETR) exhibited clear dynamic patterns over the morning. Moreover, microalgae responded to the light increase by developing the photoprotective xanthophyll cycle. The analysis of PI parameters and pigment profiles suggests that July communities were adapted to higher light environments in comparison with November ones, which could be partly explained by a shift in the taxonomic composition. Finally, differences between development stages were significant only in July. In particular, photoinhibition was less pronounced in mature assemblages, indicating that self‐shading (in relation to algal biomass) could have influenced photosynthesis in older communities.  相似文献

Photopigment radiolabcling, a useful method for measuring thein situ carbon-specific growth rates of microalgae, is basedon the determination of synthesis rates of chemosystematic (i.e.specific for microalgal phylogenetic groups) chlorophylls andcarotenoids using photosynthetically assimilated 14C as a radiotracer.The reliability of this method depends on accurate measurementsof the 14C-specific activity of individual photopigments. Typically,photopigments are separated by high-performance liquid chromatography(HPLC) with fraction collection of individual peaks, followedby further purification and standard scintillation counting.To simplify analyses, we evaluated in-line flow scintillationcounting to determine its applicability and reliability formeasuring the activity of radio-labeled photopigments. Incubationswere conducted using both pure cultures and natural phyto-planktonsamples. The radiochemical purity of photopigments was determinedby extract acidification (10% HC1) to transform chlorophyllsinto degradation products. Purity was also checked by comparingabsorbance spectra with purified standards. Although 14C-labeledcolorless compounds are a common feature in radiograms, thesecompounds do not co-elute with photopigments using our HPLCprotocol. Flow scintillation counting, coupled with a highlyselective HPLC protocol, provides an efficient, reliable andfeasible alternative to fraction collection/repurification methodsfor measuring the 14C-specific activity of microalgal photosyntheticpigments.  相似文献

In the aqueous solution of copper(II) ions, bidentate L-α-alaninehydroxamic acid (CH3CH(NH2)-CONHOHHL) binds cupric ion forming of monodimeric and bis(L-α-alaninehydroxamato)copper(II) complexes. These complexes were studied by potentiometric, ESR and spectrophotometric methods.The ESR studies provide important evidence for the formation of different Cu(II) complexes with L-α-alaninehydroxamic acid, depending on pH. The ESR spectra can be used to follow the appearance of the individual complexes, to estimate the coordination sphere around Cu(II) and to observe the equilibria between different complexes.The solution electronic spectra are reported. The experimental curve was resolved into precise- positioned absorption bands by Gaussian analysis for the bis(L-α-alaninehydroxamato)copper(II) species. These data were used in a weak tetragonal ligand field model to calculate ligand field parameters.The distribution and the relevant stability constants of species present in aqueous solutions were obtained by analytical potentiometry.  相似文献

AIMS: A previous model for adaptation and growth of individual bacterial cells was not dynamic in the lag phase, and could not be used to perform simulations of growth under non-isothermal conditions. The aim of the present study was to advance this model by adding a continuous adaptation step, prior to the discrete step, to form a continuous-discrete-continuous (CDC) model. METHODS AND RESULTS: The revised model uses four parameters: N(0), initial population; N(max), maximum population; p0, mean initial individual cell physiological state; SD(p0), standard deviation of the distribution of individual physiological states. A truncated normal distribution was used to generate tables of distributions to allow fitting of the CDC model to viable count data for Listeria monocytogenes grown at 5 degrees C to 35 degrees C. The p0 values increased with increasing SD(p0) and were, on average, greater than the corresponding population physiological states (h0); p0 and h0 were equivalent for individual cells. CONCLUSION: The CDC model has improved the ability to simulate the behaviour of individual bacterial cells by using a physiological state parameter and a distribution function to handle inter-cell variability. The stages of development of this model indicate the importance of physiological state parameters over the population lag concept, and provide a potential approach for making growth models more mechanistic by incorporating actual physiological events. SIGNIFICANCE AND IMPACT OF THE STUDY: Individual cell behaviour is important in modelling bacterial growth in foods. The CDC model provides a means of improving existing growth models, and increases the value of mathematical modelling to the food industry.  相似文献

The relationship between regional variabilities in airflow (ventilation) and blood flow (perfusion) is a critical determinant of gas exchange efficiency in the lungs. Hypoxic pulmonary vasoconstriction is understood to be the primary active regulator of ventilation-perfusion matching, where upstream arterioles constrict to direct blood flow away from areas that have low oxygen supply. However, it is not understood how the integrated action of hypoxic pulmonary vasoconstriction affects oxygen transport at the system level. In this study we develop, and make functional predictions with a multi-scale multi-physics model of ventilation-perfusion matching governed by the mechanism of hypoxic pulmonary vasoconstriction. Our model consists of (a) morphometrically realistic 2D pulmonary vascular networks to the level of large arterioles and venules; (b) a tileable lumped-parameter model of vascular fluid and wall mechanics that accounts for the influence of alveolar pressure; (c) oxygen transport accounting for oxygen bound to hemoglobin and dissolved in plasma; and (d) a novel empirical model of hypoxic pulmonary vasoconstriction. Our model simulations predict that under the artificial test condition of a uniform ventilation distribution (1) hypoxic pulmonary vasoconstriction matches perfusion to ventilation; (2) hypoxic pulmonary vasoconstriction homogenizes regional alveolar-capillary oxygen flux; and (3) hypoxic pulmonary vasoconstriction increases whole-lobe oxygen uptake by improving ventilation-perfusion matching.  相似文献

Platyrrhine (New World) monkeys possess highly polymorphic color vision owing to allelic variation of the single-locus L/M opsin gene on the X chromosome. Most species consist of female trichromats and female and male dichromats. Howlers (genus Alouatta) are an exception; they are considered to be routinely trichromatic with L and M opsin genes juxtaposed on the X chromosome, as seen in catarrhine primates (Old World monkeys, apes, and humans). Yet it is not known whether trichromacy is invariable in howlers. We examined L/M opsin variation in wild howler populations in Costa Rica and Nicaragua (Alouatta palliata) and Belize (A. pigra), using fecal DNA. We surveyed exon 5 sequences (containing the diagnostic 277th and 285th residues for λmax) for 8 and 18 X chromosomes from Alouatta palliata and A. pigra, respectively. The wavelengths of maximal absorption (λmax) of the reconstituted L and M opsin photopigments were 564 nm and 532 nm, respectively, in both species. We found one M–L hybrid sequence with a recombinant 277/285 haplotype in Alouatta palliata and two L–M hybrid sequences in A. pigra. The λmax values of the reconstituted hybrid photopigments were in the range of 546~554 nm, which should result in trichromat phenotypes comparable to those found in other New World monkey species. Our finding of color vision variation due to high frequencies of L/M hybrid opsin genes in howlers challenges the current view that howlers are routine and uniform trichromats. These results deepen our understanding of the evolutionary significance of color vision polymorphisms and routine trichromacy and emphasize the need for further assessment of opsin gene variation as well as behavioral differences among subtypes of trichromacy.  相似文献

After fixating on a colored pattern, observers see a similar pattern in complementary colors when the stimulus is removed [1-6]. Afterimages were important in disproving the theory that visual rays emanate from the eye, in demonstrating interocular interactions, and in revealing the independence of binocular vision from eye movements. Afterimages also prove invaluable in exploring selective attention, filling in, and consciousness. Proposed physiological mechanisms for color afterimages range from bleaching of cone photopigments to cortical adaptation [4-9], but direct neural measurements have not been reported. We introduce a time-varying method for evoking afterimages, which provides precise measurements of adaptation and a direct link between visual percepts and neural responses [10]. We then use in vivo electrophysiological recordings to show that all three classes of primate retinal ganglion cells exhibit subtractive adaptation to prolonged stimuli, with much slower time constants than those expected of photoreceptors. At the cessation of the stimulus, ganglion cells generate rebound responses that can provide afterimage signals for later neurons. Our results indicate that afterimage signals are generated in the retina but may be modified like other retinal signals by cortical processes, so that evidence presented for cortical generation of color afterimages is explainable by spatiotemporal factors that modify all signals.  相似文献

The photosensitivities of the bovine rhodopsin and gecko pigment 521 analogues regenerated from C-10-substituted analogues of 11-cis- and 9-cis-retinals were determined by two different methods. A similar reactivity trend was noted for both pigment systems as revealed in the photosensitivity of the gecko pigments and relative quantum yields of the bovine analogues. The 10-fluoro-11-cis photopigments had a photosensitivity less than, but approaching, that of the native (11-cis) visual pigment while the 10-fluoro-9-cis photopigments had a much lower photosensitivity than the parent 9-cis regenerated pigment. The results are interpreted in terms of recently described models of rhodopsin architecture and of the primary molecular reaction of visual pigments to light. The unusually low photoreactivity of the 10-fluoro-9-cis pigment molecule is viewed as the result of a regiospecific hydrogen-bonding interaction of the electronegative fluorine atom to the opsin.  相似文献

The aim of this study was to assess the impact of the microalgae Chlorella vulgaris on the rice seedlings at physiological conditions and under cadmium (Cd) stress. We examined the effects of C. vulgaris in the nutrient solution on rice seedlings grown hydroponically in the presence and the absence of 150 μM CdCl2, using the low (77 K) temperature and pulse amplitude modulated (PAM) chlorophyll fluorescence, P700 photooxidation measurements, photochemical activities of both photosystems, kinetic parameters of oxygen evolution, oxidative stress markers (MDA, H2O2 and proline), pigment content, growth parameters and Cd accumulation. Data revealed that the application C. vulgaris not only stimulates growth and improves the functions of photosynthetic apparatus under physiological conditions, but also reduces the toxic effect of Cd on rice seedlings. Furthermore, the presence of the green microalgae in the nutrient solution of the rice seedlings during Cd exposure, significantly improved the growth, photochemical activities of both photosystems, the kinetic parameters of the oxygen-evolving reactions, pigment content and decreased lipid peroxidation, H2O2 and proline content. Data showed that the alleviation of Cd-induced effects in rice seedlings is a result of the Cd sorption by microalgae, as well as the reduced Cd accumulation in the roots and its translocation from the roots to the shoots.  相似文献
