The morphometry, feeding habits and reproductive biology of Etroplus suratensis, an indigenous cichlid food fish, were studied for a period of one year on specimens from three different habitats in Sri Lanka. The morphometric studies implied that the length weight relationships are curvilinear. Macrophytic fragments formed the most important component of food, although detritus, diatoms, and animal matter are also ingested. First sexual maturity is attained in the length ranges 8.0-9.0 cm in females from brackish water habitats. Fecundity ranged from 700-3900 eggs in mature specimens. The wide range of the gonadosomatic index suggests that spawning occurs several times during the year.  相似文献

As previously reported, narcissus latent virus (NLV) has flexuous filamentous particles measuring c. 650 nm × 13 nm, is manually transmissible to Nicotiana clevelandii and Tetragonia expansa, and is transmitted by the aphid Myzus persicae following brief acquisition access periods. In contrast to previous reports the virus particle protein has an apparent mol. wt of c. 45 kD. Moreover, infected cells in N. clevelandii leaves contain cytoplasmic inclusion bodies resembling those of potyviruses. In vitro translation of NLV RNA produced only one major product (mol. wt c. 25 kD) which was not precipitated by antisera to virus particle protein or to cytoplasmic inclusion protein. Antisera to 12 potyviruses and nine carlaviruses failed to react with sap containing NLV particles. Similarly antiserum to NLV particles did not react with particles of seven potyviruses or four carlaviruses. A weak reaction was detected between NLV particles and antiserum to particles of maclura mosaic virus (MMV), a virus which resembles NLV in particle morphology and particle-protein size, and in inducing pinwheel inclusions. The cytoplasmic inclusion proteins (CIPs) of NLV, MMV and from narcissus plants with yellow stripe symptoms were serologically inter-related. These proteins were also serologically related to, and had mol. wt similar to, the CIP of members of the potyvirus group. Particles with the size and antigenic specificity of those of NLV were found consistently in narcissus plants with yellow stripe disease. Narcissus latent and narcissus yellow stripe viruses therefore seem to be synonymous and, together with MMV, have properties distinct from those of any previously described virus group.  相似文献

Mutations in the eIF4E homolog encoded at the pvr1 locus in Capsicum result in broad-spectrum potyvirus resistance attributed to the pvr1 resistance allele, a gene widely deployed in agriculture for more than 50 years. We show that two other resistance genes, previously known to be eIF4E with narrower resistance spectra, pvr2(1) and pvr2(2), are alleles at the pvr1 locus. Based on these data and current nomenclature guidelines, we have re-designated these alleles, pvr1(1) and pvr1(2), respectively. Point mutations in pvr1, pvr1(1), and pvr1(2) grouped to similar regions of eIF4E and were predicted by protein homology models to cause conformational shifts in the encoded proteins. The avirulence determinant in this potyvirus system has previously been identified as VPg, therefore yeast two-hybrid and GST pull-down assays were carried out with proteins encoded by the pvr1 alleles and VPg from two different strains of Tobacco etch virus (TEV) that differentially infected Capsicum lines carrying these genes. While the protein encoded by the susceptible allele pvr1+ interacted strongly, proteins translated from all three resistance alleles (pvr1, pvr1(1), and pvr1(2)) failed to bind VPg from either strain of TEV. This failure to bind correlated with resistance or reduced susceptibility, suggesting that interruption of the interaction between VPg and this eIF4E paralog may be necessary, but is not sufficient for potyvirus resistance in vivo. Among the three resistance alleles, only the pvr1 gene product failed to bind m7-GTP cap-analog columns, suggesting that disrupted cap binding is not required for potyvirus resistance.  相似文献

Lily mottle virus (LMoV), Potyvirus genus, is very difficult to purify for the preparation of diagnostic antisera. The coat protein (CP) gene of LMoV was amplified by RT-PCR from infected plants and cloned into the prokaryotic pET-30a vector to generate the recombinant plasmid pET-CP. The resulting carboxy-terminal His-tagged CP was over-expressed in Escherichia coli BL 21 cells by Isopropyl-β- d -thiogalactoside (IPTG) induction and purified over Ni-NTA affinity columns. The purified CP was used to elicit a polyclonal antiserum in rabbits. The antiserum had a titre of 1 : 128 in double diffusion tests, and specifically recognized LMoV in Western blots, Enzyme-Linked Immunosorbent Assays and Immuno-Electron Microscopy. The CP antiserum was also used to evaluate LMoV infection of lily bulbs used for commercial production in Yunnan, China. Substantial levels of infection were found in both imported and native bulbs, and provide the basis to implement flexible, rapid and large-scale virus indexing of lily plants for use in propagation and to meet virus-free quarantine regulations.  相似文献

A hitherto undescribed virus, termed rottboellia yellow mottle virus (RoYMV), affecting Rottboellia cochinchinensis (syn. R. exaltata, itch grass) at Ibadan, Nigeria, was investigated. RoYMV virions are isometric, c. 29 nm in diameter, and sediment homogeneously at 114s. In isopycnic CsCl gradients, RoYMV virions band sharply at a buoyant density of 1.379 g cm-3, but in Cs2SO4 gradients, virions band at two zones, at the densities of 1.300 g cm-3 and 1.325 g cm-3. Treatment with EDTA at pH 8.0 reduced the sedimentation value of RoYMV to c. 87s and rendered it susceptible to proteinase-K, SDS and NaCl. The apparent molecular weight of RoYMV coat protein was c. 27 000. Virions encapsidate a single-stranded RNA of mol. wt 1.4 × 106 Da. Besides R. cochinchinensis, RoYMV was mechanically transmissible only to maize (Zea mays). No reaction occurred when RoYMV was tested against antisera to 44 isometric plant viruses (belonging to 12 groups), including several that affect Gramineae. RoYMV exhibits striking similarities with other viruses belonging to the sobemovirus group, and it is tentatively designated as a new member of the sobemovirus group.  相似文献

A flexuous filamentous virus 750 nm in length was isolated from field-grown melons (Cucumis melo L.) and squirting cucumber (Ecballium elaterium L.). Infection of melon seedlings of most commercial varieties resulted in a lethal wilting, but late infections gave dieback. The virus infected systemically all economically important cucurbit crops. Its coat protein consists of a single polypeptide, 34 500 D in size, encapsidating a single stranded RNA genome of 10.1 kb. Antiserum against the Italian zucchini yellow fleck virus (ZYFV) reacted specifically with the Israeli isolate in both ISEM and Western blot. The virus was aphid transmissible in a non-persistent manner. Based on host range, host reaction and serological data, it is suggested that the virus described is an Israeli variant of ZYFV.  相似文献

Pea (Pisum sativum L.) is an important legume worldwide. The importance of pea in arable rotations and nutritional value for both human and animal consumption have fostered sustained production and different studies to improve agronomic traits of interest. Moreover, complete sequencing of the pea genome is currently underway and will lead to the identification of a large number of genes potentially associated with important agronomic traits. Because stable genetic transformation is laborious for pea, virus‐induced gene silencing (VIGS) appears as a powerful alternative technology for determining the function of unknown genes. In this work, we present a rapid and efficient viral inoculation method using DNA infectious plasmids of Bean pod mottle virus (BPMV)‐derived VIGS vector. Six pea genotypes with important genes controlling biotic and/or abiotic stresses were found susceptible to BPMV carrying a GFP reporter gene and showed fluorescence in both shoots and roots. In a second step, we investigated 37 additional pea genotypes and found that 30 were susceptible to BPMV and only 7 were resistant. The capacity of BPMV to induce silencing of endogenes was investigated in the most susceptible genotype using two visual reporter genes: PsPDS and PsKORRIGAN1 (PsKOR1) encoding PHYTOENE DESATURASE and a 1,4‐β‐D‐glucanase, respectively. The features of the ‘one‐step’ BPMV‐derived VIGS vector include (i) the ease of rub‐inoculation, without any need for biolistic or agro‐inoculation procedures, (ii) simple cost‐effective procedure and (iii) noninterference of viral symptoms with silencing. These features make BPMV the most adapted VIGS vector in pea to make low‐ to high‐throughput VIGS studies.  相似文献

Three pepper cultivars obtained from National Horticultural Research Institute (NIHORT) Idi-Ishin, Ibadan were intercropped with maize for two planting seasons between March and September in each year. These pepper cultivars were NHV1-D96, and NHV1-E96 and NHV1-F96. A 90-day maturing maize variety (DMSR-1) was used as the intercropping companion plant. The pepper seedlings were raised in a greenhouse. A randomised complete block design was used for this experiment. Each variety was intercropped with maize and replicated three times including the sole plot. The results obtained for each year were not significantly different from each other. There was a significant difference in pepper veinal mottle virus (PVMV) disease incidence and severity at a probability of less than 5% in the treatment used. PVMV disease incidence and severity was relatively higher in the sole pepper crop compared with pepper intercropped with maize. In the three varieties of peppers intercropped with maize, less than 17% disease incidence and 15% disease severity were recorded in all the varieties with a minimum yield of 4 tons per hectare compared with the sole pepper cropping of the same variety that recorded as high as 75% disease incidence and 72% disease severity with a maximum yield of 3.3 tons per hectare. There was a significant negative correlation at probability less than 0.05 between disease incidence, severity and the fruit yield of pepper. Variety NHV1-F96 in the maize intercrop recorded the highest yield of 15.99 tons/ha with a land equivalent ratio of 2.4 tons/ha. The success of the PVMV disease management evaluated in this study was judged by the extent of reduction in number of diseased plants and by an increase in vigor of the cultivated pepper crop, with an increase in fruit yield and quality. This signifies that for devising effective viral disease management for any crop it is important that the vectors of the virus present in the particular area are exactly controlled from having contact with the target plant. The reduction of pest incidence with intercropping of non-host plants should be carefully considered.  相似文献

Recombinant DNA technology was used to raise a polyclonal antiserum against the coat protein (CP) of Parietaria mottle virus (PMoV). The CP gene was expressed in Escherichia coli as a fusion to a 6xHis tag and purified by affinity chromatography. Recombinant purified protein was used as antigen to raise a polyclonal antiserum. This polyclonal antiserum consistently detected PMoV specifically infected tomato plants from different commercial tomato crops by indirect enzyme-linked immunosorbent assay (I-ELISA) and direct tissue-printing immunoassay (DTBIA).  相似文献

Helminth parasites were identified in a natural population of primates at Polonnaruwa, Sri Lanka. Five fatally wounded or recently deceased toque macaques (Macaca sinica) and three langurs (Presbytis senex andP. entellus) were autopsied. The following nematodes were found:Oesophagostomum aculeatum (Chabertiidae),Streptopharagus pigmentata (Spirocercidae),Physaloptera sp. (Spiruridae),Enterobius vermicularis (Oxyuridae), andTrichuris trichiura (Trichuridae). One cestode,Hymenolepis was also recovered fromP. entellus. Among fresh faecal samples of 210M. sinica, worm eggs ofOesophagostomum andStrongyloides were most abundant, followed byTrichostrongylus. Symptomless nature of Piper yellow mottle virus (PYMoV) infection in three varieties of black pepper (Piper nigrum) (Panniyur 1, Panniyur 5 and Panchami) and a wild species of Piper (Piper colubrinum) was confirmed by polymerase chain reaction (PCR) using PYMoV specific primers. The virus could be transmitted from these PYMoV-infected symptomless plants onto symptom producing black pepper cv. Karimunda through mealybug vector, Ferrisia virgata and by graft transmission. About 20–50% seedlings showed typical symptoms of the PYMoV at 30 days after mealybug inoculations while it was 75–94% at 90 days after inoculation. PCR test of the inoculated seedlings confirmed the presence of PYMoV in 50–64%, 76–100% and 80–100% of plants in 30, 60 and 90 days after inoculation, respectively. Similarly, 50–66%, 91–100% and 100% of graft-transmitted plants showed typical symptoms of the disease at 30, 60 and 90 days after grafting. PCR test of the graft-transmitted plants showed 100% PYMoV infection at 60 days after grafting. The results clearly demonstrated the existence of PYMoV-infected symptomless plants that can act as source for secondary spread of the virus in the field.  相似文献

Sri Lankan rice farmers rarely practise green manuring and they depend mostly on chemical fertilizers for the nutrient requirement of their crops. With the removal of government subsidy on fertilizers since 1990, they are now faced with the dilemma of meeting the cost of production to sustain previous yield targets. Therefore the present study devotes to evaluate alternative cheaper nutrient sources like green manures for rice. Results of recent work in Sri Lanka have shown that Sesbania speciosa Taub. ex Engl. is a promising green manure for lowland rice in the dry zone. It was evaluated to adopt a suitable green manuring system for rice. Its year-round biomass production, nutrient accumulation during the vegetative growth and nitrogen fixation were monitored. Field trials were conducted to investigate its ability to suppress weed growth when grown during fallow periods and its contribution to soil fertility and effect on rice yield. Highest biomass of around 100 g dry matter per plant, was achieved when S. speciosa was seeded in August and harvested at flowering in November. When this was planted during a fallow period at a spacing of 0.25×0.25 m (16 plants m-2), percentage weed reduction was as much as 40%. Its N2 fixation activity was similar to that of S. sesban (L.) Merr., Nitrogen concentration in S. speciosa increased until 70 days after establishment and then decreased towards flowering. Phosphorus and K accumulation was also effective during this period. Therefore suitable time for soil-incorporation of S. speciosa is around 70 days after establishment. S. speciosa-manured and chemically fertilized rice crops were comparable in terms of grain yield, indicating the effective nutrient supplying potential of this green manure. Straw biomass was significantly high under S. speciosa, because of the high N-allocation to vegetative growth of rice under this. This evaluation shows that S. speciosa not only supplies N, but is also an effective and complete supplier of other nutrients for rice. A scheme to introduce this into rice production system is proposed, where S. speciosa is seeded/broadcast two weeks before crop harvest (around January) in Maha (major) season as well as Yala (minor) season (around June) so that its biomass can be incorporated into soil during land preparation of subsequent seasons.  相似文献

From the characterization of the recessive resistance gene, sbm1, in pea we have identified the eukaryotic translation initiation factor, eIF4E, as a susceptibility factor required for infection with the Potyvirus, Pea seed-borne mosaic virus. A functional analysis of the mode of action of the product of the dominant allele revealed a novel function for eIF4E in its support for virus movement from cell-to-cell, in addition to its probable support for viral RNA translation, and hence replication. Different resistance specificities in two independent pea lines were explained by different mutations in eIF4E. On the modelled structure of eIF4E the coding changes were in both cases lying in and around the structural pocket involved in binding the 5'-m7G cap of eukaryotic mRNAs. Protein expression and cap-binding analysis showed that eIF4E encoded by a resistant plant could not bind to m7G-Sepharose, a result which may point to functional redundancy between eIF4E and the paralogous eIF(iso)4E in resistant peas. These observations, together with related findings for other potyvirus recessive resistances, provide a more complete picture of the potyvirus life cycle.  相似文献

Although many viruses have been crystallized and the protein capsid structures have been determined by x-ray crystallography, the nucleic acids often cannot be resolved. This is especially true for RNA viruses. The lack of information about the conformation of DNA/RNA greatly hinders our understanding of the assembly mechanism of various viruses. Here we combine a coarse-grain model and a Monte Carlo method to simulate the distribution of viral RNA inside the capsid of cowpea chlorotic mottle virus. Our results show that there is very strong interaction between the N-terminal residues of the capsid proteins, which are highly positive charged, and the viral RNA. Without these residues, the binding energy disfavors the binding of RNA by the capsid. The RNA forms a shell close to the capsid with the highest densities associated with the capsid dimers. These high-density regions are connected to each other in the shape of a continuous net of triangles. The overall icosahedral shape of the net overlaps with the capsid subunit icosahedral organization. Medium density of RNA is found under the pentamers of the capsid. These findings are consistent with experimental observations.  相似文献

A seroepidemiological study of arboviruses infecting 115 wild toque macaques (Macaca sinica) at Polonnaruwa, Sri Lanka showed a high prevalence of antibodies to dengue and Lumbo viruses. There was low seroprevalence of Chandipura (2/115) and Batai (1/115) virus antibodies, but no seropositivity to Chikungunya or Sindbis. There was no serological evidence of infection by Japanese encephalitis (JE) virus in spite of large human epidemics in the study area, indicating that toque macaques are unlikely to be an epidemiologically relevant host in the maintenance cycle of JE virus.  相似文献

The complete genome of a novel bipartite begomovirus (genus Begomovirus, family Geminiviridae) was cloned from a severely diseased yellow Peruvian chili pepper (Capsicum baccatum cv. Pendulum) plant collected in the department of La Libertad, Northern Peru and full‐length sequenced. The two genomic components share a common region of 156 nucleotides with a 100% sequence identity. Analysis of the genome organisation and phylogenetic comparisons revealed that the virus is a typical New World begomovirus. The closest related begomovirus, an isolate of Tomato yellow vein streak virus (ToYVSV), shared only 76.8% nucleotide sequence identity for the DNA‐A component. Therefore, following species demarcation criteria of the International Committee on Taxonomy of Viruses, this virus isolate belongs to a new begomovirus species for which the name pepper leafroll virus (PepLRV) is proposed. Pepper plants infected with the cloned PepLRV isolate developed leaf roll symptoms similar to those observed in field‐infected plants suggesting this virus as the causal agent of the disease syndrome observed in the field. Widespread occurrence of PepLRV throughout Peru was demonstrated, infecting plants of diverse cultivated species such as tomato, pepper, common and pallar beans, and of the weed species Nicandra physaloides. Low genetic diversity was observed among PepLRV isolates present in this country with no evident geographical or temporal structure of the population, typical of a recent founder effect. This is the first report of a begomovirus infecting pepper and bean crops in Peru.  相似文献

Spherical viruslike particles (VLP) were found in the tissues of apparently healthy tobacco thrips, Frankliniella fusca. The particles occurred in abundance in thrips from Ontario but were absent in thrips from Oklahoma reared under identical conditions. The VLP were not transmissible to any of the seven plant hosts (in four families) of F. fusca suggesting that they may be an insect virus. Transmission of tomato spotted wilt virus (TSWV) by F. fusca, a known vector, was not affected by the presence or absence of the VLP. No TSWV particles were detected in tissues of F. fusca that transmitted TSWV to test plants. The VLP occurred in several internal organs and hemocoele of the thrips and were isolated in vitro by preparing homogenates of gut tissues. Infection of oocytes and presence of VLP in young nymphs suggested transovarial transmission of the particles. The VLP measured 62 ± 4 nm in diameter and usually occurred in dense viroplasms in the cell cytoplasm. Development of the particles within the viroplasms is discussed.  相似文献

A mechanically transmissible virus was isolated from Bedford Giant blackberry plants showing chlorotic mottling and ringspot symptoms growing in Scotland. It infected several herbaceous test plants, many of them symptomlessly. This virus was also transmitted to several Rubus species and cultivars by graft inoculation with scions from the field‐infected Bedford Giant plant. Most grafted plants were infected symptomlessly, but Himalaya Giant blackberry and the hybrid berry Tayberry developed symptoms similar to those in the infected Bedford Giant plant. In the sap of infected Chenopodium quinoa, the virus lost infectivity when diluted 10?4 but not 10?3, after 6 h and 48 h when kept at 20°C and 4°C, respectively, but was infective for more than 8 days when kept at ?15°C. Preparations of purified virus from infected C. quinoa or spinach sedimented as three major nucleoprotein components and consisted of quasi‐isometric particles that varied in size from 24 to 32 nm in diameter and that were not penetrated by negative stain. Such virus particle preparations contained a major polypeptide of ca 28 kDa and three single‐stranded RNA species of estimated size 3.2, 2.8 and 2.1 kb. The complete sequence of the largest RNA (RNA 1, 3478 nt) and the partial sequence of the other RNAs (1863 and 2102 nt long, respectively) were determined and compared with sequences in databases. These findings, together with the biological and biochemical properties of this virus, indicate that it should be regarded as a distinct species in subgroup 1 of the genus Ilarvirus even though it was serologically unrelated to existing members of this subgroup. The virus showed a very distant serological relationship with prune dwarf virus (PDV) but differed significantly from it in the amino acid sequence of its coat protein, experimental host range and symptomatology and was unrelated to PDV at the molecular level. The virus, tentatively named blackberry chlorotic ringspot virus, is therefore a newly described virus and the first ilarvirus found naturally infecting Rubus in the UK.  相似文献
