Spectrophotometric data have been determined for mesoferrihaem at several pH values and over a range of concentration covering four orders of magnitude. The data reveal a dimerization process according to the equation 2 monomer in equilibrium dimer + H+, analogous to earlier findings for deuteroferrihaem and protoferrihaem. The value of K (defined as K = [dimer] [H+]/[monomer]2) was found to be 6.92.10(-2). This is close to the value for deuteroferrihaem but much less than that for protoferrihaem. This is interpreted in terms of possible additional bonding between the delocalized electron systems in protoferrihaem dimers relative to those of mesoferrihaem and deuteroferrihaem. Rate constants for dimerization were determined by temperature-jump spectrophotometry. The pH dependence of the rate constants is explained in terms of two distinct pathways for the dimerization process. These involve either direct reaction between two undissociated monomer molecules or alternatively an initial acid dissociation of a monomer molecule followed by reaction between an undissociated and dissociated molecule.  相似文献

The dimerization of deuteroferrihaem in aqueous solution has been investigated using a parameter, named the dimerization index (Robs). This is defined as the ratio of extinction coefficients at wavelengths corresponding to Soret band maxima for the monomeric and dimeric species, respectively. For solutions containing mainly monomeric species, Robs > 2, whereas for solutions containing mainly dimeric species Robs < 1.A computer programme has been applied to determine values of the dimerization constant, K, defined as: K = [dimer][H+][monomer]2.Phosphate buffer anions and Tris · HCl buffer enhanced dimerization. Monovalent and divalent cations also increased dimerization, but in a specific manner. The magnitudes of their effects increased in the order K+ < Na+ < Li+ < Sr2+ < Mg2+ ? Ca2+. Values of K were determined for several concentrations of Na+ and Sr2+. These data are interpreted in terms of a stabilization of the ferrihaem dimer by the formation of ion triplets with the added cation ‘sandwiched’ between carboxyl residues of the adjacent ferrihaem monomeric units. General guidelines are recommended for the choice of conditions which minimize dimerization.  相似文献

The dimerization of haematoferrihaem was studied in phosphate buffer in the pH range 7.02--8.14. The absorbance of dilute solutions decreased over a period of several hours due to adsorption of haematoferrihaem to glass vessels. This problem was overcome by using dilute solutions within 10 min of preparation. Spectrophotometric data were consistent with a dimerization process according to the equation 2 monomer in equilibrium dimer + H+ as found earlier for other ferrihaems studied. The value of K, defined as K = [dimer] [H+]/[monomer]2, was found to be 1.00 . 10(-2). Rate constants for the forward and reverse steps in dimerization were determined at pH values of 6.63, 7.01 and 7.44, using the temperature-jump technique. The reaction pathway for dimerization was found to be similar to those of deuteroferrihaem and mesoferrihaem, but different from that of coproferrihaem. A general appraisal of the factors influencing dimerization is attempted in the light of accumulated data on various ferrihaems. With the exception of protoferrihaem, it is suggested that dimerzation increases with the hydrophobicity of the 2,4 substituents. The additional stability of the protoferrihaem dimer is explained in terms of increased interaction due to conjugation of the vinyl groups with the porphyrin macrocycle.  相似文献

The equilibrium constant for [125I]β nerve growth factor was determined using polyacrylamide gel electrophoresis to separate the monomer and dimer. Various concentrations of the radiolabelled nerve growth factor were incubated for 24 and 48 hours. The equilibrium constants obtained for both incubation periods were the same, 3.2 ± 1.4 × 10?11M and 2.6 ± 1.6 × 10?11M, respectively. Thus, at physiological concentrations the β nerve growth factor is in the dimeric form almost exculsively.  相似文献

Rate and apparent equilibrium constants for the dissociation of pig liver carboxylesterase into three subunit molecules have been determined by complement fixation. The dependence of the dissociation equilibria on pH are consistent with dissociation reactions involving the addition of two protons per subunit, a pH-independent dissociation, and a dissociation upon the loss of one proton per subunit. The rate constants for dissociation are consistent with terms first order in hydrogen and hydroxide ions and a pH-independent path. The equilibrium constants in the range 3–35 °C at pH 7.2 exhibit no dependence on temperature; the association reaction is entropy driven with ΔS = 68 cal mol?1°K?1. The rate constants for the pH-independent dissociation follow ΔH ? 6 kcal mol?1. The order of effectiveness of concentrated salts in promoting denaturation is correlated with their effect on the activity coefficient of acetyltetraglycine ethyl ester and suggests that peptide groups become more exposed upon dissociation. The increased dissociation in the presence of urea derivatives containing alkyl substituents suggests exposure of hydrophobic regions upon dissociation; this is also consistent with ΔH = 0 for dissociation. It is likely that hydrophobic interactions contribute to the stability of the trimeric whole molecule.  相似文献

Acid dissociation constants of aqueous cyclohexaamylose (6-Cy) and cycloheptaamylose (7-Cy) have been determined at 10–47 and 25–55°C, respectively, by pH potentiometry. Standard enthalpies and entropies of dissociation derived from the temperature dependences of these pKa's are ΔH0 = 8.4 ± 0.3 kcal mol?1, ΔS0 = ?28. ± 1 cal mol?10K?1 for 6-Cy and ΔH0 = 10.0 ± 0.1 kcal mol?1, ΔS0 = ?22.4 ±0.3 cal mol?10K?1 for 7-Cy. Intrinsic 13C nmr resonance displacements of anionic 6- and 7-Cy were measured at 30°C in 5% D2O (vv). These results indicate that the dissociation of 6- and 7-Cy involves both C2 and C3 20-hydroxyl groups. The thermodynamic and nmr parameters are discussed in terms of interglucosyl hydrogen bonding.  相似文献

A non-alkalophilic mutant strain of Bacillusalcalophilus grows on L-malate over a pH range from 5.0 to 9.0. The mutant does not exhibit the energy-dependent efflux of Na+ that has been used to assay a Na+H+ antiporter in the wild type organism. The mutant also fails to transport α-aminoisobutyric acid, at pH 9.0, either in the presence or absence of Na+; at pH 5.5, the amino acid analogue is taken up by a Na+-independent mechanism. The properties of the mutant constitute strong evidence that the Na+H+ antiporter is involved in maintaining an acidified cytoplasm in B. alcalophilus.  相似文献

(1) A (K+ + H+)-ATPase containing membrane fraction, isolated from pig gastric mucosa, has been further purified by means of zonal electrophoresis, leading to a 20% increase in specific activity and an increase in ratio of (K+ + H+)-ATPase to basal Mg2+-ATPase activity from 9 to 20. (2) The target size of (Na+ + K+)-ATPase, determined by radiation inactivation analysis, is 332 kDa, in excellent agreement with the earlier value of 327 kDa obtained from the subunit composition and subunit molecular weights. This shows that the Kepner-Macey factor of 6.4·1011 is valid for membrane-bound ATPases. (3) The target size of (K+ + H+)-ATPase is 444 kDa, which, in connection with a subunit molecular weight of 110000, suggests a tetrameric assembly of the native enzyme. The ouabain-insensitive K+-stimulated p-nitrophenylphosphatase activity has a target size of 295 kDa. (4) In the presence of added Mg2+ the target sizes of the (K+ + H+)-ATPase and its phosphatase activity are decreased by about 15%, while that for the (Na+ + K+)-ATPase is not significantly changed. This observation is discussed in terms of a Mg2+-induced tightening of the subunits composing the (K+ + H+)-ATPase molecule.  相似文献

Using guanidinium and n-butylammonium cations (C+) as models for the positively charged side chains in arginine and lysine, we have determined the association constants with various oxyanions by potentiometric titration. For a dibasic acid, H2A, three association complexes may exist: K1M = [CHA][C+] [HA?]; K1D = [CA?][C+] [A2?]; K2D = [C2A][C+] [CA?]. For guanidinium ion and phosphate, K1M = 1.4, K1D = 2.6, and K2D = 5.1. The data for carboxylates indicate that the basicity of the oxyanion does not affect the association constant: acetate, pKa = 4.8, K1M = 0.37; formate, pKa = 3.8, K1M = 0.32; and chloroacetate, pKa = 2.9, K1M = 0.43, all with guanidinium ion. Association constants are also reported for carbonate, dimethylphosphinate, benzylphosphonate, and adenylate anions.  相似文献

The rate of reaction of [Cr(III)Y]aq (Y is EDTA anion) with hydrogen peroxide was studied in aqueous nitrate media [μ = 0.10 M (KNO3)] at various temperatures. The general rate equation, Rate = k1 + k2K1[H+]?11 + K1[H+]?1 [Cr(III)Y]aq[H2O2] holds over the pH range 5–9. The decomposition reaction of H2O2 is believed to proceed via two pathways where both the aquo and hydroxo-quinquedentate EDTA complexes are acting as the catalyst centres. Substitution-controlled mechanisms are suggested and the values of the second-order rate constants k1 and k2 were found to be 1.75 × 10?2 M?1 s?1 and 0.174 M?1 s?1 at 303 K respectively, where k2 is the rate constant for the aquo species and k2 is that for the hydroxo complex. The respective activation enthalpies (ΔH*1 = 58.9 and ΔH*2 = 66.5 KJ mol?1) and activation entropies (ΔS*1 = ?85 and ΔS*2 = ?40 J mol?1 deg?1) were calculated from a least-squares fit to the Eyring plot. The ionisation constant pK1, was inferred from the kinetic data at 303 K to be 7.22. Beyond pH 9, the reaction is markedly retarded and ceases completely at pH ? 11. This inhibition was attributed in part to the continuous loss of the catalyst as a result of the simultaneous oxidation of Cr(III) to Cr(VI).  相似文献

The catalytic decomposition of H2O2 by deuteroferrihaem, mesoferrihaem, coproferrihaem and haematoferrihaem was studied as a model for the mechanism of action of catalase. For haematoferrihaem, anomalous but reproducible results were obtained, which could not be adequately explained. For each of the other ferrihaems studied, both monomeric and dimeric species catalysed decomposition, although the activity of monomer (aM) was much greater than that of dimer (aD). The pH variation of aD in the range 6.5--11 was consistent with an inverse dependence on [H+]1/2. The molecular mechanism whereby such a dependence could be achieved is not apparent. A study of the pH-dependence of aM in the range 6.5--11 revealed a linear inverse relationship with [H+]. This is interpreted in terms of attack by HO2- on ferrihaem monomer. The specific pH-independent rate constants for this reaction were in the order coproferrihaem greater than protoferrihaem greater than or equal to mesoferrihaem congruent to deuteroferrihaem. The order of magnitude of these rate constants is the same as that for catalysis by Fe(H2O)63+ and the second-order rate constant for decomposition of H2O2 by catalase. The implications on the mechanism of action of catalase are discussed.  相似文献

The bimolecular decay rates (2k) of the flavosemiquinones (FH·F?) of riboflavin, FMN and FAD have been determined using pulse radiolysis. The rates (defined as d[FH·F?]dt = ?2k[FH·F?]2) for the neutral flavosemiquinones at zero ionic strength and pH 5.9 are (in units of mol?1·dm3·s?1): (1.2 ± 0.1)·109, (5.0 ± 0.2)·108 and (1.4 ± 0.1)·108; and for the anionic flavosemiquinones at pH 11.2 (5.4 ± 0.9)·108, (4.5 ± 0.3)·107 and (8.5 ± 1.3)·106, respectively. The kinetic salt effect has been used to formulate rate equations for each flavin to adjust for ionic strength effects.  相似文献

13C nuclear magnetic resonance (n.m.r.) spectral data for 13C reductively methylated N-terminal tryptic glycopeptides and for 13C reductively methylated N-terminal glyco-octapeptides derived from homozygous glycophorins AM and AN are presented. Their 13C chemical shift data are compared with the previously published 13C n.m.r. data for 13C reductively methylated homozygous glycophorins AM and AN in order to investigate the means of display of the MN blood determinants by these species. The pH dependence of the 13C resonances of Nα,N-[13C]dimethyl leucine of glyco-octapeptide AN and of Nα,N-[13C]dimethyl serine of glyco-octapepti AM indicated that only a slight structural perturbation occurs at the N-terminus when a large portion of the glycoprotein molecule is removed. However, one structural ‘state’ of 13C reductively methylated glycophorin AM is lost when the glyco-octapeptide AM is produced. The 13C resonance of Nα,N-[13C]dimethyl leucine of glycooctapeptide AN titrated with a pKa of 7.7 (Hill coefficient ~ 1). The 13C resonance of Nα,N-[13C]dimethyl serine, on the other hand, exhibited an unusual pH dependence, indicating the existence of some possible steric constraints or hydrogen bonding in this molecule. In comparison to the data obtained for 13C-labelled glycooctapeptide AM molecule, the pH dependence of the chemical shift of the 13C resonance of Nα,N-[13C]dimethyl serine of tripeptide tri-L-serine is also presented. Circular dichroism (c.d.) spectra indicated that the reductive methylation technique does not cause a large perturbation of the glycophorin A molecule.  相似文献

The dependence of the mitochondrial respiratory rate on the reduction of cytochrome c has been measured as a function of the exogenous [ATP][ADP][Pi] ratio and pH. The respiratory rate at [ADP][ADP][Pi] values of less than 10-1m-1 is proportional to the reduction of cytochrome c and independent of pH from pH 6.5 to pH 8.O. The maximal turnover number (at 100% reduction) for cytochrome c is approximately 70 s?1. As the [ATP][ADP][Pi] ratio is increased from 10?1m?1 to 104m?1, the respiration at any given level of reduction of cytochrome c is progressively inhibited. Greater inhibition is observed at more oxidized levels of cytochorme c with respiratory control values for oxidation of reduced cytochrome c exceeding 10. The behavior of mitochondrial respiratory control is shown to be quantitatively consistent with a proposed mechanism in which the regulation occurs in the reaction of oxygen with cytochrome oxidase. A steady-state rate expression is derived which fits the mitochondrial respiratory rate dependence on (i) the extramitochondrial [ATP][ADP][Pi] ratio; (ii) the level of reduction of cytochrome c (or the intramitochondrial [NAD+][NADH]) at different [ATP][ADP][Pi] values; (iii) the pH of the suspending medium. This rate expression appears to correctly predict the relationships of the cytoplasmic [ATP][ADP][Pi] ratio, the mitochondrial [NAD+][NADH] ratio, and the mitochondrial respiratory rate in intact cells as well as suspensions of isolated mitochondria.  相似文献

(1) H+/electron acceptor ratios have been determined with the oxidant pulse method for cells of denitrifying Paracoccus denitrificans oxidizing endogenous substrates during reduction of O2, NO?2 or N2O. Under optimal H+-translocation conditions, the ratios H+O, H+N2O, H+NO?2 for reduction to N2 and H+NO?2 for reduction to N2O were 6.0–6.3, 4.02, 5.79 and 3.37, respectively. (2) With ascorbate/N,N,N′,N′-tetramethyl-p-phenylenediamine as exogenous substrate, addition of NO?2 or N2O to an anaerobic cell suspension resulted in rapid alkalinization of the outer bulk medium. H+N2O, H+NO?2 for reduction to N2 and H+NO?2 for reduction to N2O were ?0.84, ?2.33 and ?1.90, respectively. (3) The H+oxidant ratios, mentioned in item 2, were not altered in the presence of valinomycinK+ and the triphenylmethylphosphonium cation. (4) A simplified scheme of electron transport to O2, NO?2 and N2O is presented which shows a periplasmic orientation of the nitrite reductase as well as the nitrous oxide reductase. Electrons destined for NO?2, N2O or O2 pass two H+-translocating sites. The H+electron acceptor ratios predicted by this scheme are in good agreement with the experimental values.  相似文献

ADP and Pi-loaded membrane vesicles from l-malate-grown Bacillus alcalophilus synthesized ATP upon energization with ascorbateN,N,N′,N′-tetramethyl-p-phenylenediamine. ATP synthesis occurred over a range of external pH from 6.0 to 11.0, under conditions in which the total protonmotive force Δ\?gmH+ was as low as ?30 mV. The phosphate potentials (ΔGp) were calculated to be 11 and 12 kcal/mol at pH 10.5 and 9.0, respectively, whereas the Δ\?gmH+ values in vesicles at these two pH values were quite different (?40 ± 20 mV at pH 10.5 and ?125 ± 20 mV at pH 9.0). ATP synthesis was inhibited by KCN, gramicidin, and by N,N′-dicyclohexylcarbodiimide. Inward translocation of protons, concomitant with ATP synthesis, was demonstrated using direct pH monitoring and fluorescence methods. No dependence upon the presence of Na+ or K+ was found. Thus, ATP synthesis in B. alcalophilus appears to involve a proton-translocating ATPase which functions at low Δ\?gmH+.  相似文献

The interaction of lanthanides and other cations with phosphatidylcholine bilayers present as single bilayer vesicles in 2H2O has been investigated in terms of stoichiometry, apparent binding constants and environmental conditions.Lanthanides are shown to form 2 : 1 (molar ratio) phosphatidylcholine to metal ion complexes.The apparent binding constant Kb varies as a function of the quantity of metal ion bound and as a function of the Cl? concentration. The apparent binding constant at “zero loading” is K0 = 1.25 · 104L2 · M?at 0.15 M KCl. It decreases exponentially with increased “loading” expressed as the molar ratio of metal ion bound to effective phosphatidylcholine concentration and increases exponential with Cl? concentration.The interaction of lanthanides and divalent cations such as Ca2+ and Mg2+ is independent of pH in the pH range 3–7+ and 3–10 respectively, but is sensitive to the nature of the anion. The presence of anions enhances the interaction with polyvalent cations, the chaotropic anions showing the largest effect. The order of enhancement is Cl? < Br? < NO3? < SCN? < I? < ClO4?. The nature of the monovalent counterion (cation) has little effect on the enhanced binding of lanthanides in the presence of the above anions.The affinity of other polyvalent cations for phosphatidylcholine bilayers has been determined by competition with lanthanides. The physiologically important divalent cations Ca2+ and Mg2+ both bind less strongly (by about an order of magnitude) to the lipid surface. The order of binding of cations reflects direct binding to the phosphodiester group, with UO22+ showing the highest affinity.  相似文献

The observed equilibrium constants (Kobs) for the l-phosphoserine phosphatase reaction [EC 3.1.3.3] have been determined under physiological conditions of temperature (38 °C) and ionic strength (0.25 m) and physiological ranges of pH and free [Mg2+]. Using Σ and square brackets to indicate total concentrations Kobs = Σ L-serine][Σ Pi]Σ L-phosphoserine]H2O], K = L-H · serine±]HPO42?][L-H · phosphoserine2?]H2O]. The value of Kobs has been found to be relatively sensitive to pH. At 38 °C, K+] = 0.2 m and free [Mg2+] = 0; Kobs = 80.6 m at pH 6.5, 52.7 m at pH 7.0 [ΔGobs0 = ?10.2 kJ/mol (?2.45 kcal/mol)], and 44.0 m at pH 8.0 ([H2O] = 1). The effect of the free [Mg2+] on Kobs was relatively slight; at pH 7.0 ([K+] = 0.2 m) Kobs = 52.0 m at free [Mg2+] = 10?3, m and 47.8 m at free [Mg2+] = 10?2, m. Kobs was insignificantly affected by variations in ionic strength (0.12–1.0 m) or temperature (4–43 °C) at pH 7.0. The value of K at 38 °C and I = 0.25 m has been calculated to be 34.2 ± 0.5 m [ΔGobs0 = ?9.12 kJ/mol (?2.18 kcal/ mol)]([H2O] = 1). The K for the phosphoserine phosphatase reaction has been combined with the K for the reaction of inorganic pyrophosphatase [EC 3.6.1.1] previously estimated under the same physiological conditions to calculate a value of 2.04 × 104, m [ΔGobs0 = ?28.0 kJ/mol (?6.69 kcal/mol)] for the K of the pyrophosphate:l-serine phosphotransferase [EC 2.7.1.80] reaction. Kobs = [Σ L-serine][Σ Pi][Σ L-phosphoserine][H2O], K = [L-H · serine±]HPO42?][L-H · phosphoserine2?]H2O. Values of Kobs for this reaction at 38 °C, pH 7.0, and I = 0.25 m are very sensitive to the free [Mg2+], being calculated to be 668 [ΔGobs0 = ?16.8 kJ/mol (?4.02 kcal/mol)] at free [Mg2+] = 0; 111 [ΔGobs0 = ?12.2 kJ/mol (?2.91 kcal/mol)] at free [Mg2+] = 10?3, m; and 9.1 [ΔGobs0 = ?5.7 kJ/mol (?1.4 kcal/mol) at free [Mg2+] = 10?2, m). Kobs for this reaction is also sensitive to pH. At pH 8.0 the corresponding values of Kobs are 4000 [ΔGobs0 = ?21.4 kJ/mol (?5.12 kcal/mol)] at free [Mg2+] = 0; and 97.4 [ΔGobs0 = ?11.8 kJ/ mol (?2.83 kcal/mol)] at free [Mg2+] = 10?3, m. Combining Kobs for the l-phosphoserine phosphatase reaction with Kobs for the reactions of d-3-phosphoglycerate dehydrogenase [EC 1.1.1.95] and l-phosphoserine aminotransferase [EC 2.6.1.52] previously determined under the same physiological conditions has allowed the calculation of Kobs for the overall biosynthesis of l-serine from d-3-phosphoglycerate. Kobs = [Σ L-serine][Σ NADH][Σ Pi][Σ α-ketoglutarate][Σ d-3-phosphoglycerate][Σ NAD+][Σ L-glutamat0] The value of Kobs for these combined reactions at 38 °C, pH 7.0, and I = 0.25 m (K+ as the monovalent cation) is 1.34 × 10?2, m at free [Mg2+] = 0 and 1.27 × 10?2, m at free [Mg2+] = 10?3, m.  相似文献

Specific binding of human β-endorphin to rabbit cerebellar and brain membranes was measured using [3H2-Tyr27]-βh-endorphin as the primary ligand. In both tissues binding was time dependent and saturable, with apparent equilibrium dissociation constants of 0.275 nM and 0.449 nM in the cerebellum and brain, respectively. The binding capacity of cerebellum is greater than that of brain. Kinetic studies showed that the association rate constants were 2.7 × 107 M?1min?1 for cerebellum and 2.4 × 107 M?1min?1 for brain. Dissociation of tritiated βh-endorphin from both cerebellum and brain is not consistent with a first order decay from a single site. In the cerebellum, these is a time-dependent increase in slowly dissociating complex. The potency of several opioid peptides and opiates to inhibit the binding of tritiated βh-endorphin was determined. Ligands with preference for μ, δ, and κ opiate receptor (morphine, Metenkephalin and ethylketocyclazocine) all have similar affinities toward βh-endorphin sites in both brain and cerebellar membranes.  相似文献

Depletion of mitochondrial divalent cations by addition of the ionophore A23187 results in a marked increase in passive 42K+K+ exchange activity. The exchange is activated by increasing pH and temperature and inhibited by added divalent cations. The reaction is independent of the amount of A23187 present, but depends on the concentration of external K+ (Km = 25 mm). Intramitochondrial 42K+ in cation-depleted mitochondria exchanges passively with external Na+ and Li+, but not with choline+. The evidence suggests that removal of mitochondrial divalent cations by A23187 activates the endogenous K+H+ exchange component of the mitochondrion and that the activated exchanger promotes cation/cation exchange in the absence of a metabolic pH gradient.  相似文献
