The experiments reported here attempted to examine in two groups of rats the effects on the taste preferences, food and fluid intake, energy balance and body weight gain of corticocerebellar lesions involving, primarily, the Lobulus VI (LVI) or the Lobulus Paramedianus (LP). The results showed that the lesions of LVI or LP did not affect the daily intake of total fluid and salty solution. The intake of sweet solution increased in both groups of lesioned rats, while the intake of deionized water and acid and bitter solutions decreased only in the LVI lesioned rats. Food intake decreased in the LVI-lesioned rats but not in the LP-lesioned animals. Body weight gain, efficiency of food utilization, caloric intake and body surface gain decreased in both groups. It seems therefore that the cerebellar cortex, which probably receives taste fibers, somehow influences taste preferences and water intake, and that it may be involved in the mechanisms of food intake, its utilization and body energy balance.  相似文献

Progesterone and LH concentrations were measured in the plasma of blood samples taken from forty-eight pregnant ewes on Days 100, 120 and 134 of gestation. The ewes, in two groups of twenty-four were maintained from Day 100 until parturition on two planes of nutrition which supplied daily energy and protein intakes of about 4-1 or 2-3 Mcal metabolizable energy and either 192 or 111 g digestible crude protein per ewe. Within the groups, the ewes carried one, two or three fetuses and the feed intake was adjusted according to litter size to produce a uniform nutritional state within the group. On Day 100, litter size affected the concentration of plasma progesterone (P less than 0-001), but had no effect on Days 120 or 134 when the ewes were fed according to litter size. The low feed intake however caused a significant increase in plasma progesterone concentrations. The LH concentrations showed no major changes during late pregnancy and no effect of nutrition or little size on the plasma hormone concentration was observed. It was concluded that the effect of litter size on plasma progesterone concentration recorded on Day 100 or gestation was partly mediated by level of nutrition.  相似文献

The significance of the increased progesterone plasma levels after day 21 on the onset of puberty in female rats is unclear. To analyze this question, female rats were injected from day 21 to 32 with the potent antiprogestagen RU486 (1 mg/day) or vehicle. Treatment with RU486 did not modify either the age at which vaginal opening or first estrus occurs. Vaginal cycles were regular. Animals sacrificed on day 40 or 50 showed reduced uterus weight, whereas body, pituitary, adrenal and ovaries were not affected. These results evidence that antagonization of progesterone action during the juvenile phase did not affect the onset of puberty.  相似文献

The minimum progesterone concentration required to maintain the pregnancy was studied by varying doses of progesterone given subcutaneously to rats ovariectomized on Day 8 of pregnancy. Injecting 3 mg progesterone plus 200 ng oestradiol benzoate daily provided serum progesterone values between 25.4 +/- 7.0 and 35.2 +/- 6.2 ng/ml throughout Days 10-19 which were significantly lower than normal levels (P less than 0.05), but resulted in 93.6% of fetal survival on Day 19 which was not significantly different from 93.3% in the control group. Injecting 2 mg progesterone plus 200 ng oestradiol benzoate daily gave progesterone values between 13.2 +/- 4.6 and 19.0 +/- 6.2 ng/ml and could not maintain fetal viability to Day 19 (14.2%, P less than 0.05 compared with control group). Critical times to supplement progesterone in rats ovariectomized on Day 8 or Day 15 were studied by varying the time of progesterone implantation after ovariectomy. Progesterone implants were administered 8, 12 and 24 h after ovariectomy on Day 8 and 24, 36 and 48 h after ovariectomy on Day 15. On Day 8, progesterone replacement could be delayed to 8 h but not 12 h, while on Day 15, progesterone replacement could be delayed up to 36 h but not 48 h after ovariectomy without affecting fetal survival.  相似文献

Rate of decline in plasma progesterone concentration may influence the success of lactogenesis in the sow. The aim of this experiment was to investigate whether progesterone concentration and rate of decline of progesterone in the periparturient sow could be manipulated by changing her feeding level. Forty-two sows received either 1.15 or 2 times maintenance energy daily from day 100 of gestation up until and including the day of farrowing. Blood samples were taken on days 98 (pre-treatment baseline) and 109 of gestation, during farrowing, 6h after farrowing and at 09:00 h for the 3 days following farrowing. Plasma progesterone concentration was determined and progesterone half-life was calculated for each sow. High intake feeding had no effect on plasma progesterone concentration at any time of sampling. Progesterone half-life averaged 41.2 +/- 3.81 h and did not differ between treatments. There was no relationship between progesterone concentration, or half-life, and litter weight gain, although there was a weak correlation between decline in progesterone in the first 6h after birth and piglet growth rate from birth to 6 days of age (R(2) = 0.109, P < 0.05). It was concluded that increasing feed intake in late gestation cannot be used to increase progesterone clearance rate and hasten the onset of lactogenesis in sows.  相似文献

This study was designed to test the hypothesis that the loss of LH surges in response to the stimulatory actions of estradiol and progesterone in middle-aged, persistent-estrous (PE) rats may be caused by chronic elevations in circulating estradiol. Five groups of regularly cycling young rats received an s.c. estradiol implant immediately after ovariectomy (Day 0). For determination of LH surges, blood samples were collected hourly between 1200-1900 h from each of the five groups at one of the following times: 3 days, or 1, 2, 4, or 8 wk later. On the next day, either progesterone (0.5 mg/100 g BW) or corn oil was injected s.c. at 1200 h, and samples were obtained as before. Incidence and amplitude of estradiol-induced LH surges decreased during the first 2 wk of estradiol treatment, after which no surges occurred. Progesterone enhanced the incidence and amplitude of estradiol-induced LH surges thus delaying their disappearance. These results support our hypothesis and demonstrate that the stimulatory actions of estradiol and progesterone on the LH surge sequentially diminish with time after exposure to estradiol in young rats. Thus, young rats chronically treated with estradiol may be a useful model for studying the mechanisms whereby LH surges are abolished in middle age during the hyperestrogenic state of PE.  相似文献

The activity of 20alpha-hydroxysteroid dehydrogenase (20alpha-HSD) was assayed in the ovaries of rats after accelerated lactation to determine its relationship to the decrease in progesteron secretion that occurs. When rats were sjbjected to accelerated lactation on Day 9 of pregnancy, activity of the enzyme was only slightly increased by Day 10, but had risen to twice the control level by Day 11, and three times the control level by Day 12. Administration of LH or progesterone prevented the increase in enzyme activity. Progesterone concentration had decreased considerably before the time at which any significant increase in 20alpha-HSD activity was detected. These findings are discussed in relation to the role of 20alpha-HSF in regulating progesterone levels in the rat.  相似文献

Neurohomonal control of prolactin release was studied in pseudopregnant and pregnant rats. Nembutal administered at 1300 hours on Day 3 of pseudopregnancy prevented prolactin release which normally occurred at 1700 hours of the same day. Antiestrogen administered the day before did not prevent prolactin release but ovariectomy did. Estrogen administered immediately after ovariectomy did not restore prolactin secretion; however, progesterone on Day 3 in the ovariectomized-estrogen treated induced an increase in prolactin at 1700 hours. Progesterone was capable of increasing prolactin release the first 5 days of pseudopregnancy but not Days 6-12 when prolactin values were low. A similar effect was seen the first 7 days of pregnancy. Progesterone, but not estrogen, modified prolactin values on Day 9 at 1700 hours. Ovariectomy on Day 19 of pregnancy induced prolactin release within 4 hours and persisted for 58 hours. Progesterone administration immediately after ovariectomy prevented prolactin release for a few hours. These results suggest that the regulation of prolactin release by the central nervous system depends on the circulating estrogen/progesterone ratio, since estrogen facilitated prolactin release when plasma progesterone was low and progesterone induced prolactin release when adequated levels of estrogen existed, but exerted an inhibitory action when estrogen was not present.  相似文献

Treatment of immature female rats with 100 micrograms 2-bromo-alpha-ergocryptine mesylate (CB-154) per ml drinking water beginning on Day 30 of age until vaginal opening delayed puberty by 6 days. Rats treated with CB-154 exhibited vaginal opening at 43.3 +/- 0.6 days whereas controls exhibited vaginal opening at 37.9 +/- 0.8 days. Most interestingly, serum levels of luteinizing hormone (LH) and prolactin (PRL) on Days 31-35, determined by a homologous radioimmunoassay were significantly lower in treated rats than in controls. The ovarian concentrations of progesterone (P) and androstenedione (A) were lower in rats treated with CB-154 than in controls; ovarian estradiol (E2) concentrations were low in both groups. Serum levels of P (but not A and E2) were reduced on Days 31-35 of the treatment period. Cessation of the CB-154 treatment on the morning of Day 35 returned the onset of puberty to normal values; steroid and gonadotropin levels also returned to normal values within 2 days after removal of the CB-154 from the drinking water. Near the time of onset of puberty, serum levels of LH in rats treated with CB-154 returned to control values. These data indicate that in the female rat the delay in puberty induced by CB-154 might be due to a reduction in the secretion of LH, especially since the onset of delayed puberty in rats treated with CB-154 correlates with an increase in the serum level of LH. Further studies are needed to elucidate the specific effects of hypoprolactinemia on ovarian function and the onset of puberty in the rat.  相似文献

In Exp. 1, PMSG was injected to 26-day-old prepubertal rats to induce ovulations. On Day 2 (2 days later, the equivalent of the day of pro-oestrus) they received at 08:00 h 5 mg hydroxyflutamide or vehicle and at 12:00 h 2 mg progesterone or testosterone or vehicle. Animals were killed at 18:00 h on Day 2 or at 09:00 h on Day 3. Progesterone but not testosterone restored the preovulatory LH surge and ovulation in hydroxyflutamide-treated rats. In Exp. 2, 2 mg progesterone or testosterone were injected between 10:30 and 11:00 h on Day 2, to advance the pro-oestrous LH surge and ovulation in PMSG-primed prepubertal rats. Injection of hydroxyflutamide abolished the ability of progesterone to advance the LH surge or ovulation. Testosterone did not induce the advancement of LH surge or ovulation. In Exp. 3, ovariectomized prepubertal rats implanted with oestradiol-17 beta showed significantly (P less than 0.01) elevated serum LH concentrations at 18:00 h over those observed at 10:00 h. Progesterone injection to these animals further elevated the serum LH concentrations at 18:00 h, in a dose-dependent manner, with maximal values resulting from 1 mg progesterone. Hydroxyflutamide treatment significantly (P less than 0.003) reduced the serum LH values in rats receiving 0-1 mg progesterone but 2 mg progesterone were able to overcome this inhibition. It is concluded that progesterone but not testosterone can reverse the effects of hydroxyflutamide on the preovulatory LH surge and ovulation. It appears that hydroxyflutamide may interfere with progesterone action in induction of the LH surge, suggesting a hitherto undescribed anti-progestagenic action of hydroxyflutamide.  相似文献

Chronically ovariectomized ewes were pretreated with progesterone and oestradiol to induce oestrus and randomly allocated into four treatment groups. Progesterone injections were given to Groups 1 and 2 on Days 1–12 and Groups 3 and 4 on Days 1–15. Ewes in Groups 2 and 4 were infused with conceptus secretory proteins (oCSP), via an intrauterine catheter, twice daily on Days 13–15. Ewes in Groups 1 and 3 were similarly infused, but with serum proteins (oSP). Endometrial oxytocin receptor (OTr) concentrations and oxytocin-induced 13,14-dihydro-15-keto-prostaglandin F (PGFM) release were measured on Day 16.Progesterone concentrations in ewes receiving 12 days of progesterone treatment declined after Day 12, reaching a nadir on Day 14. In contrast, plasma progesterone concentrations remained elevated until Day 16 in ewes receiving the extended progesterone treatment. On Day 16, endometrial OTr concentrations were significantly higher in ewes given 12 days of progesterone treatment than in ewes given 15 days of progesterone irrespective of the presence of oCSP or oSP. Treatment with oCSP significantly decreased oxytocin-induced PGFM release in ewes given 12 days of progesterone treatment compared with those ewes receiving oSP infusions. The extended 15 day progesterone treatment resulted in a further decrease in oxytocin-induced PGFM release in both oCSP and oSP infused ewes.These data indicate that, in steroid treated ovariectomized ewes, intrauterine infusion of oCSP will reduce oxytocin-induced PGFM response but not OTr concentrations. Progesterone appears to play a dominant role in the regulation of OTr as well as oxytocin-induced PGFM release.  相似文献

The influences of ovarian hormones on food intake, taste preferences, and glucoregulation were examined in intact and ovariectomized rhesus monkeys. Intake of intact monkeys was lowest in the preovulatory stage of the cycle, when estrogen levels are elevated, and exogenous estradiol transiently suppressed food intake of ovariectomized monkeys in a dose-related manner, confirming previous observations. Progesterone treatment did not affect food intake when given alone, but it did attenuate the effect of estradiol when both hormones were given concurrently. Preferences for sweet solutions were not detectably influenced by chronic treatment with estradiol or progesterone, and compensatory responses to ingestion of sugar were unaltered by hormone treatment. Glucose tolerance tests did not reveal significant influences of ovarian hormones on glycemia, but insulin levels were elevated during periods of progesterone treatment. These results do not support the suggestion that fluctuations in caloric intake during the menstrual cycle are secondary to changes in taste preference or glucoregulation. However, possible changes in sweet preference and glucoregulation shortly after initiation of estrogen treatment, i.e., during the time of suppressed food intake, remain to be examined.  相似文献

Blood samples were collected for the measurement of progesterone concentrations from 320 Holstein-Friesian heifers on Days 7 and 21 post-estrus. All animals were the recipients of either a fresh or previously frozen embryo on Day 7 and were palpated for pregnancy on Day 60 post-estrus. At the time of transer, progesterone levels were highly variable and were not strongly related to subsequent pregnancy status. There was a tendency for lower pregnancy rates in heifers receiving fresh embryos if progesterone levels were less than 1 ng/ml (33 vs 64% overall), and for previously frozen embryos when progesterone concentrations were less than 3 ng/ml (34 vs 44% overall). Progesterone concentrations were not related to subjective evaluation of corpus luteum quality by palpation per rectum. No heifers which maintained pregnancy had progesterone levels less than 1 ng/ml on Day 21. Only 41% of 247 heifers receiving either fresh or previously frozen embryos that were not pregnant on Day 60 had progesterone concentrations less than 1 ng/ml on Day 21. These data suggest that many recipients that do not maintain a pregnancy will experience an extended estrous cycle after transfer.  相似文献

Conceptus number was reduced to one on Day 7 of pregnancy in rats by aspirating all but a single conceptus (Group E) or left at greater than or equal to 8 conceptuses (Group C). In Group E rats, serum progesterone concentrations remained low from Day 12 until autopsy at Day 21. Hypophysectomy on Day 12 significantly increased serum progesterone values after Day 17 of pregnancy, and these increases were blocked by treatment with ACTH (10 U/day, i.p., Days 12-17). Adrenalectomy on Day 12 also induced slight, but statistically significant, increases in serum progesterone concentration after Day 17, and these were overcome by implantation of a 10 mg capsule of corticosterone. In Group C rats, hypophysectomy or adrenalectomy on Day 12 did not change serum progesterone concentrations, but 40 U ACTH/day inhibited progesterone secretion. We conclude from these results that the pituitary-adrenal system exerts inhibitory effects on progesterone secretion during mid-pregnancy in rats.  相似文献

The development of the adrenal corticosterone and progesterone diurnal rhythms was retarded by a single injection of cortisol acetate, dexamethasone acetate or progesterone into 3-day-old female rats. This effect was transient with the establishment of the corticosterone rhythm by day 70. The retardation of adrenal hormone rhythmicity may alter the normal process of sexual maturation, because cortisol and dexamethasone-treated animals but not progesterone treated rats exhibited a desynchronization of the events associated with puberty. Vaginal opening was delayed, and dissociated from first ovulation in these rats. Further, neonatal cortisol administration lengthened the estrous cycle in adulthood. It can be concluded from our study that the maturation of the neural centers controlling adrenal steroid secretion may be essential for the proper timing of pubertal events in the female rat.  相似文献

Adult male rats have been demonstrated to increase food intake in response to administration of drugs that interfere with oxidation of fatty acids (e.g. methyl palmoxirate and mercaptoacetate [MA]), effects that are larger in animals maintained on a high-fat diet. In contrast, while administration of MA has been reported to stimulate food intake in pre-pubertal female rats, food intake is not stimulated by MA in adult female rats. Instead, administration of MA to adult females results in changes in reproductive behavior and physiology. The present experiments were designed to examine the effects of administration of MA on food intake in adult female rats. The results demonstrated that, as previously reported, food intake was stimulated by MA in adult male rats on low-fat and high-fat diets, but food intake was not stimulated by MA in gonadally-intact adult female rats on either low-fat or high-fat diet. Further, MA did not stimulate food intake in female rats ovariectomized as adults. However, when females were ovariectomized prior to the onset of puberty (postnatal day 25–28), food intake was stimulated by administration of MA in adulthood. Finally, cyclic injections of 17-β-estradiol benzoate given to females ovariectomized prior to the onset of puberty abolished the stimulatory effects of MA on food intake in adult females. Taken together, the data suggest that exposure to estrogens during the time of puberty in female rats can persistently alter adult ingestive responding to signals related to changes in energy utilization.  相似文献

Milk samples were collected from Murrah buffalo between Day 30 and Day 120 post partum and analysed for progesterone concentration to monitor ovarian cyclicity. Progesterone levels were low (1 to 5 ng/ml) during the anestrous period. Levels were also low around estrus, but they began to increase at Day 6 postestrus; high levels (15 to 32 ng/ml) were maintained for different periods. There was a marked drop in progesterone level after Day 16 to 18 of the estrous cycle in those animals which returned to estrus. Progesterone levels remained high in buffalo which did not return to estrus, indicating that these animals were pregnant. Some of the progesterone cycles were not associated with the expression of estrus. This study indicated that a milk progesterone enzymoimmunoassay can be used to detect early pregnancy as well as conditions such as silent estrus and anovulatory estrus; it can thus help reduce the long intercalving period in buffalo.  相似文献

A completely randomized experimental design was used to investigate the effect of supplemental progesterone on pregnancy rates of recipient mares. Every other recipient mare received daily 200 mg progesterone in oil beginning the day of surgical embryo transfer and lasting until either Day 120 of pregnancy or until pregnancy failure was confirmed by ultrasound. Progesterone supplementation did not affect pregnancy rate (P > 0.05). Overall, embryos that did not result in pregnancy were of greater mean diameter than embryos that resulted in pregnancy (P < 0.05). Pregnancy rates tended (P < 0.1) to be greater in recipients that were detected to be ovulating the same day or prior to that of the donor and that had been supplemented with progesterone (75 %) as opposed to untreated control mares of the same synchrony group (40 %). Progesterone supplementation did not affect the incidence of embryonic loss; however, there was a slightly higher loss of pregnancies between Day 15 and 30 in treated versus untreated recipients. There was no effect (P > 0.05) of treatment on pregnancy rate for embryos recovered from fertile versus subfertile donor mares. However, overall, there tended (P < 0.1) to be fewer pregnancies with embryos recovered from subfertile (50 %) as compared to fertile donors (75 %). It was concluded that supplemental progesterone at the dosage and frequency described was not beneficial in improving pregnancy rates in cyclic recipient mares after surgical embryo transfer.  相似文献

The hypothesis that luteinizing hormone (LH) secretion in prepubertal females is responsive to estradiol negative feedback and that decreased feedback occurs as puberty approaches was tested in heifers. In the first experiment, seven heifers were maintained prepubertal by dietary energy restriction until 508 days of age (Day 0). All heifers were placed on a high-energy diet on Day 0 at which time they received no additional treatment (CONT), were ovariectomized (OVX) or were ovariectomized and subcutaneously implanted with estradiol-17 beta (OVX-E2). This feeding regimen was used to synchronize reproductive state in all heifers. A second experiment was performed with 16 prepubertal heifers using the same treatments at 266 days (Day 0) of age (CONT, OVX and OVX-E2) but no dietary intake manipulation. In both experiments, LH secretion increased rapidly following ovariectomy in OVX heifers. In the initial experiment, LH secretion was maintained at a low level in OVX-E2 heifers until a synchronous rapid increase was noted coincidental with puberty in the CONT heifer. In the second experiment, LH secretion increased gradually in OVX-E2 heifers and attained castrate levels coincidental with puberty in CONT heifers. A gradual increase in LH secretion occurred as puberty approached in CONT heifers. These results indicate that: a) LH secretion in prepubertal heifers is responsive to estradiol negative feedback; and b) estradiol negative feedback decreases during the prepubertal period in beef heifers.  相似文献

The aim of this study was to determine the effects of maternal diet with adequate (A) or high (H) selenium (Se) supplementation on ovarian and uterine characteristics, and onset of puberty in adolescent offspring. Sheep were fed a maintenance (M) diet with ASe or HSe levels from breeding to parturition. From Day 50 to parturition, a portion of the ewes from ASe and HSe groups was fed restricted (R, 60% of M) or excess (E, 140% of M) diet. Immediately after birth, lambs were separated from their dams and given artificial colostrum for 20 hours, followed by milk replacer. From Day 57.3 ± 0.6, ewe lambs were fed a pelleted grower diet until Day 116.3 ± 0.6 when they were transitioned to a finisher diet. From Day 99 to 180, serum samples were collected weekly from jugular vein for progesterone analysis to determine onset of puberty. Reproductive tissues were collected on Day 180.1 ± 0.4 of age. Maternal diet or Se supplementation did not affect uterine or ovarian weight and onset of puberty. However, area under the curve for progesterone was greater (P = 0.05) in ASe compared with HSe groups, and was greater in ASeM than HSeM group. In CLs, labeling index (LI; a proportion of proliferating cells) was less (P < 0.04) in HSeM than ASeM group, and in stroma was less (P < 0.05) in R and E groups than M group. Maternal diet did not affect the LI of any follicle types. For all groups combined, LI was the greatest (P < 0.001) in antral, less in early antral and secondary, and the least in atretic follicles. Our results demonstrate that maternal diet influenced ovarian but not uterine characteristics or onset of puberty. These results indicate that maternal plane of nutrition and/or Se supplementation may have specific effects on reproductive function in offspring.  相似文献
