The success of Triticum aestivumxZea mays crosses, used to producewheat doubled haploids, is influenced by light intensity. Toexamine the basis for this response, pollen tube growth, embryosurvival and indicators of photosynthetic rate were measuredin two wheat cultivars (‘Karamu’ and ‘Kotuku’)crossed with maize at two irradiance levels (250 or 750 µmolm-2s-1, PAR). Pollen tube growth was significantly affectedby light intensity in ‘Karamu’ plants but not in‘Kotuku’ plants, despite both cultivars being pollinatedby the same maize source. The percentage of pollen tubes reachingthe cavity between the ovarian wall and integuments, or in themicropyle of ‘Karamu’ plants at high light intensity(65%) was nearly three-times greater than that at low lightintensity (22%). Thus, either low light intensity can affectthe maternal wheat plant in a way that inhibits pollen tubegrowth and/or high light intensity may promote pollen tube growthin ‘Karamu’ plants. Significant differences in ratesof electron transport in plants grown at the two light intensitiesindicated that the rate of photosynthesis may also have an effecton pollen tube growth. These results have importance for improvingthe efficiency of wheat x maize crosses and other wide cerealcrosses. Species that showed marked morphological and physiological responsesby their roots to Fe-deficiency (Strategy I plants) were comparedwith others that do not exhibit these responses (Strategy IIplants). Roots from Fe-deficient cucumber (Cucumis sativusL.‘Ashley’), tomato (Lycopersicon esculentumMill.T3238FER) and pea (Pisum sativumL. ‘Sparkle’) plantsproduced more ethylene than those of Fe-sufficient plants. Thehigher production of ethylene in Fe-deficient cucumber and peaplants occurred before Fe-deficient plants showed chlorosissymptoms and was parallel to the occurrence of Fe-deficiencystress responses. The addition of either the ethylene precursorACC, 1-aminocyclopropane-1-carboxylic acid, or the ethylenereleasing substance, Ethephon, to several Fe-sufficient StrategyI plants [cucumber, tomato, pea, sugar beet (Beta vulgarisL.),Arabidopsis(Arabidopsis thaliana(L.) Heynh ‘Columbia’), plantago(Plantago lanceolataL.)] promoted some of their Fe-deficiencystress responses: enhanced root ferric-reducing capacity andswollen root tips. By contrast, Fe-deficient roots from severalStrategy II plants [maize (Zea maysL. ‘Funo’), wheat(Triticum aestivumL. ‘Yécora’), barley (HordeumvulgareL. ‘Barbarrosa’)] did not produce more ethylenethan the Fe-sufficient ones. Furthermore, ACC had no effecton the reducing capacity of these Strategy II plants and, exceptin barley, did not promote swelling of root tips. Aspects of growth and development were evaluated in the fast-developingannual Triticum aestivum L. ‘Chinese Spring’, theslow-developing perennial Lophopyrum elongatum Löve, theiramphiploid, and chromosome addition and substitution lines ofL. elongatum into ‘Chinese Spring’. Relative growthrates (RGR) of shoots of L. elongatum and the amphiploid werelower than those of ‘Chinese Spring’ (34 and 13%respectively) and main stem development was also slower. Therewas no difference in shoot RGR of any of the chromosome additionor substitution lines and that of ‘Chinese Spring’when assessed between Haun stages 2.0 and 5.0. In contrast,several aspects of plant development were observed to differin the chromosome addition and substitution lines. SubstitutingE genome chromosomes (with the exceptions of 3E and 4E) forD genome chromosomes, or adding E genome chromosomes, slowedthe rate of main stem development, at least up to Haun stage5.0. Despite these differences in the rate of main stem development,the appearance of adventitious roots commenced at approximatelyHaun stage 2.0 in all genotypes. Immature embryos (stage I) and cotyledonary somatic embryos(stage III) of black spruce [Picea mariana (Mill) B.S.P.] werebombarded with tungsten particles coated with a gene constructcontaining the fusion of gus:: nptll. GUS (ß-glucuronidase)activity was monitored histochemically with X-gluc giving ablue colour where transient gene expression was detected inthe bombarded tissues. A high transient expression of gus wasobserved in stage I embryo cultures 2 d after bombardment (202GUS foci per 300 mg tissue). GUS activity had substantiallydiminished in this material 14 d after bombardment, when grownin liquid LP maintenance medium containing BA (4.4µM),2,4-D (9µM) and 1% sucrose. However, when stage I embryoswere cultured on LP maturation medium containing BA (40 µM),IBA (1 µM), 3.4% sucrose and 0.8% agar, GUS activity after2 d was 335 GUS foci per 300 mg tissue, and the activity wasdetected until 30 d after bombardment. With stage III somaticembryos cultured on LP maintenance medium, 92% showed GUS activity2d after bombardment (16 GUS foci per embryo), and 31 % showedactivity 30 d after bombardment (4 GUS foci per embryo). GUSactivity was still evident in 12% of the embryos (2 GUS fociper embryo) 45 d after bombardment. The effects of exposure of up to 2 h with sulphur dioxide ona range of plant species was observed by measuring changes inthe rate of net photosynthesis under closely controlled environmentalconditions. Ryegrass, Lolium perenne ‘S23’ was thespecies most sensitive to SO2; significant inhibition was detectedat 200 nl l–1. Fumigations at 300 nl l–1 also inhibitedphotosynthesis in field bean (Vicia faba cv. ‘Three FoldWhite’ and ‘Blaze’) and in barley (Hordeumvulgare cv. ‘Sonja’). No effect was detected inwheat (Triticum aestivum cv. ‘Virtue’) at concentrationsup to 600 nl l–1 SO2, or in oil-seed rape (Brassica napuscv. ‘Rafal’) except at 800 nl l–1 SO2). Recoverycommenced immediately after the fumigation was terminated andwas complete within 2 h when inhibition had not exceeded 20%during the SO2 treatment. Manipulation of Grain Dormancy in Wheat   总被引:2,自引:1,他引:1  
To help in understanding the mechanisms of dormancy in wheat(Triticum aestivum L.), brief drying of intact ears has beenexamined as a technique for rapid imposition of dormancy. Beforenatural grain desiccation at 40 d post anthesis (dpa), wholeplants were moved for 15 to 20 d to ‘wet’, humidconditions (90–100% RH) or to ‘dry’ conditions(35–40% RH). The duration of subsequent dormancy was atleast doubled by the ‘dry’ treatment, however, onlyif the grain was allowed to dry before 50 dpa. In the ‘wet’ears there was a low level (up to 22%) in-ear germination by55 dpa and the remaining, non-sprouted grain no longer becamedormant on drying. Brief (4 d) drying episodes could also preventsprouting and induce some dormancy, but only over a 4 to 10d window of sensitivity before 50 dpa. Grain drying when properlytimed not only arrests development but, perhaps through damagingeffects of drying, causes dormancy. Single populations of three hexaploid species of wheat, Triticumaestivum, Triticum spelta and Triticum macha, and two populationsof the tetraploid wheat, Triticum dicoccum (Pontus and Bordeaux),were grown in a greenhouse experiment at a range of soil floodingregimes: free draining, two levels of transient flooding andcontinuous flooding. Increasing severity of flooding treatment resulted in increasedsoil reduction and an increase in the concentration of reducediron and manganese in the experimental soil, and also resultedin a reduction in vegetative growth, number of inflorescences,grain number and grain weight. There were, however, large differencesbetween the wheat populations in the degree of reduction inyield caused by flooding. The population of T. macha was muchmore flooding-tolerant than the other hexaploid species andthe ‘Pontus’ population of the emmer wheat, T. dicoccum,was more tolerant than the ‘Bordeaux’ populationof this species and than T. spelta and T. aestivum. The effect of wind sway on the mechanical characteristics ofthe anchorage roots and the stem was investigated in maturewinter wheat (Triticum aestivumL., cv. Hereward). Wheat plantswere field-grown, either supported by a frame, which preventedwind sway, or unsupported (free-standing) and the morphologyand mechanical properties of the stems and the anchorage, ‘coronal’, roots were measured. Wind sway had little influence on either the stem height orear weight of the plants but did affect the mechanical propertiesof the stem. Stems of supported plants were weaker and moreflexible than the stems of free-standing plants. There werealso differences in the anchorage systems between the treatments:supported plants had just under half as many ‘coronal’ anchorage roots as the free-standing plants. This reducedthe anchorage strength of supported plants by a third. These differences in mechanical structure meant that the free-standingplants were more resistant to stem buckling and more resistantto anchorage failure. However, considering the difference inthe need for mechanical strength in plants from the two regimes,these differences were small. This suggests that wheat has inherentmechanical integrity and, as a monocotyledon with no secondarythickening, it differs little structurally between environments. The objectives of this study were: (1) to quantify post-anthesiskernel cytokinin levels in ‘Tibet Dwarf’, a dwarfwheat (Triticum aestivumL.) that accumulates elevated quantitiesof leaf cytokinins; and (2) to measure the effects of temperatureon kernel cytokinin accumulation and mature kernel mass in thiswheat. Post-anthesis kernel cytokinin accumulation was measuredin control plants maintained at 25/12 °C (day/night) andtreated plants which received a 7 d exposure to 35/25 °Cbeginning at anthesis and grown to maturity at 25/15 °C.Zeatin (Z), dihydrozeatin (diHZ) and their respective ribosideswere the predominant cytokinins detected in control and treatedplants. Minimal quantities of isopentenyl adenine-type cytokininswere detected. Kernel cytokinin content peaked within 3 d afteranthesis in both groups and returned to baseline levels within1–2 d. Relative to controls, exposure to high temperaturereduced kernel cytokinin content approx. 80% within 1 d afteranthesis. Because kernel cytokinin in control Tibet Dwarf plantsexceeded that previously measured in other varieties by over100-fold, the reduced content of treated plants still exceededthat of untreated plants of other varieties. The increased cytokinincontent did not enhance thermotolerance. Expression of the wheat ‘Em’ genes in embryos isstrongly induced both by abscisic acid and by subjection toosmotic stress. We have examined the basis of this inductionin an homologous system by con structing fusions between thepromoter sequences of a cloned ‘Em’ gene and theGUS reporter gene. The ABA-and stress-mediated expression ofthese constructs has been assayed following delivery to intactwheat embryos by particle bombardment. Staining of bombardedembryos with the chromogenic substrate X-gluc enabled a simpleand rapid visual identification of promoter activity by scoringthe numbers of stained spots. Although not rigorously quantitative,a general correspondence between number of expression events(spots) and promoter activity could be inferred when the resultsobtained with bombarded embryos were compared with those obtainedby the fluorlmetnc measurement of GUS activity in cereal aleuroneprotoplasts transfected with the same constructs. Analysis ofa series of 5'-deletions of the ‘Em’ promoters indicatedthat the stress-responsive cis-acting regulatory elements comprisethe same sequences as those responsible for ABA-mediated geneexpression. Two tetraploid (Triticum turgidum L.emend gr. turgidum and gr.durum) and five hexaploid wheats (Triticum x aestivum L. emendgr. aestivum) with reported tendencies for ‘branched heads’(supernurnerary spikelets) exhibited variation in its expressionunder different vernalization photoperiod and temperature regimes. Two main types of supernumerary spikelets were identified, multiplesessile spikelets (MSS) with two or more complete spikeletsat a rachis node and indeterminate rachilla spikelets (IRS)with two to 13 spikelets on an extended rachilla. The degree of supernumerary spikelet expression in wheats withvernalization response differed from those without. Short photoperiods(9–14 h) both outdoors and in a glasshouse environment,were more conducive to supernumerary spikelet expression than24 h photoperiod in both environments. The 24 h photoperiodglasshouse environment (higher mean temperatures) was leastconducive to its expression except in lines with a strong vernalizationresponse. The high stability of supernumerary spikelet expression in certaingenotypes in the different environments indicated the feasibilityof incorporating this character in breeding and selecting commercialwheats to increase grain number per head. Seventy-one wild and primitive diploid accessions of the S-,A- and D-genome species of Aegilops and Triticum, one tetraploidwheat, T. turgidum L. var. durum Desf., ‘Mexicali’,and two hexaploid wheats, T. aestivum L., ‘Anza’and ‘Yecora Rojo’ were evaluated and compared forprotein and lysine contents, carbon isotope discrimination,and various agronomic traits in the 1987–88 season underfield conditions. Significant variability was observed amongthe 71 accessions and among the three genomes for all traits.For most characters, the D-genome species exhibited the mostvariation, followed by the A- and S-genome species. Aegilopssquarrosa, T. urartu, and T. boeoticum showed large variationfor harvest index. Large variation for grain yield was exhibitedby Ae. squarrosa, Ae. sharonensis and Ae. longissima, whichcould be exploited in hybridization and breeding programs withmodern cultivars. The mean protein and lysine values of thediploids were significantly higher than those of the moderncultivars. The S- and A-genome accessions had higher proteinand lysine contents than the D genome. Among genomes, the meanvalues for yield and harvest index were significantly greaterin S- and D-genome accessions than in the A-genome accessions. The correlation pattern between yield and quality traits wasdifferent in the three genomes. Superior accessions with regardto both grain yield and quality traits were identified in eachspecies studied, except Ae. longissima and Ae. sharonensis.The advantages and uses of these accessions in wheat breedingprograms are discussed. A ‘Particle Inflow Gun’ was constructed to introducethe glucuronidase (GUS) and hygromycin resistance genes intohalophytic suspension cells of Kosteletzkya virginica. The transientexpression of the GUS gene was associated with the cell cultureconditions, physical parameters during the use of the ParticleInflow Gun, and different promoters coupled to GUS. When theCaMV35S promoter was used, the cells adapted at 85 mM NaCl hada similar gene transfer efficiency to those of the non-salt-adaptedcontrol, while expression was less in the 170 mM and 255 mMNaCl-adapted cells. Both elevating bombardment pressure to 1.65mPa and shortening the distance between the cells and the particleholder from 21 cm to 9 cm enhanced GUS expression in the cellsgrown in four salinity treatments. An ABA-responsive promoterinduced the expression of the GUS gene either with 10–4M ABA or with salts in the post-bombardment medium in both controland NaCl-adapted cell tines. Stable transgenic callus lineswere isolated by using hygromycin containing medium after bombardingthe suspension cells with the Particle Inflow Gun. The presenceof the GUS gene in stable transformants was confirmed not onlyby histochemical and fluorimetric assays for the GUS activity,but also by Southern hybridization of RT-PCR amplified mANA. Stable transformation of Eucalyptus globulus after biolisticDNA delivery was investigated using zygotic embryos as targetmaterial. Conditions for plant regeneration were first investigated.Six-day-old cultured embryos, which had shown to be a good targetfor DNA transient expression, appeared to be suitable for regeneration.Whole plants were recovered through organogenesis after particlegun bombardment. Transformation experiments were performed witha linear T-DNA fragment harbouring GUS and NPTII genes, usingthe biological and physical conditions defined for optimum transientexpression. After 2 months on a culture medium, neoformed GUS-positivecalli were obtained from the ‘T-DNA’ bombarded embryos.GUS-expressing calli were also recovered after selection withkanamycin following bombardment. The integration of both GUSand NPTII genes into the Eucalyptus genome was confirmed bySouthern blot analysis, demonstrating stable transformationof Eucalyptus globulus cells for the first time. Root overproduction in embryogenic calli of wheat cv. ‘ChineseSpring’ was controlled by the addition of IAA-degradingcompounds (citric acid, MnSO4 or EDTA) to the culture media.In the presence of 20 gl–1 sucrose, the number of rootson calli decreased significantly when media for either callusmaintenance or plant regeneration were amended with citric acid.EDTA was less effective in reducing the number of roots thancitric acid. An increase in the amount of a 31kDa (P1) polypeptideunder conditions which favoured a reduction in root formationwas observed. Mixed Nitrogen Nutrition and Productivity of Wheat Grown in Hydroponics   总被引:12,自引:1,他引:11  
The objective of this study was to study the effects of nitrogen(N) supplied as either mixtures of NO3 and NH4 or as all NO3on the final yield of spring wheat. Two separate greenhouseexperiments evaluated the durum spring wheat (Triticum durumL.) cultivar ‘Inbar’ in 1986, and the hard red springwheat (Triticum aestmum L.) cultivar ‘Len’ in 1987.Nitrogen treatments consisted of all NO, or mixtures (75/25or 50/50) of NO3 and NH4. At maturity, plants were harvested,separated into leaves, stems, roots, and grain, and each partanalysed for dry matter and chemical composition Compared to plants receiving only NO3 as the source of N, mixedN nutrition resulted in greater accumulation of whole plantreduced-N (49 to 108% more), phosphorus (38 to 69% more), andpotassium (25% more) for both cultivars. In all cases, plantsproduced higher grain yields (28% for Len to 78% for Inbar)when grown with mixed N nutrition than with only NO3. The yieldincrease was not associated with heavier grains or more grainsper ear, but rather with an increase in the number of ear-bearingtillers per plant. For both cultivars, the higher yields withmixed N resulted from the production of more total biomass (36to 76%) as the partitioning of dry matter between plant partswas not altered by N treatment. The relationship between amino acid and sugar export to thephloem was studied in young wheat plants (Triticum aestivumL. ‘Pro-INTA, Isla Verde’) using the EDTA-phloemcollection technique. Plants grown with a 16 h photoperiod showeda rapid decrease in the concentration of sugars and amino acidsin the phloem exudate from the beginning of the dark period.When plants grown with a 16 h photoperiod were kept in the darkfor longer than 8 h the free amino acid content in leaves andexudate (on a dry weight basis) increased continually throughoutthe 72 h of darkness. During the first 24 h of darkness thesugars in the phloem exudate decreased to 30% of the initialvalue, and returned to the control level when plants were returnedto light. When plants grown under low light intensity for 10d were transferred to high light intensity, they showed an increasein leaf sugar content (dry weight basis) after 3 d but therewere no differences in leaf free amino acid content (dry weightbasis) compared to low-light plants. The sugar concentrationin the phloem exudate was increased by higher light intensities,but there was no difference in the amino acid concentrationof the phloem exudate, and thus the amino acid:sugar ratio inthe phloem decreased in the high-light plants. The present resultssuggest that amino acids can be exported to the phloem independentlyof the export of sugars. Stagnant nutrient solution containing 0.1% agar and with anextremely low oxygen level (‘stagnant agar solution’)was used to simulate the gaseous composition and slow gas diffusionof waterlogged soils. Comparisons were made between the growthof two wheat cultivars(Triticum aestivum,cvs. Gamenya and Kite)and one triticale cultivar(Triticosecale,cv. Muir) grown instagnant relative to aerated solution. For all genotypes tested,immersion of roots in stagnant agar solution resulted in thedeath of the entire seminal root system and led to profuse branchingof the laterals of the nodal roots. In the stagnant agar solutionaerenchyma, as a percentage of the total cross sectional areaof nodal roots, was 18% for Muir, 14% for Kite and 12% for Gamenya;the roots of species with more aerenchyma also attained a longermaximum root length as predicted by the model of Armstrong (in:Woolhouse HW, ed.Advances in botanical research, vol. 7. London:Academic Press, 1979). Muir also had a nodal root/shoot freshweight ratio of 0.5 compared with 0.2–0.3 in Kite andGamenya. Development of seeds following pollination with irradiated pollenwas studied inActinidia deliciosa(kiwifruit) ‘Hayward’.Pollinations were carried out using two different sources ofpollen (‘Tomuri’ and ‘Matua’) irradiatedwith gamma rays at doses of 700 and 900 Gy. Non-irradiated crosseswere used as controls. Pollen irradiation had little effectonin vitropollen germination. Irradiated pollen affected seedset and seed content, and induced the formation of parthenogeneticembryos. In comparison to the control, the embryo growth ratewas slower and the endosperm contained very low amounts of storageproducts. Seed set was significantly reduced following bothdoses of irradiation. Two types of seeds were observed: (1)seeds with endosperm only; and (2) seeds with both embryo andendosperm. The proportion of seeds containing endosperm onlywas almost ten-fold higher than those containing both embryoand endosperm. Embryo production by gamma-irradiated pollenwas genotype- and dose-dependent. The induction of parthenogenesiswas higher following gamma ray doses of 900 Gy than 700 Gy,which suggests the ‘Hertwig Effect’; the best efficiencywas obtained with ‘Tomuri’ pollen. Ploidy levelof parthenogenetic embryos was evaluated by nuclear size (area)with the use of image analysis. There was a large differencein embryo nuclei size between control and parthenogenetic embryos(mean size 90.8 and 49.1 µm2, respectively). Grain number in the wheat cultivar Banks was reduced by up to11 % with a rise in temperature from 21/16 °C to 30/25 °Cover a 10-d period immediately following first anthesis in general,the upper ‘d’ and ‘c’ florets were moreaffected by high temperature than the basal ‘a’and ‘b’ florets within a spikelet and florets fromthe upper spikelets were more sensitive than those lower onthe ear Grain weight and grain length at maturity were most affectedby a 10 d period of high temperature commencing 7–10 dafter anthesis However, if dry-matter accumulation between thestart of a treatment and grain maturity was used as a base forcomparison, the response was more uniform throughout development,with a peak in sensitivity 25 d after anthesis Although grainposition within an ear did not have a large effect on the responseto temperature, grains from the basal spikelets were more sensitivethan those from the apex, and the upper floret grains of a spikeletwere more sensitive to high temperature than those at the base There is a need to obtain, for a range of cultivars, more comprehensivedata on the effect of the timing and degree of temperature stressfollowing anthesis, for use in interpreting the response torising temperatures late in the development of the crop in thefield Triticum aestivum L, wheat, temperature, grain development  相似文献
