It is shown that the models for the transduction process in photoreceptors which treat latency and amplification as integrated phenomena (integrated models) yield time scales for single photon signals (quantum bumps) which distinctly conflict with the experimentally observed ones for the ventral nerve photoreceptor of Limulus: the ratio of bump duration/ latency t B/t lat is predicted by integrated models to be 3 in contrast to the experimental result of 0.5. Moreover, integrated models lead to a predicted value of an extinction rate of 50%, i.e., 50% of the absorbed photons should be expected to cause no signal in the dark adapted state of the cell. In this paper it is shown that separation of latency and amplification in such a way that the latency causing process precedes amplification in the transduction process eliminates these discrepancies. In addition, the separate modeling of latency and amplification resolves the rather large ambiguity in determining the exponent n of the initial signal current J(t) nreported in the literature to be between n2 (from noise analysis) up to n17 (from flash experiments). This paper details attempts to breed members of the L. saxatilis species complex in the laboratory and to construct interspecific hybrids between them. Success in reciprocal crosses between animals from the type localities of the species of Littorina saxatilis (Olivi) and Littorina rudis (Maton) indicates that these two taxa are synonymous. Six of the twelve possible reciprocal hybridisation crosses between the four species (L. saxatilis, L. arcana Hannaford Ellis, L. nigrolineata Gray and L. neglecta Bean) have been attempted, with only one proving successful, that between male L. saxatilis and female L. arcana. This hybrid cross produces viable offspring, although at a lower frequency than either of the parental crosses in the laboratory, while the reciprocal cross has, as yet, proved unsuccessful. Limited work on the F1 hybrids shows them to be inter-fertile and also capable of backcrossing with male L. saxatilis, but not with female L. saxatilis or L. arcana. Details are also given of attempts to find natural hybrids in sympatric populations of the species of the complex, using gel electrophoresis.  相似文献

Summary Spindles of nuclei mostly from metaphases of expiants in vitro, Set free according to Dan's method by means of solutions of Duponol (a sodium lauryl sulfonate), have been studied according to their elasticity. The isolated atractoplasm has shown a static modul of elasticityE0.8 to 1.6×106 dynes×cm–2, and, sometimes, a Poisson number of0.42, that is to say the atractoplasmic gel of the spindles of nuclei is certain elastically. The values of the modul of elongation agree satisfactory with those values recently ascertained by Crick and Hughes with cytoplasm oi explanted cells.  相似文献

The general properties of the excitable membrane on molluscan pacemaker neurons can be described on the basis of a fair amount of experimental evidence available in the literature. The neuronal membrane exhibits under voltage clamp an initial inward current carried by both Na+ and Ca2+ ions, the time- and voltage-dependent characteristics of which are similar to that of other excitable structures. The conductance mechanism for the two ion species and the transport kinetics appear to be closely similar. The time course and amplitude of the delayed outward current carried by K+ ions shows a marked dependence on the membrane potential. Characteristic for the molluscan neurons is the existence of an additional fast transient outward current which is only activated by hyperpolarizing shifts from the membrane potential. A regular beating discharge over a wide range of frequencies can be predicted by making the assumption of a metabolically controlled driving of the Na+ conductance. Bursting pacemaker characteristics can be correctly simulated by the model if sinusoidal variations of an additional Na+ and Ca2+ conductances g Na and g Ca, and periodic variations of the K+ conductance g K, governed by the known operation of a metabolic substrate cycle are introduced. The close approximation of experimentally observed impulse bursts requires that the actual inpulse-frequency and the amplitude of the after-spike hyperpolarization are determined by the temporal pattern of g Na, while the spike amplitude is controlled by g Na which (although of similar time course) is lagging in phase behing g Na. The periodic changes in additional K+ conductance g K, are responsible for burst termination and the changes in inter-burst interval, to the effect that spike doublets, triplets and multi-spike bursts can be simulated by a suitable choice for the time characteristics of g K. The model makes use of the finding that the Ca2+ inflow associated with a spike discharge actually activates g K, so that large postburst hyperpolarizations can be obtained in high-frequency bursts.Supported by the Deutsche Forschungsgemeinschaft (Grant Ch 25/1)  相似文献

Previous results on Limulus ventral photoreceptors have suggested that besides inositol trisphosphate, another unknown transmitter may also work in the transduction cascade. This assumption has been supported by the finding of two light-activated channel types. The present report furnishes further evidence of the dual transmitter mechanism in phototransduction by analyzing the kinetic properties and voltage dependency of these cation channels with conductances of 12 pS and 30 pS. Single-channel currents were recorded in Limulus ventral nerve photoreceptors in cell-attached configuration at 14°C. At V m + 80 mV the open-time histograms of both channels were fit best by the sum of two exponentials; time constants (and weights) were: 0.81 ms (0.62) and 6.20 ms (0.38) for the 12 pS channels and 2.38 ms (0.43) and 19.4 ms (0.57) for the 30 pS channels. At this potential the mean open times were 2.7 ms for the 12 pS and 13.3 ms for the 30 pS channels, about two-times larger than at hyperpolarizing potentials. The deactivation kinetics were also different for the two channels. The time constants of the decay of the channel activity, after switching off the light, were 2.5 s for the 12 pS and 12.9 s for the 30 pS channels. The 12 pS channel exhibits bursting and subconductance states at positive potentials. The subconductances are about 20%, 46% and 72% of the fully open state. Results show that the two types of light-activated channels have different kinetic parameters, voltage dependence and gating mechanisms. The two channels are suggested to be gated by different transmitters or processes. It is proposed that for the 30 pS channel the transmitter could be calcium ion or a calcium-dependent transmitter.  相似文献

The Fourier transform infrared spectra of agar, agarose, -, -, and -carrageenan, and ofChondrus canaliculatus, Iridaea ciliata, I. membranacea, I. laminarioides andGracilaria chilensis polysaccharides were recorded in the 4000–400 cm-1 region. The bands in the second derivative mode are sharper and more bands are resolved than in the normal spectra.Agar, agarose andG. chilensis phycocolloids exhibit diagnostic bands at 790 and 713 cm-1. -, - and -carrageenans, and native carrageenan-type polysaccharides fromC. canaliculatus andIridaea species exhibit bands at around 1160, 1140, 1100, 1070, 1040, 1008, 610, and 580 cm-1. Therefore, FT-IR spectroscopy in the second-derivative mode may be applied to differentiate between agar- and carrageenan-types seaweed galactans.  相似文献

The sequence of the mtDNA of the grey seal, Halichoerus grypus, was determined. The length of the molecule was 16,797 base pairs. The organization of the molecule conformed with that of other eutherian mammals but the control region was unusually long due to the presence of two types of repeated motifs. The grey seal and the previously reported harbor seal, Phoca vitulina, belong to different but closely related genera of family Phocidae, true (or earless) seals. In order to determine the degree of differences that may occur between mtDNAs of closely related mammalian genera, the 2 rRNA genes, the 13 peptide coding genes, and the 22 tRNA genes of the 2 species were compared. Total nucleotide difference in the peptide coding genes was 2.0–6.1%. The range of conservative difference was 0.0–1.5%. In the inferred peptide sequences the amino acid difference was 0.0–4.5%, and the difference with respect to chemical properties of amino acids was 0.0–3.0%. A gene that showed a limited degree of difference in one mode of comparison did not necessarily show a corresponding limited difference in another mode. The ratio for differences in codon positions 1, 2, and 3 was 2.7:1:16. The corresponding ratio for conservative differences was 1.8:1. l:1. The evolutionary separation of the two species was calculated to have taken place 2–2.5 million years ago. This dating gives the figure 8 × 10–9 as the mean rate of substitution per site and year in the entire mtDNA molecule. Comparison with the cytochrome b gene of the Hawaiian monk seal and the Weddell seal suggested that the lineage of these two species and that of the grey and harbor seals separated 8 million years ago. We have examined and dated primate divergences by applying a newly established molecular/paleontological reference, the evolutionary separation between artiodactyls and cetaceans anchored at 60 million years before present (MYBP). Owing to the morphological transformations coinciding with the transition from terrestrial to aquatic (marine) life and the large body size of the animals (which makes their fossils easier to find), this reference can be defined, paleontologically, within much narrower time limits compared to any local primate calibration marker hitherto applied for dating hominoid divergences. Application of the artiodactyl/cetacean reference (A/C-60) suggests that hominoid divergences took place much earlier than has been concluded previously. According to a homogenous-rate model of sequence evolution, the primary hominoid divergence, i.e., that between the families Hylobatidae (gibbons) and Hominidae, was dated at 36 MYBP. The corresponding dating for the divergence betweenPongo (orangutan) andGorilla-Pan (chimpanzee)-Homo is 24.5 MYBP, that forGorilla vsHomo-Pan is 18 MYBP, and that forHomo vsPan 13.5 MYBP. The split between Sumatran and Bornean orangutans was dated at 10.5 MYBP and that between the common and pygmy chimpanzees at 7 MYBP. Analyses of a single gene (cytochromeb) suggest that the divergence within the Catarrhini, i.e., between Hominoidea and Old World monkeys (Cercopithecoidea), took place >40 MYBP; that within the Anthropoidea, i.e., between Catarrhini and Platyrrhini (New World monkeys), >60 MYBP; and that between Anthropoidea and Prosimii (lemur), 80 MYBP. These separation times are about two times more ancient than those applied previously as references for the dating of hominoid divergences. The present findings automatically imply a much slower evolution in hominoid DNA (both mitochondrial and nuclear) than commonly recognized.  相似文献

Endothelin-1 (ET-1) has been found to increase cardiac -myosin heavy chain (-MyHC) gene expression and induce hypertrophy in cardiomyocytes. ET-1 has been demonstrated to increase intracellular reactive oxygen species (ROS) in cardiomyocytes. The exact molecular mechanism by which ROS regulate ET-1-induced -MyHC gene expression and hypertrophy in cardiomyocytes, however, has not yet been fully described. We aim to elucidate the molecular regulatory mechanism of ROS on ET-1-induced -MyHC gene expression and hypertrophic signaling in neonatal rat cardiomyocytes. Following stimulation with ET-1, cultured neonatal rat cardiomyocytes were examined for 3H-leucine incorporation and -MyHC promoter activities. The effects of antioxidant pretreatment on ET-1-induced cardiac hypertrophy and mitogen-activated protein kinase (MAPKs) phosphorylation were studied to elucidate the redox-sensitive pathway in cardiomyocyte hypertrophy and -MyHC gene expression. ET-1 increased 3H-leucine incorporation and -MyHC promoter activities, which were blocked by the specific ETA receptor antagonist BQ-485. Antioxidants significantly reduced ET-1-induced 3H-leucine incorporation, -MyHC gene promoter activities and MAPK (extracellular signal-regulated kinase, p38, and c-Jun NH2 -terminal kinase) phosphorylation. Both PD98059 and SB203580 inhibited ET-1-increased 3H-leucine incorporation and -MyHC promoter activities. Co-transfection of the dominant negative mutant of Ras, Raf, and MEK1 decreased the ET-1-induced -MyHC promoter activities, suggesting that the Ras-Raf-MAPK pathway is required for ET-1 action. Truncation analysis of the -MyHC gene promoter showed that the activator protein-2 (AP-2)/specificity protein-1 (SP-1) binding site(s) were(was) important cis-element(s) in ET-1-induced -MyHC gene expression. Moreover, ET-1-induced AP-2 and SP-1 binding activities were also inhibited by antioxidant. These data demonstrate the involvement of ROS in ET-1-induced hypertrophic responses and -MyHC expression. ROS mediate ET-1-induced activation of MAPK pathways, which culminates in hypertrophic responses and -MyHC expression. The feeding dynamics of males and females of the sequential protandric shrimp Nauticaris marionis were investigated in late austral summer (April) 2002. Results of gut content analysis indicated that N. marionis is an opportunistic feeder preying on a variety of prey, with a preference for detritus, benthic amphipods and gastropods. Overall, pelagic prey made only an insignificant contribution to diet, and was only taken by males. Females of N. marionis exhibited cannibalism, particularly in incubation containers. There was no diel pattern in the feeding activity of N. marionis. In-situ daily rations of males (carapace length <7 mm) and females (>7 mm) were equivalent to 10% and 5% of body dry weight, respectively.An erratum to this article can be found at  相似文献

Pigeonpea, Cajanus cajan (L.) Millsp. (Leguminosae) was found as a novel food plant of the neotropical brown stink bug, Euschistus heros (F.), in Paraná State, Brazil. Laboratory studies indicated nymphs performing better when fed on immature pods of soybean, Glycine max (L.) Merrill (Leguminosae), than on immature pods of pigeonpea. Female nymphs required 1.7 d less to complete development and tended to have a lower mortality on soybean (17.5%) than on pigeonpea (25.0%). At emergence, fresh body weight was greater on soybean than on pigeonpea. Adult E. heros performed better on soybean than on pigeonpea. After 120 d, 35% of adults were alive on soybean and 30% on pigeonpea; 93.3% of females oviposited on soybean, and 73.3% on pigeonpea, with an oviposition delay of 17 days on pigeonpea compared to soybean. Fecundity was 63% greater on soybean than on pigeonpea, and adult body weight gain for females and males at different ages was 6–36% greater on soybean than on pigeonpea. E. heros colonized soybean during January–March, from the pod-setting and filling stages (R3–R6) to maturity (R7–R8), and pigeonpea during April–June. These results indicate that E. heros has expanded its host range to pigeonpea in Brazil; pods of this plant are suitable food, and can support the insect during autumn-early winter, after soybean harvest, and before it enters into partial hibernation.  相似文献

Summary To identify ion transport systems involved in the maintenance of vascular smooth muscle cell volume the effects of incubation medium osmolality and ion transport inhibitors on the volume and 86Rb and 22Na transport in cultured smooth muscle cells from rat aorta (VSMC) have been studied. A decrease of medium osmolality from 605 to 180 mosm increased intracellular water volume from 0.6 to 1.3 l per 106 cells. Under isosmotic conditions, cell volume was decreased by ouabain (by 10%, P< 0.005) but was not influenced by bumetanide, furosemide, EIPA and quinidine. These latter compounds were also ineffective in cell volume regulation under hypotonic buffer conditions. Under hyperosmotic conditions, cell volume was decreased by bumetanide (by 7%, P<0.05) and by ethylisopropyl amiloride (by 13%, P< 0.005). Ouabain-sensitive 86Rb influx was decreased by 30–40% under hypoosmotic conditions. An increase in medium osmolality from 275 to 410 mosm resulted in an eightfold increase in bumetanide-inhibited 86Rb influx and 86Rb efflux. The (ouabain and bumetanide)-insensitive component of 86Rb influx was not dependent on the osmolality of the incubation medium. However (ouabain and bumetanide)-insensitive 86Rb efflux was increased by 1.5–2 fold in VSMC incubated in hypotonic medium. Ethylisopropyl amiloride-inhibited 22Na influx was increased by sixfold following osmotic-shrinkage of VSMC. The data show that both Na+/H+ exchange and Na+/K+/2Cl cotransport may play a major role in the regulatory volume increase in VSMC. Basal and shrinkage-induced activities of Na+/K+/2Cl cotransport in VSMC were similarly sensitive to inhibition by either staurosporin, forskolin, R24571 or 2-nitro4-carboxyphenyl N,N-diphenylcarbomate (NCDC). In contrast basal and shrinkage-induced Na+/K+/2Cl cotransport were differentially inhibited by NaF (by 30 and 65%, respectively), suggesting an involvement of guanine nucleotide binding proteins in the volume-sensitive activity of this carrier. Neither staurosporin, forskolin, R24571 nor NCDC influenced shrinkage-induced Na+/H+ exchange activity. NaF increased Na+/H+ exchanger activity under both isosmotic and hyperosmotic conditions. Summary It has been previously demonstrated that taurine inhibits the phosphorylation of an 20K apparent molecular weight protein present in the mitochondrial fraction of the rat retina (Lombardini, 1991). In the present studies, various analogues of taurine were tested for their inhibitory activity on the phosphorylation of this 20K protein. The most potent analogues were (±)trans-2-aminocyclopentanesulfonic acid (TAPS) and 1,2,3,4-tetrahydroquinoline-8-sulfonic acid (THQS) which were approximately 21 and 7 times more potent than the parent compound, taurine. Median-effect plots were used to calculate the inhibitory median effect and combination index values for the combined effects of taurine and taurine analogues. From these studies, it was determined that the inhibitory taurine analogues were antagonistic to taurine when used in combination with taurine to inhibit the phosphorylation of the 20K apparent molecular weight protein. It was also concluded that: 1) the distance between the nitrogen and sulfur atoms in the taurine structure was important for inhibitory activity; 2) if the nitrogen atom is either within or attached to an unsaturated ring structure the inhibitory potency was significantly decreased, and 3) if both the sulfur and nitrogen atoms are present within the ring structure the analogue has no activity.  相似文献

Summary The growth factor receptor-bound protein-2 (Grb2) is an adaptor protein that mediates signal transduction pathways. Chemical shift assignments were obtained for the SH2 domain of Grb2 by heteronuclear NMR spectroscopy, employing the uniformly 13C-/15N-enriched protein as well as the protein containing selectively 15N-enriched amino acids. Using the Chemical Shift Index (CSI) method, the chemical shift indices of four nuclei, 1H, 13C, 13C and 13CO, were used to derive the secondary structure of the protein. Nuclear Overhauser enhancements (NOEs) were then employed to confirm the secondary structure. The CSI results were compared to the secondary structural elements predicted for the Grb2 SH2 domain from a sequence alignment [Lee et al. (1994) Structure, 2, 423–438]. The core structure of the SH2 domain contains an antiparallel -sheet and two -helices. Summary The conductance of the Ca2+-activated K+ channel (g K(Ca)) of the human red cell membrane was studied as a function of membrane potential (V m ) and extracellular K+ concentration ([K+]ex). ATP-depleted cells, with fixed values of cellular K+ (145mm) and pH (7.1), and preloaded with 27 m ionized Ca were transferred, with open K+ channels, to buffer-free salt solutions with given K+ concentrations. Outward-current conductances were calculated from initial net effluxes of K+, correspondingV m , monitored by CCCP-mediated electrochemical equilibration of protons between a buffer-free extracellular and the heavily buffered cellular phases, and Nernst equilibrium potentials of K ions (E K) determined at the peak of hyperpolarization. Zero-current conductances were calculated from unidirectional effluxes of42K at (V m –E K)0, using a single-file flux ratio exponent of 2.7. Within a [K+]ex range of 5.5 to 60mm and at (V m –E K) 20 mV a basic conductance, which was independent of [K+]ex, was found. It had a small voltage dependence, varying linearly from 45 to 70 S/cm2 between 0 and –100 mV. As (V m –E K) decreased from 20 towards zero mVg K(Ca) increased hyperbolically from the basic value towards a zero-current value of 165 S/cm2. The zero-current conductance was not significantly dependent on [K+]ex (30 to 156mm) corresponding toV m (–50 mV to 0). A further increase ing K(Ca) symmetrically aroundE K is suggested as (V m –E K) becomes positive. Increasing the extracellular K+ concentration from zero and up to 3mm resulted in an increase ing K(Ca) from 50 to 70 S/cm2. Since the driving force (V m –E K) was larger than 20 mV within this range of [K+]ex this was probably a specific K+ activation ofg K(Ca). In conclusion: The Ca2+-activated K+ channel of the human red cell membrane is an inward rectifier showing the characteristic voltage dependence of this type of channel.  相似文献

We determined the structure of the Schizosaccharomyces pombe centromere cen3 using direct genomic mapping and cosmid walking. The repetitive region of cen3 is approximately 110 kb, much longer than that of the previously determined cen1 and cen2 regions. The 30 kb long left and 60 kb right repetitive sequences are arranged with an inverted symmetry and flank the 1520 kb central domain. The repeat motifs in cen3, although they consist of the common centromeric repeat elements, are slightly different from those in cen1 and cen2. The cen3 repeat motifs appear to be reiterated four times in the left and nine times in the right side repetitive regions. We found that the central domain consists of the common 5 kb core sequence associated with the pair of innermost inverted sequences, most of which are reiterated only twice in the genome. Although their sizes differ significantly, the general features of cen1, cen2 and cen3 are similar, and a prototype, consensus structure for the fission yeast centromere may be deduced.by J.A. We have examined the effects of culturing neonatal rat-brain astrocytes in medium containing delipidated serum, with or without added linoleic acid (LA, 18:26), on membrane fatty-acid composition and functions. After 18–21 days in culture, polyunsaturated fatty acids (PUFA) constituted24 mol% of the total fatty acids in the astrocytes grown in delipidated media (controls); these proportions were increased by 35–40% to33 mol% when the cells were supplemented with 35M LA. Notable differences in the PUFA profiles of the cells cultured with or without added LA included: (a) higher proportions of 6 PUFA in the LA-supplemented astrocytes (25%, relative to10% in controls) that were accompanied by an increase in the ratio of 6/3 PUFA (from <2 in controls to 5), and (b) higher proportions of 20:39 and 22:39 in the control astrocytes (>5%) relative to the LA-supplemented cells (1%). The major metabolites in the 6 PUFA-enriched cells were arachidonic (20:46), adrenic (22:46) and docosapentaenoic (22:56) acids (15, 5 & 3 mol%, respectively). Enrichment of the astrocytes in 6 PUFA did not alter basal levels of cAMP, nor did it affect the amounts of cAMP formed in response to forskolin, isoproterenol, adenosine or histamine. However, dopamine-dependent increases in cAMP formation in the presence of the phosphodiesterase inhibitor, Ro 20-1724, were reduced by 25% relative to those in controls. LA supplementation modified uptake of [3H]adenosine into the astrocytes; values for Kt for a high affinity transport were increased relative to controls, and maximum capacity of a lower affinity process was reduced. Uptake of [3H]glutamate was not altered in the 6 PUFA-enriched astrocytes. Summary Annual seed consumption by a small mammal community in the semi-arid Karoo, South Africa, was calculated using population, biomass and dietary estimates and published estimates of field metabolic rate. The community comprised a macroscelid elephant shrew Macroscelides proboscideus, and two rodents Gerbillurus paeba and Desmodillus auricularis. Only G. paeba consumed seed during the study, and then in low amounts (annual mean = 0.4% of diet, insects = 53%, herbage = 47%), with M. proboscideus and D. auriocularis consuming mainly insects (88%) and herbage (90%), respectively. Energy requirements for the G. paeba population averaged 568 kJ ha–1 day–1. Total seed consumption was estimated to be 140 g ha–1, or 155 000 seeds ha–1 which represents 0.5% of the annual seed production for this site. These levels of granivory by small mammals are significantly lower than those recorded for North American deserts, and raises the question as to why granivory varies between these systems. These findings also refute the hypothesis that as a result of the harshness and similarity of physical conditions in deserts, the properties of desert communities are convergent.  相似文献

Summary The foraging strategies of four naturally co-existing heteromyid rodent species were investigated: Dipodomys deserti (100 g), D. merriami (38 g), Microdipodops pallidus (13 g), and Perognathus longimembris (7 g). In 208 over-night laboratory foraging trials animals were provided with millet seed distributed in clumped and scattered patterns. Net removal of seeds from the foraging arena and amounts of seeds in cheek pouches and in caches were determined. When alone in an areana none of these species specialized extensively on either clumped or scattered seeds, although each tended to take more clumped than scattered seeds. When placed together with other individuals, animals once again tended to cache more clumped than scattered seeds in all but one paired combination of species: P. longimembris cached more scattered than clumped seeds when opposed by D. deserti. This suggests that the smaller species obtained a less preferred distribution of seeds in the face of competition. The two smaller species showed a great reduction in general foraging success in the presence of either of the two larger species. In general, a species cached less seeds when faced by larger opponent species.  相似文献

Effects of barnacle epibionts on the periwinkle Littorina littorea (L.)   总被引:1,自引:0,他引:1  
In a sandy bay with mussel beds in the Wadden Sea (Island of Sylt, eastern North Sea), periwinkles Littorina littorea (L.) were often strongly overgrown with the barnacle Balanus crenatus Bruguière in the lower intertidal zone. Consequences of this epibiosis on mobility, reproduction and mortalityof the snail were examined. B. crenatus growing on L. littorea increased snail volume up to 4-fold and weight up to 3.5-fold and crawling speed of fouled L. littorea was significantly slowed down. The epibiotic structure also caused a decrease in reproductive output. In laboratory experiments, egg production of fouled L. littorea was significantly lower than in snails free of barnacles. Presumably, copulation of the periwinkles is hampered by the voluminous and prickly cover of barnacles. Field studies demonstrated an increased mortality of overgrown L. littorea. A decrease in reproductive output and a lower survival of snails with a cover of barnacles suggest that B. crenatus epibionts may have a significant impact on the population of L. littorea.
