帘蛤科贝类rDNA内转录间隔区序列的研究   总被引:5,自引:0,他引:5  
根据18SrDNA、5.8SrDNA和28SrDNA保守序列设计引物,应用聚合酶链式反应(PCR)扩增了文蛤(Meretrix meretrix L.)、青蛤(Cyclina sinensis G)、硬壳蛤(Mercenaria mercenaria L.)和江户布目蛤(Protothaca jedoensis L.)4种帘蛤科贝类的第一内转录间隔区(ITS1)和第二内转录间隔区(ITS2)序列,并进行了测序。结果表明,文蛤、青蛤、硬壳蛤和江户布目蛤的ITS1扩增产物大小分别为978bp、663bp、757bp和942bp,GC含量分别为61.55%、60.78%、62.48%和64.86%~64.97%,其中ITS1序列长度分别为900bp、585bp、679bp和864bp,是迄今已报道双壳贝类中变化范围最大的,GC含量分别为61.67%、61.03%、63.03%和65.51%~65.62%,江户布目蛤种内ITS1序列有个体差异;ITS2扩增产物大小分别为644bp、618~620bp、593bp和513~514bp,GC含量分别为61.18%、61.29%~61.81%、62.73%和61.48%61.60%,其中ITS2序列长度分别为412bp、386~388bp、361bp和281~282bp,GC含量分别为65.29%、65.21%~66.06%、67.87%和67.38%~67.62%,青蛤和江户布目蛤种内ITS2序列有个体差异。4种蛤ITS1和ITS2序列种间差异很大,有明显的长度多态性,ITS2种间序列相似度73.0%~89.1%,与ITS1的种间序列相似度48.7%~81.5%相比略高。此外,在4种蛤ITS1和ITS2序列中各发现2个与rRNA加工有关的保守区。通过对ITS1和ITS2序列的组装获得了4种蛤5.8SrRNA基因完整序列,序列长度都是157bp,GC含量57.96%~58.60%,4种蛤5.8SrRNA基因相对保守,种间序列差异度0-6.0%,共有10个变异位点,其中转换4处,颠换6处,硬壳蛤和江户布目蛤5.8SrRNA基因序列完全相同。以ITS2序列(包含5.8SrRNA和28SrRNA基因部分序列)为标记,调用北极蛤科的Arctica islandica相应序列数据作外群,构建了帘蛤科贝类的系统发育树,其拓扑结构显示江户布目蛤与硬壳蛤亲缘关系最近,青蛤与其他3物种的亲缘关系最远。  相似文献

岩蕨科(Woodsiaceae)和球盖蕨科(Peranemaceae)历来被多数学者认为是亲缘关系密切的两个科,它们的系统位置一直存在争议,两科曾被放入三叉蕨科(Aspidiaceae)、碗蕨科(Dennstaedtiaceae)、鳞毛蕨亚科(Dryopteridoidaae)及鳞毛蕨科(Dryopteriaceae)蹄盖蕨亚科(Athyrioideae)中.根据岩蕨科1种、球盖蕨科3种、蹄盖蕨科8种、鳞毛蕨科2种及1种外类群假大羽铁角蕨(Asplenium pseudolaserpitiifolium)的cpDNA基因trnL-F区序列建立的系统树,结果显示,岩蕨科和球盖蕨科的亲缘关系较远,岩蕨科与蹄盖蕨科有密切的亲缘关系,球盖蕨科与鳞毛蕨科亲缘关系较近,岩蕨科和球盖蕨科是处于不同进化线上的植物.秦仁昌(1978)和Pichi-Sermolii(1977)的系统将二者分别作为独立的科是合适的.  相似文献

从rbcL序列探讨单室茱萸属的系统位置   总被引:1,自引:1,他引:1  
为了探讨单室茱萸属Mastixia的系统位置,本文对八蕊单室萸(Mastixia euony moides),云南单室茱萸(M.pentandra subsp,chinensis)及近缘物种云南紫树(Nyssa yunnanensis)和毛八角枫(Alangium kurzii)的cpDNA中编码1,5-二磷酸核酸糖羧化酶大亚基的rbcL基因序列进行了双脱氧测定。通过对这些序列与广义山茱萸科Cornaceae和Mastixia曾被归入的科的类群的rbcL序列进行PAUP(Phylogenetic Analysis Using Parsimony)分析,得到24棵最简约的分支树图。PAUP分析表明单室茱萸属与马蹄参属Diplopanax,紫树属Nyssa,喜树属Camptotheca和珙桐属Davidia的亲缘关系最近,并由分支图支持可以将Mastixia提升为山茱萸目Cornales中的一个科-单室萸科Masstixiaceae。  相似文献

五针白皮松(Pinus squamata X.W.Li)是上世纪90年代描述的一种中国特有松树,目前野外只有立木32株,处于极度濒危状态。前人认为这个种可能是白松亚属(subgen.Strobus)的白皮松(P.bungeana Zucc.ex Endl.)与松亚属(subgen.Pinus)的云南松(P.yunnanensis Franch.)的过渡类型,并将其归入白皮松组狐尾松亚组(subsect.Balfourianae)。本文试图在前人分子系统学工作的基础上,检测五针白皮松5个DNA片段,叶绿体基因rbcL、matK、rpl20-rps18间隔区和trn V内含子以及核糖体ITS,将五针白皮松放在整个松属中探讨其系统位置。4个叶绿体基因单独分析结果和它们的联合数据分析结果以及根据ITS得到的系统树均表明,五针白皮松是白皮松亚组(subsect.Gerardianae)的一个稳定成员,其可能的姐妹群是西藏白皮松(P.gerardiana Wall.)。对白皮松亚组的地理分布作了初步探讨。  相似文献

以玫瑰香葡萄(Vitisvinifera L.)为材料,克隆了含有叶绿体rbc L基因的3.1 kb Bam HⅠ片段,构建了该基因的限制性酶切图谱,测定了该基因的核苷酸序列。所测的核苷酸序列总长度为2004 bp,其中基因的编码区为1428 bp,编码一个含475 个氨基酸的蛋白质,其分子量约为53 kD;测定基因的5上游含启动子的部分共358 bp,包括- 10 区(TAAAAT)、- 35区(TTGCGC)和SD 序列(GGAGG);基因的3下游区共218 bp,含有3 个转录茎环终止结构。玫瑰香葡萄rbc L基因编码区的核苷酸序列与烟草、矮牵牛、菠菜、苜蓿、水稻和玉米之间的同源性分别为91.5% 、91.4% 、90.2% 、89.8% 、86.3% 和84.5% ;推导出的氨基酸序列的同源性分别为92.2% 、91.6% 、92.2% 、93.7% 、93.5% 和90.1% 。  相似文献

Abstract: Micromorphological analysis reveals that A. jahandiezii, which occurs as a local endemic taxon in the Grand Canyon du Verdon in southern France, shows substantial differences to A. bourgaei, from the eastern Mediterranean region, in the surface structure of perispores, sporangial indehiscence and the size of stomatal subsidiary cells. Nucleotide sequences and genetic distances (0.9 % nucleotide substitutions) of the rbc L gene support the view that both taxa are related but distinct sibling species. Both species form a monophyletic group together with Asplenium fontanum and A. majoricum.  相似文献

The phylogeny of Ectocarpus and Kuckuckia strains representing widely separated populations from both hemispheres was inferred from sequence analysis of the internal transcribed spacers of the nuclear ribosomal DNA (ITS 1—5.8S-ITS 2) and the spacer region in the plastid-encoded ribulose- bis -phosphate-carboxylase (RUBISCO) cistron (partial rbc L-spacer-partial rbc S ). Both sequences resulted in matching phylogenies, with the RUBISCO spacer region being most informative at the level of genera and species and the internal transcribed spacer sequences at the level of species and populations. Three major clades were formed by strains previously described by morphology and physiology as Kuckuckia, E. fasciculatus, and E. siliculosus, confirming the validity of these taxa . Ectocarpus and Kuckuckia are regarded as sibling taxa with respect to the outgroup species Feldmannia simplex, Hincksia mitchelliae, and Pilayella littoralis. The clade formed by sexual E. siliculosus strains and most asexual Ectocarpus strains was subdivided into several clades that are consistent with geographical races within E. siliculosus. The inferred phylogeny of Ectocarpus corresponds generally with results from cross-fertilization experiments, morphology, and lipid analysis. A hypothesis on the origin and dispersal of E. siliculosus suggests several natural dispersal events during periods of global cooling as well as recent and possibly anthropogenic dispersal events .  相似文献

基于叶绿体trnL_F序列单独分析以及trnL_F和rbcL序列联合分析重建了木通科的分子系统发育。本研究的系统发育拓扑结构与覃海宁和塔赫他间的族划分系统非常一致。猫儿屎族和串果藤族在系统发育树上位于本科的基部。由分布于南美的勃奎拉藤属和拉氏藤属组成的拉氏藤族得到了trnL_F序列分析 (10 0 % )和联合序列分析 (99% )的很好支持。木通族在两个分析里都得到了 10 0 %的靴带支持率。新建立的长萼木通属在trnL_F树上嵌套在木通属内 ;然而 ,在联合分析的树上 ,它与木通属形成姐妹群并得到很高的支持率。在系统发育上关系密切的 3个属 :牛藤果属、八月瓜属和野木瓜属之间的关系仍未解决。牛藤果与八月瓜在两个分析里都形成姐妹群 ,但支持率低。小花鹰爪枫嵌套在野木瓜属内 ,并与西南野木瓜形成姐妹群。木通族内这 3个属可能都不是单系 ,它们的属间界限和属的界定需要更多的分子和形态数据的进一步研究。  相似文献

Partial sequences (1032 bp) of the nuclear-encoded large ribosomal RNA gene (LSU) were determined for 16 gelidialean species, and analyzed separately and in combination with plastid rbcL and nuclear SSU gene sequences. The number of informative characters and levels of sequence divergence among taxa are intermediate in LSU sequences as compared to that for rbcL and SSU. Analyses of the separate LSU, and a combined LSU, SSU, and rbcL data sets have identified early-diverging lineages within the Gelidiales including Gelidiella, Pterocladia, Pterocladiella, and a lineage including Gelidium and species classified in other genera. The relationships among most gelidialean taxa are well-resolved and well-supported by analyses of the combined data; however, the relationships of Ptilophora and Capreolia remain unclear. It is speculated that these two lineages have diverged from a common ancestor over an evolutionarily short period of time.
