The present study focusses on allozyme variation in the commensal house mouseMus musculus, the pygmy field miceM. booduga andM. terricolor, and the spiny mouseM. platythrix. Genetic heterozygosity was estimated using a set of 24 polymorphic biochemical genetic markers. The extent of variability present inM. booduga, M. terricolor andM. platythrix has been compared with that in theM. musculus complex. Levels of allozyme variation at species level indicate thatM. musculus has the maximum heterogeneity, followed byM. booduga andM. terricolor, whileM. platythrix shows comparatively homogeneous genetic make-up. Gene frequency data have been used to trace phylogenetic relationships among these four species.  相似文献

Restriction maps of milochondrial DNA of the Indian pygmy field miceMus booduga andM. terricolor, and the house mouseM. musculus tytleri were determined with seven six-cutter restriction enzymes. The restriction map of the mitochondrial DNA of the laboratory mouseM. m. domesticus was used as a reference. Pairwise comparison was made of the mitochondrial DNAs for the presence or absence of the restriction sites, and per cent sequence divergence was calculated. The results show that the sequence divergence betweenbooduga andterricolor is 8-7% while thedomesticus-tytleri andbooduga-terricolor groups are divergent by 16-3%. The mtDNA sequence divergences we have obtained suggest that thebooduga-terricolor lineage might not have diverged before the Southeast Asiancervicolor-cookii-caroli lineage during evolution of these lineages of the subgenusMus as inferred by others earlier. On the other hand, it seems likely that these lineages evolved in parallel.  相似文献

Mus booduga and Mus terricolor both have 2n=40. Unlike M. booduga, with all acrocentric chromosomes, M. terricolor invariably has large submetacentric X and acrocentric Y due to an increase of heterochromatin. In contrast to the conservative karyotype of the co-existing sibling species booduga, three chromosome types of terricolor are found in different populations and their divergent karyotypes have autosomal heterochromatin variations established in the homozygous condition. The average genetic distance determined from electrophoretic study of 20 protein loci ranges from lowest (D=0.106) between chromosome types I & II to highest (D=0.185) between types II & III. In terricolor, booduga and M. m. tytleri high mean values of variations per locus (range A=1.604 to 1.928) and heterozygosity per individual per locus (range H=0.180 to 0.336) have been observed. Sequence divergence of 0.39 to 1.2%, calculated from restriction profiles of mtDNA, shows that the terricolor chromosome types have diverged recently. Hybridizations between type I females and type III males gave a preponderance of males in the F1 with varying degrees of sterility. The terricolor complex is an interesting system for critical probing for the role of heterochromatin in the process of speciation. MtDNA, protein loci and AT-rich musculus-related major and minor satellite DNA data indicate that progenitors of the booduga-terricolor lineage might have evolved simultaneously with the caroli-cookii-cervicolor lineage in the evolution of the subgenus Mus.  相似文献

Mus terricolor I, II and III are the three chromosomal species which differ in stable autosomal short-arm heterochromatin variations established in homozygous condition. Analysis of meiosis in the laboratory-generated F1 male hybrids from crosses (both ways) betweenM. terricolor I and II and betweenM. terricolor I and III shows high frequencies of pairing abnormalities at pachytene. The backcross (N3 generation) male hybrids betweenM. terricolor I and II have meiotic abnormalities as in the F1male hybrids, though to a lesser extent. They show difference in pairing abnormalities in the different karyotypic forms; the backcross hybrids heterozygous for the heterochromatic short arms have more anomalies compared to the homokaryotypic hybrids. This suggests a negative influence of the heterochromatin heterozygosity in meiotic pairing. The results indicate a role for heterochromatin variations in the development of a reproductive barrier in the speciatingM. terricolor complex.  相似文献

The Indian pygmy field mouse, Mus terricolor, is a tiny, yet economically and ecologically important crop pest found throughout South-East Asia. There are no systematic reports exploring its reproductive physiology. We report the presence of distinct periods of annual reproductive activity and quiescence in M. terricolor. Body weight in males and females, relative weights of testis, epididymis and seminal vesicle in males, ovarian and uterine weight in females, gonadal histomorphic changes, testicular and ovarian cholesterol, sialic acid in epididymis, fructose in seminal vesicle, uterine protein content, melatonin in males and females, testosterone in males, estradiol, and progesterone in females were studied over a period of three years in both wild-caught and lab-acclimated mice. The number of Graafian follicles and corpora lutea, and plasma estradiol and progesterone, along with relative weights of ovary and uterus in females exhibited a peak in the months of October–January, compared to June. Based on histomorphic and hormonal status, the major reproductively active season is the winter (short-day breeding). There is a brief period of sub-maximal reproductive activity in April. M. terricolor is reproductively inactive in the summer, monsoons, and autumn. The results establish M. terricolor as a seasonal breeder in the field, with interesting implications for pest management.  相似文献

Inhibitory and stimulatory adenosine receptors have been identified and characterized in both membranes and intact rat C6 glioma cells. In membranes, saturation experiment performed with [3H]DPCPX, selective A1R antagonist, revealed a single binding site with a K D = 9.4 ± 1.4 nM and B max = 62.7 ± 8.6 fmol/mg protein. Binding of [3H]DPCPX in intact cell revealed a K D = 17.7 ± 1.3 nM and B max = 567.1 ± 26.5 fmol/mg protein. On the other hand, [3H]ZM241385 binding experiments revealed a single binding site population of receptors with K D = 16.5 ± 1.3 nM and B max = 358.9 ± 52.4 fmol/mg protein in intact cells, and K D = 4.7 ± 0.6 nM and B max = 74.3 ± 7.9 fmol/mg protein in plasma membranes, suggesting the presence of A2A receptor in C6 cells. A1, A2A, A2B and A3 adenosine receptors were detected by Western-blotting and immunocytochemistry, and their mRNAs quantified by real time PCR assays. Giα and Gsα proteins were also detected by Western-blotting and RT-PCR assays. Furthermore, selective A1R agonists inhibited forskolin- and GTP-stimulated adenylyl cyclase activity and CGS 21680 and NECA stimulated this enzymatic activity in C6 cells. These results suggest that C6 glioma cells endogenously express A1 and A2 receptors functionally coupled to adenylyl cyclase inhibition and stimulation, respectively, and suggest these cells as a model to study the role of adenosine receptors in tumoral cells.  相似文献

Mitochondrial DNA control region of Mus terricolor, three aboriginal species M. spretus, M. macedonicus, M. spicilegus; the Asian lineage M. caroli, M. cervicolor, M. cookii; and the two house mice, M. musculus domesticus and M. m. castaneus were analysed to estimate the substitution rate, phylogenetic relationship and the probable time of divergence. Results showed that M. spretus, M. caroli and M. terricolor are highly diverged from each other (caroli/terricolor = 0.146, caroli/spretus = 0.147 and terricolor/spretus = 0.122), whereas M. spretus showed less divergence with two house mice species (0.070 and 0.071). Sequence divergence between M. terricolor and the Palearctic group were found to be ranging from 0.121 to 0.134. Phylogenetic analysis by minimum evolution, neighbour-joining, unweighed pair group method with arithmetic mean and maximum parsimony showed almost similar topology. Two major clusters were found, one included the Asian lineage, M. caroli, M. cookii and M. cervicolor and the other included the house mice M. m. domesticus, M. m. castaneus and the aboriginal mice M. macedonicus and M. spicilegus along with M. spretus, forming the Palearctic clade. M. terricolor was positioned between the Palearctic and Asian clades. Results showed that Palearctic-terricolor and the Asian lineages diverged 5.47 million years ago (Mya), while M. terricolor had split around 4.63 Mya from their ancestor. M. cervicolor, M. cookii and M. caroli diverged between 4.70 and 3.36 Mya, which indicates that M. terricolor and the Asian lineages evolved simultaneously. M. spretus is expected to have diverged nearly 2.9 Mya from their most recent common ancestor.  相似文献

A new species of fossil Mus (Rodentia, Mammalia) is reported from the Late Pleistocene fluvio-lacustrine deposits of the Kumaun sector of Indian Himalaya. The fossiliferous horizon, medium-coarse grained sand lenses embedded in the massive carbonaceous mud, is exposed at locality Dulam and is Late Pleistocene (ca. 30 ka) in age. A new species, Mus dulamensis n. sp., is proposed. Characterized by strongly distorted and moderately elongated M1 (about 170% of M2 length) with prominent conules in front of the t2, higher magnitude of stephanodonty (between the t1 and t4 and between the t3 and t6), moderately reduced M3 (about 56% of M2 length) and stronger connection between the cusps, this species is distinct from all other species of the genus and is included in the “booduga group”. I suggest that M. dulamensis n. sp. is comparatively more derived than M. auctor, M. flynni, M. jacobsi and Mus sp. and it may have given rise to M. booduga or M. dunni. It is further suggested that presently living M. booduga and M. dunni are more specialized than Mus dulamensis n. sp.  相似文献

Thewm7 haplotype of the major histocompatibility complex (MHC), derived from the Japanese wild mouseMus musculus molossinus, enhances recombination specific to female meiosis in theK/A interval of the MHC. We have mapped crossover points of fifteen independent recombinants from genetic crosses of thewm7 and laboratory haplotypes. Most of them were confined to a short segment of approximately 1 kilobase (kb) of DNA between theA 3 andA 2 genes, indicating the presence of a female-specific recombinational hotspot. Its location overlaps with a sex-independent hotspot previously identified in theMus musculus castaneus CAS3 haplotype. We have cloned and sequenced DNA fragments surrounding the hotspot from thewm7 haplotype and the corresponding regions from the hotspot-negative B10.A and C57BL/10 strains. There is no significant difference between the sequences of these three strains, or between these and the published sequences of the CAS3 and C57BL/6 strains. However, a comparison of this A3/A2 hotspot with a previously characterized hotspot in theE gene revealed that they have a very similar molecular organization. Each hotspot consists of two elements, the consensus sequence of the mouse middle repetitive MT family and the tetrameric repeated sequences, which are separated by 1 kb of DNA.The nucleotide sequence data reported in this paper have been submitted to the DNA Data Bank of Japan nucleotide sequence database and have been assigned the accession numbers d90007-9. Understanding some aspects of the ecology of the house mouseMus musculus Linnaeus, 1758 in an urban area may be crucial to decide materials or strategies for pest control. The aim of this study was to examine microhabitat use byM. musculus in vacant lots in the urban area of Río Cuarto, Córdoba, Argentina. We livetrapped M. musculus in five vacant lots in summer and autumn of 2006, and in four vacant lots in summer and autumn of 2007. Variables influencing the use and not-use of trapping stations by the house mouse by season were identified with a logistic regression.M. musculus presence was closely associated with vegetation variables. In three of four analyzed seasons, the house mouse presence was positively associated with high values of vegetation volume and negatively associated with traps located close to walls. To control mouse presence in vacant lots, it would be important to minimize habitat complexity through the maintenance of a short vegetation cover and the avoidance of the important plant species growth forM. musculus.  相似文献

Black rats are of outstanding interest in parasitology and infective disease analysis. We used chromosome paints from both the mouse(Mus musculus) and the Norway rat(Rattus norvegicus) to characterize the genome of two Black rat subspecies from Italy. Both subspecies have two large metacentrics (n. 1, 4) not present in the Norway rat (2n = 42).Rattus rattus rattus has a diploid number of 2n = 38, whileRattus rattus frugivorous has two small metacentric “supernumerary” or B chromosomes for a diploid number of 2n = 38 + 2B. The 21 mouse paints gave 38 signals on theR. r. rattus karyotype and 39 signals in theR. r. frugivorous karyotype. The two metacentrics, not present inR. norvegicus, were hybridized by mouse 16/1/17 and mouse 4/10/15. These chromosomes are homologous to: RRA1 = RNO 5/7, and RRA4 = RNO 9/11 and not “4/7” and “11/12” as previously reported. Furthermore, the synteny of Chr 13 of theR. r. frugivorous withR. norvegicus Chr 16 and mouse Chrs 8/14 is not complete, because there is a small pericentromeric insertion of RNO Chr 18 (mouse Chr 18). If we consider only the two metacentrics, RRA1 and RRA4, the principal differences betweenR. norvegicus andR. rattus, then we can propose the derived synteny of 124 genes in the black rat. A comparison of the Z index between rats and mice shows an acceleration of genomic evolution among genus, species, and subspecies. The chromosomal differences betweenR. r. rattus xR. r. frugivorous suggest that they may be classified as different species because hybrids would produce 50% unbalanced gametes.  相似文献

A complementary approach of stomach content and stable isotope analyses was used to characterize the foraging ecology and evaluate niche overlap between pygmy (Kogia breviceps) and dwarf (K. sima) sperm whales stranded on the U.S. mid‐Atlantic coast between 1998 and 2011. Food habits analysis demonstrated both species were primarily teuthophagous, with 35 species of cephalopods, and 2 species of mesopelagic fishes represented in their overall diets. Pianka's Index of niche overlap suggested high overlap between whale diets (On = 0.92), with squids from the families Histioteuthidae, Cranchidae, and Ommastrephidae serving as primary prey. Pygmy sperm whales consumed slightly larger prey sizes (mean mantle length [ML] = 10.8 cm) than dwarf sperm whales (mean ML = 7.8 cm). Mean prey sizes consumed by pygmy sperm whales increased with growth, but showed no trend in dwarf sperm whales. Significant differences were not detected in δ15N and δ13C values of muscle tissues from pygmy (10.8‰ ± 0.5‰, ?17.1‰ ± 0.6‰), and dwarf sperm whales (10.7‰ ± 0.5‰, ?17.0‰ ± 0.4‰), respectively. Isotopic niche widths also did not differ significantly and dietary overlap was high between the two species. Results suggest the feeding ecologies of the pygmy and dwarf sperm whales are similar and both species occupy equivalent trophic niches in the region.  相似文献

Single 2h administration of diazepam (benzodiazepine) in 3.5% ethanol solution was found to evoke advance and delay phase shifts in the locomotor activity rhythm in the field mouseMus booduga. Through such pulsed administration of diazepam at various phases of circadian rhythm a phase response curve could be constructed. Phase advance occurred during early subjective day (CT 2) and phase delays were observed in the remaining phases. The shape of the diazepam phase response curve is similar to the general shape of the phase response curves generated by intraperitoneal injections of other benzodiazepines in hamsters. The phase shifting action of diazepam may be explained by its agonistic action on the neurotransmitter gamma-aminobutyric acid.  相似文献

Paternal care is generally, although not exclusively, associated with monogamy in mammals. The Algerian mouse, Mus spretus, is a nocturnal murid living in xeric habitats in northern Africa, the Iberian peninsula and the south of France. We compared the amount of paternal care induced by removing young in Mus spretus and in the polygamous house mouse M. musculus domesticus in terms of the evolution of paternal care during the first 10-days post-partum and absolute (father alone) and relative (both parents) values. We then recovered indirect evidence of young being cared for by males from field data. The results were unambiguous in showing that the two species differed dramatically in paternal behaviour, with no variation being observed during the initial pup development. Mus spretus dedicated half of its time to caring from the young against less than 10% in M. m. domesticus. The former species showed the same number of retrieving acts and direct care bouts both in the presence and in the absence of the mother. Merging these results with field observations of spatial association “male-young” until subadult age, we drew conclusions on the likely occurrence of paternal care in M. spretus. Taken together with our previous results on socio-spatial organisation and pair bonding, we propose that social monogamy is advanced for this species.  相似文献

Abstract: High-affinity μ-opioid receptors have been solubilized from rat brain membranes. In most experiments, rats were treated for 14 days with naltrexone to increase the density of opioid receptors in brain membranes. Occupancy of the membrane-associated receptors with morphine during solubilization in the detergent 3-[(3-cholamidopropyl)dimethyl]-1-propane sulfonate appeared to stabilize the μ-opioid receptor. After removal of free morphine by Sephadex G50 chromatography and adjustment of the 3-[(3-cholamidopropyl)dimethyl]-1-propane sulfonate concentration to 3 mM, the solubilized opioid receptor bound [3H][d -Ala2,N-Me-Phe4,Gly-ol5]-enkephalin ([3H]DAMGO), a μ-selective opioid agonist, with high affinity (KD = 1.90 ± 0.93 nM; Bmax = 629 ± 162 fmol/mg of protein). Of the membrane-associated [3H]-DAMGO binding sites, 29 ± 7% were recovered in the solubilized fraction. Specific [3H]DAMGO binding was completely abolished in the presence of 10 µM guanosine 5′-O-(3-thiotriphosphate). The solubilized receptor also bound [3H]diprenorphine, a nonselective opioid antagonist, with high affinity (KD = 1.4 ± 0.39 nM, Bmax = 920 ± 154 fmol/mg of protein). Guanosine 5′-O-(3-thiotriphosphate) did not diminish [3H]diprenorphine binding. DAMGO at concentrations between 1 nM and 1 µM competed with [3H]diprenorphine for the solubilized binding sites; in contrast, [d -Pen2,d -Pen5]-enkephalin, a δ-selective opioid agonist, and U50488H, a κ-selective opioid agonist, failed to compete with [3H]diprenorphine for the solubilized binding sites at concentrations of <1 µM. In the absence of guanine nucleotides, the DAMGO displacement curve for [3H]diprenorphine binding sites better fit a two-site than a one-site model with KDhigh = 2.17 ± 1.5 nM, Bmax = 648 ± 110 fmol/mg of protein and KDlow = 468 ± 63 nM, Bmax = 253 ± 84 fmol/mg of protein. In the presence of 10 µM guanosine 5′-O-(3-thiotriphosphate), the DAMGO displacement curve better fit a one- than a two-site model with KD = 815 ± 33 nM, Bmax = 965 ± 124 fmol/mg of protein.  相似文献

Abstract: G protein activation mediated by serotonin 5-HT1A and 5-HT1B/D receptors in guinea pig brain was investigated by using quantitative autoradiography of agonist-stimulated [35S]GTPγS binding to brain sections. [35S]GTPγS binding was stimulated by the mixed 5-HT1A/5-HT1B/D agonist L694247 in brain structures enriched in 5-HT1A binding sites, i.e., hippocampus (+140 ± 14%), dorsal raphe (+70 ± 8%), lateral septum (+52 ± 12%), cingulate (+36 ± 8%), and entorhinal cortex (+34 ± 5%). L694247 caused little or no stimulation of [35S]GTPγS binding in brain regions with high densities of 5-HT1B/D binding sites (e.g., substantia nigra, striatum, central gray, and dorsal subiculum). The [35S]GTPγS binding response was antagonized by WAY100635 (10 µM) and methiothepin (10 µM). In contrast, the 5-HT1B inverse agonist SB224289 (10 µM) did not affect the L694247-mediated [35S]GTPγS binding response, and the mixed 5-HT1B/D antagonist GR127935 (10 µM) yielded a partial blockade. The distribution pattern of the [35S]GTPγS binding response and the antagonist profile suggest the L694247-mediated response in guinea pig brain to be mediated by 5-HT1A receptors. In addition to L694247, 8-hydroxy-2-(di-n-propylamino)tetralin, and flesinoxan also stimulated [35S]GTPγS binding; their maximal responses varied between 46 and 52% compared with L694247, irrespective of the brain structure being considered. Sumatriptan, rizatriptan, and zolmitriptan (10 µM) stimulated [35S]GTPγS binding in the hippocampus by 20–50%. Naratriptan, CP122638, and dihydroergotamine stimulated [35S]GTPγS binding to a similar level as L694247 in hippocampus, lateral septum, and dorsal raphe. It appears that under the present experimental conditions, G protein activation through 5-HT1A but not 5-HT1B/D receptors can be measured in guinea pig brain sections.  相似文献

To forecast the potential impact of plant community and dry-stone wall restoration on an insular population of the lesser white-toothed shrewCrocidura suaveolens Pallas, 1811, shrew and house mouseMus musculus Linnaeus, 1758 abundances were assessed in 3 anthropogenic habitats on Béniguet Island, Brittany, France, by a standardised annual trapping system checked yearly for 9 years and in 6 “natural” habitats by trap-lines. The standardised trapping system showed that abundances of both species fluctuated synchronously for nine years, suggesting that interactions between the species had little impact if any on abundances. Mice were trapped in all habitats, but shrews only in “stone” habitats except for rare occurrences in one damp depression. Ruderal habitat was rarely used by either species. Data suggest that on Béniguet Island: (1)M. musculus is associated with anthropogenic habitats but is not as strictly tied to them as at nearby continental sites; (2)C. suaveolens is synanthropic, as has been reported in continental northern France; (3) shingle beaches and seashore food resources are particularly important forC. suaveolens; (4) the relationship betweenC. suaveolens andM. musculus could not be determined by the experiments and, if it exists at all, appears to be more competitive than predatory. Grassland restoration is unlikely to affect shrew populations. Dry-stone wall restoration may temporarily affect shrews but should ultimately benefit them.  相似文献

When organisms are maintained under constant conditions of light and temperature, their endogenous circadian rhythms free run, manifesting their intrinsic period. The phases of these free-running rhythms can be shifted by stimuli of light, temperature, and drugs. The change from one free-running steady state to another following a perturbation often involves several transient cycles (cycles of free-running rhythm drifting slowly to catch up with the postperturbation steady state). Although the investigation of oscillator kinetics in circadian rhythms of both insects and mammals has revealed that the circadian pacemaker phase shifts instantaneously, the phenomenon of transient cycles has remained an enigma. We probed the phases of the transient cycles in the locomotor activity rhythm of the field mouse Mus booduga, evoked by a single light pulse (LP), using LPs at critically timed phases. The results of our experiments indicate that the transient cycles generated during transition from one steady state to another steady state do not represent the state of the circadian pacemaker (basic oscillator) controlling the locomotor activity rhythm in Mus booduga. Homozygous In(10)17Rk mice contain a paracentric inversion within Chromosome (Chr) 10 and exhibit a pygmy phenotype, suggesting that the distal inversion breakpoint is within the pygmy (pg) locus. In order to obtain the pygmy gene by positional cloning procedures, In(10)17Rk DNA was subjected to RFLP analysis with single-copy probes derived from the wild-type pygmy locus. This analysis identified a DNA polymorphism in the DBA/2J mouse strain on which the In(10)17Rk mutation was originally induced. A detailed characterization of this polymorphism revealed the presence of a novel, tandemly repeated DNA element. Copy number estimation experiments indicate that there are approximately 100,000 copies of this element in the haploid DBA/2J genome. PCR typing studies revealed the presence of the repeat at the pygmy locus of 6 of the 18 Mus domesticus strains analyzed. The absence of the repeat from the pygmy locus of 12 strains of the M. domesticus species and from the M. caroli, M. spretus, M. castaneus, and M. molossinus species suggests that the repeat could serve as a strain-specific hybridization probe in genetic mapping studies. Finally, the novel tandem DNA repeat is conserved in both rat and human genomes as indicated by Southern hybridization experiments.  相似文献

Adrenocorticotrophin secreting cells are identified in the hypophysis of the brown spiny mouseMus platythrix by conventional methods of light microscopy. Quantitative data showed that certain smaller acidophilic cells in thepars distalis, under conditions provoking their hypersecretion such as unilateral adrenalectomy and metopirone treatment, increase in number and size from the pre-existing corticotrophs. There is no evidence for the transmigration of these cells from the chromophobes, basophils or any other cell type. Thepars intermedia revealed two types of cells of which the type II cells are histochemically identical to adrenocorticotrophin secreting cells of thepars distalis  相似文献
