Cyclodextrins (CDs) can improve the productivity of steroid biotransformation by enhancing substrate solubility. CDs can be recycled by grafting them with appropriate carriers. Loofah fiber is an excellent grafting material for CDs, and can be applied to the biotransformation and recycling of β-cyclodextrin (β-CD). In this work, a technique for recycling β-CD in cortisone acetate (CA) biotransformation by Arthrobacter simplex CPCC 140451 was studied. Loofah fiber-grafted β-CD (LF-β-CD) was prepared using epichlorohydrin, which is a cross-linking agent. The grafting yield of β-CD was 74.8 mg g?1 dried fibers. LF-β-CD could increase the solubility of CA and enhance biotransformation. The initial conversion rate of CA was 1.5-fold higher than that of the blank group. LF-β-CD was also used in biocatalytic reactions for eight cycles, and it maintained the conversion ratio of CA at approximately 90%. Given the above positive results, LF-β-CD can be utilized in biotechnological recycling applications. This method can also be applied to CD derivatives and hydrophobic compounds.  相似文献

The complexation between two isomers of citral in lemongrass oil and varying types of cyclodextrins (CDs), α-CD, β-CD, and HP-β-CD, were studied by molecular modeling and physicochemical characterization. The results obtained revealed that the most favorable complex formation governing between citrals in lemongrass oil and CDs were found at a 1:2 mole ratio for all CDs. Complex formation between E-citral and CD was more favorable than between Z-citral and CD. The thermal stability of the inclusion complex was observed compared to the citral in the lemongrass oil. The release time course of citral from the inclusion complex was the diffusion control, and it correlated well with Avrami’s equation. The release rate constants of the E- and Z-citral inclusion complexes at 50 °C, 50% RH were observed at 1.32×10?2 h?1 and 1.43×10?2 h?1 respectively.  相似文献

CD437 (6-[3-(1-adamantyl)-4-hydroxyphenyl]-2-naphthalene carboxylic acid) is a novel synthetic retinoic acid derivative that has been shown to selectively induce apoptosis in human lung cancer cells. This compound, however, is limited in its application due to its low solubility in aqueous solutions. One technique for increasing the solubility and bioavailability of a cytotoxic agent is the formation of inclusion complexes with cyclodextrins. Herein, we report the formation and characterization of a 2:1 complex between β-cyclodextrin (β-CD) and CD437. It is shown that CD437 is a tight binder of β-CD with an overall association constant of 2.6 ± 0.6 × 107 M−2. In addition, we demonstrate (a) that β-CD-derived complexation enhances the aqueous solubility of CD437, and (b) that a significant increase in the toxicity of CD437 against a human lung adenocarcinoma cell line can be achieved by co-treatment with β-CD.  相似文献

Novel adsorbents that are composed of ligand, spacer and support were chemically synthesized and the two consecutive screenings of them made it possible to determine the adsorbents that were most recommendable for α- and β-cyclodextrin (CD) production. First, ligands of high adsorption selectivity for each CD were screened from among the candidates (carboxylic acids) that are tied in ionic bonds to two types of strongly basic anion exchange resins as support. Secondly, ligand derivatives (as ligand and spacer) were bound in covalent bonds onto chilosan beads as support and then, the most suitable spacer length for the CDs' adsorption selectivity and capacity were investigated. An optimal mol ratio of ligand to amino group of the beads was also examined. Stearic acid was the most effective ligand for α-CD, whereas cyclohexanepropanamide-n-caproic acid was the best for β-CD. Adsorption selectivity of adsorbents derived from carboxylic acids (stearic and/or palmitic) and chitosan beads was almost 100%, while their adsorption capacities were large enough to meet the demand for economic production and purification of CDs on an industrial scale.  相似文献

Enantioselective host-guest complexation between five racemic Ru(II) trisdiimine complexes and eight derivatized cyclodextrins (CDs) has been examined by NMR techniques. The appearance of non-equivalent complexation-induced shifts of between the Δ and Λ-enantionomers of the Ru(II) trisdiimine complexes and derivatized CDs is readily observed by NMR. In particular, sulfobutyl ether-β-cyclodextrin sodium salt (SBE-β-CD), R-naphtylethyl carbamate β-cyclodextrin (RN-β-CD), and S-naphtylethyl carbamate β-cyclodextrin (SN-β-CD) showed good enantiodiscrimination for all five Ru complexes examined, which indicates that aromatic and anionic derivatizing groups are beneficial for chiral recognition. The complexation stoichiometry between SBE-β-CD and [Ru(phen)3]2+ was found to be 1:1 and binding constants reveal that Λ-[Ru(phen)3]2+ binds more strongly to SBE-β-CD than the Δ-enantiomer. Correlations between this NMR method and separative techniques based on CDs as chiral discriminating agents (i.e., selectors) are discussed in detail.  相似文献

Knowledge of the complexation process of oxyresveratrol with β-cyclodextrin (β-CD) under different physicochemical conditions is essential if this potent antioxidant compound is to be used successfully in both food and pharmaceutical industries as ingredient of functional foods or nutraceuticals, despite its poor stability and bioavailability. In this paper, the complexation of oxyresveratrol with natural CDs was investigated for first time using RP-HPLC and mobile phases to which α-, β-, and γ-CD were added. Among natural CDs, the interaction of oxyresveratrol with β-CD was more efficient than with α- and γ-CD. The decrease in the retention times with increasing concentrations of β-CD (0–4 mM) showed that the formation constants (KF) of the oxyresveratrol/β-CD complexes were strongly dependent on both the water–methanol proportion and the temperature of the mobile phase employed. However, oxyresveratrol formed complexes with β-CD with a 1:1 stoichiometry in all the physicochemical conditions tested. Moreover, to obtain information about the mechanism of the oxyresveratrol affinity for β-CD, the thermodynamic parameters ΔG°, ΔH° and ΔS° were obtained. Finally, to gain information on the effect of the structure of different compounds belonging to the stilbenoids family on the KF values, the complexation of other molecules, resveratrol, pterostilbene and pinosylvin, was studied and compared with the results obtained for the oxyresveratrol/β-CD complexes.  相似文献

α-D-Mannosyl-maltotriose (Man-G3) were synthesized from methyl α-mannoside and maltotriose by the transfer action of α-mannosidase. (Man-G3)-βCD and (Man-G3)2-βCD were produced in about 20% and 4% yield, respectively when Aerobacter aerogenes pullulanase (160 units per 1 g of Man-G3) was incubated with the mixture of 1.6 M Man-G3 and 0.16 M βCD at 50°C for 4 days. The reaction products, (Man-G3)-βCD were separated to three peaks by HPLC analysis on a YMC-PACK A-323-3 column and (Man-G3)2-βCD were separated to several peaks by HPLC analysis on a Daisopak ODS column. The major product of (Man-G3)-βCDs was identified as 6-O-α-(63-O-α-D-mannosyl-maltotriosyl)-βCD by FAB-MS and NMR spectroscopies. The structures of (Man-G3)2-βCDs were analyzed by TOF-MS and NMR spectroscopies, and confirmed by comparison of elution profiles of their hydrolyzates by α-mannosidase and glucoamylase on a graphitized carbon column with those of the authentic di-glucosyl-βCDs. The structures of three main components of (Man-G3)2-βCDs were identified as 61,62-, 61,63- and 61,64-di-O-(63-O-α-D-mannosyl-maltotriosyl)-βCD.  相似文献

The configurational stability of (+)- and (−)-diethylpropion [(+)- and (−)-2-(diethyl)-1-phenyl-1-propanone or (+)- and (−)-DEP ] was investigated systematically from chemical, pharmaceutical, and pharmacological aspects. The enantiomeric ratio was monitored directly with a recently developed stability-indicating enantioselective HPLC method. In aqueous solutions, the rate of racemization increased non-linearly with increasing pH and with increasing phosphate buffer concentration. The racemization rate showed a positive slope with increasing temperature and decreasing ionic strength. The racemization rates of (+)- and (−)-DEP in the presence of cyclodextrins (CDs) did not differ significantly. CDs that were added to (+)- and (−)-DEP in a molar ratio 5:1 showed the following effects after dissolution in 10 mM phosphate buffer (final pH 6.7): sulfobutyl ether-β-CD (SBE-β-CD) and methylated-β-CD (Me-β-CD) retarded racemization; whereas hydroxypropyl-β-CD (HP-β-CD), acetyl-γ-CD (Ac-γ-CD), acetyl-β-CD (Ac-β-CD), γ-CD, and β-CD showed a weak destabilising effect. In contrast to the described CDs, α-CD distinctly accelerated the rate of racemization. The configurational stability of (+)- and (−)-DEP was also studied under physiological conditions. The half-life of racemization in heparinised human plasma was for both enantiomers determined to be approximately 23–25 min. In phosphate buffer (10 mM, pH 7.4), rac-DEP showed a high, but unselective affinity towards human α1-acid glycoprotein (orosomucoid) immobilised on silica (Chiral AGP). The rate of racemization of the free base of (−)-DEP dissolved in organic solutions generally increases with the polarity of the solvating agent. The accessible inclusion sites of insoluble copolymers containing β-cyclodextrin (β-CD) were studied in aqueous solutions by measuring the absorbance changes (decolourization) of phenolphthalein (phth) at pH 10.5. The various copolymers were reacted at different β-CD:crosslinker mole ratios with five individual types of crosslinker agents (epichlorohydrin (EP), sebacoyl chloride (SCL), terephthaloyl chloride (TCL), glutaraldehyde (GLU), and poly(acrylic) acid (PAA), respectively). The decolourization provided estimates of the 1:1 binding constants (K1) for the β-CD monomer/phth complex. Comparable values of K1 were measured for copolymer/phth complexes with highly accessible β-CD inclusion sites as compared with the 1:1 β-CD/phth complex. The surface accessibility of the β-CD inclusion binding sites for the polymers ranged from ∼10 to 72%. The observed variability of the inclusion sites was attributed to: (i) steric effects in the annular hydroxyl region of β-CD, (ii) the degree of crosslinking of the copolymer and (iii) the accessibility of the micropore sites within the copolymers. The Gibbs free energy (ΔG°) and site occupancy (θ) of phth adsorbed to the copolymer materials was estimated independently using the Sips isotherm model. The ΔG° values ranged between −27.6 and −30.9 kJ mol−1 for the copolymers and are in close agreement with the value for the 1:1 β-CD/phth complexes (ΔG° = −27 kJ mol−1) in aqueous solution.  相似文献

Efavirenz (EFV) is an oral antihuman immunodeficiency virus type 1 drug with extremely poor aqueous solubility. Thus, its gastrointestinal absorption is limited by the dissolution rate of the drug. The objective of this study was to characterize the inclusion complexes of EFV with β-cyclodextrin (β-CD), hydroxypropyl β-CD (HPβCD), and randomly methylated β-CD (RMβCD) to improve the solubility and dissolution of EFV. The inclusion complexation of EFV with cyclodextrins in the liquid state was characterized by phase solubility studies. The solid-state characterization of various EFV and CD systems was performed by X-ray diffraction, differential scanning calorimetry, and scanning electron microscopy analyses. Dissolution studies were carried out in distilled water using US Pharmacopeia dissolution rate testing equipment. Phase solubility studies provided an AL-type solubility diagram for β-CD and AP-type solubility diagram for HPβCD and RMβCD. The phase solubility data enabled calculating stability constants (K s) for EFV-βCD, EFV-HPβCD, and EFV-RMβCD systems which were 288, 469, and 1,073 M−1, respectively. The physical and kneaded mixtures of EFV with CDs generally provided higher dissolution of EFV as expected. The dissolution of EFV was substantially higher with HPβCD and RMβCD inclusion complexes prepared by the freeze drying method. Thus, complexation with HPβCD and RMβCD could possibly improve the dissolution rate-limited absorption of EFV.  相似文献

Di-O-α-maltosyl-β-cyclodextrin ((G2)2-β-CD) was synthesized from 6-O-α-maltosyl-β-cyclodextrin (G2-β-CD) via a transglycosylation reaction catalyzed by TreX, a debranching enzyme from Sulfolobus solfataricus P2. TreX showed no activity toward glucosyl-β-CD, but a transfer product (1) was detected when the enzyme was incubated with maltosyl-β-CD, indicating specificity for a branched glucosyl chain bigger than DP2. Analysis of the structure of the transfer product (1) using MALDI-TOF/MS and isoamylase or glucoamylase treatment revealed it to be dimaltosyl-β-CD, suggesting that TreX transferred the maltosyl residue of a G2-β-CD to another molecule of G2-β-CD by forming an α-1,6-glucosidic linkage. When [14C]-maltose and maltosyl-β-CD were reacted with the enzyme, the radiogram showed no labeled dimaltosyl-β-CD; no condensation product between the two substrates was detected, indicating that the synthesis of dimaltosyl-β-CD occurred exclusively via transglycosylation of an α-1,6-glucosidic linkage. Based on the HPLC elution profile, the transfer product (1) was identified to be isomers of 61,63- and 61,64-dimaltosyl-β-CD. Inhibition studies with β-CD on the transglycosylation activity revealed that β-CD was a mixed-type inhibitor, with a Ki value of 55.6 μmol/mL. Thus, dimaltosyl-β-CD can be more efficiently synthesized by a transglycosylation reaction with TreX in the absence of β-CD. Our findings suggest that the high yield of (G2)2-β-CD from G2-β-CD was based on both the transglycosylation action mode and elimination of the inhibitory effect of β-CD.  相似文献

In this work, a phage-displayed peptide library was applied to identification of β-cyclodextrin (CD)-binding peptide tag, capable of being combined to target peptides or proteins in a homogeneous way by established methods such as peptide synthesis and the recombinant DNA technique. Four enriched sequences were obtained after five rounds of biopanning against polymeric β-CD beads. One of the sequences showed high binding affinity to β-CD beads with a dissociation constant of approximately 7 × 10–6 M. The β-CD-binding sequence was used for immobilization of a hepatitis C virus (HCV) antigenic peptide on β-CD beads. The functionalized β-CD beads were successfully used for immunoassay of anti-HCV antibody with a detection limit of 1 ng. These results demonstrate that the identified peptide sequence has the potential of being used as an affinity tag to β-CD-containing surfaces.  相似文献

Reported herein are the systematic design and characterization of several novel polyurethane (PU) copolymers containing a macrocyclic porogen (β-cyclodextrin; β-CD). These copolymers were synthesized from the reaction between β-CD with different types of diisocyanate linker molecules (e.g., 1,6-hexamethylene diisocyanate (HDI), 4,4′-dicyclohexylmethane diisocyanate (CDI), 4,4′-diphenylmethane diisocyanate (MDI), 1,4-phenylene diisocyanate (PDI) and 1,5-naphthalene diisocyanate (NDI)) at variable synthetic conditions. The copolymers were characterized using diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS), solid state 13C CP-MAS NMR, 1H/13C solution NMR spectroscopy, thermogravimetric analysis (TGA) and elemental analyses (CHN). The PU copolymers were generally insoluble in water and the optimal preparation of copolymer materials for sorption-based applications is for β-CD/linker synthetic mole ratios from 1:1 to 1:3. The practical upper limit of the crosslink density (∼1:7, β-CD/linker) depends on the steric bulk of the cross linker units.  相似文献

Bacillusfirmus strain 37 produces the cyclomaltodextrin glucanotransferase (CGTase) enzyme and CGTase produces cyclodextrins (CDs) through a starch cyclization reaction. The strategy for the cloning and expression of recombinant CGTase is a potentially viable alternative for the economically viable production of CGTase for use in industrial processes. The present study used Bacillus subtilis WB800 as a bacterial expression host for the production of recombinant CGTase cloned from the CGTase gene of B. firmus strain 37. The CGTase gene was cloned in TOPO-TA® plasmid, which was transformed in Escherichia coli DH5α. The subcloning was carried out with pWB980 plasmid and transformation in B. subtilis WB800. The 2xYT medium was the most suitable for the production of recombinant CGTase. The enzymatic activity of the crude extract of the recombinant CGTase of B. subtilis WB800 was 1.33 µmol β-CD/min/mL, or 7.4 times greater than the enzymatic activity of the crude extract of CGTase obtained from the wild strain. Following purification, the recombinant CGTase exhibited an enzymatic activity of 157.78 µmol β-CD/min/mL, while the activity of the CGTase from the wild strain was 9.54 µmol β-CD/min/mL. When optimal CDs production conditions for the CGTase from B. firmus strain 37 were used, it was observed that the catalytic properties of the CGTase enzymes were equivalent. The strategy for the cloning and expression of CGTase in B. subtilis WB800 was efficient, with the production of greater quantities of CGTase than with the wild strain, offering essential data for the large-scale production of the recombinant enzyme.

An intensive and systematic investigation of the oxidation of cholesterol (CL) to cholest-4-en-3-one (CN) by Rhodococcus erythropolis was undertaken in the presence of natural and chemically modified cyclodextrins (CDs) in a stirred bioreactor. The biotransformation was found to be strongly affected by the mode of addition of the natural CDs. While simultaneous addition of CL with either - or -CD led to a limited enhancement effect, the microbial oxidation of - and -CD complexes of CL was totally inhibited. In contrast, the alkylated CDs- dimethyl-, trimethyl- and hydroxypropyl--CD exhibited a remarkable enhancement of the microbial oxidation, irrespective of their mode of addition. The performance of the alkylated CDs was interpreted in the light of the measured phase solubility diagrams of CL and CN. It was thus shown that unlike the low solubilising power of hydroxypropyl--CD, dimethyl- and trimethyl--CD at 90 mm each, dissolved 9.3 and 8.7 g/l of CL and CN, respectively. Further investigation focused on the formation of CD complexes with CL and CN, analysed by X-ray powder diffractometry, differential scanning calorimetry and 1H-nuclear magnetic resonance. It was thus shown that -CD forms a 2:1 CD:CL and CD:CN water-insoluble complexes. A mechanism of the biotransformation in homogeneous and heterogeneous CD media was presented while suggesting a direct interaction of the CD-substrate complex with microbial cells. Alkylation of cyclomaltohexaose (α-cyclodextrin, α-CD) with allyl or cinnamyl bromide, followed by peracetylation of remaining hydroxyl groups and separation of isomers, resulted in the set of peracetylated 2I-O-, 3I-O- and 6I-O-alkylated α-CDs in up to 27% yields. Ozonolysis or oxidative cleavage of peracetylated allyl or cinnamyl derivatives resulted in a complete set of peracetylated 2I-O-, 3I-O- and 6I-O-formylmethyl or carboxymethyl derivatives that are useful precursors for preparation of regioselectively monosubstituted derivatives of α-CD. Moreover, a quick method to recognize single 2I-O-, 3I-O- and 6I-O-monosubstituted peracetylated CDs from one another using only their 1H NMR spectra has been proposed.  相似文献

The aim of this work was to study the influence of β-cyclodextrin (β-CD) on the biopharmaceutic properties of diclofenac (DCF). To this purpose the physicochemical characterization of diclofenac-β-cyclodextrin binary systems was performed both in solution and solid state. Solid phase characterization was performed using differential scanning calorimetry (DSC), powder x-ray diffractometry (XRD), and Fourier transform infrared spectroscopy (FTIR). Phase solubility analyses, and in vitro permeation experiments through a synthetic membrane were performed in solution. Moreover, DCF/β-CD interactions were studied in DMSO by1H nuclear magnetic resonance (NMR) spectroscopy. The effects of different preparation methods and drug-to-β-CD molar ratios were also evaluated. Phase solubility studies revealed 1∶1 M complexation of DCF when the freeze-drying method was used for the preparation of the binary system. The true inclusion for the freeze-dried binary system was confirmed by1H NMR spectroscopy, DSC, powder XRD, and IR studies. The dissolution study revealed that the drug dissolution rate was improved by the presence of CDs and the highest and promptest release was obtained with the freeze-dried binary system. Diffusion experiments through a silicone membrane showed that DCF diffusion was higher from the saturated drug solution (control) than the freeze-dried inclusion complexes, prepared using different DCF-β-CD molar ratios. In the present study, we investigated the feasibility of preparation of novel controlled release systems for the delivery of essential oil used as ambient odors. The inclusion interactions of cyclodextrins (CDs) and β-cyclodextrin polymers with linalool and camphor in Lavandula angustifolia essential oil were investigated by static headspace gas chromatography (SH-GC). The stability constants with monomeric CD derivatives were determined for standard compounds and for the compounds in essential oil. All studied CDs and CD polymers reduce the volatility of the aroma compounds and stable 1:1 inclusion complexes are formed. The retention capacity of the CD derivatives was measured in static experiments. The feasibility of preparation of novel controlled release systems for the delivery of fragrances was investigated by multiple headspace extraction (MHE) experiments.  相似文献
