目的:从青藏高原冰川雪中筛选出一菌多酶的菌株.方法:对恢复出的4个细菌,通过平板透明法研究其产淀粉酶、脂肪酶和蛋白酶的特性.结果:LHG-C-9为惟一可以产淀粉酶的菌株,所产脂肪酶活性最高.4个菌株均不产蛋白酶.结论:LHG-C-9最适生长温度为15℃,属于耐冷菌.对该菌所产淀粉酶和脂肪酶的性质进行了初步研究,其随产淀粉酶的最适作用温度为50℃;最佳产酶pH值为7.0,该pH值所产酶活为83.9U/mL;在60℃的高温下温浴10min后酶活为0%.该菌株所产脂肪酶的最适作用温度为20℃;最佳产酶pH值为7.0,该pH值所产酶活为9.2U/mL;50℃温浴1h后酶活力不足34%.  相似文献

[目的]以纤维素为唯一碳源,从四川省阿坝自治州黄龙沟的高山低温环境中分离筛选产纤维素酶的耐冷菌,并研究菌株的产酶特征.[方法]根据菌株的ITS序列分析及形态特征,对菌株进行鉴定.利用DNS法测定纤维素酶酶活性.[结果]从四川省阿坝自治州黄龙沟的高山腐殖土中筛选出一株产纤维素酶的耐冷菌HD1031,经鉴定该菌为玫红假裸囊菌(Pseudogymnoascus roseus).该菌可在4℃-25℃生长,最适生长温度为16℃-17℃.该菌在以微晶纤维素和玉米芯粉为碳源、硫酸铵和Tryptone为氮源的培养基中,17℃、160 r/min摇瓶发酵8d后产生纤维素酶,其中内切葡聚糖酶酶活为366.67 U/mL,滤纸酶酶活87.6 U/mL,β-葡萄糖苷酶酶活90.8 U/mL,酶最适反应pH为6.0,最适反应温度为50℃.[结论]筛选获得一株产纤维素酶的耐冷菌HD1031,此菌株所产纤维素酶在20℃-40℃下活性较高,对热敏感,具有低温纤维素酶的特点.  相似文献

高温蛋白酶产生菌的筛选及其产酶条件和酶学性质分析   总被引:2,自引:0,他引:2  
从徂徕山温泉附近土样中分离到9株产高温蛋白酶菌株,选取一株碱性蛋白酶高产菌株L7为出发菌株,进行显微形态、16S rRNA基因序列分析,将其初步鉴定为短芽孢杆菌(Brevibacillus sp.)。研究该菌株发酵条件,确定产酶的最佳培养基组成为葡萄糖40 g/L,蛋白胨20 g/L,磷酸氢二钠1.4 g/L,氯化钙0.6 g/L,硫酸镁0.4 g/L,通过培养基优化,酶活达到103.08 U/mL。最佳培养条件为250 mL三角烧瓶中装液量50 mL、pH8.0、培养温度为55℃、培养时间为24 h。对该菌株酶学性质研究,L7菌株所产高温蛋白酶的最适温度为55℃,最适pH为10,并且具有良好的温度稳定性和pH稳定性,酶活性受PMSF强烈抑制。  相似文献

灵芝作为传统中药,具有重要的医药和经济价值。本研究以一株野生灵芝属真菌BSU01为实验材料,通过形态特征和ITS序列聚类分析对其进行了鉴定,探讨了该菌株菌丝生长的最适碳源、氮源、C/N、pH及温度等生物学特征,以及该菌株产木质纤维降解素酶的能力。结果表明,菌株BSU01的形态特征与有柄灵芝相符;且与有柄灵芝ITS序列一致性达99%,并在系统发育树上聚在一起;菌株BSU01菌丝生长最适碳源为果糖或者葡萄糖,氮源为酵母提取物,C/N比为20/1到30/1,温度为30℃,pH值为5.5;菌株BSU01的漆酶、木质素过氧化物酶和Mn依赖过氧化物酶分别为12.13 U/L、0.52 U/L和0.33 U/L;CMCase和木聚糖酶酶活分别为7.14 U/mL和1.88 U/m L。本研究结果将为该菌的进一步开发利用提供数据参考。  相似文献

目的:筛选鉴定产胆固醇氧化酶的菌株并对其酶的性质及发酵条件进行初步的研究。方法:利用唯一碳源的胆固醇平板筛选,酶活测定比较得酶活力最高的菌株;生理生化试验结合16S rDNA序列分析鉴定其种属,单因素及正交实验优化培养基及发酵条件。结果:所得菌株H4与产不动杆菌(Acinetobacter)有最近的亲缘关系,其胆固醇氧化酶作用的最适温度和pH分别为37℃和8.0,金属离子Mg2+、Zn2+、Fe2+对该酶具有一定激活作用,菌株产酶的最适培养基为(g/L):胆固醇1.5,蔗糖5,蛋白胨7,硝酸铵3,吐温1.0,pH7.5;最适培养条件为33℃,15mL培养基/100mL三角瓶,摇床培养(200r/min)48h,优化后发酵液酶活达135.8U/L。结论:获得了1株产胆固醇氧化酶的菌株H4,并初步鉴定为不动杆菌(Acinetobacter)。  相似文献

本实验从虫生真菌中筛选出金龟子绿僵菌M a83菌株,它的几丁质酶合成能力最强。其产酶的适宜条件是,碳源为胶体几丁质加葡萄糖,氮源为NaNO3,培养温度为28℃,培养基起始pH 6.0;接种量为5 mL液态种,最适装液量为5 mL,添加维生素C可以提高酶活;正交实验表明培养因子的最佳组合是:NaNO31 g/L,胶体几丁质0.6 g/L,酵母膏0.05 g/L,葡萄糖0.10 g/L。根据液态培养产酶过程结果可知,当M a83菌培养6天时,几丁质酶活力达到8.1 U/mL。  相似文献

从渤海湾盐碱地被油污染的土样中分离筛选出6株产热稳定碱性脂肪酶菌株。其中菌株1-7产脂肪酶能力较强,其最高酶活为8.67U/mL。根据其16S rDNA序列分析和Biolog生理生化分析,初步鉴定为醋酸钙不动杆菌(Acinetobacter calcoaceticus)。初步酶学性质研究表明该菌所产脂肪酶具有较好的热稳定性,最适作用pH为9.0。摇瓶实验表明,该菌株最适产酶培养基为(g/L):玉米粉10,黄豆饼粉20,K_2HPO_4 1,NaNO_3 5,橄榄油10。最适产酶条件为:初始pH 8.0,培养温度37℃,培养时间29 h。接种量为2%,250 mL摇瓶装液量为30 mL。  相似文献

采用黑曲霉 (Aspergillusniger)GD 6液体发酵生产纤维素酶 ,研究了碳源、氮源、培养基起始 pH值、接种量、摇床转速、通气量对该菌株产纤维素酶活力的影响。结果表明 ,GD 6的最适发酵温度为 2 8~ 3 0℃ ,产酶pH为 5 .5~ 6.0 ,摇床最适转速为 1 5 0r/min ,最佳接种量为 1 0 %。在以 6.0 %稻草粉为碳源、1 %豆饼粉为氮源时产酶活力最高。在最适培养条件下 ,发酵周期为 1 2 0h,发酵液中CMC酶活为 1 88.6U/mL ,FP酶活为 2 7.0U/mL。  相似文献

通过刚果红染色产脱色圈初筛出能分解纤维素的菌株,再利用DNS法分别测定纤维素酶的酶活,得到分解纤维素能力最强的一株真菌Lv-1。该菌株菌丝为黄绿色,孢子为深绿色,有多分支的无隔菌丝,长孢子囊孢子,用DNS法测定其酶活为62.42 U/m L,后对其进行了产酶条件优化。经单因素试验和正交试验得到该菌的最佳产酶条件为:在以10 g/L羧甲基纤维素钠为碳源,4 g/L蛋白质,2 g/L硫酸铵为氮源,1 g/L硫酸二氢钾为无机盐的最适产酶培养基中,初始p H为6.0,培养温度37℃,装液量100 m L/250 m L,发酵36 h。在此发酵条件下,其产酶活力可达96.898U/m L,试验结果显示,该菌株在产纤维素酶能力上具有显著优势,且菌株Lv-1产酶活力较优化前高出34.478 U/m L,提高了55.24%。  相似文献

目的 从黑胸散白蚁肠道内筛选获得具有降解纤维素性能的菌株,并对菌株最佳产酶条件进行优化.方法 采用筛选性培养基进行筛选,通过培养性状、显微观察及16S rDNA部分片段同源性分析进行菌种鉴定,利用正交试验优化该菌株的最佳产酶培养基配方以及单因子试验优化产酶培养条件.结果 通过鉴定,获得的菌株属柠檬酸杆菌属(Citrobacter sp.B03),最适产酶的碳氮源为CMC-Na和蛋白胨.该菌株最佳产酶培养基的配方为CMC-Na5.0 g/L、蛋白胨5.0 g/L、NH4Cl 0.6 g/L、KH2P04 0.9 g/L、MgSO4 0.9 g/L;最佳产酶培养条件为起始pH 5.0,温度35℃,装液量20~ 30 mL/150 mL.结论 经过优化,可将该菌株产生的纤维素酶酶活力从0.184 U/mL提高到0.311 U/mL,该研究结果对纤维素酶的工业开发具有一定的指导意义.  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
