In the last three years, more than 70,000 scientific articles have been published in peer reviewed journals on the application of histochemistry in the biomedical field: most of them did not appear in strictly histochemical journals, but in others dealing with cell and molecular biology, medicine or biotechnology. This proves that histochemistry is still an active and innovative discipline with relevance in basic and applied biological research, but also demonstrates that especially the small histochemical journals should likely reconsider their scopes and strategies to preserve their authorship. A review of the last three years volumes of the European Journal of Histochemistry, taken as an example of a long-time established small journal, confirmed that the published articles were widely heterogeneous in their topics and experimental models, as in this journal''s tradition. This strongly suggests that a journal of histochemistry should keep its role as a forum open to an audience as broad as possible, publishing papers on cell and tissue biology in a wide variety of models. This will improve knowledge of the basic mechanisms of development and differentiation, while helping to increase the number of potential authors since scientists who generally do not use histochemistry in their research will find hints for the applications of histochemical techniques to novel still unexplored subjects.Key words: Basic and applied histochemistry  相似文献

Histochemistry provides the unique opportunity to detect single molecules in the very place where they exert their structural roles or functional activities: this makes it possible to correlate structural organization and function, and may be fruitfully exploited in countless biomedical research topics. Aiming to estimate the impact of histochemical articles in the biomedical field, the last few years citations of articles published in a long-established histochemical journal have been considered. This brief survey suggests that histochemical journals, especially the ones open to a large spectrum of research subjects, do represent an irreplaceable source of information not only for cell biologists, microscopists or anatomists, but also for biochemists, molecular biologists and biotechnologists.Key words: Basic and applied histochemistry, Biomedical research  相似文献

Journal impact factors have become an important criterion to judge the quality of scientific publications over the years, influencing the evaluation of institutions and individual researchers worldwide. However, they are also subject to a number of criticisms. Here we point out that the calculation of a journal’s impact factor is mainly based on the date of publication of its articles in print form, despite the fact that most journals now make their articles available online before that date. We analyze 61 neuroscience journals and show that delays between online and print publication of articles increased steadily over the last decade. Importantly, such a practice varies widely among journals, as some of them have no delays, while for others this period is longer than a year. Using a modified impact factor based on online rather than print publication dates, we demonstrate that online-to-print delays can artificially raise a journal’s impact factor, and that this inflation is greater for longer publication lags. We also show that correcting the effect of publication delay on impact factors changes journal rankings based on this metric. We thus suggest that indexing of articles in citation databases and calculation of citation metrics should be based on the date of an article’s online appearance, rather than on that of its publication in print.  相似文献

More than 25 years have passed since publication of the first comprehensive multi-authored landmark volume on the population biology and evolution of clonal organisms (Jackson et al. 1985). Since then, no less than eight symposium volumes or special issues have appeared in scientific journals reporting on advances in the field of clonal plant research, indicating that the study of clonal organisms has remained an important topic in ecological research. The three most recent overviews were published in special issues of this journal (Stuefer et al. 2000; Tolvanen et al. 2004; Sammul et al. 2008), and these are now supplemented with a fourth special issue of Evolutionary Ecology. The articles published here reflect some of the most important contributions to a workshop on clonal plant biology held in Leuven (Belgium) in July 2009 and they illustrate some major advances that have been made over the last few years. In the following paragraphs, we first summarize some representative contributions to the current issue, and second, we put forward some personal ideas about promising and underexplored research lines in clonal plant research.  相似文献

The distribution of acid phosphatase in liver cirrhosis, as well as in its reverse development, was investigated in mice using histochemistry and electron histochemistry methods. Histochemistry demonstrated a sharp activity increase of acid phosphatase (as compared with the same in the material of partial hepatectomy) in liver cells (especially hepatocytes) during liver cirrhosis regression 10 days after a partial hepatectomy. Electron histochemistry has shown the enzyme withdraw out of hepatocytes and connective tissue cells of fibrotic stratum in the extra-cell medium. The reaction product localized on the neighbouring collagen fibres giving evidence that during reverse development of liver cirrhosis the lisosomal enzyme release from specified cells by means of exocytosis and they are involved in the lysis of collagen.  相似文献

The journal impact factor is an annually calculated number for each scientific journal, based on the average number of times its articles published in the two preceding years have been cited. It was originally devised as a tool for librarians and publishers to provide information about the citation performance of a journal as a whole, but over the last few decades it has increasingly been used to assess the quality of specific articles and the research performance of individual investigators, institutions, and countries. In addition to this clear abuse of the journal impact factor, several conceptual and technical issues limit its usability as a measure of journal reputation, especially when journals are compared across different fields. An author’s decision regarding the suitability of a scholarly journal for publication should, therefore, be based on the impact that this journal makes in the field of research, rather than on the journal impact factor.  相似文献

Summary Bridging between the two bodies of cell science, histochemistry has brought together knowledge of morphological structures and biochemical constituents and provided insight into the function of one or the other. Where it has localized an enzyme, hormone or other entity of known biological activity to a cell type, histochemistry has contributed insight into the cell's function. By detecting heterogeneity in the content of an enzyme or glycoconjugate within a presumed uniform population of cells, the histochemical approach has differentiated among these cells subtypes with demonstrated or presumed differences in function. On the other hand, in instances where it has located an isozyme, antigen, glycoconjugate or other entity of uncertain significance in a cell or organelle of known function, histochemistry has suggested a possible role for the constituent related to that of the structure. Histochemical examination provides the observer with a different view of body tissues and their composition from that obtained by strictly morphological or chemical techniques. Information acquired through this advantage is cited wherein an immunohistochemical method disclosed previously undiscovered neural organs and carbohydrate histochemistry detected previously unrecognized glycoconjugates. This article explores forensic anthropological and bioarchaeological publishing patterns in the American Anthropologist (n.s.). Early contributions by Harris Hawthorne Wilder on both subjects are considered in detail, including previously unrecognized discussions of taphonomic variables. Articles on forensic anthropology in the American Anthropologist appear only during the first four decades of publication. While relatively well represented in early issues, bioarchaeological articles decrease in number over time. Comparative data from six other journals suggest that their presence has had a profound effect on the American Anthropologist . The impact of the "new" physical anthropology on publishing patterns in bioarchaeology is also assessed. 在2006年10月26日,期待已久的蜜蜂的基因组序列在英国杂志《自然》上发表了。这一事件对于全世界蜜蜂生物学家来说,标志着蜜蜂研究“后基因组时代”的开始。这一重大项目由倍乐医学院完成,耗资800万美元,历时2年。而实际上对于基因组的注释又经过近2年的时间,由代表15个国家、来自64个实验室的170名科学家完成。这一等待是值得的基因组序列最终发表后,生物学家通过互联网能做他们自己的“数据采掘”,用这一可得的公共资源验证自己的假说。事实上,在蜜蜂基因组文章在《自然》上发表的当周,其姊妹刊上发表的约50篇相关论文中,有一半以上是基于生物信息学的研究,这些刊物包括《科学》、《基因组研究》、《美国国家科学院院报》及《昆虫分子生物学》。蜜蜂与人类享有一些共性劳动分工,复杂的通信系统包括语言,发达的保卫和防御系统,精妙的建筑以及为了共同利益自我牺牲的特点。由于有这些共性,蜜蜂基因组的发表将使科学家不仅能够深入研究蜜蜂生物学,而且能深入了解我们人类自身的生物学。  相似文献

The field of developmental biology has a history that spans the last 500 years. Within the last 10 years, our understanding of developmental mechanisms has grown exponentially by employing modern techniques of genetics and molecular biology, frequently combined with experimental embryology and the use of molecular markers, rather than solely morphology, to identify critical populations of cells and their state of differentiation. Three main principles have emerged. First, mechanisms of development are highly conserved, both among developing rudiments of a variety of organ systems and among diverse organisms. This conservation occurs both at the level of tissue and cellular mechanisms, and at the molecular level. Second, the development of organ rudiments is influenced by surrounding tissues through interactions called inductive interactions. Such interactions are mediated by highly conserved growth factors and signaling systems. Third, development is a life-long process and can be reawakened in events such as wound healing and regeneration, and in certain diseases. Advances in understanding normal development provide hope that diseases in which development runs amuck, such as cancer, may soon be preventable and fully treatable. Histochemistry and cytochemistry of glucose-6-phosphate dehydrogenase has found many applications in biomedical research. However, up to several years ago, the methods used often appeared to be unreliable because many artefacts occurred during processing and staining of tissue sections or cells. The development of histochemical methods preventing loss or redistribution of the enzyme by using either polyvinyl alcohol as a stabilizer or a semipermeable membrane interposed between tissue section and incubation medium, has lead to progress in the topochemical localization of glucose-6-phosphate dehydrogenase. Optimization of incubation conditions has further increased the precision of histochemical methods. Precise cytochemical methods have been developed either by the use of a polyacrylamide carrier in which individual cells have been incorporated before staining or by including polyvinyl alcohol in the incubation medium. In the present text, these methods for the histochemical and cytochemical localization of glucose-6-phosphate dehydrogenase for light microscopical and electron microscopical purposes are extensively discussed along with immunocytochemical techniques. Moreover, the validity of the staining methods is considered both for the localization of glucose-6-phosphate dehydrogenase activity in cells and tissues and for cytophotometric analysis. Finally, many applications of the methods are reviewed in the fields of functional heterogeneity of tissues, early diagnosis of carcinoma, effects of xenobiotics on cellular metabolism, diagnosis of inherited glucose-6-phosphate dehydrogenase deficiency, analysis of steroid-production in reproductive organs, and quality control of oocytes of mammals. It is concluded that the use of histochemistry and cytochemistry of glucose-6-phosphate dehydrogenase is of highly significant value in the study of diseased tissues. In many cases, the first pathological change is an increase in glucose-6-phosphate dehydrogenase activity and detection of these early changes in a few cells by histochemical means only, enables prediction of other subsequent abnormal metabolic events. Analysis of glucose-6-phosphate dehydrogenase deficiency in erythrocytes has been improved as well by the development of cytochemical tools. Heterozygous deficiency can now be detected in a reliable way. Cell biological studies of development or maturation of various tissues or cells have profited from the use of histochemistry and cytochemistry of glucose-6-phosphate dehydrogenase activity.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Advances in the field of histochemistry, a multidisciplinary area including the detection, localization and functional characterization of molecules in single cells and complex tissues, often drives the attainment of new knowledge in the broadly defined discipline of cell biology. These two disciplines, histochemistry and cell biology, have been joined in this journal to facilitate the flow of information with celerity from technical advancement in histochemical procedures, to their utilization in experimental models. From the multitude of oral presentations at major medical meetings, the most informative and highest-quality studies make it to full publication in peer-reviewed journals. The rate of publication may be regarded as an indicator of the scientific level of the meeting. Study of the publication rates of consecutive annual meetings allows for the evaluation of the consistency of the scientific level of these meetings and for comparison with publication rates of other meetings in the same field of interest. To grade how useful any publication is to other authors, one can furthermore measure how frequently they cite it in their own publications. Finally, the time lag between oral presentation and full publication is of importance to both its authors and the audience at the meeting. The main objectives of this study were to determine the publication rate of papers of various fields of interest as presented at five consecutive annual meetings of the European Association of Plastic Surgeons (EURAPS) and the time lag between these presentations and their publication. The authors compared their overall findings to those reported for other surgical specialties. Moreover, they identified and classified the journals in which the full publications appeared as an indicator of the scientific value of the meeting. They conclude that a greater than average number of papers presented at the 1995 through 1999 annual EURAPS meetings went on to full publication in peer-reviewed journals. Among these journals, Plastic and Reconstructive Surgery was the best source for information presented at the meetings. Although approximately 90 percent of the publications appeared before 3 years had passed after a meeting, additional publications may be expected to appear even more than 6 years after the meeting. Given the high publication rate and the high average normalized impact factor of the journals in which the presentations appeared, the five studied EURAPS meetings overall had high scientific value.  相似文献

The procurement of new knowledge and understanding in the ever expanding discipline of cell biology continues to advance at a breakneck pace. The progress in discerning the physiology of cells and tissues in health and disease has been driven to a large extent by the continued development of new probes and imaging techniques. The recent introduction of semi-conductor quantum dots as stable, specific markers for both fluorescence light microscopy and electron microscopy, as well as a virtual treasure-trove of new fluorescent proteins, has in conjunction with newly introduced spectral imaging systems, opened vistas into the seemingly unlimited possibilities for experimental design. Although it oftentimes proves difficult to predict what the future will hold with respect to advances in disciplines such as cell biology and histochemistry, it is facile to look back on what has already occurred. In this spirit, this review will highlight some advancements made in these areas in the past 2 years.  相似文献
