The objective of this study was to determine the reproductive performance of lactating dairy cows treated with GnRH and/or PGF2a for synchronization of estrus and ovulation. Between Days 43 and 57 post partum, a total of 374 dairy cows was divided into 4 groups. Cows in Group 1 (n = 62) were treated with 25 mg, i.m. PGF2a on Days 43 and 57; cows in Group 2 (n = 65) were not treated at this time; cows in Group 3 (n = 118) were treated with 100 ug, i.m. GnRH on Day 50, 25 mg, i.m. PGF2a on Day 57, 100 ug, i.m. GnRH on Day 59, and time-inseminated 16 h later; cows in Group 4 (n = 129) were treated with 25 mg, i.m. PGF2a once on Day 57. Cows in Groups 1 and 4 were inseminated at an induced estrus within 7 d after the last PGF2a treatment, and cows in Group 2 were inseminated at a noninduced estrus within a corresponding period of time. Conception rate, estrus detection rate and pregnancy rate were analyzed using logistic regression, and controlled for lactation number, body condition score and time of year. Days from calving to conception were analyzed using the GLM procedures of SAS, and the model included group, body condition score, lactation number, time of year, and their interactions. Cows in Group 3 had a significantly higher pregnancy rate than cows in Groups 1, 2 and 4. Orthogonal contrasts of mean days from calving to conception showed that cows in Group 3 had significantly (P < 0.01) less days from calving to conception than cows in Group 1 and Group 4. There was a significant effect of time of year on pregnancy rate and days from calving to conception, but there was no interaction between time of year and these reproductive characteristics. There was no effect of body condition score and lactation number on the reproductive characteristics evaluated. From the results of this study, it was concluded that better reproductive performance was observed in cows inseminated at a synchronized ovulation than in those inseminated at a synchronized estrous period.  相似文献

The effects of milk yield, body condition score (BCS) and lactation number on the number of oocytes recovered and blastocysts formed were studied following in vitro maturation, fertilization and culture of bovine oocytes collected from 48 high and 46 medium genetic merit dairy cows in their first and third lactation. The cows were slaughtered between 125 and 229 d post partum. Ovaries were recovered, and 2- to 10-mm follicles were aspirated. Cleavage rate and number of blastocysts were determined at 44 h and 7 d after insemination, respectively. Oocytes from high genetic merit cows formed fewer blastocysts and had lower cleavage and blastocyst formation rates than those from medium genetic merit cows (0.36 +/- 0.19, 70.4 and 6.8% vs 0.85 +/- 0.22, 77.4 and 11.4%, respectively). The effect of milk production was tested by grouping cows in their third lactation into high and low groups. There was no difference in number of oocytes recovered and subsequent development into blastocysts between the cows in the high milk production group (4559 to 5114 kg, n = 20) and cows in the low yield (3162 to 3972 kg, n = 20) group (6.9 +/- 1.34 vs 8.9 +/- 1.32, respectively). The effect of BCS was tested by grouping cows in their first or third lactation into high and low groups. Cleavage and blastocyst formation rates were greater for oocytes from cows with a high BCS (3.3 to 4.0, n = 20) than a low BCS (1.5 to 2.5, n = 20) (75.7 vs 61.9% and 9.9 vs 3.0%, respectively). Cows in the first lactation yielded fewer oocytes (5.7 +/- 1.24) than cows in the third lactation (7.8 +/- 0.79). Thus, the quality of oocytes probably contributes to reduced fertility, often evident in high genetic merit dairy cows.  相似文献

This study investigated the profile of locomotion score and lameness before the first calving and throughout the first (n=237) and second (n=66) lactation of 303 Holstein cows raised on a commercial farm. Weekly heritability estimates of locomotion score and lameness, and their genetic and phenotypic correlations with milk yield, body condition score, BW and reproduction traits were derived. Daughter future locomotion score and lameness predictions from their sires’ breeding values for conformation traits were also calculated. First-lactation cows were monitored weekly from 6 weeks before calving to the end of lactation. Second-lactation cows were monitored weekly throughout lactation. Cows were locomotion scored on a scale from one (sound) to five (severely lame); a score greater than or equal to two defined presence of lameness. Cows’ weekly body condition score and BW was also recorded. These records were matched to corresponding milk yield records, where the latter were 7-day averages on the week of inspection. The total number of repeated records amounted to 12 221. Data were also matched to the farm’s reproduction database, from which five traits were derived. Statistical analyses were based on uni- and bivariate random regression models. The profile analysis showed that locomotion and lameness problems in first lactation were fewer before and immediately after calving, and increased as lactation progressed. The profile of the two traits remained relatively constant across the second lactation. Highest heritability estimates were observed in the weeks before first calving (0.66 for locomotion score and 0.54 for lameness). Statistically significant genetic correlations were found for first lactation weekly locomotion score and lameness with body condition score, ranging from −0.31 to −0.65 and from −0.44 to −0.76, respectively, suggesting that cows genetically pre-disposed for high body condition score have fewer locomotion and lameness issues. Negative (favourable) phenotypic correlations between first lactation weekly locomotion score/lameness and milk yield averaged −0.27 and −0.17, respectively, and were attributed to management factors. Also a phenotypic correlation between lameness and conception rate of −0.19 indicated that lame cows were associated with lower success at conceiving. First-lactation daughter locomotion score and/or lameness predictions from sires’ estimated breeding values for conformation traits revealed a significant linear effect of rear leg side view, rear leg rear view, overall conformation, body condition score and locomotion, and a quadratic effect of foot angle.  相似文献

Dairy cows (n = 199) were assigned randomly at Day 14 post partum to a control group or a programmed reproductive treatment group (PRT). Cows in the PRT group received 8 ug of GnRH on Days 14 and 50 post partum and 25 mg of PGF2alpha injected on Days 21, 34 and 57 post partum. Cows in the PRT group had a greater frequency of progesterone (P4) concentrations > 1 ng/ml (50% vs 30%; P < 0.01). Frequency of cows having P4 > 1 ng/ml at both Days 21 and 34 post partum was greater in the PRT group than in the control group (39% vs 20%; P < 0.01). Accumulation of days with a palpable cystic ovary was lower in the PRT than the control group (11.4% vs 6.5%; P < 0.05). The frequency of anestrous cows through Day 57 post partum was not different between the 2 groups. Cows that were anestrous had a lower body condition score throughout the postpartum period than the cyclic cows (2.7 vs <3.0; P < 0.01). Conception rate to first service, conception rate to all services, services per conception and days open were not different between the groups. Intensity of estrus detection was low for the study based on a low estrus detection rate (42%) and a low palpation pregnancy rate index (below 68%) throughout the study period. Accuracy of estrus detection was low based on the distribution of normal interestrus intervals for the study (19 to 23 d; 13.5%). It is concluded that programmed reproductive treatments during the postpartum period are functionally effective relative to altered ovarian activity; however, potential advantages to such a system are not evident without good estrus detection practices in a large commercial herd.  相似文献

Beef cows (n = 294) calving between November and April in six states were used to evaluate the effects of postpartum diet and calf separation on body weight, body condition score (BCS), reproductive performance, and weaning weight of calves. In each state, half of the 48 cows that calved during 60 d were group fed an additional 4.5 kg of a 20% crude protein supplement daily for 28 d starting an average of 30 d post partum (flush). Calves were separated from half of the flush and half of the nonflush cows for 48 h at 14 and 28 d after the beginning of flush. Progesterone was quantified in plasma samples obtained weekly during a 56-d breeding period to assess ovarian luteal activity. The breeding period started at the first calf separation. BCS ranged from 3.3 to 5.6 among states (on a scale of 1 to 9) at the start of the flush but was similar for treatments within a state. There was a state x flush (P < 0.008) effect on body weight at the end of the flush period. Weaning weights were influenced by state x separation x flush (P < 0.06) and were greatest for flush nonseparated calves in five of six states. There were state x flush (P < 0.08) and separation (P < 0.04) effects on ovarian luteal activity at the start of the breeding period. Flush and separation tended to increase ovarian luteal activity. During the breeding period, ovarian luteal activity was influenced only by state but there was a state x separation x flush effect (P < 0.001) on the number of weeks post partum to onset of ovarian luteal activity. Conception rate and days postpartum to conception were not influenced by either separation or flush but were affected by state (P < 0.001). These data indicate that flushing may increase weaning weights of calves and calf separation may hasten the onset of postpartum ovarian luteal activity, but conception rate and days postpartum to conception for cows in thin to moderate body condition were not influenced by the calf separation or flushing treatments.  相似文献

To induce cyclicity in dairy cattle with prolonged postpartum anestrous, repeated dosages of gonadotrophin releasing hormone (GnRH) were administered. Twenty-one (21) Holstein dairy cows and heifers calving between October 1, 1989, and January 1, 1990, at the Louisiana State University Dairy were used in the study. The animals were defined as anestrous if their plasma progesterone remained < 1.0 ng/ml until 32 to 36 days post partum. They were randomly assigned to one of two treatment groups. Group 1 (n=6) received two injections 1 hour apart of a GnRH analogue (50 mug) (i.m.). The treatment was repeated twice weekly at 3- to 4-day intervals. Group 2 controls (n=6) received saline (1 ml, i.m.) on the same schedule as Group 1. A maximum of 12 to 13 treatments were given. Cattle that had plasma progesterone >1.0 ng/ml by 32 to 36 days post partum were identified as Group 3, or cyclic contemporaries (n=9). Postpartum anestrous in the herd was 46.2% (18 39 ). Cows in Group 1 had significantly fewer days to first plasma progesterone > 1.0 ng/ml than those in Group 2 (P < 0.05), but more days than Group 3. Cows in Group 1 also had significantly fewer treatments to induce plasma progesterone > 1.0 ng/ml than those in Group 2 (P < 0.05). There were no significant differences among treatment groups in the number of days from calving to first observed estrus or the number of days open (P > 0.05).  相似文献

We concurrently evaluated early weaning and once-daily suckling as options for shortening postpartum interval to estrus in primiparous cows calving in good body condition. Over 2 consecutive years, a total of 77 primiparous cows were assigned to 1 of 3 treatments: control (ad libitum suckling); once-daily suckling (for 30 d prior to start of breeding); or early weaning (calves weaned 30 d prior to start of breeding). All cows had a body condition score (BCS) of > or = 5 at parturition and were maintained in good condition throughout the breeding season. Cows were observed for estrus twice daily and blood samples for subsequent measurement of progesterone were collected at 48-h intervals. For data analysis, cows were divided into early- (> or = 85 d post partum) and late-calving (< 85 d post partum) groups at the onset of the breeding season. All calves were weighed at birth, at the onset of treatment, at weekly intervals until the start of the breeding season, and within 45 d of 205 d of age. Early weaning or once-daily suckling shortened the postpartum interval to first estrus of late-calving primiparous cows by 17 (P < 0.01) and 12 d (P < 0.08), respectively, compared with control group cows. However, among early-calving cows, neither early weaning nor once-daily suckling influenced the postpartum interval. Postpartum intervals of once-daily suckled cows and early-weaned cows were similar (P > 0.10).  相似文献

Two experiments were conducted to examine the effects of repeated low-dose injections of gonadotropin releasing hormone (GnRH) 30 to 40 d post partum on reproductive characteristics in multiparous suckled Brahman cross cows. In Experiment I, 39 cows were injected (i.v.) with GnRH (5 mug/injection) at 2-h intervals for either 0 (control), 6, 12, or 24 h at 30 to 37 d post partum. GnRH injections for short periods (6h) increased the number of cows exibiting estrus within 45 d of treatment, but cows injected for 24 h failed to exhibit estrus during this period. The period from treatment to first estrus was shorter in the 6-h GnRH group compared to the control group. Injections for 6h significantly (P < 0.05) increased in serum luteinizing hormone (LH) concentrations 1 d after GnRH treatment. In Experiment II we examined the effect of i.v. GnRH injections (5 mug/injection at 2-h intervals) for 6h in a larger group of cows (n = 70). The days from treatment to first estrus were reduced (P < 0.05) in GnRH-treated cows; however, first-service conception rates were significantly lower (P < 0.01) in treated compared to control cows (46.4 and 80.0%, respectively). The results led us to believe that GnRH injections for short periods reduce postpartum interval to first estrus, but fertility at first estrus is lowered.  相似文献

The aim of this study was to investigate the effect of treating repeat breeder dairy crossbred cows with different doses of GnRH analogue through i.m. at the time of artificial insemination, on pregnancy rates from their first service after treatment and overall pregnancy rates. One hundred and thirty seven crossbred dairy cows with a history of repeat breeding and eligible after 6-8 infertile services but clinically free of diseases were selected for the study. The animals were randomly divided into three groups. Group 1 (n = 55) cows were treated intramuscularly with each 20 microg Buserelin-acetate (Receptal, Hoechst Roussel Vet GmbH) at the time of artificial insemination. Group 2 (n = 40) cows were treated intramuscularly with each 10 microg Buserelin-acetate at the time of artificial insemination. Group 3 (n = 42) cows were treated intramuscularly with saline as control at the time of artificial insemination. The first service pregnancy rates in Groups 1-3 were 45, 25 and 17%, respectively. Similarly, the overall conception rates in Groups 1-3 were 87, 58 and 48%, respectively. The results indicated that the pregnancy rate in crossbred cows could be improved by the GnRH treatment. The higher dose of GnRH significantly increased (P < 0.05) the first service as well as overall pregnancy rate in a dose dependent manner in repeat breeder crossbred cow bred previously 6-8 times unsuccessfully.  相似文献

Lactating Holstein dairy cows (n=1,533) were allocated to one of three treatment groups, with Group I (n=514) receiving 10 mug of a GnRH analogue (buserelin) at artificial insemination (AI) and Group II (n=503) receiving 10 mug of the same analogue at both the time of AI and at 12 days post AI. Herdmates in Group III (n=516) were inseminated on the same day and served as contemporary AI controls. The trial was conducted on five large dairy farms during the spring and summer months in Saudi Arabia. Pregnancy rates were determined by palpation per rectum between 33 and 50 days following AI. The first service pregnancy rate for the control cows (42.4%) was lower (P<0.05) than that for cows treated with the GnRH analogue at AI (48.8%) or for the combined treatment at AI and at Day 12 post AI (51.5%). No additive effect on the pregnancy rate was noted from the combined analog treatment. The overall increase in pregnancy rate from the analogue treatment at AI resulted from an 11% increase in pregnancy rate in first parity cows over that of contemporary controls (P<0.05) and a 14.7% increase in pregnancy for cows mated at 40 to 59 days post partum and treated with the analogue at AI over that of the corresponding controls (P<0.05). We determined whether insertion of a CIDR for 7 days prior to the breeding season enhanced pregnancy rates and altered the date of conception in suckled beef cows mated naturally. Suckled beef cows (n=2033) from 15 locations were randomly assigned to one of two treatments: (1) cows received a CIDR 7 days prior to the breeding season for 7 days (CIDR; n=999); (2) cows received no treatment (Control; n=1034). On the first day of the breeding season bulls were introduced to herds at a rate of 15-25 cows per yearling bull or 20-30 cows per mature bull. Pregnancy status and the date of conception were determined via transrectal ultrasonography at 56 and 120 days after initiation of the breeding season. Overall pregnancy rates ranged from 59.3 to 98.9% among the 15 locations. The percentage of cows becoming pregnant during the first 30 days of the breeding season was similar between CIDR (68.2%) and Control (66.7%) cows, and overall pregnancy rates were similar between CIDR (88.9%) and Control (88.6%) cows. The average day of conception after initiation of the breeding season was shorter (P<0.01) for CIDR (20.1+/-0.8 days) compared to Control cows (23.2+/-0.8 days). Of cows conceiving during the breeding season, more (P<0.05) CIDR cows (35.9%) conceived during the first 10 days of the breeding season than Control cows (30.8%). Neither body condition score and nor parity affected pregnancy rates or days to conception, whereas pregnancy rates and days to conception were affected (P<0.01) by location and days postpartum. Days to conception were greater for cows that calved within 40 days (31.6+/-1.2 days) of initiation of the breeding season compared to cows calving between 40 and 50 days (25.3+/-1.2 days) prior to initiation of the breeding season, which were greater than those cows calving between 50-60 days (20.0+/-0.8 days) and 60-70 days (21.3+/-1.0 days) prior to initiation of the breeding season. Cows calving greater than 70 days (17.3+/-1.5 days) from initiation of the breeding season had the shortest interval to conception. We concluded that insertion of a CIDR prior to the breeding season failed to increase overall pregnancy rates, but did influence the average day of conception.  相似文献

A controlled trial investigating the effect on conception of administration of 250 μg of gonadotropin-releasing hormone (GnRH) at artificial insemination (AI) in dairy cows in seasonal or split calving herds was conducted. Time of detection of estrus, body condition, extent of estrous expression, treatment, breed, age and milk production from the most recent herd test of the current lactation was recorded. Cows were tested for pregnancy with fetal aging between 35 and 135 days after AI. Sixteen herds provided 2344 spring-calved cows and 3007 inseminations. Logistic regression adjusting for clustering at herd level was used to examine the effect of treatment for first (2344) and second (579) inseminations separately. For first AI, treatment significantly improved conception rate in cows with milk protein concentrations of 3.75% or greater and for cows with milk protein concentrations between 3.00% and 3.50% and less than 40 days calved; increased conception rate from 41.2% to 53.4%. Treatment reduced conception rates in cows with milk protein concentrations of 2.75% or less. Treating only cows identified as responding positively to treatment (11% of all study cows) was estimated to increase first service conception rate in herds from 48.1% to 49.4%. There was no significant effect of treatment on conception to second AI, nor any significant interactions. These findings indicate that GnRH at AI should be limited to the sub-group cows most likely to respond. The positive effect of GnRH at AI may be mediated through improved oocyte maturation and/or improved luteal function, rather than by reducing AI-to-ovulation intervals.  相似文献

The benefit of using timed-insemination in lactating dairy cows for the treatment of ovarian cysts lies in the fact that cows do not have to be detected in estrus for insemination and achieving pregnancy. We compared the effectiveness of synchronization of ovulation with timed-insemination and induction of estrus with insemination at estrus in the treatment of bovine ovarian cysts in lactating dairy cows. After Day 65 post partum, a total of 368 lactating dairy cows was divided into 3 groups. Cows in Group 1 (n = 209, normal, noncystic) were treated with 100 ug, i.m. GnRH on Day 0; 25 mg, i.m. PGF2 alpha on Day 7; and 100 ug, i.m. GnRH on Day 9 and then were time-inseminated 16 h later. Cows in Group 2 (n = 76, abnormal, cystic) were treated with 100 ug, i.m. GnRH on Day 0; 25 mg, i.m. PGF2 alpha on Day 7; and 100 ug, i.m. GnRH on Day 9 and time-inseminated 16 h later. Cows in Group 3 (n = 83, abnormal, cystic) were treated with 100 ug, i.m. GnRH on Day 0; 25 mg, i.m. PGF2 alpha on Day 7; and inseminated at induced estrus within 7 d after treatment with PGF2 alpha. Day 0 was the day of initiation of the study. Conception and pregnancy rates among groups were compared using logistic regression and adjusted for parity, time of year and days in milk. Conception and pregnancy rates of Group 1 cows (31.5%) were not significantly different from those of Group 2 cows (23.6%). However, the pregnancy rate in normal cows (Group 1) was higher (P < 0.01) than in cystic cows (Groups 2 and 3). Cows in Group 3 had a higher conception rate than cows in Group 2 (51.7% > 23.6%; P < 0.01). However, pregnancy rates for cows in Groups 2 (23.6%) and 3 (18%) were not significantly different. The finding indicated that synchronization of ovulation and timed-insemination resulted in pregnancy rates similar to those of synchronization of estrus and insemination at an induced estrus within 7 d for the treatment of ovarian cysts in lactating dairy cows.  相似文献

Seasonal effects on luteal activity during post partum were evaluated in two consecutive studies in 253 dairy cows in Northern Italy. In study 1, plasma progesterone concentrations were determined on days 14, 21, 28, 35, 42, 49 and 56 post partum and in study 2 cows were synchronized and inseminated at a fixed time using two regimes based on the 'Ovsynch' protocol. Study 1: Animals were classified as luteal (progesterone >1.5 ng/ml in at least two consecutive samples) or non-luteal (progesterone <1.5 ng/ml in all samples). The proportion of cows without luteal activity from calving to day 56 post partum was 47/253 (18.5%). Of the 47 cows without luteal activity, 42 (89%) were detected during the warm months of the year and five were detected during the cold months of the year, and the effect of season was highly significant (P < 0.001). Study 2: Three study groups were established; control (CONT, untreated cows, n = 92), GPG (cows receiving gonadotropin-releasing hormone (GnRH) on day 0, PGF2α on day 7 followed by a second dose of GnRH 24 h later, n = 80); and HPH (the same as the GPG group, but with human chorionic gonadotropin (hCG) substituted for GnRH, n = 81). In the GPG and HPH groups, cows were inseminated 16 to 22 h after the second GnRH or hCG injection. Untreated cows were inseminated at first estrus after a voluntary weaning period. Because the effects of the GPG and HPH regimes on pregnancy rate were not significantly different, data were pooled into a single treatment group (TREAT). Pregnancy rates during the warm months of the year were 16% and 15% at first service and 65% and 66% at day 135 post partum for CONT and TREAT groups, respectively. Pregnancy rates during the cold months of the year were 36% and 38% at first service and 72% and 76% at day 135 post partum for CONT and TREAT groups, respectively. There was an effect of season (P < 0.05) but not of treatment on pregnancy rate. Treatment reduced the number of days from calving to conception during both the cold (101 ± 3.2 v. 121 ± 3.1 days; P < 0.001) and warm seasons (122 ± 3.2 v. 145 ± 3.1 days; P < 0.001). In conclusion, the present study shows that (i) heat stress during the warm season can compromise luteal activity and (ii) that regimes based on the Ovsynch protocol did not improve pregnancy rate at first service or by 135 post partum, but they had a positive effect on the calving-to-conception interval.  相似文献

A study was conducted to determine whether treatment with gonadotropin releasing hormone (GnRH) in combination with prostaglandin F(2)alpha (PGF(2)alpha) could enhance ovarian activity and uterine involution in postpartum dairy cows to reduce the calving interval. Cows were randomly assigned to one of three treatment groups. Cows (n = 8) in Group 1 received 100 mug GnRH intramuscularly (i.m.) twice on Day 20 and Day 35 postpartum, and 25 mg PGF(2)alpha i.m. on Day 47 postpartum. Group 2 (n = 8) received a single i.m. injection of 100 mug GnRH on Day 25 postpartum and 25 mg PGF(2)alpha i.m. on Day 37 postpartum. The Control Group (n = 9) did not receive hormonal treatment. Palpation per rectum of the reproductive organs and serum progesterone (P) determination were performed twice a week to monitor ovarian activity and uterine involution. Postpartum interval to the first ovulation was short in treated groups (Group 1, 21.0 d; Group 2, 26.3 d) compared with Control Group (30.1 d, P < 0.05). Likewise, mean frequency of ovulation was increased in both treated groups compared with the Control Group (P < 0.05). Cows in treated groups required fewer days to complete uterine involution than in the Control Group. The mean interval to the first service, the conception rate at first service and the number of services per conception showed no significant differences among the three groups, but the mean days from calving to conception were shorter for the treated groups (78.7 d in Group 1; 83.3 d in Group 2) than (109.1 d, P < 0.05) for the Control Group. Our results suggest that combined treatment with GnRH and PGF(2)alpha may enhance ovarian activity in the postpartum cow, resulting in improved reproductive performance.  相似文献

We investigated the effect of body condition loss from the dry to near calving periods on the subsequent body condition change, the occurrence of postpartum diseases, the serum metabolic parameters total cholesterol, triglyceride, glucose, and urea nitrogen and the number of days to first breeding after calving in Holstein dairy cows. Body condition scoring (using a 5-point scale with quarter-point divisions) was performed on 67 pregnant Holstein dairy cows. Cows were scored once for body condition during the dry period (prepartum day 35 +/- 16), near calving (postpartum day 5 +/- 4), and subsequently at months 1 (+/- day 4), 2 (+/- day 5), 3 (+/- day 5), and 4 (+/- day 4) of lactation. At the same time, blood samples were collected to evaluate serum total cholesterol, triglyceride, glucose and urea nitrogen concentrations. Cows were categorized based on body condition loss from the dry to near calving periods into two groups: the moderate condition loss group (0-0.75 points, n=41), or the marked condition loss group (1.0-1.5 points, n=26). Regular reproductive health examination was conducted by the same investigator once a week. The marked condition loss group lost more body condition (P<0.01) than the moderate condition loss group consistently until month 1 of lactation. The recovery of body condition in the marked condition loss group was more delayed (P<0.01) than in the moderate condition loss group from months 1 to 4 of lactation. The occurrence of metritis and metabolic diseases (including abomasal displacement, milk fever, ketosis) was greater (P<0.01) in the marked condition loss group (62 and 23%) than in the moderate condition loss group (27 and 2%), respectively. The total cholesterol concentration was lower (P<0.05) in the marked condition loss group (167 +/- 5.3 mg/dl) than in the moderate condition loss group (183 +/- 5.8 mg/dl) at month 1 of lactation, however, the cholesterol levels at the other periods were not different (P>0.05) between the two groups. The triglyceride, glucose, and urea nitrogen concentrations were not different (P>0.05) from the dry period to month 4 of lactation between the two groups. The number of days to first breeding after calving was longer (P<0.05) in the marked condition loss group (103 +/- 7.8 day) than in the moderate condition loss group (87 +/- 5.3 day). We conclude that marked body condition loss from the dry to near calving periods results in the increased occurrence of postpartum metabolic and reproductive diseases, decreased serum total cholesterol concentrations at month 1 of lactation and a longer interval to first breeding after calving in Holstein dairy cows, probably due to the more severe energy deficit reflected by unfavorable body condition score change during early lactation.  相似文献

This study was designed to determine conception rates in dairy cows after timed-insemination and simultaneous treatment with gonadotrophin releasing hormone (GnRH) and/or prostaglandin F2 alpha (PGF2alpha). A total of 2352 cows was randomly assigned to six groups. Cows in Groups 1 to 5 were palpated per rectum to determine the presence of a corpus luteum (CL) on the ovary, and blood samples were obtained for the determination of plasma progesterone (P4) concentrations. Cows with a CL and P4 concentrations >1 ng/ml were treated (Day 0) with PGF2alpha (25 mg, i.m.) and were observed for estrus. Cows in estrus prior to 72 hours after treatment (Group 5, n = 106) were bred, but were not treated. Cows not observed in estrus by 72 hours were divided into four remaining groups, were bred between 72 and 80 hours and were assigned as follows: Cows in Group 1 (n = 203) were not treated; Cows in Group 2 (n = 200) were treated with GnRH (100 ug, i.m.); Cows in Group 3 (n = 201) were treated with PGF2alpha (25 mg, i.m.); and cows in Group 4 (n = 202) were treated with both GnRH and PGF2alpha. Cows in Group 6 (n = 1440) were not treated with PGF2alpha on Day 0 and were estrual cows that were bred on days when cows in Groups 1 to 5 were time-inseminated. The percentage of cows in all groups pregnant at 45 to 50 days after one insemination was compared using analysis of variance (P<0.05). The conception rate of cows in Group 2 was significantly higher than that of cows in Groups 1 to 4. There was a significant group-by-season interaction. Cows treated with GnRH during the spring had a higher conception rate than at other times of the year. Conception rates of cows in Groups 1 to 4 that were inseminated during the summer were low and not significantly different from each other. Conception rates of cows in Groups 5 and 6 inseminated during the summer were not significantly different from each other, but were significantly higher than that of cows in Groups 1 to 4 that were inseminated during the summer.  相似文献

The objective of this study was to determine the value of using plasma concentrations of PGFM to diagnose subclinical endometritis in the dairy cow, and its relationship to subsequent fertility. A total of 274 cows between 24 to 29 d post partum was divided into 4 groups on the basis of clinical features of the uterus and ovary. Cows in Group 1 (n = 74) had a normal, involuting uterus and a CL on the ovary; cows in Group 2 (n = 51) had a normal, involuting uterus but no CL on the ovary; cows in Group 3 (n = 83) did not have a normal, involuting uterus but had a CL on the ovary; and cows in Group 4 (n = 66) did not have a normal, involuting uterus or a CL on the ovary. A blood sample was obtained from each cow on the day they were placed on the study, and plasma concentrations of PGFM and P4 were determined using RIA. Cows were artificially inseminated (AI) at the first observed estrus after Day 60 post partum, and pregnancy was determined by palpation of the uterus per rectum between 45 and 50 d postAI. Reproductive responses evaluated were conception rate to first service, days open, and percentage of cows pregnant by 90, 120, 150 and 180 d post partum. Data were analyzed using GLM procedures of SAS and a 2 x 2 factorial with contrast procedures. Polynomial regression analysis was used to determine the shape of the PGFM, P4 and fertility curves. There was no difference among mean PGFM concentrations of cows in each group. The rate of decline of plasma PGFM concentrations was lower in cows with an abnormal uterus and a CL on the ovary compared with those without a CL. A lower percentage of cows with abnormal uteri was pregnant by 90 d post partum compared with cows with normal uteri. From the results of this study, it was concluded that plasma PGFM concentrations between Days 24 to 29 post partum were not effective in identifying cows with subclinical endometritis.  相似文献

The milk production, energy balance (EB), endocrine and metabolite profiles of 10 New Zealand Holstein Friesian (NZ) cows and 10 North American Holstein Friesian (NA) cows were compared. The NA cows had greater peak milk yields and total lactation milk yields (7387 v. 6208 kg; s.e.d. = 359), lower milk fat and similar protein concentrations compared with the NZ cows. Body weight (BW) was greater for NA cows compared with NZ cows throughout lactation (596 v. 544 kg; s.e.d. = 15.5), while body condition score (BCS) tended to be lower. The NA strain tended to have greater dry matter intake (DMI) (17.2 v. 15.7 kg/day; s.e.d. = 0.78) for week 1 to 20 of lactation, though DMI as a proportion of metabolic BW was similar for both strains. No differences were observed between the strains in the timing and magnitude of the EB nadir, interval to neutral EB, or mean daily EB for week 1 to 20 of lactation. Plasma concentrations of glucose and insulin were greater for NA cows during the transition period (day 14 pre partum to day 28 post partum). Plasma IGF-I concentrations were similar for the strains at this time, but NZ cows had greater plasma IGF-I concentration from day 29 to day 100 of lactation, despite similar calculated EB. In conclusion, the results of this study do not support the premise that the NZ strain has a more favourable metabolic status during the transition period. The results, however, indicate that NZ cows begin to partition nutrients towards body reserves during mid-lactation, whereas NA cows continue to partition nutrients to milk production.  相似文献

Three hundred and one Holstein cows (n=301), calving at a commercial free-stall dairy farm, were randomly assigned to 1 of 3 prostaglandin treatment groups or a placebo group. The placebos were packaged 3 ways to mimic the 3 commercial prostaglandin preparations. Group 1 received 1 mg fenprostalene and 1.6 mg oxytetracycline; Group 2 received the fenprostalene placebo (2 ml polyethylene glycol and 1.6 mg oxytetracycline); while Group 3 was given 25 mg dinoprost. Group 4, the dinoprost placebo received 5 ml saline; Group 5 received 500 ug cloprostenol; and Group 6 the cloprostenol placebo received 2 ml saline. The treatments were administered between Days 24 and 31 post partum. Double blind techniques were used in administering treatments and in assessing the response to treatment. There were no significant differences among treatment groups with respect to incidence of retained fetal membranes, endometritis, pyometra, anestrus, number of services per pregnancy, calving-to-first estrus interval, services per conception, number of prostaglandin treatments other than those administered between Days 24 and 31 post partum, the percentage culled for reproductive reasons and all factors combined. Cows receiving fenprostalene, dinoprost or cloprostenol had a decreased calving-to-conception interval compared with that of the controls (P = 0.05). It is concluded that, in the herd studied, treatment with any of the 3 commercially available prostaglandin products between Days 24 and 31 post partum was beneficial for reproductive performance.  相似文献
