Hedgerows play important roles in agricultural landscapes and they increase biodiversity by providing habitat refugia for species sensitive to agricultural disturbance. We have studied the characteristics of the communities of harvestmen (Opiliones) inhabiting hedgerows. Harvestmen are terrestrial arachnids associated with soil surface and subsurface and sensitive to land-use and disturbance. We were specifically interested in quantifying how hedgerow characteristics (e.g., elevation, length, width, connectivity, and plant diversity) affect harvestman diversity and community composition. We expected harvestman diversity and community composition to be positively related to both hedgerow size (area, length, width) and biological attributes of hedgerows (e.g., connectivity, plant community diversity). We surveyed hedgerow characteristics and harvestman communities of 20 hedgerows in an upland agricultural landscape in the Western Carpathians. Hedgerow characteristics were measured in the field or derived from GIS layers and we used correlation and ordination methods to relate them to harvestman community metrics. We found surprisingly high taxonomic richness of harvestmen within the studied hedgerows (15 species). Importantly, the Shannon index of harvestman communities was positively related to hedgerow length and hedgerow tree layer species richness and diversity. Harvestman community composition varied with hedgerow area and width. Despite their small total area, hedgerows represented an important habitat for diverse harvestman communities and hedgerow attributes such as size and tree diversity significantly affected the composition and the Shannon index of harvestman communities. Thus, greater lengths and widths of hedgerows can provide habitats for a higher Shannon index of harvestman communities, within surrounding agricultural landscapes.  相似文献

Mistletoes offer a unique model to study interactions among Al and nutrients in vascular plants, because they grow and reproduce on hosts with distinct Al uptake strategies. We investigated Al distribution and nutrient relations of mistletoes on Al‐accumulating and non‐accumulating hosts. We hypothesised that mistletoes would exhibit similar leaf nutrient and Al concentrations as their host plants, but a strong compartmentalisation of Al when growing on Al‐accumulators. We measured concentrations of N, P, K, Ca, Mg, Cu, Fe, Mn, Zn in leaves and Al in leaves, seeds and branches of Phthirusa ovata and Psittacanthus robustus infecting Miconia albicans, an Al‐accumulator, and Ph. ovata infecting Byrsonima verbascifolia, a non‐Al‐accumulator. High leaf concentrations of Al in Ph. ovata only occurred while parasitizing the Al‐accumulating host; there was no accumulation in branches or seeds. In P. robustus, large concentrations of Al were found in leaves, branches and seeds. Mistletoe seed viability and leaf nutrient concentrations were not affected by Al accumulation. Passive uptake of Al, Ca, Mg, Mn and Cu in mistletoes was evidenced by significant correlations between mistletoes and host leaf concentrations, but not of N, P and K. Al was retranslocated to different plant organs in P. robustus, whereas it was mostly restricted to leaves in Ph. ovata. We suggest that Al might have some specific function in P. robustus, which only parasitizes Al‐accumulator hosts, while the host generalist Ph. ovata can be considered a facultative Al‐accumulator.  相似文献

Plants can defend themselves against herbivorous insects before the larvae hatch from eggs and start feeding. One of these preventive defence strategies is to produce plant volatiles, in response to egg deposition, which attract egg parasitoids that subsequently kill the herbivore eggs. Here, we studied whether egg deposition by Pieris brassicae L. (Lepidoptera: Pieridae) induces Brussels sprouts plants to produce cues that attract or arrest Trichogramma brassicae Bezdeko (Hymenoptera: Trichogrammatidae). Olfactometer bioassays revealed that odours from plants with eggs did not attract or arrest parasitoids. However, contact bioassays showed that T. brassicae females were arrested on egg‐free leaf squares excised from leaves with 72 h‐old egg masses, which are highly suitable for parasitisation. We tested the hypothesis that this arresting activity is due to scales and chemicals deposited by the butterflies during oviposition and which are thus present on the leaf surface in the vicinity of the eggs. Indeed, leaf squares excised from egg‐free leaves, but contaminated with butterfly deposits, arrested the wasps when the squares were tested 1 day after contamination. However, squares from egg‐free leaves with 72 h‐old butterfly deposits had no arresting activity. Thus, we exclude that the arresting activity of the leaf area near 72 h‐old egg masses was elicited by cues from scales and other butterfly deposits. We suggest that egg deposition of P. brassicae induces a change in the leaf surface chemicals in leaves with egg masses. A systemic induction extending to an egg‐free leaf neighbouring an egg‐carrying leaf could not be detected. Our data suggest that a local, oviposition‐induced change of leaf surface chemicals arrests T. brassicae in the vicinity of host eggs.  相似文献

1. Understanding of animal movement behaviour is critical for developing appropriate conservation strategies because of its profound implications for predicting species' responses to perturbations. To date there is a substantial knowledge gap of the movement behaviour of subterranean invertebrates. 2. In this study temporary emigration (TE), the probability that an individual is absent from the cave on a given sampling occasion, was used as a method to inform on the movement and behaviour of cave‐dwelling invertebrates. Because these animals are difficult to study with traditional tracking techniques, the capture–recapture (CR) modelling framework was used to assess TE and to account for imperfect detectability and unobservable states. 3. Specifically, the influence of season, sex, and surface weather variables on the TE of the facultative cave‐dwelling harvestman species, Paranemastoma sillii sillii (Herman, 1871), was investigated. Multistate CR models were used while accounting for individual heterogeneity on 999 individually marked adults of two populations inhabiting two caves, located in southwestern Romania. 4. Harvestmen demonstrated clear heterogeneity and seasonality in TE. Findings showed that the TE of harvestmen exhibited variation between caves and there was little support for surface weather variables influencing TE and for sex‐specific TE patterns. 5. These results show that the study of TE patterns is useful to gain valuable insights into movement and intrinsic behavioural processes of cave‐dwelling harvestmen. The method could potentially be used for other invertebrate groups with similar movement characteristics and when traditional tracking techniques are difficult.  相似文献

Upon inoculation with pathogenic microbes, plants induce an array of metabolic changes that potentially contribute to induced resistance or even enhance susceptibility. When analysing leaf lipid composition during the Arabidopsis thaliana–Pseudomonas syringae interaction, we found that accumulation of the phytosterol stigmasterol is a significant plant metabolic process that occurs upon bacterial leaf infection. Stigmasterol is synthesized from β‐sitosterol by the cytochrome P450 CYP710A1 via C22 desaturation. Arabidopsis cyp710A1 mutant lines impaired in pathogen‐inducible expression of the C22 desaturase and concomitant stigmasterol accumulation are more resistant to both avirulent and virulent P. syringae strains than wild‐type plants, and exogenous application of stigmasterol attenuates this resistance phenotype. These data indicate that induced sterol desaturation in wild‐type plants favours pathogen multiplication and plant susceptibility. Stigmasterol formation is triggered through perception of pathogen‐associated molecular patterns such as flagellin and lipopolysaccharides, and through production of reactive oxygen species, but does not depend on the salicylic acid, jasmonic acid or ethylene defence pathways. Isolated microsomal and plasma membrane preparations exhibited a similar increase in the stigmasterol/β‐sitosterol ratio as whole‐leaf extracts after leaf inoculation with P. syringae, indicating that the stigmasterol produced is incorporated into plant membranes. The increased contents of stigmasterol in leaves after pathogen attack do not influence salicylic acid‐mediated defence signalling but attenuate pathogen‐induced expression of the defence regulator flavin‐dependent monooxygenase 1. P. syringae thus promotes plant disease susceptibility through stimulation of sterol C22 desaturation in leaves, which increases the stigmasterol to β‐sitosterol ratio in plant membranes.  相似文献

Predation by naturally occurring predatory arthropods was investigated to explain variations in population numbers of twospotted spider mite (Tetranychus urticae Koch) between first and second season strawberry crops. Araneomorph spiders. European harvestman [Phalangium opilio (L.)], Tasmanian lacewing [Micromus tasmaniae (Walker)] and Pacific damsel bug [Nabis kinbergii Reuter] were the only predators found in high numbers. However, spiders and harvestment were more prevalent than lacewings and nabids. Laboratory feedings trials indicated spiders build horizontal webs in the plants and prey predominantly on small flying insects that shelter in the crops. Similar feeding trials cofirmed the palatability of TSSM to spiders and harvestmen. Immunological testing for proteins of TSSM, aphids, Collembola and leafrollers in the intestines of field collected European harvestman, spiders, Tasmanian lacewing and Pacific damsel bug confirmed European harvestman includes TSSM in its diet, but not in large enough quantities to exert a significant regulating pressure on TSSM populations. Lacewings and nabids include TSSM in their diets but only in very small quantities (2% and 1% respectively). Spiders do not take TSSM unless they drop or spin down onto the spider webbing. The immunological testing also showed European harvestman to be a polyphagous and opportunistic feeder. Prey residues were detected more frequently in harvestmen intestines at times of prey abundance which indicated a seasonality to harvestmen diet.   相似文献

External characteristics of the leaf epidermis and their effects on behaviour of Aphis gossypii Glover were evaluated in two Cucumis melo L. genotypes, ‘Bola de Oro’ (aphid susceptible) and TGR‐1551 (aphid resistant) in order to explore their role in the early rejection of TGR‐1551 by this aphid. No differential effects of epicuticular waxes on aphid behaviour were observed. The type, distribution and number of trichomes on melon leaves were also studied. Pubescence in melon, measured as the number of non‐glandular trichomes per cm2, was not sufficient to prevent aphid settling. However, there was a high density of type I glandular trichomes on leaves of the aphid‐resistant genotype. According to microscopic observations and stain testing, these trichomes store and secrete phenols and flavonoids. Free‐choice tests were conducted to determine the effect of these glandular trichomes on A. gossypii preference, revealing that aphids reject leaf disks of TGR‐1551 from the onset of the experiment. Additional experiments after removal of leaf type I glandular trichome exudates showed that A. gossypii preferred washed TGR‐1551 leaf disks over unwashed disks, while this effect was not observed in experiments using washed and unwashed ‘Bola de Oro’ leaf disks. These results suggest that a high density of glandular trichomes and chemicals secreted by them deter A. gossypii and disturb aphid settling on TGR‐1551.  相似文献

Increasing evidence suggests that individuals of the same plant species occurring in different microhabitats often show a degree of phenotypic and phytochemical variation. Consequently, insect herbivores associated with such plant species must deal with environment‐mediated changes or variability in the traits of their host plants. In this study, we examined the effects of habitat condition (shaded vs. full‐sun habitats) on plant traits and leaf characteristics of the invasive alien plant, Chromolaena odorata (L.) King & Robinson (Asteraceae). In addition, the performance was evaluated in two generations of a specialist folivore, Pareuchaetes insulata (Walker) (Lepidoptera: Erebidae: Arctiinae), on leaves obtained from both shaded and full‐sun habitats. The study was done in an area where the insect was introduced as a biological control agent. Leaves growing in shade were less tough, had higher water and nitrogen content, and lower total non‐structural carbohydrate, compared with leaves growing in full sun. Plants growing in shade had longer leaves and were taller, but above‐ground biomass was significantly reduced compared with plants growing in full sun. In both generations (parents and offspring), P. insulata developed faster and had larger pupal mass, increased growth rate, and higher fecundity when reared on shaded foliage compared with full‐sun foliage. Although immature survival and adult longevity did not differ between habitats, Maw's host suitability index indicated that shaded leaves were more suitable for the growth and reproduction of P. insulata. We suggest that the benefits obtained by P. insulata feeding on shaded foliage are associated with reduced toughness and enhanced nitrogen and water content of leaves. These results demonstrate that light‐mediated changes in plant traits and leaf characteristics can affect insect folivore performance.  相似文献

Many species have been reported to form roosting (resting, sleeping) aggregations at ‘traditional’ sites, but the alternative hypothesis that specific sites are used repeatedly because of habitat limitation is rarely tested. We studied the roosting behavior of a species of harvestman (Opiliones, Prionostemma sp.) at a lowland rainforest site in Nicaragua. Both sexes roosted by day in spiny palm trees, dispersed at dusk to forage, and rejoined aggregations just before dawn. The distribution of harvestmen among spiny palms was significantly clumped, and harvestman density did not correlate with spiny palm density. Aggregations formed repeatedly in a small subset of the available spiny palms and the same sites were used in two different years (2001, 2003). Nevertheless, the membership of aggregations was fluid; individual harvestmen were found at multiple roosts and moved up to 0.2 km per night. Translocated animals often returned to the roost where they had been released or nearby roosts but were never found at previously unused sites. The high consistency of site use but low site fidelity of individuals suggests that roost sites differed conspicuously (to the harvestmen) from sites that were not used. We found no univariate or multivariate differences between used and unused sites, however, in the characteristics of the trees or microclimate. These results conflict with the habitat limitation hypothesis but are consistent with the traditional site use hypothesis. The tradition may be mediated by a site‐labeling chemical, a mechanism that does not require individual site fidelity. We discuss these results in relation to the proposed functions of roosting aggregations.  相似文献

Aims: To screen micro‐organisms for inducing the production of dragon’s blood, which is normally produced by stem xylem and by leaf of Dracaena cochinchinensis, and to evaluate the product by comparing with the standard. Methods and Results: Thirty microbial strains were isolated from D. cochinchinensis leaves. Three of them were confirmed to elicit the leaf of D. cochinchinensis producing dragon’s blood after inoculation. Upon elicitation, all of the 6‐month‐old leaves of the inducible trees produced dragon’s blood; 60–70% of the 1‐year‐old leaves elicited produced the resin. All the three strains were identified as Colletotrichum gloeosporioide by morphological and molecular methods. The leaf resin had a similar TLC profile and antioxidant activities to the standard resin. In particular, it had a higher total flavonol content and antimicrobial activity than the standard. Conclusions: Upon the induction of the screened C. gloeosporioide mycelia, D. cochinchinensis leaf produced dragon’s blood with higher total flavone content and antimicrobial activity than the standard dragon’s blood. Significance and Impact of the Study: This work has provided a strategy for producing dragon’s blood in a sustainable way using leaves of C. gloeosporioides by fungal elicitation.  相似文献

Over a period of 19 yr, the harvestman (Opiliones) community associated with the lowbush blueberry agro-ecosystem in Maine was studied. Eight species representing five genera, four subfamilies, and two families of harvestmen belonging to the suborder Eupnoi were collected. The harvestman community was dominated by two introduced, synanthropic species: Phalangium opilio in all but 1 yr (that year dominated by Rilaena triangularis). Rilaena was recorded for the first time from eastern North America. Relative abundance of harvestman adults increases throughout the season and the temporal pattern of trap capture does not refute speculated life cycles of the harvestmen being univoltine with overwintering eggs. Some blueberry management practices were found to affect trap capture. We did find that on average (with opposite results 1 yr) trap captures are greater in pruned fields than in fruit-bearing fields. Organic fields were found to have higher relative abundance of harvestmen than conventionally managed fields. Conventionally managed fields with reduced-risk insecticides showed no difference in harvestmen relative abundance compared with those conventionally managed fields using the older more persistent organophosphate insecticides. Insecticide trials with common insecticides used in blueberry insect pest management showed that the organophosphate insecticide, phosmet, and the pyrethroid insecticide, esfenvalerate, were detrimental to P. opilio adults when exposed to leaf residues, whereas the reduced-risk insecticide, spinosad, showed no negative effects compared with nonsprayed foliage.  相似文献

Of 70 micro‐organisms (fungi, bacteria and actinomycetes) isolated from soil using vegetable tissue baits, 16 produced substances in culture fluids capable of preventing the development of blast caused by Magnaporthe oryzae on rice leaves with little or no inhibitory effect on the conidial germination of the pathogen. Isolate KS‐F14, which secreted substances capable of activating resistance in untreated leaves, was selected and identified as Fusarium solani. The resistance‐inducing substances were effective at pH values ranging from 5 to 10 and were stable under high temperatures, maintaining approximately the same level of activity even after autoclaving for 20 min. After application, the activated resistance in rice leaves persisted for 14 days. The polar solvent extracts of freeze‐dried KS‐F14 secretions were effective in activating resistance against M. oryzae in rice plants. The non‐polar solvent extracts were also effective, albeit not as effective as the polar solvent extracts, indicating that although the majority of the secreted resistance‐inducing compounds are hydrophilic, some of the compounds are hydrophobic. Treating secretions with cation or anion exchange resins only partially reduced their resistance‐inducing ability, suggesting that the resistance‐inducing components include both charged and non‐charged compounds. The resistance‐inducing compounds produced by F. solani have the potential to be developed into a commercial product for the control of rice blast and possibly other plant diseases.  相似文献

Harvestmen have a general distribution pattern with more species and higher abundance in forests than in open habitats, as previously verified in mountain Cantabrian areas of northern Spain, of the Orocantabrian Province. The study of harvestmen of the low Cantabrian areas of the adjacent biogeographic Cantabro-Atlantic Province is highly appropriate to determine the influence of biogeographic position on the main diversity parameters and the effect of different land uses on harvestman assemblages. The main types of managed habitats (forest plantations and secondary grasslands), together with natural forests, non-planted young forests, shrublands and habitat boundaries were continuously sampled with seven pitfall traps during 1 year at 28 sites. The harvestman assemblages were classified using six different analyses, and indicator species were identified. The spatial patterns of harvestman diversity in low areas differed from those of mountain areas, although they have 15 species in common. Remarkably, higher average harvestman species richness was measured in low Cantabrian areas than in mountain areas. Shrublands and boundaries were the most species-rich habitats. Forested areas were the poorest in abundance, and were not the habitats richest in harvestman species, though they had higher average richness than mountain Cantabrian forests. Grasslands had a unique harvestman composition with significant extraordinary abundances, in particular of Homalenotus quadridentatus (indicator species of this habitat) and H. laranderas. Interestingly, H. laranderas, Paroligolophus agrestis and Ischyropsalis hispanica, indicator species of some open habitats in low Cantabrian areas, have recently been shown to be indicators of shady forests in mountain Cantabrian territories.  相似文献

Harvestmen (Arachnida, Opiliones) are especially dependent on chemical cues and are often regarded as animals that rely mainly on contact chemoreception. Information on harvestman sensilla is scarce when compared to other arachnid orders, especially concerning internal morphology. Using scanning (SEM) and transmission (TEM) electron microscopy, we investigated tarsal sensilla on the distal tarsomeres (DT) of all leg pairs in Heteromitobates discolor (Laniatores, Gonyleptidae). Furthermore, we explored the typological diversity of sensilla present on the DT I and II in members of the suborder Laniatores, which include two thirds of the formally described opilionid fauna, using species from 17 families representing all main laniatorian lineages. Our data revealed that DT I and II of H. discolor are equipped with wall-pored falciform hairs (two types), wall-pored sensilla chaetica (two types) and tip-pored sensilla chaetica, while DT III and IV are mainly covered with trichomes (non-sensory) and tip-pored sensilla chaetica. The ultrastructural characteristics support an olfactory function for all wall-pored sensilla and a dual gustatory/mechanoreceptive function for tip-pored sensilla chaetica. Based on our comparative SEM survey, we show that wall-pored sensilla occur in all investigated Laniatores, demonstrating their widespread occurrence in the suborder and highlighting the importance of both legs I and II as the sensory appendages of laniatorean harvestmen. Our results provide the first morphological evidence for olfactory receptors in Laniatores and suggest that olfaction is more important for harvestmen than previously thought.  相似文献

It has been suggested that plant physical and chemical traits vary considerably in space and time. Hence, leaf‐mining insects may adjust their oviposition in response to leaf attributes representing high quality. Moreover, herbivorous insects can modify leaf morphology by acting as stressors, increasing, for example, fluctuating asymmetry (FA) levels. Here, we investigate oviposition preference in Agnippe sp.2, a leaf‐mining moth of Erythroxylum tortuosum, in relation to differences in leaf nutritional quality (i.e. levels of water, nitrogen and tannin content), leaf area (i.e. quantity of resource hypothesis) and FA. We also verify whether temporal variation in plant nutritional quality emerges as an alternative hypothesis to explain oviposition distribution in time, and whether this leaf miner is a stress‐causing agent, increasing FA during larval development. Mined leaves and leaves with and without eggs were periodically collected from plants located in a Cerrado fragment in Brazil. In the laboratory, leaf traits were assessed (using image analysis software) and quantified (biochemical analysis) according to the aims previously determined. Oviposition probability did not change in relation to variations in nitrogen, tannins and FA of leaves. However, leaf‐miner females preferred to oviposit on leaves having large areas and low water contents. It was also verified that new leaves of E. tortuosum, which carried most leaf‐miner eggs, presented significantly lower tannins and greater levels of nitrogen and water than old leaves. The oviposition choice exhibited by leaf miners was found to be non‐random because they appear to use resource quantity and water content as cues as where to lay their eggs. The temporal variation of plant nutritional quality is likely to influence the time of leaf‐miner oviposition; and leaf FA was not increased during larval feeding, suggesting that these herbivores do not cause variations in FA levels.  相似文献

Mitopus morio is a widespread harvestman species occurring in most of Europe and in moderate and cold‐moderate zones of Asia and North America. The species is characterized by extreme variability in body size and leg length. As leg length is correlated with habitat temperature, M. morio has been considered as an example of Allen's rule. Recently, observations for a single location in Tyrol, Austria, indicated the absence of mating between short‐ and long‐legged individuals. This study examines for signs of putative cryptic species in M. morio using an integrative approach that combines mating trials, amplified fragment length polymorphism whole‐genome scans, mitochondrial sequences and morphometrics. The mating trials did not corroborate the initial hypothesis of a reproductive barrier associated with leg size. Both types of genetic data revealed the existence of three distinct groups, in line with the mating results but largely unrelated to leg morphology and geographical origin of specimens. Morphometric characters supporting the findings of the other disciplines were identified using a supervised approach. We infer from all data together the existence of strongly diverged cryptic lineages among the analysed individuals, cautiously interpret them as three sympatric species and conclude that in these harvestmen Allen's rule applies at different levels. Due to the unexpected amount of differentiation found within a geographical scale very small compared with the distribution of M. morio, we suggest a thorough revision of the genus prior to formal taxonomic changes. Our case study underlines the general applicability of the integrative taxonomic protocol used and highlights the relevance of several rationales implemented in the protocol.  相似文献

The red spider mite, Tetranychus marianae McGregor (Acari: Tetranychidae), has been an important pest of eggplant Solanum melongena L. (Solanaceae) and other vegetables in the Mariana Islands. The damage due to T. marianae has been severe and caused huge economic losses. Because no threshold levels are available for T. marianae, many growers are applying up to 12 chemical applications per eggplant cropping period. This is not only expensive, but also results in lower yields because of extensive foliar damage and development of resistance in mites to chemicals. To diminish the calendar‐based chemical applications and to preclude damage to foliage and fruit quality, this study was undertaken for the development of a threshold level for optimum timing of chemical applications for T. marianae. In the direction of this aim, an attempt was made to generate different threshold levels by applying chemical spray (Sun‐spray 6E, 5 ml/l) within 12 h after reaching the threshold levels at 2, 4, 6, 8 and 10 mites/leaf, as well as current recommended calendar‐based sprays and non‐sprayed control in a replicated block design for the dry and wet seasons of 2010 at two locations (Yigo and Inaranjan) in Guam (USA). Based on T. marianae‐infested leaves, incidence of T. marianae and yield levels, the plots sprayed at 2 or 4 mites/leaf in the dry season and 2–8 mites/leaf during the wet season had significantly lower leaf damage and incidence of T. marianae compared with a greater number of mites/leaf in calendar‐based sprays and control plots. At the greater threshold levels, the mean yield was significantly reduced in comparison with the mean yield obtained when plots were sprayed at a threshold of 2–8 mites/leaf. Therefore, it is concluded that the optimum threshold chemical spray for T. marianae on eggplant is 4 mites/leaf during the dry season and 8 mites/leaf in the wet season.  相似文献

The occurrence of geranium rust (caused by Puccinia pelargonii‐zonalis) in commercial greenhouses can result in unmarketable plants and significant economic losses. Currently, detection of geranium rust relies solely on scouting for symptoms and signs of the disease. The purpose of this research was to develop a rapid detection assay for P. pelargonii‐zonalis‐infected tissues or urediniospores on greenhouse‐grown geraniums. Two oligonucleotide primers were designed based on internal transcribed spacer sequence data from three isolates of P. pelargonii‐zonalis. The primers amplified a 131‐bp product from genomic DNA from each isolate of P. pelargonii‐zonalis but did not amplify a product from genomic DNA from twelve other rust fungi or four other plant pathogenic fungi. A PCR product was amplified consistently from solutions that contained 1 ng or 100 pg/ml of purified P. pelargonii‐zonalis DNA in conventional PCR and at 1 pg/ml using real‐time PCR. The detection threshold was 102 urediniospores/ml for real‐time PCR and 104 urediniospores/ml for conventional PCR using urediniospores collected by vacuum from sporulating lesions. Puccinia pelargonii‐zonalis DNA was amplified by real‐time PCR from urediniospores washed from a single inoculated leaf, but recovered urediniospores were below detection threshold from one inoculated leaf with 5, 10, 25 and 50 non‐inoculated leaves. Conventional and real‐time PCR did not detect P. pelargonii‐zonalis in infected leaf tissues, presumably due to PCR inhibitors in the geranium leaf tissue. The inhibition of both conventional and real‐time PCR by geranium tissues suggests that a detection assay focusing on urediniospore recovery and microscopic examination with subsequent species verification by PCR may be the most efficient method for assessing the presence of geranium rust in greenhouses.  相似文献

Optimal foraging and optimal oviposition are two major forces leading to plant selection by insect females, but the contribution of these forces to the host‐selection process has been little studied for sucking herbivores. We studied feeding and oviposition behavior of a global pest, the bronze bug, Thaumastocoris peregrinus Carpintero & Dellapé (Heteroptera: Thaumastocoridae), using dual‐choice bioassays to evaluate the preference of females between host species, developmental leaf stage, or prior plant exposure to conspecifics. We assessed the link between these preferences and the performance of the offspring, by comparing survival and developmental time of nymphs reared on the various treatments. Finally, we compared the composition of the leaf wax of healthy and damaged leaves, and tested the effects of leaf wax on female preference behavior. Using healthy adult leaves of Eucalyptus tereticornis Sm. (Myrtaceae) as a reference, we found that females prefer to feed on Eucalyptus grandis W. Hill ex Maiden and E. tereticornis adult leaves that had been previously damaged by female conspecifics, whereas they reject juvenile leaves of E. tereticornis as food. Females also prefer to oviposit on leaves previously damaged by conspecifics but they rejected E. grandis as oviposition substrate. Nymphal performance varied among leaf treatments, suggesting a correlation with oviposition preference (but not feeding preference). Epicuticular wax extracts from damaged leaves contained higher concentrations of long‐chain, saturated linear alkanes, aldehydes, and alcohols than extracts from undamaged leaves. However, a choice assay failed to demonstrate an oviposition preference based on leaf surface wax chemistry. We discuss these findings in the context of the preference‐performance relationship.  相似文献

Light is considered a non‐limiting factor for vascular epiphytes. Nevertheless, an epiphyte's access to light may be limited by phorophyte shading and the spatio‐temporal environmental patchiness characteristic of epiphytic habitats. We assessed the extent to which potential light interception in Rodriguezia granadensis, an epiphytic orchid, is determined by individual factors (plant size traits and leaf traits), or environmental heterogeneity (light patchiness) within the crown of the phorophyte, or both. We studied 104 adult plants growing on Psidium guajava trees in two habitats with contrasting canopy cover: a dry tropical forest edge, and isolated trees in a pasture. We recorded the number of leaves and the leaf area, the leaf position angles, and the potential exposure of the leaf surface to direct irradiance (silhouette area of the leaf blade), and the potential irradiance incident on each plant. We found the epiphytes experience a highly heterogeneous light environment in the crowns of P. guajava. Nonetheless, R. granadensis plants displayed a common light interception strategy typical of low‐light environments, resembling terrestrial, forest understory plants. Potential exposure of the total leaf surface to direct irradiance correlated positively with plant size and within‐plant variation in leaf orientation. In many‐leaved individuals, within‐plant variation in leaf angles produced complementary leaf positions that enhanced potential light interception. This light interception strategy suggests that, in contrast to current wisdom, enhancing light capture is important for vascular epiphytes in canopies with high spatio‐temporal heterogeneity in light environments.  相似文献
