The whole plant of Arnebia hispidissima DC. (Boraginaceae) is used for the treatment of tongue and throat ailments in Indian traditional medicine. The present paper deals with the plants phytochemical constituents, the arnebins, and antimicrobial activities of its root extract. The antimicrobial activities were tested against Gram-positive and Gram-negative bacteria and fungi. The crude hexane extract demonstrated a potent antimicrobial effect against bacteria and a mild effect against fungi. Likewise, the hexane extract of cell cultures of A. hispidissima also showed mild bioefficacy against the select microorganisms.  相似文献

Arnebia hispidissima, which belongs to the family Boraginaceae, is an important medicinal and dye yielding plant. The alkannin, a red dye, are root-specific secondary metabolites of A. hispidissima. Shoots were regenerated from callus derived from immature inflorescence explants obtained from field grown plants. MS medium containing 4.52 μM 2, 4-D and 3.33 μM BAP was found to be most effective for the proliferation of callus, induced on medium containing 4.52 μM 2, 4-D. Maximum number (43.1 ± 0.25) with average length (5.2 ± 0.23) of shoots regenerated when callus was transferred to MS medium supplemented with 1.11 μM BAP, 1.16 μM Kin and 0.57 μM IAA. About 75.5 % of in vitro regenerated shoots were rooted on half-strength MS medium supplemented with 9.84 μM of IBA and 200 mg l−1 of activated charcoal. In comparison to in vitro, higher percent (90.2 %) of shoots were rooted under ex vitro conditions when treated with IBA (0.98 mM) for 5 min. Plantlets rooted in vitro as well as ex vitro were acclimatized successfully under the green house conditions. Ex vitro rooted plants exhibited higher survival percentage (75 %) as compared to in vitro rooted plantlets (60 %). Present study may be applicable in the large-scale root-specific red dye (alkannin) production via root induction under ex vitro condition.  相似文献

The olive tree is usually hermaphrodite but self-incompatible. In the Western Mediterranean some cultivars are totally male-sterile. Three different male-sterile phenotypes have been recognised. To infer the genetic basis of male sterility we studied its inheritance and cytoplasmic diversity in wild (oleaster) and cultivated Mediterranean olive. In the cross Olivière×Arbequina, the male-sterile trait was maternally inherited and affected all progenies. We also checked that both chloroplast and mitochondrial DNAs are maternally inherited. RFLP studies on chloroplast and mitochondrial DNAs revealed several cytotypes: two chlorotypes and four mitotypes in cultivars and oleaster (wild or feral Mediterranean olive). Furthermore, a total linkage desequilibrium between the CCK chlorotype and the MCK mitotype in cultivars and oleaster from different regions supports the fact that paternal leakage of organelles was not observed. The male sterility (ms 2) displayed by Olivière, plus six other cultivars and three oleaster was strictly associated with the CCK chlorotype and the MCK mitotype. These facts suggest that Olivière carries cytoplasmic male sterility. Male-fertile and male-sterile oleasters carrying this cytotype showed the presence of restorer alleles. This CMS might be due to a distant cross between olive taxa. The two other male-sterile phenotypes displayed by Lucques (ms 1) and Tanche (ms 3) were associated with the ME1 mitotype but we have not demonstrated CMS. The method described here allows the study of cell migration under confinement in one dimension. It is based on the use of microfabricated channels, which impose a polarized phenotype to cells by physical constraints. Once inside channels, cells have only two possibilities: move forward or backward. This simplified migration in which directionality is restricted facilitates the automatic tracking of cells and the extraction of quantitative parameters to describe cell movement. These parameters include cell velocity, changes in direction, and pauses during motion. Microchannels are also compatible with the use of fluorescent markers and are therefore suitable to study localization of intracellular organelles and structures during cell migration at high resolution. Finally, the surface of the channels can be functionalized with different substrates, allowing the control of the adhesive properties of the channels or the study of haptotaxis. In summary, the system here described is intended to analyze the migration of large cell numbers in conditions in which both the geometry and the biochemical nature of the environment are controlled, facilitating the normalization and reproducibility of independent experiments.  相似文献

Summary Restriction patterns of chloroplast (cp) and mitochondrial (mt) DNA in Brassica napus rapeseed reveal the alloplasmic nature of cytoplasmic male sterility in this crop. Both the Shiga and Bronowski systems probably exploit cytoplasmic diversity in B. napus cultivars arising from introgression of cytoplasm from the other rapeseed species, B. campestris. Nuclear genes specific to these systems do not cause sterility in maintainers (Bronowski and Isuzu-natane) because they have a campestris cytoplasm, but give rise to sterility in napus cytoplasms. In the course of hybridization to napus cultivars a line with the triazine resistant cytoplasm (a campestris cytoplasm) has undergone an alteration in the mt genome rendering its restriction pattern more similar than previously to that of napus. The alteration may be an inversion between 7.2 and 3.4 kb in length.  相似文献

为探讨巨噬细胞集落刺激因子(M-CSF)对人宫颈癌细胞(He La细胞)侵袭和迁移的影响及机制,采用胞质定位空载体p CMV/cyto/myc与重组载体p CMV/cyto/myc-M-CSF稳定转染He La细胞株,建立稳定高表达胞质M-CSF的细胞系(He La-M细胞).经Transwell实验观察胞质M-CSF对He La细胞侵袭和迁移能力的影响,逆转录-聚合酶链式反应及蛋白质印迹检测细胞Rho三磷酸鸟苷酶(Rho GTPases)及基质金属酶的表达,明胶酶谱法检测基质金属蛋白酶2的活性.结果显示,与转染空载体的He La细胞(He La-C细胞)和对照组He La细胞比较,胞质M-CSF的高表达可明显增强He La细胞在体外的侵袭和迁移能力,其机制与Rho GTPases的活化,以及MMP2表达上调及其活性增高密切相关.  相似文献

Summary Allozyme variation was examined in 22 populations of Pinus densiflora (Sieb, et Zucc.) and four geographic varieties of P. sylvestris (L.): var lapponica (Fries, Hartman), var armena (Komarov), var mongolica (Litvinov) and var sylvestriformis (Takenouchi). In addition, we developed paternal chloroplast (cp) DNA markers that distinguish P. densiflora from var lapponica, var armena and var mongolica. UPGMA cluster analysis based on Nei's distances between all pairwise combinations of the 22 populations revealed patterns corresponding strictly to geographic origin and taxonomic status. Analysis of allozyme variation in var lapponica, var armena and var mongolica demonstrated a high level of intrapopulational variability but a low level of interpopulational differentiation. It appears that the late Pleistocene blending of genetically diverse populations was responsible for the observed variation patterns. The constructed phylogenetic trees also showed late divergence of these three varieties. The var sylvestri formis was genetically distinct from the other three P. sylvestris varieties. The genetic distances separating var sylvestriformis from P. densiflora and the other taxa lend support to a separate taxonomic status for var sylvestriformis and a close relation with P. densiflora. We found that var sylvestriformis harbors admixtures of allozymes and cpDNA from both P. sylvestris and P. densiflora, which suggests an introgressive nature of this variety. Levels of intrapopulational variability were similar in P. sylvestris and P. densiflora, but interpopulational differentiation was much higher in P. densiflora. In the constructed phylogenetic trees, populations of this species were characterized by relatively long internode distances and branch lengths. The present results suggest that P. densiflora has a more advanced evolutionary age than P. sylvestris.  相似文献

Cell migration/proliferation processes associated with wound healing were measured in BeWo cells at 6 h, when mitosis is still scarce. Cells were cultured in medium with 1% fetal bovine serum to minimize proliferation. BeWo cell migration covered 20.6 ± 7.0%, 38.0 ± 5.4%, 16.6 ± 4.8% and 13.7 ± 3.6% of the wound when cultivated under control, aldosterone (100 nM, 12 h), aldosterone plus amiloride (10 μM) and amiloride treatments, respectively. When BeWo cells were treated with aldosterone, there was an increase in wound healing (P < 0.05), which was prevented by adding the ENaC blocker amiloride (P < 0.05, n = 16). Immunocytochemistry studies showed that the three ENaC subunits showed greater expression at the leading edge of the wound 3 h after injury, supporting the notion that these proteins participate in a postinjury signal. Antisense oligonucleotides directed against the α-ENaC subunit decreased the migratory response of the cells compared to the sense treated cells or the cells without oligonucleotides (P < 0.001, n = 16): 30.2 ± 3.7%, 17.6 ± 1.3%, 27.5 ± 1.5% and 20.2 ± 1.5% reinvasion of the wound with aldosterone, aldosterone plus antisense, aldosterone plus sense treatments and control conditions, respectively. Aldosterone and amiloride influence wound healing in BeWo cells, probably by their effects upon ENaCs, transmitting a signal to the cell cytoplasm for the release of several agents that promote cell migration.  相似文献

The N-terminal cytoplasmic region of the Kv11.1a potassium channel contains a Per-Arnt-Sim (PAS) domain that is essential for the unique slow deactivation gating kinetics of the channel. The PAS domain has also been implicated in the assembly and stabilization of the assembled tetrameric channel, with many clinical mutants in the PAS domain resulting in reduced stability of the domain and reduced trafficking. Here, we use quantitative Western blotting to show that the PAS domain is not required for normal channel trafficking nor for subunit-subunit interactions, and it is not necessary for stabilizing assembled channels. However, when the PAS domain is present, the N-Cap amphipathic helix must also be present for channels to traffic to the cell membrane. Serine scan mutagenesis of the N-Cap amphipathic helix identified Leu-15, Ile-18, and Ile-19 as residues critical for the stabilization of full-length proteins when the PAS domain is present. Furthermore, mutant cycle analysis experiments support recent crystallography studies, indicating that the hydrophobic face of the N-Cap amphipathic helix interacts with a surface-exposed hydrophobic patch on the core of the PAS domain to stabilize the structure of this critical gating domain. Our data demonstrate that the N-Cap amphipathic helix is critical for channel stability and trafficking.  相似文献

Summary The investigations were carried out with ten genetically diverse maize varieties and all possible crosses between them, including reciprocals, at two agro-climatically different locations in Punjab (India). The materials were studied in a split-split plot design with plant population level of 59200, 74000 and 98700 plants per hectare. The variances due to reciprocal cross effects were significant for plant height, ear height and ear girth in the pooled analysis and the effects were quite consistent over plant population levels and locations. The data on days to silk were recorded only at one location and highly significant reciprocal effects were observed. The cytoplasmic effects, however, did not depend on the cytoplasm alone but also on the interaction of genotype with cytoplasm. The reciprocal effects were more distinct in early × late combinations of varieties than in early × early or late × late combinations. Early parents when used as female tended to reduce plant and ear height and days to silk, indicating a common developmental pathway for these three traits. Since the cytoplasmic effects for these characters were not associated with significant effects on yield, the results can be of practical significance. The cytoplasmic effects for days to silk were maintained even in the F2 and back-crosses. These findings encourage the use of particular cytoplasm in developing early maturing varieties.  相似文献

The accumulation of the Kunitz-type chymotrypsin inhibitor WCI-3 in winged bean seeds is controlled developmentally. In vitro translation experiments showed that the WCI-3 mRNA was present in 35- and 40-day-old immature seeds after flowering. The size of the in vitro translation product is about 2 000 Da larger than that of the mature WCI-3 protein. The WCI-3 cDNA clones were isolated from a gtll cDNA library of 35-day-old immature seeds by immunoscreening. A nearly full-length cDNA clone was obtained containing an open reading frame of 207 amino acid residues. The deduced sequence of the 183 carboxy terminal amino acids coincides precisely with the amino acid sequence determined for purified WCI-3. The amino terminal extension of 24 residues has the characteristics of a signal peptide. Northern hybridization analysis of total poly(A)+ RNA showed that the WCI-3 mRNA is approximately 900 nucleotides long and accumulates in 35- and 40-day-old but not in 30-day-old immature seeds.  相似文献

The callosic wall which covers microsporocyte mother cells during meiotic division has been studied using different fluorochromes as alternatives to the widely used aniline blue. We have confirmed that both acridine orange and 4', 6' diamidino-2-phenylindole (DAPI) produce a fluorescent response to callose which is comparable in specificity and intensity to that of aniline blue: therefore, they can be used to study callose wall formation. Staining properties of these fluorochromes, as well of those of curcumin and sirofluor, reported earlier as fluorescent stains for callose, are discussed. We also discuss the efficacy of the combined use of sirofluor and DAPI to study particular aspects of the deposition of callose.  相似文献

Three partial S-RNase genes, MSRN-1, MSRN-2, and MSRN-3, in the Japanese apricot (Prunus mume Sieb. et Zucc.) were isolated from the three cultivars Nankou, Gyokuei, and Kairyouuchidaume, respectively. The structural characteristics revealed that S-RNase genes from the Japanese apricot were in the T2/SRNase-type S-RNase family with five conserved regions (C1, C2, C3, RC4, and C5) and one variable region (RHV) as reported in the other rosaceous plants. In the phylogenetic tree of T2/S SRNase-type RNases, three S-RNase genes of the Japanese apricot did not form a species-specific subgroup but the Prunus subfamily did. At least seven S-allelic genes were present in the Japanese apricot, and S-genotypes of six representative cultivars, including Nankou, Gyokuei, Kairyouuchidaume, Baigou, Kagajizou, and Oushuku were first established in this study as S 1 S 7, S 2 S 6, S 3 S 4, S 3 S 6, S 3 S 6 and S 1 S 5, respectively. Inward rectifier K+ (Kir) channels can be functionally categorized into two groups: those that are constitutively active and those that are constitutively inactive, with examples such as Kir2.x and Kir3.x, respectively. Their cytoplasmic regions are thought to be critical for control of channel gating, but a structural basis for this hypothesis is not known. In this study, we report a structure for the cytoplasmic region of a G protein-gated Kir channel, Kir3.2, and compare it with those of Kir3.1 and Kir2.1 channels. The isolated cytoplasmic region of Kir3.2 forms a tetrameric assembly in solution and also in the crystal. While the secondary structure arrangement and the subunit interface of the Kir3.2 crystal structure are found to be nearly identical to those of Kir3.1 and Kir2.1, it is quite different at and around loops between βC- and βD-strands and between βH- and βI-strands. These structural elements are located at the interface with the plasma membrane. Therefore, these structural elements could associate with the Kir channel transmembrane helices and be involved in the regulation of Kir channel gating.  相似文献

Lindera glauca (Sieb. et Zucc.) Blume (Lauraceae) has been used to treat rheumatic arthritis, stroke, and cardiac pain. Phytochemical investigation of twigs of L. glauca (Sieb. et Zucc.) Blume resulted in the isolation and identification of a new cerebroside, glaucerebroside (1). The structure of 1 was elucidated by a combination of extensive spectroscopic analyses, including extensive 2D NMR, HR-MS, chemical reactions, and LC/MS analysis. Compound 1 is a relatively rare cerebroside with l-threo-configuration of the sphingosine part. This is the second example of identification of a cerebroside from the family Lauraceae. Compound 1 significantly inhibited nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated BV-2 cells, with an IC50 value of 23.84 μM without inducing cell toxicity. This study suggests that glaucerebroside (1) can be an excellent candidate for development of novel anti-neuroinflammatory agents.  相似文献

The role of the cytoplasmic pH (pHc) of Eremosphaera viridisin the signal transduction chain after light-off from the chloroplaststo the K+ channels in the plasmalemma of this unicellular algawas investigated. The temporary opening of K+ channels is indicatedby a transient hypcrpolarization (TP). To record rapid changesof pHc, continuous measurements with pH sensitive micro-electrodeswere carried out. (i) Under normal conditions pHc in the light(7·56 ±0·2) did not differ from pHc inthe dark (7·62 ±0·2). (ii) The vacuolepH ranged between 4·8 and 5·2. (iii) After light-offa rapid transient acidification of pHc O19±0·07occurred and a TP was released, (iv) In every case, the startof the transient acidification after light-off preceded thehyperpolarization by about 3s. (v) Light-on caused a rapid transientalkalinization but never a TP. (vi) Change to acid externalmedium (3.2) transiently acidified the cytoplasm and was ableto release a TP. (vii) After addition of NH4Cl, pHc again showeda rapid transient acidification and the release of a TP. The origin of the protons appearing in the cytoplasm after light-offis discussed critically with respect to the buffer capacity.Either direct or indirect translocation is a possible mechanismfor the movement of H+ from the chloroplasts into the cytoplasm.The intracellular acidification and its relation to the openingof potassium channels in the plasmalemma leads us to suggestthat a sudden change of pHc is a potent internal signal factorin Eremosphaera viridis. Three crystalline compounds have been isolated from the Desmodium triquetrum (L.) DC. On the basis of their spectroscopic data and chemical correlations, the structures have been identified as friedelin (Ⅰ), epi-friedelinol (Ⅱ), stigmasterol (Ⅲ).
