Factor F430 is a yellow compound of unknown structure present in methanogenic bacteria. It has recently been shown to contain nickel. In this communication the influence of the nickel concentration in the growth medium on the factor F430 content of Methanobacterium thermoautotrophicum and on the nickel content of factor F430 was studied. It was found: (1) The content of factor F430 in the cells was strongly dependent on the nickel concentration of the growth medium. Cells grown on media with 2.5 M NiCl2 contained 28 times as much factor F430 per g as those grown on media with 0.075 M NiCl2; (2) factor F430 was synthesized in nickel deprived cells only upon the addition of nickel Nickel uptake paralleled factor F430 synthesis; (3) independent of the nickel concentration in the growth medium, the extinction coefficient at 430 nm of factor F430 per mol nickel was always near 22,500 cm-1 (mol Ni)-1. F430 is the prosthetic group of the methylcoenzyme M reductase of methanogenic bacteria. The compound isolated from Methanosarcina barkeri appears to be identical to the one obtained from the only distinctly related Methanobacterium thermoautotrophicum. F430 is thermolabile and in the presence of acetonitrile or C10 in4 sup- two epimerization products are obtained upon heating; in the absence of these compounds F430 is oxidized to 12, 13-didehydro-F430. The latter is stereoselectively reduced under H2 atmosphere to F430 by cell-free extracts of M. barkeri or M. thermoautotrophicum. Factors F430 from methanogenic bacteria have recently been shown to contain nickel and it has been speculated that they may have a nickel tetrapyrrole structure. This assumption was tested by determining whether succinate is incorporated by growing Methanobacterium thermoautotrophicum into three factors F430. Succinate is assimilated by Methanobacterium thermoautotrophicum into the amino acids glutamate, arginine and proline and into tetrapyrroles rather than other cell components. It was found that per mol nickel 8–9 mol of succinate were incorporated into the three factors F430 which is the amount predicted for a tetrapyrrole structure. Since the three factors F430 only contained significant amounts of glutamate rather than arginine or proline, the incorporation data suggest that factors F430 are nickel tetrapyrrole compounds. Spectral properties of the three factors F430, apparent molecular weights, and the absence of phosphor in these compounds are also described.  相似文献

Archaeoglobus fulgidus, a sulfate-reducing Archaeon with a growth temperature optimum of 83°C, uses the 5-deazaflavin coenzyme F420 rather than pyridine nucleotides in catabolic redox processes. The organism does, however, require reduced pyridine nuclcotides for biosynthetic purposes. We describe here that the Archaeon contains a coenzyme F420-dependent NADP reductase which links anabolism to catabolism. The highly thermostable enzyme was purfied 3600-fold by affinity chromatography to apparent homogeneity in a 60% yield. The native enzyme with an apparent molecular mass of 55 kDa was composed of only one type of subunit of apparent molecular mass of 28 kDa. Spectroscopic analysis of the enzyme did not reveal the presence of any chromophoric prosthetic group. The purified enzyme catalyzed the reversible reduction of NADP (apparent K M 40 M) with reduced F420 (apparent K M 20M) with a specific activity of 660 U/mg (apparent V max) at pH 8.0 (pH optimum) and 80°C (temperature optimum). It was specific for both coenzyme F420 and NADP. The distribution of the F420-reactive and F420-nonreactive hydrogenases from the methylotrophic Methanosarcina strain Gö1 indicated a membrane association of the F420-nonreactive enzyme. The membrane-bound F420-nonreactive hydrogenase was purified 42-fold to electrophoretic homogeneity with a yield of 26.7%. The enzyme had a specific activity of 359 mol H2 oxidized · min-1 · mg protein-1. The purification procedure involved dispersion of the membrane fraction with the detergent Chaps followed by anion exchange, hydrophobic and hydroxylapatite chromatography. The aerobically prepared enzyme had to be reactivated anaerobically. Maximal activity was observed at 80°C. The molecular mass as determined by native gel electrophoresis and gel filtration was 77000 and 79000, respectively. SDS gel electrophoresis revealed two polypeptides with molecular masses of 60000 and 40000 indicating a 1:1 stoichiometry. The purified enzyme contained 13.3 mol S2-, 15.1 mol Fe and 0.8 mol Ni/mol enzyme. Flavins were not detected. The amino acid sequence of the N-termini of the subunits showed a higher degree of homology to cubacterial uptake-hydrogenases than to F420-dependent hydrogenases from other methanogenic bacteria. Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. The responses to irradiance of photosynthetic CO2 assimilation and photosystem 2 (PS2) electron transport were simultaneously studied by gas exchange and chlorophyll (Chl) fluorescence measurement in two-year-old apple tree leaves (Malus pumila Mill. cv. Tengmu No.1/Malus hupehensis Rehd). Net photosynthetic rate (P N) was saturated at photosynthetic photon flux density (PPFD) 600-1 100 (mol m-2 s-1, while the PS2 non-cyclic electron transport (P-rate) showed a maximum at PPFD 800 mol m-2 s-1. With PPFD increasing, either leaf potential photosynthetic CO2 assimilation activity (Fd/Fs) and PS2 maximal photochemical activity (Fv/Fm) decreased or the ratio of the inactive PS2 reaction centres (RC) [(Fi – Fo)/(Fm – Fo)] and the slow relaxing non-photochemical Chl fluorescence quenching (qs) increased from PPFD 1 200 mol m-2 s-1, but cyclic electron transport around photosystem 1 (RFp), irradiance induced PS2 RC closure [(Fs – Fo)/Fm – Fo)], and the fast and medium relaxing non-photochemical Chl fluorescence quenching (qf and qm) increased remarkably from PPFD 900 (mol m-2 s-1. Hence leaf photosynthesis of young apple leaves saturated at PPFD 800 mol m-2 s-1 and photoinhibition occurred above PPFD 900 mol m-2 s-1. During the photoinhibition at different irradiances, young apple tree leaves could dissipate excess photons mainly by energy quenching and state transition mechanisms at PPFD 900-1 100 mol m-2 s-1, but photosynthetic apparatus damage was unavoidable from PPFD 1 200 mol m-2 s-1. We propose that Chl fluorescence parameter P-rate is superior to the gas exchange parameter P N and the Chl fluorescence parameter Fv/Fm as a definition of saturation irradiance and photoinhibition of plant leaves.  相似文献

A fluorescent pigment was isolated from the culture fluid of Methanobacterium thermoautotrophicum strain H. This pigment was shown to be 7,8-didemethyl-8-hydroxy-5-deazariboflavin by various spectroscopic and chromatographic techniques. This compound was previously described as the FO acid hydrolysis fragment of coenzyme F420. On the basis of the time of appearance of the pigment in the course of fermentation, it is suggested that this substance may be an over-produced biosynthetic precursor of F420.  相似文献

A toxic factor released from disrupted cells of Vibrio parahaemolyticus was partially purified by gel filtration after precipitation with (NH4)2SO4 at 40% saturation. The factor, which was a thermostable protein of 63 kDa, lysed human erythrocytes at a concentration of 0.15 g ml-1. Its LD50 by intravenous injection into mice was 6.4 g. Fluid accumulated in suckling mice force-fed with the toxic material (1 to 25 g). Haemolytic activity, which occurred maximall at 37°C and pH 7.0 was enhanced by Ca2+, Cu2+ and Zn2+, each at 1 mm. Anti-toxic-factor serum agglutinated V. parahaemolyticus cells. The factor may play a role in the pathogenesis of V. parahaemolyticus infections and in the host's defence mechanisms against infection by the microorganism.  相似文献

Methanopyrus kandleri belongs to a novel group of abyssal methanogenic archaebacteria that can grow at 110°C on H2 and CO2 and that shows no close phylogenetic relationship to any methanogens known so far. N 5 N 10 -Methylenetetrahydromethanopterin reductase, an enzyme involved in methanogenesis from CO2, was purified from this hyperthermophile. The apparent molecular mass of the native enzyme was found to be 300 kDa. Sodium dodecylsulfate/polyacrylamide gel electrophoresis revealed the presence of only one polypeptide of apparent molecular mass 38 kDa. The ultraviolet/visible spectrum of the enzyme was almost identical to that of albumin indicating the absence of a chromophoric prosthetic group. The reductase was specific for reduced coenzyme F420 as electron donor; NADH, NADPH or reduced dyes could not substitute for the 5-deazaflavin. The catalytic mechanism was found to be of the ternary complex type as deduced from initial velocity plots. V max at 65°C and pH 6.8 was 435 U/mg (kcat=275 s-1) and the K m for methylenetetrahydro-methanopterin and for reduced F420 were 6 M and 4 M, respectively. From Arrhenius plots an activation energy of 34 kJ/mol was determined. The Q 10 between 40°C and 90°C was 1.5.The reductase activity was found to be stimulated over 100-fold by sulfate and by phosphate. Maximal stimulation (100-fold) was observed at a sulfate concentration of 2.2 M and at a phosphate concentration of 2.5 M. Sodium-, potassium-, and ammonium salts of these anions were equally effective. Chloride, however, could not substitute for sulfate or phosphate in stimulating the enzyme activity.The thermostability of the reductase was found to be very low in the absence of salts. In their presence, however, the reductase was highly thermostable. Salt concentrations between 0.1 M and 1.5 M were required for maximal stability. Potassium salts proved more effective than ammonium salts, and the latter more effective than sodium salts in stabilizing the enzyme activity. The anion was of less importance.The N-terminal amino acid sequence of the reductase from M. kandleri was determined and compared with that of the enzyme from Methanobacterium thermoautotrophicum and Methanosarcina barkeri. Hydrogenase was solubilized from the membrane of acetate-grown Methanosarcina barkeri MS and purification was carried out under aerobic conditions. The enzyme was reactivated under reducing conditions in the presence of H2. The enzyme showed a maximal activity of 120±40 mol H2 oxidized · min–1 · min–1 with methyl viologen as an electron acceptor, a maximal hydrogen production rate of 45±4 mol H2 · min–1 · mg–1 with methyl viologen as electron donor, and an apparent K m for hydrogen oxidation of 5.6±1.7 M. The molecular weight estimated by gel filtration was 98,000. SDS-PAGE showed the enzyme to consist of two polypeptides of 57,000 and 35,000 present in a 1:1 ratio. The native protein contained 8±2 mol Fe, 8±2 mol S2–, and 0.5 mol Ni/mol enzyme. Cytochrome b was reduced by hydrogen in a solubilized membrane preparation. The hydrogenase did not couple with autologous F420 or ferredoxin, nor with FAD, FMN, or NAD(P)+. It was recently reported that the extreme thermophile Methanopyrus kandleri contains only a H2-forming N 5, N 10-methylenetetrahydromethanopterin dehydrogenase which uses protons as electron acceptor. We describe here the presence in this Archaeon of a second N 5,N 10-methylenetetrahydromethanopterin dehydrogenase which is coenzyme F420-dependent. This enzyme was purified and characterized. The enzyme was colourless, had an apparent molecular mass of 300 kDa, an isoelectric point of 3.7±0.2 and was composed of only one type of subunit of apparent molecular mass of 36 kDa. The enzyme activity increased to an optimum with increasing salt concentrations. Optimal salt concentrations were e.g. 2 M (NH4)2SO4, 2 M Na2HPO4, 1.5 M K2HPO4, and 2 M NaCl. In the absence of salts the enzyme exhibited almost no activity. The salts affected mainly the V max rather than the K m of the enzyme. The catalytic mechanism of the dehydrogenase was determined to be of the ternary complex type, in agreement with the finding that the enzyme lacked a chromophoric prosthetic group. In the presence of M (NH4)2SO4 the V max was 4000 U/mg (k cat=2400 s-1) and the K m for N 5,N 10-methylenetetrahydromethanopterin and for coenzyme F420 were 80 M and 20 M, respectively. The enzyme was relatively heat-stable and lost no activity when incubated anaerobically in 50 mM K2HPO4 at 90°C for one hour. Methylene-H4MPT reductase was found to be present in Archaeoglobus fulgidus in a specific activity of 1 U/mg. The reductase was purified 410-fold. The native enzyme showed an apparent molecular mass of approximately 200 kDa. Sodium dodecylsulfate/polyacrylamide gel electrophoresis revealed the presence of only 1 polypeptide of apparent molecular mass 35 kDa. The ultraviolet/visible spectrum of the reductase was almost identical to that of albumin indicating the absence of a chromophoric prosthetic group. The reductase was dependent on reduced coenzyme F420 as electron donor. Neither NADH, NADPH, nor reduced viologen dyes could substitute for the reduced deazaflavin. From reciprocal plots, which showed an intersecting patter, a K m for methylene-H4MPT of 16 M, a K m for F420H2 of 4 M, and a V max of 450 U/mg (Kcat=265 s-1) were obtained. The enzyme was found to be rapidly inactivated when incubated at 80°C in 100 mM Tris/HCl pH 7. The rate of inactivation, however, decreased to essentially zero in the presence of either F420 (0.2 mM), methylene-H4MPT (0.2 mM), albumin (1 mg/ml), or KCl (0.5 M). The N-terminal amino acid sequence was determined and found to be similar to that of methylene-H4MPT reductase (F420-dependent) from the methanogens Methanobacterium thermoautotrophicum, Methanosarcina barkeri, and Methanopyrus kandleri. The marine green sulfur bacterium Chlorobium vibrioforme strain 1930 produced H2 and elemental sulfur from sulfide or thiosulfate under N limitation in the light. H2 production depended on nitrogenase and occurred only in the absence of ammonia. Methionine sulfoximine, an inhibitor of glutamine synthetase, prevented the switch-off by ammonia. In defined syntrophic cocultures of the acetate-oxidizing, sulfur-reducing bacterium Desulfuromonas acetoxidans with green sulfur bacteria, H2 was produced from acetate via a light-driven sulfur cycle. The sulfur-reducing bacterium could not be replaced by sulfate-reducing bacteria in these experiments. In a coculture of the marine Chlorobium vibrioforme strain 1930 and the sulfur-reducing bacterium Desulfuromonas acetoxidans strain 5071, optimum long-term H2 production from acetate was obtained with molecular nitrogen as N source, at low light intensity (110 mol · m-2 · s-1), in sulfide-reduced mineral medium (2 mM Na2S) at pH 6.8. Traces of sulfide (10 M) were sufficient to keep the sulfur cycle running. The coculture formed no poly--hydroxyalkanoates (PHA), but 20%–40% polysaccharide per cell dry mass. Per mol acetate added, the coculture formed 3.1 mol of H2 (78% of the theoretical maximum). Only 8% of the reducing equivalents was incorporated into biomass. Exchange of CO2 and O2 and chlorophyll fluorescence were measured in the presence of 360 1 · 1–1 CO2 in nitrogen in Helianthus annuss L. leaves which had been preconditioned in the dark or at a photon flux density (PFD) of 24 mol · m–2 · s–1 either in 21 or 0% O2. An initial light-dependent O2 outburst of 6 mol · m–2 was measured after aerobic dark incubation. It was attributed to the reduction of electron carriers, predominantly plastoquinone. The maximum initial rate of O2 evolution at PFD 8000 mol · m–2 · s–1 was 170 mol · m–2 · s–2 or about four times the steady CO2-and light-saturated rate of photosynthesis. Fluorescence measurements showed that the rate was still acceptor-limited. Fast O2 evolution ceased after electron carriers were reduced in the dark-adapted leaf, but continued for a short time at the lower rate of 62 mol · m–2 · s–1 in the light-adapted leaf. The data are interpreted to show that enzymes involved in 3-phosphoglycerate reduction are dark-inhibited, but were fully active in low light. In a dark-adapted leaf, respiratory CO2 evolution continued under nitrogen; it was partially inhibited by illumination. Prolonged exposure of a leaf to anaerobic conditions caused reducing equivalents to accumulate. This was shown by a slowly increasing chlorophyll fluorescence yield which indicated the reduction of the PSII acceptor QA in the dark. When the leaf was illuminated, no O2 evolution was detected from short light pulses, although transient O2 production was appreciable during longer light pulses. Growing cultures of Methanobacterium thermoautotrophicum were supplemented with [U-14C]adenosine or [1-14C]adenosine. 7,8-Didemethyl-8-hydroxy-5-deazariboflavin (factor F0) and 7-methylpterin were isolated from the culture medium. Hydrolysis of cellular RNA yielded purine and pyrimidine nucleotides. The ribose side chain of proffered adenosine is efficiently incorporated into cellular adenosine and guanosine nucleotide pools but not into pyrimidine nucleotides. Thus, M. thermoautotrophicum can utilize exogenous adenosine by direct phosphorylation without hydrolysis of the glycosidic bond, and AMP can be efficiently converted to GMP. Factor F0 and 7-methylpterin had approximately the same specific activities as the purine nucleotides. It follows that the ribityl side chain of factor F0 is derived from the ribose side chain of a nucleotide precursor by reduction. Methyl viologen (MV) (20–150 M), a generator of superoxide anion (O2 ), but not hydrogen peroxide (H2O2) (10 M–2 mM) triggered the formation of cryptotanshinone (a phytoalexin) in cultures of both crown galls and hairy roots of Salvia miltiorrhiza. MV also inhibited the biomass formation and decreased the contents of phenolic acids in both cultures whereas H2O2 did not. In addition, MV and yeast elicitor induced cryptotanshinone formation synergistically only in crown gall cultures. Treatment of the cultures with 3.3 M diphenylene iodonium, an inhibitor of NAD(P)H oxidase, did not exhibit any detrimental effect on the yeast elicitor-induced cryptotanshinone formation in hairy root cultures whereas 1 M diphenylene iodonium was inhibitory on yeast elicitor-induced cryptotanshinone formation in crown gall cultures.  相似文献

An assay is described that allows the direct measurement of the enzyme activity catalyzing the transfer of the methyl group from N 5-methyltetrahydromethanopterin (CH3–H4MPT) to coenzyme M (H–S–CoM) in methanogenic archaebacteria. With this method the topology, the partial purification, and the catalytic properties of the methyltransferase in methanol- and acetate-grown Methanosarcina barkeri and in H2/CO2-grown Methanobacterium thermoautotrophicum were studied. The enzyme activity was found to be associated almost completely with the membrane fraction and to require detergents for solubilization. The transferase activity in methanol-grown M. barkeri was studied in detail. The membrane fraction exhibited a specific activity of CH3–S–CoM formation from CH3–H4MPT (apparent K m=50 M) and H–S–CoM (apparent K m=250 M) of approximately 0.6 mol·min-1·mg protein-1. For activity the presence of Ti(III) citrate (apparent K m=15 M) and of ATP (apparent K m=30 M) were required in catalytic amounts. Ti(III) could be substituted by reduced ferredoxin. ATP could not be substituted by AMP, CTP, GTP, S-adenosylmethionine, or by ATP analogues. The membrane fraction was methylated by CH3–H4MPT in the absence of H–S–CoM. This methylation was dependent on Ti(III) and ATP. The methylated membrane fraction catalyzed the methyltransfer from CH3–H4MPT to H–S–CoM in the absence of ATP and Ti(III). Demethylation in the presence of H–S–CoM also did not require Ti(III) or ATP. Measurement of the light response of photosynthetic CO2 uptake is often used as an implement in ecophysiological studies. A method is described to calculate photosynthetic parameters, such as the maximum rate of whole electron transport and dissimilative respiration in the light, from the light response of CO2 uptake. Examples of the light-response curves of flag leaves and ears of wheat (Triticum aestivum cv. Grain sorghum [Sorghum bicolor (L.) Moench. cvs. TX430 and KS82] was grown in a Haynie very fine sandy loam (coarse-silty, mixed, superactive, calcareous, mesic Mollic Udifluvents) under constant 47 % shade or full irradiance in a greenhouse under two watering regimes to see the combined and individual effects of low irradiance (LI) and low water (LW) on the sorghum genotypes. Under the high-irradiance (HI) and high-water (HW) treatment (control) and the LI-HW treatment, TX430 grew taller than KS82. Both LI and LW reduced several times the fresh and dry masses. Under the control conditions, TX430 reached its maximum net photosynthetic rate (P Nmax) of 28.93 mol m–2 s–1 at a photosynthetic photon flux density (PPFD) of 1 707 mol m–2 s–1, and KS82 reached its P Nmax of 28.32 mol m–2 s–1 at a PPFD of 2 973 mol m–2 s–1. The fact that TX430 had P Nmax under a lower PPFD than KS82 may relate to its taller growth under LI conditions. Hence genotypes of sorghum might be selected for low irradiance using curves relating P N to PPFD.  相似文献
