通过实验和临床观察幽门螺杆菌(Helicobacter pylori)对胃上皮细胞间隙连接超微结构的影响,从细胞间隙连接角度探讨H. pylori致癌机制.将不同H. pylori菌株与BGC-823细胞共培养24 h或 48 h,用原位固定与原位包埋法透射电镜观察细胞间隙连接超微结构变化.对70例胃癌患者,用快速尿素酶试验、碱性品红染色和14C尿素呼气实验检测H. pylori,PCR法检测H. pylori CagA基因,及透射电镜观察胃上皮细胞间隙连接超微结构变化.结果显示,未加H. pylori组BGC-823细胞可见较多细胞连接及连接复合体,加H. pylori各组细胞的连接数、单位周长连接数与单位周长连接长度均小于未加H. pylori组,而细胞间隙最小宽度大于未加H. pylori组(P < 0.001或P < 0.005),且CagA+ 的NCTC J99组、临床株GC 01组和NCTC 11639组细胞连接数、单位周长连接数均小于CagA- 的NCTC 12908组(P < 0.001或P < 0.05),NCTC J99组与临床株GC 01组细胞单位周长连接长度短于NCTC 12908组(P < 0.001).胃癌患者H. pylori感染组细胞连接数、单位周长连接数与单位周长连接长度均小于无H. pylori感染组,细胞间隙最小宽度大于无H. pylori感染组(P < 0.001),且CagA+ H. pylori感染者细胞连接数、单位周长连接数与单位周长连接长度均小于CagA- H. pylori感染者,细胞间隙最小宽度大于CagA- H. pylori感染者.上述结果表明,胃上皮细胞间隙连接改变与H. pylori感染,特别是CagA+ H. pylori感染有关.  相似文献

研究幽门螺杆菌 (Helicobacterpylori,Hp)ureB基因重组子转染胃上皮细胞后对胃上皮细胞的作用。用PCR方法从Hp标准株NCTC116 37中获取ureB全长基因 ,将其开放读码框架定向克隆入真核表达载体pcDNA3 1;获得的重组子转染SGC 790 1细胞 ,筛选耐潮霉素的细胞克隆 ,用RT PCR方法检测细胞内ureB基因在转录水平的表达 ;分别用荧光染色技术、MTT、流式细胞术检测UreB对细胞表型、增殖、凋亡及细胞周期的影响。UreB阳性表达的细胞 (SureB)胞膜出芽、细胞皱缩 ;用MTT法检测细胞增殖 ,结果表明 ,SureB细胞与SpcDNA3 1细胞比较 (pcDNA3 1转染的细胞 ) ,生长增殖无显著性差异 (P >0 0 5 ) ,流式细胞术检测细胞凋亡结果显示 ,SureB的凋亡率显著高于SpcDNA3 1(P值为 0 0 0 7) ;细胞周期分析显示 ,SureB细胞有S期比率增高、G2 M、G0 G1 期比率下降的趋势。ureB在培养细胞内的表达可促进细胞凋亡  相似文献

目的和方法 :以Giemsa染色法、DNA条带分析、流式细胞仪分析和TUNEL法观察幽门螺杆菌 (Helicobacterpylor ,Hp)菌体超声粉碎提取物对人胃上皮细胞株BGC 82 3凋亡的影响。 结果 :四种方法的检测结果均显示Hp提取物能诱导BGC 82 3细胞凋亡 ,效应具有剂量依赖和时间依赖关系。结论 :Hp可能对胃粘膜细胞凋亡有直接的诱导作用。  相似文献

目的 探讨幽门螺杆菌(Helicobacter pylori)急性感染对GES 1细胞凋亡的影响,揭示H. pylori引起GES 1细胞凋亡变化的分子机制。 方法 将H. pylori临床分离株SBK与胃上皮细胞GES 1按不同比例(感染复数MOI分别为50∶1和100∶1)共培养24 h,建立H. pylori急性感染模型。采用流式细胞仪分析GES 1的凋亡,通过Western Blot检测凋亡相关蛋白Bcl xL、Bcl 2、Bax、Caspase 3和NF κB p65的表达。经H. pylori感染的GES 1细胞为处理组细胞,未经H. pylori感染的GES 1细胞即为对照细胞。使用SPSS 21.0软件对结果进行统计学分析。 结果 GES 1细胞经H. pylori处理24 h后,与对照细胞相比,MOI为50∶1(t=11.040,P结论 H. pylori急性感染通过改变线粒体途径中凋亡相关蛋白Bax、Bcl 2及Bcl xL的表达促进GES 1细胞凋亡,且GES 1细胞的凋亡程度与H. pylori的感染复数有关。  相似文献

幽门螺杆菌是公认的胃相关疾病的病原菌,而空泡毒素(VacA)是其主要的致病因子,它除了可以引起细胞空泡样病变外,现已被广泛认为是一种凋亡诱导因子,它能通过线粒体、MAPK、NO等信号转导通路介导细胞凋亡,在诱导细胞凋亡的同时又与机体的免疫系统相互作用。从细胞动力学角度探讨VacA和细胞凋亡的关系已成为研究的热点,本文总结了VacA诱导细胞凋亡机制的最新研究进展,对研究VacA损伤胃黏膜机制以及相关疫苗的研制具有重要意义。  相似文献

目的:探讨肝癌与H.pylori感染的相关性.方法:采用金标免疫斑点法和酶免疫检测法(EIA)分别检测33例肝癌患者及169例普通成人Hp-Ab和CagA-Hp-IgG抗体.结果:肝癌组Hp-Ab和CagA -Hp-IgG检出率分别为78.8%和45.5%,而普通成人组分别为55.0%和27.2%,经x2检验,P<0.05,差异均有显著性.结论:肝癌患者由于整体免疫力低下,易受H.pylori感染.  相似文献

目的:表达和纯化幽门螺杆菌不同菌株的CagA蛋白N端片段,检测其与磷脂酰丝氨酸(PS)的相互作用及亲和力。方法:用PCR方法从幽门螺杆菌3个菌株中扩增出CagA蛋白N端基因,并连接到表达载体pET-28a上;转化大肠杆菌BL21,经IPTG诱导可溶性表达CagA蛋白N端880残基片段;经镍柱亲和纯化后,利用PLOA法检测CagA蛋白与PS的相互作用。结果:构建了3种幽门螺杆菌菌株cagA基因的原核表达质粒pET-28a/cagAJ99、pET-28a/cagA11637及pET-28a/cagASS1,并在大肠杆菌中获得可溶性表达,SDS-PAGE和Western印迹证实得到目标融合蛋白,亲和纯化得到高纯度CagA蛋白。PLOA结果表明,CagA蛋白与PS有明显的相互作用。结论:3种幽门螺杆菌菌株CagA蛋白与PS之间存在相互作用,且不同的CagA与PS有不同的亲和力。  相似文献

Helicobacter pylori has been proposed as a causative agent of gastric cancer. The aim of this study was to define serum antibodies response against different H. pylori antigens in patients with gastric cancer. Serum samples were collected from 115 Lithuanian patients with non-cardia gastric cancer and 110 age- and sex-matched controls without cancer. Heat-stable, low-molecular-mass, and outer membrane proteins were used as antigens to analyze serum IgG antibody response against H. pylori by enzyme-linked immunosorbent assay. Seroprevalence of H. pylori using low-molecular-mass antigen was significantly higher in gastric cancer patients, compared to controls (77% versus 57%, p<0.05). Significant differences in the prevalence of H. pylori infection between gastric cancer patients and controls were found in females using all three studied antigens: heat-stable (98% versus 84%, p<0.05), low-molecular-mass (88% versus 48%, p<0.05) and outer membrane proteins (78% versus 57%, p<0.05). In males, no significant differences were revealed between gastric cancer patients and controls. There may be other cofactors in addition to H. pylori that are important for the development of gastric cancer. H. pylori seems, however, to be a more important for development of gastric cancer in females than in males or males may have more confounding risk factors for gastric cancer than females.  相似文献

除幽门螺杆菌之外,胃黏膜内还定居着大量细菌,占主导地位的是厚壁菌门、变形菌门、拟杆菌门、放线菌门和梭杆菌门。幽门螺杆菌和胃黏膜菌群之间可通过竞争营养和空间、扰乱抑菌肽的分泌以及改变宿主胃生理环境等直接或间接相互影响。本研究总结了胃内正常菌群的组成特征,分析了胃黏膜菌群与幽门螺杆菌之间的相互关系及其潜在机制,并进一步探讨了胃黏膜菌群对幽门螺杆菌相关胃部疾病的影响,有利于深入理解慢性胃病的发病机制,为疾病预防及治疗提供理论依据。  相似文献

目的:了解球形幽门螺杆菌对体外培养细胞增殖的影响.方法:采用抗生素诱导幽门螺杆菌标准菌株NCTC11637球变,将弯曲形和球形菌的菌悬液(1×108个/ml)作5倍梯度稀释,而后分别加入胃癌上皮细胞SGC-7901,共孵育3 d后,用MTT法检测SGC-7901细胞增殖的情况.结果:高浓度的弯曲形和球形幽门螺杆菌(>8×107个/ml)可明显抑制细胞的增殖(P<0.05);低浓度的球形幽门螺杆菌(<1.28×105个/ml)可明显促进细胞的增殖(P<0.05).结论:球形幽门螺杆菌在体外可影响SGC-7901细胞的增殖.  相似文献

目的 观察幽门螺杆菌(Helicobacter pylori, H. pylori)感染的慢性非萎缩性胃炎(no-atrophic gastritis,NAG)→萎缩性胃炎(chronic atrophic gastritis,CAG)→肠上皮化生(intestinal metaplasia,IM)→非典型增生(dysplasia,DYS)→胃癌(gastric cancer,GC)五个不同阶段miR-1、miR-20a、miR-34a、miR-423-5p表达变化规律及其与临床的关系。方法 收集胃镜及病理证实的上述胃癌发生五个不同阶段且H. pylori感染的患者(依次为44、47、43、50、45例),胃癌无H. pylori感染者46例,胃黏膜正常(normal gastric mucose,NGM)63例的血清标本,采用Real-time PCR法检测无H. pylori感染者miR-1、miR-20a、miR-34a、miR-423-5p表达。结果 H. pylori感染NAG→GC不同阶段,miR-1、miR-20a、miR-34a、miR-423-5p表达逐渐升高(P<0.05),GC阶段最高,miR-1、miR-20a CAG→GC阶段均高于NGM(P<0.05),与NGM比较差异有统计学意义(P<0.05);其表达程度与GC发生阶段呈正相关(P<0.001);GC组H. pylori感染者较无H. pylori感染者miR-1、miR-20a、miR-34a、miR-423-5p表达升高(P<0.05)。结论 H. pylori感染CAG→GC阶段miR-1、miR-20a、miR-34a、miR-423-5p表达升高,向胃癌演进中呈逐渐升高趋势,miR-1、miR-20a、miR-34a、miR-423-5p高表达可能是H. pylori感染后导致胃癌发生发展的重要机制,miR-1、miR-20a、miR-34a可作为诊断早期胃癌的标记物。  相似文献

Helicobacter pylori is a major factor for the development of gastric cancer. The aim of this study was to define serum antibody patterns associated with H. pylori infection in patients with gastric cancer using a Western blot technique. Serum samples collected from 115 patients with gastric cancer and 110 age- and gender-matched patients without gastrointestinal diseases were tested for IgG antibodies to H. pylori antigens (outer membrane proteins and whole cell preparations). No significant differences were found between patients with and without gastric cancer using outer membrane proteins (82% and 73%, P>0.05) or whole cell antigens (84% and 76%, P>0.05), respectively. The significant differences between patients with and without gastric cancer were associated with bands of 94 kDa (54% and 20%, P<0.001) and 30 kDa (65% and 44%, P<0.01). A combination of antibodies to 85 kDa (VacA) and 120 kDa (CagA) was significantly (P<0.01) more frequent in gastric cancer patients than in patients without gastric cancer. The detection of antibodies to 94- and 30-kDa bands, in association with the determination of serum antibodies to CagA+/VacA+, may have a prospective value in assessment of the risk of developing of gastric cancer.  相似文献

Background. Relatives of gastric cancer patients have an increased risk of gastric cancer, possibly related to genetically‐related strains of Helicobacter pylori or a common environment. Methods. The pattern of gastritis and H. pylori from gastric cancer patients and their first‐degree relatives were compared using detailed DNA fingerprints and vacA, cagA, and iceA genotyping. Results. Sixteen index cases from Korea, the US, or Colombia and their 38 first‐degree relatives (brothers, sisters, sons and daughters) were studied. No definite, or consistent, relationship between the pattern of gastritis and the relatedness of the H. pylori strain was observed (i.e. relatives could have an identical or a totally different pattern of gastritis regardless if they were infected with identical or highly similar organisms). For example, three elderly siblings of an index case with atrophic pangastritis had identical H. pylori isolates and environments in childhood and yet two had antral predominant nonatrophic gastritis, which is typically associated with duodenal ulcer instead of gastric cancer. Conclusions. The results of this study are not consistent with the hypothesis that specific virulence factors or similar H. pylori strains correlate with a specific histologic pattern or outcome even among those sharing the same environment in childhood.  相似文献

目的探讨胃息肉与幽门螺杆菌(Helicobacter pylori,H.pylori)感染关系。方法对1218例胃息肉同时进行H.pylori检查患者进行回顾性分析,分析胃息肉患者H.pylori感染率、胃息肉部位与H.pylori感染关系、胃息肉病理类型与H.pylori感染关系。结果发现胃息肉Hpylori感染患者532例,Hpylori感染率为43.7%。男性胃息肉患者H.pylori感染率为47.5%(216/455),女性Hpriori感染率感染率为41.4%(316/763)(P〉0.05),年龄〈20岁、20~39岁、40—59岁和≥60岁胃息肉H.priori感染率分别为41.7%、44.7%、41.6%和47.2%(P〉0.05);胃窦胃角息肉H.pylori感染率高于其他部位(胃体、胃底和贲门)(P〈0.05);炎性和增生性胃息肉H.priori感染率高于胃底腺和腺瘤性息肉(P〈0.05)。结论H.pylori感染可能与部分胃息肉发生有一定关系,需要进一步深入研究胃息肉的发生机制。  相似文献
