Whole cells of the methylotrophic bacteriumMethylophilus methylotrophus cultured under methanol-limited conditions contain approximately equal amounts of two majorc-type cytochromes,c H andc L. Virtually all of the cytochromec H, and over one-third of the cytochromec L, are loosely attached to the periplasmic surface of the respiratory membrane whence they can be released by sonication or by washing cells in ethylenediaminetetraacetate (EDTA). The latter causes inhibition of methanol oxidase activity and stimulation of ascorbate-N,N,N,N-tetramethyl-p-phenylenediamine (TMPD) oxidase activity, neither of which effects are reversible by divalent metal ions. Kinetic analyses indicate that ascorbate-TMPD is oxidised via two routes, viz. a slow low-affinity pathway involving loosely membrane-boundc-type cytochromes plus cytochrome oxidaseaa 3, and a faster higher-affinity pathway involving the firmly membrane-bound cytochrome oxidasec L o complex; the former route predominates in the presence of divalent metal ions, and the latter route after exposure to EDTA. The respiratory pigments of Prototheca zopfii include seven cytochromes: two c-type cytochromes, a soluble c(549) and a membrane bound c(551); three b-type cytochromes, b(555), b(559) and b(564); and cytochromes a and a3. Cytochromes a and a3 could be resolved spectrally in the α-band region by reducing the cells in the presence of methanol and cyanide. Methanol shifted the absorption maximum of cytochrome a from 598 to 603 nanometers and permitted dithionite (or substrate) to reduce the cyanide-cytochrome a3 complex to give a well defined 595-nanometer absorption band. Methanol did not interfere with CO binding by cytochrome a3, and CO did not alter the methanol effect on cytochrome a. Azide and cyanide, which partially inhibited exogenous respiration, stimulated endogenous respiration. Frozen steady states of the electron transport chain in the presence of cyanide and azide indicated that the stimulation by these inhibitors was due to an increased autooxidation of one of the b-type cytochromes, possibly b(564).  相似文献

Seven strains of bacteria belonging to the Beggiatoa-Vitreoscilla group were studied for their respiratory activity and for the presence of electron transport conponents. All strains tested oxidized [1-14C] and [2-14C] acetate to 14CO2 at relatively high rates. All strains tested were N,N,N,N-tetramethylphenylenediamine (TMPD)-oxidase positive and contained spectra representing a-type and carbon monoxide-binding cytochromes. Most of the strains also contained spectra representing c-type and b-type cytochromes. Beggiatoa alba B18LD contained b-type, a-type, c-type and CO-binding cytochromes, the latter two being located in the 144,000 x g soluble fraction. The cytochromes of the filamentous bacteriaStreptomyces clavuligerus andSaccharopolyspora erythraea have been studied by low-temperature (77K) difference spectra and room-temperature potentiometric titrations. Difference spectra of membranes fromSa. erythraea indicate the presence of twoc- and twob-type cytochromes. A furtherb-type cytochrome is revealed by potentiometric titration. The soluble and membrane-bound forms of a green pigment, believed to be involved in electron transport to oxygen, are each shown to have complex absorption spectra which, in the case of the soluble form, may be potentiometrically resolved into several components. Threeb-type and threec-type cytochromes are shown to be present in the membranes ofSt. clavuligerus.  相似文献

1. Purified mitochondria have been prepared from wild type Paramecium tetraurelia and from the mutant Cl1 which lacks cytochrome aa3. Both mitochondrial preparations are characterized by cyanide insensitivity. Their spectral properties and their redox potentials have been studied.2. Difference spectra (dithionite reduced minus oxidized) of mitochondria from wild type P. tetraurelia at 77 K revealed the α peaks of b-type cytochrome(s) at 553 and 557 nm, of c-type cytochrome at 549 nm and a-type cytochrome at 608 nm. Two α peaks at 549 and 545 nm could be distinguished in the isolated cytochrome c at 77 K. After cytochrome c extraction from wild type mitochondria, a new peak at 551 nm was unmasked, probably belonging to cytochrome c1. The a-type cytochrome was characterized by a split Soret band with maxima at 441 and 450 nm. The mitochondria of the mutant Cl1 in exponential phase of growth differed from the wild type mitochondria in that cytochrome aa3 was absent while twice the quantity of cytochrome b was present. In stationary phase, mitochondria of the mutant were characterized by a new absorption peak at 590 nm.3. Cytochrome aa3 was present at a concentration of 0.3 nmol/mg protein in wild type mitochondria and ubiquinone at a concentration of 8 nmol/mg protein both in mitochondria of the wild type and the mutant Cl1. Cytochrome aa3 was more susceptible to heat than cytochromes b and c,c1.4. CO difference spectra at 77 K revealed two different Co-cytochrome complexes. The first, found only in wild type mitochondria, was a typical CO-cytochrome a3 complex characterized by peaks at 596 and 435 nm and troughs at 613 and 450 nm. The second, found both in mitochondria of the wild type and the mutant, was a CO-cytochrome b complex with peaks at 567, 539 and 420 nm and a trough at 558-549 nm. Both complexes are photo-dissociable.5. Spectral evidence was obtained for interaction of cyanide with the a-type cytochrome (shift of the α peak at 77 K from 608 to 605 nm), but not with the b-type cytochrome.6. The mid-point potentials of the different cytochromes at neutral pH are as follows: cytochrome aa3 235 and 395 mV, cytochrome c,c1 233 mV, cytochromes b 120 mV.  相似文献

The role(s) of copper in a bacterial cytochrome oxidase of the aa 3-type was investigated by growth of Paracoccus denitrificans NCIB 8944, in batch and steady state continuous culture, in a medium from which the bulk of the copper had been extracted. In a medium containing approximately 0.02 M copper, cellular copper content, cytochromes a+a 3 and cytochrome a 3 were reduced to 55%, 58% and 33% respectively of control values and there were also less marked decreases in cytochromes c+c 1 (to 85%) and a CO-binding b-type cytochrome, possibly cytochrome o (to 71%). Copper deficiency elicited in reduced minus oxidized difference spectra a shift to shorter wavelengths and narrowing of the band width of the -band of the oxidase, and loss of a (negative) band near 830 nm attributable to CuA (the copper functionally associated with haem a in the oxidase complex). The oxidase in copper-deficient cells reacted with oxygen to form the oxy Compound A at rates similar to that in control cells but CO recombination to ferrous haem a 3 was slowed 4-fold in the copper deficient case. The results are interpreted as indicating loss of CuA and changes in the proportions of haems a and a 3 with retention of catalytic activity. Titrations of respiration rates with antimycin suggested that copper deficiency did not result in diversion of electron flux through an antimycin A-insensitive, cytochrome o-terminated branch of the respiratory chain.  相似文献

Summary The plasma membrane of higher plants contains more than one kind ofb-type cytochromes. One of these has a high redox potential and can be fully reduced by ascorbate. This component, the cytochromeb 561 (cytb 561), has its characteristic -band absorbance close to 561 nm wavelength at room temperature. Cytb 561 was first isolated from etiolated bean hook plasma membranes by two consecutive anion exchange chromatography steps. During the first step performed at pH 8, cytb 561 did not bind to the anion exchange column, but otherb-type cytochromes did. In the second step performed at pH 9.9, cytb 561 was bound to the column and was eluted from the column at an ionic strength of about 100 mM KCl. However, when the same protocol was applied to the solubilized plasma membrane proteins fromArabidopsis thaliana leaves and maize roots, the ascorbate-reducible cytb 561 bound already to the first anion exchange column at pH 8 and was eluted also at an ionic strength of about 100 mM KCl. Otherb-type cytochromes than the ascorbate-reducible cytb 561 from the plasma membranes of Arabidopsis leaves and maize roots showed similar Chromatographic characteristics to that of bean hypocotyls. The cytochromes of the bacteriumStreptomyces erythreus have been investigated. Membrane-bounda-, b-, andc-type cytochromes were found together with a green pigment, which was found in both a soluble and membrane-bound form. Cells containing the green pigment exhibited cyanide-insensitive oxygen uptake. The CO-binding pigments included cytochromea 3, ab-type cytochrome, cytochrome P450, and the green pigment. Photodissociation spectra at various low temperatures, in the presence or absence of oxygen, revealed cytochromeaa 3 to be the predominant cytochrome terminal oxidase. The green pigment was capable of electron transport; the relationship of the pigment to the remainder of the electron transport chain remains to be ascertained.  相似文献

Thiobacillus tepidarius was shown to contain cytochrome(s) c with absorption maxima at 421, 522 and 552 nm in room temperature reduced minus oxidized difference spectra, present at 1.1–1.2 nmol per mg dry wt and present in both membrane and soluble fractions of the cell. The membrane-bound cytochrome c (1.75 nmol per mg membrane protein) had a midpoint potential (Em, pH 7.0) of 337 mV, while the soluble fractions appeared to contain cytochrome(s) c with Em (pH 7.0) values of about 270 and 360 mV. The organism also contained three distinct membrane-bound b-type cytochromes (totalling 0.33 nmol per mg membrane protein), each with absorption maxima in reduced minus oxidized difference spectra at about 428, 532 and 561 nm. The Em (pH 7.0) values for the three cytochromes b were 8 mV (47.8% of total), 182 mV (13.7%) and 322 mV (38.5%). No a- or d-type cytochromes were detectable spectrophotometrically in the intact organism or its membrane and soluble fractions. Evidence is presented for both CO-binding and CO-unreactive cytochromes b or o, and CO-binding cytochrome(s) c. From redox effects observed with CO it is proposed that a cytochrome c donates electrons to a cytochrome b, and that a high potential cytochrome b or o may be acting as the terminal oxidase in substrate oxidation. This may be the 445 nm pigment, a photodissociable CO-binding membrane haemoprotein. Substrate oxidation was relatively insensitive to CO-inhibition, but strongly inhibited by cyanide and azide. Thiosulphate oxidation couples directly to cytochrome c reduction, but tetrathionate oxidation is linked (probably via ubiquinone Q-8) to reduction of a cytochrome b of lower potential than the cytochrome c. The nature of possible electron transport pathways in Thiobacillus tepidarius is discussed. The respiratory electron-transport chain of heterotrophically dark-grown Chloroflexus aurantiacus has been investigated. Membranes isolated from these cells have been shown to contain at least three c-type cytochromes (Em, 7.0 255,180, and 10 mV), three b-type cytochromes (Em, 7.0 of 210, 60 and −65 mV) and two cytochromes of the a type with Em, 7.0 of 330 and 190 mV. Spectroscopic evidence from CO-difference spectra, CN-duference spectra and spectra at fixed oxidation-reduction potentials suggests that the two a-type components may be analogous to cytochromes a and a3 of mitochondria. The analyses of the effects induced by CN, myxothiazol and antimycin A on both steady-state respiratory activities and semi-rapid oxidation-reduction kinetic patterns of c- and a-type cytochromes indicate the presence of a branched respiratory chain. Growth of Chloroflexus in medium lacking added copper diminished the concentration of the a-type cytochromes but not those of cytochromes of the b and c type.  相似文献

Oxidized particles ofA. vinelandii show high-spin ferric signals with an axial and a rhombically distorted component with g-values at 5.94 and 6.24, 5.51, respectively. The signals behave similarly on variation of temperature and/or power and are, assigned to cytochromed. The addition of ligands such as cyanide and carbon monoxide to oxidized particles mainly affects the rhombic component of the signal in the g=6 region. Prolonged, incubation of cyanide with oxidized particles results in the appearance of two new low-spin ferric heme signals at g=2.99 and at g=3.23 which are tentatively assigned to low-spin forms of cyanide-liganded cytochromed. With computer signal-averaging of the EPR spectrum of oxidized particles, the presence of resonances in the g=3–4 region could be demonstrated. These resonances are assigned to cytochromeb 1 (g-values at 3.68, 3.43),c-type cytochromes (g-values at 3.43, 3.25) and cytochromea 1, and possibly a low-spin form of ac-type cytochrome (g-value at 3.03). These EPR results represent, to our knowledge, the only such studies reported on the membrane-boundb 1 andc-type cytochromes of a bacterial respiratory-linked phosphorylating electron-transport chain.  相似文献

The cytochrome composition of membrane vesicles ofLegionella pneumophila has been examined by low temperature (77°K) and room temperature difference spectroscopy, and cytochromes of thec, b, a, andd types have been detected. The presence ofc-type cytochrome was verified by formation of the pyridine ferrohemochromogen. A carbon monoxide-bindingc-type cytochrome was detected in CO-reduced minus reduced difference spectra and may also function in cytochromec reductase activity. Respiratory activities were determined for membrane vesicles, and reduced nicotinamide adenine dinucleotide (NADH) was the most rapidly oxidized substrate (199 nmol per min per mg protein), followed by succinate and malate. Cytochrome oxidase activity was demonstrated using ascorbate andN,N,N,N-tetramethyl-p-phenylenediamine (TMPD) (39 nmol per min per mg of protein). High levels of cyanide (K i =10 mM) inhibited NADH oxidation, while low levels of 2-heptyl-4-hydroxyquinoline-N-oxide (HQNO, 18 and 37 M) inhibited NADH oxidation by nearly 90%. The respiratory chain appeared to be complex and terminated by at least three terminal oxidases. Superoxide dismutase activity, but not catalase activity, was detected in cellular extracts.  相似文献

The respiratory system of chemolithoautotrophically-grown Alcaligenes latus contains a, b, and c type cytochromes. Two cytochrome oxidases were identified by their carbon monoxide difference spectra and their differing sensitivities to cyanide and carbon monoxide. The oxidases were cytochrome o and an a-type cytochrome. Ubiquinone was present in A. latus membranes and could be reduced by H2. Visible region of an absorption spectrum was followed in cells of original strains and of rough mutants ofSaccharomyces cerevisiae andS. cerevisiae var.ellipsoideus. It was found that there are no substantial differences in relative content of cytochromesb andc in aerobically grown rough and smooth yeast forms, in spite of the fact that both forms differ substantially in the metabolic oxygen quotient. If the cytochromes present were not reduced in washed cells by dithionite or by substrate addition, the rough forms exhibited a lower cytochrome b:c ratio than the smooth forms. Under anaerobic conditions of cultivation, the rough forms retained a typical aerobic spectrum, lacking, however, the cytochromea and a3 band; the ratio of cytochromesb andc was changed in favour of cytochromeb (from the original 1.7: 1 up to 3.4: 1). The inability of the rough mutants to produce anaerobic cytochrome spectrum represented by cytochrome b1 was connected with their inability to reproduce under anaerobic conditions.  相似文献

(1) Cells of ThiobacillusA2 grown chemoautotrophically on thiosulfate or heterotrophically on succinate with oxygen contained b-, c-, o-, a- and a3-type cytochromes. The amount of cytochrome per mg of cell protein was much greater in thiosulfate-grown cells and differences in the relative concentrations of cytochromes were observed for the different growth conditions. (2) The half-reduction potentials at pH 7.0 (Em,7.0) and spectral maxima of c-, b-, a- and a3-type cytochromes were similar in cells grown aerobically with thiosulfate or with succinate as the growth substrate. (3) The half-reduction potential of the ‘invisible’, or high-potential copper, as determined from the potentiometric behavior of the carbon monoxide-reduced cytochrome a3 complex at pH 8.0, was 365 mV. (4) Reducing equivalents from thiosulfate appear to enter the respiratory chain at the cytochrome c level; however, studies in cell-free extracts were limited due to a loss in respiratory activity with thiosulfate as a substrate upon cell disruption.  相似文献

Im Entwicklungsabschnitta stieg nur der Sauerstoffverbrauch exponentiell an. Eine-Galaktosidase konnte zu dieser Zeit noch nicht gefunden werden.4. Die mit dem Einsetzen des Herzschlages erfolgende Änderung der Syntheserate der-Galaktosidase wird als hormonale Steuerung der Genaktivität gedeutet.
Metabolism during embryonic and early post-embryonic development
The embryonic development of the snailLymnaea stagnalis may be divided into three main stages: (a) cleavage and gastrulation, (b) larvae and (c) older embryos with beating hearts. During these three stages the following were measured: respiration by means of a Cartesian diver apparatus, embryo dry weight by means of a quartz fiber balance, and activity of-galactosidase (enzymatically with o-Nitrophenyl--D-galactopyranoside as substrate). During stagesb andc a two-phase exponential increase occurs in respiration, in dry weight and in the activity of-galactosidase. Change of phases coincides with the beginning of heart beats. The increase of enzyme activity is explained by assuming a hormonal regulation of the activity of the genes responsible for enzyme synthesis. During stagea only respiration rises exponentially; activity of-galactosidase is not demonstrable.

The homoacetogenic bacteria Sporomusa ovata and Sporomusa sphaeroides were grown on betaine, betaine + formate, and acetoin in the absence of carbon dioxide, and the formation of membrane-bound cytochromes was determined. In S. sphaeroides, the growth substrate had little influence on the expression of cytochromes. In contrast, membranes from betaine-or acetoin-grown S. ovata cells had an 11-or 3-fold higher cytochrome b content than cells grown on betaine + formate. The cytochrome c content was reduced below the detection level after growth on the latter two substrates. The cytochromes in the membranes of S. sphaeroides and S. ovata were characterized by low-temperature difference spectroscopy, hemochrome difference spectroscopy, and redox potentiometry. Membranes of S. ovata were shown to contain two b-type cytochromes with Em,7=-153±10 mV and Em,7=-226±14 mV and two c-type cytochromes with Em,7=-86±6 mV and Em,7=-265±10 mV. In S. sphaeroides also two b-type cytochromes with Em,7=-165±7 mV and Em,7=-241±2 mV and two c-type cytochromes with Em,7=-101±4 mV and Em, 8.5=-338±9 mV could be distinguished. Cell extracts of S. sphaeroides were shown to contain all the enzymes of the acetyl-CoA (Wood) pathway. Summary 1. Nitrate reductase activity in cell-free extracts ofAzotobacter vinelandii was obtained. The enzyme is TPNH-linked and shows some stimulation by molybdenum and FMN.2. The cell-free preparations showed a strong DPNH-oxidase activity and also a slight TPNH oxidation following the addition of distilled water. The latter activity could, however, be removed by ultra-centrifugation of the enzyme extracts. However, nitrate reductase seems to be only to a small extent soluble as its main activity was associated with particles. The particles spun down were red in colour suggesting the presence of cytochromes.3. Thick cell suspensions ofA. vinelandii, A. agile, andA. chroococcum showed similar cytochrome spectra. The max. observed suggest the presence of cytochromes of thec type (max. at 524 and 552 mµ) anda type (max. at 590 and 632 mµ).4. No apparent differences were observed between the cytochrome spectra ofA. vinelandii cells grown on molecular and nitrate nitrogen.  相似文献

Group BNeisseria meningitidis (SDIC) was grown aerobically in Mueller-Hinton medium to stationary phase and then broken either by sonic oscillation or high pressure extrusion from a French pressure cell. The particulate fraction (345,000×g, 3 h) contained cytochromesc, b, o, and evidence of ana-type. The supernatant fraction contained a solublec-type cytochrome (c 549). The pyridine hemochrome derivatives of acid-acetone-extracted supernatant fractions were shown to be free of other cytochrome types. The solublec cytochrome increased during the growth cycle of the organism, reaching a maximum after cells entered stationary phase, whereas the cellular level of other cytochromes from the particulate fraction started to decline as cells progressed from log into stationary phase. Osmotic shock, as well as other treatments, failed to selectively release the solublec cytochrome from the cell. When untreated, thec 549 in the supernatant fraction remained in a reduced form. While it was not air oxidizable, it was readily oxidizable with K3Fe(CN)6. In the reduced form, thec 549 bound either CO or NO, but in the oxidized form, only NO was bound. The results obtained from this study indicate that the solublec 549 is probably best classified as a cytochromec′.  相似文献
