Stand-replacing fires influence soil nitrogen availability and microbial community composition, which may in turn mediate post-fire successional dynamics and nutrient cycling. However, fires create patchiness at both local and landscape scales and do not result in consistent patterns of ecological dynamics. The objectives of this study were to (1) quantify the spatial structure of microbial communities in forest stands recently affected by stand-replacing fire and (2) determine whether microbial variables aid predictions of in situ net nitrogen mineralization rates in recently burned stands. The study was conducted in lodgepole pine (Pinus contorta var. latifolia) and Engelmann spruce/subalpine fir (Picea engelmannii/Abies lasiocarpa) forest stands that burned during summer 2000 in Greater Yellowstone (Wyoming, USA). Using a fully probabilistic spatial process model and Bayesian kriging, the spatial structure of microbial lipid abundance and fungi-to-bacteria ratios were found to be spatially structured within plots two years following fire (for most plots, autocorrelation range varied from 1.5 to 10.5 m). Congruence of spatial patterns among microbial variables, in situ net N mineralization, and cover variables was evident. Stepwise regression resulted in significant models of in situ net N mineralization and included variables describing fungal and bacterial abundance, although explained variance was low (R2<0.29). Unraveling complex spatial patterns of nutrient cycling and the biotic factors that regulate it remains challenging but is critical for explaining post-fire ecosystem function, especially in Greater Yellowstone, which is projected to experience increased fire frequencies by mid 21st Century.  相似文献

Soil nitrogen (N) mineralization in wetlands is sensitive to various environmental factors. To compare the effects of salinity and temperature on N mineralization, wetland soils from a tidal freshwater marsh locating in the Yellow River Delta was incubated over a 48-d anaerobic incubation period under four salinity concentrations (0, 10, 20 and 35‰) and four temperature levels (10, 20, 30 and 40°C). The results suggested that accumulated ammonium nitrogen (NH4+-N) increased with increasing incubation time under all salinity concentrations. Higher temperatures and salinities significantly enhanced soil N mineralization except for a short-term (≈10 days) inhibiting effect found under 35‰ salinity. The incubation time, temperature, salinity and their interactions exhibited significant effects on N mineralization (P<0.001) except the interactive effect of salinity and temperature (P>0.05), while temperature exhibited the greatest effect (P<0.001). Meanwhile, N mineralization processes were simulated using both an effective accumulated temperature model and a one-pool model. Both models fit well with the simulation of soil N mineralization process in the coastal freshwater wetlands under a range of 30 to 40°C (R2 = 0.88–0.99, P<0.01). Our results indicated that an enhanced NH4+-N release with increasing temperature and salinity deriving from the projected global warming could have profound effects on nutrient cycling in coastal wetland ecosystems.  相似文献

Understanding the interactions of plant-parasitic nematodes with antagonistic soil microbes could provide opportunities for novel crop protection strategies. Three arable soils were investigated for their suppressiveness against the root knot nematode Meloidogyne hapla. For all three soils, M. hapla developed significantly fewer galls, egg masses, and eggs on tomato plants in unsterilized than in sterilized infested soil. Egg numbers were reduced by up to 93%. This suggested suppression by soil microbial communities. The soils significantly differed in the composition of microbial communities and in the suppressiveness to M. hapla. To identify microorganisms interacting with M. hapla in soil, second-stage juveniles (J2) baited in the test soil were cultivation independently analyzed for attached microbes. PCR-denaturing gradient gel electrophoresis of fungal ITS or 16S rRNA genes of bacteria and bacterial groups from nematode and soil samples was performed, and DNA sequences from J2-associated bands were determined. The fingerprints showed many species that were abundant on J2 but not in the surrounding soil, especially in fungal profiles. Fungi associated with J2 from all three soils were related to the genera Davidiella and Rhizophydium, while the genera Eurotium, Ganoderma, and Cylindrocarpon were specific for the most suppressive soil. Among the 20 highly abundant operational taxonomic units of bacteria specific for J2 in suppressive soil, six were closely related to infectious species such as Shigella spp., whereas the most abundant were Malikia spinosa and Rothia amarae, as determined by 16S rRNA amplicon pyrosequencing. In conclusion, a diverse microflora specifically adhered to J2 of M. hapla in soil and presumably affected female fecundity.  相似文献

Conceptual models of river–floodplain systems and biogeochemical theory predict that floodplain soil nitrogen (N) and phosphorus (P) mineralization should increase with hydrologic connectivity to the river and thus increase with distance downstream (longitudinal dimension) and in lower geomorphic units within the floodplain (lateral dimension). We measured rates of in situ soil net ammonification, nitrification, N, and P mineralization using monthly incubations of modified resin cores for a year in the forested floodplain wetlands of Difficult Run, a fifth order urban Piedmont river in Virginia, USA. Mineralization rates were then related to potentially controlling ecosystem attributes associated with hydrologic connectivity, soil characteristics, and vegetative inputs. Ammonification and P mineralization were greatest in the wet backswamps, nitrification was greatest in the dry levees, and net N mineralization was greatest in the intermediately wet toe-slopes. Nitrification also was greater in the headwater sites than downstream sites, whereas ammonification was greater in downstream sites. Annual net N mineralization increased with spatial gradients of greater ammonium loading to the soil surface associated with flooding, soil organic and nutrient content, and herbaceous nutrient inputs. Annual net P mineralization was associated negatively with soil pH and coarser soil texture, and positively with ammonium and phosphate loading to the soil surface associated with flooding. Within an intensively sampled low elevation flowpath at one site, sediment deposition during individual incubations stimulated mineralization of N and P. However, the amount of N and P mineralized in soil was substantially less than the amount deposited with sedimentation. In summary, greater inputs of nutrients and water and storage of soil nutrients along gradients of river–floodplain hydrologic connectivity increased floodplain soil nutrient mineralization rates.  相似文献

Understanding the chronological changes in soil microbial and biochemical properties of tea orchard ecosystems after wasteland has been reclaimed is important from ecological, environmental, and management perspectives. In this study, we determined microbial biomass, net N mineralization, and nitrification, enzyme (invertase, urease, proteinase, and acid phosphatase) activities, microbial community diversity assessed by denaturing gradient gel electrophoresis (DGGE) of 16S rDNA polymerase chain reaction (PCR) products, and related ecological factors in three tea orchard systems (8-, 50-, and 90-year-old tea orchards), adjacent wasteland and 90-year-old forest. Soil microbial biomass C (Cmic) and activity, i.e., soil basal respiration (Rmic), microbial biomass C as a percent of soil organic C (Cmic/Corg), N mineralization, invertase, urease, proteinase, and acid phosphatase, significantly increased after wasteland was reclaimed; however, with the succeeding development of tea orchard ecosystems, a decreasing trend from the 50- to 90-year-old tea orchard became apparent. Soil net nitrification showed an increasing trend from the 8- to 50-year-old tea orchard and then a decreasing trend from the 50- to 90-year-old tea orchard, and was significantly higher in the tea orchards compared to the wasteland and forest. Urea application significantly stimulated soil net nitrification, indicating nitrogen fertilizer application may be an important factor leading to high-nitrification rates in tea orchard soils. The Shannon’s diversity index (H) and richness (S) based on DGGE profiles of 16S rRNA genes were obviously lower in all three tea orchards than those in the wasteland; nevertheless, they were significantly higher in all three tea orchards than those in the forest. As for the three tea orchard soils, comparatively higher community diversity was found in the 50-year-old tea orchard.  相似文献

Methyl bromide (MB) and other alternatives were evaluated for suppression of Fusarium spp., Phytophthora spp., and Meloidogyne spp. and their influence on soil microbial communities. Both Fusarium spp. and Phytophthora spp. were significantly reduced by the MB (30.74 mg kg-1), methyl iodide (MI: 45.58 mg kg-1), metham sodium (MS: 53.92 mg kg-1) treatments. MS exhibited comparable effectiveness to MB in controlling Meloidogyne spp. and total nematodes, followed by MI at the tested rate. By contrast, sulfuryl fluoride (SF: 33.04 mg kg-1) and chloroform (CF: 23.68 mg kg-1) showed low efficacy in controlling Fusarium spp., Phytophthora spp., and Meloidogyne spp. MB, MI and MS significantly lowered the abundance of different microbial populations and microbial biomass in soil, whereas SF and CF had limited influence on them compared with the control. Diversity indices in Biolog studies decreased in response to fumigation, but no significant difference was found among treatments in PLFA studies. Principal component and cluster analyses of Biolog and PLFA data sets revealed that MB and MI treatments greatly influenced the soil microbial community functional and structural diversity compared with SF treatment. These results suggest that fumigants with high effectiveness in suppressing soil-borne disease could significantly influence soil microbial community.  相似文献

Increased N inputs through chronic atmospheric deposition has enriched temperate forest ecosystems, altering critical ecosystem functions such as decomposition and potentially resulting in a shift to P limitation. We used a combination of microbial biomass stoichiometry and enzymatic activity analyses to evaluate the potential for microbial nutrient limitation over the course of a growing season in response to multi-decadal, whole-watershed N enrichments and a one time, plot-scale P addition that occurred in the 22nd year of whole-watershed treatments. The one-time P addition increased microbial biomass threefold and reduced N-acetyl-glucosaminidase (NAG) and acid phosphatase (AP) activity 1 week after application, but there was no interaction with long-term experimental N enrichment to indicate a shift to P limitation. However, both N and P treatments increased C limitation independently of each other over the duration of the study based on measured increases in β-1,4-glucosidase (BG) activity relative to NAG and AP. Microbial biomass stoichiometry and enzyme activity indicated that BBWM is P limited regardless of N status. Our findings highlight the complex interactions between C, N, and P use and limitation in a forested ecosystem subjected to long-term N enrichment.  相似文献

This research addressed the effect of mineral and organic soil constituents on the fate of organic compounds in soils. Specifically, it sought to determine how the associations between organic chemicals and different soil constituents affect their subsequent biodegradation in soil. Four 14C-labeled surfactants were aseptically adsorbed to montmorillonite, kaolinite, illite, sand, and humic acids. These complexes were mixed with a woodlot soil, and 14CO2 production was measured over time. The mineralization data were fitted to various production models by nonlinear regression, and a mixed (3/2)-order model was found to most accurately describe the mineralization patterns. Different mineralization patterns were observed as a function of the chemical and soil constituents. Surfactants that had been preadsorbed to sand or kaolinite usually showed similar mineralization kinetics to the control treatments, in which the surfactants were added to the soil as an aqueous solution. Surfactants that had been bound to illite or montmorillonite were typically degraded to lesser extents than the other forms, while surfactant-humic acid complexes were degraded more slowly than the other forms. The desorption coefficients (Kd) of the soil constituent-bound surfactants were negatively correlated with the initial rates of degradation (k1) and estimates of 14CO2 yield (Po) as well as actual total yields of 14CO2. However, there was no relationship between Kd and second-stage zero-order rates of mineralization (ko). Microbial community characteristics (biomass and activity) were not correlated with any of the mineralization kinetic parameters. Overall, this study showed that environmental form had a profound effect on the ultimate fate of biodegradable chemicals in soil. This form is defined by the physicochemical characteristics of the chemical, the composition and mineralogy of the soil, and the mode of entry of the chemical into the soil environment.  相似文献

Abstract Forest soil ecology was studied in Fennoscandinavian dry Scots pine forests grazed by reindeer to varying extents (ungrazed, lichen-dominated-sites; grazed sites; and bryophyte-dominated sites). We hypothesized that the productivity parameters of the site (i.e., tree growth and soil nutrient concentrations), the vegetation composition, and the microbial activities are directly correlated. Since the productivity of the lichen-dominated ecosystem is low, microbial activities are assumed to be naturally low. Grazing was expected to decrease both the amount of Scots pine fine roots and the soil microbial activities. Several variables on the characteristics of the soil microbial community, Scots pine fine roots, soil nutrients, and tree growth were studied in relation to vegetation composition by using non-metric multidimensional scaling (NMDS). Basal respiration (Bas), metabolic quotient of the microbial community (qCO2), and pine fine root parameters increased toward the ungrazed, nutrient-poor, lichen-dominated sites, which were grouped at one end of the first axis in the NMDS ordination. Soil nutrient and tree growth parameters and thickness of the humus layer increased toward bryophyte-dominated sites, which were grouped at the other end of the first axis in the ordination. The grazed sites fell between them. These were characterized by lower Bas and qCO2 values and longer lag, compared to ungrazed lichen- or bryophyte-dominated sites, probably due to decreased carbon input and microclimatic change (the soil without lichen carpet is exposed to direct sunlight and wind). Microbial biomass (Cmic), fungal biomass (ergosterol concentration), and the specific growth rate (μCO2) were not related to vegetation ordination. The high fine root production is the most plausible explanation for the high microbial activities at nutrient-poor, lichen-dominated sites, which produce qualitatively poor and slowly decomposing litter, as fine roots secrete considerable amounts of organic substances. At bryophyte-dominated sites, the higher soil nutrient concentrations and the higher production of easily decomposable substrates are likely to maintain the microbial activities.  相似文献

Laboratory tests were conducted with four organophosphorus insecticides, Bay 37289 (O-ethyl O-2,4,5-trichlorophenyl ethylphosphonothioate), diazinon [O,O-diethyl O-(2-isopropyl-4-methyl-6-pyrimidinyl) phosphorothioate], Dursban (O,O-diethyl O-3,5,6-trichloro-2-pyridyl phosphorothioate), and Zinophos (O,O-diethyl O-2-pyrazinyl phosphorothioate), applied to a sandy loam at rates of 10 and 100 mug/g to determine whether these materials caused any serious effects on microbial activities related to soil fertility. All insecticides showed an effect on fungi and bacteria for the first and second week of incubation, but, subsequently, the populations returned to levels similar to those obtained in the controls. All insecticide applications increased ammonium production, but, in some instances, there appeared to be a slight depression of nitrification. Sulfur oxidation was equal to or better than that obtained with untreated soil in most cases. There was no significant effect on phosphorus mineralization. Oxygen consumption indicated that microbial respiration increased in proportion to the concentration of insecticides, suggesting the possibilities of microbial degradation of the insecticides or their degradation products and of uncoupling oxidative phosphorylation.  相似文献

Changes in the quantity and/or quality of soil labile organic matter between and after different types of cultivation system could play a dominant role in soil nitrogen (N) mineralization. The quantity and quality of particulate organic matter (POM) and potentially mineralizable-N (PMN) contents were measured in soils from 16 paired rice-rapeseed (RR)/cotton-rapeseed (CR) rotations sites in Hubei province, central China. Then four paired soils encompassing low (10th percentile), intermediate (25th and 75th percentiles), and high (90th percentile) levels of soil PMN were selected to further study the effects of POM on soil N mineralization by quantifying the net N mineralization in original soils and soils from which POM was removed. Both soil POM carbon (POM-C) and N (POM-N) contents were 45.8% and 55.8% higher under the RR rotation compared to the CR rotation, respectively. The PMN contents were highly correlated with the POM contents. The PMN and microbial biomass N (MBN) contents concurrently and significantly decreased when POM was removed. The reduction rate of PMN was positively correlated with changes in MBN after the removal of POM. The reduction rates of PMN and MBN after POM removal are lower under RR rotations (38.0% and 16.3%, respectively) than CR rotations (45.6% and 19.5%, respectively). Furthermore, infrared spectroscopy indicated that compounds with low-bioavailability accumulated (e.g., aromatic recalcitrant materials) in the soil POM fraction under the RR rotation but not under the CR rotation. The results of the present study demonstrated that POM plays a vital role in soil N mineralization under different rotation systems. The discrepancy between POM content and composition resulting from different crop rotation systems caused differences in N mineralization in soils.  相似文献

Soil properties, microbial communities, and enzyme activities were studied in soil planted with transgenic or nontransgenic papaya under field conditions. The transgenic papaya contained a replicase (RP) mutant gene of the papaya ringspot virus (PRSV), which conferred resistance to the virus, the neomycin phosphotransferase II (NPT II) marker gene, which conferred Km resistance, and a cauliflower mosaic virus 35S promoter (CaMV 35S). There were significant differences (P < 0.05) in the total number of colony forming units (CFUs) of bacteria, actinomycetes, and fungi between soils planted with RP-transgenic and nontransgenic plants; total CFUs of bacteria, actinomycetes, and fungi in soil planted with transgenic papaya were significantly higher by 0.43, 0.80, and 0.46 times, respectively. Significantly higher (P < 0.05) CFUs of bacteria, actinomycetes, and fungi resistant to kanamycin (Km) were present in soils planted with the transgenic papaya than in those planted with nontransgenic papaya. Resistance quotients (CFU in the presence of a chemical relative to that without) of Km-resistant bacteria, actinomycetes and fungi were higher in soil planted with transgenic papaya, and the resistance quotients of Km-resistant bacteria, actinomycetes, and fungi in soils planted with transgenic papaya increased statistically significantly (P<0.05) from 1.5 to 2.5, from 1.2 to 2.6, and from 0.9 to 2.8 times, respectively. Soils planted with transgenic papaya had significantly higher enzyme activities of arylsulfatases (+5.4 times), alkaline phosphatases (+0.5 time), invertase (+0.5 time) and phosphodiesterases (+0.2 time), but lower enzyme activities of proteases (−2.1 times), polyphenol oxidases (−1.4 times), urease (−0.2 time) than the soils planted with nontransgenic papaya. Our results suggest that transgenic papaya could alter chemical properties, enzyme activities, and microbial communities in soil.  相似文献

We used long-term laboratory incubations and chemical fractionation to characterize the mineralization dynamics of organic soils from tussock, shrub, and wet meadow tundra communities, to determine the relationship between soil organic matter (SOM) decomposition and chemistry, and to quantify the relative proportions of carbon (C) and nitrogen (N) in tundra SOM that are biologically available for decomposition. In all soils but shrub, we found little decline in respiration rates over 1 year, although soils respired approximately a tenth to a third of total soil C. The lack of decline in respiration rates despite large C losses indicates that the quantity of organic matter available was not controlling respiration and thus suggests that something else was limiting microbial activity. To determine the nature of the respired C, we analyzed soil chemistry before and after the incubation using a peat fractionation scheme. Despite the large losses of soil C, SOM chemistry was relatively unchanged after the incubation. The decomposition dynamics we observed suggest that tundra SOM, which is largely plant detritus, fits within existing concepts of the litter decay continuum. The lack of changes in organic matter chemistry indicates that this material had already decomposed to the point where the breakdown of labile constituents was tied to lignin decomposition. N mineralization was correlated with C mineralization in our study, but shrub soil mineralized more and tussock soil less N than would have been predicted by this correlation. Our results suggest that a large proportion of tundra SOM is potentially mineralizable, despite the fact that decomposition was dependent on lignin breakdown, and that the historical accumulation of organic matter in tundra soils is the result of field conditions unfavorable to decomposition and not the result of fundamental chemical limitations to decomposition. Our study also suggests that the anticipated increases in shrub dominance may substantially alter the dynamics of SOM decomposition in the tundra. There has been much work on plant-feeding nematodes, and less on other soil nematodes, their distribution, abundance, intrinsic properties, and interactions with biotic and abiotic factors. Seasonal variation in nematode fauna as a whole is correlated with factors such as moisture, temperature, and plant growth; at each site nematode distribution generally reflects root distribution. There is a positive correlation between average nematode abundance and primary production as controlled by moisture, temperature, nutrients, etc. Soil nematodes, whether bacterial feeders, fungivores, plant feeders, omnivores, or predators, all influence the populations of the organisms they feed on. Although soil trematodes probably contribute less than 1% to soil respiration they may play an important role in nutrient cycling in the soil through their influence on bacterial growth and plant nutrient availability.  相似文献

Observations of atypical developmental and anatomical characteristics have been recorded for many taxa of soil nematodes. They include the unusual occurrence of extra feeding structures, aberrant configuration of features of both male and female reproductive systems, and the occurrence of intersexes assumed to be functionally female, functionally male, or non-functional. In many cases, hypotheses have been advanced regarding the genetic or developmental mechanisms and environmental stimuli that control, regulate, or facilitate abnormalities, but many are quite speculative and lack experimental verification. Further, the fitness costs or advantages, and the heritability of aberrant characters are largely unknown, except where they clearly preclude reproduction, either apomictic or amphimictic. Underlying mechanisms and ecological consequences may be difficult to study in organisms that are not readily cultured under axenic or sterile laboratory conditions, however information on developmental processes in Caenorhabditis elegans represents an important resource in which to seek homologies.  相似文献

采用氯仿熏蒸-浸提法,以宁夏盐池荒漠草原3种典型群落(柠条、沙蒿、短花针茅)类型为研究对象,分析了不同生境(冠下、丛间)和不同土层间(0~5、5~10、10~15cm)土壤理化性质及微生物量——微生物量碳(MBC)和微生物量氮(MBN)的变化特征。结果表明:(1)3种群落土壤微生物量变化差异较大,柠条、沙蒿和短花针茅群落土壤MBC含量分别为77.00~393.18、17.27~221.71和81.05~173.37mg/kg,MBN含量分别为7.59~64.81、1.43~13.95和4.25~22.13mg/kg,MBC和MBN含量均表现为:冠下丛间,且随土层深度的增加而降低,有明显的"沃岛效应"。(2)群落类型对土壤微生物量碳氮含量的变化有显著影响,3种典型群落类型下土壤微生物量熵(qMB)、碳氮比(C/N)、微生物量碳氮比(MBC/MBN)分别在0.76~4.10、15.02~52.50、5.34~23.07范围内变化,其比值在不同生境和不同土层深度的分布特征有明显差异。(3)3种典型群落类型的土壤MBC与SOC、MBN、qMB具有显著相关关系,土壤C/N与MBC/MBN呈显著正相关关系,表明土壤MBC、MBN具有一定的生物学指示特性,可以作为评价土壤质量的生物学指标。  相似文献

Nematodes of three genera (Acrobeloides sp., Aphelenchus avenae, and Scutellonema brachyurum) were induced to coil and enter anhydrobiosis in drying soil of two types: sandy loam and loamy sand. Coiling was studied in relationship to soil moisture characteristics. Coiling and the physiological state of anhydrobiosis occurred before the water in sandy soils reached a water potential of -15 bars. Coiling was maximum at 3-6 bars, depending on the soil type and nematode species. It appeared that induction of coiling and anhydrohiosis were determined by the physical forces exerted by the water film surrounding the nematode, which, for these three species, was 6-9 monomolecular layers of water, rather than the % moisture and relative humidity of the soil per se.  相似文献

Elevated nitrogen (N) deposition in humid tropical regions may exacerbate phosphorus (P) deficiency in forests on highly weathered soils. However, it is not clear how P availability affects soil microbes and soil carbon (C), or how P processes interact with N deposition in tropical forests. We examined the effects of N and P additions on soil microbes and soil C pools in a N-saturated old-growth tropical forest in southern China to test the hypotheses that (1) N and P addition will have opposing effects on soil microbial biomass and activity, (2) N and P addition will alter the composition of the microbial community, (3) the addition of N and P will have interactive effects on soil microbes and (4) addition-mediated changes in microbial communities would feed back on soil C pools. Phospholipid fatty acid (PLFA) analysis was used to quantify the soil microbial community following four treatments: Control, N addition (15 g N m−2 yr−1), P addition (15 g P m−2 yr−1), and N&P addition (15 g N m−2 yr−1 plus 15 g P m−2 yr−1). These were applied from 2007 to 2011. Whereas additions of P increased soil microbial biomass, additions of N reduced soil microbial biomass. These effects, however, were transient, disappearing over longer periods. Moreover, N additions significantly increased relative abundance of fungal PLFAs and P additions significantly increased relative abundance of arbuscular mycorrhizal (AM) fungi PLFAs. Nitrogen addition had a negative effect on light fraction C, but no effect on heavy fraction C and total soil C. In contrast, P addition significantly decreased both light fraction C and total soil C. However, there were no interactions between N addition and P addition on soil microbes. Our results suggest that these nutrients are not co-limiting, and that P rather than N is limiting in this tropical forest.  相似文献
