It is well known that algal growth rates decrease with increasing cell size. Most of these findings were, however, obtained under laboratory conditions. It is not clear if these allometric relationships are also applicable to in situ conditions. In the present study, the relationship between growth rates and cell size of algal species was examined seasonally in Lake Biwa by in situ dilution bioassays. The bioassays revealed that the highest growth rate of each species throughout the experiments was negatively correlated with cell size consistent with known allometric relationships. At each incubation experiment, however, growth rates were not necessarily correlated with cell size. This was true even when both macro- and micronutrients were added, although a substantial number of species responded to nutrient enrichment. These results showed that nutrient supplies affected algal species differently regardless of cell size and that factors other than nutrient supplies limited the growth rate of some algal species. Due to such species-specific differences in limiting factors, at any given time in situ growth rates of algae are not determined exclusively by cell size. Main effects of growth factors (GFs) on proliferation seem to be mediated by second messengers (SMs). A variety of feed-backs in the SM system allows to suggest that cell cycling is determined by self-oscillations of levels of the SMs and SM-dependent proteins. GFs perform roles of parameter switches of the system functioning regimes; self-oscillations occur only if the GF levels are within certain range. This approach makes it possible to explain the GF synergy, dual roles of GFs as activators and inhibitors of proliferation, and cell cycle duration variability. It predicts that GFs must accelerate some cell cycle phases and decelerate other phase. When the GF levels are sharply changed, cells may become terminally differentiated, which allows to explain a limited number of cell divisions in culture. We can also explain why some oncogenes decrease the dependence of cells on GFs but cells undergo a limited number of divisions, and other oncogenes enable cells to divide unlimitedly.  相似文献

Summary Tomato juice contains a growth factor (TJF) active for some strains of Leuconostoc oenos but not required by other lactic acid bacteria. TJF does not appear to be identical with any known growth factor, is of limited distribution in natural products and is destroyed by the growth of most strains of Leuconostoc oenos and by Lactobacillus plantarum and Pediococcus cerevisiae.TJF is essential for the growth of Leuconostoc oenos NCDO 1674 when organic acids are in a medium with an initial pH of 4.8 and incubated at 22° C and also with an initial pH of 6.0 and incubated at 30° C. Under the first conditions only and in the absence of organic acids Tween 80 is essential for growth if tomato juice is absent.  相似文献

Many biological systems can be expected to exhibit resonance behavior involving the mechanical vibration of system elements. The natural frequencies of such resonances will, generally, be in the microwave frequency range. Some of these systems will be coupled to the electromagnetic field by the charge distributions they carry, thus admitting the possibility that microwave exposures may generate physiological effects in man and other species. However, such microwave excitable resonances are expected to be strongly damped by interaction with their aqueous biological environment. Although those dissipation mechanisms have been studied, the limitations on energy transfers that follow from the limited coupling of these resonances to the electromagnetic field have not generally been considered. We show that this coupling must generally be very small and thus the absorbed energy is so strongly limited that such resonances cannot affect biology significantly even if the systems are much less strongly damped than expected from basic dissipation models.  相似文献

1. Climate change and other human-driven environmental perturbations are causing reductions in biodiversity and impacting the functioning of ecosystems on a global scale. Metacommunity theory suggests that ecosystem connectivity may reduce the magnitude of these impacts if the regional species pool contains functionally redundant species that differ in their environmental tolerances. Dispersal may increase the resistance of local ecosystems to environmental stress by providing regional species with traits adapted to novel conditions. 2. We tested this theory by subjecting freshwater zooplankton communities in mesocosms that were either connected to or isolated from the larger regional species pool to a factorial manipulation of experimental warming and increased salinity. 3. Compensation by regional taxa depended on the source of stress. Warming tolerant regional taxa partially compensated for reductions in heat sensitive local taxa but similar compensation did not occur under increased salinity. 4. Dispersal-mediated species invasions dampened the effects of warming on summer net ecosystem productivity. However, this buffering effect did not occur in the fall or for periphyton growth, the only other ecosystem function affected by the stress treatments. 5. The results indicate that regional biodiversity can provide insurance in a dynamic environment but that the buffering capacity is limited to some ecosystem processes and sources of stress. Maintaining regional biodiversity and habitat connectivity may therefore provide some limited insurance for local ecosystems in changing environments, but is unable to impart resistance against all sources of environmental stress.  相似文献

POPULATION ECOLOGY OF SEALS: RETROSPECTIVE and PROSPECTIVE VIEWS   总被引:1,自引:0,他引:1  
This review focuses on population ecology, with critical accounts of past work and future possibilities in age determination, body growth and condition, estimating abundances, mortality rates and lifespans, reproduction, comparative life histories, population dynamics, population modelling and seals in ecosystems. We suggest ways to reduce errors in age determination and to improve methods of obtaining and presenting growth data. Generalized von Bertalanffy growth equations are promoted as a basis for analysing species differences and intra-population variation in body lengths. Indices other than blubber thickness may be better for following body condition. Catch-effort and survival-index methods of estimating abundances have limited applicability, total counts are only locally useful, and sample counts may only be accurate for scattered, ice-breeding species. Some new techniques for population indices are promising. Pre-adult mortality remains difficult to assess. Although not always recognized, adult mortality rates do increase with age, as well described by Gompertz functions. Existing estimates of lifespans are unreliable, and a new approach is outlined. There are methodological problems in estimating ages of maturity. Corpora albicantia should not be used for back-extrapolation, and more study is needed of use of teeth annuli as indicators of maturity. Age-specific proportions of females parous based on reproductive tracts may disagree with proportions recruited in breeding groups, suggesting that the former may often be in error. Allometric relationships among body sizes and life-history variables need more reliable data, especially since the residuals of such relationships are of greatest interest. Brain size may be a better scalar. Direct evidence of density dependence in population growth of seals is sparse. Early survival has been more widely shown to be density-dependent, but only among polygynous species where crowding on land may be a byproduct of sexual selection; there is as yet no good evidence of trophic restraints. Evidence of density dependence of ages of maturity is generally unconvincing. Predation, especially by sharks, may be critical in some species. Characteristics of equilibrium populations might profitably be sought in mass remains in middens and historic kill sites. More attention should be paid to the search for density-independent influences. Supposed impacts of fisheries and pollutions are not wholly convincing. Natural epidemics may keep some populations below resource or space saturation, and some high-latitude species may show large year-to-year variations in recruitment and abundances. Evidence for such density-independent effects should be sought in residuals of growth curves and in teeth layers. Although surplus yield and production/biomass models have been tried, realistic pinniped models must be completely age-structured and time-dependent. Simple models have questionably assumed stationarity to derive life-history parameters. The best available estimates of density dependence of such parameters give no resolution when extrapolated toward equilibrium, and only limited efforts have been made to introduce stochasticity. Better data, not improved model structures, are needed for better understanding. Recent work has contradicted the assumed voraciousness of seals, but their system impacts and dependencies are not well understood. Extended Lotka-Volterra equations used to model Antarctic food webs, including seals, are merely heuristic. Fixed seal biomasses enter as top-down, driving functions in a Bering Sea model, which accordingly cannot be used to analyse or manage their populations. Some Soviet models are tantalizing but ill-specified. The introduction of harbor seals in well-chosen lakes might give mote insights into system roles than would more elaborate modelling. We wonder if pinniped ecology is well served by too many enthusiasts operating under too many restraints.  相似文献

On line control coupled with an expert system was constructed for the control of a fed-batch culture with the aim of achieving a high cell density. During the cultivation, the expert system could monitor the extents of sufficiencies in the amounts of chemical elements in the medium every 10 min, and suggest a modification to the feeding control policy if the amount of a particular component was inadequate for cell growth. However, we often encounter such a kind of cultivations in which some particular carbon source such as glucose and ethanol should be controlled at low concentration in many culture processes, because the excess feeding might cause the growth inhibition by its own accumulation in the culture broth, or the biproduct accumulation like as lactate or acetate, unsuitable substances for smoothed growth. In this study, we developed an online control system based on production rules which managed the glucose feed rate from DO signal. The online control was carried out by the control computer connected with the other computer for expert system, because a relatively long time (several minutes) was needed for the inference of the expert system and the influence of the starvation of carbon source on the cell growth is not negligible even in several minutes. The on-line control system with expert system was applied to the production of cell mass of E. coli W3110 and the final cell concentration reached 137 g-dry cell weight/l.  相似文献

Tuberculosis (TB), which is caused by Mycobacterium tuberculosis, remains one of the most prevalent infectious diseases worldwide which causes high morbidity and mortality. However, there is still limited understanding of the physiological processes that allow M. tuberculosis to survive in its host environment. One of the challenges is the limited availability of molecular probes that can be used to study some of the complex systems in mycobacteria. One such system is the ESX-3 secretion system, a specialized type VII secretion (T7S) system. This system is essential for optimal growth of pathogenic mycobacteria in low iron environments similar to that encountered by mycobacteria in macrophages during infection. EsxG, a protein of unknown function, is both encoded within the ESX-3 locus and secreted by the ESX-3 system.  相似文献

Summary Techniques available for the calculation of the time variation of the number of viable mammaliar cells in a cell population are reviewed. Events in the course of the cell's growth may include one or more exposures to ionizing radiations or other cytotoxic agents. The dependence of cell killing upon the cell's position in the cell cycle is emphasized, and a unified model for calculation of cell kinetics and cell survival is discussed. For a cell population not limited in growth by contact inhibition or by nutritional factors, experimental data agree with predictions of the model.The possibility of utilizing the model to arrive at optimum treatment schedules for the management of some malignant diseases is discussed. The conclusion drawn is that the state of knowledge with respect to cellular events in solid tumors is such as to leave most such applications in the realm of speculation.This work was supported by the National Institutes of Health, United States Public Health Service, under Grants CA 5008 and CA 4542.  相似文献

It has been recently suggested that there are likely generic features characterizing the emergence of systems constructed from the self-organization of self-replicating agents acting under one or more selection pressures. Therefore, structures and behaviors at one length scale may be used to infer analogous structures and behaviors at other length scales. Motivated by this suggestion, we seek to characterize various "animate" behaviors in biochemical networks, and the influence that these behaviors have on genomic evolution. Specifically, in this paper, we develop a simple, chemostat-based model illustrating how a process analogous to associative learning can occur in a biochemical network. Associative learning is a form of learning whereby a system "learns" to associate two stimuli with one another. Associative learning, also known as conditioning, is believed to be a powerful learning process at work in the brain (associative learning is essentially "learning by analogy"). In our model, two types of replicating molecules, denoted as A and B, are present in some initial concentration in the chemostat. Molecules A and B are stimulated to replicate by some growth factors, denoted as G(A) and G(B), respectively. It is also assumed that A and B can covalently link, and that the conjugated molecule can be stimulated by either the G(A) or G(B) growth factors (and can be degraded). We show that, if the chemostat is stimulated by both growth factors for a certain time, followed by a time gap during which the chemostat is not stimulated at all, and if the chemostat is then stimulated again by only one of the growth factors, then there will be a transient increase in the number of molecules activated by the other growth factor. Therefore, the chemostat bears the imprint of earlier, simultaneous stimulation with both growth factors, which is indicative of associative learning. It is interesting to note that the dynamics of our model is consistent with certain aspects of Pavlov's original series of conditioning experiments in dogs. We discuss how associative learning can potentially be performed in vitro within RNA, DNA, or peptide networks. We also describe how such a mechanism could be involved in genomic evolution, and suggest relevant bioinformatics studies that could potentially resolve these issues.  相似文献

Many models of honest signaling, based on Zahavi's handicap principle, predict that if receivers are interested in a quality that shows continuous variation across the population of signalers, then the distribution of signal intensities will also be continuous. However, it has previously been noted that this prediction does not agree with empirical observation in many signaling systems, where signals are limited to a small number of levels despite continuous variation in the trait being signaled. Typically, there is a critical value of the trait, with all individuals with trait values on one side of the threshold using the same cheap signal, and all those with trait values on the other side of the threshold using the same expensive signal. It has already been demonstrated that these classical models naturally predict such "all-or-nothing signaling" if it is additionally assumed that receivers suffer from perceptual error in evaluating signal strength. We show that such all-or-nothing signaling is also predicted if receivers are limited to responding to the signals in one of two ways. We suggest that many ecological situations (such as the decision to attack the signaler or not, or mate with the signaler or not) involve such binary choices.  相似文献

An experimental testing of material from thin-layered, transparent in passing light, colonies which appear with some frequency after plating Bacillus subtilis cells on agar medium with limited enrichment, has shown that such colonies are formed by auxotrophic mutants. The growth requirements for many of them has been identified. The most of mutants can be reversed to original phenotype by UV-irradiation. The frequency of auxotrophs increases after UV-irradiation of suspension of original cells. The sensitivity of auxotrophic mutants to inactivating action of UV-light is near to that of original cells, hence the increase of the frequency of mutants with dose is a result of induction, but not of selection of preexisting spontaneous auxotrophic mutants. The frequency of induced auxotrophs, in contrast to that of suppressor revertants, badly give way to declining in the time of temporary inhibition of postradiation growth. In the case of Bac, subtilis, the system of induced auxotrophic mutants on the medium with limited enrichment is rather comfortable in use and can be recommended for studying UV-induced mutagenesis in structural genes as well as for testing mutagenic activities.  相似文献

Vascular endothelial growth factor (VEGF) plays a dominant role in angiogenesis. While inhibitors of the VEGF pathway are approved for the treatment of a number of tumor types, the effectiveness is limited and evasive resistance is common. One mechanism of evasive resistance to inhibition of the VEGF pathway is upregulation of other pro-angiogenic factors such as fibroblast growth factor (FGF) and epidermal growth factor (EGF). Numerous in vitro assays examine angiogenesis, but many of these assays are performed in media or matrix with multiple growth factors or are driven by VEGF. In order to study angiogenesis driven by other growth factors, we developed a basal medium to use on a co-culture cord formation system of adipose derived stem cells (ADSCs) and endothelial colony forming cells (ECFCs). We found that cord formation driven by different angiogenic factors led to unique phenotypes that could be differentiated and combination studies indicate dominant phenotypes elicited by some growth factors. VEGF-driven cords were highly covered by smooth muscle actin, and bFGF-driven cords had thicker nodes, while EGF-driven cords were highly branched. Multiparametric analysis indicated that when combined EGF has a dominant phenotype. In addition, because this assay system is run in minimal medium, potential proangiogenic molecules can be screened. Using this assay we identified an inhibitor that promoted cord formation, which was translated into in vivo tumor models. Together this study illustrates the unique roles of multiple anti-angiogenic agents, which may lead to improvements in therapeutic angiogenesis efforts and better rational for anti-angiogenic therapy.  相似文献

Environmental effects on body size are of widespread ecological and economic importance but our understanding of these effects has been obscured by an apparent paradox. Life history analysis suggests that it is adaptive for adults to emerge smaller if reared in conditions that slow down juvenile growth. However, whereas smaller adults emerge if growth is limited by food availability, the reverse is usually observed if growth is limited by temperature. The resolution of this apparent paradox may be that the response of adult size to temperature is adaptive, but is constrained by a trade-off that can be understood in terms of von Bertalanffy's classic theory of growth. Alternatively, the response may be the unavoidable consequence of a fundamental relationship between cell size and temperature.  相似文献

It is often claimed that some organisms maximise individual growth rates (within limits set by physiological and environmental constraints) whereas others do not. Adequate experimental protocol to test for these hypotheses has not been available. We proceeded from the idea that a maximiser should be unable to further improve its growth performance, i.e. there should be no adaptive plasticity in relative growth rates. In the case of an insect larva always maximising its weight gain, we should thus expect no increase in growth rate following a short-term environmental perturbation. Another prediction is that initial and final weights of a larval instar should be highly correlated. We applied this approach to test for the growth rate maximiser status of the geometrid moth Epirrita autumnata . Based on various lines of ecological evidence it had previously been suggested that this spring-feeding species is strongly selected for high growth rates, and may be growing at its maximum physiological potential. Contrary to expectations, the larvae responded to starvation treatments by a subsequent compensatory increase in their relative growth rates. Such a plasticity may indicate that the rates of gaining weight are not maximised even in a 'classical' example a time-constrained organism. This observation supports the idea that endogenous regulation of growth rates is widespread, and it remains to be shown if true maximisation ever exists in nature. In contrast, possibilities to compensate for adverse conditions experienced early in larval development appeared to be limited. This was indicated by the strong correlation between initial weight in the last instar, and pupal weight. Though consistent with our expectations, such an inability to compensate can hardly be ascribed to growth rate maximisation, as indicated by quantitatively similar patterns found in an unrelated species with different ecology, the butterfly Polygonia c-album .  相似文献

Mg2+-availability in Staphylococcus aureus cells decreased significantly with increasing NaCl concentration in growth media. Cells grew in a NaCl-free, Chelex resin-treated complex medium only if the medium was supplemented with 50 microM MgCl2, while, growth was limited when the medium was further supplemented with 1.0 M NaCl. Cells grown in such a high-NaCl/low-Mg2+ medium exhibited the morphologic abnormality of larger than normal cells. Both sufficient growth and normal cell morphology were restored by increasing Mg2+ concentration in a high-NaCl medium, or by supplementation with either CaCl2 or MnSO4 in a high-NaCl/low-Mg2+ medium. Supplementing with BaCl2, SrCl2 or FeSO4, however, had no effect. These results indicate that Ca2+ and Mn2+ might play some essential role in the growth of Staphylococcus aureus in a high-NaCl/low-Mg2+ environment.  相似文献

Novel therapeutics in oncology stem from a rational design of drugs targeting selective pathways that stimulate and maintain tumor cell growth. Many of these agents are cytostatic in action and also have a limited toxicity profile. However, some can be cytotoxic if they successfully modulate molecular pathways of apoptosis, such as bcl-2. This article discusses points to consider in the design of one class of cytostatic agents, antisense therapy. Our purpose is to stimulate designs that answer the question specifically with regard to proof-of-concept, and the concepts proposed should be viewed as ideas in development rather than firm recommendations.  相似文献
