Maitake mushroom (Grifola frondosa [Dicks.] Gray) is generally cultured using the sawdust of broadleaf trees. The maitake strain Gf433 has high production efficiency, with high-quality of fruiting bodies even when 30% of the birch sawdust on the basal substrate is replaced with conifer sawdust. We performed metabolome analysis to investigate the effect of different cultivation components on the metabolism of Gf433 and Mori52 by performing CE–MS on their fruiting bodies in different cultivation conditions to quantify the levels of amino acids, organic acids, and phosphorylated organic acids. We found that amino acid and organic acid content in Gf433 were not affected by the kind of sawdust. However, Gf433 contained more organic acids and less amino acids than Mori52, and Gf433 also contained more chitin compared with Mori52. We believe that these differences in the metabolome contents of the two strains are related to the high production efficiency of Gf433.  相似文献

The lectin extracted from Vicia graminea seeds has been purified by conventional techniques but such procedures did not give a satisfactory yield. We describe a new purification which involves 3 steps after obtention of the crude extract. The first step is based on affinity chromatography on con A—Sepharose. Further purification steps were performed on DEAE-Sephacel chromatography and ultrogel AcA44 gel filtration. The homogeneity of the lectin was demonstrated by polyacrylamide gel electrophoresis. Purification of the lectin by this new method was less time consuming, the yield was higher and the specific activity increased.  相似文献

Abstract

Sorption of lead(II) or cadmium(II) ions on humic acid particles (at pH 3.5) yields surface complexes which are sparingly soluble in the pH 3.55 region. Interaction of these species with acetic acid or dilute nitric acid released ASV labile metal species (mainly hydrated metal ion). When the pH was adjusted between 4 and 7, increasing amounts of the metal humate species (and humic acid substrate) dissolved and low levels of ASV labile species were detected. Overnight interaction with an excess of acetate ions (0.01 M) led to higher lability values (attributed to acetate/humate ligand exchange). The percentage of labile metal ion detected in acetate solutions varied with the amount of metal ion initially sorbed (range 100 to 500 mmol g?1). With the lower loadings, the ASV labile levels peaked in the pH 6–7 region (at around 12% of total sorbed cadmium, and 4% of total lead). In alkaline solutions, the percentage of labile Cd fell to about half of the peak value, but with Pb, lability values increased at pH > 8, due possibly to the formation of hydroxy complex species. The ASV lability of the Cd and Pb humates, and their 24 hour lability values (determined using a transfer to cation exchanger technique) has been compared with the behaviour of Cu and Zn humates (using similar conditions).  相似文献

Giberellic acid (GA) induced germination of hazel seeds is accompanied by early increases in the specific and total activities of MVA kinase in the embryonic axes. This is followed by an increase in the activity of decarboxylation of MVA by the whole axes. The activity of MVA kinase in the cotyledons is not affected by GA treatment although increased uptake of MVA results in increased decarboxylation by cotyledon slices. The effects of cofactors and inhibitors on the activities of MVA kinase and MVA decarboxylation in a cell free extract of hazel cotyledons are described.  相似文献

The pH optima of mevalonate kinase and phosphatases in green leaves, cotyledons and chloroplasts of French bean, and in green leaves and chloroplasts of maize, have been studied. Whereas in chloroplasts the pH optimum for mevalonate kinase is at pH 7·5 with little or no activity at pH 5·5, there is with leaf and cotyledon preparations appreciable activity at the lower pH. Under some circumstances isoelectric focusing studies have given fractions showing mevalonate kinase activity at only pH 7·5 or 5·5. Acid phosphatase and ATPase activity in preparations is maximal at pH 5·5 and is much reduced in the presence of high levels of phosphate. Other investigations reported concern the stability of mevalonate kinase and phosphatase activity at pH 5·5 and 7·5 on ageing of extracts, and the activity of mevalonate kinase on greening of etiolated French bean cotyledons. The influence of metal cofactors and fluoride on mevalonate kinase and phosphatase are reported.  相似文献

菜粉蝶(Pieris rapae)体内存在着超氧物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶POD)等保护酶系统,SOD活力水平与虫龄关系密切。溴氰菊酯处理菜粉蝶后10分钟,SOD、CAT及POD的活力水平均高于正常虫体。溴氰菊酯处理褐边绿刺蛾 (Parasa consocia) 和褐刺蛾 (Thoseapastornata)后,中毒的虫体内SOD及CAT活力随着中毒程度加重而逐渐上升,接近死亡时又急剧下降;未中毒的活虫体内SOD及CAT活力水平比对照活虫为高,表明与昆虫耐药性有关。在褐边绿刺蛾和褐刺蛾体内未测得POD。  相似文献

马尾松毛虫(Dendrolimus Punctatus Walder)幼虫蛋白质含量,羧酸酮酶和多酚氧化酶活力与虫龄大小成正相关;氰戊菊酯处理后,兴奋期的蛋白质含量和羧酸酯酶活力上升,到抑制期均降低到正常虫体的水平,而多酚氧化酶的变动不大。正常虫体中脱氧核糖核酸酶(Dnase)和核糖核酸酶(Rnase)的活力存在差异:Dnase从3-5龄幼虫随虫体增大而上升,到6龄时明显降低;Rnase与虫龄大小成负相关;氰戊菊酯处理后,Dnase便开始下降并低于正常虫体的水平,而Rnase在兴奋期上升,到抑制期下降亦低于正常虫体水平。结果说明,除多酚氧化酶外,蛋白质、核酸酶和羧酸酯酶均与耐药性存在一定的相关性,研制酶的抑制剂具有实用意义。根据毒力测定结果,马尾松毛虫幼虫随虫龄增大而耐药力增加,氰戊菊酯对5龄幼虫是3龄的2.8倍,6龄是4龄的2.3倍。因此,掌握在4龄前进行药物防治是合理的。  相似文献

大豆蚜 Aphis hlycines 有翅和无翅孤雌生殖蚜为其寄主植物大豆叶和鼠李叶气味所引诱,而非寄主植物棉花叶和黄瓜叶气味处于中性,丝瓜叶和南瓜叶气味具有明显的排斥作用。非寄主植物气味可以遮蔽寄主植物气味的引诱作用。大豆蚜触角感受器对植物气味具有嗅觉生理反应,对一些化合物的最小感觉阈值达10-5至10-6体积比浓度,表明大豆蚜触角上存在识别植物气味的嗅觉受体细胞。由此证明,嗅觉在大豆蚜选择寄主植物过程中起重要作用。  相似文献

Polyadenylated mRNA was isolated from germinating watermelon cotyledons and translated in a wheat germ protein synthesizing system. The synthesis of glyoxysomal malate dehydrogenase was detected by direct immunoprecipitation and electrophoretic analysis of the precipitate. In addition to a small amount of the authentic isoenzyme (subunit molecular weight = 33 000), the major part of the incorporated [35S] methionine was observed in a polypeptide with a molecular weight of 38 000. The possible role of the larger molecule as a precursor of glyoxysomal malate dehydrogenase is discussed.  相似文献

本文报道杀斜纹夜娥(Prodenia lirura)δ-内毒素和杀鞘翅目昆虫δ-内毒素经昆虫肠液蛋白酶及胰蛋白酶作用后,其毒性肽及毒力特异性的变化。 以Sephadex G75柱层析提纯的130kD原毒素,经斜纹夜蛾幼虫肠液蛋白酶作用后,产生的70kD与75kD抗蛋白酶多肽(PRP)对斜纹夜蛾和家蚕皆有毒;经家蚕幼虫肠液蛋白酶作用后,产生的62kD与65kD的PRP失去对斜纹夜蛾的毒性,仅对家蚕有毒;经胰蛋白酶作用后产生的65kD与68kD的PRP,其毒力特性与经家蚕肠液蛋白酶作用后的相似。斜纹夜蛾肠液蛋白酶作用后产生的PRP,可进一步被家蚕肠液蛋白酶或胰蛋白酶降解为63-65kD的PRP,此种多肽对斜纹夜蛾无毒,对家蚕有毒。杀鞘翅目昆虫的原毒素不能为胰蛋白酶和粘虫肠液所完全降解。证明不同种类昆虫的肠道蛋白酶对占-内毒素蛋白质的作用位点不同,δ-内毒素对宿主昆虫的毒力特异性与其肠道蛋白酶的特性密切相关。  相似文献

刚羽化的淡色库蚊(Culex pipiens pallens)去首后,其第I卵泡停滞在N期,不能进一步发育。 5天以上日龄滞育蚊去首后,第I卵泡约在去首后24小时即从N期开始发育。 组织切片发现,滞育蚊咽侧体(CA)体积小,形态瘦长,略呈圆锥形,胞核着色深,排列紧密,胞质少,似处于失活状态。 滞育蚊卵巢转种到发育蚊体内,其卵泡可以发育,而转种到滞育蚊体内则不能发育。类保幼激素(JHA,ZR515)及从发育蚊血淋巴液中提取的粗制保幼激素都能使滞育蚊的卵泡发育。  相似文献

松毛虫赤跟蜂 Trichogramma dendrolimi Matsumura 白跗平腹Pseudanastatusalbitarsis Ashmead松毛虫宽缘金小蜂Pachyneuron olitarium(Harig), 松毛虫黑卵蜂 Telenomus dendrolimusi Chu和大蛾卵跳小蜂Ooenvyrtus Kuwanae(Howard)是松毛虫卵期几种主要寄生蜂。通过对上述几种寄生蜂寄生习性的研究表明:赤眼蜂和平腹小蜂不但能寄生新鲜害虫卵,还可以寄生对方已寄生并巳发育1-3天的寄生卵,但羽化串均不高;松毛虫宽缘金小蜂不寄生新鲜害虫卵,专门寄生赤眼蜂寄生后巳发育1-7,天的寄生卵,其中又以赤眼蜂发育3-5天的卵寄生串和羽化率为最高。除赤眼蜂和平腹小蜂可以混合使用外,其它蜂种混用都不同程度地存在相互排挤的现象。如果先让平腹小蜂寄生尔后再让赤眼蜂寄生或先让赤眼蜂寄生,尔后再让跳小蜂或黑卵蜂寄生,能够提高卵块寄生串和充分发挥各自天敌的作用。  相似文献

本文针对苜蓿夜娥Autographa californica核多角体病毒的多角体蛋白mRNA设计并合成了两个ribozyme体外实验结果表明它们都能准确、高效地切割体外转录得到的mRNA片段。实验表明,在一定的范围内,切割百分率随着温度的升高、时间的延长和ribozyme浓度的提高而增加。Ca2+、Mn2+可以代替Mg2+作为ribozyme切割反应所必需的二价金属离子。  相似文献
