Using molecular fingerprinting (amplified fragment length polymorphism [AFLP] method), we explored the potential of small-scale population analysis for understanding colonization patterns of herb layer species in forests after canopy disturbance. We investigated three common forest understorey species with different life forms (Trientalis europaea, Calamagrostis villosa, and Vaccinium myrtillus) in the Harz Mountains in Germany in three different gap age classes and undisturbed forest. For two of them (T. europaea and C. villosa), we analyzed clone sizes and clonal structure. We hypothesized that clone sizes depend on age since gap formation and are affected by light availability. Mean patch sizes of V. myrtillus, T. europaea, and C. villosa formed were 3.7 m(2), 27.9 m(2), and 40.6 m(2), respectively. Trientalis europaea and C. villosa patches consisted mostly of more than one genet. Largest clone sizes of T. europaea were encountered in gaps of intermediate successional age (15-60 years, averaged minimum estimation of clone sizes: 6.56 m(2)) whereas clone size of C. villosa was found to be independent from gap age and had a mean minimum clone size of 0.49 m(2). In both species, clone size was positively related to light availability. Additionally, there was a positive relationship between clone size and ramet density for T. europaea and C. villosa. Genetic variation was higher within populations of T. europaea and C. villosa than among populations. Trientalis europaea was the only species with a clear genetic isolation by distance, pointing at an equilibrium between gene flow and genetic drift. In conclusion, we showed that forest canopy gap dynamics clearly affect the small-scale structure of populations of understorey plants. Species with high lateral growth rates, such as T. europaea offer the possibility to serve as "ecological clock" for dating ecological processes.  相似文献

Cultivated rice (Oryza sativa) is an AA genome Oryza species that was most likely domesticated from wild populations of O. rufipogon in Asia. O. rufipogon and O. meridionalis are the only AA genome species found within Australia and occur as widespread populations across northern Australia. The chloroplast genome sequence of O. rufipogon from Asia and Australia and O. meridionalis and O. australiensis (an Australian member of the genus very distant from O. sativa) was obtained by massively parallel sequencing and compared with the chloroplast genome sequence of domesticated O. sativa. Oryza australiensis differed in more than 850 sites single nucleotide polymorphism or indel from each of the other samples. The other wild rice species had only around 100 differences relative to cultivated rice. The chloroplast genomes of Australian O. rufipogon and O. meridionalis were closely related with only 32 differences. The Asian O. rufipogon chloroplast genome (with only 68 differences) was closer to O. sativa than the Australian taxa (both with more than 100 differences). The chloroplast sequences emphasize the genetic distinctness of the Australian populations and their potential as a source of novel rice germplasm. The Australian O. rufipogon may be a perennial form of O. meridionalis.  相似文献

Many animals use photoperiod cues to synchronize reproduction with environmental conditions and thereby improve their reproductive success. The circadian clock, which creates endogenous behavioral and physiological rhythms typically entrained to photoperiod, is well characterized at the molecular level. Recent work provided evidence for an association between Clock poly-Q length polymorphism and latitude and, within a population, an association with the date of laying and the length of the incubation period. Despite relatively high overall breeding synchrony, the timing of clutch initiation has a large impact on the fitness of swallows in the genus Tachycineta. We compared length polymorphism in the Clock poly-Q region among five populations from five different Tachycineta species that breed across a hemisphere-wide latitudinal gradient (Fig. 1). Clock poly-Q variation was not associated with latitude; however, there was an association between Clock poly-Q allele diversity and the degree of clutch size decline within breeding seasons. We did not find evidence for an association between Clock poly-Q variation and date of clutch initiation in for any of the five Tachycineta species, nor did we found a relationship between incubation duration and Clock genotype. Positive demographic responses have been reported in several species where the immigration or supplementation of genetically distinct individuals into wild populations has resulted in a genetic rescue effect. However, rarely have researchers incorporated what could be considerable risk of outbreeding depression into planning for genetic management programs. We assess the genetic effects of an experiment in genetic management involving replicate populations of California bighorn sheep (Ovis canadensis californiana) in Oregon, USA, which previously experienced poor productivity and numerical declines. In the experiment, two declining populations were supplemented with ewes from a more genetically diverse population of California bighorn sheep in Nevada. We incorporated analysis of genetic samples representing both experimental populations prior to supplementation, samples from the supplemented individuals, and samples collected from both experimental populations approximately one generation after supplementation. We used genetic analyses to assess the integration of supplemented and resident populations by identifying interpopulation hybrids. Further, we incorporated demographic simulations to assess the risk of outbreeding depression as a result of the experimental augmentation. Finally, we used data from microsatellites and mitochondrial sequences to determine if genetic management increased genetic diversity in the experimental populations. Our analyses demonstrated the success of genetic management by documenting interpopulation hybrids, identifying no evidence for outbreeding depression as a result of contact between the genetically distinct supplemented and resident populations, and by identifying increased population-level metrics of genetic diversity in postsupplementation populations compared with presupplementation levels.  相似文献

Sexual selection has the potential to contribute to population divergence and speciation. Most studies of sexual selection in Drosophila have concentrated on a single signaling modality, usually either courtship song or cuticular hydrocarbons (CHCs), which can act as contact pheromones. We have examined the relationship between both signal types and reproductive success using F(1-3) offspring of wild-collected flies, raised in the lab. We used two populations of the Holarctic species Drosophila montana that represent different phylogeographic clades that have been separate for ca. 0.5 million years (MY), and differ to some extent in both traits. Here, we characterize the nature and identify the targets of sexual selection on song, CHCs, and both traits combined within the populations. Three measures of courtship outcome were used as fitness proxies. They were the probability of mating, mating latency, and the production of rejection song by females, and showed patterns of association with different traits that included both linear and quadratic selection. Courtship song predicted courtship outcome better than CHCs and the signal modalities acted in an additive rather than synergistic manner. Selection was generally consistent in direction and strength between the two populations and favored males that sang more vigorously. Sexual selection differed in the extent, strength, and nature on some of the traits between populations. However, the differences in the directionality of selection detected were not a good predictor of population differences. In addition, a character previously shown to be important for species recognition, interpulse interval, was found to be under sexual selection. Our results highlight the complexity of understanding the relationship between within-population sexual selection and population differences. Sexual selection alone cannot predict differences between populations.  相似文献

Phylogeographic studies indicate that many marine invertebrates lacking autonomous dispersal ability are able to achieve trans-oceanic colonization by rafting on buoyant macroalgae. However, less is known about the impact of rafting on on-going population-genetic connectivity of intertidal species associated with buoyant macroalgae. We hypothesize that such species will have higher levels of population-genetic connectivity than those exploiting nonbuoyant substrates such as rock. We tested this hypothesis by comparing nuclear multilocus population-genetic structuring in two sister topshell species, which both have a planktonic larval phase but are fairly well segregated by their habitat preference of low-tidal bull-kelp holdfasts versus mid-to-low tidal bare rock. We analyzed population samples from four sympatric sites spanning 372 km of the east coast of southern New Zealand. The sampled region encompasses a 180 km wide habitat discontinuity and is influenced by a stable, northward coastal current. The level of connectivity was high in both species, and neither of them showed significant correlation between genetic and geographic distances. However, a significant negative partial correlation between genetic distance and habitat discontinuity was found in the rock-associated species, and estimates of migrant movement between sites were somewhat different between the two species, with the kelp-associated species more often yielding higher estimates across the habitat discontinuity, whereas the rock-associated species more often exhibited higher estimates between sites interspersed by rock habitats. We conclude that for species with substantial means of autonomous dispersal, the most conspicuous consequence of kelp dwelling may be enhanced long-distance dispersal across habitat discontinuities rather than a general increase of gene flow.  相似文献

Species occupying a broad latitudinal range may show greater phenotypic plasticity in behavior than species with smaller ranges or more specific habitat requirements. This study investigates for the first time the occurrence of conspecific brood parasitism (CBP) in sympatric tropical populations of the common moorhen (Gallinula chloropus pauxilla Bangs) and the American purple gallinule (Porphyrula martinica L.). CBP occurred in at least 20% (N = 76) of common moorhen nests on the Rio Chagres in Panama. Half (N = 20) of the parasitic eggs were accepted, but 10 were destroyed or ejected from host nests. Introductions of experimental eggs into nests revealed hosts were more likely to accept parasitism later in the host's laying period and during incubation, consistent with expectation of an adaptive response. CBP was not detected in a small sympatric population of American purple gallinules. Members of this population did not eject experimental eggs, suggesting a lack of experience with costly CBP. Contrasting ecological factors help explain why these two species of rail (Family Rallidae) differ in regard to CBP. Purple gallinule territories were sparse, owing to the distribution of preferred habitat. Moorhens flocked outside of the breeding season. They nested more synchronously, at higher densities, and primarily in ephemeral floating vegetation. Further, moorhens suffered a rate of nest loss nearly double that of American purple gallinules, and this increased over the course of the breeding season. Moorhen clutches were larger on average, and more variable in size than those of purple gallinules. Reproductive effort and rate (seasonality) constitute important life history differences between these species that may constrain the evolution of reproductive tactics. Comparing these sympatric populations, and others differing in life-history traits and ecological constraints, highlights the role of risk management in the evolution of CBP.  相似文献

Myotis lucifugus, once among the most widespread and common bats in North America, has been forecast to be extirpated east of the Rockies in as few as 16 years by the spread of white-nose syndrome. Recent genetic research has demonstrated that this species is paraphyletic and part of a broader species complex; however, only one lineage (Myotis lucifugus lucifugus [M. l. lucifugus]) is present in eastern North America. I used molecular tools and niche modeling to validate this and investigate the role that historical biogeography has played in the phylogenetic and population genetic structure of this species to determine if the eastern subspecies represents an evolutionarily distinct population.To establish the genetic structure within M. l. lucifugus, I densely sampled maternity colonies in Minnesota and sequenced 182 individuals for a portion of cytochrome b. Phylogenetic reconstruction and a haplotype network were used to infer the relationships among mitochondrial haplotypes. Population growth statistics were calculated to determine if there was evidence of significant expansion, and an environmental niche model (ENM) was constructed based on conditions during the last glacial maximum (LGM) to illustrate potential glacial refugia. All individuals derived from a single mitochondrial lineage. Genetic evidence points to population growth starting approximately 18 kya. ENM results show that there was likely a single large southern refugium extending across the southeastern United States and possibly several isolated refugia in western North America. Myotis lucifugus lucifugus likely maintained both a large range and a large population during the peaks of the glacial cycles, and its population appears to have expanded following the retreat of the Laurentide ice sheet. This imperiled lineage likely diverged in isolation from other members of the M. lucifugus/western long-eared Myotis during the Pleistocene.  相似文献

The use of marine protected area (MPA) networks to sustain fisheries and conserve biodiversity is predicated on two critical yet rarely tested assumptions. Individual MPAs must produce sufficient larvae that settle within that reserve's boundaries to maintain local populations while simultaneously supplying larvae to other MPA nodes in the network that might otherwise suffer local extinction. Here, we use genetic parentage analysis to demonstrate that patterns of self-recruitment of two reef fishes (Amphiprion percula and Chaetodon vagabundus) in an MPA in Kimbe Bay, Papua New Guinea, were remarkably consistent over several years. However, dispersal from this reserve to two other nodes in an MPA network varied between species and through time. The stability of our estimates of self-recruitment suggests that even small MPAs may be self-sustaining. However, our results caution against applying optimization strategies to MPA network design without accounting for variable connectivity among species and over time.  相似文献

The objectives of this study were to describe and evaluate potential drivers of genetic structure in Canadian breeding populations of the Ovenbird, Seiurus aurocapilla. We performed genetic analyses on feather samples of individuals from six study sites using nuclear microsatellites. We also assessed species identity and population genetic structure of quill mites (Acariformes, Syringophilidae). For male Ovenbirds breeding in three study sites, we collected light‐level geolocator data to document migratory paths and identify the wintering grounds. We also generated paleohindcast projections from bioclimatic models of Ovenbird distribution to identify potential refugia during the last glacial maximum (LGM, 21,000 years before present) as a factor explaining population genetic structure. Birds breeding in the Cypress Hills (Alberta/Saskatchewan) may be considered a distinct genetic unit, but there was no evidence for genetic differentiation among any other populations. We found relatively strong migratory connectivity in both western and eastern populations, but some evidence of mixing among populations on the wintering grounds. There was also little genetic variation among syringophilid mites from the different Ovenbird populations. These results are consistent with paleohindcast distribution predictions derived from two different global climate models indicating a continuous single LGM refugium, with the possibility of two refugia. Our results suggest that Ovenbird populations breeding in boreal and hemiboreal regions are panmictic, whereas the population breeding in Cypress Hills should be considered a distinct management unit.  相似文献

Various instars of Choristoneura occidentalis were fed with a range of doses of Nosema fumiferanae and reared at 20, 24 and 28 degrees C to determine the influence of temperature and dose on the time to spore egestion and the number of spores egested in the frass. When larvae were fed in the third stadium, as few as 10(2) spores per larva initiated infection, and both onset of spore egestion and the number of spores egested were affected by a complex relationship between temperature and inoculation dose. Onset of spore egestion varied from 11 to 15 days postinoculation. At 20 degrees C, the onset was delayed and spore production decreased with increasing inoculation dose whereas at higher temperatures spores were first egested at the lowest dose and spore production increased with dose. When larvae were fed spores in the fifth and sixth stadium, no spores were egested because pupation occurred before completion of the incubation period. To assess the effect of temperature on horizontal transmission, Choristoneura fumiferana larvae fed with 10(4) N. fumiferanae spores per larva were reared with uninfected larvae at 15, 20 and 25 degrees C. At 15 degrees C, we observed the highest degree of horizontal transmission, defined by the largest change in N. fumiferanae prevalence, even though the density of spores available for horizontal transmission was the lowest. Infected adults eclosed later than uninfected adults and the time to eclosion was also dependent on sex and temperature. We relate our experimental findings to consequences for horizontal and vertical transmission of N. fumiferanae in spruce budworm populations.  相似文献

Climate change is predicted to negatively impact wildlife through a variety of mechanisms including retraction of range. We used data from the North American Breeding Bird Survey and regional and global climate indices to examine the effects of climate change on the breeding distribution of the Rusty Blackbird (Euphagus carolinus), a formerly common species that is rapidly declining. We found that the range of the Rusty Blackbird retracted northward by 143 km since the 1960s and that the probability of local extinction was highest at the southern range margin. Furthermore, we found that the mean breeding latitude of the Rusty Blackbird was significant and positively correlated with the Pacific Decadal Oscillation with a lag of six years. Because the annual distribution of the Rusty Blackbird is affected by annual weather patterns produced by the Pacific Decadal Oscillation, our results support the hypothesis that directional climate change over the past 40 years is contributing to the decline of the Rusty Blackbird. Our study is the first to implicate climate change, acting through range retraction, in a major decline of a formerly common bird species.  相似文献

Hybridization has been repeatedly put forward to explain the invasiveness of Rhododendron ponticum L. in the British Isles. The present study investigates the pattern of ecotypic differentiation and hybridization among native North American R. catawbiense and R. maximum, native R. ponticum from Georgia and Spain, and invasive R. ponticum from Ireland and aims to assess the contribution of hybridization for Rhododendron invasion in the British Isles. Six populations per taxon were analyzed with AFLP markers for genetic dissimilarity, subjected to germination and growth experiments, and tested for frost hardiness. We assessed variation in morphological and ecological characteristics to identify traits displaying evidence of hybridization, thus, promoting invasiveness. Molecular marker analyses revealed a clear distinction between North American R. catawbiense and R. maximum on the one hand, and all R. ponticum populations on the other hand, displaying a complete intermixture of native Spanish and invasive Irish populations. Multivariate analyses of traits revealed leaf length-width ratio, relative growth rates (RGRs) in leaf length, root biomass, and shoot-root ratio to significantly discriminate between the different taxa and unequivocally assigned invasive Irish R. ponticum to the Spanish phenotypes. While the Irish R. ponticum had similar growth traits as conspecific native R. ponticum provenances, germination and biomass allocation were more similar to North American R. catawbiense and R. maximum. Hybridization did not contribute to explaining invasiveness of R. ponticum in Ireland. The similarity in germination and biomass allocation of invasive Irish R. ponticum and North American species has evolved independently and can more probably be attributed to an independent shift within the Ponticum cluster in Ireland.  相似文献

We have isolated a microsporidium from a laboratory colony of the eastern spruce budworm, Choristoneura fumiferana (Clem.) (Lepidoptera: Tortricidae). Light and electron microscopic investigations showed that gross pathology and ultrastructure of our isolate are similar to those described for Cystosporogenes legeri from the European grape vine moth, Lobesia botrana. Comparative phylogenetic analysis of the small subunit rDNA using maximum likelihood, maximum parsimony, and neighbour joining distance methods revealed perfect homology with the C. legeri sequence. The microsporidian was infectious to other Choristoneura species, as well as Malacosoma disstria, Lymantria dispar, and Lambdina fiscellaria. Incubation of infected egg masses at 41 degrees C for 20 min followed by 30 min in 33% formaldehyde did not reduce disease incidence in larval offspring. Exposure of one or two generations to fumagillin at 6000 ppm or higher eliminated infection in adult moths, but also reduced colony fitness. A clean colony was established by conducting individual matings and selecting disease-free offspring.  相似文献

Clinal variation can result from primary differentiation or secondary contact and determining which of these two processes is responsible for the existence of a cline is not a trivial problem. Samples from a coastal transect of New Zealand geckos (Woodworthia maculata) identified for the first time a body size cline 7-10 km wide. The larger geckos are almost twice the mass of the small adult geckos. Clines in allele and haplotype frequency were found at two of the four genetic loci examined. Estimated width of the morphological cline was concordant with neither the narrower mtDNA cline (3-7 m) nor the wider nuclear cline (RAG-2; 34-42 km), and cline centers were not coincident. Although the body size cline is narrow compared to the entire range of the species, it is 2-3 orders of magnitude greater than estimates of dispersal distance per generation for these geckos. No evidence of assortative mating, nor of hybrid disadvantage was identified, thus there is little evidence to infer that endogenous selection is maintaining a hybrid zone. We cannot distinguish secondary contact from primary origin of this body size cline but conclude that secondary contact is likely due to the occurrence of mtDNA haplotypes from three distinct clades within the coastal transect and the presence of two frequency clines within this region.  相似文献

Little is known about the microevolutionary processes shaping within river population genetic structure of aquatic organisms characterized by high levels of homing and spawning site fidelity. Using a microsatellite panel, we observed complex and highly significant levels of intrariver population genetic substructure and Isolation-by-Distance, in the Atlantic salmon stock of a large river system. Two evolutionary models have been considered explaining mechanisms promoting genetic substructuring in Atlantic salmon, the member-vagrant and metapopulation models. We show that both models can be simultaneously used to explain patterns and levels of population structuring within the Foyle system. We show that anthropogenic factors have had a large influence on contemporary population structure observed. In an analytical development, we found that the frequently used estimator of genetic differentiation, F(ST), routinely underestimated genetic differentiation by a factor three to four compared to the equivalent statistic Jost's D(est) (Jost 2008). These statistics also showed a near-perfect correlation. Despite ongoing discussions regarding the usefulness of "adjusted"F(ST) statistics, we argue that these could be useful to identify and quantify qualitative differences between populations, which are important from management and conservation perspectives as an indicator of existence of biologically significant variation among tributary populations or a warning of critical environmental damage.  相似文献

The Culex pipiens complex includes two widespread mosquito vector species, Cx. pipiens and Cx. quinquefasciatus. The distribution of these species varies in latitude, with the former being present in temperate regions and the latter in tropical and subtropical regions. However, their distribution range overlaps in certain areas and interspecific hybridization has been documented. Genetic introgression between these species may have epidemiological repercussions for West Nile virus (WNV) transmission. Bayesian clustering analysis based on multilocus genotypes of 12 microsatellites was used to determine levels of hybridization between these two species in Macaronesian islands, the only contact zone described in West Africa. The distribution of the two species reflects both the islands' biogeography and historical aspects of human colonization. Madeira Island displayed a homogenous population of Cx. pipiens, whereas Cape Verde showed a more intriguing scenario with extensive hybridization. In the islands of Brava and Santiago, only Cx. quinquefasciatus was found, while in Fogo and Maio high hybrid rates (~40%) between the two species were detected. Within the admixed populations, second-generation hybrids (~50%) were identified suggesting a lack of isolation mechanisms. The observed levels of hybridization may locally potentiate the transmission to humans of zoonotic arboviruses such as WNV.  相似文献

We aim to show how a combination of molecular systematics and ecological niche modelling approaches can be used to test historical biogeographical hypotheses for species of conservation concern. We focus on the land snail genus Oreohelix (Oreohelicidae), a group found throughout the Rocky Mountains. In addition to its larger distribution, a group of Oreohelix is also found in the Black Hills of Wyoming and South Dakota, an isolated, easternmost extension of the Rocky Mountains. We determine the number, distribution, and relationships of Black Hills Oreohelicids, which are a current conservation concern due to their fragmented distribution. We compared Black Hills groups to those in the main part of the Rockies to test historical biogeographical patterns that explain current diversity. We collected mtDNA data (COI and 12S sequences) from multiple populations of Oreohelix throughout the Black Hills and in adjacent populations in the Rocky Mountains to construct phylogenetic hypotheses. To determine whether favourable environmental conditions currently exist between the Black Hills and the north-eastern Rocky Mountains, we used DesktopGARP to generate an ecological niche model for distinct lineages discovered in the molecular phylogenetic analysis. Results show that all Black Hills populations are likely Oreohelix cooperi and that little genetic differentiation exists within this clade. In addition, Black Hills groups are genetically similar or identical to populations found in the Judith Mountains and Bighorn Mountains (north-eastern Rockies). Ecological niche models show that suitable environmental conditions may exist between eastern Rockies and Black Hills O. cooperi samples. Taken together, the phylogenetic and niche model data, along with the low vagility of the snails, support passive long-distance dispersal as a likely explanation for current arrangement of biodiversity.  相似文献

Despite current interest in population genetics, a concrete definition of a "population" remains elusive. Multiple ecologically and evolutionarily based definitions of population are in current use, which focus, respectively, on demographic and genetic interactions. Accurate population delimitation is crucial for not only evolutionary and ecological population biology, but also for conservation of threatened populations. Along the Pacific Coast of North America, two contrasting patterns of geographic variation in deer mice (Peromyscus maniculatus) converge within the state of Oregon. Populations of these mice diverge morphologically across an east-west axis, and they diverge in mitochondrial DNA haplotypes across a north-south axis. In this study, we investigate these geographically contrasting patterns of differentiation in the context of ecological and evolutionary definitions (paradigms) of populations. We investigate these patterns using a new and geographically expansive sample that integrates data on morphology, mitochondrial DNA, and nuclear DNA. We found no evidence of nuclear genetic differentiation between the morphologically and mitochondrially distinct populations, thus indicating the occurrence of gene flow across Oregon. Under the evolutionary paradigm, Oregon populations can be considered a single population, whereas morphological and mitochondrial differentiations do not indicate distinct populations. In contrast, under the ecological paradigm morphological differentiation indicates distinct populations based on the low likelihood of demographic interactions between geographically distant individuals. The two sympatric but mitochondrially distinct haplogroups form a single population under the ecological paradigm. Hence, we find that the difference between evolutionary and ecological paradigms is the time-scale of interest, and we believe that the more chronologically inclusive evolutionary paradigm may be preferable except in cases where only a single generation is of interest.  相似文献
