Treatment of suspensions of vesicular stomatitis virus with Tween-ether results in a rapid and considerable loss of infectivity (ca. 4 logs in 2 min), but the residual infectivity is comparatively stable to further treatment with ether. The infectivity remaining after the short exposure to Tween-ether is not due to virus for the following reasons. (i) It is much less infective for tissue cultures than for mice, whereas the intact virion is equally infective for both hosts. (ii) The residual infectivity is much less stable than virus infectivity in both sucrose and tartrate gradients. (iii) Virus immune serum does not neutralize its activity. (iv) The infectivity is associated with material which sediments further in sucrose gradients and has a greater buoyant density in tartrate gradients than the virion. Experiments with (32)P-labeled virion showed that the infective substructure contains ribonucleic acid with the same sedimentation characteristics as that extracted from the virion. Electron microscopy shows that the infective component has the same overall bullet-like structure as the virion but lacks the outer envelope and fringe structure.  相似文献

Rabies remains a major public health threat around the world. Once symptoms appear, there is no effective treatment to prevent death. In this work, we tested a recombinant parainfluenza virus 5 (PIV5) strain expressing the glycoprotein (G) of rabies (PIV5-G) as a therapy for rabies virus infection: we have found that PIV5-G protected mice as late as 6 days after rabies virus infection. PIV5-G is a promising vaccine for prevention and treatment of rabies virus infection.  相似文献

Isometric virus-like particles (VLPs) have been purified from cucumber leaf tissue. Three dsRNA segments with estimated molecular weights of 1.8, 1.1 and 1.0 × 106d have been isolated from VLPs occurring in CsCl density gradient fractions but were also readily detected in preparations from as little as 1 g of fresh leaf tissue. The VLPs resemble dsRNA containing cryptic viruses and have been named Cucumis sativus cryptic virus (CsCV).  相似文献

Marked cytopathic changes were induced by challenge with Newcastle disease (ND) virus in bovine testicle or kidney cell cultures which were previously infected with non-cytopathogenic strains of bovine diarrhea (BD) virus. No cytopathic changes were induced by ND virus in similar cells not infected with BD virus. The development of cytopathic effect was shown to be associated with enhancement of ND virus replication. This exalting effect of BD virus appears to be dependent on infectivity, since the effect was inhibited when infection of the cells with BD virus was blocked by specific antiserum. Various factors involved in the phenomenon were investigated and an in vitro method (END) for the assay of BD virus and its antibodies was developed. The use of this method eliminates the difficulties in recognizing non-cytopathogenic strains of BD virus which hampered systematic investigations of the nature and behavior of BD virus as well as of the natural history and pathogenesis of the infection in cattle.  相似文献

Powassan virus isolations were achieved from three of 60 pools of Ixodes cookei ticks removed from 286 groundhogs (Marmota monax) which were collected some 200 miles north of Toronto between May 5 and September 5, 1966. Virus yields per pool of one to 11 ticks ranged from 102.5 to 106.0 TCD50 for primary swine kidney tissue cultures, and positive pools were collected on June 24, July 15 and August 10. Powassan neutralizing antibodies were detected by mouse inoculation tests in 143 of 362 animals including 127 of 286 groundhogs, 14 of 45 red squirrels (Tamiasciurus hudsonicus) and two of 31 other forest mammals. The monthly prevalence of antibody in the current season''s groundhogs increased from 0 to 25% with the progression of summer, but in older animals the incidence remained between 38 and 62% throughout the season. These results substantiate earlier findings which pointed towards the maintenance of Powassan virus in nature by a cycle involving groundhogs and squirrels as reservoirs, with ticks as vectors, from which human infections occurred tangentially.  相似文献

Virus strain No. 12, one of the new isolates from Japanese cattle described previously, was studied for its physicochemical properties. The new isolate was shown to be very small in size by centrifugation and filtration, being filtrable through Millipore filters of 50 mμ pore size. It appears to be an RNA virus as its replication was not inhibited by 5-iodo-2′-deoxyuridine. The virus was readily inactivated by ether and deoxycholate, and partially by trypsin; was labile at pH 3, not stabilized by 1 m MgCl2 at 50 C, was inactivated by ultraviolet, and withstood repeated freeze-thawing. Further it was readily inactivated at 56 C but more slowly at 37 C, and was stable at lower temperatures. These findings support the identification of the isolated virus as the bovine diarrhea (BD) virus. The properties of BD virus, i.e. size, type of nucleic acid, ether, chloroform and deoxycholate sensitivities, and acid lability, appear to be similar to those of arboviruses. The trypsin sensitivity of BD virus is similar to the B group of arboviruses, which, unlike the A group, sensitive to trypsin. For the classification of BD virus as well as hog cholera virus, which is closely related, further elucidation of properties, fine structure of the virion, etc., is needed.  相似文献

Enhancement of tumor induction by oncornaviruses through a dual viral infection has been described by several investigators. The mechanism(s) of this enhancement has not as yet been determined. By using a murine sarcoma virus and a Bunyawera group arbovirus (Guaroa virus) in an in vitro system, evidence was obtained for enhancement of the oncogenic potential of the viruses by genetic interaction with the nononcogenic virus as well as the production of increased amounts of the oncogenic virus. These results confirm and extend similar responses obtained in in vivo systems.  相似文献

Pseudotype virus vectors serve as a powerful tool for the study of virus receptor usage and entry. We describe the development of murine leukemia virus (MuLV) particles pseudotyped with the visna virus envelope glycoprotein and encoding a green fluorescent protein reporter as a tool to study the expression of the visna virus receptor. Functional MuLV/visna virus pseudotypes were obtained when the cytoplasmic tail of the visna virus envelope TM protein was truncated to 3, 7, or 11 amino acids in length. MuLV/visna virus particles were used to transduce a panel of cell types from various organisms, including sheep, goat, human, hamster, mouse, monkey, and quail. The majority of the cells examined were susceptible to MuLV/visna pseudotype viruses, supporting the notion that the visna virus cellular receptor is a widely expressed protein found in many species. Of 16 different cell types tested, only mouse embryo fibroblast NIH 3T3 cells, hamster ovary CHO cells, and the human promonocyte cell line U937 cells were not susceptible to transduction by the pseudotyped virus. The production of functional MuLV/visna virus pseudotypes has provided a sensitive, biologically relevant system to study visna virus cell entry and envelope-receptor interactions.  相似文献

为探讨肝炎病毒(HV)与EB病毒(EBV)重叠感染的状况和后果,我们用免疫酶法对154例各型病毒性肝炎患者作了EBVIgA抗体检测。结果发现,急性肝炎、慢性轻度肝炎、慢性中度肝炎、肝炎肝硬化、慢性重型肝炎和原发性肝癌VGA-IgA抗体的阳性率分别为24.0%、30.0%、53.3%、63.3%、40.0%和72.7%,与健康人(5.3%)比较,有非常显著升高(P<0.01);原发性肝癌又较急性肝炎和慢性轻度肝炎高,并有非常显著意义差异(P<0.01)。HBV和HAV+HBV感染者比较,前者又较后者低(P<0.01)。重叠感染者的临床表现均为“肝炎型”,未见咽炎、腺热、胃肠、肺炎、肾炎、神经等类型。重叠感染者的CD+3及CD+4T细胞下降,CD+8T细胞及IgG,IgM升高,与健康人比较差异非常显著意义(P<0.01)。结果提示:HV感染,不仅因免疫失调易感EBV,又可因重叠感染而进一步使免疫功能失调;对病毒性肝炎的处理应强调免疫调节治疗。  相似文献

We have studied the binding of the tumor antigen (T-antigen) of simian virus 40 to simian virus 40 chromatin (minichromosomes). The minichromosomes isolated from infected cells by a modification of standard techniques were relatively free of contaminating RNA and cellular DNA and had a ratio (by weight) of protein to DNA of approximately 1; their DNA was 50 to 60% digestible to an acid-soluble form by staphylococcal nuclease. Cleavage of this chromatin with restriction endonucleases indicated that the nuclease-resistant regions were randomly distributed in the population of minichromosomes, but were not randomly distributed within minichromosomes. Only 20 to 35% of these minichromosomes adsorbed nonspecifically to nitrocellulose filters, permitting binding studies between simian virus 40 T-antigen and chromatin to be performed. Approximately two to three times as much T-antigen was required to bind chromatin as to bind an equivalent amount of free DNA. When T-antigen was present in excess, both chromatin and free DNA were quantitatively retained on the filters. On the other hand, when DNA or chromatin was present in excess, only one-third as much chromatin as DNA was retained. We suggest that T-antigen-chromatin complexes may be formed by the cooperative binding of T-antigen to chromatin, whereas T-antigen-DNA complexes may be formed by simple bimolecular interactions.  相似文献

Viruses, despite being relatively simple in structure and composition, have evolved to exploit complex cellular processes for their replication in the host cell. After binding to their specific receptor on the cell surface, viruses (or viral genomes) have to enter cells to initiate a productive infection. Though the entry processes of many enveloped viruses is well understood, that of most non-enveloped viruses still remains unresolved. Recent studies have shown that compared to direct fusion at the plasma membrane, endocytosis is more often the preferred means of entry into the target cell. Receptor-mediated endocytic pathways such as the dynamin-dependent clathrin and caveolar pathways are well characterized as viral entry portals. However, many viruses are able to utilize multiple uptake pathways. Fluid phase uptake, though relatively non-specific in terms of its cargo, potentially aids viral infection by its ability to intersect with the endocytic pathway. In fact, many viruses despite using specialized pathways for entry are still able to generate productive infection via fluid phase uptake. Macropinocytosis, a major fluid uptake pathway found in epithelial cells and fibroblasts, is stimulated by growth factor receptors. Many viruses can induce these signaling cascades in cells leading to macropinocytosis. Though endocytic trafficking is utilized by both enveloped and non-enveloped viruses, key differences lie in the way membranes are traversed to deposit the viral genome at its site of replication. This review will discuss recent developments in the rapidly evolving field of viral entry.  相似文献

马传染性贫血病毒(equine infectious anemia virus,EIAV)引起马传染性贫血(简称马传贫),导致马持续性感染和反复病毒血症[1].EIAV与人免疫缺陷病毒Ⅰ型(HIV-1)同属反转录病毒科慢病毒属,二者有很多相似的特性[2].在反转录病毒前病毒基因组两端含有长末端重复序列(long terminal repeat,LTR).LTR含有真核启动子,其中含有病毒转录调控顺式作用位点,病毒编码的反式作用因子与其结合后可以反式激活基因的表达,对病毒基因的表达和其它生命活动起重要调控作用[3,4].因此,LTR序列的变异可能会引起病毒转录和复制方式的改变,进而引起其细胞嗜性和致病性的改变[5,6].为了探讨LTR在EIAV病毒复制和转录过程中的作用,并进一步研究EIAV的致病和免疫机制,用EIAV强毒L株LTR置换了以前构建的EIAV DLA(弱毒)感染性分子克隆中的LTR,构建了马传贫强/弱毒嵌合分子克隆,并获得了具有感染性的强/弱毒嵌合病毒.  相似文献

Nucleoprotein complexes containing viral DNA and cellular histones were extracted from nuclei of permissive cells infected with polyoma virus or simian virus 40 (SV40) and examined by electron microscopy. Polyoma and SV40 nucleoprotein complexes are almost identical. They appear as relaxed circular molecules consisting of 20 to 21 globular particles interconnected by thin filaments. Their contour length in 0.02 M salt is 2.7 times shorter than that of viral DNA form I obtained after dissociation of the proteins in 1 M NaCl. The nucleosomes have an average diameter of 12.5 nm. Each nucleosome contains 175 to 205 DNA base pairs condensed fivefold in length. The nucleosomes are regularly spaced on the circular molecule. The internucleosomal filaments are made of naked DNA, and each filament contains about 55 base pairs. The partial sensitivity of the nucleoprotein complex to cleavage by EcoR1 endonuclease suggests that the nucleosomes are not formed at specific sites on the viral genome. Faster sedimenting nucleoprotein complexes containing replicative intermediates were studied. Isopycnic centrifugation in metrizamide gradients in the absence of aldehyde fixation showed that these molecules conserved the same DNA-to-protein ratio as the form I DNA-containing complexes.  相似文献

Abstract

Sendai virus was the first virus to encounter liposomes. Gangliosides when incorporated into liposomes act as Sendai virus receptors even at 0–4°C. When receptor-containing liposomes are incubated with virus at 37°C, they envelop the virus. At 37°C liposomes also fuse with Sendai virus membrane.

Virus binding initially involves weak adhesion, which may allow the virus to “browse” the cell, and which is followed by adhesion strengthening. MicrogrΔpHs of Sendai virus fusion with liposomes after one minute at 37°C indicate that fusion occurs at the very curved leading edge of the region of the liposome enveloping virus. A model of fusion is proposed that emphasizes the role of the curvature and membrane tension in this localized region of “host” membrane. The curvature assists close approach and destabilizes the outer monolayer. The proposed intermediates are consistent with the “stalk” hypothesis.  相似文献

The use of a Spinco L-4 zonal centrifuge with the B-XVI continuous-flow rotor for the purification of canine distemper and infectious canine hepatitis viruses is described. Up to 68 liters of virus was processed at one time. Infectious canine hepatitis virus was found to band at 39% sucrose and canine distemper virus banded between 32 and 48% sucrose. The virus was concentrated 10-fold, and the purity of the virus, as measured by protein concentration, was increased by 99%.  相似文献

IT has been proposed that lymphocytic choriomeningitis (LCM) virus and immunologically related viruses should be placed in a new taxonomic group, with LCM virus as the prototype virus1,2. They were called arenoviruses because they contain, as a unique feature, electron-dense, sand-like or ribo-some-like granules2. Hepadnaviruses (hepatitis B viruses) cause transient and chronic infections of the liver. Transient infections run a course of several months, and chronic infections are often lifelong. Chronic infections can lead to liver failure with cirrhosis and hepatocellular carcinoma. The replication strategy of these viruses has been described in great detail, but virus-host interactions leading to acute and chronic disease are still poorly understood. Studies on how the virus evades the immune response to cause prolonged transient infections with high-titer viremia and lifelong infections with an ongoing inflammation of the liver are still at an early stage, and the role of the virus in liver cancer is still elusive. The state of knowledge in this very active field is therefore reviewed with an emphasis on past accomplishments as well as goals for the future.  相似文献
