The photosynthetic performance of the cyanobacterium Synechocystis sp. PCC 6803 exposed to intermittent light was studied by measuring oxygen evolution, respiration and the fluorescence parameters for maximum efficiency of excitation energy capture by photosystem II (PSII) reaction centres ( F v/ F m), PSII quantum yield (ΔF/ F m 1) and non-photochemical quenching (NPQ). Cultures were pre-acclimated to constant light conditions. Block and sinusoidal light regimes were tested using four photon-flux densities (PFDs) applied in light/dark intervals of 1:1, 5:5 and 10:10 min. Light use was higher under the sinusoidal light regime compared with the block regime. The accumulated gross photosynthesis of the cyanobacterium was lower under intermittent light conditions compared with predictions from the photosynthesis-irradiance curve (PI curve). The respiration rates were similar for all light/dark intervals tested. However, the respiration slightly increased with increasing oxygen production for both block and sinusoidal light regime. NPQ, ΔF/ F m' and F v/ F m depended on the PFD rather than on the duration of the light/dark intervals tested, and there was no detected influence of the two applied light regimes.  相似文献

In photobioreactors and natural systems, microalgae are subjected to rapidly changing light intensities (LI) due to light attenuation and mixing. A controlled way to study the effect of rapidly changing LI is to subject cultures to flashing light. In this study, series of flashing-light experiments were conducted using Synechocystis sp. PCC6803 with constant overall average LI of approximately 84 μmol·m−2·s−1 and relative times in the light and dark varied. The results were also compared with simulated results using a mathematical model including an absorbed pool of light energy, photoacclimation, and photoinhibition. With equal time in light and dark, the specific growth rate (μ) systematically decreased with increasing light duration, and µ decreased further when the ratio of light to dark was decreased. The model captured both trends with the mechanistic explanation that when the light duration was very short the changes in the pool of absorbed LI were smoothed out across the light and dark periods, whereas longer durations caused the biomass to experience discrete light and dark conditions that lead to reduced light absorption, more energy loss to nonphotochemical quenching, and more photodamage. These growth effects were accentuated as the ratio of light to dark decreased.  相似文献

The xanthophyll cycle and NPQ in diverse desert and aquatic green algae   总被引:1,自引:0,他引:1  
It has long been suspected that photoprotective mechanisms in green algae are similar to those in seed plants. However, exceptions have recently surfaced among aquatic and marine green algae in several taxonomic classes. Green algae are highly diverse genetically, falling into 13 named classes, and they are diverse ecologically, with many lineages including members from freshwater, marine, and terrestrial habitats. Genetically similar species living in dramatically different environments are potentially a rich source of information about variations in photoprotective function. Using aquatic and desert-derived species from three classes of green algae, we examined the induction of photoprotection under high light, exploring the relationship between nonphotochemical quenching and the xanthophyll cycle. In liquid culture, behavior of aquatic Entransia fimbriata (Klebsormidiophyceae) generally matched patterns observed in seed plants. Nonphotochemical quenching was lowest after overnight dark adaptation, increased with light intensity, and the extent of nonphotochemical quenching correlated with the extent of deepoxidation of xanthophyll cycle pigments. In contrast, overnight dark adaptation did not minimize nonphotochemical quenching in the other species studied: desert Klebsormidium sp. (Klebsormidiophyceae), desert and aquatic Cylindrocystis sp. (Zygnematophyceae), and desert Stichococcus sp. (Trebouxiophyceae). Instead, exposure to low light reduced nonphotochemical quenching below dark-adapted levels. De-epoxidation of xanthophyll cycle pigments paralleled light-induced changes in nonphotochemical quenching for species within Klebsormidiophyceae and Trebouxiophyceae, but not Zygnematophyceae. Inhibition of violaxanthin–zeaxanthin conversion by dithiothreitol reduced high-light-associated nonphotochemical quenching in all species (Zygnematophyceae the least), indicating that zeaxanthin can contribute to photoprotection as in seed plants but to different extents depending on taxon or lineage.  相似文献

The crossbreeding activities of the Schistosoma mansoni vector snail Biomphalaria glabrata were counted in a laboratory aquarium throughout the year under two regimes of 12h light: 12h dark from 7 A. M. to 10 P. M. Mating increased significantly in Autumn and Winter and just missed a significant inverse correlation with temperature and a direct one with locomotion. Other similar experiments were carried out to compare mating under various illumination conditions in complete daily cycle measurements. Mating counts decreased under the regimes which submitted snail to a total exposure of 12h light and 12h dark during a daily cycle in the following sequence: 12h light:12h dark alternating hourly with light gradient, 12h light:12h dark, 1h light:1h dark and 12h dark:12h light. Under two constant illuminations, the mating scored less than under the previous conditions, except under 12h dark:12h light. Under darkness the mating count was lower than under light conditions. There was no way to differentiate the night and day rhythms of mating on different days in each regime, except for mating under 12h light:12h dark alternating with light gradient, constant dark and 12h dark:12h light conditions. Mating increased in certain light and temperature conditions, in which the intensities should have an optimum value.  相似文献

Protonation of the lumen-exposed residues of some photosynthetic complexes in the grana membranes occurs under conditions of high light intensity and triggers a major photoprotection mechanism known as energy dependent nonphotochemical quenching. We have studied the role of protonation in the structural reorganization and thermal stability of isolated grana membranes. The macroorganization of granal membrane fragments in protonated and partly deprotonated state has been mapped by means of atomic force microscopy. The protonation of the photosynthetic complexes has been found to induce large-scale structural remodeling of grana membranes—formation of extensive domains of the major light-harvesting complex of photosystem II and clustering of trimmed photosystem II supercomplexes, thinning of the membrane, and reduction of its size. These events are accompanied by pronounced thermal destabilization of the photosynthetic complexes, as evidenced by circular dichroism spectroscopy and differential scanning calorimetry. Our data reveal a detailed nanoscopic picture of the initial steps of nonphotochemical quenching.  相似文献

A conserved regulatory mechanism protects plants against the potentially damaging effects of excessive light. Nearly all photosynthetic eukaryotes are able to dissipate excess absorbed light energy in a process that involves xanthophyll pigments. To dissect the role of xanthophylls in photoprotective energy dissipation in vivo, we isolated Arabidopsis xanthophyll cycle mutants by screening for altered nonphotochemical quenching of chlorophyll fluorescence. The npq1 mutants are unable to convert violaxanthin to zeaxanthin in excessive light, whereas the npq2 mutants accumulate zeaxanthin constitutively. The npq2 mutants are new alleles of aba1, the zeaxanthin epoxidase gene. The high levels of zeaxanthin in npq2 affected the kinetics of induction and relaxation but not the extent of nonphotochemical quenching. Genetic mapping, DNA sequencing, and complementation of npq1 demonstrated that this mutation affects the structural gene encoding violaxanthin deepoxidase. The npq1 mutant exhibited greatly reduced nonphotochemical quenching, demonstrating that violaxanthin deepoxidation is required for the bulk of rapidly reversible nonphotochemical quenching in Arabidopsis. Altered regulation of photosynthetic energy conversion in npq1 was associated with increased sensitivity to photoinhibition. These results, in conjunction with the analysis of npq mutants of Chlamydomonas, suggest that the role of the xanthophyll cycle in nonphotochemical quenching has been conserved, although different photosynthetic eukaryotes rely on the xanthophyll cycle to different extents for the dissipation of excess absorbed light energy.  相似文献

The photosynthetic apparatus in plants is protected against oxidative damage by processes that dissipate excess absorbed light energy as heat within the light-harvesting complexes. This dissipation of excitation energy is measured as nonphotochemical quenching of chlorophyll fluorescence. Nonphotochemical quenching depends primarily on the [delta]pH that is generated by photosynthetic electron transport, and it is also correlated with the amounts of zeaxanthin and antheraxanthin that are formed from violaxanthin by the operation of the xanthophyll cycle. To perform a genetic dissection of nonphotochemical quenching, we have isolated npq mutants of Chlamydomonas by using a digital video-imaging system. In excessive light, the npq1 mutant is unable to convert violaxanthin to antheraxanthin and zeaxanthin; this reaction is catalyzed by violaxanthin de-epoxidase. The npq2 mutant appears to be defective in zeaxanthin epoxidase activity, because it accumulates zeaxanthin and completely lacks antheraxanthin and violaxanthin under all light conditions. Characterization of these mutants demonstrates that a component of nonphotochemical quenching that develops in vivo in Chlamydomonas depends on the accumulation of zeaxanthin and antheraxanthin via the xanthophyll cycle. However, observation of substantial, rapid, [delta]pH-dependent nonphotochemical quenching in the npq1 mutant demonstrates that the formation of zeaxanthin and antheraxanthin via violaxanthin de-epoxidase activity is not required for all [delta]pH-dependent nonphotochemical quenching in this alga. Furthermore, the xanthophyll cycle is not required for survival of Chlamydomonas in excessive light.  相似文献

The response of CO2 fixation to a sudden increase in ambient CO2 concentration has been investigated in intact leaf tissue from spinach (Spinacia oleracea) using a dual channel infrared gas analyzer. Simultaneous with these measurements, changes in fluorescence emission associated with a weak, modulated measuring beam were recorded. Application of brief (2-3 seconds) dark intervals enabled estimation of the dark fluorescence level (Fo) under both steady state and transient conditions. The degree of suppression of Fo level fluorescence in the light was strongly correlated with nonphotochemical quenching under all conditions. During CO2-induced oscillations in photosynthesis under 2% O2 the changes in nonphotochemical quenching anticipate changes in the rate of uptake of CO2. At such low levels of O2 and constant illumination, changes in the relative quantum efficiency of open photosystem II units were estimated as the ratio of the rate of CO2 uptake and the photochemical quenching coefficient. Under the same conditions the relative quantum efficiency of photosystem II was found to vary inversely with the degree of nonphotochemical quenching. The relationship between changes in the rate of CO2 uptake: photochemical quenching coefficient and nonphotochemical quenching was altered somewhat when the same experiment was conducted under 20% O2. The results suggest that electron transport coupled to reduction of O2 occurs to varying degrees with time during oscillations, especially when ambient O2 concentrations are high.  相似文献

Local burning induces generation and propagation of variation potential (VP) in higher plants. VP induces transient inactivation of photosynthesis, which is possibly connected with proton signal in plant cell. Analysis of the role of changes in intracellular and extracellular pH in the VP-induced photosynthetic response in pea seedlings was the aim of this work. It was shown that local burning induced VP propagation, which was accompanied with a decrease of intracellular pH and increase of extracellular pH. VP induced photosynthesis inactivation that included an increase in the nonphotochemical fluorescence quenching and a decrease in the CO2 assimilation rate. Analysis of photosynthetic responses under control and low external CO2 concentration and changes in pH showed that there were two components in the responses. The first component appeared as a fast decrease of the CO2 assimilation and increase of nonphotochemical quenching. It depended on the activity of the dark stage of photosynthesis and was connected with apoplast alkalization. The second component was presented as a slow increase of nonphotochemical quenching. It weakly depended on a dark stage and was connected with a decrease of intracellular pH.  相似文献

The transition from dark to light involves marked changes in the redox reactions of photosynthetic electron transport and in chloroplast stromal enzyme activity even under mild light and growth conditions. Thus, it is not surprising that redox regulation is used to dynamically adjust and coordinate the stromal and thylakoid compartments. While oxidation of regulatory proteins is necessary for the regulation, the identity and the mechanism of action of the oxidizing pathway are still unresolved. Here, we studied the oxidation of a thylakoid-associated atypical thioredoxin-type protein, ACHT1, in the Arabidopsis thaliana chloroplast. We found that after a brief period of net reduction in plants illuminated with moderate light intensity, a significant oxidation reaction of ACHT1 arises and counterbalances its reduction. Interestingly, ACHT1 oxidation is driven by 2-Cys peroxiredoxin (Prx), which in turn eliminates peroxides. The ACHT1 and 2-Cys Prx reaction characteristics in plants further indicated that ACHT1 oxidation is linked with changes in the photosynthetic production of peroxides. Our findings that plants with altered redox poise of the ACHT1 and 2-Cys Prx pathway show higher nonphotochemical quenching and lower photosynthetic electron transport infer a feedback regulatory role for this pathway.  相似文献

Efficient light to biomass conversion in photobioreactors is crucial for economically feasible microalgae production processes. It has been suggested that photosynthesis is enhanced in short light path photobioreactors by mixing‐induced flashing light regimes. In this study, photosynthetic efficiency and growth of the green microalga Chlamydomonas reinhardtii were measured using LED light to simulate light/dark cycles ranging from 5 to 100 Hz at a light‐dark ratio of 0.1 and a flash intensity of 1000 µmol m−2 s−1. Light flashing at 100 Hz yielded the same photosynthetic efficiency and specific growth rate as cultivation under continuous illumination with the same time‐averaged light intensity (i.e., 100 µmol m−2 s−1). The efficiency and growth rate decreased with decreasing flash frequency. Even at 5 Hz flashing, the rate of linear electron transport during the flash was still 2.5 times higher than during maximal growth under continuous light, suggesting storage of reducing equivalents during the flash which are available during the dark period. In this way the dark reaction of photosynthesis can continue during the dark time of a light/dark cycle. Understanding photosynthetic growth in dynamic light regimes is crucial for model development to predict microalgal photobioreactor productivities. Biotechnol. An important factor in photosynthetic ecophysiology is the light regime that a photobiont is acclimated to exploit. In a wide range of cyanobacteria and cyano-lichens, the easily measured fluorescence parameters, coefficient of nonphotochemical quenching of photosystem II variable fluorescence (qN) and nonphotochemical quenching, decline to a minimum near the acclimated growth light intensity. This characteristic pattern predicts the integrated light regime to which populations are acclimated, information that is particularly useful for cyanobacteria or cyano-lichens from habitats with highly variable light intensities. qN reflects processes that compete with photosystem II photochemistry for absorbed excitation energy. In cyanobacteria, we find no evidence for energy-dependent quenching mechanisms, which are the predominant components of qN in higher plants. Instead, in cyanobacteria, qN correlates closely with the excitation flow from the phycobilisome to photosystem I, indicating that qN reflects the state transition mechanism for equilibration of excitation from the phycobilisome to the two photosystems.  相似文献

设置100%光环境(L100)、40%光环境(L40)、8%光环境(L8)3种光照梯度,分析万木林闽楠(Phoebe bournei)幼树的光合特性及生物量分配特征对不同光环境的响应。结果表明:(1)闽楠幼树在不同光环境下的最大净光合速率(Pnmax),表观量子效率(AQY),光饱和点(LSP),光补偿点(LCP),暗呼吸速率(Rd)均有显著差异(P<0.05),且随着光照强度的降低,Pnmax,LSP,LCP,Rd随之降低,而AQY却呈现升高的趋势。(2)总生物量、茎生物量、叶生物量、根生物量均表现为L100最大,根冠比、叶生物量比、茎生物量比及根生物量比在3种光环境下无显著差异。(3)闽楠幼树通过改变光合特性,生物量积累来适应光环境的变化,其中光照强度的降低限制了生物量的积累,但并未显著改变生物量地上地下分配比例。闽楠幼树在3种光环境下生物量分配比例并无显著的改变,生物量分配可塑性极低可能是闽楠零散分布的一个重要非人为干扰因素。(4)闽楠幼树所采取的生存策略以地上部分生长为主,光照强度降低时则采取保守策略进行缓慢的资源获取和消耗,全光照条件下采取快速的资源获取和消耗策略。未来造林时可以将闽楠与毛竹(Phyllostachys pubescens)混交来减少郁闭度,促进闽楠幼树的生长。  相似文献

We used an apparatus in which pieces of dark-adapted amphibian retinas (Rana pipiens, Bufo marinus) obtained under infrared illumination were exposed to precise intervals of 500-nm illuminations, and then frozen by contact of their outer segment surface with a liquid helium-cooled copper mirror. Sections of the frozen outer segment layer were obtained in a cryostat and then assayed for total extractable cyclic 3',5'-guanosine monophosphate (cGMP). Significant losses of cGMP with respect to the dark level were evident as early as 60 ms after light onset. With dim subsecond illuminations these losses were surprisingly large, which suggests a previously underestimated magnification in the cGMP cascade, or a transient substantial inhibition of guanylate cyclase activity in combination with increased cyclic GMP phosphodiesterase activity. Within the subsecond period, significant losses that were proportional to light intensity (2-log-unit range) and duration (60-550 ms) were generally not evident. However, losses significantly proportional to these factors became evident with durations of 1 s or longer. When pieces of retina were first illuminated (10 or 60 ms), then held in darkness for increasing periods before freezing, we observed a continuous loss of cGMP during the early postillumination dark period, followed by a recovery of the total cGMP level. The times for recovery to the preillumination level appear to be significantly longer than times reported for the recovery of the photoreceptor membrane potential after similar light exposures.  相似文献

The diatom algae, responsible for at least a quarter of the global photosynthetic carbon assimilation in the oceans, are capable of switching on rapid and efficient photoprotection, which helps them cope with the large fluctuations of light intensity in the moving waters. The enhanced dissipation of excess excitation energy becomes visible as non-photochemical quenching (NPQ) of chlorophyll a fluorescence. Intact cells of the diatoms Cyclotella meneghiniana and Phaeodactylum tricornutum, which show different NPQ induction kinetics under high light illumination, were investigated by picosecond time-resolved fluorescence under dark and NPQ-inducing high light conditions. The fluorescence kinetics revealed that there are two independent sites responsible for NPQ. The first quenching site is located in an FCP antenna system that is functionally detached from both photosystems, while the second quenching site is located in the PSII-attached antenna. Notwithstanding their different npq induction and reversal kinetics, both diatoms showed identical NPQ via both mechanisms in the steady-state. Their fluorescence decays in the dark-adapted states were different, however. A detailed quenching model is proposed for NPQ in diatoms.  相似文献

The relationship between steady-state photosynthetic efficiency, as moles CO2 per mole of incident visible photons under 2% O2, and chlorophyll fluorescence quenching has been investigated in intact leaf tissue of Spinacia oleracia. Fluorescence yield was measured using a pulse amplitude modulation technique that permitted rapid and sensitive resolution and quantitation of photochemical and nonphotochemical quenching coefficients. A highly linear relationship was observed between photosynthetic efficiency and the ratio of photochemical:nonphotochemical quenching coefficients for values of the latter less than 1.6. This relationship applied whether irradiance or CO2 concentration was varied. The observed relationships between photochemical yield and fluorescence yield were compatible with the photosystem II model proposed by Butler and Kitajima (1975 Biochim Biophys Acta 376: 116-125). The results are discussed with respect to the proposed role of nonphotochemical quenching in regulating radiant energy utilization and also the applicability of fluorescence measurements as a means of estimation of the rate of photosynthetic electron transport.  相似文献

This study investigated whether changes in illumination modify perception of day and night conditions in a diurnal species, the Indian weaver bird. Birds were initially subjected to a 12-h light:12-h dark regime (12L:12D; L=20 lux, D =0.5 lux). After every 2 wks, the combinations of light illumination in L and D phases were changed as follows: 20:2 lux, 20:5 lux, 20:10 lux, 20:20 lux, 20:100 lux, and 20:200 lux. Finally, birds were released into dim constant light (0.5 lux) for 2 wks to determine the phase and period of the circadian activity rhythm. They were also laparotomized at periodic intervals to examine the effects of the light regimes on the seasonal testicular cycle. All individuals showed a consistently similar response. As evident by the activity pattern under these light regimes, both in total activity during contrasting light phases and during the 2?h in the beginning and end of first light phase, birds interpreted the period of higher light intensity as day, and the period of lower intensity as the night. During the period of similar light intensity, i.e., under LL, birds free-ran with a circadian period ( ~ 24 h). In bright LL (20 lux), the activity rhythm was less distinct, but periodogram analysis revealed the circadian period for the group as 24.46 (+/-) 0.41 h (mean???SE). However, in dim LL at the end of the experiment, all birds exhibited a circadian pattern with average period of 25.52 (+/-) 0.70 h. All birds also showed testicular growth and regression during the 16-wks study. It is suggested that weaver birds interpret day and night subjectively based on both the light intensity and contrast between illuminations during two phases over the 24 h.  相似文献

A portable Chi fluorescence imaging system was used to characterise nonuniform Chi fluorescence quenching in Abutilon striatum leaves infected with phloem-localised abutilon mosaic virus. The instrument was used to observe fluorescence emission at intervals during induction transients, and to map nonphotochemical quenching during saturating pulses applied in the course of these transients. Two symptom types were distinguished: yellow vein-associated motifs that showed lower maximum Chi fluorescence than nearby green tissues, but virtually zero nonphotochemical quenching, and vein-defined mosaics (pale green) that initially showed normal maximum Chi fluorescence but strongly impaired nonphotochemical quenching. Mature vein-defined mosaics (yellow to white areas) resembled vein-associated symptoms with zero nonphotochemical quenching. Islands of apparently healthy green tissue enclosed by mosaic symptoms showed slower nonphotochemical quenching than controls. Possible effects of localised carbohydrate accumulation, thought to follow from infection by the phloem-limited virus, on photosynthetic processes as well as the synthesis and stability of chloroplast protein complexes, are discussed in the context of symptom development.  相似文献

We report on the changes the photosynthetic apparatus of Chlamydomonas reinhardtii undergoes upon acclimation to different light intensity. When grown in high light, cells had a faster growth rate and higher biomass production compared with low and control light conditions. However, cells acclimated to low light intensity are indeed able to produce more biomass per photon available as compared with high light-acclimated cells, which dissipate as heat a large part of light absorbed, thus reducing their photosynthetic efficiency. This dissipative state is strictly dependent on the accumulation of LhcSR3, a protein related to light-harvesting complexes, responsible for nonphotochemical quenching in microalgae. Other changes induced in the composition of the photosynthetic apparatus upon high light acclimation consist of an increase of carotenoid content on a chlorophyll basis, particularly zeaxanthin, and a major down-regulation of light absorption capacity by decreasing the chlorophyll content per cell. Surprisingly, the antenna size of both photosystem I and II is not modulated by acclimation; rather, the regulation affects the PSI/PSII ratio. Major effects of the acclimation to low light consist of increased activity of state 1 and 2 transitions and increased contributions of cyclic electron flow.  相似文献

The ability of purple photosynthetic bacteria to form phase variants (dissociants) was discovered. The R and M phase variants were isolated from the Rhodobacter sphaeroides population, and their affiliation with the parent strain was confirmed by the PCR of their 16S rRNA gene fragments. The R and M variants were different both in colony morphology and in resistance to some physical and chemical factors. The R variant of Rhb. sphaeroides had advantages when grown in the light, at elevated temperature, under aeration, and UV irradiation, whereas the M variant had advantages when grown aerobically in the dark or at increased NaCl concentrations.  相似文献
