Investigations have been made on the changes in the levels ofprotochlorophyll, chlorophyll a and chlorophyll b in relationto the kinetin induced expansion of isolated pumpkin cotyledonsin the presence and absence of chloramphenicol. It has been shown that rise in pigment level keeps pace withexpansion growth of the cotyledons. Kinetin markedly promotes the synthesis of protochlorphyll withoutmuch affecting the rate of its photoreduction to chlorophyll. Chloramphenicol strongly inhibits the development of both chlorophylla and b. The inhibition seems to be due to its interferenceboth with the synthesis of protochlorophyll and its subsequentconversion to chlorophyll. The inhibitory effect of chloramphenicol on the formation ofchlorophyll a is greater than on that of chlorophyll b, suggestingthereby the probability of divergent pathways for the formationof the two chlorophylls. N6-Benzyladenine (BA) was applied to intact bean (Phaseolusvulgaris L.) primary leaves at 2 and 6 days after imbibition,when they were in the cell division and post-cell division stages,respectively. BA treatment at day 2 temporarily inhibited an increase in chlorophyllcontent in the following day, but stimulated it in later days.No such inhibition by BA was observed for changes with timein DNA, RNA, and protein content and f. wt. On the other hand,BA treatment at day 6 enhanced RNA and protein content, withoutsignificant influence on DNA and chlorophyll content and f.wt. The mode of cytokinin action on greening in leaves during cell-divisiongrowth seems to be different from that in etiolated cotyledons. ATP concentration increased rapidly during the first 6h of imbibitionin cotyledons of cucumber and mung bean seeds. The increasewas strongly inhibited by 1-h treatment of tissues with cyanidein both species. Carbonylcyanide m-chlorophenylhydrazone, anuncoupler of oxidative phosphorylation, showed little effectduring the first hour of the treatment, but its inhibitory effecton ATP synthesis became significant after 3 h. Mitochondrialfractions prepared from 6-h-old cucumber cotyledons were capableof phosphorylating ADP to ATP. These results suggest that mitochondrialoxidative phosphorylation may be involved in ATP synthesis duringthe early hours of imbibition in both cucumber and mung beanseeds. Regeneration of photosynthetic activity and phycobilin pigmentswas studied with cells of Anabaena variabilis lacking photosystemII activity and phycobilin pigments. Regeneration was achievedonly when the cells were incubated in the presence of nitrateor nitrite. The addition of ammonium salts or urea was far lesseffective. Nitrate-directed regeneration was independent oflight and inhibited by chlorate. Dark-regenerated cells, however,differed from light-regenerated ones in that the former wereincapable of excitation transfer from phycocyanin to pigmentsystemII chlorophyll a, although they emitted fluorescence of pigmentsystem II chlorophyll a origin, if illuminated by the lightabsorbed by chlorophyll. The regeneration process inAnabaenacells is assumed to consist of two steps: [1] light-independent,nitratesupported synthesis of phycobilin pigments and photosystemII integrity, followed by [2] light-directed formation of excitationtransfer from phycocyanin to pigment system II chlorophyll a.An antibiotic study revealed that the former is associated withprotein synthesis, while the latter isnot. 1 Present address: Ocean Research Institute, University of Tokyo,Nakano, Tokyo 164, Japan. Carotenogenesis in mung been seedlings has been shown to beunder phytochrome control. Dark-grown seedlings contain moreindividual carotenoids than light-treated ones, but total carotenoidsare greatly increased by light. As expected, chlorophyll wassynthesized after a lag period on transferring etiolated seedlingsto the light, and there was an increase in total chlorophylland a parallel increase in total carotenoids. CPTA [2-(4-chlorophenylthio)-triethylamine hydrochloride]-treatedseedlings showed a decrease in both total carotenoids and totalchlorophylls. The usual effect of CPTA was also observed withmung bean seedlings, namely that lycopene, normally absent,appeared and increased to 17 per cent of total carotenoids afteran eight-day treatment. A cis-lycopene, possibly poly-cis-lycopene,was observed for the first time in CPTA-treated tissues. Onlyin the presence of CPTA but not on its removal was it possibleto show an inverse relationship between ß-caroteneand lycopene. This suggests that CPTA may not act as a cyclaseinhibitor in green tissues, cyclase being the enzyme/s involvedin the cyclization of carotenes. Etiolated cotyledons of squash (Cucurbita moschata Duch. var.melonaeformis Makino cv. Tokyo) treated with kinetin, KCl orfusicoccin in the dark increased both transpiration in the darkand chlorophyll synthesis after exposure to light. The minimumperiod of kinetin treatment to stimulate transpiration was similarto that to accelerate chlorophyll synthesis. On the other hand,treatment of cotyledons with vaseline or abscisic acid markedlyinhibited both transpiration and chlorophyll synthesis, suggestingthat cytokinin-induced stimulation of chlorophyll synthesiscould be mediated through the effect on the stomatal opening. Completely anhydrous fixation with acrolein vapour or osmiumtetroxide vapour was applied to tissues of air-dry seeds: thecoleoptile of wheat (Trilicum aestivum), and plumule and radicleof mung bean (Vigna radiata). Great shrinkage of cells and organelleswas noted, but all the usual organelles could be identified,except for Golgi bodies and (in most cases) ribosomes. The endoplasmicreticulum was very abundant and endoplasmic reticulum tubuleswere closely associated with the storage organelles, namelylipid bodies in the wheat coleoptile, and protein bodies inthe mung bean embryo axis. An in vitro procedure promoting the rapid growth and proteinincrease of soya bean cotyledons has been developed. The amountof protein synthesized varied greatly depending on the nitrogen(N) source provided. Glutamine was the most effective N source,while inorganic forms of N were ineffective. Growth and proteinsynthesis were both more rapid in vitro than in vivo. Underthe best conditions, soya bean cotyledons increased 8-fold bothin dry weight and in protein in 6 days. The formation of the7S and 11S storage proteins in vitro was similar to that invivo. Hence, this in vitro culture method is appropriate forstudying legume seed storage protein synthesis under controlledconditions.  相似文献

萌发绿豆的子叶自然衰老期间,核酸含量降低,RNA降低的幅度比DNA大。电泳分析结果表明,子叶衰老期间细胞核主带DNA明显降低;而迁移慢的卫星带DNA变化不大。在RNA各组分中,18S rRNA从衰老前期就开始降低;25S rRNA和4~5S小分子RNA到衰老后期才缓慢下降。DNase和RNase活性在子叶整个衰老期间都明显升高,是导致核酸含量下降的主要原因。~3H-核苷掺入试验表明,核酸的合成速率在子叶衰老前期有所上升,到衰老后期又降低。poly(A)~ -mRNA含量在子叶开始衰老时明显上升。  相似文献

Trypsin inhibitor was purified to homogeneity from seeds of the mung bean (Vigna radiata [L.] Wilczek). The protease inhibitor has the following properties: inhibitory activity toward trypsin, but not toward chymotrypsin; isoelectric point at pH 5.05; molecular weight of 11,000 to 12,000 (sodium dodecyl sulfate gel electrophoresis) or 14,000 (gel filtration); immunological cross-reactivity against extracts of black gram and black-eyed pea, but not against soybean; no inhibitory activity against vicilin peptidohydrolase, the principal endopeptidase in the cotyledons of mung bean seedlings.

The trypsin inhibitor content of the cotyledons declines in the course of seedling growth and the presence of an inactivating factor can be demonstrated by incubating crude extracts in the presence of β-mercaptoethanol. This inactivating factor may be a protease as vicilin peptidohydrolase rapidly inactivates the trypsin inhibitor. Removal of trypsin inhibitory activity from crude extracts by means of a trypsin affinity column does not result in an enhancement of protease activity in the extracts.

The intracellular localization of trypsin inhibitor was determined by fractionation of crude extracts on isopycnic sucrose gradients and by cytochemistry with fluorescent antibodies. Both methods indicate that trypsin inhibitor is associated with the cytoplasm and not with the protein bodies where reserve protein hydrolysis occurs. No convincing evidence was obtained which indicates that the catabolism of trypsin inhibitor during germination and seedling growth is causally related to the onset of reserve protein breakdown.

The nucleic acid, protein and uncombined amino acid contentof seeds of soya-bean (Glycine max L. Merr.), garden pea (Pisumsativum L.), kidney bean (Phaseolus vulgaris L.) and peanut(Arachis hypogaea L.) were measured at various times duringseed formation in an effort to understand why the soya-beanhas nearly twice as much protein as the other legume seeds.In all these species the concentration of deoxyribonucleic acid,ribonucleic acid and uncombined amino acids decreased duringseed formation. The protein level of kidney bean was relativelyconstant during development whereas the protein levels of pea,peanut and soya-bean increased during development. The proteincontent of the soya-bean increased throughout development whereasthe protein increase in peanut took place early and that inpea took place later in development. The ratio of protein toribonucleic acid was highest in peanut, less in soya-bean, andlowest in pea and kidney bean. Similarly, the ratio of proteinto deoxyribonucleic acid was higher in kidney bean than in soya-bean.Soya-beans had a lower amino acid content than any of the otherseeds at all stages of development. These results indicate thatneither total deoxyribonucleic acid, ribonucleic acid nor uncombinedamino acid content is responsible for the higher protein contentof soya-beans.  相似文献

Effluxes of K+ and Ca2+ from root segments of both wheat, Triticunaestivum L. cv. Capelle and mung bean, Vigna radiata (L.) Wilczek,were measured in the presence or absence of 20 mol m–3para-fluorophenylalanine (p-FPA). The results were used to estimatethe compartment contents and transmembrane K+ and Ca2+ fluxesin root cortex cells. Using the Ussing-Teorell flux equationas the criterion, it was concluded that entry of K+ from theoutside solution to the cytoplasm, and from the cytoplasm tothe vacuole were active in both wheat and mung bean. Also, inboth species, Ca2+ entered the cytoplasm passively across theplasmalemma and was actively pumped back to the external solution.However, interpretation of the direction of active transportacross the tonoplast depends upon an assumption about Ca2+ activityin the cytoplasm. The only qualitative effect of p-FPA was to alter the drivingforce for K+ influx, across the plasmalemma in wheat, from anactive to a passive one. Quantitative effects of the analoguewere seen for K+ fluxes in both wheat and mung bean and forCa2+ fluxes in wheat. The p-FPA reduced transport of K+ in bothspecies, while transport of Ca2+ was unaffected. The implicationsof these results for the ‘two pump hypothesis’ arediscussed. The development of mitochondrial NAD+-malate dehydrogenase (EC 1.1.1.37) in mung bean and cucumber cotyledons was followed. using the antibody raised against it, during and following germination. The developmental patterns were quite different between the two. In cucumber, the content of mitochondrial malate dehydrogenase continued to increase through 3–4 days after the beginning of imbibition. This was, at least in part, due to active synthesis of the enzyme protein, and the synthesis seemed to be regulated by the availability of the translatable mRNA for the enzyme. In mung bean, on the other hand, the enzyme was present in dry cotyledons at a rather high concentration, and remained at a constant level between day 1 and day 3 after the reduction of the content to one-half its initial level during the first day. De novo synthesis of the enzyme could not be detected in mung bean cotyledons by pulse-labeling experiment.  相似文献

Isolated cotyledons of Pinus pinaster were cultured under bud-formingconditions. The morphology, histology, lipid content and thechlorophyll pools of the cotyledons were followed during buddevelopment. Comparison with cotyledons cultured continuouslyon hormone-free or hormone-supplemented medium was performed.Three periods were distinguished during bud formation: (i) theinitiation period, characterized by a drastic decrease of triacylglycerols,enhanced by the presence of the hormones; (ii) the differentiationperiod, during which the intense cell divisions and the increaseof the polar lipid and chlorophyll contents appeared stronglycorrelated and (iii) bud development, characterized by stablelevels of both chlorophylls and neutral lipids. Moreover, itappeared that the continuous presence of hormones in the culturemedium inhibited the synthesis of molecular components, in particularchlorophylls and membrane lipids, required for normal cell differentiationand physiology. Four cytokinins have been separated from extracts of root nodulesof Phaseolus mungo by thin-layer chromatography. Their activitywas determined on the basis of their ability to induce betacyaninsynthesis in cotyledons of Amaranthus caudatus. Zeatin and itsriboside showed greater activity than N6 (2-(isopentenyl)) aminopurine and its riboside in the bioassay. Photosynthetic pigments extracted from the paniculate materialof the water column of Lake Kinneret were studied throughoutthe periods of May 1988-June 1989, and November 1993-November1994, by means of HPLC. The temporal and vertical variationof the pigment suite found agreed with the microscopically determinedphytoplankton record. The regression calculations of taxon-specificbiomass with the corresponding signature pigments suggest thatpigment analysis may be a useful tool for the monitoring ofbloom-forming species, e.g. the dinoflagellate Peridinium gatunenseNygaard. The HPLC pigment analysis permitted the identificationand quantification of chlorophyll degradation products, providingfor the first time information about their composition in LakeKinneret. Chlorophyllide a was the major detectable degradationproduct of chlorophyll a, varying between 1 and 9% of the chlorophylla concentration. Other chlorophyll a derivatives appeared mostlyin minor quantities. Pheophytin a was virtually lacking in allthe samples. Removal rates of pigments, measured by sedimentationtraps, indicated that the degradation of chlorophyll a via chlorophyllidea is a dynamic process that continues during the sedimentationof the phytoplankton particles.  相似文献

Germinated seeds of Lactuca sativa (L.) were placed in Petri-dishesin (2-chloroethyl)-trimethyl-ammonium chloride (CCC; 0.005–0.05M), KN03 (0.01 M), and KC1 (0.01 M) solutions, and incubatedfor 2 or 5 days under continuous light. CCC strikingly arrestedchlorophyll accumulation, and retarded cotyledon growth relativelylittle. The retardant inhibited 14C-leucine incorporation intobulk proteins of the cotyledons. KN03 and KC1 promoted cotyledongrowth and chlorophyll synthesis per cotyledon by about 150per cent, and about doubled protein synthesis. Potassium saltscompletely reversed the inhibitory effects of CCC on chlorophylland protein synthesis. It is suggested that the inhibition ofgreening by CCC is dependent on a prior inhibition of proteinsynthesis.  相似文献

By using the Giemsa C-banding technique, chromosome bandingpatterns on the somatic chromosomes of eight important pulsecrops, pea, lentil, guar (cluster bean), chick pea, pigeon pea,mung bean (green gram), urd (black gram) and cowpea have beenstudied. Each species has a characteristic C-banding pattern.The significance of such banding patterns which correlate withthe position of pachytene knobs, in chromosome identification,and in assigning relationships at the cytological level in thepulses of genus Vigna is stressed. 在绿豆子叶衰老达到不归点(萌发后5~6d)前切除上胚轴可使开始衰老的子叶中核酸和蛋白质含量回升,衰老短期逆转。衰老不归点后的子叶中核酸和蛋白质变化的主要特征是:丧失了较多的核主带DNA、25S、18S rRNA以及大部分可溶性蛋白质组分,一种小分子DNA 组分完全消失。不归点前切除上胚轴可使上述核酸和蛋白质组分明显增加,表明子叶衰老的逆转可能与这些重要功能物质的回升有关。在切除上胚轴的茎顶涂抹IAA,能阻止子叶核酸和蛋白质回升,也消除了切除上胚轴对子叶裹老的逆转作用。  相似文献

We studied the action mechanism of lycoricidinol, a plant growthinhibitor isolated from Lycoris radiata Herb. Lycoricidinolinhibited protein synthesis in mung bean hypocotyls, but notRNA synthesis. Protein synthesis in Escherichia coli was notaffected by the inhibitor. Results of in vitro translation experimentswith the wheat germ system and the E. coli system indicatedthat lycoricidinol inhibited only eukaryotic but not prokaryotictranslation. Use of specific inhibitors of initiation and polypeptidechain elongation of polypeptide synthesis revealed that chainelongation was inhibited by lycoricidinol. 1Permanent address: Department of Biology, Yonsei University,Seoul 120, Korea.
