Brenneria salicis resides in symptomless willow ( Salix spp.) and other tree species, but only willow trees develop watermark disease. To understand the conversion of B. salicis into a pathogen, its pathogenicity and differential growth in the various tree species are studied. Brenneria salicis was detected by plating and polymerase chain reaction-based techniques. Cell wall degradation and quorum sensing (QS) were assayed as possible pathogenicity mechanisms in wood . Differences in B. salicis growth capacities were tested in wood sap of the trees. Watermark diseased willow wood contained high concentrations of B. salicis with QS-induced cellulase activity. In the fall, wood sap of willow, and not of poplar and alder, promoted high density growth of B. salicis . In situ , B. salicis was the dominant bacterial type in willow wood during the fall and winter period. Willow sustains high densities of B. salicis at the time of leaf shedding. The cellulase in the immobilized wood sap has then a long-lasting contact with the xylem cell wall. Timing of dormancy and subsequent winter conditions might interfere with sap composition, B. salicis density, activity and survival, and be the reason, at least partly, for the variable occurrence of the disease.  相似文献

Brenneria salicis has been studied in willow wood only in relation to watermark disease. In this pathogenic condition, the bacterium occurs at high concentrations. Pathogenicity of B. salicis is still uncontrollable and the disease unpredictable because the plant–bacteria interaction is not understood. Thanks to molecular techniques B. salicis can be detected at low concentrations, which are found in most non-pathogenic interactions. Brenneria salicis was identified and traced with a new specific three-primer polymerase chain reaction and its identity and relative concentration in biological samples confirmed through denaturing gradient gel electrophoresis profiling. Brenneria salicis was found in symptomless willows sampled randomly in Flanders agricultural areas, in young nursery willows, and also in poplar ( Populus ) and alder ( Alnus ). It harboured the nitrogenase reductase gene NifH and promoted growth and chlorophyll in willow. Inoculated luminescence-marked B. salicis circulated through the whole plant without inducing disease and exuded at the leaf margins. Other willow endophytes identified were Rahnella , Sphingomonas and Methylobacterium . In conclusion, because endophytic B. salicis is generally observed in willow, disease must not be dependent on infection. Leaf-to-leaf spread is proposed as an important mechanism for spread of B. salicis .  相似文献

The bacterium Brenneria salicis is the causal agent of watermark disease in willow. This work shows the importance of in situ studies and high-resolution separation of biological samples with ultrahigh performance liquid chromatography combined with ion trap mass spectrometry to unambiguously identify molecular compounds associated with this disease. Approximately 40 oligolignols accumulated in wood sap of watermark diseased willow, and are indicative for degradation of the xylem cell wall, of which 15 were structurally assigned based on an earlier study. Many bacteria are known to produce and release quorum sensing signal molecules that switch on the expression of specific, sometimes pathogenic functions. Two quorum sensing signal molecules, N-(3-oxohexanoyl)-l-homoserine lactone and N-(hexanoyl)-l-homoserine lactone, were present in 4/1 ratios in diseased wood and in high-density in vitro cultures of B. salicis at 0.13-1.2muM concentrations, and absent in healthy wood and in low-density in vitro cultures of B. salicis. Although it is not a proof, it can be an indication for involvement of quorum sensing in B. salicis pathogenesis. Cyclic dipeptides were present at high concentrations in high-density in vitro cultures of B. salicis, but not in situ, and were found not to be involved in quorum sensing signaling, therefore, the attribution of quorum signal properties to cyclic dipeptides isolated from in vitro cultures of pathogenic bacteria should be reconsidered.  相似文献

Eight apple genotypes, including cultivars and breeding selections resistant and susceptible to Venturia inaequalis on foliage, were screened for shoot infection and the development of wood pustules following inoculation of shoot tips of 1–year maiden trees in the greenhouse. Where genotypes were highly resistant in terms of foliar symptoms (cvs Prima and Gavin), no shoot infection was observed. Where genotypes were highly susceptible in terms of foliar symptoms, then shoot tissue was either resistant (cv. Shinko) or susceptible (cv. Starking). In one experiment, shoot tips were inoculated sequentially as shoots extended. No pustules developed where inoculations were made later than May. In a further experiment, two leaf internode positions were inoculated on one occasion. Pustules were only observed where inoculations were made above the youngest unrolled leaf. The results suggested that shoot tips were more susceptible during early extension growth of the shoot. Pustules were noted in abundance on petioles of susceptible cultivars, and these probably contributed to early leaf abscission.  相似文献

The effect of two species of stem‐feeding aphid on the yield, phenology and water‐use of soil‐grown and hydroponic‐cultured willow were investigated. The effects of Pterocomma salicis were less marked than those of Tuberolachnus salignus. The latter reduced not only the above and below‐ground growth of trees made during and subsequent to infestation, but also altered the mass of previously developed woody tissue. The negative effects of T. salignus on the shoots and roots of established and establishing trees were drastic and were both quantitative and qualitative. They were also observed to reduce the survival of infested trees. P. salicis had negative effects on the growth of roots and shoots during infestation, but the influence of this species did not persist nor did it influence previous growth.  相似文献

1 In 1996, 7000 ha of pine forests were defoliated by the pine looper Bupalus piniaria in south‐western Sweden. 2 The susceptibility of trees of different defoliation classes (0, 30, 60, 90 and 100% defoliation) to beetle‐vectored blue‐stain fungi was tested in inoculation experiments. Forty and 120‐year‐old Scots pine trees were inoculated with ‘single’, i.e. a few inoculations of Leptographium wingfieldii and Ophiostoma minus, two blue‐stain fungi associated with the pine shoot beetle Tomicus piniperda. The young trees were also ‘mass’ inoculated with L. wingfieldii at a density of 400 inoculation points per m2 over a 60 cm stem belt. 3 Host tree symptoms indicated that only trees with 90–100% defoliation were susceptible to the mass inoculation. 4 Single inoculations did not result in any consistent differences in fungal performance between trees of different defoliation classes, regardless of inoculated species or tree age class. 5 Leptographium wingfieldii produced larger reaction zones than O. minus, and both species produced larger lesions in old than in young trees. 6 As beetle‐induced tree mortality in the study area occurred only in totally defoliated stands, mass inoculations seem to mimic beetle‐attacks fairly well, and thus seem to be a useful tool for assessing host resistance. 7 As even severely defoliated pine trees were quite resistant, host defence reactions in Scots pine seem to be less dependent on carbon allocation than predicted by carbon‐based defence hypotheses.  相似文献

Control of the invasive wasp, Sirex noctilio Fabricius using the parasitic nematode Deladenus siricidicola Bedding is a well known example of a successful classical biological control program. Despite its wide-scale success, this control method has recently had poor success in the summer rainfall areas of South Africa. Data from previous studies showed variation in nematode parasitism from inoculated trees (inoculation success) between different tree sections and among inoculation times. They also pointed to moisture content of the wood or virulence of the nematode as the most likely underlying factors influencing variations in inoculation success. The results from our study showed that the highest levels of parasitism was obtained from early inoculations and from the bottom sections of trees, where moisture content of the wood was highest, supporting the hypothesis that moisture content influences parasitism. However, even when moisture content was adequate, average inoculation success remained below 25% and was often 0%, suggesting that there are other barriers to inoculation success. Different sources from which the nematodes were produced did not influence inoculation success, indicating that nematode virulence is most likely not the cause of the low success. Another interesting finding was that parasitized wasps were larger than unparasitized wasps. Background parasitism was present despite the poor success with past inoculations, but the data also suggest that the natural build-up of this population could be constrained by the same factors that influence inoculations.  相似文献

Controlled inoculation of spruce seedling needle crowns and of shoots of 4-year-old spruce trees by Sirococcus conigenus led to disease symptoms (discoloration and necrosis) and to the induction of phenolic metabolites. Even upon complete infection, as proved by re-isolation of the pathogen from inoculated seedlings, only 40% of the plants developed visible disease symptoms after 38 days. A Sirococcus-specific polymerase chain reaction (PCR) primer pair, SIRO1 and SIRO6, was designed based on sequences of a RAPD fragment. The primer pair permitted the detection of 1 pg fungal DNA (10-40 genomes) in 1 mg fresh weight spruce tissues (needles, bark, wood), regardless of visible disease symptoms. The amounts of the major phenolic compound of spruce needles, catechin, increased significantly in all of the five spruce provenances as a response to inoculation with Sirococcus. The second major phenolic compound, picein, increased in three of the provenances, whereas the remaining two had high concentrations to begin with and showed no reaction. Minor phenolic compounds increased in response to infection regardless of provenance. In a preliminary field study, Sirococcus infection of spruce was detectable by PCR even in the presence of massive infection by other fungi, such as Rhizospaera spp. and Lophodermium spp.  相似文献

Injury or inoculation of apparently healthy citrus trunk bark and wood with Fusarium solani immediately prior to and after the December 1983 freeze resulted in cankers similar to natural branch and trunk cankers which formed on trees following the freeze. Fusarium solani was consistently isolated from active lesions of natural and induced cankered bark. Phytophora spp. were not isolated from cankered tissue. Earliest cankers appeared as a water-soaked area under the bark around the point of inoculation, later F. solani sporodochia formed on the bark surface. Older cankers became dry and cracked but produced no gum. Canker size ranged from 10–90 cm vertical diameter in spring 1984 inoculations but were only several cm diameter following late summer inoculations. Active cankers developed again after inoculation following the January 1985 freeze. Similar cankers were produced on Hamlin sweet orange stems inoculated with F. solani.  相似文献

Longan witches’ broom (LgWB) syndrome was observed in all longan-growing provinces of Vietnam. Key symptoms include: small, stunted shoots with curved, rolled up margins, deformed leaves with blisters and hairy patches on the underside; abnormal development of flower structures, flower abortion, failure to produce fruits or developing small fruits. During 2012–2015, field surveys and study of the relationship between LgWB symptoms and Eriophyes dimocarpi mites presence were carried out. Moreover, molecular analysis, electron microscopy observations, E. dimocarpi inoculations, pruning of affected spikelets and miticide experiments were performed. An average of 36.58% of longan seedlings inoculated with E. dimocarpi mites showed typical symptoms of LgWB at about 27.5 days after inoculation. The use of several pesticides in different combinations showed high efficacy on E. dimocarpi mites elimination and of symptomatology incidence reduction. These results suggest a significant association of E. dimocarpi with LgWB syndrome in Vietnam.  相似文献

Willows in the Flemish region are threatened by the watermark disease, a wood disease caused by the bacterium Brenneria salicis (Hauben et al. Syst Appl Microbiol 21:384–397, 1998). Affected trees are observed to grow at specific sites which seem more sensitive to periodical watermark symptom development than other sites. In this study we aim at revealing abiotic site factors that could play a role in triggering the watermark disease. During three consecutive years we analyzed stands of young isogenic willows (Salix alba L. cv. Lichtenvoorde), which were planted on four different sites in an agricultural environment. These test sites were chosen, based on the occurrence/absence of watermark disease in the old willows that grow on these sites. Two sites had old willow pollards with clear watermark symptoms, and on the two other sites the old willows were unaffected. Differences in abiotic characteristics between the sites were analysed and their effects on the nutrient status in willow were measured in the isogenic willows that were newly planted. Brenneria salicis-specific multiplex PCR revealed the presence of this pathogenic bacterium in both the old and the freshly planted young willows. At each site, the physical and chemical characteristics of soil and groundwater were investigated; the young tree growth was monitored and nutritional status of the willow plantings was assessed by foliar analysis. Physical soil properties including soil type and profile water availability, and chemical soil analysis were unable to explain differences between healthy and affected sites. Diseased sites, however, had significantly higher N concentrations and traces of herbicides in the groundwater. In young trees, foliar levels of N and S were always greater at the affected sites, while P contents were lower. Dry mass based N concentration in the leaves of the newly planted trees increased systematically over the years, but more at affected than at healthy sites. Also, chlorophyll content was significantly greater at the affected sites. Growth rate of the young isogenic trees was twice as fast at the diseased sites. The presented data suggest that application of fertilisers in agriculture causes an imbalance in the nutritional status of Salix alba L. Excess nitrogen makes this species fast-growing, reflected by an average foliar N:P ratio >10, and more susceptible for watermark disease, indicated by an average foliar N:Ca ratio exceeding 1.5.  相似文献

The influence of Pleurotus ostreatus inoculation on wood degradation and on fungal community structure was studied. The experiments were performed on an organically poor fly ash deposit covered with a 10 cm layer of beech wood chips inoculated with P. ostreatus isolate ZIM76. Compared to non-inoculated wood chips, inoculation increased the temperatures and relative humidities and, in the first 6 months, accelerated Klason lignin degradation by 9% and also, after 17 months, increased iron translocation into wood chips by 30%. After 6 months, PCR-DGGE showed 22-28 and 13-21 fungal taxa in non-inoculated and P. ostreatus-inoculated beech chips, respectively. The differences in number of taxa and in the fungal community structure (based on Dice coefficient) between non-inoculated and inoculated wood chips diminished with time. The results indicate that the naturally occurring processes of wood degradation are as efficient as those occurring in sites inoculated with P. ostreatus.  相似文献

Metabolic fingerprinting to discriminate diseases of stored carrots   总被引:1,自引:0,他引:1  
Volatile metabolites from headspace gas of carrot cv. Vita‐treat inoculated with water or four different pathogens Botrytis cinerea, Erwinia carotovora subsp. carotovora, Aspergillus niger and Fusarium avenaceum were profiled using gas chromatography and mass spectrometry to develop a technology to discriminate diseases. The inoculation of carrot roots with water or different pathogens released a total of 137 different volatile metabolites. Among them, 39 compounds were relatively consistent and 11 were specific to one or more diseases/inoculations. E. carotovora subsp. carotovora produced seven disease‐specific metabolites: 1‐butanol, 3‐methyl; 1‐pentanol; 1‐propanol, 2‐methyl; 2,3‐butanedione; boronic acid, ethyl; butane, 1‐methoxy‐3‐methyl; and ethane, ethoxy. Some metabolites were disease/inoculation discriminatory and were not detected in all treatments: 1,2‐dimethoxy‐ethene was common in carrots inoculated with E. carotovora subsp. carotovora and B. cinerea, while 2‐butanone, 3‐chloro‐4‐hydroxy‐1,4‐diphenyl was common in carrots inoculated with E. carotovora subsp. carotovora, F. avenaceum and water‐inoculated control. The significant mass ions, based on univariate analysis, from a total of 150 (46–195 m/z) and compounds from a total of 32 were further subjected to stepwise discriminant analysis and discriminant analysis. The models for 3 days after inoculation (DAI) were better than those for 6 DAI and 3 + 6 DAI, where up to 90% of the observations were correctly classified into respective inoculations. The disease‐discriminatory compounds from different diseases/inoculations and discriminant analysis models developed here have the potential for the early detection and discrimination of postharvest diseases of carrot cv. Vita‐treat, after validation under commercial conditions.  相似文献

The atypical symptoms first described by Bryan (1932) of the angular leaf spot disease of cotton caused by Xanthomonas malvacearum (E. F. Sm.) Dowson were reproduced by inoculation into seeds, stem apices or buds. The lesions that developed on the veins of the newly produced leaves were elongated and water-soaked, becoming dark brown. The leaf tissue dependent upon infected veins became yellow, flaccid and withered. The development of these symptoms was enhanced when inoculations were made into opening buds or germinating seeds as compared with inoculations into closed buds or dormant seeds.
In other bacterial diseases caused by Xanthomonas spp., somewhat atypical symptoms could also be produced by bud inoculation into the appropriate host. Those produced by X. ricini (Yoshi & Takimato) Dowson on castor, closely resembled the vein lesions described above on cotton but resulted only from bud inoculations; inoculations into stem apices and seeds failed to produce them. In dolichos bean inoculated with X. phaseoli (Smith) Dowson, atypical symptoms were produced only by seed inoculations and were confined to the first simple leaves (prophylls).
The differences in the production of atypical symptoms on the three hosts are correlated with differences in host structure and with the degree of virulence of the pathogen. The leaf parasite X. ricini , for example, which cannot infect castor bean stems, does not produce atypical symptoms when inoculated below the stem apex.
From the data discussed below, the incidence of atypical symptoms is attributed to infection either of an actively growing tissue or of a telescoped structure which subsequently completes its development.
The atypical symptoms of the cotton disease are not caused by a special strain of X. malvacearum. Further, they are not a peculiarity of this disease but may also develop in other necrotic diseases under similar conditions.  相似文献

Interactions of Puccinia hordei and Erysiphe graminis on seedling barley   总被引:1,自引:0,他引:1  
The development of Puccinia hordei on the first leaf of barley seedlings previously inoculated with Erysiphe graminis was compared with that on uninoculated leaves of comparable age. On cv. Zephyr, more rust pustules developed when leaves were inoculated with both fungi within 24 h but fewer pustules if the period between the two inoculations was longer than 2 days. The reduction in numbers of rust pustules was especially marked where leaves were previously inoculated with many conidia of E. graminis. The size of rust pustules was reduced whatever the period between the two inoculations. Arresting mildew development by applying ethirimol as a soil drench to pots of seedlings inoculated with E. graminis 6 days previously, or floating segments of leaves inoculated with both fungi on 2% sucrose, in part counteracted these effects on rust pustule size. Similar effects were observed with cv. Mazurka where inoculations with E. graminis produced only small necrotic flecks but did induce premature loss of chlorophyll. On this cultivar (in contrast to Zephyr) the inoculation of one leaf surface affected the development of P. hordei on the other. In comparable experiments using Zephyr, E. graminis produced smaller colonies with fewer conidiophores on leaves previously inoculated with P. hordei. These effects could be alleviated by arresting rust development with a spray containing benodanil or by floating segments of leaves inoculated with both fungi on 2% sucrose. Germination of the conidia of E. graminis, formation of appressoria and initiation of colonies were not affected by the presence of P. hordei.  相似文献

The influence of controlled temperatures on levels of sorbitol and other carbohydrates was determined to provide further information on dormancy of apple trees (Malus sylvestris Mill.). For 3 years, 2-year-old “Red Delicious” apple shoots were collected from mature trees in an orchard at intervals during the autumn and winter, and shoots were stored for 6 hr to 1 week at temperatures from 18 to ?60 °C. Sorbitol and other carbohydrates were estimated in the sap or wood by gas chromatography.Controlled temperatures had a marked influence on the carbohydrate content of excised 2-year-old apple shoots. Levels of sorbitol in the sap were maximum at ?0.6 °C. The increase was greatest at the earliest sampling before complete hardening had occurred in each of the 3 years tested. Total sorbitol in the wood was less influenced by storage at various controlled temperatures than sorbitol in the sap. Levels of fructose, glucose, and sucrose in the wood were higher at temperatures below ?0.6 °C than at warmer temperatures. Levels of starch were usually inversely related to soluble sugars.  相似文献

Preparations of methyl benzimidazol-2-ylcarbamate (MBC) hydrochloride stabilized with HC1, MBC nitrate stabilized with lactic acid and MBC bisul-phate stabilized with KHS04 were made from technical MBC. As it was the least phytotoxic to elm shoots, the hydrochloride was selected for field experiments on the control of Dutch elm disease using 6 m elms. In protectant experiments using 1.51 of 0.25 or 0.5 % MBC per tree, only five of eighteen injected trees became infected while all the control trees were severely diseased. In curative experiments, injection with 0.25 % MBC 2 and 4 wk after inoculation with Ceratocystis ulmi kept symptoms to < 1 and 5% respectively, compared with 76% in untreated inoculated trees. Pseudomonas fluorescens EPS62e was selected during a screening procedure for its high efficacy in controlling infections by Erwinia amylovora, the causal agent of fire blight disease, on different plant materials. In field trials carried out in pear trees during bloom, EPS62e colonized flowers until the carrying capacity, providing a moderate efficacy of fire-blight control. The putative mechanisms of EPS62e antagonism against E. amylovora were studied. EPS62e did not produce antimicrobial compounds described in P. fluorescens species and only developed antagonism in King's B medium, where it produced siderophores. Interaction experiments in culture plate wells including a membrane filter, which physically separated the cultures, confirmed that inhibition of E. amylovora requires cell-to-cell contact. The spectrum of nutrient assimilation indicated that EPS62e used significantly more or different carbon sources than the pathogen. The maximum growth rate and affinity for nutrients in immature fruit extract were higher in EPS62e than in E. amylovora, but the cell yield was similar. The fitness of EPS62e and E. amylovora was studied upon inoculation in immature pear fruit wounds and hypanthia of intact flowers under controlled-environment conditions. When inoculated separately, EPS62e grew faster in flowers, whereas E. amylovora grew faster in fruit wounds because of its rapid spread to adjacent tissues. However, in preventive inoculations of EPS62e, subsequent growth of EPS101 was significantly inhibited. It is concluded that cell-to-cell interference as well as differences in growth potential and the spectrum and efficiency of nutrient use are mechanisms of antagonism of EPS62e against E. amylovora.  相似文献

We examined the effects of trunk cutting on the regrowth responses of two dominant willow species, Salix eriocarpa and S. gilgiana , and the subsequent effects on the community structure (abundance and species richness) of herbivorous and predaceous arthropods. We studied ten randomly selected pairs of cut and uncut (control) trees of each willow species. Field observations showed that when the trunks were cut, shoots sprouted from the base and developed rapidly. These shoots continued to grow until July, producing lateral shoots and leaves, whereas current-year shoots on uncut trees essentially stopped growing by April. In July, the upper leaves of cut trees were less tough and had a greater water and nitrogen content than leaves of uncut trees. Leaf consumption and abundance and species richness of both herbivorous and predaceous arthropods were significantly greater on cut trees than on uncut trees in both willow species. Overall, trunk cutting resulted in at least a two-fold increase in both relative abundance and species richness within the arthropod community on the willow species. We concluded that the severe physical damage caused by trunk cutting greatly increased the biodiversity of herbivorous and predaceous arthropods through bottom-up cascading effects.  相似文献

  • There are no records of established plant pathogenic Phytophthora species in Finnish forests, but they are likely in the future. Therefore, the effects of Phytophthora inoculations on young, ca. 2‐month‐old silver birch (Betula pendula) seedling roots and shoots were investigated.
  • Visual inspection of dark discoloration, direct PCR and re‐isolation, and detailed root morphology analyses were used to evaluate the effects of Phytophthora inoculation on roots. Symptoms in leaves and stems were also recorded.
  • Phytophthora was successfully re‐isolated from 67% of the surface‐sterilized roots of inoculated seedlings, but not from the non‐inoculated control seedlings. Dark discolorations were found more often in the root segments of inoculated seedlings than in control seedlings. In the Phytophthora‐treated seedlings, discoloured root segments were usually linked and found primarily in the main root or lateral roots attached to it, whereas in the control seedlings a few single discoloured root segments were scattered throughout the root systems. The number of root segments was lower in the inoculated than in the control seedlings, indicating root loss after Phytophthora inoculation. In the shoots of inoculated birches, leaf and shoot wilting was observed.
  • The appearance of wilting in shoots without visible dark discoloration in the base of stems indicated that symptoms originated from roots inoculated with Phytophthora.
